Full textThis thesis considers the development of a variable speed drive motor based on the Switched Reluctance Motor (SRM) 8/6 for an application in an automotive air-conditioning (A/C) system. A complete A/C system model + cabin in Matlab-Simulink are established to study the performance in a refrigeration cycle. It is complemented by the inclusion of a SRM drive model, designed particularly for the application. We present a set of guidelines for the development and application of the SRM when operating at 42V. This means to take into account the effects of magnetic saturation of the material (Imax=65A). The nonlinearities of the machine are developed modelling the nonlinear characteristic flux-current-rotor position (-i-θ) and electromagnetic torque (T-i-θ), for which it has used commercial software of electric machines design. The SRM drive uses a speed/torque control; besides the temperature control in the cabin utilizes a fuzzy control. The validation uses a similar feature motor with different load points.

Full textThe main aim of the study is to determine the minimal fuel consumption of a parallel hybrid vehicle that fulfill the driver's request. First, a model representing the energy flows inside the vehicle was set. Then local control algorithms were developed to determine the optimal gear ratio and torque repartition between the motors by minimizing a criterion including consumption and driveability aspects. A second point of the thesis consists in extracting the optimal behavior of the car on a planned trip. A global optimization that computes the minimal consumption on the planned trip was developed. Considering a vehicle with a navigation system, the analysis of the results allows to improve the gear ratio and the battery charging management of the local algorithms. Fast prototyping tests and vehicle tests were made to validate this approach

Este plan de Marketing ha sido desarrollado con la finalidad de aprovechar la coyuntura de la industria minera (baja en el precio del cobre1) e introducir, en Chile, un respirador de alta calidad adecuado a esa coyuntura, con el objetivo de lograr incrementar la participación en el segmento de respiradores reutilizables filtrantes de MSA. MSA es el líder mundial en el desarrollo, fabricación y suministro de productos de seguridad que protegen a las personas y las infraestructuras de las instalaciones. Muchos productos de MSA integran una combinación de electrónica, sistemas mecánicos y materiales avanzados para proteger a los usuarios contra las situaciones peligrosas o potencialmente mortales. Debido a la baja del precio del cobre2, reducción de personal en la industria minera3 y a la búsqueda de la reducción de costos de las compañías4, se ha configurado un escenario atractivo para la introducción de un respirador adecuado que le permitirá a MSA quitar participación de mercado a las demás marcas. Este plan de marketing está orientado a soportar ese objetivo. Full textObjective: The aim of this study is to compare in vitro the antibacterial activity and the cytotoxic effect of polyalcohols (Xylitol E-967, Sorbitol E-420, Mannitol E-421) on the bacterial strain Streptococcus mutans (ATCC® 35668 ™). Materials and Methods: The present study was experimental in vitro. To determinate the antibacterial effect was used Cerebral Heart Infusion Agar (BHI) plates with differents concentrations of polyalcohols (Xylitol E-967, Sorbitol E-420 and Mannitol E-421) inoculated with strains of Streptococcus mutans (ATCC® 35668 ™). The minimum inhibitory concentration (MIC) and the antibacterial effect was evaluated by the microdilution method. The cytotoxic effect (CC50) of the polyalcohols was evaluated against the MDCK cell line by the MTT colorimetric test. Results: The antibacterial effect was observed after 72 hours, the Xylitol polyalcohol had antibacterial effect on strains of Streptococcus mutans up to the concentration 1x10-5 with 0 colony forming units (CFU), while Sorbitol and Mannitol had countless CFU in all concentrations. The MIC of the Xylitol Polyalcohol was 1x10-5. In the evaluation of the cell viability of Xylitol, Sorbitol and Mannitol, the result was greater than 85% in all concentrations. Conclusion: Xylitol polyalcohol showed a higher antibacterial effect than Sorbitol and Manitol polyalcohols. Regarding the cytotoxicity of the polygons, none presented toxicity at the cellular level.

