Abstract:
Abstract Background: Alpelisib and fulvestrant are used as a combination treatment option for postmenopausal PIK3CA-mutated, hormone receptor positive (HR+), human epidermal growth factor receptor 2-negative (HER2-), advanced or metastatic breast cancer (a/mBC) patients. However, despite the presence of activating mutations in PIK3CA, the majority of patients do not derive benefit, or ultimately progress while on alpelisib therapy. Here, we investigate the genomic landscape of PIK3CA-mutated, HER2- a/mBC using next-generation sequencing (NGS) to provide insight into possible mechanisms of therapeutic resistance to alpelisib/fulvestrant and to identify potential targetable pathways. Methods: We utilized the Tempus LENS platform to retrospectively analyze de-identified NGS data from 2,918 a/mBC patients with formalin-fixed, paraffin-embedded tumor biopsies sequenced using the Tempus|xT solid tumor assay (DNA-seq of 595-648 genes at 500x coverage; full transcriptome RNA-seq). Mutations identified included germline and/or somatic single nucleotide variants, insertions/deletions and copy number variations (gains defined as ≥8 copies). We used curated clinical data to determine HER2 and hormone receptor (ER/PR) status. Results: Among 2,918 a/mBC patients, we identified somatic mutations in PIK3CA in 782 (26.8%). Within these tumors, 629 (80.4%) had one of the 11 mutations currently included in the alpelisib companion diagnostic, and we focused on this population (here defined as mut-PIK3CA). Of these 629, 546 (86.8%) were HER2-, with 176 (32.3%) and 370 (67.7%) derived from primary and metastatic tumors, respectively. Cases were further classified as HR+ (defined as ER+ or PR+) or triple negative (TNBC). While the majority of mutPIK3CA samples were identified in HR+ disease, 10% of the cases occurred in TNBC. Within the mutPIK3CA cohort, tumor mutational burden high (TMB-H; defined as ≥10 mutations/MB) was detected in 11.5% of samples, while microsatellite instability high (MSI-H) was detected in 0.5%. MSI-H was detected at a higher frequency in TNBC compared to HR+. Overall, the most commonly co-mutated genes among mutPIK3CA, HER2- samples were TP53 (34.6%), CDH1 (21.6%), ESR1 (12.3%), KMT2C (11%), MAP3K1 (9.5%), ARID1A (8.1%), PTEN (6.8%), GATA3 (6.6%), NF1 (5.9%), and TBX3 (5.9%) among others (Table 1); some of these genes have previously been implicated in resistance to endocrine therapy or PI3K inhibitor. In addition, in HR+ disease, metastatic samples had a higher frequency of mutations in genes implicated in endocrine resistance, such as ESR1 (18.7% vs 1.9%), ERBB2 (3.3% vs 2.6%), NF1 (6.8% vs 2.6%), compared to primary tumors. We also identified copy number gains (CNG) in several cell cycle genes, including: CCND1 (15.2%), CDK4 (2.7%), and AURKA (2.6%) (Table 1). Finally, further analyses at the transcript-level are the subject of on-going research. Conclusions: Our study highlights that there is substantial genomic heterogeneity among mutPIK3CA, HER2- a/mBCs. Across a series of comparisons between primary and metastatic samples, as well as HR+ and TNBC subtypes, we identified a number of co-mutations that occur alongside mutPIK3CA and which could be potentially exploited by targeted therapies. Future studies are needed to assess the prognostic/predictive role of these and other candidate gene alterations. Table 1. Abstract:
