Abstract:
On a beef carcass, Escherichia coli may sequentially encounter acid- and heat-intervention steps. This study tested whether acid stress (1.5% [vol/vol] acetic acid, pH 4.0, 37°C, 15 min) would enhance subsequent heat resistance of E. coli. Initially, cells (E. coli O157:H7 ATCC 43894, nonpathogenic E. coli B [strain FRIK-124], and rpoS-deficient mutant 813-6 [derived from E. coli O157:H7 ATCC 43895]) were acid stressed and transferred to 54°C tiypticase soy broth (TSB), and survivors were immediately enumerated after at least three intervals of 12, 2, and 6 min, respectively, by plating. The ATCC 43894 and 813-6 strains survived the acid stress but strain FRIK-124 did not. Acid-stressed ATCC 43894 had significantly lower D values than the non-acid-stressed controls. Strain 813-6 had significantly lower D values than strain ATCC 43894, with no significant difference between acid-stressed and non-acid-stressed cells. In a second experiment, cooling of cells prior to plating resulted in an increased D value for acid-stressed ATCC 43894 cells, such that it was not significantly different from the D value for non-acid-stressed Controls. Using this protocol, there was no significant difference in D values between acid-stressed and non-acid-stressed ATCC 43894 cells in prewarmed TSB (54, 58, and 62°C), in prewarmed ground beef slurry (GBS; 58°C), or in TSB and GBS inoculated at 5°C and heated to 58°C. The acid stress tested does not enhance subsequent heat resistance of E. coli.

Abstract:
The surveillance and virological characterization of H5N8 avian influenza viruses are important in order to assess their zoonotic potential. The genetic analyses of the Egyptian H5N8 viruses isolated through active surveillance in wild birds and domestic poultry in the winter of 2016/2017 showed multiple introductions of reassortant viruses. In this study, we investigated and compared the growth kinetics, infectivity, and pathogenicity of the three reassortant forms of H5N8 viruses detected in wild birds and domestic poultry in Egypt during the first introduction wave in the winter of 2016/2017. Three representative H5N8 viruses (abbreviated as 813, 871, and 13666) were selected. The 871/H5N8 virus showed enhanced growth properties in vitro in Madin Darby canine kidney (MDCK) and A549 cells. Interestingly, all viruses replicated well in mice without prior adaptation. Infected C57BL/6 mice showed 20% mortality for 813/H5N8 and 60% mortality for 871/H5N8 and 13666/H5N8, which could be attributed to the genetic differences among the viruses. Studies on the pathogenicity in experimentally infected ducks revealed a range of pathogenic effects, with mortality rate ranging from 0% for 813/H5N8 and 13666/H5N8 to 28% for 871/H5N8. No significant differences were observed among the three compared viruses in infected chickens. Overall, different H5N8 viruses had variable biological characteristics, indicating a continuous need for surveillance and virus characterization efforts.

The purpose of this study was to investigate the prevalence and etiology of hearing loss in primary and middle school students in the Hubei province of China. During a 2-year period, 504,348 students were examined by a speech audiometry test, and 813 deaf students were detected. Among the 813 deaf students, 232 cases were diagnosed with congenital deafness and 560 cases had acquired deafness, among which 276 cases had aminoglycoside-antibiotic-induced deafness. The severity of deafness could be further ascertained in 804 other students, with 402 profound, 363 severe, 21 moderate and 18 mild deafness cases. The age at deafness onset of most students was under 3 years. The prevalence of congenital and acquired deafness was 0.046% (232/504,348) and 0.111% (560/504,348), respectively, much lower than previously reported in other regions of China. This study aims to adapt the Gratitude Questionnaire (GQ-6) to the Philippines, gathering evidence of its validity and reliability. Two studies were conducted. Participants in Study 1 were 340 college students (Mage= 20.63; 62.1% female), who completed the GQ-6 and demographic questions. The exploratory factor analysis was performed, indicating a one-factor solution (a= .80). Participants in Study 2 were 813 college students (Mage= 19.99 years; 50.1% male), who answered the GQ-6, the Life Orientation Test-Revised, the Subjective Happiness Scale, and the Spirituality/Religiousness items. Results corroborated the one-factor structure (e.g., CFI= .98, RMSEA= .05) showing evidence of its association with life orientation (r= .29), subjective happiness (r= .08), and religiosity (r= .31). The scores from the GQ-6 also exhibited invariance across gender. In conclusion, the GQ-6 provide evidence of factorial and criterion validity and reliability, justifying its use in the Philippines.

