Hur bevarar man bäst en stads karaktär? Slottsarkitekten Jakob Strömholm visade oss runt i Mariefred och pekade på alla idéer han hade stoppat. Det finns ett stort engagemang i staden. Ett tag skrev Jakob beslutsdokument om huruvida ett träds grenar skulle klippas eller inte. Besluten fattas av hänsyn till Gripsholmsslott. Lättheten i byggnaden speglar den välkomnande gesten. Strukturen bärs av de svarta ramarna i betong och släppet på 700mm från marken förtydligar nättheten. Släppet ger även ljus till hotellets personalytor på källarplan. På bottenplan finns restaurang, hotellreception och en offentlig scentrappa. Tomtens nya stadsrum erbjuder en park och uteservering kopplad till restaurangen. Jag skulle vilja sätta ett nytt kapitel för staden - låta det gamla vara det gamla och det nya bli det nya, utan att förstöra varandra. Som en gest har jag delat upp tomten och skapat två stadsrum. Byggnaden definierar och introducerar det ena rummet till det andra. Med hjälp av öppningar i strukturen erbjuds ett välkomnande flöde genom och runt byggnaden.How to preserve a city's character? Todays architect of Gripsholm castle Jakob Strömholm showed us around in Mariefred and pointed out all the ideas he had stopped. There is a strong commitment in the city. At one point Jakob wrote decision documents whether a tree's branches would be cut or not. The decisions are made for the sake of Gripsholm castle. The weightlessness of the building reflects the welcoming gesture. The structure is carried by the black frames in concrete and the release of 700mm from the ground clarifies the weightlessness. The release does also provide light to the hotel staff in the basement. On the ground floor there is a restaurant, hotel reception and a public scene. The plot’s new urban space offers a park and outdoor terrace connected to the restaurant. I would like to put a new chapter for the city - let the old be old and new become the new without destroying each other. As a gesture, I have divided the site and created two urban spaces. The building defines and introduces one room to the other. The openings in the structure offers a welcoming flow through and around the building.

Resumo: Todos sabemos que é difícil tentar expressar num discurso verbal o que é arte. Quando indagado sobre esse mesmo dilema, o músico contemporâneo Frank Zappa (1940-1993) afirmou que "escrever sobre música é como dançar sobre arquitetura". Ainda assim, gostaria de arriscar essa empreitada com a intensão de explicitar parte de meu processo de construção das imagens, principalmente as que foram realizadas durante essa investigação sobre os cadernos de desenho. A construção de tais cadernos impõe um processo de reflexão específico, ligado aos meios expressivos que permitem uma aproximação mais adequada com a imposição determinada pelas configurações técnicas de formatos e suportes que os cadernos de desenho impõem, tanto na sua confecção, como no manuseio e, até mesmo, nas formas de expor. Visando expressar-me textualmente da forma mais racional, clara e sistemática possível sobre minha própria produção, dentro da perspectiva de um olhar pautado pela poética visual proposta, decidi organizar esse olhar, essa reflexão por meio da seleção, apresentação e análise de sete cadernos separados em capítulos distintos. Eles evidenciam eixos comuns importantes, como as questões afetivas que direcionam e moldam a construção das imagens, as escolhas cromáticas carregadas de informações visuais ou assumindo uma postura de franca vocação documental, como a proposta praticada há muito pelos artistas viajantes, acadêmicos ou contemporâneos. A confecção ritualizada dos cadernos, prevê posturas diversas em várias etapas do trabalho, da escolha dos materiais, procedimentos técnicos, tomada de decisões, direcionamento do olhar, da memória ou de experimentações mais livres, até a sua completa finalização
Abstract: We all know that it is difficult to try to express a verbal discourse what is art. When asked about this same dilemma, the contemporary musician Frank Zappa (1940-1993) said that "writing about music is like dancing about architecture." Still, I would risk this venture with the intention of explaining part of my images of the construction process, especially those carried out during the investigation into the sketchbooks. The construction of such notebooks impose a particular reflection process on the expressive means to a more appropriate approach to the imposition determined by technical configurations of formats and supports the sketchbooks impose both in its making, as the handling and, even in the forms of display. In order to express myself literally the most rational, clear and systematic as possible on my own production, from the perspective of a look marked by poetic visual proposal, decided to organize this look, this reflection through the selection, presentation and analysis of seven separate notebooks in separate sections. They show important common axes as affective issues that drive and shape the construction of images, loaded chromatic choices of visual information or assuming an attitude of frank documentary vocation, as the proposal practiced long by artists travelers, students or contemporary. The ritualized manufacture of notebooks, provides for various positions in various stages of work, the choice of materials, technical procedures, decision-making, direction of gaze, memory or more free trials until its full completion

I will also mention few examples of periods of art in which artists were touched by the same phenomena as me.

