Abstract:
Abstract Antibody-drug conjugates (ADCs) have become increasingly adopted clinically, with 13 drugs FDA-approved and over 100 under clinical development. Despite this momentum and major advances in payload and linker technology, ADCs are limited by off-cancer target-mediated toxicities incurred by currently available targets. In contrast to genomic and proteomic strategies, our unique drug discovery approach accounts for a disease’s native context. This led to the identification of cancer-specific plectin (CSP) as not only overexpressed in malignant tissue compared to healthy, but exclusively present on the surface of cancer cells and absent in normal or pre-malignant tissue. A Phase 0 imaging trial (Biodistribution of Novel Imaging for Resectable Pancreatic Cancer - NCT01962909) evaluating a radiolabeled CSP-targeted peptide has revealed that CSP is bioavailable and abundant, with over a million CSP molecules per cancer cell in pancreatic cancer. Previously, we generated ZB131, a humanized monoclonal antibody that targets CSP. ZB131 demonstrated potent monotherapy efficacy in pancreatic, ovarian, and bile duct preclinical cancer models, indications that have high CSP expression and are being evaluated in a first-in-human Phase 1/2 clinical trial (NCT05074472). Here, we show that CSP is an ideal target for an ADC: it is abundantly and selectively expressed in many indications, bioavailable, and its inhibition is predicted to synergize with FDA-approved payloads. ZB131 binds specifically to CSP, rapidly internalizes, displays linear pharmacokinetics in pre-clinical models, and demonstrates a strong safety profile. We describe the development of two ZB131 ADCs, ZB131-MMAE (monomethyl auristatin E) and ZB131-DXd (Deruxtecan), with drug-to-antibody ratios of 3 – 4 and 7 – 8, respectively. After binding, both ADCs are rapidly and specifically internalized by CSP-positive cells resulting in drug payload release and enhancing cancer cell death compared to ZB131 alone. Moreover, we characterize cytotoxic activity in high and low CSP cell lines to evaluate ADC activity in relation to CSP abundance. In preclinical xenograft models, ZB131-ADC enhanced tumor regression compared to controls at clinically relevant doses. Furthermore, anti-huIgG staining revealed selective target engagement by ZB131-ADC. Taken together, these data show that ZB131-ADCs demonstrate potent antitumor activity and support their evaluation in a Phase 1 clinical trial. Citation Format: Samantha M. Perez, Brian P. Abstract:
To avail the possible pharmacological actions of Brideliaferruginea Benth., the present investigation was designed to quantitatively analyze the total flavonoid and phenolic contents and assess the various antioxidant and enzyme inhibition properties of leaf and stem bark extracts (ethyl acetate, water and methanolic) of B. ferruginea. Anti-proliferative effect was also investigated against human colon cancer cells (HCT116) as well as the antimicrobial potential against multiple bacterial and fungal (yeasts and dermatophytes) strains. The methanolic and water extracts of the stem bark demonstrated the highest phenolic content (193.58 ± 0.98 and 187.84 ± 1.88 mg/g, respectively), while the leaf extracts showed comparatively higher flavonoid contents (24.37–42.31 mg/g). Overall, the methanolic extracts were found to possess the most significant antioxidant potency. Compared to the other extracts, methanolic extracts of the B. ferruginea were revealed to be most potent inhibitors of acetyl- and butyryl-cholinesterases, tyrosinase α-amylase, except α-glucosidase. Only the ethyl acetate extracts were found to inhibit glucosidase. Additionally, the stem bark methanolic extract also showed potent inhibitory activity against E. coli and gram-positive bacteria (MIC (minimum inhibitory concentration): 2.48–62.99 µg/mL), as well as all the tested fungi (MIC: 4.96–62.99 µg/mL). In conclusion, B. ferruginea can be regarded as a promising source of bioactive compounds displaying multifunctional pharmacological activities and thus is a potential candidate for further investigations in the endeavor to develop botanical formulations for pharmaceutical and cosmeceutical industries.

