Abstract:
Objective: Intraplaque neovascularization is an important feature of unstable human atherosclerotic plaques. However, its impact on plaque formation and stability is poorly studied. Because proliferating endothelial cells generate up to 85% of their ATP from glycolysis, we investigated whether pharmacological inhibition of glycolytic flux by the small-molecule 3PO (3-[3-pyridinyl]-1-[4-pyridinyl]-2-propen-1-one) could have beneficial effects on plaque formation and composition. Approach and Results: ApoE −/ − (apolipoprotein E deficient) mice treated with 3PO (50 µg/g, ip; 4×/wk, 4 weeks) showed a metabolic switch toward ketone body formation. Treatment of ApoE −/− Fbn1 C1039G+/− mice with 3PO (50 µg/g, ip) either after 4 (preventive, twice/wk, 10 weeks) or 16 weeks of Western diet (curative, 4×/wk, 4 weeks) inhibited intraplaque neovascularization by 50% and 38%, respectively. Plaque formation was significantly reduced in all 3PO-treated animals. This effect was independent of intraplaque neovascularization. In vitro experiments showed that 3PO favors an anti-inflammatory M2 macrophage subtype and suppresses an M1 proinflammatory phenotype. Moreover, 3PO induced autophagy, which in turn impaired NF-κB (nuclear factor-kappa B) signaling and inhibited TNF-α (tumor necrosis factor-alpha)–mediated VCAM-1 (vascular cell adhesion molecule-1) and ICAM-1 (intercellular adhesion molecule-1) upregulation. Consistently, a preventive 3PO regimen reduced endothelial VCAM-1 expression in vivo. Furthermore, 3PO improved cardiac function in ApoE −/− Fbn1 C1039G+/− mice after 10 weeks of treatment. Conclusions: Partial inhibition of glycolysis restrained intraplaque angiogenesis without affecting plaque composition. However, less plaques were formed, which was accompanied by downregulation of endothelial adhesion molecules—an event that depends on autophagy induction. Inhibition of coronary plaque formation by 3PO resulted in an overall improved cardiac function.

The transitions between 4p and 4d levels have been studied to examine the possibility of laser wavelength production in the soft X-ray region. The oscillator strengths of the transitions 3p4p (3S,3P,3D) → 3p4d (3Po,3Do,3Fo) have been calculated using CIV3 code. Using calculated oscillator strengths, the excitation rate coefficients, reduced populations, and gain coefficients for Si-like Ni XV, Zn XVII, Ga XVIII, Ge XIX, and As XX at electron temperatures 1/4, 1/2, and 3/4 the ionization potential have been calculated. The method by Palumbo and Elton has been used in the calculation of laser parameters. The present calculated data show promising values for the production of laser wavelengths in the soft X-ray region. The maximum values of the gain factor lie within the range α ∼ 109 cm−1 and wavelength λ ≤ 469 Å.

