Abstract:
The lung cancer was most deadly tumor in the world and the suvival rate needs to be improved clinically and urgently. The abnormal miR-340 expression is found in several solid tumors. Our study was aimed to explore miR-340’s role in lung cancer. 14 cases of patients with lung cancer was selected to measure miR-340 level by RT-PCR and analyze its correlation with clinical characteristics. The relation between the miR-340 and DICER1 was detected by dual luciferase assay and cell proliferation was measured by MTT assay along with analysis of cell migration and invasive by Scratch-Wound experiment. The miR-340 in lung cancer cells was reduced significantly and DICER1 was predicted to be a potential target of miR-340. DICER1 level was found to be negatively related with miR-340 level. The DICER1 as the direct target gene of miR-340 was conducive to improve miR-340 function through overexpression and knock-out experiment further. Abnormal miR-340 level affected lung cancer cell proliferation and migration as well as MAPK signaling. miR-340 could affect the biological morphology and transformation of physiological function of lung cancer cells mainly through restraining the expression of apoptosis and prompting the cellular proliferation, indicating that it might be a novel target to improve the treatment of lung cancer.

Abstract:
e14587 Background: PSMA, a transmembrane protein highly expressed on the surface of prostate cancer cells, is a validated target in prostate cancer; however, achieving a favorable therapeutic index has been challenging for PSMA-directed T-cell engagers (TCE’s). AMG 340, a PSMA-targeted TCE with a novel low-affinity CD3 binding domain specifically engineered to mitigate cytokine release syndrome (CRS) toxicity and maintain efficacy was investigated in a phase 1, open-label, dose-escalation study (NCT04740034). Methods: Patients (pts) with mCRPC who had received ≥2 prior lines of therapy were treated with AMG 340 monotherapy (1.5–800 mg intravenously every 3 weeks [Q3W]) during dose escalation. The primary outcome measures were dose-limiting toxicities, treatment-emergent adverse events (TEAEs), and pharmacokinetic profile. Secondary outcomes included prostate antigen specific (PSA) response and objective response per response evaluation criteria in solid tumors (RECIST). Results: As of September 6, 2023, a total of 42 pts (mean ± SD age, 68.5±7.8 years) received AMG 340 step up dosing across 11 cohorts. At doses up to 800 mg Q3W, AMG 340 had a manageable side effect profile. Any grade TEAEs reported in ≥30% of pts included CRS (52%), nausea (52%), vomiting (52%), fatigue (48%), diarrhea (38%), increased aspartate aminotransferase (36%), increased alanine aminotransferase (33%), and hypocalcemia (31%). Treatment-related toxicities included CRS (52%; Grade 1, 26%; Grade 2, 24%; Grade ≥3, 2%) and thrombocytopenia (24%; Grade 2, 10%; Grade ≥3, 14%). Step dosing was implemented at doses >40 mg and successfully mitigated CRS incidence and severity after the first dose. The maximal tolerated dose was not reached, as increasing the dose beyond 800 mg was predicted to minimally increase the potential AMG 340 activity. Exposure increased in an approximately dose-proportional manner with no evidence of clinically significant anti-drug antibodies. AMG 340 demonstrated evidence for pharmacodynamic engagement as shown by cytokine induction. Of the 41 PSA evaluable pts, PSA responses were seen across dose levels (PSA50, n=4), but were not dose dependent. Eight of 42 pts remained on treatment after 6 months. Among 27 RECIST evaluable pts, no RECIST responses were observed (n=14 stable disease, n=0 partial or complete response). Conclusions: Despite reaching high doses, no dose response was observed, and AMG 340 did not generate sufficient clinical activity to warrant further exploration. However, AMG 340 demonstrated manageable CRS toxicity showing that decreased CD3-binding strength mitigates CRS, the main toxicity of TCEs. Clinical studies are ongoing evaluating the PSMA protein as a molecular target for prostate cancer. Clinical trial information: NCT04740034 .

Field trials were conducted at the Irrigation Research Station, Kadawa to evaluate the response of sesame varieties to stress period and NPK fertilization in Sudan savannah. The treatments were two sesame varieties (NCRIBEN 01 and NCRIBEN 03) three water stress period (at seedling, flowering and podding stages), three NPK application rates (200, 270 and 340 kg NPKha-1). The experimental design used was randomized complete block design (RCBD) laid out in a split plot arrangement where the factorial combination of fertilizer rates and water stress periods were assigned to the main plot and varieties allocated to the sub plot and replicated three times. Results showed that variety NCRIBEN 03 recorded more capsules and higher capsule weight per plant and a grain yield of 1111.4 kg ha-1 and 1168.8kgha-1, in 2012/13 and 2013/14 dry seasons respectively. Stressing sesame at podding lead to 17 and 29% decline in number of pods and pod weight per plant, respectively and31% drop in grain yield. Application of 340 kg NPK fertilizer ha1 had highest yield of 1018 and 1078 kg ha-1 in 2012/13 and 2013/14 seasons, respectively. The use of variety NCRIBEN 03 + 340 kg NPK fertilizer ha1+ stressing sesame at seedling stage will give best grain yield of sesame.

