Abstract:
The purpose of this research is to increase the iodine content of processed foods such as ice cream with the addition of seaweed variations Euchema cotonii. Seaweed is one of the food sources that contain high fiber and iodine so good for health. Variables research done by adding seaweed to the dough ice cream with the amount of 25%, 50% and 75%. The test results showed the water content, protein, fat concentration increases with seaweed. Test results proximat ie water content ranged 34.55% -37.15%, protein 1.6% -4% range, fat 20.25% - 26.45 % range, ranged from 20.6 to 22.6% carbohydrate. The addition of seaweed showed elevated levels of iodine which is quite high due to the amount of seaweed is added. Test results iodine levels without seaweed ice cream of 2.07 ppm to 19.91 ppm with the addition of seaweed as much as 75%. Test hedonic/A indicates aroma, color, texture and taste almost preferred by the panelists. ABSTRAKTujuan penelitian ini adalah melakukan peningkatan kandungan yodium pada olahan pangan berupa es krim dengan variasi penambahan rumput laut jenis Euchema Cotonii. Rumput laut merupakan salah satu sumber bahan pangan yang mengandung serat dan yodium yang tinggi sehingga baik untuk kesehatan. Variabel penelitian dilakukan dengan penambahan rumput laut ke dalam adonan es krim dengan jumlah 25%, 50% dan 75%. Hasil uji menunjukan kadar air, protein, lemak meningkat seiring dengan bertambahnya konsentrasi rumput laut. Hasil uji proximat yaitu kadar air berkisar 34,55%-37,15%, protein berkisar 1,6%-4%, lemak berkisar 20,25%-26,45%, karbohidrat berkisar 20,6-22,6%. Penambahan rumput laut menunjukan peningkatan kadar yodium yang cukup tinggi seiring dengan jumlah rumput laut yang ditambahkan. Hasil uji kadar yodium es krim tanpa rumput laut sebesar 2,07 ppm menjadi 19,91 ppm dengan penambahan rumput laut sebanyak 75%. Uji hedonik/kesukaan menunjukan aroma, warna, tekstur dan rasa hampir disuka oleh panelis. Kata kunci : es krim, rumput laut Euchema Cottonii, yodium

Abstract:
Abstract Venetoclax (VEN), an inhibitor of anti-apoptotic protein BCL2 is an important therapeutic intervention in acute myeloid leukemia (AML). However, development of resistance to VEN and its combinations is common, which poses a significant clinical challenge to the management of AML. Heterogeneity in resistance can exist not only across different tumors but also within the same tumor. While some mechanisms of resistance have been revealed, others remain elusive. Here, we derive a VEN resistance signature from AML cell-lines in CCLE, and utilize non-negative matrix factorization to decompose the signature and systematically define four transcriptionally distinct clusters of VEN resistant AML patient using the BeatAML cohort, consisting of 186 patients. By integrating gene expression and drug response profiles from these patients, we elucidate distinct expression programs underlying VEN resistance in these clusters and their associated vulnerabilities. Samples in Cluster 1 are characterized by high frequency of DNMT3A and RAS (KRAS and NRAS) mutations, strong monocytic phenotype and high PI3K-mTOR signaling, oxidative phosphorylation (OXPHOS) and fatty acid metabolism, indicating improved metabolic fitness. These samples also show remarkable sensitivity to panobinostat, an HDAC inhibitor, which suppresses mTORC1 signaling and OXPHOS. Cluster 3, in contrast, is characterized by TP53 and SRSF2 mutations, high CTL infiltration and activation of JAK-STAT and MAPK signaling, which regulate pro-survival pathways and proliferation. They also show selective sensitivity to JAK family inhibitors. Cluster 3-like AML cell-lines also show remarkable sensitivity to the JAK inhibitor, ruxolitinib. Cluster 2 samples exhibit high rates of NRAS mutations and suppression of HOX genes. Finally, Cluster 4 samples are transcriptionally similar to VEN sensitive patients, but show higher interferon signaling and higher rates of DNMT3A and IDH1 mutations. We validated these clusters using the TCGA AML (n=173) and a second BeatAML cohort (n=182).We further confirmed the existence and therapeutic value of these clusters by projecting them onto single-cell RNA-seq data obtained from a patient before and after VEN+azacitidine treatment. In pre-treatment cells, both VEN sensitive (41%) and resistance (59%) signatures were present in the patient tumor, however, post-treatment we observed that Cluster 1 (5%) and Cluster 2 (93%) like resistant cells accounted for most of the tumor cells. While Cluster 1 like cells were characterized by activation of metabolic pathways, the more proliferative Cluster 2 like cells dominated the tumor after relapse. Our study systematically identified and characterized mechanistic heterogeneity in venetoclax resistant AMLs and can aid therapeutic decision making and development of effective combination therapies