Abstract:
Abstract Metabolic reprogramming is one of the hallmarks of cancer, as these rapidly dividing cells need to adapt their metabolism to cope with an increased energy demand. We have previously showed that in mouse models of myeloproliferative neoplasms (MPN), JAK2-mutant cells display metabolic alterations, including increased oxidative phosphorylation and glycolysis. The glutamine-glutamate-alphaKetoglutarate (aKG) axis, besides fueling the Krebs cycle and anabolic processes, contributes to the synthesis of the heme precursor 5-aminolevulinic acid (5-ALA), thus making glutaminolysis a potential target for MPN therapy. Here, we examined the effects of pharmacological inhibition of the conversion of glutamine to glutamate by glutaminase 1 by Telaglenastat (CB-839) in a mouse model of JAK2-V617F driven MPN. We performed competitive transplantations of bone marrow co-expressing JAK2-V617F with a GFP-reporter and wildtype mice in a 1:10 mixture into lethally irradiated mice. At 8 weeks post-transplantation, the mice developed MPN phenotype and were randomized into 4 treatment arms. They were then treated for 6 weeks with vehicle, CB-839, 3PO (6-phosphofructo-2-kinase inhibitor) and the combination of CB-839 + 3PO. We found that hemoglobin levels were significantly reduced in CB-839 treated mice and were fully normalised in mice that received a combination of CB-839 + 3PO (Fig. 1A). Other peripheral blood cell counts did not change (not shown). Hypoglycemia was noted in the vehicle group, as previously described, but an increase in blood glucose levels was seen already after one week of treatment with CB-839. 3PO alone had less impact on hypoglycemia and mice receiving CB-839 + 3PO had similar values to CB-839 alone (Fig. 1A). In line with these findings, splenomegaly was also reduced (Fig. 1A). To characterise the changes in metabolic activity, we determined the metabolic profiles of unfractionated bone marrow (BM) cells using seahorse assays. As expected, basal and maximal glycolysis and oxidative phosphorylation were higher in vehicle treated MPN cells than cells from wildtype controls. However, when BM cells from CB-839 or CB-839 + 3PO groups were examined, they displayed a strong downmodulation of both glycolysis and oxidative phosphorylation, indicating that chronic exposure to CB-839 reprograms the metabolic machinery by reducing not only the basal glycolytic and respiratory levels, but also the maximal capacity (Fig. 1B). Glutamine is the primary source of aKG needed to produce the heme precursor 5-ALA, and hemoglobin synthesis consumes the majority of ATP in erythroid progenitors. We therefore hypothesize that 5-ALA shortage caused by CB-839 limits hemoglobin production, leading to reduced catabolic activity and drop in ATP demand. To evaluate this model, we examined the stages of erythroid differentiation from vehicle or treated mice. Early and intermediate erythroid cells were enriched at the expense of later stages in CB-839 and CB-839+3PO treatment arms (Fig. 1C). The accumulation of erythroid precursors at the stage before hemoglobin is assembled supports the hypothesis that targeting glutamine usage impairs excessive erythropoiesis by limiting hemoglobinization rate. Our data show that inhibiting glutaminolysis ameliorated MPN phenotype and that the combination with the glycolytic inhibitor 3PO was able to induce complete hematological response. Since CB-839 is already in clinical trials for other cancers, our data suggest that it could also be tested for treatment of patients with polycythemia vera. Figure 1 Figure 1. Abstract Background There is no widely accepted clinical tool to safely predict skull fracture in young children who sustained a head trauma. The C-3PO rule is a clinical decision rule which reported that age up to 3 months, or parietal/occipital scalp hematoma are predictors of skull fracture in young children. Objectives The present study prospectively evaluated the C-3PO rule in multiple new settings. Design/Methods This was a prospective multi-centre observational study conducted in three tertiary Emergency Departments (ED) in Canada and Switzerland. Patients were included if they were up to the age of 24 months of age and had a head trauma without high risk factors for clinically important intracranial injury according to the PECARN head trauma rule. The primary outcome was a radiologically identified skull fracture identified by any radiographic imaging modalities. The independent variables were the three components of the C-3PO rule as assessed by the treating physician. The patients were treated as per the physician’s discretion and followed up by phone if there was no initial skull radiological imaging. Criterion validity of the rule was calculated. Results A total of 844 patients were included with a median age of 8 months. Thirty percent of these patients (n=263) received radiological imaging, and 52 (6.2%) patients had a skull fracture. The C-3PO rule identified 51/52 children with a fracture for a sensitivity of 98% (95% CI: 90-100%), and a specificity of 70% (95% CI: 67-74%). Applying the rule would have reduced radiological imaging from 263 to 211 patients (-20%; 95%CI: 15-25%). Conclusion The C-3PO rule can safely be applied by pediatric emergency physicians evaluating children younger than 24 months with a head trauma. It helps ED clinicians to identify young children at low risk for skull fracture, improving decision making regarding the use of CT scan for children at intermediate risk of clinically important traumatic brain injury.

