Abstract:
Redox imbalance plays an important role in the pathogenesis of CKD progression. Previously, we demonstrated that microRNA-382 (miR-382) contributed to TGF-β1-induced loss of epithelial polarity in human kidney epithelial cells, but its role in the development of renal tubulointerstitial fibrosis remains unknown. In this study, we found that with 7 days of unilateral ureteral obstruction (UUO) in mice, the abundance of miR-382 in the obstructed kidney was significantly increased. Meanwhile, the protein expression of heat shock protein 60 (HSPD1), a predicted target of miR-382, was reduced after 7 days of UUO. Expression of 3-nitrotyrosine (3-NT) was upregulated, but expression of thioredoxin (Trx) was downregulated. Anti-miR-382 treatment suppressed the upregulation of miR-382, attenuated renal interstitial fibrosis in the obstructed kidney, and reversed the downregulation of HSPD1/Trx and upregulation of 3-NT after UUO. Furthermore, in vitro study revealed that overexpression of HSPD1 significantly restored Trx expression and reversed TGF-β1-induced loss of E-cadherin, while in vivo study found that direct siRNA-mediated suppression of HSPD1 in the UUO kidney promoted oxidative stress despite miR-382 blockade. Our clinical data showed that upregulation of miR-382/3-NT and downregulation of HSPD1/Trx were also observed in IgA nephropathy patients with renal interstitial fibrosis. These data supported a novel mechanism in which miR-382 targets HSPD1 and contributes to the redox imbalance in the development of renal fibrosis.

Abstract:
Previously we have shown that microRNA miR-382 can facilitate loss of renal epithelial characteristics in cultured cells. This study examined the in vivo role of miR-382 in the development of renal interstitial fibrosis in a mouse model. Unilateral ureteral obstruction was used to induce renal interstitial fibrosis in mice. With 3 days of unilateral ureteral obstruction, expression of miR-382 in the obstructed kidney was increased severalfold compared with sham-operated controls. Intravenous delivery of locked nucleic acid-modified anti-miR-382 blocked the increase in miR-382 expression and significantly reduced inner medullary fibrosis. Expression of predicted miR-382 target kallikrein 5, a proteolytic enzyme capable of degrading several extracellular matrix proteins, was reduced with unilateral ureteral obstruction. Anti-miR-382 treatment prevented the reduction of kallikrein 5 in the inner medulla. Furthermore, the protective effect of the anti-miR-382 treatment against fibrosis was abolished by renal knockdown of kallikrein 5. Targeting of kallikrein 5 by miR-382 was confirmed by 3′-untranslated region luciferase assay. These data support a completely novel mechanism in which miR-382 targets kallikrein 5 and contributes to the development of renal inner medullary interstitial fibrosis. The study provided the first demonstration of an in vivo functional role of miR-382 in any species and any organ system.

Abstract:
Compost sources were used to determine long-term influence on common vegetable cropping systems (tomato, pepper, and cucumber). Three sources of Controlled Microbial Compost (CMC) (20 yd3/A) amended with fumigant Telone-C35 (35 gal/A) and Trichoderma-382 [2.5 oz/yd.3 (T-382)] were used during 3 consecutive years. Tomato showed statistic differences (1%) among compost treatments with higher total yields when CMC was combined with Telone-C35 (21%) and T-382 (8.2%). All treatments but Bio-Compost and control presented al least 25% more marketable yield per acre. No differences in fruit size were found for tomato, except for medium-size fruit when Telone C-35 was added. The CMC alone or combined with Telone C-35 and T-382 increased the total plant dry weight at least 18.6%. Pepper crop showed statistic differences with higher number of No. 1 fruit size when CMC was combined with Telone C-35 and T-382. Number of culls per acre decreased for all three compost sources, with no differences from the control. Cucumber yields differed among treatments for total and marketable yields and No.1 size fruit per acre. Best yields were achieved with CMC and when mixed with Telone C-35 and T-382. The lower numbers of culls per acre were found with Bio-Compost and Lexington sources and CMC+T-382. Total plant dry weight was increased in at least 24% when Bio-Compost or CMC compost were used alone or combined with Telone-C35 or T-382. CMC increased root knot nematode soil counts and percentage of root galling, but tended to improve root vigor in cucumbers. It seems that compost sources combined with Telone C-35 or T-382 could improve the cropping management as alternative to methyl bromide. Weed responses will also be discussed.