ABSTRACT We report on the discovery and validation of TOI 813 b (TIC 55525572 b), a transiting exoplanet identified by citizen scientists in data from NASA’s Transiting Exoplanet Survey Satellite (TESS) and the first planet discovered by the Planet Hunters TESS project. The host star is a bright (V = 10.3 mag) subgiant ($R_\star =1.94\, R_\odot$, $M_\star =1.32\, M_\odot$). It was observed almost continuously by TESS during its first year of operations, during which time four individual transit events were detected. The candidate passed all the standard light curve-based vetting checks, and ground-based follow-up spectroscopy and speckle imaging enabled us to place an upper limit of $2\, M_{\rm Jup}$ (99 per cent confidence) on the mass of the companion, and to statistically validate its planetary nature. Detailed modelling of the transits yields a period of $83.8911 _{ - 0.0031 } ^ { + 0.0027 }$ d, a planet radius of 6.71 ± 0.38 R⊕ and a semimajor axis of $0.423 _{ - 0.037 } ^ { + 0.031 }$ AU. The planet’s orbital period combined with the evolved nature of the host star places this object in a relatively underexplored region of parameter space. We estimate that TOI 813 b induces a reflex motion in its host star with a semi-amplitude of ∼6 m s−1, making this a promising system to measure the mass of a relatively long-period transiting planet.

The effect of surface modification on the tensile strength of the bonded interface of Al alloy and SUS304 stainless steel was investigated by SEM observations of interfacial microstructures and fractured surfaces. Aluminum surfaces were modified by boiling in 5% aqueous solution of NaOH for 20 s and 99.7% Acetic acid for 60 s. Bonding was performed at bonding temperature of 753 ~ 813 K under a pressure of 6 MPa (bonding time of 1.8 ks). As a result of surface modification, bonded joints were obtained at a bonding temperature 20 K lower than that required for non-modified surfaces, and the bonded strength was comparable to that of the maximum load.

LiNiVO4 was synthesized using Li2CO3, NH4VO3 and Ni(CH3COO)2.4H2O as starting reagents and citric acid as a chelating agent. Mole ratios of metals to citric acid used for the preparation were 1 : 1 - 1 : 4. Carboxylate precursors were calcined at high temperatures to form powder. TGA data showed weight loss due to the evaporation and decomposition processes. FTIR showed the stretching bands of VO4 tetrahedron at 642, 713 and 813 cm-1. For 1 : 3 and 1 : 4 mole ratios, nano-crystals of inverse spinel LiNiVO4 (produced at 450 oC calcination for 6 h) were detected by XRD, electron diffraction, SEM and TEM.

Rietveld neutron powder profile analysis of the compound Na0.5Bi0.5TiO3 (NBT) is reported over the temperature range 5–873 K. The sequence of phase transitions from the high-temperature prototypic cubic structure (above 813 K), to one of tetragonal (673–773 K) and then rhombohedral structures (5–528 K) has been established. Coexisting tetragonal/cubic (773–813 K) and rhombohedral/tetragonal (with an upper temperature limit of 145 K between 528 and 673 K) phases have also been observed. Refinements have revealed that the rhombohedral phase, space group R3c, with a H = 5.4887 (2), c H = 13.5048 (8) Å, V = 352.33 (3) Å3, Z = 6 and D x = 5.99 Mg m−3, exhibits an antiphase, a − a − a − oxygen tilt system, ω = 8.24 (4)°, with parallel cation displacements at room temperature. The tetragonal phase, space group P4bm, with a T = 5.5179 (2), c T = 3.9073 (2) Å, V = 118.96 (1) Å3, Z = 2 and D x = 5.91 Mg m−3, possesses an unusual combination of in-phase, a 0 a 0 c + oxygen octahedra tilts, ω = 3.06 (2)°, and antiparallel cation displacements along the polar axis. General trends of cation displacements and the various deviations of the octahedral network from the prototypic cubic perovskite structure have been established and their systematic behaviour with temperature is reported. An investigation of phase transition behaviour using second harmonic generation (SHG) to establish the centrosymmetric or non-centrosymmetric nature of the various phases is also reported.

ABSTRACT: In April 1948, Progressive Party presidential candidate Henry A. Wallace arrived in Indiana to much controversy. The conservative state did not welcome Wallace, and veterans’ organizations actively organized to disrupt his speaking engagements. On April 6, at the Progressive Party’s Evansville campaign event, a mob attacked Wallace supporters, causing injuries and pushing the isolated town into the national spotlight. In the wake of the riot, a local professor was fired for his involvement in the Wallace campaign, and the radical CIO Local 813 became the subject of U. S. House committee hearings. Anticommunist hysteria gripped the Evansville community. What happened in Evansville on April 6 was part of a populist fascism in the United States propelled by anti-communism and enacted by veterans’ organizations. While national politicians dominate histories of anti-communism, some of the greatest damage done during the 1940s and 1950s occurred when other Americans, specifically veterans’ groups, violated the constitutional rights of their fellow citizens.