Abstract Sepsis is characterized by a severe shift in metabolism, characterized by low IGF-I levels. We have studied the influence of caecal ligation and puncture (CLP) on the levels of circulating IGF-I and hepatic IGF-I and IGF-binding protein (IGFBP)-1, -2 and -3 mRNA in adult male Wistar rats (n=12) and compared it with sham-operated rats (n=6). In order to exclude anorexia-induced changes we also studied animals pair-fed to both groups. IGF-I levels were measured by RIA. Steady-state hepatic IGF-I, IGFBP-1, IGFBP-2 and IGFBP-3 mRNA levels were measured by Northern blot analysis using specific rat cDNA probes. Food intake averaged 13·0 ± 2·0 g/day in the sham-operated rats fed ad libitum during the study period, with a sharp decline in food intake in the CLP animals (2·3 ± 1·3 g/day). After CLP, there was a significant reduction in circulating IGF-I levels (467·2 ± 50·9 μg/l) compared with sham-operated animals (924·0 ± 75·3 μg/l; P=0·04) or those pair-fed to the CLP rats (612·5 ± 52·9 μg/l; P=0·04). Total hepatic IGF-I mRNA levels were significantly reduced (2·57 ± 0·05 densitometric units (DU)) after CLP compared with the sham-operated group (2·71 ± 0·04; P=0·04), or their pair-fed controls (2·75 ± 0·08 DU; P<0·05). Hepatic IGFBP-3 mRNA levels were lower after CLP (0·37 ± 0·04 DU) than in the sham-operated animals (0·66 ± 0·09 DU; P=0·04) or their pair-fed controls (0·61 ± 0·05 DU; P=0·04). On the other hand, hepatic IGFBP-2 mRNA levels were increased after CLP (0·91 ± 0·11 DU) compared with sham-operated animals (0·28 ± 0·06 DU; P=0·01) or with their pair-fed controls (0·22 ± 0·22; P=0·01), as were hepatic IGFBP-1 mRNA levels (CLP animals 0·95 ± 0·11 DU; sham-operated 0·30 ± 0·04 DU, P=0·01; pair-fed 0·30 ± 0·02 DU, P=0·01). No significant difference between sham-operated animals and their pair-fed controls was observed in circulating IGF-I levels (888·0 ± 109·3 μg/l; P=not significant (N.S.)), hepatic mRNA levels for IGF-I (2·72 ± 0·06 DU; P=N.S), IGFBP-3 (0·71 ± 0·07 DU; P=N.S.), IGFBP-2 (0·25 ± 0·07 DU; P=N.S.) or IGFBP-1 (0·27 ± 0·06 DU; P=N.S.). In summary, after CLP there was a reduction in both circulating and hepatic IGF-I mRNA levels associated with a specific and differential regulation of hepatic IGFBP-1, -2 and -3 mRNA levels. Although we cannot eliminate a possible effect of surgical stress combined with malnutrition, our results suggest that these changes are a specific effect of sepsis rather than simply a result of surgical stress or poor nutrition alone. Objective. To describe the yield of smear-microscopy and radiological findings by male and female patients with symptoms of tuberculosis in Abuja, Nigeria.Methods. Patients ≥15 years old with cough for >3 weeks submitted 3 sputum samples for smear microscopy. One specimen was cultured using MGIT-960. All patients had lung X-rays and screened for HIV.Results. were more likely to be smear-positive than females (262/774 [34%] and 137/547 [25%],P<.01), but similar proportions of males and females were culture-positive (437/691 [63%] and 294/495 [59%],P=.09). 317/626 (50.6%) males and 249/419 (59.4%) females were HIV-positive (P<.005). Among culture-positives patients, HIV-infected males were less likely to have positive smears than HIV-negative males (49.2% versus 66%,P=.001). Among females, smear positivity did not vary with HIV (46.4% for HIV-positive and 52.9% for HIV-negative,P=.38). Of 274 culture-confirmed TB cases, 226 (82.5%) had cavities, and 271 (99%) had ≥1 lung areas affected. HIV-positive males were more likely to have lung cavities than HIV-positive females (85% versus 69%,P<.04) and to have ≥3 lung areas affected (P=.03).Conclusion. Differences in the yield of smear-microscopy, culture and X-rays on presentation are due to several factors including HIV coinfection and gender.