Abstract Background Non-small cell lung cancer (NSCLC) is a leading threat to human lives with high incidence and mortality. Circular RNAs were reported to play important roles in human cancers. The purpose of this study was to investigate the role of circ_0005962 and explore the underlying functional mechanisms. Methods The protein levels of Beclin 1, light chain3 (LC3-II/LC3-I), Pyruvate dehydrogenase kinase 4 (PDK4), Cleaved Caspase 3 (C-caspase 3), and proliferating cell nuclear antigen were examined using western blot analysis. Glycolysis was determined according to the levels of glucose consumption and lactate production. Xenograft model was constructed to investigate the role of circ_0005962 in vivo. Result circ_0005962 expressed with a high level in NSCLC tissues and cells. circ_0005962 knockdown inhibited proliferation, autophagy, and glycolysis but promoted apoptosis in NSCLC cells. miR-382-5p was targeted by circ_0005962, and its inhibition reversed the role of circ_0005962 knockdown. Besides, PDK4, a target of miR-382-5p, was regulated by circ_0005962 through miR-382-5p, and its overexpression abolished the effects of miR-382-5p reintroduction. circ_0005962 knockdown suppressed tumor growth in vivo. Conclusion circ_0005962 knockdown restrained cell proliferation, autophagy, and glycolysis but stimulated apoptosis through modulating the circ_0005962/miR-382-5p/PDK4 axis. Our study broadened the insights into understanding the mechanism of NSCLC progression.

Abstract:
У статті досліджено особливості покарання за невиконання судового рішення за кримінальнімй законодавством України. Виконання судових рішень – один із основних критеріїв, за яким громадяни оцінюють спроможність, ефективність і чесність системи правосуддя в будь-якій державі. Натомість відсутність дієвого механізму забезпечення виконання судових рішень негативно впливає на авторитет судової влади, оскільки відтак не досягається кінцева мета правосуддя – захист інтересів громадян і реальне поновлення їхніх порушених прав. У контексті предмету дослідження оарактеризовано окремі положення про загальне поняття покарання, зміст і форма покарання у вітчизняному кримінальному законодавстві. На цій теоретичній основі проаналізовано види покарання за невиконання судового рішення і підстави для їх застосування. Вивчено пом'якшуючі і обтяжуючі обставини і вплив ст. 69 КК України на призначення покарання за вчинення вказаного злочину. Наголошено на необхідності чіткого обґрунтування судами застосування судового розсуду при прийнятті рішення у кримінальному провадженні про невиконання судового рішення. Межі покарань за злочин, передбачений ст. 382 КК України, встановленні у санкціях цієї статті є необґрунтованими, оскільки вчинення злочину, передбаченого ч. 2 ст. 382 КК України, спеціальним суб’єктом є більш суспільно небезпечним, аніж учинення того ж посягання загальним суб’єктом. Якщо порівнювати санкції ч. 2 та ч. ч. 3, 4 ст. 382 КК України, то очевидно, що за менш суспільно небезпечне діяння судом може бути призначено суворіше покарання. Автор запропонував збільшити розмір покарання за вчинення злочину, передбаченого ч. 2 ст. 382 КК України, від однієї тисячі до двох тисяч неоподатковуваних мінімумів доходів громадян та збільшити мінімальний розмір основного покарання у санкції ч. 3 і 4 ст. 382 КК України до п’яти років позбавлення волі.