Foram avaliados o rendimento de grãos, a estabilidade e adaptabilidade ambiental, a tolerância ao alumínio tóxico e a qualidade tecnológica da farinha de vinte genótipos de trigo em três regiões tritícolas do Estado de São Paulo (Zonas A e B, sequeiro; Zona H, com irrigação por aspersão), no biênio 1994-95. Os resultados mostraram que: (a) os genótipos tiveram comportamento regional diferenciado quanto ao rendimento de grãos; (b) os genótipos IAC 289, IAC 340 e BR 18 mostraram-se, em média os mais produtivos, não diferindo do 'IAC 304', 'OCEPAR 14', 'IAC 350', 'Panda', 'IAC 60' e 'IAC 348'; (c) os genótipos apresentaram, em geral, boa estabilidade produtiva nas diferentes condições ambientais; (d) os genótipos IAC 289, IAC 340 e IAC 304 foram responsivos à melhoria ambiental e 'IAC 227', adaptado a ambientes desfavoráveis; (e) os genótipos IAC 24, IAC 120, IAC 227, OCEPAR 14, IAC 348, Panda, BR 18, IAC 319, IAC 346, IAPAR 60, IAC 332 e IAC 347 foram tolerantes à toxicidade de Al3+; (f) a farinha dos diferentes genótipos apresentou baixa atividade amilásica; (g) os genótipos IAC 340, IAC 227, IAC 319 e IAC 60 propiciaram farinha com baixos valores de W (energia de deformação da massa) e baixa estabilidade, características próprias de farinha de glúten fraco. Objetivou-se com este estudo avaliar economicamente a terminação em confinamento de novilhos Red Angus superjovens alimentados até atingirem os pesos de abate pretendidos de 340, 370, 400 e 430 kg (pesos reais obtidos: 340, 373, 401 e 434 kg). O número de dias necessários para atingir os respectivos pesos foi de 114, 144, 168 e 209 dias aos 12, 13, 14 e 15 meses de idade. Os animais foram alimentados com dieta contendo 13,26% de PB e relação volumoso (silagem de milho):concentrado de 56,21:43,79 na MS. Na formação e condução da lavoura para produção de volumoso, as operações mais representativas do custo total/ha foram, em ordem decrescente, adubação de plantio (21,21%), adubação de cobertura (12,86%), sementes (11,40%) e controle de plantas daninhas (5,78%) e de pragas (4,52%). A alimentação (volumoso mais concentrado) foi o componente de custo mais representativo (média de 77,2%) se desconsiderado o custo de compra do animal. Da alimentação, o concentrado foi o componente mais oneroso (58,3%). O custo/kg de ganho de peso incrementou linearmente com o aumento no peso de abate, sendo de 1,94; 2,06; 2,15 e 2,34 R$/kg, respectivamente, para os pesos de abate de 340, 373, 401 e 434 kg. O incremento no peso de abate de 340 para 434 kg reduziu a lucratividade da terminação em confinamento de novilhos Red Angus superjovens.

Abstract:
The utilization of rubber wood waste in the form of liquid smoke has the potential to overcome environmental problems caused by the industry. Therefore, this study aimed to determine the potential of liquid smoke from rubberwood clone PB 340 to be used as a coagulant and preservative. Rubber wood waste was processed into liquid smoke using the pyrolysis method with a temperature of 400°C. Determination of the composition of liquid smoke was carried out using gas chromatography-mass spectrometry (GC-MS) analysis. The results showed that the rubber wood clone PB 340 contained 57.78% cellulose, 12.16% hemicellulose, and 19.01% lignin. Furthermore, volatile analysis with GC-MS showed that liquid smoke from rubber wood clone PB 340 contained 58 organic compounds. Some compounds in liquid smoke were phenols, furans, furfurals, acetic acid, and cyclopentene. The product was then tested for its performance as a latex coagulant at several concentrations of 5%, 10%, 15%, 20%, and 25% v/v, as well as storage time of 1, 7, and 14 days. The treated latex samples that had turned into coagulums were examined for their sheet quality parameters, including initial plasticity (P0), plasticity retention index (PRI), total volatile compounds, impurity, and ash content. The outcomes from all comparisons of pure liquid smoke concentrations and storage time of up to 14 days of the coagulum samples showed that in the initial plasticity value (P0), the plasticity retention index (PRI), volatile matter, ash content, and dirt content had met the applied Standard Indonesian Rubber (SIR).