with venetoclax. La relevancia que han cobrado las entidades y actividades financieras no bancarias en los últimos años ha centrado la atención de supervisores y reguladores a efectos de preservar la estabilidad financiera. Se ha iniciado un debate a escala internacional sobre la conveniencia de desarrollar la política macroprudencial en el ámbito de los fondos de inversión. Los riesgos más importantes para la estabilidad financiera que pueden emanar de estas instituciones se originan, principalmente, por los desajustes entre las condiciones de liquidez de su cartera y las de sus reembolsos o por un apalancamiento elevado. Las herramientas macroprudenciales disponibles en este ámbito son múltiples y de diferente naturaleza, y existe una gran disparidad entre las jurisdicciones. Actualmente están en marcha relevantes iniciativas internacionales, impulsadas por la Organización Internacional de Comisiones de Valores y el Consejo de Estabilidad Financiera, destinadas a promover y homogeneizar el conjunto de herramientas disponibles y su aplicación. En este artículo se describen las herramientas existentes en España, detallando el objetivo que persiguen y la posibilidad de acción por parte del regulador. Asimismo, se compara la situación de España con la de otras jurisdicciones europeas. La comparativa revela la amplia disponibilidad en España tanto de información como de herramientas para la gestión de los riesgos.

Abstract:
Streptomyces lavendulae IMRU 3455 contains two large linear plasmids designated pSLV45 (45 kb) and pSLV195 (195 kb). A cosmid, pSPRX604, containing 42 kb from pSLV45 was cloned and sequenced. pSLV45 was tagged with a hygromycin-resistance marker by homologous recombination to generate the derivatives pSLV45.680 and pSLV45.681. An apramycin-resistance marker was introduced into S. lavendulae IMRU 467 using the pSPR910 integration vector to yield the recipient strain SPW910. The self-transmissible nature of pSLV45 was determined by transfer of pSLV45.680 and pSLV45.681 from the donor strains SPW680 and SPW681 into the recipient strain SPW910. Southern analysis indicated the presence of hygromycin- and pSLV45-hybridizing sequences within SPW910 exconjugants. PFGE analysis confirmed pSLV45.680 and pSLV45.681 were transferred intact and formed freely replicating linear plasmids. Sequence analysis of pSPRX604 revealed genes predicted to be involved in plasmid transfer, partitioning and regulation. The transfer of the linear plasmid pSLV45 from S. lavendulae IMRU 3455 into S. lavendulae IMRU 467 may allow the development of pSLV45 as an actinomycete-to-actinomycete conjugative shuttle vector.

Abstract:
217 Background: Recent data indicate that biomarker driven use of targeted therapy and I/O-therapy among patients with GI cancer is associated with improved outcome. The presence of biomarkers varies broadly between different GI tumor types, highlighting the importance of comprehensive molecular profiling. To analyze the presence of various alterations in GI cancer samples of a large database, comparing congruency between various tumor types. Methods: A retrospective data analysis of 34855 GI cancer patients profiled at CARIS Life Sciences obtained from Jan 1, 2010 till Sep 14, 2018 was performed. GI tumors were classified as CRC, esophageal/gastric/GIST, small intestine, pancreatic/hepatobiliary/liver. Technologies used to analyze the biomarkers: IHC for PD-L1, MMR and Her2, and DNA-NGS for EGFR, BRAF, KRAS, NRAS, MET-CNV, TMB, MSI, POLE and BRCA1/2. Results: Median age was 61 range (18-89 years). 51.5% was CRC (n=18047), 15.5 % was esophageal/Gastric/GIST (n=5470), 3% was small intestinal cancer (n=886) and 30% was pancreatic/hepatobiliary/liver cancer (n=10452). Information on biomarkers was available from 2931 cases for MET amplification to 28536 for RAS mutation. Overall, the most common finding was a pathogenic RAS-mutation in 7650 cases (26.81%), the rarest one was a mutation in EGFR in 12 cases (0.06%). Higher rate of HER2 amplification was observed among pts with esophageal/gastric/GIST tumors (6.5%) in comparison to tumors at other GI sites ( ~1.5%). High TMB was seen among patient with CRC and small intestine tumors (~7%) while it was lowest among pancreatic cancer (1.8%). Compared to other GI sites lower MSI /MMR deficiency rates were observed in pancreatic /hepatobiliary tumors, significantly higher PD-L1 positivity was observed in gastrooesophageal cancer types, increased MET-amplifications in gastrooesophageal and small intestinal cancer types and lower RAS-mutation rate in gastrooesophageal cancer. Conclusions: Molecular profiling analyzing druggable biomarkers can help identify patients with increased likelihood for benefit from immune-checkpoint-inhibitors and targeted therapies. Further investigations are needed to evaluate the different findings in various GI cancer types.