Full textENGLISH ABSTRACT: Worldwide, bacteriocins, particularly those produced by food-related lactic acid bacteria, are receiving attention due to the possible use of these peptides as natural preservatives in food, replacing potentially harmful chemical preservatives. Bacteriocins are ribosomally synthesized proteins or peptides that inhibit closely related microorganisms. Most bacteriocins produced by lactic acid bacteria are small, heat resistant peptides that inhibit other Gram-positive bacteria, including food-borne pathogens such as Listeria monocytogenes, Bacillus cereus, Clostridium perfringens and Staphylococcus aureus, but do not inhibit Gram-negative bacteria, molds or fungi. Bacteriocins are produced as inactive prepeptides that become active after the N-terminal leader peptide is cleaved off. Small heat resistant bacteriocins are either lantibiotics (Class I), containing unusual posttranslationally modified amino acids, or peptides that are non-Ianthionines (Class II). The Class II bacteriocins are further divided into four different groups: Class lIa, the anti-listerial bacteriocins containing the YGNGV consensus sequence in the N-terminal of the protein, Class lib, bacteriocins consisting of two peptides, Class IIc, bacteriocins that are secreted via the sec pathway, and Class lid, bacteriocins that do not belong in the previous three subgroups. A bacteriocin producing lactic acid bacterium was isolated in our laboratory from traditionally home fermented South African sorghum beer. The producing bacterium was found to be a facultative heterofermentative Lactobacillus sp. and was identified as Lactobacillus plantarum or Lactobacillus pentosus by using the API 50 CHL carbohydrate fermentation system and numerical analysis of total soluble cell protein patterns. RAPD-PCR analysis identified the strain as L. plantarum, but 16S rRNA sequencing confirmed its identification as L. pentosus. The bacteriocin, first designated plantaricin 423 and later bacteriocin 423, was identified as a Class lIa small heat resistant anti-listerial bacteriocin containing the YGNGV consensus motif. Bacteriocin 423 inhibited a variety of Gram-positive bacteria, including Lactobacillus spp., Leuconostoc spp., Oenococcus oeni, Pediococcus spp., Enterococcus spp., Propionibacterium spp., Staphylococcus spp., Bacillus spp., Clostridium spp. and Listeria spp. The bacteriocin was inactivated by proteolytic enzymes and active over a wide pH range (pH 1-10). Bacteriocin 423 lost 50 % of its activity after autoclaving for 15 min at 121°C, but was not affected by lesser heat treatments. Bacteriocin production was increased by optimizing the growth medium, which consisted of glucose, tryptone, yeast extract, potassium phosphate, sodium acetate, ammonium citrate, manganese sulphate, Tween 80 and casamino acids. The bacteriocin was found to be plasmid-encoded. Genetic analysis of the bacteriocin operon indicated a high percentage of homology to the operon of another Class lIa bacteriocin, pediocin PA-1, although the structural genes of the two bacteriocins were markedly different. The structural gene of bacteriocin 423 was amplified by PCR and cloned into a yeastJE. coli vector between the ADH1 promoter and terminator sequences and fused in-frame to the MFa1 secretion signal sequence. Saccharomyces cerevisiae transformed with this plasmid expressed the bacteriocin. The sequence of prebacteriocin 423 (MMKKIEKL TEKEMANIIGGKYYGNGVTCGKHSCSVN WGOAFSCSVSHLANFGHGKC) is similar, but not identical to any other reported Class lIa anti-listeria I peptide.
AFRIKAANSE OPSOMMING: Bakteriosiene, veral dié wat deur melksuurbakterieë geproduseer word, wek belangstelling as gevolg van die moontlike gebruik van hierdie natuurlike antimikrobiese proteiëne as preserveermiddels in voedselprodukte, in plaas van potensieël gevaarlike chemiese preserveermiddels. Bakteriosiene is ribosomaal-vervaardigde proteiëne wat naverwante bakterieë inhibeer. Die meeste bakteriosiene wat deur melksuurbakterieë geproduseer word, is klein en hittebestand. Hierdie bakteriosiene inhibeer ander Gram-positiewe bakterieë, insluitend patogene soos Listeria monocytogenes, Bacillus cereus, Clostridium perfringens en Staphylococcus aureus, maar inhibeer nie Gram-negatiewe bakterieë, giste of swamme nie. Bakteriosiene word as onaktiewe prepeptiede geproduseer, wat ge-aktiveer word wanneer die N-terminale leierpeptied afgesplits word. Klas lie is sec-afhanklike bakteriosiene, en Klas lid sluit in al die bakteriosiene wat nie in die eerste drie groepe geklassifiseer kan word nie. 