Intestinal barrier dysfunction is associated with the occurrence and development of sepsis. Further, aerobic glycolysis plays an essential role in inflammation and cell death. This study is aimed at investigating the protective effect and mechanism of PFKFB3 inhibition on intestinal barrier dysfunction in sepsis mice. Sepsis mouse models were established by cecal ligation and puncture (CLP) in wild-type mice and Gsdmd-/- mice. The results showed that the expression of 6-phosphofructo-2-kinase/fructose-2,6-biphosphatase 3 (PFKFB3) in the small intestines was significantly upregulated in sepsis. 3-(3-Pyridinyl)-1-(4-pyridinyl)-2-propen-1-one (3PO), the specific inhibitor of PFKFB3, and Gsdmd gene knockout significantly inhibited the inflammatory response and cell death caused by sepsis, thus alleviating intestinal damage and barrier dysfunction. 3PO was also shown to significantly inhibit oxidative stress and NLRP3/caspase-1/GSDMD-dependent cell pyroptosis in the small intestines. The in vitro studies revealed that 3PO reduced NLRP3/caspase-1/GSDMD-dependent cell pyroptosis by inhibiting ROS. Taken together, our results suggest that PFKFB3 is involved in inflammation, oxidative stress, and pyroptosis during sepsis and enhances intestinal damage, which may provide important clues about the potential targets to be exploited in this highly lethal disease.

Abstract:
The paper presents the results of the study of decaying neon plasma by the method of kinetic spectroscopy. Experimental conditions: neon pressure 0.2–152 Torr, electron density in the initial stage of decay [e] ≤ 5 × 1010 cm-3. The plasma was created by a pulsed barrier discharge with electrodes on the outer surface of a cylindrical glass tube. Discharge frequency 40–160 Hz. On the basis of a comparative analysis of the dependences of the intensities of spectral lines on the time and temperature of electrons in the afterglow, it was shown that in terms of the mechanisms of population, the levels of 2p54p are clearly divided into two groups. Lower levels: from 3p10 to 3p3 (Paschen notation) are associated with the dissociative recombination of molecular ions with electrons and, up to pressures of 0.6 Torr, the lines emitted by them behave identically to the lines of 2p53s ← 2p53p and 2p53p ← 2p53d transitions. The behavior of the upper levels (3p5, 3p2, 3p4, and 3p1) was more complex, and at low pressures, the populations of all 2p54p levels turned out to be related to the impact-radiation recombination of Ne + ions. Significant differences in the dependences of the relative populations of 3d and 4p levels on the neon pressure are discussed.

Via mental simulation, imagined events faithfully reproduce the neural and behavioral activities that accompany their actual occurrence. However, little is known about how fundamental characteristics of mental imagery—notably perspectives of self—shape neurocognitive processes. To address this issue, we used fMRI to explore the impact that vantage point exerts on the neural and behavioral correlates of imaginary sensory experiences (i.e., pain). Participants imagined painful scenarios from three distinct visual perspectives: first-person self (1PS), third-person self (3PS), and third-person other (3PO). Corroborating increased ratings of pain and embodiment, 1PS (cf. 3PS) simulations elicited greater activity in the right anterior insula, a brain area that supports interoceptive and emotional awareness. Additionally, 1PS simulations evoked greater activity in brain areas associated with visual imagery and the sense of body ownership. Interestingly, no differences were observed between 3PS and 3PO imagery. Taken together, these findings reveal the neural and behavioral correlates of visual perspective during mental simulation.