This creative project and essay for a Masters degree in Media Theory and Practice is focused around the design of a South African TV series called 'Schedule 7' which is set in Cape Town's south peninsula in a (fictional) non-governmental, psychiatric clinic. The premise is that a troubled handful of patients' perspectives on life change after they encounter what they believe to be God's salvation when they accidentally almost burn themselves to death. The format follows their endeavours to follow and serve God, who they term 'GOGO' (God of Goodness Only), with mixed results. The format is an hour slot comedy-drama series designed for television broadcast (in that it is structured around commercial breaks). A comprehensive series bible is included, as well as two full sample episodes: the pilot and episode 3.Accompanying the creative components, the academic research essay firstly looks at the significance of mental illness: particularly in terms of stigma, South African context and places of treatment. It argues that an NGO psychiatric clinic makes for an interesting setting for a TV series as it allows characters from a range of socio-economic backgrounds to be rendered equal in power and united under a 'patient' status. This section also looks at mental illness intersecting with religion or faith. Secondly, the essay gives an overview of trends in and significant instances of on-screen representations of mental illness for which it offers a critique. Thirdly, in light of the issues raised surrounding mental illness, religion and the strengths and weaknesses of on-screen fiction around the subject so far, it offers an explication of the creative choices made in the design of the TV series, which were made with the aim of a resulting TV series that is enjoyable, relevant, diverse, and open-ended in its exploration of both experiences of mental illness and religious perception and practice.

These nuggets have a single presentation in a box of 8 units. The operational plan will explain the processes necessary to carry out efficient production by developing a dynamic relationship with suppliers and other key partners. The marketing plan will establish the appropriate guidelines to establish a relationship with potential customers through traditional and digital marketing strategies, establishing social networks, websites and fairs as sales channels. Finally, the analysis of the viability of the project is developed in the financial plan which will show all the projected income and expenses. Atherosclerosis is an inflammatory disease in which an accumulation of fatty acids and cholesterol occurs to form a plaque in small and large arteries. Monocyte polarization to classic M1 macrophages or alternative M2 macrophages is an important area of research that can determine the severity of disease progression. BMP-7 is a key growth factor responsible for directing differentiation of mesenchymal stem cells into brown fat cells, suggesting a role of BMP-7 in cellular plasticity; however, its role in monocyte polarization is yet to be revealed. In the current study, we hypothesize that monocyte treatment with BMP-7 will significantly result in increased polarization of monocytes into M2 macrophages and increased expression of anti-inflammatory cytokines. To that effect, we have established a stress induced cell culture system with monocytes (THP-1 cells) and apoptotic conditioned medium (ACM), simulating injury, to understand the effects of BMP-7 on M2 macrophage polarization from monocytes. Our data demonstrates that the BMP type 2 receptor (BMPR2) is found on monocytes and its activation is significantly (p<0.05) increased in both monocytes and M2 macrophages following treatment with BMP-7. Furthermore, a significant (p<0.05) increase of M2 macrophages in the BMP-7 treated group was shown following immunostaining with CD206 and arginase-1, two M2 macrophage markers, whereas a significant (p<0.05) decrease of iNOS expression, an M1 macrophage marker, was shown. Moreover, treatment with BMP-7 resulted in significantly (p<0.05) increased expression of IL-10 and IL-1ra, two anti-inflammatory cytokines, but significantly (p<0.05) decreased levels of the pro-inflammatory cytokines, MCP-1, IL-6 and TNF-?. We also hypothesize that polarization of monocytes to M2 macrophages occurs through activation of SMAD1/5/8 and PI3K-Akt-mTOR pathways. Upon BMP-7 binding to its receptor, BMPR2, activation of SMAD1/5/8 occurs which then activates the p85 subunit of PI3K resulting in downstream activation of Akt and mTOR. Our data shows that following treatment with BMP-7, expression of p-SMAD1/5/8, p-PI3K, p-Akt and p-mTOR is significantly (p<0.05) increased compared to controls whereas p-PTEN, an inhibitor of the PI3K pathway, is significantly (p<0.05) decreased in the BMP-7 treated group compared to controls. In conclusion, our data reveals that BMP-7 polarizes monocytes into M2 macrophages and it achieves this through activation of the PI3K-Akt-mTOR pathway, which will have significant applications for atherosclerosis treatment.M.S.

This work uses STM-scanning tunneling microscope and computers to study the interaction of two different materials. The equipment used are STM-scanning tunneling microscopy and computers. The materials are the silicon (111)-7 x 7 surface and the carbon 60. The C₆₀ topic has been widely discussed since 1990. The basic properties of C₆₀ molecules have been reviewed in this work. The silicon surface is important for the current electronic industry, but the silicon surface structure was not well known until 1985. STM is the best technique to image C₆₀ on the silicon surface in the real space domain. In chapter one and chapter two, I discuss the background of this topic and the principles of my experiment. The interaction of C₆₀ with the Si (111)-7 x 7 surface has been seen clearly on STM images, but the nature of the interaction is not well known. The primary question is where these C₆₀ molecules prefer to stay on the Si (111)-7 x 7 surface? I made a computer to answer this question by using a sophisticated program package to do image recognition. To explain the interesting statistical result, a very simple physical model has been constructed and calculations by the theoretical model are fitted to the experiment result. Molecular dynamical computer simulation is one of the most powerful tools for studying assemblies of particles interacting through realistic inter-particle forces. I used this method to calculate the C₆₀ and silicon (111)-7 x 7 in van der Waals force interaction to see if the result can fit the experiment or not. The conclusion is that the interaction between C₆₀ and Si (111)-7 x 7 is not dominated by van der Waals forces. Image processing techniques are very important in this work. I devoted one chapter to describe the image processing in my data analysis. The technique I used here is not only good for the current work, it also could be used in other research areas. Chapter six discusses a simulation calculation to study scanning force microscope tip and sample interaction. The whole chapter is a paper that has been published in Nanotechnology.