We report results showing that several gamma gene promoter elements participate in the developmental control of gamma-globin genes. Four gamma gene constructs with 5' truncated at -141, -201, -382, and -730 of the A gamma gene promoter linked to a micro locus control region (microLCR) cassette were used for production of transgenic mice and analysis of gamma gene expression during development. Mice carrying a microLCR -141 A gamma construct displayed downregulation of gamma gene expression in the adult stage of development, indicating that the proximal promoter contains elements participating in gamma gene silencing. Mice carrying a microLCR -201 A gamma or a microLCR -382 A gamma construct displayed high gamma gene expression in the fetal stage of development and complete loss of gamma gene downregulation in the adult stage, suggesting that the -141 to -201 gamma gene sequence contains elements which upregulate gamma gene expression and are dominant over the negative element 3' to -141. Extension of the promoter to -730 resulted in reappearance of gamma gene downregulation, suggesting that the -382 to -730 sequences contain an adult-stage-specific silencer. gamma gene expression in the microLCR -201 A gamma and the microLCR -382 A gamma transgenic mice was copy number dependent. All the microLCR -730 A gamma transgenic mice expressed gamma mRNA; however, gamma gene expression was copy number independent, indicating that levels of gamma gene expression were modulated by the surrounding chromatin. Our results suggest that multiple elements participate in gamma gene silencing. The findings in the microLCR-201 A gamma and microLCR -382 A gamma transgenic mice are interpreted to indicate that the LCR interacts not only with the minimal gamma gene promoter but also with sequences of the upstream promoter. We postulate that gamma gene downregulation is achieved when the interaction between LCR and the upstream promoter is disturbed by the silencer located in the -382 to -730 region. We propose that gamma gene silencing is achieved by the combined effect of negative elements located 3' to -141, the negative element located between -382 and -730, and the competition by the beta gene promoter during the adult stage of development.

Abstract:
Owing to a lack of appropriate therapeutic regimens, prostate cancer (PC) is a global health concern with a high incidence and mortality rate in elderly men. Combination treatment seems to have the highest clinical benefit and avoids unwanted side effects. The current study focused on the chemotherapeutic efficacy of Zoledronic acid (ZA) in combination with 5-fluorouracil (5-FU) or Flutamide on prostate cancer cells, as well as its effect on apoptosis. The MTT assay was used to determine the cytotoxic effect of Zoledronic acid (ZA), 5-FU, and flutamide on PC-3 and DU-145 cells, as well as the combined therapy of ZA with 5-FU or flutamide. Additionally, immunofluorescence staining analysis was used to assess changes in Bcl-2 and p53 expression. Furthermore, the western blotting method was extensively used to evaluate Bax, caspase 3, and cyclin D1. Furthermore, quantitative real-time polymerase chain reaction (qRT-PCR) was applied to determine the relative expression of miRNA-382 (miR-382) and miRNA-18a (miR-18a). Instead of (13.47, 8.23, and 9.42 μM) for PC-3 or (38.77, 17.6, and 8.47 μM) for DU145 cells, the combination therapy improved cytotoxicity with doses approximately half of IC50 (6.74, 4.12, and 7.07 μM) in PC-3 and (19.38, 8.8, and 6.33 μM) in DU145 cells for ZA, 5-FU, and flutamide, respectively. When compared to a single therapy, the combination therapy significantly up-regulated the pro-apoptotic Bax, cleaved caspase 3 and p53 levels while down-regulated the cyclin D1 and Bcl-2 expression. In addition, the combination therapy was linked to changes in miR-382 and miR-18a expression. Our findings suggest that combining ZA with 5-FU or flutamide improves chemotherapeutic efficacy against prostate cancer cells, at least in part by encouraging apoptosis and modulating miRNA expression, especially miR-382 and miR-18a.

Abstract:

The 3:12-provisions are part of a legal system specified for owners of close corporations (companies owned and managed by a small group of businesspeople), for taxation of capi-tal gains and dividend. The reason for special rules for the taxation of these owners is to prevent them from transforming their income, to only be subject of the lower taxation of capital gains, instead of income of service. Since the rules were put in force, they have been subject to several changes. The most recent changes took effect on the 1 January 2006. They involve a higher importance for the rule of salary-based taxation. The rule of salary-based taxation means that compensation paid out to employees may increase the part of a company owner’s income which is taxed at the lower taxation of capital gains. The salary-base is calculated including compensation paid out in cash to employees based within the area of the European Economic Community.

When applying the rule several problems in the interpretation arise. Questions concerning whether the part of per diem and allowance for expenses, which are subject to taxation, should be considered compensation paid out in cash, as well as whether board members are to be considered as employees. In addition, it is not certain how to deal with salaries in subsidiaries, which are trading companies, and salaries paid to employees outside of the European Economic Community.