AbstractObjectiveTo describe the burden, clinical characteristics and prognostic factors of severe malnutrition in children under the age of 5 years.DesignRetrospective study of hospital-based data systematically collected from January 2001 to December 2010.SettingRural Mozambican district hospital.SubjectsAll children aged <5 years admitted with severe malnutrition.ResultsDuring the 10-year long study surveillance, 274 813 children belonging to Manhiça's Demographic Surveillance System were seen at out-patient clinics, almost half of whom (47 %) presented with some indication of malnutrition and 6 % (17 188/274 813) with severe malnutrition. Of these, only 15 % (2522/17 188) were eventually admitted. Case fatality rate of severe malnutrition was 7 % (162/2274). Bacteraemia, hypoglycaemia, oral candidiasis, prostration, oedema, pallor and acute diarrhoea were independently associated with an increased risk of in-hospital mortality, while malaria parasitaemia and breast-feeding were independently associated with a lower risk of a poor outcome. Overall minimum community-based incidence rate was 15 cases per 1000 child-years at risk and children aged 12–23 months had the highest incidence.ConclusionsSevere malnutrition among admitted children in this Mozambican setting was common but frequently went undetected, despite being associated with a high risk of death. Measures to improve its recognition by clinicians responsible for the first evaluation of patients at the out-patient level are urgently needed so as to improve their likelihood of survival. Together with this, the rapid management of complications such as hypoglycaemia and concomitant co-infections such as bacteraemia, acute diarrhoea, oral candidiasis and HIV/AIDS may contribute to reverse the intolerable toll that malnutrition poses in the health of children in rural African settings.

Laparoscopic, transgastric, stapler-assisted mucosectomy (SAM) with the retrieval of a circumferential specimen is proposed. Methods SAM was evaluated in two phases. The feasibility of SAM and the quality of specimens were assessed in eight animals. The mucosal healing was evaluated in a 6-week survival experiment comparing SAM (n = 6) with EMR (n = 6). The ratio of the esophageal lumen width (REL) at the resection level measured on fluoroscopy at 6 weeks divided by the width immediately after resection was compared. Results In all animals, a circular mucosectomy specimen was successfully obtained, with a median area of 492 mm2 (interquartile range [IQR] 426 – 573 mm2) and 941 mm2 (IQR 813 – 1209 mm2) using a 21 mm and 25 mm stapler, respectively. In the survival experiments, symptomatic stenosis developed in two animals after EMR and in none after SAM. The REL was 0.27 (0.18 – 0.39) and 0.96 (0.9 – 1.04; P < 0.0001) for EMR and SAM, respectively. Conclusions SAM provides a novel technique for en bloc mucosectomy in BE. In contrast to EMR, mucosal healing after SAM was not associated with stenosis up to 6 weeks after intervention.

COVID-19 is a global threat with an increasing number of infections. Research on IgG seroprevalence among health care workers (HCWs) is needed to re-evaluate health policies. This study was performed in three pandemic hospitals in Istanbul and Kocaeli. Different clusters of HCWs were screened for SARS-CoV-2 infection. Seropositivity rate among participants was evaluated by chemiluminescent microparticle immunoassay. We recruited 813 non-infected and 119 PCR-confirmed infected HCWs. Of the previously undiagnosed HCWs, 22 (2.7%) were seropositive. Seropositivity rates were highest for cleaning staff (6%), physicians (4%), nurses (2.2%) and radiology technicians (1%). Non-pandemic clinic (6.4%) and ICU (4.3%) had the highest prevalence. HCWs in “high risk” group had similar seropositivity rate with “no risk” group (2.9 vs 3.5 p = 0.7). These findings might lead to the re-evaluation of infection control and transmission dynamics in hospitals.

Tujuan dari penelitian ini adalah untuk mengetahui efek penggunaan biokoagulan serbuk biji kelor (moringa oleifera) untuk perbaikan kualitas air tanah di Desa Tunggunjagir Lamongan. Treatment serbuk biji kekor 0,5 mg menggunakan metode jar test dalam 500 ml air sampel yang diambil dari 4 titik sumur sendang. Kekeruhan memiliki rata rata penurunan nilai kekeruhan sebesar 38%, penurunan nilai TDS sebesar 4%, kenaikkan nilai pH sebesar 6%, dan kenaikan nilai nitrat sebesar 813%. Berdasarkan hasil penelitian penggunaan serbuk biji kelor (moringa oleifera) mampu meningkatkan kualitas fisik air tanah,tetapi ada satu parameter yang melebihi standar baku mutu yakni nitrat sedangkan untuk range nilai parameter lainnya tidak melebihi standart baku mutu.Kata kunci : Biokoagulan, moringa oleifera, jar test, air tanah.