Background. This study aimed to establish the expression profile of circulating microRNAs (miRNAs) during nerve allotransplantation in the presence and absence of FK506 immunosuppression.Methods. A 1 cm BALB/c donor sciatic nerve graft was transplanted into the sciatic nerve gaps created in recipient C57BL/6 mice with or without daily FK506 immunosuppression [1 mg/(kg·d)]. At 3, 7, and 14 d after nerve allotransplantation, serum samples were collected for miRNA expression analysis by Illumina small RNA deep sequencing.Results. Sequence analysis showed that the dominant size of circulating small RNAs after nerve allotransplantation was 22 nucleotides, followed by 23-nucleotide sequences. Nine upregulated circulating miRNAs (let-7e-5p, miR-101a-3p, miR-151-5p, miR-181a-5p, miR-204-5p, miR-340-5p, miR-381-3p, miR-411-5p, miR-9-5p, and miR-219-2-3p) were identified at 3 d, but none was identified at 7 or 14 d. Among them, miR-9-5p had the highest fold-change of >50-fold, followed by miR-340-5p with 38.8-fold. The presence of these nine miRNAs was not significant at 7 and 14 d after nerve allotransplantation with or without immunosuppression, showing that these miRNAs are not ideal biomarkers for monitoring rejection of deep-buried nerve allografts, a response usually observed later.Conclusions. We identified nine upregulated circulating miRNAs, which may have a biological function, particularly during the early stages after nerve allotransplantation under FK506 immunosuppression.

Two series of very large discotic complexes of tetrakis (2,3,6,7-tetraalkoxy)triphenylenocyaninato copper(II) (abbreviated as ( C n O )16 TcCu ; n = 8, 10, 12 and 14) and tetrakis(2,3,6,7-tetraalkoxy)-1,4-diazatriphenylenocyaninato copper(II) (abbreviated as ( C n O )16 TzCu ; n = 8, 10, 12 and 14), were synthesized to investigate their mesomorphic properties by differential scanning calorimetry, thermogravimetry analysis, polarizing optical microscope observation and temperature-dependent X-ray studies. The results show that each of the ( C n O )16 TcCu derivatives exhibits two or three disordered rectangular columnar ( Col rd ) mesophases from room temperature to the decomposition temperature at ca. 330-340°C under nitrogen gas. However, each of the ( C n O )16 TzCu complexes exhibits only one ordered tetragonal columnar ( Col tet.o ) mesophase in an extremely wide temperature region from room temperature to the decomposition temperature at ca 300°C and ca. 340-350°C under the atmosphere and nitrogen gas, respectively. It is very interesting that the Col tet.o mesophase of ( C n O )16 TzCu for n = 10, 12 and 14 shows remarkable spontaneous homeotropic alignment in a large area, without disclinations and polydomains, in the temperature range from room temperature to the decomposition temperature. As far as we know, these discotic liquid crystals are the first examples to show homeotropic alignment at room temperature. Furthermore, the difference between the mesomorphic properties of ( C n O )16 TcCu and ( C n O )16 TzCu originated only from the additional eight nitrogen atoms in the central core in ( C n O )16 TzCu .

The metal complex between Bi3+ and 8-hydroxyquinoline-based azo dye (Bi3+ -8-HDQ) was prepared and characterized by UV-Visible, IR, SEM and powder XRD analysis. A spectroscopic peak of complex formation appears at 340 nm. The azo dye of 8-hydroxy-quinoline is a typical indicator to determine bismuth ions (III) using the complexometric method. The antimicrobial activity of the complex results in a high zone of inhibition in the range of 12 mm to 14 mm.

Out of 340 genotypes/varieties screened against black rot disease in rapeseed-Indian mustard under field conditions, 15 genotypes found resistant, viz. RTM 1624, RTM 1626, TM EPAU, DRMRAB-7-116, NPJ-177, LES-55, DRMRIJ 16-1, RH 1514, BASANTI, NDRS 1-2, TMR 14-1, TMR- 14-4, TMR 14-5, TMR 14-3, T 27, can be utilized as a donor for the development of black rot disease resistant varieties in crucifer crops. All genotypes of Eruca sativa were found resistant against black rot disease. Under in vitro conditions, streptocycline @200 ppm had maximum inhibition zone 3.8 cm diameter with 73.48% inhibition over control. In vivo, minimum disease severity 13.29% and 14.29%, lowest AUDPC 185.8% and 201.4% and highest yield 16.45 q/ha and 16.62 q/ha was recorded in the treatment T1, Streptocycline @200 ppm on rapeseed-mustard var. NDR 8501 (Narendra Rai) in 2017–18 and 2018–19 respectively.