'n Bakteriosien-produserende melksuurbakterie is uit tradisionele tuisgefermenteerde Suid- Afrikaanse sorghumbier geïsoleer. Die bakterie is as 'n fakultatief heterofermentatiewe Lactobacillus sp. geïdentifiseer. Die bakterie is verder as 'n Lactobacillus plantarum of Lactobacillus pentosus geïdentifiseer deur middel van die API 50 CHL-koolhidraat fermentasiesisteem en numeriese analiese van totale oplosbare selproteiënprofiele. Met RAPD-PCR analiese is die organisme as L. plantarum geïdentifiseer, maar 168 rRNA nukleotiedopeenvolging het die identiteit van die organisme as L. pentosus bevestig. Bakteriosien 423, aanvanklik geklassifiseer as plantaricin 423, is fn klein Klas lIa, hittebestande en anti-listeriese bakteriosien met die YGNGV motief, wat verskeie Grampositiewe bakterieë inhibeer. Bakteriosien 423 het verskeie Gram-positiewe organismes geïnhibeer, onder andere Lactobacillus spp., Leuconostoc spp., Oenococcus oeni, Pediococcus spp., Enterococcus spp., Propionibacterium spp., Staphylococcus spp., Bacillus spp., Clostridium spp., en Listeria spp. Proteolitiese ensieme inaktiveer die bakteriosien. Die peptied was oor 'n pH reeks van 1-10 aktief. Outoklavering vir 15 min by 121°C het die aktiwiteit van die peptied halveer, maar die bakteriosien is nie geïnaktiveer met ander hittebehandelings nie. Produksie van die bakteriosien is verhoog deur die groeimedium te optimiseer. Die groeimedium het bestaan uit glukose, triptoon, gisekstrak, kaliumfosfaat, natriumasetaat, ammoniumsitraat, mangaansulfaat, Tween 80 en casaminosure. Die bakteriosien se genetiese determinante is op In plasmied gesetel. Genetiese analiese van die bakteriosien operon het 'n hoë homologie met In ander Klas lIa bakteriosien, pediocin PA-1, getoon, maar die strukturele gene van die twee bakteriosiene verskil merkbaar. Die strukturele geen van bakteriosien 423 is met PKR ge-amplifiseer en in 'n gistE. coli-vektor tussen die ADH1 promotor- en termineerderopeenvolgings, in leesraam met die MFa1 sekresiesein, gekloneer. Saccharomyces cerevisiae wat met hierdie plasmied getransformeer is, het bakteriosien 423 uitgedruk. Die aminosuurvolgorde van prebakteriosien 423 (MMKKIEKL TEKEMANIIGGKYYGNGVTCGKHSCSVNWGOAFSCSVSHLANFGHGKC) is verwant aan, maar nie identies aan, ander Klas lIa anti-listeriese peptiede.

Full textENGLISH ABSTRACT: Lactic acid bacteria play an essential role in the majority of fermented foods by producing organoleptic compounds and increasing the shelf life. The best-studied antimicrobial compounds are bacteriocins, i.e. ribosomally synthesized peptides. Most of these peptides have a narrow spectrum of activity and are usually only active against bacteria from the same ecological niche. The fact that all bacteriocins are degraded by proteolytic enzymes enlarges their potential use as natural food preservatives. The ideal would be to replace or reduce chemical preservatives such as sulfur dioxide, nitrates and nitrites. Bacteriocins are classified into four groups according to their structural and functional characteristics. Plantaricin 423, produced by Lactobacillus plantarum 423, is heat stable, plasmid encoded, relatively small (3.5 kDa) and is classified as a class Iia bacteriocin. The peptide is active from pH 1.0 to 10.0 and inhibits Gram-positive bacteria, including Lactobacillus spp., Leuconostoc spp., Oenococcus oeni, Pediococcus spp., Enterococcus spp., Propionibacterium spp. and pathogens such as Bacillus cereus, Clostridium spp. and Listeria monocytogenes. Production of bacteriocins may occur constitutively or may be regulated by a cell-density dependent system called quorum sensing. Plantaricin 423 is produced throughout logarithmic growth, with no apparent change in production levels when the producer strain is cultured in the presence of plantaricin 423 or Listeria innocua and Lactobacillus sakei. This led us to believe that plantaricin 423 may be produced constitutively. A reporter system was constructed which consisted of the plantaricin 423 promoter, P423, fused to the luxAB genes and cloned into a shuttle vector, pTRKH2. The newly constructed plasmid, pTAB4, was transformed to a bacteriocin-negative mutant of L. plantarum (423 B} Despite several repeats, no luciferase activity was recorded and no RNA homologous to the luxAB genes was detected. The region necessary for expression of plantaricin 423 may be located stream-up of the -80 region homologous to the -80 and -40 conserved repeats of regulated class II bacteriocins. Inclusion of the latter region in the reporter construct may result in the successful expression of luxAB.