Objective.To explore 5 possible criteria for remission in rheumatoid arthritis (RA) based on a patient self-report index, the Routine Assessment of Patient Index Data (RAPID3), with a careful joint examination and possible physician global estimate (DOCGL), but without a formal joint count or laboratory test.Methods.The ESPOIR early RA cohort of 813 French patients recruited in 2002–2005 was analyzed to identify patients in remission 6 months after enrollment, according to 2 American College of Rheumatology/European League Against Rheumatism (ACR/EULAR) criteria: Boolean ≤ 1 for total tender joint count-28, swollen joint count-28, C-reactive protein, and patient global estimate (PATGL), and Simplified Disease Activity Index (SDAI) ≤ 3.3. Substantial agreement with the Boolean criteria was seen for SDAI, CDAI, RAPID3R+SJ1, RAPID3R+SJ1+D1, RAPID3R+SJ0, and RAPID3R+SJ0+D1 (92.2%–94.7%, kappa 0.67–0.79), versus only moderate agreement for DAS28 or RAPID3R (79.9%–85.8%, kappa 0.46–0.55).Conclusion.Remission according to CDAI and RAPID3R+SJ1, but not DAS28 or RAPID3R, is similar to that of the ACR/EULAR criteria. RAPID3 scores require a complementary careful joint examination for clinical decisions, do not preclude formal joint counts or other indices, and may be useful in busy clinical settings.

Children’s height, weight and waist circumference were measured along with their PROP taster status. PROP was measured using two paper discs, one impregnated with NaCl (1·0 mol/l) and the other with PROP solution (0·50 mmol/l).ResultsA significant PROP sensitivity by socio-economic status (SES) interaction term (P = 0·010) was detected wherein supertasters had the largest BMI percentile and Z-score, but only among the group with highest SES.ConclusionsThe results suggest that other factors overwhelmed the influence of PROP sensitivity on adiposity in lower-SES groups. The percentage of variance accounted for by the interaction term was about 1 %. Thus, PROP supertasters had the largest BMI percentile and Z-score, but only among the highest-SES group.

Mitral valve prolapse has been studied extensively in the adult population, but less is known about it in children. Therefore, 813 children between 9 and 14 years of age were examined by a team of cardiologists and technicians. The children also responded to a questionnaire concerning the presence of symptoms and the What I Think and Feel anxiety instrument. The prevalence of mitral valve prolapse using auscultatory criteria was 4.2% (6.2% for girls, 2.3% for boys). Of those with mitral valve prolapse, 85% had a solitary click, 9% had a click and systolic murmur, and 6% had multiple clicks. Children with auscultatory mitral valve prolapse were less likely to have symptoms than those free of cardiac abnormalities. No difference in average anxiety scores was detected between the two groups. It is concluded that auscultatory mitral valve prolapse is common in children and not accompanied by an increased likelihood of symptoms or anxiety.

Tujuan dari penelitian ini adalah untuk menghitung ulang kelebihan air tampungan berdasarkan kondisi hidrologis terkini, menggunakan metode perhitungan debit intake di menara Bendungan Semantok, menghitung kebutuhan air irigasi, dan menghitung kehilangan air di sungai, sehingga nilainya kelebihan air tambahan dapat digunakan untuk mengembangkan potensi lahan pertanian. Penelitian ini dilakukan di bagian hilir Bendungan Semantok tepatnya di 6 (enam) Daerah Irigasi (D.I) yaitu D.I Rejoso, D.I Margomulyo, D.I Jatirejo, D.I Jati, D.I Janeng, dan D.I Ngomben untuk menghitung kebutuhan air irigasi. However, influence of the polymorphisms on survival in metastatic colorectal cancer (mCRC) patients (pts) treated with IRI-containing therapy remains controversial. Methods: We retrospectively reviewed data of consecutive mCRC pts who received FOLFIRI/IRI with or without an anti-VEGF agent as 2nd line therapy at the Shizuoka Cancer Center between April 2008 and December 2015. The primary selection criteria were histologically confirmed adenocarcinoma, prior history of oxaliplatin-containing therapy, and adequate organ function. UGT1A1 polymorphisms were screened and pts with homozygous (*6/*6, *28/*28) or compound heterozygous (*6/*28) were excluded. Results: Among 103 pts found eligible for this analysis, 63 were wild type (W) (−/−) for UGT1A1, and 40 were heterozygous type (H) (-/*6, -/*28), with the following characteristics: median age (range), 60 (30–77) vs 67 (37–84); male/female, 41/22 vs 18/22; PS 0/1/2, 40/21/2 vs 21/18/1; tumor location right/left, 16/47 vs 9/31; peritoneal metastasis −/+, 49/14 vs 33/7; disease status metastasis/recurrence, 49/14 vs 23/17; RAS wild type/mutant, 25/37 vs 18/21; IRI regimen doublet/mono, 51/12 vs 33/7; anti-VEGF agent use −/+, 16/47 vs 12/28; and median IRI dose intensity, 63.7 vs 60 mg/m2/week. Incidences of grade 3/4 neutropenia, febrile neutropenia, and diarrhea in W/H pts were 36%/48%, 5%/9%, 5%/13% (initial IRI dose 180 mg/m2). Median PFS was 6.2 M in W, and 3.9 M in H (p = 0.36). Median OS was 12.1 M in W and 10.9 M in H (p = 0.86). RR was 13% in W, and 10% in H (p = 0.68). DCR was 81% in W, and 58% in H (p = 0.012). Multivariate analysis identified no liver-limited disease, disease status (metastatic), UGT1A1 H type, IRI monotherapy, and no anti-VEGF as independent predictors of poorer PFS (HR 1.76, 2.01, 1.67, 2.17, 2.58), and initial dose of CPT < 180mg/m2 and no anti-VEGF as independent predictors of poorer OS (HR 1.85 and 1.85). UGT1A1 polymorphism was not detected as an independent predictor of RR or DCR. Conclusions: UGT1A1 polymorphism may be useful in predicting PFS in mCRC pts treated with IRI-containing therapy.