The thesis shows the first monographs of Aleix Clapés and Manuel Sayrach, two important individuals of Catalonian culture, whose works were developed between 1880 and 1937; a long period which saw many importan historical events take place which were reflected in their work. These works on their merit alone, deserve a level of careful attention which they have never before received . This is the principal jusfitication of this thesis and the core reason why both bodies of work have been united within the scope of a single study. But there are other important parallels between them : Originality, anachronism, stylish ambiguity and extravagance; common characteristichs that allow us to understand a priori the problems faced by historians when trying to classify and interpret these works : They are such outsiders of the traditional catalogue of modern style that they are paid little attention. Nevertheless, there is another important connection between them : Antoni Gaudi. Competitor, close friend and colleague in important projects for Clapés; while for Sayrach, a mythical and inspirational figure , against whom he contrasted his works and architectural theories.lf we consider the difficulty with which the extraordinarily creative output of Gaudi was taken on board by the traditionally conservative local historiography, then the works of Clapés and Sayrach, being so close both formally and ideologically to those of Gaudi come up against a similar, but even greater, problem in that is not easy to place into the historiography of Catalonian art works that, while eccentric, are also "inferior" to those of Gaudi.
For this reason , our theory distances its elf from the conventions of the traditional discourse of modernism with the ¡ntention of getting closer to the historical truth. This new perspective allows the works and their authors to be truly considered artistic productions of great value and worthy of protection. The abundance of mentions and commentaries about Clapés' work in the contemporary press demonstrates that our initial hypotesis was correct: Clapés' place in history is merely a pale shadow of the real individual in his time and in his world . On the other hand , ¡nterest is awakened more and more in "casa Sayrach", the main work of Sayrach which highlights the disconnect between the role given to him by history and the authentic dimension of the individual.
Aleix Clapés (1846-1920) and Manuel Sayrach (1886-1937) were born and died in the province of Barcelona. They followed different reative, literary and cultural activities . Aleix Clapés with his pictures , advanced the local syrnbolism and he can be considered expressionist avant la lettre. The preflerence that Eusebi Güell and Pere Mila gave to his pictorial work, expresses de high esteem in which he was held by the Catalonian bourgeois. Clapés also designed the furniture for the entrance and living room for the Ibarz family (1902), the most powerful and beautiful of Catalonian syrnbolism, and in addition , bought and ran the influential magazine publication "Hispania" in its second period (1903). Manuel Sayrach, received the title of architect in 1917, when he had already reformed the family retreat known as "La Torre dels dimonis" (Sant Feliu de Llobregat, Barcelona , 1909-1913) and when he was finishing building the "casa Sayrach" (Barcelona, 1914-1918). Some years later, Sayrach built the "casa Montserrat" (Barcelona , 1926). He also conceived a dramatic project: "Els drames de la llum", from this project Sayrach published "Abelard i Eloïsa" (1919) and "Reigzel" (1920). He entered the field of political ideology with his work "República i Constitució" (1931).

Mainstream media has markedly depicted Salafism in quite monolithic ways, giving an impression of uniformity in ideology and practice of this community. This picture has moreover been framed in particularly negative ways when destructive phenomena like transnational terrorism, violent Jihadism and radical insurgency are closely associated with the movement. I decided to study how British Salafist responses to the London 7/7 bombings have been represented. The aim has been to study how such an atrocious act with the capacity to divide society has stimulated the will to live in peace or conflict; and moreover, I aim to look at how these responses have been portrayed by Salafis themselves over against the depictions given by mainstream media. I have however chosen to focus on the visual iconographic representations of these responses because I found them intriguing: “a picture paints a thousand words.” The thesis conducts a critical discourse analysis of these visual representations of Salafist responses to 7/7.