3:12-reglerna utgör en del av ett skattesystem för beskattning av kapitalvinst och utdelning för fåmansföretagsdelägare. Anledningen till att dessa skall beskattas enligt särskilda regler är att förhindra inkomstomvandling, så att exempelvis tjänsteinkomster inte beskattas som kapitalinkomster, och därmed med en lägre skattesats.

Sedan regelverkets tillkomst har flera förändringar skett. De senaste förändringarna trädde i kraft den 1 januari 2006, och innebär bl.a. att löneunderlaget får en ökad betydelse vid be-skattning av fåmansföretagsdelägare. Löneunderlagsregeln innebär att ersättningar utbetal-da till arbetstagare får öka den delen av fåmansföretagsdelägarens inkomster som beskattas som inkomst av kapital, och därför till en lägre skattesats än inkomst av tjänst.

I löneunderlaget ingår kontanta ersättningar utbetalda till arbetstagare inom EES-området. Denna bestämmelse innebär flera tolkningssvårigheter vid dess tillämpning. Frågor uppstår om den skattepliktiga delen av kostnadsersättningar och traktamenten skall räknas som en kontant utbetald ersättning, samt om styrelseledamöter räknas som arbetstagare. Dessutom är rättsläget osäkert vad gäller löner utbetalade i dotterbolag som är handelsbolag, och löner utbetalade till arbetstagare utanför EES-området. Vad gäller den senaste är det troligt att löneunderlagsregeln står i strid med EG-rätten.

Det finns inte något klart och tydligt svar att finna för dessa tolkningsproblem varken i lag-text, förarbete, praxis eller doktrin. Rättsläget vid tillämpningen av löneunderlagsregeln är därför osäkert. Eftersom bestämmelsen utgör en sådan viktig del vid beskattning av få-mansföretagare är det anmärkningsvärt att lagstiftaren inte lade mer vikt vid att utforma re-geln på ett annat sätt.