We hypothesized that using industrial engineering techniques and a modified dynamic sustainability framework for implementation, we can implement an electronic medical record (EMR) based pathway in the management of MBO. Methods: A workflow in the management of MBO was developed using iterative meetings from 8/2018 to 4/2019 including gateway stakeholders (surgical oncology, gynecological oncology and medical oncology), interventional stakeholders (gastroenterology, interventional radiology) and supportive stakeholders (hospital medicine, palliative care, nutrition, nursing). Industrial engineers were utilized to study human factors, and perform a method study. EMR integration was performed using EPIC systems Agile MD pathway and educational materials were created. Interventions such as early placement of gastrostomy tubes, total parenteral nutrition and medications were protocolized. Results: Since implementation, over 6 months the pathway and order set has been activated 56 times. Orders have been employed 21 times through the AgileMD pathway demonstrating a pathway drift of 62.5%. Educational materials have been accessed routinely during this time. Conclusions: Feasibility of implementing an EMR integrated MBO pathway is demonstrated with early suggestion of pathway drift. Utilizing tools of implementation science are necessary to facilitate widespread adoption of evidence based interventions in the management of patients with MBO.

We tested the hypothesis that prior heavy-intensity exercise reduces the difference between asymptotic oxygen uptake (VO2) and maximum oxygen uptake (VO2max) during exhaustive severe-intensity running lasting ≍2 minutes. Ten trained runners each performed 2 ramp tests to determine peak VO2 (VO2peak) and speed at venti-latory threshold. They performed exhaustive square-wave runs lasting ≍2 minutes, preceded by either 6 minutes of moderate-intensity running and 6 minutes rest (SEVMOD) or 6 minutes of heavy-intensity running and 6 minutes rest (SEVHEAVY). Two transitions were completed in each condition. VO2 was determined breath by breath and averaged across the 2 repeats of each test; for the square-wave test, the averaged VO2 response was then modeled using a monoexponential function. The amplitude of the VO2 response to severe-intensity running was not different in the 2 conditions (SEVMOD vs SEVHEAVY; 3925 ± 442 vs 3997 ± 430 mL/min, P = .237), nor was the speed of the response (τ; 9.2 ± 2.1 vs 10.0 ± 2.1 seconds, P = .177). VO2peak from the square-wave tests was below that achieved in the ramp tests (91.0% ± 3.2% and 92.0% ± 3.9% VO2peak, P < .001). There was no difference in time to exhaustion between conditions (110.2 ± 9.7 vs 111.0 ± 15.2 seconds, P = .813). The results show that the primary VO2 response is unaffected by prior heavy exercise in running performed at intensities at which exhaustion will occur before a slow component emerges.

The effects of exercise training on hepatic function in horses were determined by studying the plasma clearance of antipyrine (20 mg/kg iv) in adult mares that either underwent treadmill training for 5 wk ( n = 7) or remained in box stalls for the same time period ( n = 6). Training consisted of treadmill exercise at 60% (12 min/day) and 90% (3 min/day) of pretraining maximal oxygen consumption (V˙o 2 max) for 6 days/wk for 5 wk.V˙o 2 max and velocity to obtain a blood lactate concentration of 4 mmol/l were significantly increased (from 129 to 149 ml ⋅ min−1 ⋅ kg−1and from 5.6 to 6.1 m/s, respectively) as a result of training. The plasma clearance and volume of distribution of antipyrine increased significantly in the trained group (from 5.5 to 6.4 ml ⋅ min−1 ⋅ kg−1and from 813 to 881 ml/kg, respectively) and decreased significantly in the untrained group. Elimination half-lives did not change as a result of training or box rest. Increases in plasma antipyrine clearance were indicative of an increase in hepatic metabolism of antipyrine. Increases in the volume of distribution of antipyrine suggest that total body water increases as a result of exercise training.

Among the problems that need to be addressed is the need to design and develop environmentally friendly products that are energy efficient and easy to recycle. In this study, the effect of metal salt generation processing on the tensile strength of the bonded interface of Al/Al and Al/Cu was investigated by SEM observations of interfacial microstructures and fractured surfaces. Aluminum surfaces were modified by boiling in 5% aqueous solution of NaOH for 30 s and 98% formic acid for 60 s. Copper surface were modified by boiling 98% formic acid for 60 s. Solid-state bonding was performed at bonding temperature of 673 ~ 813 K and under a pressure of 6 MPa ( bonding time of 1.8 ks). Using metal salt generation bonding technique, the bonded joint is able to reach 0.2% proof stress at lower bonding temperature and with less deformation.