ABSTRAKKabupaten Muaro Jambi merupakan kabupaten di Provinsi Jambi dengan luas karhutla terbesar dengan luas 4.054 hektar (34,55%). Hal ini dikarenakan sebagian besar jenis tanah di Kabupaten Muaro Jambi merupakan jenis tanah gambut. Pada penelitian bertujuan untuk melihat hubungan antara parameter cuaca dan distribusi titik panas di Kabupaten Muaro Jambi. Parameter cuaca yang akan dianalisis yaitu curah hujan pada tahun 2019 dan 2020 dimana tahun 2019 mewakili tahun terjadinya El Nino dan tahun 2020 merupakan tahun tanpa El Nino. Data yang digunakan adalah antara lain data curah hujan tahun 2019 dan tahun 2020, data ENSO (El Nino-Shouthern Oscillation) dan IOD (lndian Ocean Dipole), dan data titik panas. Pada tahun 2019, curah hujan terendah terjadi pada bulan september sebesar 39,3 mm/bulan dan pada tahun 2020, terendah pada bulan Juli sebesar 154,5 mm/bulan. Jumlah titik panas pada bulan September tahun 2019 sebanyak 3000 titik atau sekitar 77,88% dari total titik panas pada tahun 2019. Sedangkan pada tahun 2020, jumlah titik panas terbanyak hanya sebanyak 13 titik panas di bulan April. Hal ini menunjukkan bahwa peristiwa El Nino yang menyebabkan rendahnya curah hujan sangat berpengaruh terhadap pertambahan jumlah titik panas sebagai potensi terjadinya kebakaran hutan dan lahan. Kata kunci: kebakaran hutan dan lahan; el nino; curah hujan; titik panas. ABSTRACTMuaro Jambi Regency is a district in Jambi Province with the largest area of forest and land fires with an area of 4,054 hectares (34.55%). Most of the soil types in Muaro Jambi Regency are peat soil types. This study aims to look at the relationship between weather parameters and the distribution of hotspots in Muaro Jambi Regency. The weather parameters to be analyzed are rainfall in 2019 and 2020 where 2019 represents the year El Nino occurs and 2020 is the year without El Nino. The data used includes rainfall data for 2019 and 2020, ENSO (El Nino-Southern Oscillation) and IOD (Indian Ocean Dipole) data, and hotspot data. In 2019, the lowest rainfall occurred in September of 39.3 mm/month and in 2020, the lowest was in July of 154.5 mm/month. The number of hotspots in September 2019 was 3,000 points or around 77.88% of the total hotspots in 2019. Meanwhile, in 2020, the highest number of hotspots was only 13 hotspots in April. This shows that the El Nino event which causes low rainfall greatly influences the increase in the number of hotspots as a potential for forest and land fires. Abstract:
ABSTRACT Health workers as the frontline facing the covid-19 outbreak professionally treat patients so that they have concerns about the health of individuals and their families that have the potential to cause mental health problems such as fear, anxiety, stigma, marginalization of people with diseases and stress due to job demands. This study aims to determine the impact of COVID-19 on the mental health of health workers at UNUD hospital in 2021. This research is cross sectional research using Google Form link questionnaire. The sample is health workers who work and registered in UNUD hospital using convenience sampling technique with data collected as many as 110 people. The results showed that 92.73% of health workers did not experience depression, 7.27% mild depression, 65.45% did not experience anxiety, 34.55% mild anxiety, 76.36% did not experience stress and 23.64% mild stress. Factors of profession, having family members with chronic diseases, availability of information on the use of PPE, family support, incentives related to the impact of mental health (p<0.05). Factors not related to the impact of mental health are gender, education, marital status, living with children, living with the elderly, installation, length of Service, Compensation, hours and rest facilities (p>0.05). It is necessary to pay special attention by holding free counseling in order to still create conducive and healthy working conditions. Keyword : Health Workers, Mental Health, Depression, Anxiety, Stress ABSTRAK Tenaga kesehatan sebagai garda terdepan menghadapi wabah COVID-19 secara profesional merawat pasien sehingga mempunyai kekhawatiran mengenai kesehatan individu dan keluarganya yang berpotensi menimbulkan masalah kesehatan mental seperti ketakutan, kecemasan, stigma, marginalisasi terhadap penderita penyakit dan stress akibat tuntutan pekerjaan. Penelitian ini bertujuan untuk mengetahui dampak