Abstract:
Vascular dysfunction is one of the primary factors in the onset and progression of atherosclerosis and other vascular-related diseases such as acute myocardial infarction (AMI) and chronic thromboembolic pulmonary hypertension (CTEPH). Emerging evidence indicates that pathological blood vessel responses and endothelial dysfunction are associated with metabolic alterations in endothelial cells (ECs). The identification of the insights and causes resulting in dysfunctional ECs is crucial for the understanding of the disease and to the development of new therapeutic tools. Therefore, to study the characteristics of such EC dysfunction in AMI and CTEPH diseases, we aim to determine the protein profile and/or the altered redox status of patient-derived EC lines by using mass spectrometry-based label-free quantitative proteomic and/or, spectrophotometric and fluorometric approaches. Moreover, quantitative proteomic approach was used to explore the underlying mechanisms of 3PO [(3-(3-Pyridinyl)-1-(4-pyridinyl)-2-propen-1-one), PFKFB3 inhibitor] at cellular level. 3PO is a compound able to inhibit the glycolytic flux partially and transiently and to reduce pathological angiogenesis in a variety of disease models. Bioinformatic and network analyses performed on pathologic HCAEC-AMI cells revealed the alteration of a) metabolism of RNA, b) platelet activation, signaling and aggregation, c) neutrophil degranulation, d) metabolism of amino acids and derivatives, e) cellular responses to stress and, f) response to elevated platelet cytosolic Ca2+ pathways. Similarly, network analysis in pathologic CTEPH-ECs, revealed the differentially regulation of pathways related to a) neutrophil and platelet degranulation, b) metabolism of lipids, amino acids and selenoamino acids, c) response to elevated cytosolic Ca2+, d) detoxification of reactive oxygen species. In addition, the main parameters, indicators of the redox status of both HCAEC-AMI and CTEPH-ECs were significantly increased: advanced oxidation protein products (AOPPs), protein carbonyls (PCO) content, and intracellular reactive oxygen species (ROS). Interestingly, the amount of GSH/GSSG (reduced glutathione/oxidized glutathione) and NADPH/NADP (reduced/oxidized form of nicotinamide adenine dinucleotide phosphate) ratios in dysfunctional ECs were reduced, a clear indication of oxidative stress involvement in both the pathological ECs. Finally, 3PO has multiple targets in the ECs, targeting mitochondrial inner membrane and it inhibits the important cellular pathways including the tricarboxylic acid cycle, the mitochondrial respiratory chain, and vasculogenesis, that may be useful for understanding the inhibitory effect of 3PO on EC proliferation and migration. Therefore, the present data suggest a potential application of this molecule as a starting point in designing novel molecules to prevent diseases where inflammatory reactions are involved, such as in atherosclerosis, cancer or neurodegenerative diseases.

Services of 3PL providers are nowadays very commonly used by production and merchant firms. It is happening due to companies' concentration on their own core-business and outsourcing of many of their activities to external suppliers. This thesis is focused on marketing activities of a 3PL provider, which is a specific part of the B2B market, as well as the logistics market. The thesis consists of three parts. First chapter is focused on theoretical definition of marketing and individual aspects, which marketing is concerned with. In the second part, the logistics, the specifies of 3PL logistics and the logistic market in the Czech Republic are presented. These theoretical findings are applied to a particular firm. Its marketing strategy is evaluated and a set of recommendations is proposed to enhance marketing position of the company.

In the theoretical part of the thesis there is a general description of outsourcing of logistics services, advantages and disadvantages of logistics outsourcing, history and current state of the logistic market and attitude of providers of logistics services towards sustainable development. Next part of the thesis describes phases of outsourcing process, types of contracts and tools that help control outsourcing relationship. In the practical part of the thesis in-depth interviews are used to analyze needs of 3PL client. Based on the information gathered from the in-depth interviews recommendations are created. These recommendations are supposed to raise client's satisfaction and evolve and improve cooperation. The aim of this Master's Thesis is to determine gaps between client's request and real state of provided services.

Based on analysis of a particular outsourcing warehousing contract held between global company and 3PL, this thesis has three goals: evaluate current contract compared to standard contractual requirements, evaluate the option of switching the outsourcing relationship from open book to closed book approach, and suggest solution that would use the potential of the provider most. Break event point analysis, labour productivity analysis and method of managerial decision making were used in this thesis. Closed book and open book approaches were analysed in original way using previously mentioned methods. The recommendation to the company is to keep current open book contract with implementing quality KPI's and performance-based outsourcing in accordance with vested outsourcing approach.