SummaryPure and mixed swards of Centrosema pubescens (centro), Pueraria phaseoloides (puero) and Brachiaria mutica (para grass) established under a 12-year-old mango plantation were sampled at 6-week intervals for 2 years to determine crude-protein (CP) content, dry-matter (D.M.) and CP yields and apparent digestible energy and apparent digestible crude-protein (DCP) contents. Daily dry-matter intake (DMI) and apparent digestible energy and digestible crude-protein intakes (DEI and DCPI), as well as live-weight gain (LWG) by 11·5 kg penned West African cross-bred weaner lambs, were also determined.The pure legumes were better than the mixed swards in respect of CP (mean: 22·1, 18·8, 14·4 and 13·8% for centro, puero, centro-para grass and puero-para grass respectively) and DCP (mean: 16·2, 13·4, 10·4 and 10·0% respectively) contents, and daily DMI (mean: 50·8, 49·7, 41·8 and 41·3 g/kg W0·75 respectively), DEI (mean: 562, 531, 418 and 410 kj/kg W075 respectively) and DCPI (mean: 8·09, 6·55, 4·04 and 3·94 g/kg W0·75 respectively). On the other hand the legume-grass mixed swards, especially puero-para grass, were superior to the corresponding pure-legume swards with regard to D.M. yield (mean: 11·90, 7·69, 7·13 and 4·54 t/hafor puero-para grass, centro-para grass, puero and centro respectively) and CP yield(mean: 1·62,1·09, 1·32 and 0·99 t/ha respectively). The pure-legume stands gave the highest LWG per lamb (mean: 45·7, 3·89, 2·63 and 25·3 g/day for centro, puero, centro–para grass and puero-para grass respectively) but it was estimated that the legume-grass mixed swards would give higher returns in terms of lamb LWG on forages harvested per unit sward area than the corresponding pure-legume cover.It was concluded that legume-grass covers would be preferred to all-legume covers if livestock were to be integrated into tree crop farming systems. However, the choice of companion grass would have to be given proper consideration as shade-intolerant grass species, such as para grass, would detract from profit maximization.

10089 Background: Masitinib is a tyrosine kinase inhibitor that in vitro has greater activity and selectivity than imatinib against KIT. This multicenter, open label, phase 2 study evaluated efficacy and safety of masitinib as a first-line treatment of advanced GIST. Initial results with a median follow-up of 34 months, were previously reported in EJC 2010. We present here 5-year follow-up data from the same series with updated safety and survival data. Methods: Imatinib-naïve patients (pts) with inoperable, advanced GIST received oral masitinib (7.5 mg/kg/day) until progression, refusal or toxicity. Results: Thirty pts with a median age of 58 years (60% of males) were included from 06/2005 to 04/2007 in 5 institutions. At the cut-off date of 13/09/2011, 7/30 pts (23%) were still under treatment with median follow-up of 65 months and the median overall survival (OS) had not yet been reached (NR). The 5-year OS rate was 61.5% (95% CI [41.1;76.6]) (see Table). Among patients with confirmed KIT exon 11 mutation, 8/9 pts (89%) were still alive with one pt having died from non-treatment related causes (surgical complication) following a complete response while receiving masitinib. No additional progressions have been reported giving a total of 14 events (13 progressions and 1 death). Updated median PFS was 41.3 months (95% CI [17.5;53.8]).No additional grade 3/4 adverse events have been reported. Conclusions: Long term results of masitinib confirm an interesting activity with prolonged PFS and OS. The median PFS rate in masitinib compares very favorably to that of imatinib, which is reported as 18 months (JCO 26:626, 2008). The 5-year OS rates for masitinib in the overall and exon 11 populations also compare favorably to imatinib, both of which are reported at ≤50% for imatinib (JCO 26:626, 2008; JCO 28:1247, 2010). These results support the head to head comparison with imatinib in the currently ongoing phase 3 randomized clinical trial in first line advanced GIST pts. [Table: see text]