Abstract:

Abstract:
A new approach to detailed statistical inference for the two-decision and multiple decision problem was developed by Chatterjee and Chattopadhyay (1992, 1993). The procedure developed for the multipledecision case is used here to find a solution to, what may be called, the problem of estimation by a ‘confidence set’ when the parameter space is finite. The confidence set problem in which we are required to state a datadependent measure of ‘confidence’ which can be interpreted in terms of betting odds that are ‘optimal’ under repeated experimentation, is considered. A truncated form of utility function is introduced. The case of the truncated form of logarithmic utility function is considered in detail. AMS {2000} Subject Classification: 62A01, 62F99.

Abstract:
In this paper, we concentrate on the statistical properties of Gamma-X family of distributions. A special case of this family is the Gamma-Weibull distribution. Therefore, the statistical properties of Gamma-Weibull distribution as a sub-model of Gamma-X family are discussed such as moments, variance, skewness, kurtosis and Rényi entropy. Also, the parameters of the Gamma-Weibull distribution are estimated by the method of maximum likelihood. Some sub-models of the Gamma-X are investigated, including the cumulative distribution, probability density, survival and hazard functions. The Monte Carlo simulation study is conducted to assess the performances of these estimators. Finally, the adequacy of Gamma-Weibull distribution in data modeling is verified by the two clinical real data sets. Mathematics Subject Classification: 62E99; 62E15

Abstract:
Designs combioatorially balanced for residual treatment effects of various orders, (especialJy first and second orders) with and without interactions were discussed in considerable details by williams (1949). Construction of combinatorially balanced designs for residual treatment effects was also considered by Patterson (1952); Hedayat and Afsarinejad (1975, 1978); Cheng and Wu (1980). An excellent account of such designs is given by Jones and Kenward (1989). In this paper, a method of construction of generalized cyclic designs combinatorially balanced for residual treatment effects of the first order without interactions, is presented. The method is based on an extension of theorems on symmetricaliy repeated difference sets developed by Bose (1939) and others. Abstract:
Abstract: The paper addresses the problem of speaker identification based on spectrograms in the text dependent case. Using spectrogram segmentation, this paper, mainly, focusses on understanding the complex patterns in frequency and amplitude in an utterance of a given word by an individual. The features used for identifying a speaker based on an observed variable extracted from the spectrograms, rely on the distinct speaker effect, his/her interaction effect with the particular word and with the frequency bands of the spectrogram. Performance of this novel approach on spectrogram samples, collected from 40 speakers, show that this methodology can be effectively used to produce a very high success rate in a closed set of speakers for text-dependent speaker identification. Abstract:
The sanitary sewage is treated with four different vegetation types by means of artificial surface runoff. Results show soil+ Cynodon dactylon L. + Festuca arundinacea Schreb is the best vegetation type in treating the sanitary sewage, and the total removal of it is 80.24% TN, 62.99% TP, 74.85% BOD5, 39.73% COD and 58.64% TOC respectively in the course of trial, which indicates that the mixed vegetation type made up of plants and soil is better than the pure soil vegetation type, and the complementary collocation of complex plants is better than the single plant.Five water-quality indexes of the sanitary sewage removed by the single vegetation type are also studied, and it shows that there are some differences among them from high to low as follows: TN>BOD5>TP>TOC>COD.