However, apprehension exists that variants of concern (VOCs) may evade vaccine protection, due to evidence of reduced neutralization of the VOCs B.1.1.7 and B.1.351 by vaccine sera in laboratory assays. We performed a matched cohort study to examine the distribution of VOCs in infections of BNT162b2 mRNA vaccinees from Clalit Health Services (Israel) using viral genomic sequencing, and hypothesized that if vaccine effectiveness against a VOC is reduced, its proportion among breakthrough cases would be higher than in unvaccinated controls. Analyzing 813 viral genome sequences from nasopharyngeal swabs, we showed that vaccinees who tested positive at least 7 days after the second dose were disproportionally infected with B.1.351, compared with controls. Those who tested positive between 2 weeks after the first dose and 6 days after the second dose were disproportionally infected by B.1.1.7. These findings suggest reduced vaccine effectiveness against both VOCs within particular time windows. Our results emphasize the importance of rigorously tracking viral variants, and of increasing vaccination to prevent the spread of VOCs.

Background: Infant and young child feeding practices directly affect the nutritional status of children under two years of age and, ultimately, impact child survival. Therefore, the objective of this study was to assess the WHO infant and child feeding indicators in a rural area of Ludhiana, Punjab, India. Methods: A community based cross sectional study was conducted in fifteen villages of Ludhiana district under rural health training centre of Department of Community Medicine, Dayan and Medical College & Hospital, Ludhiana, Punjab. The data was obtained from mothers/primary caregivers of 813 living children 0-23 months of age group and information about seven core and three optional WHO infant and young child feeding indicators was collected using WHO questionnaire. Prevalence estimates with 95% confidence interval were calculated. A Χ2 test and Odds Ratio (OR) with 95% CI was calculated wherever required. Results: Only 56.7% of infants were put to breast within one hour of birth, while three fourths of infants younger than 6 months were exclusively breastfed. About seventy percent of children aged 12-15 months and 54.7% of children 20-23 months continued breastfeeding at 1 & 2 years respectively. Almost all the children born in the last 23 months were ever breastfed. Nearly ninety percent of infants 6-8 months of age were fed with complimentary foods at the recommended daily frequency. Conclusion: To improve breastfeeding practices, health education needs to be strengthened for target population having sub-optimal breastfeeding indicators. Introduction: Healthcare workers (HCWs) colonized with Staphylococcus aureus may serve as a reservoir of infection. This study was carried to determine the genetic make-up of S. aureus nasal colonizers in HCWs. Methodology: Nasal swabs were obtained from 93 HCWs and molecular characterization of identified S. aureus isolates was carried out using the StaphyType DNA microarray (Alere Technologies GmbH, Jena, Germany). Results: Twenty-nine HCWs (31%) were colonized with S. aureus (MSSA = 23; MRSA = 6). Thus the overall MRSA carriage rate was 6.5% (n/N = 6/93) and 20.7% (n/N = 6/29) of those colonized with S. aureus harboured MRSA. The S. aureus isolates belonged to 16 clonal complexes (CC). MSSA isolates included three each for CC15, CC188, ST2867; two each for CC5, CC97, CC367 as well as one each for CC1, CC8, CC30, CC45, CC101, CC121, ST291/813 and CC1153. The staphylococcal cassette chromosome recombinase genes ccrA-1; ccrB-1 and the fusidic acid resistance gene (fusC) were present in two MSSA isolates (CC1 and CC8). The six MRSA isolates included CC5-MRSA-[VI+fusC] (n = 2); one each of CC5-MRSA-V; CC22-MRSA-IV (tst1+); CC80-MRSA-IV [pvl+] (“European CA-MRSA Clone”) and CC97-MRSA-[V+fusC]. Conclusion: There is wide clonal diversity of S. aureus colonizers with associated high MRSA carriage among the HCWs. The presence of genetically stable MSSA isolates with the capability to transform into MRSA isolates is of concern.