Background: Women suffering from seizure-related illnesses are prescribed phenytoin during pregnancy if the benefits outweigh the risks. But the teratogenic effect of this drug on limb development needs exploration. This study was designed to investigate the time-mediated teratogenic effect of sodium phenytoin on limb development of chick embryos on days 04 and 09 of development using the stereomicroscope and at the time of hatching. Material and Methods: This randomized control trial comprised of two main groups of fertilized chicken eggs (Egyptian Fayoumi breed), control group A and experimental group B, each having 90 eggs. Each experimental egg was injected with sodium phenytoin (3.5 mg, teratogenic dose) just before incubation. Both the groups were divided into three subgroups each A1, A2, & A3/B1, B2 & B3. Stereomicroscope was used to observe the limb buds and cartilaginous elements in subgroups 1 and 2 on days 04 and 09 of development respectively. The embryos of subgroups 3 were assessed for gross limb deformities on hatching. Survival was noted in all the subgroups.Results: The difference in limb bud size between experimental and controls was statistically insignificant. There was no variation or deformity in the bones. The experimental group 3 had gross limb defects on hatching and the difference in survival was also statistically significant in subgroups 3.Conclusion: Prenatal administration of sodium phenytoin induces limb defects in chick embryos as evident after hatching but limb anomalies are not observed on days 04 and 09 of development as investigated by stereomicroscope. We conclude that the teratogenic effect of phenytoin is time-mediated.

Abstract Background: Flaxseed has been extensively studied in in vitro and in vivo models for possible association with reducing the risk of breast cancer. Clinical trials have been limited. There has been confusion regarding whether it is safe and beneficial for breast cancer survivors to take flaxseed. Therefore, we conducted a systematic review to evaluate the existing randomized controlled trial to understand the safety and efficacy of using flaxseed for symptom control and reducing the risk of recurrence among breast cancer survivors. Patients and Methods: We conducted a literature search of English language publications in MEDLINE, EMBASE, and Cochrane from the databases’ inception to May 2021 using the keywords flaxseed, breast cancer, randomized clinical trial, and human studies. Results: We identified three randomized controlled trials (RCT) that include patients with a history of breast cancer. Sample size ranged from 24 to 178 patients and treatment duration ranged from 13 to 42 days. The primary end points were hot flash severity (one study) and tumor biomarkers (two studies). There was no statistically significant reduction in mean hot flash score in the flaxseed group compared to the placebo group. One study showed a statistically significant reduction in Ki-67 (34.2%; P=0.001) and in c-erbB2 expression (71.0%; P=0.003), and an increase in apoptosis (30.7%; P=0.007) in the group that received flaxseed, but not in the placebo group. On the contrary, another study showed no difference in Ki-67, caspase, the Ki-67/caspase ratio, and Estrogen-receptor β. Two studies reported mild adverse events as shown in Table 1. None of the studies assessed the change in estrogen level. Risk of bias assessment showed that the overall risk of bias associated with the included studies is moderate to high. The high risk of bias originates from lack of blinding of participants and personnel. Conclusion: Current clinical research in breast cancer survivors is too limited to recommend flaxseed for cancer prevention. Flaxseed appears to be safe with mild side effects, however, the effect of flaxseed on estrogen concentration has not been established. Two pathology classifications were used: one with 4 categories and one with 5 categories (Goldhirsch et al 2011). Based on BP 3 subtypes are formed: Luminal, HER2 and Basal. The Luminal subtype is further split into Luminal A (MP low risk) and Luminal B (MP high risk). Results: See table. Conclusions: All pathological Basal cases are BP Basal, apart from 1 BP HER2 case. Of the BP Basal cases, 15 are not pathological Basal: 1 is Luminal A, 11 are Luminal B (of which 8 are IHC ER/PR borderline (≥1% and < 10%)) and 3 are HER2. No primeiro ensaio aplicou-se 1/3 da dosagem de fertilizante utilizada no segundo. O delineamento experimental foi inteiramente casualizado, com três repetições. As linhagens foram classificadas quanto à eficiência na absorção de nutrientes e reposta à adubação baseando-se nos incrementos de índice DRIS e nos incrementos de produtividade. Os valores críticos para eficiência na absorção e resposta à adubação foram as médias de incremento de índice DRIS e produtividade, respectivamente. As linhagens diferenciaram-se quanto à eficiência na absorção dos nutrientes e quanto à resposta à adubação. Foram consideradas responsivas à adubação e eficientes na absorção de nutrientes as linhagens 03, 04, 05, 09 e 22, para o N; 03, 04, 09, 13, 15 e 29, para o P; 03, 05, 10, 21, 22, 25 e 27, para o K; 05, 10, 21, 22, 25, 27 e 29, para o Ca; 04, 13, 15, 27 e 29, para o S e B; e 03, 05, 09, 10 e 27, para o Cu. As linhagens com os melhores desempenhos foram a 27, na absorção dos nutrientes, e 03, 04, 05 e 29, na responsividade à adubação.