OBJECTIVE: Generalized anxiety disorder (GAD) during pregnancy is associated with several adverse maternal and perinatal outcomes. A reliable and valid screening tool for GAD should lead to earlier detection and treatment. Among pregnant Peruvian women, a brief screening tool, the GAD-7, has not been validated. This study aims to evaluate the reliability and validity of the GAD-7. METHODS: Of 2,978 women who attended their first perinatal care visit and had the GAD-7 screening, 946 had a Composite International Diagnostic Interview (CIDI). The Cronbach's alpha was calculated to examine the reliability. We assessed the criterion validity by calculating operating characteristics. The construct validity was evaluated using factor analysis and association with health status on the CIDI. The cross-cultural validity was explored using the Rasch Rating Scale Model (RSM). RESULTS: The reliability of the GAD-7 was good (Cronbach's alpha = 0.89). A cutoff score of 7 or higher, maximizing the Youden Index, yielded a sensitivity of 73.3% and a specificity of 67.3%. One-factor structure of the GAD-7 was confirmed by exploratory and confirmatory factor analysis. Concurrent validity was supported by the evidence that higher GAD-7 scores were associated with poor self-rated physical and mental health. The Rasch RSM further confirmed the cross-cultural validity of the GAD-7. CONCLUSION: The results suggest that the Spanish-language version of the GAD-7 may be used as a screening tool for pregnant Peruvian women. The GAD-7 has good reliability, factorial validity, and concurrent validity. The optimal cutoff score obtained by maximizing the Youden Index should be considered cautiously; women who screened positive may require further investigation to confirm GAD diagnosis.: This research was supported by an award from the Eunice Kennedy Shriver Institute of Child Health and Human Development (R01-HD-059835) at the National Institutes of Health (NIH). Esta investigación estudia en profundidad cómo fue su primitivismo, pasando por el análisis formal de algunas de sus obras con influencias directas del arte africano, hasta llegar a un desarrollo teórico sobre la performance ritual que amplía el primitivismo como un recurso del arte más allá del objeto estético. El contexto histórico durante la formación académica de Ana Mendieta y su paso por Nueva york son las claves que proporcionan una visión de sus obras hasta ahora inédita. La aplicación de una metodología comparativa ha permitido establecer conexiones entre el arte, la antropología, la arqueología, la historia colonial de España, la historia de las religiones y el feminismo.The career of Ana Mendieta is part of the aesthetic movements of the late twentieth century. Primitivism in art is one of them. This research examines in depth how her primitivism was, from the formal analysis of some of her artworks directly influenced by African art, to developing a theoretical rationale of the ritual performance that extends primitivism as an artistic resource beyond the aesthetic scope. The historical context behind the academic training of Ana Mendieta and the time she spent in New York are the keys that provide a glimpse into her hitherto unpublished works. This thesis proves that the evolution of life, thoughts and work of Joan Miró in the 1968-1983 period is deeply related with his historical context in its political,economic, social, cultural and artistic. A solid conductor is his social and artistic commitment, based on four fluent influences in ideological which were depositing from childhood to old age: Catalan nationalism, Catholicism, moral conservadurism and progressive political. Such influences were sometimes contradictory and this was a personal tension, that he resolve in this period of 1968-1983 with a greater involvement in the struggle for human rights and the Spanish democracy, which is manifested in his support for acts of opposition against Franco, with the milestone of his participation in the closure of Montserrat in 1970 and the aid to the oppressed victims, and his commitment to achieving the autonomy of Catalonia and the Balearic Islands, which eventually leads him, after moving Transition democracy in 1975, to become one of the leading artists of Catalonia and even Spain, as evidenced by the significant public commissions, the great exhibitions, the largest projects of two foundations at Barcelona and Palma of Majorca, and his excellent relations with the Kings of Spain, the Culture ministers of Spanish governments, the presidents of the Generalitat of Catalonia, and the mayors of Barcelona and Palma of Majorca.And all this is reflected in an artistic response with a renewed openness to the latest avant-garde artistic movements, which are now the Arte Povera, especially the use of debris; ephemeral art in 1969 the murals of the College of Architects of Barcelona and the Expo in Osaka; brutalism as the "toiles brûlées" in 1973; the return to dramatic expressionism of "wild paintings" which had already manifested in the most tragic times of the Civil War; the growing predominance of black and gestualism, what had already experienced in the sixties; the alternation between thought and spontaneous styles; the approach of his work in series and groups both in painting and other arts; the highest propensity to make larger works; and the use of almost all artistic useful means to achieve more widespread popular. The painting was always his favorite art, which shaped and informed the other arts. The original Miro's works are fertilized by his lively internal debate between tension to express universal ideas, with a tendency to abstraction, while his personal emotions, more prone to the use of figuration, using motives and methods of artistic tradition, the personal and collective subconscious, and the experimental will of the avant-garde transgression.