Abstract:
Abstract Background: Disease-free survival (DFS) and overall survival (OS) follow 1st order kinetics. Curve Popkin assessment yields biological insights. Chemotherapy (chemo) improves outcome in resected NSCLC. We performed adjuvant DFS and OS PopKin assessments. Methods: We digitized published DFS and OS curves. We used GraphPad Prism for curve exponential decay nonlinear regression analysis (EDNLRA). Results: We assessed 1 trial with adjuvant osimertinib and 5 with platinum-based postoperative chemo and control arms. With osimertinib, DFS and OS curves were convex on log-linear plots, suggesting rapid tumor regrowth once therapy is discontinued. In chemo trials, 10 of 10 DFS curves and 9 of 10 OS curves fit 2-phase decay EDNLRA models (Table 1). Popkin calculations indicate that of control patients destined to recur, 12% would recur by 2 months after surgery, 22% by 4 months and 31% by 6 months. Conclusion: PopKin suggests osimertinib delays recurrence rather than preventing it. Combined chemo analyses suggest chemo converts 15% of patients from rapidly to slowly recurring, but many “shifted” patients eventually recur. Hence, chemo may induce reversible tumor senescence rather than eradicating micrometastases. Anti-senescent cell strategies might prove beneficial. Frequent scans are warranted in the 1st postoperative year for early detection of recurrences. Table 1. Abstract:
This paper describes the analysis of fatal incidents of Indian coal mines from April 1989 to March 1998. It is found that Indian mines have considerably higher accident and fatality rates compared to those in USA and South Africa, respectively. While open cast mines are generally known to be safer than underground mines, the Indian open cast mines are shown to be at least as hazardous to the workers as the Indian underground mines. Analysis of the accident rates is made via a few regression models involving the effects of working shifts, the various companies, the types of mine, manshift and production. The accident-prone combinations of mine type and company are identified for follow-up action. The break-up of the accidents by cause is also studied. Abstract:
The urgency of the issue of grain production as the most important branch of agricultural production is described. Equally important for refined grains has such an indicator of quality of grain as it is damage that at harvest and post-harvest handling especially when grain reaches significant values. Therefore, we evaluated the quality of grain handling of various cultures on the production line of winnowing machine 3AB-40 LLC "Fakel" of Petropavlovsky district of Voronezh region. The results of studies of the 3AB-40 are presented, quality performance of machines are defined that are part of the production line. Nature of changes in the purity of the grain at the output of each machine of production line is considers. Detailed table of the fractional composition of the grain heap of cultures during processing on machines 3AB-40 is presented, in particular the composition of the grain heap entering the post-harvest treatment on the content the whole grain, crushed grain and weeds. A plot of the purity of the grain heap is presented when it is processed. Also the level of injury of seeds separately for each culture, and as it passes through each machine 3AB-40 is detailed. Analyzing numerous studies it was found that post-harvest technology of grain heap and technical resources for its implementation should ensure the allocation of the weeds at a very early stage of processing, biologically inferior grain and most of the injured grain. From above it follows the need to improve this production line in order to increase the purity of grain at the output and reduce its damage. Recommendations for installing in the process line of pre-cleaning machine МПО-50, better air-sieved cleaning machine, for example, ОЗФ-50 and ОЗФ-80. That will provide almost complete separation of light impurities, large impurities and partially puny grain, thereby improving the operation of subsequent machines that will surely affect the purity of the grain at the output of the line.

Abstract:
The present study aimed to determine the optimum extraction conditions of conventional solvent extraction (CSE) and microwave-assisted extraction (MAE) techniques to obtain maximum total phenolic (TPC), total flavonoid (TFC), total anthocyanin (TAC) and antioxidant capacity (AA). Response surface methodology (RSM) and central composite design (CCD) were used to determine optimum points of CSE and MAE. Both two extraction methods, all parameters significantly affected TPC, AA, TFC and TAC (p<0.05). MAE showed higher bioactive compounds yield than that of CSE. Optimum point for CSE and MAE was found to be 60°C and 30 min and 591.83 W and 2.98 min respectively. TPC, AA, TFC and TAC were obtained as 33.81mg/g, 160.27 mg/g, 11.89 mg/g and 331.01 mg/kg for CSE and 62.99 mg/g, 214.62 mg/g, 21.80 mg/g and 3462.93 mg/kg for MAE respectively. This study recommended that the MAE should be used for the extraction of PBL to increase phenolic extraction yield.