Dit is moontlik dat die area nodig vir uitdrukking van plantarisien 423 verder stroom-op van die -80 area, homoloog aan die -80 en -40 gekonserveerde herhalings van reguleerbare klas II bakteriosiene, gesetel is. Insluiting van laasgenoemde area in die verklikker-konstruk mag lei tot die suksesvolle uitdrukking van luxAB.

O CERES-Rice é um modelo que simula o desenvolvimento e produtividade de uma cultivar, para realizar tal feito, é necessário obter os coeficientes genéticos da cultivar, através do processo de calibração. Os coeficientes genéticos descrevem o comportamento da planta no sistema solo-planta-atmosfera. Nesse contexto, o objetivo do presente trabalho foi calibrar e validar o modelo CERES-Rice, presente na plataforma DSSAT, em relação à estimativa de produtividade para a cultivar IRGA 424 no Rio Grande do Sul, utilizando apenas informações encontradas na literatura. O trabalho também visa o desenvolvimento de um algoritmo para auxiliar na formatação dos dados meteorológicos, a comparação do resultado obtido através modelo CERES-Rice com o estimado pelo SimulArroz e também avaliar a resposta obtida pelo modelo CERES-Rice ao estimar a data de florescimento. Após a aplicação do algoritmo, os dados meteorológicos para uso no DSSAT e SimulArroz foram exportados com sucesso. Apesar de não ter sido implementado nenhum recurso extra, o método utilizado proporciona a possibilidade de novas funções, como por exemplo, técnicas de preenchimento de dados. Para o processo de calibração dos coeficientes genéticos, foi utilizado o módulo GENCALC, também incluso no DSSAT. Após o processo de calibração, foi realizada a validação nos demais. Os resultados para produtividade foram considerados aceitáveis, uma vez que maior parte dos valores encontravam-se abaixo de 15%, apenas um dos resultados apresentou erro de 16,88%. Os valores simulados pelo modelo CERES-Rice apresentou um erro menor que o SimulArroz. Entretanto, a análise estatística aponta que ambos os resultados foram considerados bons. Apesar disso, o trabalho não aconselha a substituição do uso do modelo SimulArroz nas simulações do Rio Grande do Sul, mas propõe uma análise no modelo CERES-Rice, para averiguar a possibilidade de incorporar partes do seu código fonte no modelo SimulArroz, assim como também há o do ORYZA2000 e InfoCrop. O resultado da simulação obteve valores em dias após a semeadura inferiores ao dos valores experimentais, com uma média de 23 dias para os dados de Cachoeirinha e 25 para os de Uruguaiana. Esse resultado com os coeficientes genéticos utilizados no presente trabalho, o modelo não foi capaz de estimar a data de florescimento dentro dos limites aceitáveis, sendo necessário realizar um novo ajuste de coeficientes.
CERES-Rice is a model that simulates the development and productivity of a cultivar. To achieve this, it is necessary to obtain the genetic coefficients of the cultivar through the calibration process. The genetic coefficients describe the behavior of the plant in the soil-plant-atmosphere system. The gaol of the present work was to calibrate and validate the CERES-Rice model, included in the DSSAT platform, to estimate the productivity for the cultivar IRGA 424 in Rio Grande do Sul, using only information found in the literature. The work also aims to develop na algorithm to aid in the formatting of meteorological data, the comparison of the result obtained through the CERES-Rice model with that estimated by SimulArroz and also to evaluate the response obtained by the CERES-Rice model when estimating the date of flowering. After the application of the algorithm, the meteorological data for use in the DSSAT and SimulArroz were exported successfully. Although no extra features have been implemented, the method used provides the possibility of include new functions, such as data-filling techniques. For the calibration process of the genetic coefficients, the GENCALC module was used, also included in the DSSAT. After the calibration process, the other validations were performed. The results for productivity were considered acceptable, since most of the values were below 15%, only one of the results showed an error of 16,88%. The values found by the CERES-Rice model obtained a smaller error than the SimulArroz. However, the statistical analysis indicates that both results were considered good. Despite this, the paper does not recommend replacing the SimulArroz model in the simulations of Rio Grande do Sul, but proposes an analysis in the CERES-Rice model, to investigate the possibility of incorporating parts of its source code into the SimulArroz model, as well as there are from ORYZA2000 and InfoCrop. The results of the simulation obtained values in days after sowing inferior to those of the experimental values, with a mean of 23 days for the data of Cachoeirinha and 25 for those of Uruguaiana. This result with the genetic coefficients used in the present study, the model was not able to estimate the date of flowering within the acceptable limits, being necessary to perform a new adjustment of coefficients.