La idea es estudiar la estructura atómica de la superficie plana del Si (111) 77, así como la estructura de sus escalones. También estudiaremos mediante el microscopio de efecto túnel la diferencia entre los estados electrónicos ocupados y desocupados en las imágenes obtenidas variando el potencial aplicado por el microscopio entre la punta y la muestra. Lag (1936:81) om skuldebrev är en av få lagar som reglerar betalning. Skuldebrevslagen stiftades på 1930-talet, när de flesta betalningar gjordes med hjälp av sedlar och mynt. Idag sker emellertid betalningar ofta genom moderna betalningsmetoder såsom girering, konto- och kreditkortbetalning och e- betalning. 7 § Skuldebrevslagen reglerar betalningar av skulder ställda i en utländsk valuta, och paragrafen ger gäldenären en rätt att, istället för den avtalade valutan, betala i en gångbar valuta på betalningsorten. I uppsatsen utreds vilken funktion som 7 § Skuldebrevslagen kan och bör fylla när betalning görs via en modern betalningsmetod. Ändamålet med 7 § Skuldebrevslagen skall analyseras mot bakgrund av 1930-talets ekonomiska verklighet och utifrån denna analys skall vidare undersökas vilken funktion paragrafen fyller i dagens samhälle. Moderna betalningsmetoder omfattas av paragrafens tillämpningsområde eftersom dessa till stor del har samma funktion som traditionella former för betalning. En modifiering av 7 § Skuldebrevslagen skulle dock, enligt författarens mening, vara eftersträvansvärd eftersom den riskfördelning som paragrafen innebär till viss del blir oklar och kan medföra tillämpningsproblem när betalning görs via moderna betalningsmetoder.

Integreringen kommer utföras med hjälp av ett tilläggspaket till PCS 7, SIMATIC PCS7/Open OS. Styrsystemen som ska integreras i detta arbete är vanligt förekommande på industrier idag, SIMATIC S7-300/1500. Dessa styrs normalt inte via processkontrollsystem, mer vanligt är att de styrs lokalt via en operatörspanel intill maskinen. Genom att integrera dessa till processkontrollsystem kommer de att kunna styras från större kontrollrum, samtidigt som fördelarna med att integrera moderna styrsystem med ny teknik erhålls i produktionen. En annan del av studien har för avsikt att jämföra tidigare fall av liknande integreringar där andra typer av tekniker har använts. Dessa fall har visat att det finns idag standardiserade tekniker för att integrera enstaka fältenheter så att dessa kan hanteras av övergripande kontrollsystem. I en av studierna som undersöks, där det har gjorts försök att integrera funktionen av ett styrsystem som kontrollerar flertalet fältenheter, finns det enligt studien begränsningar i tekniken som används. Enligt Siemens AG ska dock tillägget SIMATIC PCS 7/Open OS klara att integrera ett styrsystem med dess fulla funktionalitet till processkontrollsystemet PCS 7. Resultatet av arbetet visar att integrering av styrsystemen till PCS 7 genom att använda tillägget är möjlig och styrsystemets fulla funktion kunde erhållas från ett PCS7-HMI. För att integreringen med tilläggspaketet ska ske smidigare rekommenderas att förberedelser, såsom att skapa bibliotek både i PCS 7 och i styrsystemet utförs innan integreringsarbetet påbörjas. Biblioteken bör innehålla funktionsblock förexempelvis motordrifter i styrsystemet och grafikelement som motsvarar dessa i PCS7.This study will focus on how Siemens controllers that are not listed in the productlibrary of the process control system PCS 7, can be integrated. The integration willbe performed using an option for PCS 7 (SIMATIC PCS 7/Open OS). The controllers to be integrated in this work are commonly found in industries today, the Siemens controllers SIMATIC S7-300/1500. These are usually not controlled from a processcontrol system, they are more commonly controlled via an operator panel next to the machine. By integrating these controllers into the process control system, they can be controlled from larger control rooms, and the production will acquire the benefits of using modern controllers. Previous cases with similar components, where other integration techniques was used has been evaluated. In these cases the integration have shown that there are today standardized techniques for integrating single field devices so that they can be managed by process control systems. In another study investigated, where attempts have been made to integrate the function of a controller that is controlling several field devices, the study showed limitations in the technique used. According to Siemens AG however, SIMATIC PCS 7/Open OS will be able to integrate acontroller with its complete functionality into the process control system PCS 7. The result of the work shows that integration of the controllers to PCS 7 by using the option package is possible, and the controller could be fully operated from a PCS 7-HMI. In order to make the integration with the optional package more user-friendly, it is recommended that preparations be made before integrating. Such as preparing a library in both PCS 7 and in the controller to be integrated, which are containing for example function blocks in the controller side and graphic elements corresponding to these function blocks in the PCS 7-HMI side.