Based on data on the composition of azeotropic solutions for the acetic acid - acrylic acid system, the parameters of the semi-empirical Wilson model for temperatures 320, 330, 340, 350 K were calculated using the mathematical software package Mathcad 14 by iterations. The obtained parameters made it possible to describe the vapor-liquid equilibrium in the acetic acid-acrylic acid system: to calculate the composition of the equilibrium phases, the activity coefficients of the solution components, the separation coefficient and to construct p-x isothermal boil diagrams.

Abstract:
Background: To find out the prevalence of multiorgan dysfunction in cases of severe falciparum malaria in children, correlating the organ dysfunction in different age group and relationship with case fatality rate (CFR).Methods: This prospective cross-sectional study was conducted from Jan 2014 to June 2016 in the Department of Pediatrics, Maharaja Krushna Chandra Gajapati medical college and hospital (MKCG MCH), Berhampur, Odisha, India. Total 340 cases belonging to <14 yrs having severe malaria (as per the WHO criteria) were included in the study. Mixed malaria and Plasmodium falciparum (Pf) positive cases having other significant disease e.g. chronic hemolytic anaemia, cardiac, renal or other chronic diseases were excluded. The study subjects were thoroughly screened for presence of organ dysfunction clinically and by appropriate laboratory tests and analysed.Results: Out of 340 subjects, maximum no of cases (56.76%) were found in >5yr age group. Hyperparasitemia (>10%) was detected in 12.35% cases. Cerebral malaria was most common organ dysfunction found in 1-5 years, followed by anaemia in 5-10 years, acute renal failure (ARF) in 10-14 years age group. Multiorgan dysfunction (MOD) was detected in 32.94% cases of severe malaria. Overall CFR was 8.82%, being directly proportional to no. of organ involvement, 31.81% with 4 organs and 100 % with five organs involvement respectively which was statistically significant.Conclusions: MOD is of grave outcome in severe Pf malaria increasing the case fatality rate proportionate to number of organs involved.

Abstract:
Limited data are available regarding the patient-based outcomes of SDF therapy in the kindergarten setting. This study aims to evaluate the dental fear and anxiety (DFA) of preschool children after participating in a school-based outreach service using SDF to arrest early childhood caries (ECC). The study recruited 3- to 5-year-old children with untreated ECC. A trained dentist performed a dental examination and applied SDF therapy to the carious lesions. ECC experience was measured using the dmft index. Questionnaires for parents were used to collect the children’s demographic information and their dental treatment experiences. The children’s DFA before and immediately after SDF therapy was assessed using the self-reported Facial Image Scale (FIS) on a Likert scale of 1 (very happy) to 5 (very distressed). The association between the children’s DFA after SDF therapy and the potentially related factors, including demographic background, caries experience, and DFA before SDF therapy, were analysed using bivariate analysis. Three hundred and forty children (187 boys, 55%) joined this study. Their mean (SD) age and dmft scores were 4.8 (0.9) and 4.6 (3.6), respectively. Most of them (269/340, 79%) never had a dental visit. After SDF therapy, 86% (294/340) of the children exhibited no or low DFA (FIS ≤ 3), whereas 14% (46/340) reported high DFA (FIS > 3). No factor was associated with children’s DFA after SDF therapy (p > 0.05). This study showed most preschool children with ECC exhibited no or low DFA after SDF therapy in a school setting.

Abstract We present images at 6 and 14 GHz of Source I (SrcI) in the Kleinmann–Low Nebula in Orion. At higher frequencies, from 43 to 340 GHz, images of this source are dominated by thermal emission from dust in a 100 au diameter circumstellar disk, but at 6 and 14 GHz the emission is elongated along the minor axis of the disk, aligned with the SiO bipolar outflow from the central object. Gaussian fits to the 6, 14, 43, and 99 GHz images find a component along the disk minor axis whose flux and length vary with frequency consistent with free–free emission from an ionized outflow. The data favor a broad outflow from a disk wind, rather than a narrow ionized jet. SrcI was undetected in higher-resolution 5 GHz e-MERLIN observations obtained in 2021. The 5–6 GHz structure of SrcI may be resolved out by the high sidelobe structure of the e-MERLIN synthesized beam, or be time variable.