Background and Objective: Chemokine (C-C motif) ligand (CCL)2, the prototype Th2 chemokine, is secreted by tumor cells, and has growth promoting effects. Whether CCL2 protumorigenic activities will be validated, then CCL2 and its receptor CCR2 may be therapeutic targets in cancer. Methods: We tested in “primary human anaplastic thyroid carcinoma (ATC) cells” (ANA) versus “normal thyroid follicular cells” (TFC): a) CCL2 secretion basally, after IFN-γ and/or TNF-α stimulation; b) PPARγ activation by thiazolidinediones (TZDs), rosiglitazone or pioglitazone, on CCL2 secretion, and on proliferation and apoptosis in ANA. Results: ANA produced basally CCL2, at a higher level versus TFC. IFN-γ or TNF-α dose-dependently induced the CCL2 release in 3/6 or 5/6 ANA, respectively, but in all TFC. IFN-γ+TNF-α induced a synergistic release of CCL2 in all TFC, but only in 1/6 ATC. TZDs exerted an inhibition of CCL2 release in 3/6 ANA, while had no effect in TFC. Pioglitazone inhibition of ANA proliferation was not associated with the effect on CCL2; NF-κB and ERK1/2 were basally activated in ANA, increased by IFN-γ+TNF-α, and pioglitazone inhibited IFN- γ+TNF-α activation. CCL2 serum levels were higher in 6 ATC patients than in 5 controls (813±345 versus 345±212, pg/mL; respectively; P<0.01, ANOVA). Conclusion: ANA produce CCL2 basally and after cytokines stimulation, with an extremely variable pattern of modulation, suggesting different types of deregulation in the chemokine modulation. Serum CCL2 is increased in ATC patients. Further studies will be necessary to evaluate if CCL2 might be used as a marker in the followup of ATC patients.

El objetivo de este estudio fue realizar un análisis descriptivo del rendimiento de la velocidad, la fuerza, la resistencia y la flexibilidad en futbolistas chilenas de primera división en función a la posición de juego. Las participantes de este estudio fueron 30 jugadoras con edades comprendidas entre los 18 y 28 años, las cuales se encontraban iniciando el periodo de preparación. Fueron agrupadas según la posición de juego en 3 guardametas (PO), 4 centrales (CT), 6 laterales (LT), 6 mediocentros (MC), 5 mediocentros de banda (MB) y 6 delanteras (DC). Las jugadoras completaron las pruebas de velocidad en 30 metros lineales (V30M), Arrowhead Agility Test (AAT), Counter Movement Jump (CMJ), Intermittent Fitness Test (30-15IFT) y Elevación de la Pierna Recta (EPR). No se observaron diferencias significativas para las variables de velocidad, fuerza, resistencia y flexibilidad según la posición de juego (p<.05), pero si se encontraron relaciones significativas entre V30M y AAT (r=.435, p=.001), CMJ (r=-.599, p=.000), VIFT (r=-.621, p=.000), VO2máx (r=-.562, p=.001) y EPR-D (r=-.418, p=.019), entre EPR-ND y AAT (r=.448, p=.012), EPR-D y CMJ (r=.537, p=.002), VIFT y VO2máx (r=.934, p=.000), EPR-D y EPR-ND (r=.813, p=.000). ABSTRACT Background Current literature examining the prospective relation of circulating omega-3 (n–3) and omega-6 (n–6) PUFAs and atrial fibrillation (AF) is limited to predominantly white populations. Objectives We investigated the association of circulating n–3 and n–6 PUFAs with incident AF in participants from the Multi-Ethnic Study of Atherosclerosis. Methods A total of 6229 participants (mean age = 62 y; 53% female; 39% white, 27% black, 22% Hispanic, and 12% Chinese) who were free of baseline AF and with plasma phospholipid PUFAs measured at baseline using GC were prospectively followed for the development of AF. Incident AF was ascertained using International Classification of Diseases-9 codes from hospital discharge records and Medicare claims data with follow-up through 2014. Multivariable Cox proportional hazards regression analysis was performed to determine the risk of incident AF. Results During a median follow-up of 12.9 y, 813 (13%) participants developed AF. Each higher SD increment in arachidonic acid (AA; 20:4n–6) concentrations was associated with an 11% decreased risk of incident AF (HR: 0.89; 95% CI: 0.82, 0.96). Similarly, higher overall n–6 PUFA concentrations were also associated with a reduced AF risk (HR per SD increment: 0.93; 95% CI: 0.87, 1.00). Although no significant overall associations were observed for any individual n–3 PUFAs, higher circulating concentrations of DHA (22:6n–3) and EPA (20:5n–3) were associated with a decreased AF risk in blacks and Hispanics (DHA only) but not whites or Chinese Americans. Conclusions In a multiethnic cohort of individuals free of baseline cardiovascular disease, higher plasma concentrations of n–6 PUFAs, particularly AA, were associated with a reduced risk of incident AF. Important differences in AF risk were also noted across race/ethnicity for the n–3 PUFAs DHA and EPA.