L'ataxie spinocérébelleuse de type 7 (SCA7) est une maladie neurodégénerative due à une expansion de CAG traduit en polyQ dans la protéine ataxine-7. La SUMOylation, modification post-traductionnelle que nous avons identifiée moduler l'agrégation de la protéine mutante, est facilitée par une SUMO E3 ligase.Nous avons identifié RanBP2, une nucléoporine appartenant au complexe du pore nucléaire en tant que SUMO E3 ligase, via SUMO-1 de l'ataxine-7. En effet, le silencing de RanBP2 induit l'agrégation de l'ataxine-7 mutante, ce qui démontre l'implication de RanBP2 dans la physiopathologie de SCA7. Nous montrons également que l'ataxine-7 endogène est une cible modifiée par SUMO-1 et -2. L'ataxine-7 poly-SUMOylée, grâce à la présence de chaines SUMO2/3, est capable de recruter RNF4. Cette protéine conduit à la dégradation de l'ataxine-7 mutante par la voie du protéasome. La dégradation est abolie en présence d'un mutant de RNF4.Dans un modèle murin KI SCA7, nous avons quantifié l'expression des gènes impliqués dans la voie de la SUMOylation au niveau des régions les plus touchées du cerveau. Le niveau d'expression des ARNs messagers montre des altérations dépendantes des répétitions CAG du gène SCA7. A 6 mois (avant le début de la pathologie), les premières dérégulations sont observées; à 12 mois (à un stade avancé de la maladie), on note une diminution statistiquement significative de Sumo-1 dans le cervelet des souris Atxn7100Q/5Q. Ces résultats, alliés à l'observation de l'accumulation anormale des protéines SUMO-1 et RanBP2 dans le cervelet d'un patient SCA7, suggèrent que les voies de la SUMOylation in vivo peuvent être perturbées dans SCA7Spinocerebellar ataxia type 7 (SCA7) is a neurodegenerative disease caused by a CAG expansion (polyQ) in the protein ataxin-7. SUMOylation, a post-translational modification that we identified to modulate mutant protein aggregation in a SCA7 cellular model, is facilitated by a SUMO E3 ligase. Here, we identified RanBP2 (Nup358), a nucleoporin belonging to the nuclear pore complex, as the major E3 enzyme implicated in ataxin-7 modification by SUMO-1. Indeed, RanBP2 silencing renders mutant ataxin-7 more prone to aggregation, thus demonstrating the implication of RanBP2 in SCA7 pathophysiology. We also show that endogenous ataxin-7 is a target for both SUMO-1 and -2 modification. Poly-SUMOylated ataxin-7 presents a docking site composed of SUMO2/3 chains for the recruitment of RNF4: this protein is a SUMO E3 ubiquitin ligase that mediates degradation of mutant ataxin-7 by the proteasome pathway. The degradation is abolished in presence of a mutant form of RNF4. In a SCA7 knock-in mouse model we quantified expression of SUMO-pathway related genes in cerebellum and retina, the most affected regions using quantitative RT-PCR. SUMO-related genes show expanded repeat-dependent alterations in expression patterns. At 6 months (before onset), deregulations begin to occur; by 12 months (late stage of disease), there is a statistically significant impairment in Sumo-1 levels in Atxn7100Q/5Q cerebellum. These results, together with the observation that SUMO-1 and RanBP2 protein accumulate abnormally in the cerebellum of a SCA7 patient, suggest that in vivo SUMO-modifying pathways may be perturbed in SCA7

Deux types de reacteurs de depot chimique en phase vapeur ont ete utilises pour la realisation de films minces d'yba#2cu#3o#7## et prba#2cu#3o#7# ## #, ainsi que de multicouches alternant ces deux composes. La source du premier reacteur est constituee de quatre fours contenant chacun un des precurseurs organometallique (m-tetramethyl-heptane-dionate) sous forme de poudre. La source du second reacteur est constituee d'un reservoir contenant un melange liquide solvant - precurseur, sous pression, a temperature ambiante, relie a une micro-vanne ou injecteur. Cette derniere source permet de controler avec precision la quantite de precurseur injectee vers la zone de depot. Des couches minces de ybco et de pbco ont ete deposees. Dans les deux cas, les films sont epitaxies sur le substrat (srtio#3 ou laalo#3) et presentent les caracteristiques suivantes : pour yba#2cu#3o#7## t#c = 92 k, < 0,5 k et j#c(77k) = 5. 10#6 a/cm#2, pour prba#2cu#3o#7##, (100 k) < 5. Cm. Un lien direct a ete mis en evidence entre la composition des films deposes et la composition de la solution source dans le cas de la cvd a injection. D'un point de vue experimental, cette derniere technologie parait plus adaptee pour la croissance de composes aussi complexes que ybco ou pbco. Des multicouches alternant supraconducteur et isolant, caracterisees par des periodes variant de 50 a 330 a ont ete realisees et caracterisees. Des analyses de diffraction de rayons x (/2, -scan, -scan et reflectometrie) indiquent que les empilements sont reguliers et epitaxies sur le substrat (laalo#3 et srtio#3). Une analyse en spectrometrie d'ions retrodiffuses (rbs) et en microscopie electronique a transmission permet de confirmer la regularite des couches d'ybco et de pbco constitutives de la multicouche. Enfin l'anisotropie des empilements a pu etre determinee par l'analyse du comportement de la resistivite des multicouches sous champ magnetique intense (16 t) inclines.