Abstract:
RIASSUNTO BREVE Il differenziamento epidermico prevede la migrazione dei cheratinociti dallo strato basale dell’epidermide a quelli sopra basali fino al raggiungimento dello strato più superficiale dove formano l’involucro corneo. Durante questo processo i futuri corneociti subiscono una serie di cambiamenti nella loro struttura e tali modifiche sono mediate dagli enzimi transglutaminasi. Le transglutaminasi (TGasi) rappresentano una famiglia di enzimi funzionalmente e strutturalmente correlati che catalizzano la reazione di trasferimento acilico Ca2+-dipendente tra il gruppo γ- carbossiamidico di residui di glutammina e il gruppo amminico di varie ammine primarie. In questo modo le transglutaminasi catalizzano la formazione di legami crociati (cross-linking) fra proteine che compongono l’involucro corneo. Quattro sono le TGasi espresse a livello epidermico (TGasi 1, TGasi 2, TGasi 3 e TGasi 5) e tutte, esclusa la TGasi 1, mostrano anche un’attività GTPasica. Nello strato basale dell’epidermide svolge invece una funzione molto importante il gene p63. Quest’ultimo è un membro della famiglia di p53, la cui trascrizione da due differenti promotori origina due diverse proteine:TAp63 che contiene un dominio di transattivazione all’N-terminale e ∆Np63che non lo possiede. Tale dominio è importante per l’attivazione dei geni bersaglio che presentano un motivo di legame per p63 di conseguenza TAp63 è considerato l’attivatore trascrizionale dei geni indotti durante il differenziamento, mentre l’espressione di ∆Np63 potrebbe essere correlata con il mantenimento dell’attività proliferante delle cellule staminali epidermiche localizzate nella membrana basale. Ognuna di queste due isoforme va inoltre incontro a splicing alternativo al C-terminale originando tre differenti varianti di splicing:α, β, γ. Topi p63-/- presentano severe anomalie nella pelle e negli arti. Lo scopo di questo progetto di ricerca è sia quello di fornire nuove conoscenze sul ruolo svolto dalla TGasi 3 durante il differenziamento epidermico che quello di individuare nuovi geni la cui attività risulta essere regolata da p63. Per quanto riguarda la TGasi 3 abbiamo deciso di realizzare topi knock-out condizionali attraverso l’utilizzo della ricombinasi Cre e dei siti loxP. Tale sistema consente infatti l’eliminazione tessuto-specifica dell’espressione del gene bersaglio. Durante gli esperimenti preliminari per la creazione di tali topi abbiamo inoltre scoperto due nuove varianti di splicing per il gene TGM3. La prima, denominata ∆6∆7, isolata nel topo manca della cisteina catalitica e sia l’attività di cross-linking che quella GTPasica risultano essere compromesse. La variante ∆9∆10, isolata nell’uomo, pur conservando intatta la triade catalitica presenta una bassa attività transamidasica, mentre l’attività GTPasica risulta essere persa. Nella seconda parte di questo lavoro di tesi abbiamo invece identificato un nuovo gene bersaglio denominato Scotin, la cui espressione risulta essere regolata positivamente in vitro dall’isoforma TA, ma non da ∆N. L’espressione di Scotin è localizzata nel reticolo endoplasmatico e nell’involucro nucleare. Questo gene è inoltre in grado di indurre apoptosi attraverso l’induzione di stress del reticolo endoplasmatico. Esperimenti condotti su colture epiteliali primarie (murine ed umane) hanno evidenziato come l’espressione di Scotin durante il differenziamento sia accompagnata, come atteso, dall’incremento dell’isoforma TA e dalla dimuzione di ∆N. In conclusione i nostri dati hanno portato all’isolamento di Scotin come nuovo gene bersaglio indotto da p63 durante il differenziamento epidermico. Tale meccanismo attiva la risposta allo “stress” da parte del reticolo endoplasmatico,di cui Scotin è un induttore, correlando così per la prima volta queste due differenti vie.
SHORT ABSTRACT Epidermal differentiation begins with the migration of keratinocytes from the basal layer and ends with the formation of the cornified layer. Cell proliferation, differentiation and death occur sequentially and each process is characterized by the expression of specific proteins. Transglutaminases (TGases) play an important role during the differentiation program. These enzymes catalyse post-translational modifications of proteins by the formation of isopeptide bonds (a cross-linking reaction). The cross-linked products, often of high molecular mass, are highly resistant to mechanical challenge and proteolytic degradation. In mammals, nine distinct TGase isoenzymes have been identified at the genomic level. Four TGases are expressed in the epidermis (TGase 1, TGase 2, TGase 3 and TGase 5) and the last three TGases show also a GTPase activity. TGM1-/- knock-out mice show severe epidermal abnormalities and they dye soon after birth, while TGM2 -/- knock-out mice dont’show any epidermal abnormality. During the complex mechanism that leads to epidermis formation an important role is played by the action of p63. This transcription factor, belonging to the p53 gene family, is transcribed from two different promoters, generating two classes of proteins: one containing an N-terminal transactivation domain (TA) and another that lacks this domain (∆N). The first isoform is considered the transcriptional inducer whereas ∆N could exert a role of dominant negative. In addition alternative splicing at the 3’ end of both transcripts (TA and ∆N) generates three different C-terminal splicing variants: α, β and γ. Mice p63-/- show severe limb, cranofacial and skin defects and die soon after birth. The aim of this work is to acquire new knowledges in the complex mechanism that leads to epidermal differentiation thanks to the investigation of TGase 3 and p63 function during the skin process formation. Regarding TGase 3 we plan to create conditional knock-out mice for this ezyme using the Cre/loxP system. During our work we isolated also two novel splicing variants for the TGM3 gene. The first one, isolated in mouse, lacks exons six and seven (∆6∆7), while the second one, isolated in NHEK cells, lacks of exons nine and ten (∆9∆10). The experiment demonstrated that the ∆9∆10 is at least 4 times less active than the wild type one, while ∆6∆7 is inactive because it lacks of the catalytic cysteine Regarding to the GTPase, it is lost in both splicing variants because several residues, that are important for the creation of the GTP pocket, are missed. In the second part of this work, we isolated a new p63-target gene: Scotin. This gene was previously isolated as a p53-inducible proapoptotic gene and the protein is located in the endoplasmic reticulum and in the nuclear membrane. Scotin expression is induced in response to endoplasmic reticulum (ER) stress in a p53 dependent or independent manner and this event lead to apoptosis. RT-qPCR showed that Scotin transcript is positively regulated by TAp63α in vitro, while ∆N isoform inhibits its expression. Western blot experiments, performed on differentiating mouse and human primary keratinocytes, revealed Scotin upregulation following the differentiation program. Immunofluorescence performed on mouse new born skin sections demontrated that Scotin expression is located in the suprabasal layer of epidermis where TAp63, but not ∆Np63 is expressed. In conclusion our data lead to isolation of a new p63 target gene induced during the epithelial differentiation program, a complex process that also involves endoplasmic reticulum stress induction.