Aims:To contrast the outcomes of olanzapine- and valproate-treated patients in an observational study of acute mania with the results of a RCT assessing the same treatments (Tohen et al., 2002).Methods:EMBLEM (European Mania in Bipolar Evaluation of Medication) was a 2-year, prospective, observational study of health outcomes associated with treatment of mania. Severity of mania and depression was assessed at baseline and 6 weeks using the YMRS and 5-item version of the HAMD, respectively. The RCT was a 3-week, randomised, double-blind comparison of olanzapine (n=125) and divalproate-treated (n=123) patients hospitalised for acute manic or mixed episodes. The YMRS and HAMD were used to quantify manic and depressive symptoms, respectively.Results:621 EMBLEM patients were analysed (n=107 valproate, n=514 olanzapine). Both observed groups improved from baseline to 6 weeks in mean YMRS and HAMD-5 total scores, with significantly greater mean improvements in the olanzapine compared with the valproate group using linear regression to adjust for baseline differences. The RCT reported significantly greater mean YMRS improvement (but not HAMD) in the olanzapine-treated group. EMBLEM patients treated with olanzapine experienced significantly greater weight gain than patients treated with valproate, similar to RCT results. There was a significantly greater incidence of treatment-emergent gastrointestinal adverse events in EMBLEM patients treated with valproate.Conclusions:The EMBLEM results support those of the RCT, which suggest that olanzapine monotherapy may be more effective than valproate monotherapy in the treatment of acute mania. Contrasting observational and RCT results present methodological challenges but can provide important complementary information.

Penelitian ini menggunakan pendekatan kuantitatif dengan metode SEM-PLS serta melibatkan responden sebesar 100 responden dengan menggunakan Purposive Sampling dengan kriteria seorang muslim usia 18-35 dan seorang mahasiswa. Software analisis untuk mengolah data dalam penelitian ini menggunakan Smartpls 3.3, untuk data diperoleh dengan menggunakan kuesioner online menggunakan skala likert 1 sampai 5, dengan keterangan sangat setuju hingga tidak setuju. Hasil dari penelitian ini menunjukkan bahwa kepercayaan, citra website, kebermanfaatan aplikasi, dan kemudahan penggunaan berhubungan signifikan terhadap niat penggunaan wakaf online. Implikasi hasil penelitian ini diharapkan dapat memberikan tambahan pengetahuan terutama dalam ilmu pemasaran Islam terkait penggunaan behavioral intention dalam penggunaan layanan wakaf online, serta diharapkan penelitian ini dapat memberikan masukan bagi manajemen lembaga donasi terutama nazir untuk lebih memperhatikan kemudahan penggunaan konsumen, manfaat yang dirasakan, kepercayaan lembaga, citra perusahaan serta niat dalam menggunakan layanan wakaf secara online.Kata kunci: Wakaf online, technology acceptance model, trust, image. ABSTRACTThe purpose of this study was to examine the effect of trust and website image, which is integrated with perceived ease of use and perceived usefulness, on a student's intention to pay waqf online. What makes this research different from previous research is using website images to determine student intention in using this online waqf service. This study uses a quantitative approach with the SEM-PLS method and involves 100 respondents using purposive sampling with the criteria of a Muslim aged 18-35 and a student. The analysis software to process data in this study used Smartpls 3.3 for data obtained using an online questionnaire using a Likert scale of 1 to 5, with statements strongly agreeing to disagree. This study indicates that trust, website image, application usefulness, and ease of use are significantly related to the intention to use online waqf. The implications of the results of this study are expected to provide additional knowledge, especially in Islamic marketing related to the use of behavioral intention in the use of online waqf services. The composition of leukocytes reflects the species and ecological characteristics of fish, while the ratio of individual cell types reflects