COVID-19 pada kesehatan mental tenaga kesehatan di RS Unud tahun 2021. Penelitian ini adalah penelitian cross sectional menggunakan kuesioner link google form. Sampel adalah tenaga kesehatan yang bekerja dan terdaftar di RS Unud menggunakan teknik convenience sampling dengan data terkumpul sebanyak 110 orang. Hasil penelitian menunjukkan 92,73% tenaga kesehatan tidak mengalami depresi, 7,27% depresi ringan, 65,45% tidak mengalami kecemasan, 34,55% kecemasan ringan, 76,36% tidak mengalami stress dan 23,64% stress ringan. Faktor profesi, memiliki anggota keluarga dengan penyakit kronis, ketersediaan informasi tentang penggunaan APD, dukungan keluarga, insentif berhubungan dengan dampak kesehatan mental (p<0,05). Faktor-faktor yang tidak berhubungan dengan dampak kesehatan mental yaitu jenis kelamin, pendidikan, status pernikahan, tinggal dengan anak, tinggal dengan lansia, instalasi, masa kerja, kompensasi, jam dan fasilitas istirahat (p>0,05). Perlu memberikan perhatian khusus dengan mengadakan konseling gratis supaya tetap menciptakan kondisi kerja yang kondusif dan sehat. Abstract:
For more than two decades, knowledge management (KM) has been examined in the literature and considered a basic factor in business management. The purpose of this article is to explore the ability of knowledge management to achieve small and medium-sized enterprise (SME) innovation and business performance. Data analysis procedures of PLS-SEM was used to analyze the data based on responses from the owners of 903 companies in industry, construction, services, and trade in the Region of Murcia (Spain) through a self-directed survey. The results show that KM has a significant influence on innovation but the influence on the level of performance of SMEs is insignificant. However, training employees as part of KM showed no significant influence on innovation in SMEs. It is important for leaders and managers to have greater openness to changes in the implementation and proper execution of KM. This work contributes primarily to the development of the literature on knowledge management and its relationship with innovation and business performance.

The protection of children at work was one of the first topics addressed by labor legislation in the world. However, the regulation of child labor remains one of the insufficiently regulated areas in labor law. Within that, one of the newer topics is light work of children. This form of child labor has existed since time immemorial, especially in the family and household, so it is considered useful for acquiring work habits and experience. However, it can be both dangerous and harmful. In this paper, we present the light work topic, with an emphasis on the existing regulations in the world and the possibilities for a best solutions for regulation of this form of work.

Secara mikro, alih fungsi lahan mengakibatkan petani yang semula mengusahakan tanaman pangan dan dapat memenuhi sendiri ketersediaan pangan (beras) bagi rumah tangganya menjadi tidak memiliki beras dan harus membeli. Dampak lain dari alih fungsi lahan adalah hilangnya mata pencahariannya sebagai petani, hilangnya kesempatan kerja pada usaha tani. Dengan hilangnya mata pencaharian sebagai petani maka menurunnya pendapatan dan daya beli serta berdampak pada menurunnya aksesibilitas ekonomi rumah tangga tani terhadap pangan. Megaproyek pembangunan Bandara NYIA di Provinsi D.I. Yogyakarta mengharuskan pembebasan lahan baik dari lahan masyarakat maupun dari Pakualaman. Lokasi pembebasan lahan ini ada di lima desa Kecamatan Temon Kabupaten Kulon Progo. Penelitian ini bertujuan menganalisis dampak alih fungsi lahan terhadap tingkat ketahanan pangan rumah tangga tani di Kecamatan Temon Kabupaten Kulon Progo. Data yang digunakan data primer. Data primer yang dimaksut adalah data cross sction pada waktu tertentu. Pengambilan data menggunakan kuesioner. Alat analisis yang digunakan uji beda dua rata-rata dengan paired sample t-test. Hasil penelitian ini adalah tidak terdapat perbedaan ketahanan pangan sebelum dan sesudah alih fungsi lahan di Kecamatan Temon Kabupaten Kulon Progo. Artinya alih fungsi lahan tidak memberikan pengaruh terhadap ketahanan pangan rumah tangga tani di Kecamatan Temon Kabupaten Kulon Progo namun jika dilihat dari olah data statistika deskriptif terlihat ada penambahan jumla rumah tangga tani yang rawan pangan setelah adanya alih fungsi lahan.