As before, his account of Christ’s saving work relies upon chains of biblical imagery and metaphors, but in the Antirrheticus against Apollinarius and other works such as Epistle 3, the point of transformation is placed at the nativity rather than merely after the passion. The model of mixture accordingly shifts away from one of humanity’s absorption into divinity. A new account is offered for the situation lying behind Epistle 3, and the connection of that work to both the Council of Constantinople in 382 and the dispute over Cyril of Jerusalem’s legitimacy.

Flowserve and its heritage companies have supplied valves for many of the critical applications within commercial nuclear power generation plants since the beginning of commercial nuclear power generation. Two of these highly critical applications are the Main Steam Isolation Valves (MSIVs) and the Main Feedwater Isolation Valves (MFIVs). As the requirements of these two applications have evolved, so have the applicable valve and actuator designs and their qualifications. Although functional qualification standards (ASME QME-1) for power operated valve applications have been developed within the US nuclear industry, their use is relatively new within the US. In order to globally supply these valves, Flowserve has functionally qualified its MSIVs and MFIVs to this standard. In addition to the valves, the actuators have also evolved. Flowserve’s type A, gas / hydraulic actuator has been updated to improve its reliability and provide better performance for the power plants. The updated Flowserve type A, type A, gas / hydraulic actuator has completed the Environmental Qualification testing in accordance with the requirements of IEEE 323, 344 and 382. The Flowserve MSIV and MFIV designs have been selected for installation in many of the new Generation 3 and Generation 3+ nuclear power generation plants. This paper briefly discusses: • The history of the MSIV and MFIV valve applications within the nuclear power generation plants and • The Flowserve MSIV and MFIV, ASME QME-1 functional qualifications. Ghosting is a student behaviour characterised by enrolling in a subject but never participating. Hence, a ghost student who remains enrolled receives a zero-fail grade. From 2022, the Job ready Graduates Package will require that only genuine students have access to Commonwealth assistance at Australian Universities and an institution may need to refund the fees of what are referred to as non-genuine students. In 2019 and 2020, 382 first-year subjects were monitored to identify disengaged students in weeks 3 and 4 of the session using learning analytics and nonsubmission of an early assessment item. Disengaged students were contacted via phone and 2-way SMS and offered timely and targeted support pre census. The total number of domestic undergraduate students receiving a zero fail has decreased during this time. To further reduce the number of ghost students, once identified as disengaged, an engagement should be mandatory to remain in the subject.

Introdução: O diabetes mellitus (DM) destaca-se, atualmente, como importante causa de morbidade e mortalidade. Estimativas globais indicam que 382 milhões de pessoas vivem com DM (8,3%), e esse número poderá chegar a 592 milhões em 2035(1). Com destaque para o diabetes mellitus tipo II (DM2), doença multifatorial associada a fatores genéticos e ambientais(2). Caracterizado pela resistência à insulina e hiperglicemia, o DM2 é alvo de diversos estudos para compreender os principais fatores genéticos envolvidos em seu desenvolvimento(3). Dentre eles, os polimorfismos em certos genes estão relacionados a uma maior susceptibilidade ao desenvolvimento da doença, com destaque para alterações no gene da subunidade beta 3 da proteína G humana (GNB3), uma molécula sinalizadora envolvida na regulação dos níveis de glicose pela sinalização da via metabólica da insulina. Já foi comprovado que polimorfismos nesse gene estariam associados ao desenvolvimento da doença e de suas complicações(4). Objetivos: Este resumo tem como objetivo relacionar o impacto do polimorfismo no gene GNB3 contribuindo para a DM2. Métodos: Foi realizada pesquisa nas bases públicas de dados de artigos científicos: PubMed e Google Acadêmico usando os descritores ‘’type 2 diabetes mellitus’’ e ‘’type 2 diabetes mellitus and genetics’’ durante o período de 26 de agosto à 4 de setembro. Resultados: O marcador rs5443 localizado no gene GNB3 foi associado a uma série de condições metabólicas, incluindo a obesidade, doença arterial coronariana, resistência à insulina e DM2(3). A alteração no gene GNB3 devido a alguma mutação ou polimorfismo pode levar a defeitos na proteína G codificada por este gene. Essas alterações polimórficas estão associadas ao desenvolvimento de DM2, bem como às complicações secundárias associadas(4). Conclusão: O polimorfismo GNB3 rs5443 pode conferir efeitos no DM2 tais como algumas condições metabólicas associadas como a obesidade, hipertensão e arteriosclerose. Desta forma sua detecção nos indivíduos indicam maior risco para o desenvolvimento da doença.