Malgré leur efficacité pour réduire la réplication du VIH, les traitements antirétroviraux ne s’accompagnent pas systématiquement de la restauration du compartiment des lymphocytes T CD4+ périphériques. L’espérance et la qualité de vie des individus en échec immunologique sont grandement impactées. Concomitante avec les anomalies périphériques, une atteinte des progéniteurs hématopoïétiques rend compte des fréquentes cytopénies observées au cours des stades tardifs de l’infection. Si l’infection directe des progéniteurs CD34+ reste marginale, les études qualitatives menées in vitro évoquent la perturbation du potentiel de différenciation de ces cellules.Nous avons sélectionné et apparié les patients infectés par le VIH à chaque extrême quand au profil de leur restauration immunitaire et un groupe de donneurs non infectés. Les répondeurs (IR) et non répondeurs immunologiques (INR) traités depuis plus de 8 ans présentaient les caractéristiques similaires pour chaque paramètre pouvant impacter la magnitude de la reconstitution du système immunitaire. Les INR montraient l’activation chronique du système immunitaire, l’inflammation persistante et les signes de l’atteinte de la thymopoïèse. La fréquence des progéniteurs hématopoïétiques circulants n’étant pas différente entre les deux groupes de patients, nous avons analysé le potentiel de ces cellules aux stades pré-thymiques de la différenciation.En utilisant un système de co-culture des progéniteurs hématopoïétiques avec une lignée stromale OP9-DL1 (différenciation T) ou MS5 (différenciation B et NK) avec un cocktail cytokinique approprié, nous avons mis en évidence l’altération du potentiel de différenciation T des cellules issues de patients INR impactant leur restauration périphérique. Ce n’était pas le cas chez les patients IR qui étaient similaires aux donneurs non-VIH. En revanche, le potentiel NK était impacté chez tous les patients infectés en comparaison aux donneurs. Finalement aucune anomalie de potentiel B n’était révélée.En étudiant les voies moléculaires de l’engagement des précurseurs T (Notch), de leur prolifération (IL-7/IL7R) et leur survie (Fas/FasL, TNFR, caspase-1, P2X7) nous avons constaté une diminution de la viabilité des progéniteurs hématopoïétiques chez les patients VIH+ qui présentaient d’avantage d’activation de la caspase-1 qui orchestrait la mort cellulaire par pyroptose. De plus, l’expression de certains gènes-cibles de Notch était clairement Notch-indépendante. Néanmoins, les différences dans le profil transcriptionnel de BCL11B entre les patients IR et INR nous ont permis de proposer un modèle selon lequel la différenciation T était promue chez les patients IR au dépit de celle des précurseurs NK. Enfin, les progéniteurs CD34+ de patients INR présentait la surexpression du P2X7 (récepteur à l’ATP extracellulaire) et l’absence de l’ectonucléotidase CD73 (hydrolyse de l’ATP) ce qui suggérait leur susceptibilité accrue aux nucléotides extracellulaires.L’ensemble de données nous permet de postuler qu’il existe un microenvironnement hautement inflammatoire dans la niche médullaire des progéniteurs hématopoïétiques chez les patients VIH+ qui perturbe leur survie et différenciation. Cette mortalité accrue des cellules CD34+ et probablement des cellules voisines amplifie l’inflammation locale. Ce processus est compensé chez certains patients par la meilleure différenciation T des progéniteurs CD34+ et la réponse immunologique qui s’en suit. Quand ce n’est pas le cas l’atteinte de la lymphopoïèse est importante et l’absence de la reconstitution de la population des lymphocytes T CD4+ périphériques est observée. Ainsi, nous pensons avoir identifié une population de patients infectés par le VIH pour qui les interventions ponctuelles avec les médicaments anti-inflammatoires (par exemple, les antagonistes du P2X7) peuvent s’avérer d’un bénéfice clinique irréfutableDespite the efficient reduction of the HIV replication, the administration of combination antiretroviral therapy (c-ART) is not systematically accompanied by the restoration of the peripheral T CD4+ lymphocyte compartment. The life expectancy and quality are severely impacted in individuals with immunological failure. Together with peripheral abnormalities, an alteration of CD34+ hematopoietic progenitor may explain the frequency of the cytopenia observed in the latest stages of the disease. While a direct infection of CD34+ progenitors is thought to be extremely rare, quantitative studies performed in vitro have highlighted the impairment of the differentiation potential of these cells.We selected and matched individuals infected with HIV presenting extremely opposite immunological profile in response to c-ART as well as non-infected donors. The Immune Responders (IR) and Immune Non Responders (INR) treated since more than 8 years, presented similar characteristics for each parameter known to be involved in poor reconstitution of immune system. INR patients showed chronic immune activation, persistent inflammation and thymic regeneration failure. The frequency of circulating CD34 hematopoietic progenitors being not different between both groups of patients, we analyzed the differentiation potential of these cells at pre-thymic stages of lymphopoiesis.Using a co-culture system of hematopoietic progenitors with stromal cell lines OP9-DL1 (T-cell assay) or MS5 (B- and NK-cells assay) with appropriate cytokines, we highlighted an alteration of T-cell differentiation potential in INRs impacting their peripheral restoration. This was not observed in IRs who were similar to non-HIV donors. On the other hand, NK-cell differentiation potential was impaired in both groups of patients in comparison to non-HIV donors. Lastly, no abnormalities in B-cell potential were revealed.Studying molecular pathways involved in T-cell specification (Notch), proliferation (IL-7/IL7R) and survival (Fas/FasL, TNFR, caspase-1, P2X7) we observed the decreased viability of hematopoietic progenitors in HIV patients with increased caspase-1 activation involved in cellular death by pyroptosis. Moreover, expression of some Notch target genes was clearly Notch-independent. However, differences in transcriptional profile of BCL11B between IRs and INRs allowed us to postulate that T-cell differentiation is promoted over NK-cell differentiation in IR patients. Finally, CD34+ cells from INRs presented P2X7 overexpression (extracellular ATP receptor) and absence of CD73 ectonucleotidase (ATP hydrolysis) pointing out their increased susceptibility to extracellular nucleotides.Taken together our data, we postulate that highly inflammatory microenvironment of hematopoietic progenitor’s bone marrow niche disturbs their survival and differentiation in HIV patients. Thus, increased cellular death of CD34+ cells and probably neighboring cells amplifies the local inflammation. This is compensated in some patients by enhanced T-cell differentiation of CD34+ progenitors and results in immunological success. When it is not the case, the alteration of lymphopoiesis is important and the absence of reconstitution of peripheral T CD4+ lymphocyte compartment is noted. We believe have identified the population of HIV-infected individuals who will benefit from occasional administration of anti-inflammatory drugs (such as P2X7 antagonists)