the functional state of the organism and the nature of the influence of biotic and abiotic stress factors. The work is devoted to the issues of hematopoiesis of the bream Abramis brama — one of the main commercial species of the Volga. The fish were caught in September 2019 at the Saratov Reservoir during a complex hydrobiological expedition aboard the Akademik Topchiev research vessel of the IBIW RAS. In total, 18 sexually mature bream were examined, caught at 3 standard trawling stations: 1 — Shirokiy Buerak, 2 — Khvalynsk, 3 — Kashpir-Rudnik. In fish, peripheral blood was sampled from the tail vein and tissues of the spleen and head kidney. Blood smears and organ imprint smears were performed for the study. The preparations were examined under a light microscope and the leukocyte formula was determined. To determine the index of the abundance of leukocytes in a peripheral blood smear, 100 visual fields were examined at different sites. The leukocyte abundance index allows the intensity of leukopoiesis to be recorded and indirectly assesses the leukocyte level in a blood volume unit. The results of the study showed that lymphocytes predominate in the leukocyte formula, a significant part are blast forms and a small percentage of granulocytes and monocytes/macrophages. The established differences depended on the structural and functional features of the organs: in the kidneys, as the main organ of hematopoiesisin teleost fish, a higher content of blast forms of cells, granulocytes and macrophages was noted, and in the spleen, which serves as a “depot” of blood, lymphocytes and macrophages. Blood, as an integral system, bears all types of cells circulating in the immune system at the moment. The intensity of leukopoiesis was higher in fish from the upper part of the reservoir.

The study attempted to analyze the role of right to information in ensuring good governance in Pakistan. The study objectives were to investigate the experiences of public information officers and civil society actors regarding the right to information contribution to good governance and to analyze the challenges in the implementation of right to information law. For the selection of sampling size, purposive sampling was used and data were collected from 11 ministries public information officers and 3 civil society actors through in depth interviews in Islamabad, Pakistan. Being qualitative study, data were analyzed by using thematic analyses. Jurgen Habermas theory of ‘Public Sphere” was used to throw light on main theme of the study. The findings indicated that right to information is an effective tool in promoting good governance along with all its features like, accountability, transparency, mutual trust between the government and general public, common man facilitation, rule of law and corruption elimination. Study concluded that right to information would be an effective tool in promoting the good governance only when hurdles that prevent it from its true implementation were removed.

All anthropometric and biochemical assessments were performed twice during a year (summer, winter). High BMI (Z-score >1), low HDL cholesterol (<40 mg/dl, males; <50 mg/dl, females) and high TAG (>150 mg/dl) were considered cardiometabolic risk factors.ResultsSerum 25-hydroxyvitamin D (25(OH)D) showed between-season variation, with significantly higher concentrations (mean (sd)) in summer v. winter (43 (29) v. 27 (18) nmol/l; P<0·001). Change of circulating 25(OH)D between summer and winter was negatively correlated with change of BMI (r=−0·16; P<0·001), TAG (r=−0·09; P=0·04) and total cholesterol (r=−0·10; P=0·02) and directly correlated with change of height-for-age Z-score (r=0·09; P=0·04). Multiple stepwise linear regression analysis (β; 95 % CI) showed that winter serum 25(OH)D (−0·3; −0·4, −0·2; P<0·001), gender (boys v. girls: 9·7; 5·2, 14·1; P<0·001) and latitude (>33°N v. <33°N: 4·5; 0·09, 9·0; P=0·04) were predictors of change of serum 25(OH)D between two seasons.ConclusionsSummertime improvement of vitamin D status was accompanied by certain improved cardiometabolic risk factors, notably serum TAG, total cholesterol and BMI, in children.