Ce travail s'inscrit comme une contribution a l'etude des terres rares dans les particules et les sediments marins. Deux domaines oceaniques ont ete etudies : la marge continentale et la dorsale medio-oceanique. L'etude de la marge continentale a ete realisee dans le but d'estimer le transfert des particules des continents vers le milieu oceanique profond. Des campagnes se sont deroulees dans le golfe de gascogne pour evaluer les flux de matieres traversant le canyon du cap ferret, principal site local de transfert. Les sediments du plateau, les particules sedimentant dans le canyon et les depots sedimentaires du talus continental presentent des spectres en terres rares identiques a ceux rencontres dans les shales. Un enrichissement en terres rares legeres est observe. Il se degage de ces informations que les materiels particulaires collectes sur le plateau continental, dans la colonne d'eau du canyon et sur la pente ont un caractere terrigene dominant et que leur composition en terres rares ne change pas lors de leur transfert des continents vers les oceans. L'etude du comportement des terres rares a proximite de la dorsale medio-atlantique a ete realisee dans le cadre de la campagne geofar. L'echantillonnage s'est deroule a proximite du volcan lucky strike (37\20n) et dans les plaines pelagiques environnantes. Ce travail permet de souligner l'impact de la dorsale sur la sedimentation locale. Les apports detritiques d'origine basaltique semblent peu importants et essentiellement localises a la dorsale elle-meme (volcan lucky strike). Les acores constitueraient une seconde source de produits volcaniques. Cette these porte sur l'ionisation penning des molecules co, n#2 et c#2h#2 par impact d'atomes d'helium et de neon dans des etats metastables. Dans la premiere partie, les modeles theoriques ainsi que les differents mecanismes d'ionisation coexistants lors du processus penning sont exposes. Le dispositif experimental utilise est ensuite etudie en detail. Il permet l'obtention des spectres en energie des electrons ejectes ainsi que leur distribution angulaire. Nous avons plus particulierement etudie les niveaux vibrationnels des ions crees. Lors de l'etude des interactions he#* (2#1#,#3s) - co, ne#* (#3p#0#,#2) - co, n#2 et c#2h#2, nous avons montre la superposition de l'ionisation penning directe (appelee voie covalente) et d'un processus d'ionisation indirecte. Nos observations mettent en evidence l'existence de transferts d'excitation vers des etats autoionisants. Ce travail montre que ces etats sont des etats de rydberg pour les interactions ne#* - co, ne#* - n#2 et he#* - co, et des resonances de forme pour les interactions he#* - co et ne#* - c#2h#2. Cette these prouve que l'etude conjointe des populations des niveaux vibrationnels et de la distribution angulaire des electrons correspondant permet la mise en evidence de l'intervention de transferts d'excitation lors de l'ionisation penning.

Atomic oxygen plays an important role in chemical and collisional processes in the earth's mesosphere and thermosphere. Current techniques, both in situ and remote, for measuring oxygen atom densities in this altitude regime have produced results with unexplained large disparities. We are investigating the feasibility of an approach wherein vertical profiles of translational temperature and oxygen atom density are recovered from measurements of the earth’s limb radiance profile near 147 micrometers and/or 63 micrometers wavelength, corresponding to the OI (3P0-3P1 and 3P1-3P2) transitions of the ground electronic state of atomic oxygen. The assumption that the 3P fine structure levels are in thermodynamic equilibrium (LTE) with the local translational temperature is crucial to the proposed technique, but seems a reasonable one based on the very long radiative lifetimes of the level transitions.