We have studied the deposition of Ge on two types of Si(111)-(7×7) surfaces: one with regularly spaced steps and the other consisting of alternating structure of high step density region (step-bunch region) and a step-free terrace. The surface with step-bunching structure is induced by a DC current heating with electric current running in the step-down direction whereas the surface with evenly spaced steps has the heating current running in the step-up direction. Atomic force microscopy and scanning electron microscopy are employed to investigate the Si surfaces after Ge being deposited at different temperatures and annealing times. For deposition of Ge at 300°C, we have found that Ge clusters form uniformly on the Si surface, regardless of the types of the surfaces. Though we cannot resolve the detailed structure of small Ge clusters, the surfaces of large Ge clusters are composed of many facets, presumably low-energy surfaces of Ge. For 500°C deposition, the surface displays a completely different morphology where Ge wires on the order of ~µm in length form for both types of surfaces that we have studied. It is noted that the micron-size Ge wires always grow in the direction parallel to surface steps. For a surface with evenly spaced steps, the Ge wires distribute randomly. In comparison, the Ge wires form exclusively in the step-bunching region for the other type of surface. This directional growth indicates the diffusion of Ge atoms is highly anisotropic, a result of the presence of surface steps. The top layer of the Ge wire is rough, i.e., different regions have significant height differences, indicating the formation of a long Ge wire may result from the coalescence of short Ge wires. Annealing smoothens the wire’s outer layer.

Perhaps, the most prominent neurotransmitters can be ascribed to serotonin and melatonin. In many neuropsychiatric disorders, the serotonin or melatonin system may play a key modulatory role. Although the pathophysiology of many neuropsychiatric disorders remains enigmatic, the abnormalities in serotonin or melatonin signaling have been strongly implicated. Such a small molecule like serotonin or melatonin acting as a vital element in brain function has attracted much attention in the synthetic chemistry. Focus on derivatization of serotonin or melatonin has been one of the intensive areas in the medicinal chemistry. While most of the approaches lie in the substitution effect on the parent serotonin or melatonin, we have long been interested in replacing the parent indole moiety by the 7-azaindole chromophore, forming 7-azaserotonin, 7-azamelatonin. Herein, we elaborated first synthesis of 7-azaserotonion in ~10 % total yield and rose up the yield of 7-azamelatonin by the synthetic route different from the synthetic pathway of Guillaumet in ~6 % total yield. Subsequently, we further investigated the photophysical properties of 7-azaserotonin and 7-azamelatonin, namely the excited state proton transfer reaction. Differentiation of photophysics between alcohol and water are intriguing. In view of bio-application, we suspect that once 7-azaserotonin and 7-azamelatonin are in the hydrophobic medium, water catalyzed proton transfer reaction may take place, serving as suitable molecular probes. We also performed the molecular dynamic approaches for 7-azaserotonin (and 7-azamelatonin) to investigate the difference of interaction to the 5-HT1B receptor (and calmodulin) between serotonin (melatonin) and 7-azaserotonin (7-azamelatonin). Our preliminary data indicate that 7-azaserotonin (7-azamelatonin) has larger association strength than that of serotonin (melatonin). Thus, 7-azaserotonin may serve as a powerful 5-HT1B agonist or antagonist to treat aggression or depression, respectively. 7-azamelatonin could be taken to treat estrogen responsive cancers more efficiently. We believe that exploration of 7-azaserotonin and 7-azamelatonin may spark a broad spectrum of interest in the fields of medicinal chemistry and biophysics.