The Intergroupe Francophone du Myélome (IFM) initiated 2 trials in 1999 to study patients with high-risk (β2-microglobulin level greater than 3 mg/L and chromosome 13 deletion at diagnosis) de novo multiple myeloma. In both protocols, the induction regimen consisted of vincristine, doxorubicin, and dexamethasone (VAD) followed by first autologous stem cell transplantation (ASCT) prepared by melphalan 200 mg/m2. Patients with an HLA-identical sibling donor were subsequently treated with dose-reduced allogeneic stem cell transplantation (IFM99-03 trial), and patients without an HLA-identical sibling donor were randomly assigned to undergo second ASCT prepared by melphalan 220 mg/m2 and 160 mg dexamethasone with or without anti–IL-6 monoclonal antibody (IFM99-04 protocol). Two hundred eighty-four patients—65 in the IFM99-03 trial and 219 in the IFM99-04 trial—were prospectively treated and received at least one course of VAD. On an intent-to-treat basis, overall survival (OS) and event-free survival (EFS) did not differ significantly in the studies (medians 35 and 25 months in the IFM99-03 trial vs 41 and 30 months in the IFM99-04 trial, respectively). With a median follow-up time of 24 months, the EFS of the 166 patients randomly assigned in the tandem ASCT protocol was similar to the EFS of the 46 patients who underwent the entire IFM99-03 program (median, 35 vs 31.7 months), with a trend for a better OS in patients treated with tandem ASCT (median, 47.2 vs 35 months; P = .07). In patients with high-risk de novo MM, the combination of ASCT followed by dose-reduced allogeneic transplantation was not superior to tandem dose–intensified, melphalan-based ASCT.

The article presents the results of the first research on artificial reproduction in industrial conditions of the endemic, narrow-areal subspecies of Brachymystax lenok Savinovi, which lives in lake Markakol, East Kazakhstan region. The indicators of the heat sum characteristic for each stage of development, the rate of development and growth of the Markakolsky lenok from the moment of pre-breeding to late juveniles are described. To develop technological approaches for artificial fish reproduction, one of the most important points is to determine the optimal conditions for each stage and assess the risks (loss of fish products). At the stage of insemination and transportation of eggs to the place of incubation, the loss was 50 %. The largest losses of fish products were registered during the incubation stage. The most painlessly passed the period of holding and lifting on the float, where the loss was only 3 %. When growing pre-larvae and larvae in the pool, the daily waste did not exceed 1 %. Small-sized animals that were unable to adapt to artificial feeds fell into the waste. Losses during this period amounted to 15 % of the previous stage. In General, the yield of juveniles from the moment of fertilization to the end of the experiment was 16 %. The crucial factor in the development and growth of Lenok Markakolosky is the temperature regime. For the period of embryonic development, the most favorable water temperature is 7–8 °C. From the moment of hatching, the water temperature must be increased to 10–12 °C, and the optimal temperature for the cage growing of fingerlings varies from 12 to 14 °C.