Air pollution monitoring and management is one of the key challenges for urban sectors, especially in developing countries. Measuring pollution levels requires significant investment in reliable and durable instrumentation and subsequent maintenance. On the other hand, there have been many attempts by researchers to develop image-based pollution measurement models which have shown significant results and established the feasibility of the idea. But, taking image-level models to a city-level system presents new challenges, which include scarcity of high-quality annotated data and a high amount of label noise. In this paper, we present a low-cost, end-to-end system for learning pollution maps using images captured through a mobile phone. We demonstrate our system for parts of New Delhi and Ghaziabad. We use transfer learning to overcome the problem of data scarcity. We investigate the effects of label noise in detail and introduce the metric of in-interval accuracy to evaluate our models in presence of noise. We use distributed averaging to learn pollution maps and mitigate the effects of noise to some extent. We also develop haze-based interpretable models which have comparable performance to mainstream models. With only 382 images from Delhi and Ghaziabad and single-scene dataset from Beijing and Shanghai, we are able to achieve a mean absolute error of 44 ug/m^3 in PM2.5 concentration on a test set of 267 images and an in-interval accuracy of 67% on predictions. Going further, we learn pollution maps with a mean absolute error as low as 35 ug/m^3 and in-interval accuracy as high as 74% significantly mitigating the image models' error. We also show that the noise in pollution labels emerging from unreliable sensing instrumentation forms a significant barrier to the realization of an ideal air pollution monitoring system. Моногенный сахарный диабет (МСД) – это гетерогенная группа нарушений углеводного обмена, в ос- нове которых лежит дефект генов, ассоциированных с секрецией и/или действием инсулина, закладкой и развитием поджелудочной железы. Частота МСД достоверно не известна, по данным различных иссле- дований в детском возрасте его частота составляет 1–6%. Наиболее распространенными формами МСД являются MODY и неонатальный сахарный диабет (НСД). МАТЕРИАЛЫ И МЕТОДЫ: проанализированы данные реестра пациентов с МСД, подтвержденным в ходе молекулярно-генетического исследования (МГИ), за период с 2012 по 2022 годы. Пациенты были обследованы с проведением МГИ в ФГБУ «НМИЦ эндокринологии» Минздрава России. Возраст на момент обследования составил от 0 до 18 лет. РЕЗУЛЬТАТЫ: за период с 2012 по 2022 года обследовано 417 детей с МСД (386 пробандов, 29 сибсов), из которых 382 (91,7%) пациентов с MODY, 17 (4,1%) – с НСД, 9 (2,2%) – с DIDMOAD, 4 (0,9%) – с липоатрофи- ческим сахарным диабетом, 3 (0,7%) – с синдромом инсулинорезистентности типа А, 1 (0,2%) – с синдромом Рабсона-Менденхолла и 1 (0,2%) – с синдромом Альстрема. Наиболее часто у пациентов с фенотипом MODY наблюдался GCK-MODY и HNF1A-MODY, мутации в гене GCK были выявлены у 74% пациентов (n=282), в гене HNF1A выявлены у 12% пациентов (n=45). Более ред- кими причинами MODY были мутации в гене ABCC8, которые выявлены в 1,8% (n=7), в гене HNF4β – в 1,6% (n=6), в гене HNF4A – в 1% (n=4), в гене RFX6 – в 1% (n=4), в гене INSR – в 0,9% (n=3). В единичных случаях были идентифицированы мутации в генах KLF11, NEUROD1, PAX4, PDX1, PTF1A, WFS1 – в 0,5% (n=3) каждая, в генах BLK, CEL, GLIS3, INS – в 0,2% случаев каждая. В 3,5% (n=13) диагностирован MODY с дигенным на- следованием, в 0,4% (n=2) с олигогенным наследованием. Наиболее частой причиной НСД были мутации в гене KCNJ11, которые выявлены у 