Abstract:
The geochemical processes of fractional crystallization of a magma, partial fusion of a rock and assimilation or hybridization of rock(s) and/or magma(s) are generally termed as petrological mixing process. In the present paper a unified attempt has been made to describe the three processes under the purview of regression model. As the data involved are essentially compositional in nature, their suitable log-ratio transforms have been utilized and the constrained least squares principle has been applied to reach a meaningful solution. A highly accommodative procedure is suggested so as to describe any specified d~gree of fractional crystallization of magma or of partial fusion of a rock. The paper concludes with applications of the proposed method on data available from i) Rajmahal Trap, Eastern India (fractional crystallization) ii) Bihar Mica Belt, Eastern India (partial fusion) and iii) Kilauea Volcano, Hawai (formation of hybrid magma). Abstract:
Abstract Spirituality is a social determinant of health associated with psychosocial adjustment to serious illnesses including cancer but is not often assessed in clinical care. Here we perform psychometric evaluation of a tool developed to report spiritual well-being in chronic illness in a cohort of 248 patients with primary brain tumors (PBT) enrolled in a natural history study (NCT02851706, PI: T. Armstrong). Participants completed the Functional Assessment of Chronic Illness Therapy-Spiritual Well-Being 12 (FACIT-SP 12) as part of yearly clinical outcome assessments. Total score, Meaning/Peace and Faith scores were calculated. Internal consistency (Cronbach’s alpha) and construct validity (principal components analysis with varimax rotation) evaluated psychometric properties of the FACIT-SP 12; one-way ANOVAs, one-sample t-test and Wilcoxon signed rank tests evaluated group differences (sig. p<0.05) on patient report of spirituality/beliefs including belief in god(s)/higher power, being religious, and prayer and in comparison to scores of previously reported studies. Participants were majority male (57%), white (79%) with high grade tumor (59%) and a median age of 48 (range: 20-82). Seven different religions were reported (majority Christianity [55%]) but also spiritual non-religious (9%), no beliefs (13%), and 6% preferring not to answer. The principal components analysis confirmed the original 2 factor solution (Meaning/Peace and Faith) and explained 66.8% of the variance (KMO =0.87, 43% of residuals < 0.05), with one item not loading cleanly on either factor (“I know that whatever happens with my illness, things will be okay”). Cronbach’s alphas were 0.90, 0.90, and 0.89, for the total, Meaning/Peace, and Faith scales, respectively. For all three spiritual/belief questions, patients who responded “yes” had higher total and faith scores than those who responded “no"; but not in the Meaning/Peace score when reporting religion (p=0.108). This cohort reported lower total scores (mean=32.5), Meaning/Peace scores (mean=23.4, median=24.0) and Faith scores (mean=9.2, median=10.0) compared to the initial validation in other solid tumors and HIV/AIDs (means=38.5, 25.2, 13.6, respectively) and to a smaller, less diverse PBT sample (medians: Meaning/Peace=25.8, Faith=14.0). The FACIT-SP 12 demonstrated internal consistency and construct validity, with lower scores in comparison to populations in prior studies. More diverse belief systems than previously reported may contribute to differences among studies. Future analysis should explore the increased percentage of “prefer not to answer” respondents compared to earlier reports, associations with clinical and self-reported outcomes including symptoms and quality of life, evaluation of illness-related factors that contribute to poor spiritual well-being, and a longitudinal look at spiritual well-being. Abstract:
Antimicrobial coated fabrics have a wide spectrum of use in grooming, biomedical and water purification applications. In this work, we report the antimicrobial property of a nano blend prepared from crab-shell based chitosan and nano ZnO and dip-coated on a silk fabric sample. All the synthesized materials were subjected to FT-IR, XRD and TGA studies to assess the functionality, crystallinity and thermal stability. Deacetylation of chitin to chitosan was revealed by the decrease in the intensity of the acetyl group peak at 1641 cm-1 in chitosan. The crystallite size of the synthesized nano ZnO was found be 15 nm from XRD analysis. Appearance of the characteristics peaks of chitosan (9.58°, 19.98°) and ZnO (31.86°, 34.71°, 36.44°, 56.65°, 62.99° and 68.02°) together in XRD pattern of the blended sample confirmed the blending of chitosan and nano ZnO. The deposition of the nanoblend on the coated silk fabrics was shown by SEM analysis. An inhibition zone of 3mm around the nanoblend coated silk fabric sample against E. coli bacteria clearly indicated that synthesized chitosan/ZnO nanoblend possessed good antibacterial activity.