A study of changes in some immunological parameters of fish blood serum depending on the average monthly air temperature was carried out. The studies were carried out on bream aged 3+, with an average weight of 375 ± 30 g and a length of 290 ± 20 cm, caught in the Volga reach of the Rybinsk reservoir. The fish were caught using fixed nets in February for several years and the average monthly air temperature was recorded. The studied fish were sampled from the tail vein. Blood was collected in a dry sterile test tube and kept at room temperature. After blood coagulation and retraction of the clot, the serum was taken with a syringe with a fine needle and transferred to another tube. To assess the state of natural immunity in the blood serum, the level of bactericidal activity and nonspecific immune complexes was analyzed. Based on the data obtained, the number of immunodeficient individuals was determined. Variations of the studied parameters in years different in average monthly air temperature were established. The greatest differences between the indicators were revealed in the years with the highest (2003) and lowest (2006) mean monthly temperatures. The minimum indicators of bactericidal activity were recorded in 2003, and the maximum — in 2006. The highest content of immune complexes and immunodeficient individuals was established in 2003, and the lowest in 2006. The study showed the relationship between an increase in the average monthly air temperature and a decrease in functional activity nonspecific humoral immunity. Based on the data obtained, it was concluded that the temperature factor influences the mechanisms of innate immunity of fish.

SummaryOur study evaluated thigh circumference (TC), stifle range of motion (ROM), and lameness in dogs one to five years after unilateral tibial plateau levelling osteotomy (TPLO). We hypothesised that TC, stifle ROM, and lameness would not be different to the unoperated limb (control), one to five years after surgery. Patients that were one to five years post-TPLO were reviewed and were included if they had a unilateral TPLO, and no additional clinical evidence of orthopaedic disease. Standing mid-thigh TC measurements and stifle extension and flexion angles were made in triplicate. Clinical lameness was graded blindly. Data were evaluated statistically using paired t-tests for TC and stifle flexion and extension. Significance was set at p <0.05. Twenty-nine dogs met the inclusion criteria. Mean results for the surgery limbs and control limbs were 39.5 ± 5.5 cm and 40.1 ± 5.6 cm for TC, 36.6 ± 6.8° and 28.6 ± 4.3° for stifle flexion, and 155.2 ± 6.6° and 159.8 ± 4.9° for stifle extension, respectively. The mean TC for the operated limb was 98.5% of the control limb. A significant difference was found between the operated and the control limbs for all measurements. Time after surgery had no apparent affect on outcome. Four of 29 dogs (14%) exhibited some lameness in the TLPO limb during evaluation (one dog was 1 to 2 years postoperative and three dogs were 2 to 3 years postoperative). These results indicate that TC and stifle ROM in the TLPO limb do not return to control-limb measurements one to five years after a TPLO surgery. The clinical significance is unknown as TC returned to 98.5% of control, and the source of lameness in the lame dogs was not identified.

The article discusses some aspects of the domestication of common catfish (Silurus glanis) in the conditions of cyprinid fish farms. Under the pressure of selection and directed cultivation in pond conditions of carp fish farms, changes in ethological, morphological, and physiological indicators are observed. The variability of the majority of morphological indicators and development indices (8–14%) indicates a high degree of consolidation of the resulting breeding offspring and indicates the completion of the first stage of domestication in the conditions of fish-breeding carp farms. Physiological and biochemical assessment of different age categories of catfish showed low variability of physiological indicators. it Can be assumed that the decrease in the coefficient of variability is a consequence of domestication. Studies of the immunological status have shown its optimal level, which allows us to accept the obtained indicators as indicative standards for evaluating brood flocks and their offspring. There was a significant increase in the body weight of commercial three-year-olds of the 4th breeding generation, compared with the first generation. It is established that the reproduction of catfish planted in carp summer-fallow ponds is more successful than in natural reservoirs, since ponds of this category are often small, well-warmed, which contributes to the rapid maturation of producers and the development of caviar. When growing in pond conditions for 2–3 generations, changes in behavioral reactions were noted — there was a decrease in aggressiveness and the development of conditioned reflexes to the specified food objects. The level of food availability affects the metabolism and maturation period, so along with the use of the natural feed base of the pond, it is recommended to use additional feeding and directed formation of the natural feed base.