35,5% пациен- тов (n=6), у 17,8% (n=3) идентифицирована мутация в гене INS, у 11,7% (n=2) – мутация в гене ABCC8, у 1 (n=5,8%) – в гене GATA6. В 11,7% (n=2) НСД был ассоциирован с аномалиями хромосомы 6q24. У одной пациентки НСД диагностирован в составе синдрома Уолкотта-Раллисона, подтвержденного при МГИ (вы- явлена гомозиготная мутация в гене EIF2AK3). У 2 пациентов генетическая причина НСД не установлена. ВЫВОДЫ: под данным реестра МСД в детском возрасте наиболее частой формой МСД является MODY, которая встречается в 91,7% случаях всех форм МСД. Гетерозиготные мутации в гене GCK являются наи- более распространенной причиной MODY у детей (74%). Второй по распространенности подтип у детей – HNF1A- MODY, который выявлен в 12% всех случаях MODY. Частота НСД среди МСД в детском возрасте составила 4,1%, наиболее распространенной причиной НСД являются мутации в гене KCNJ11. Abstract:
The reduction of CO2 concentration in our atmosphere consists in a big challenge for researchers, who are trying to explore novel technologies in order to transform CO2 into high added-value products. CO2 conversion into methane using microwave plasma (MWP) manifests as a very promising solution due to the ease of transport of methane and its storage. Microwave plasma represents a source of high-energy electrons, active ions and radicals that enhance or enable chemical reaction. It can be supplied by electricity generated from renewable resources. Then, MWP does not require any electrode to be generated and thus, the cost of those electrodes and of maintenance is reduced compared to glow discharge or DBD plasmas. MWP also can be generated over wide range of pressure (between 10 mbar-1bar). In addition, in the case of MWP, more electrons and active species are produced in comparison with other type of plasma[1–4]. MWP is a very suitable medium for this chemical reaction and leads to an efficient dissociation of CO2. The catalytic reduction of CO2 with H2 using MWP has been investigated in this work and the synergetic effects between the plasma and several catalysts were studied. First, the reaction was carried out without any catalysts and the effect of CO2/H2 ratio, total flow rate and input energy were evaluated. Then, a microwave generated plasma process was coupled with several Nickel catalysts that we prepared and characterized [5] in order to lead the reaction into methane formation. Multiple configurations were studied by changing the position of the catalyst bed. Obtained results were compared with conventional catalytic tests made with the same catalysts. It was found that the conversion of CO2 and energy efficiency increased using plasma assisted catalytic methanation of CO2 in comparison with conventional process. Abstract:
Inom ramen för IBTS genomförs trålundersökningar i Skagerrak och Kattegatt och på senare år även vissa områden i östra Nordsjön i kvartal 1 och 3. Under expeditionen utfördes totalt 49 giltiga tråldrag i det undersökta området, 26 i Skagerrak och 19 i Kattegatt samt fyra i Nordsjön. I totalfångsten ingick 61 olika fiskarter, bland annat 10,2 ton sill, 4,1 ton skarpsill, 3,8 ton kolja och 382 kg torsk, totalt 36,6 ton. Biologiska parametrar på individnivå samlas in på sill- och torskfiskar samt rödspätta och tunga. Totalt provtogs 3 883 individer från 11 olika arter med avseende på ålder och könsmognad. Både sill- och skarpsillfångsterna var större i år än föregående år, så också rekryteringen. Torskens 0-grupp i både Skagerrak och Kattegatt ökade kraftigt. Däremot är 2+-guppen av torsk (torsk över ca 37 cm) i stort sett frånvarande. Kolja visar en mycket stor 0-grupp i Skagerrak och 0-gruppen vitling i Kattegatt är avsevärt större än föregående år.