Abstract A Ca2+-activated and phospholipid-dependent protein kinase (PKC) has been described in several cell systems, including the human neutrophil. We found that less than 30 pmol 32P/10 min/10(6) cell equivalents of phorbol ester- or 1-oleoyl-2-acetylglycerol-stimulated PKC activity was obtained when neutrophil homogenates were used as an enzyme source. However, detergent-soluble and detergent-insoluble fractions prepared from the same homogenates, respectively, catalyzed 578 +/- 50 and 136 +/- 40 pmol 32P/10 min/10(6) cell equivalents. Recombining detergent-soluble and detergent-insoluble fractions resulted in the complete loss of activity. We therefore explored the possibility of an endogenous inhibitor of PKC in neutrophils. Homogenates from neutrophil cytoplasts, which lack the nuclei and intracellular granules of whole neutrophils, yielded 813 +/- 28 pmol 32P/10 min/10(6) cell equivalents. In addition, total neutrophil homogenates from a patient with a specific granule deficiency yielded high activities, namely 424 +/- 48 pmol 32P/10 min/10(6) cell equivalents. Specific granule-deficient neutrophils possessed translocated and activated PKC, and phosphoprotein patterns from these cells resembled those from activated normal neutrophils. Our results suggested the existence of an inhibitor of previously active PKC. That this inhibitor is primarily associated with neutrophil-specific granule membranes was suggested by our finding of high PKC activity associated with cell preparations or combinations of cell fractions that were free of specific granules, but not necessarily of other cellular organelles. Preliminary characterization of the endogenous inhibitor indicated that it was protease- and heat-sensitive, and did not exhibit either protease or phosphatase activity. We speculate that the inhibitor may play a physiologic role in regulating the activity of its target enzyme.

Background: This study aimed at monitoring the kinetics of serum total anti-SARS-CoV-2 (severe acute respiratory syndrome coronavirus 2) antibodies in a cohort of healthcare workers after voluntary vaccination with Pfizer-BioNTech coronavirus disease 2019 (COVID-19) mRNA-based vaccine. Methods: The study population consisted of 787 healthcare workers (mean age 44±12 years; 66% females), who received two 30 mg doses of Pfizer-BioNTech COVID-19 vaccine, 3 weeks apart. Venous blood was drawn before the first vaccine dose, immediately before the second vaccine dose, and then at 1, 3 and 6 months after the second vaccine dose. Serological testing employed the total antiSARS-CoV-2 antibodies measurement with Roche Elecsys Anti-SARS-CoV-2 S chemiluminescent immunoassay. Results: The median serum levels of total anti-SARS-CoV-2 antibodies reached the peak (1762 kU/L) 1 month after the second vaccine dose, but tended to progressively decline at the 3-month (1086 kU/L) and 6-month (802 kU/L) follow-up points. Overall, the values after 3and 6months were 37% and 57% lower than the corresponding concentrations measured at the peak. No healthcare worker had total anti-SARS-CoV-2 antibodies below the method-dependent cut-off after 6 months. The decline compared to the peak was more accentuated in baseline seropositive persons than in those who were baseline seronegative (74% vs. 52%) cohort. The 6-month post-vaccination anti-SARS-CoV-2 antibodies in subjects aged <65 years remained over 2-fold higher than in those aged ≥65 years (813 vs. 343 kU/L) and also remained consistently higher in women than in men. Conclusions: Gradual decline of total anti-SARS-CoV-2 antibodies occurred 6 months after Pfizer-BioNTech COVID-19 vaccination, though values remained higher than the method-dependent cut-off, with no case of sero-negativization.

One way to overcome the scarcity of clean water through sustainable approach is by utilizing a solar distillation system. This easy-to-use technology is adopting tubular solar distillation. The three main components, which are the most essential for producing the amount of permeate, are the solar collector, tubular and heat exchanger (HE). This study aims to determine the performance of a tubular solar distillation device equipped with HE using a parabolic trough collector (PTC). The PTC has an area of 5.1 m2 covered with a solar reflective chrome film. Aluminum tubular acts as the feedwater heater. The HE is placed inside the tubular, which acts as a coolant to convert the steam phase into freshwater/permeate and as a feedwater heater to flow into the tubular. In the present study, several parameters were tested: comprise temperature, solar radiation, pressure, humidity, mass flow rate, permeate productivity and efficiency. This study demonstrated the production of a sufficient amount of permeate, which was 5.32 L for 6 h. The efficiency of this device yielded a peak of 48.2% during solar radiation of 813 W/m2 in an average ambient temperature of 32 °C, with an overall average of 44.59%.

Daar and L.E. Maquat, Mol. Cell. Biol. 8:802-813, 1988). For all mutations in this group, the decrease is to the same extent, i.e., to approximately 20% of the normal level. We show here that a second group of nonsense mutations that cause translation to terminate prematurely at or downstream of codon 208, in exon 6, did not affect TPI mRNA abundance. Deletion analysis demonstrated that the abundance of translationally active TPI mRNA is a function of both the distance and the polarity of the nonsense codon relative to the final intron in TPI pre-mRNA. Our results indicate that if translating ribosomes are unable to progress to at least a certain position within the penultimate exon relative to the final intron, then the level of the corresponding mRNA will be abnormally low. Studies inhibiting RNA synthesis with dactinomycin demonstrated that a block in translation does not affect the half-life of mature TPI mRNA. The simplest interpretation of our data is that the translation of TPI mRNA in the cytoplasm facilitates the splicing of TPI pre-mRNA or the transport of TPI mRNA across the nuclear envelope or both.