Atomic oxygen and translational temperature play vital roles in the chemistry and heat balance of the earth's atmosphere above 80 km. However, existing techniques, both in situ and remote, for measuring vertical profiles of the temperature and O-atom density at these altitudes have severe limitations; see, eg., Zachor and Sharma (1989). Described in the present paper is a new remote sensing technique in which the data is a pair of limb radiance profiles corresponding to the spectrally unresolved 147 μm and 63 μm lines of atomic oxygen (the inter-multiplet transitions 3P0 → 3P1 and 3P1 → 3P2 of the ground electronic state). We outline a retrieval procedure that uses both the onion peeling and global-fit methods to recover temperature and O-atom density between 90 km and 300 km altitude. The global-fit technique (Carlotti, 1988), which provides solution values simultaneously for several altitudes, is used only near 200 km altitude, where a singular Jacobian causes the onion peeling method to diverge. The retrieval procedure was applied to noise-contaminated synthetic data in order to establish basic system requirements for a spaceborne sensor. It is concluded that the proposed remote sensing technique can be implemented with a small cryogenic sensor system based on (non-scanning) Fabry-Perot etalons and detectors having current state-of-the-art performance.

We report an investigation of a nonlinear optical absorption phenomenon observed in LaCl3 crystals doped with Pr. When these crystals are illuminated with a laser beam of 529 nm wavelength and intensity in excess of a certain critical value, the light is strongly absorbed. The laser radiation is resonant with a Pr3+ transition from the first excited state (3H5) to a high optical state (3P1), and absorption is accompanied by strong fluorescence output from 3p1 and 3p0 to several lower-lying levels. Since the resonant transition does not include the ground state, it is not expected a priori that any absorption should occur (see Fig. 1). This is an example of the so-called photon avalanche phenomenon1.

For the first time by means of two-photon hybrid excitation in the Na2-Na system, which is distinguished from the usual two-photon excitation in atomic vapor, tunable UV coherent radiation (between 337.6 and 339.1 nm and between 358.0 and 359.6 nm) and tunable infrared stimulated radiation (3.38-3.46 µm) were generated. By optically pumping with a pulsed dye laser scanning from 614.5 to 617.4 nm for sodium vapor in a heat-pipe oven, Na2 was excited by the first photon absorption from the X ′ Σ g + to the A ′ Σ u + state. The energy was transferred to Na by Na2*-Na collision to populate the levels 3p1/2,3/2 in Na, which was followed by another photon excitation from the same beam up to near the 5S level, resulting in SERS (3P1/2-4P and 3P3/2-4P) and OPSE(5S → 4P and 4P → 4S) radiation. Assuming the detunings of pumping frequency ω1, from ω(5S,3P3/2) and ω(5s,3p1/2) to be Δ1 and Δ2, respectively, the three UV coherent radiation components were supposed to be generated by the following mixing processes: ωUV = 2ω1 − ω(5S − 4P); ω U V ′ = 2 ω 1 − [ ω ( 5 S → 4 P ) + Δ 1 ] ; ω U V ′ ′ = 2 ω 1 − [ ω ( 5 S → 4 P ) + Δ 2 ] , and also ω U V ′ ′ ′ = 2 ω 1 − ω ( 4 P → 4 S ) . The SERS and the UV signals were tuned as the pumping wavelength was tuned. The measured wavelengths agreed well with the calculated values.

Abstract:
The possibility of a neutral atom-based "ultimate" frequency standard in the optical domain has improved dramatically with the discovery of sub-Doppler cooling mechanisms. Unfortunately, the atoms with clock transitions of order 1 Hz that have been identified will be extremely difficult to laser cool. We have identified a two-photon transition in metastable xenon (3P2→3P0) at 2.19 microns with a calculated linewidth of 2 Hz. Metastable xenon can be laser-cooled with a cycling transition at 882 nm, convenient for Ti:sapphire lasers, with cooling parameters almost identical to cesium. Expected temperatures should be about 3 microkelvin, which will allow a fountain to be constructed that has little loss of signal due to spatial spreading of the atoms. After making the 2-photon clock transition, the atoms that are in the 3P0 metastable state can be detected with high efficiency with a metastable detector. Atoms in the lower metastable state can be completely removed by applying small amounts of diode laser light at 828 nm to drive atoms into the ground state, allowing high efficiency, essentially background-free detection. Two-photon transition rates and AC stark shifts have been calculated and will be presented. Possible laser sources for the clock transition will be discussed, as well as a connection from this optical frequency to the microwave.