Abstract:
Summary The mechanical strength of a reservoir formation is the most crucial information required for predicting sand production and recommending sand control completion. So far, the only reliable technique to obtain the formation strength quantitatively is to perform laboratory tests on core samples. The laboratory tests require substantial volumes of cores, which in most cases are not available. In this study, we present a new method to avoid this restriction. The significance of this approach is in its simplicity and efficiency in constructing a reliable mechanical failure envelope. The key results of this study, based on measurements on a variety of sandstones, are the following.A single normalized failure envelope characterizes sandstone formations. This universal curve makes it possible to construct the failure envelope for a sandstone formation from the knowledge of critical pressure. There exists a correlation between the critical pressure and the compressional wave velocity (at equivalent depths of burial). The failure envelope for a sandstone formation can then be constructed simply from compressional wave velocities. These velocities are generally accessible from conventional logging data. Introduction A major problem encountered during hydrocarbon production is the influx of sand, or sand production.1 It can cause severe damage to both production equipment and the producing formation. Furthermore, remediation processes after sanding are extremely difficult, costly or often impossible. Although significant research has been conducted on sand production, we are still at an embryonic stage in predicting sand influx. Several factors determine sand production. The most critical factors are (1) formation strength; (2) in-situ stress; and (3) production rate. The hydrocarbon production process is associated with reservoir depletion, which results in a decrease of reservoir pore pressure. Consequently, the effective overburden pressure, defined as total overburden pressure minus pore pressure, increases. Formation collapse is most likely if the effective stress exceeds the formation strength. In addition, production rate increase, which is associated with large fluid pressure gradients near the borehole, tends to draw the sand into the wellbore. Generally, one can estimate the in-situ stress. For example, the horizontal minimum stress can be measured from hydraulic fracture testing,2-4 and the overburden pressure from overburden density data. The production rate is a controllable parameter. The parameter of concern is the formation strength, which is the focus of this study. The most reliable technique for obtaining mechanical strength data is triaxial testing of core samples in the laboratory. With appropriate arrangements of applied stresses one can determine a failure envelope in stress space. Such a failure envelope quantifies the stress conditions under which the material fails. Although the laboratory test can provide dependable mechanical strength data, it is not followed routinely simply because it is time-consuming and costly. Moreover, in most cases, a sufficient amount of core is not available. Traditionally the mechanical strength, or Mohr-Coulomb failure criterion, is estimated from P- and S-wave velocities and density log data5-8 based on the correlation of Deere and Miller.9 This approach estimates uniaxial compressive strength, and assumes a constant frictional angle. In this study, we seek an alternative method of estimating overall nonlinear mechanical strength in a three-dimensional stress space. Basic Concept of Failure Envelope Formation collapse is an indication that the in-situ stress is beyond the failure limit of the formation material. This failure stress limit is a quantitative parameter that defines the formation mechanical strength. For a one-dimensional state of stress, the mechanical strength can be simply quantified with a single parameter: the uniaxial compressive strength. However, because the in-situ formation stress is three dimensional and anisotropic, a more complicated mathematical expression involving all the stresses is required to quantify the mechanical strength. This quantitative expression of mechanical strength is known as the failure envelope or failure criterion.