We present deep 1.6 and 2.2 μm images of the central parsec of the Galaxy at a resolution of 0.15″. Most of the flux in earlier seeing limited images comes from about 340 unresolved stellar sources with K≤14. The IRS 16 and 13 complexes are resolved into about two dozen and half a dozen sources, a number of which are probably luminous hot stars. We confirm the presence of a blue near infrared object (K≈13) at the position of the compact radio source Sgr A∗. The spatial centroid of the source number distribution is consistent with the position of Sgr A∗ but not with a position in the IRS 16 complex. The stellar surface density in the central 10″ is very well fitted by an isothermal cluster model with a well defined core radius. The derived core radius of all 340 sources is 0.15±0.05 pc. The central stellar density is a few times 107 M⊙ pc−3. Buildup of massive stars by merging of lower mass stars and collisional disruption of giant atmospheres are very probable processes in the central 0.2 pc.

Abstract Introduction Ring sideroblasts (RS) are common findings in myeloid malignancies as MDS or MDS/MPN, especially in RARS-T, and are associated with SF3B1 mutations. However, the incidence of both RS and SF3B1 mutations have not yet been assessed properly in acute myeloid leukemia (AML). Aim Determine the frequency of RS and SF3B1 mutations in 1857 patients with de novo and therapy-related AML (t-AML). Define their impact on survival and association with other frequently mutated genes, as well as with cytogenetic abnormalities. Patients and Methods From a total of 1857 AML patients (excluding those with cytogenetically-defined entities according to WHO), bone marrow assessment revealed 473 (25%) with RS ≥ 1% and thereof 183 (10% of all) with ≥15 RS. Sequencing or melting curve analysis were performed in a subcohort of 340/473 patients for the detection of mutations in: SF3B1, ASXL1, DNMT3A, FLT3- TKD, IDH1 R132, IDH2 R140, IDH2 R172, KRAS, NPM1, NRAS, RUNX1, TET2, FLT3 -ITD and MLL -PTD. These 340 cases were subject to the study. Out of these 340, 141 cases (42%) had RS ≥ 15% and the remaining 199/340 (59%) had RS ≥1 to <15%. The cohort consisted of 303 (89%) de novo AML (FAB: M0 n=18, M1 n=67, M2 n=165, M4 n=30, M5 n=3, M6 n=20) and 37 (11%) t-AML. 148 were female and 192 were male, with median age 74 years, range: 20-93 years. Chromosome banding analysis (assisted by FISH if needed) was performed in all 340 cases. Results The percentage of bone marrow blasts correlated inversely with the percentage of RS present (r 0.213, p<0.001). 136 (40%) patients had a normal karyotype. Intermediate cytogenetics according to MRC criteria were found in 193 (57%), and adverse in 147 (43%). Patients with RS ≥15% more frequently had adverse cytogenetics in comparison to those with RS between 1-14% (54% vs 36%, p=0.001). The frequencies of gene mutations were as follows: TP53 103/331 (31%), RUNX1 84/315 (26%), DNMT3A 86/337 (25%), TET2 68/330 (20%), ASXL1 58/334 (17%), IDH2 R140 53/338 (15%), NPM1 43/340 (12%), SF3B1 34/334 (10%), FLT3 -ITD 33/340 (10%), NRAS 29/340 (8%), IDH1 R132 21/339 (6%), MLL -PTD 22/337 (6%), FLT3 -TKD 18/333 (5%), KRAS codon 12 13/299 (4%), IDH2 R172 13/338 (3%) and KRAS codon 61 3/299 (1%). Moreover, in total 30 variants in 28 patients were identified in DNMT3A, RUNX1, TET2 and TP53, which according to current knowledge cannot be assigned to mutations or SNPs yet. Patients with ≥15% more frequently had TP53 mutations (mut) (44% vs 22%, p<0.001) and less frequently IDH2 R140 mut (11% vs 19%, p=0.094) and MLL -PTD (2% vs 6%, p=0.006). Accordingly, patients with TP53 mutations had higher percentages of RS as compared to those without (28% vs 16%, p<0.001) and patients with IDH2 R140 mut and MLL -PTD, respectively, had lower percentages of RS as compared to those without (15% vs 21%, p=0.043 and 11% vs 21%, p=0.025, respectively). Furthermore, patients with mutations in the following genes had fewer RS than patients with the respective wild-types: ASXL1 (15% vs 21%, p=0.040), FLT3 -ITD (14% vs 21%, p= 0.049), IDH2 R140 (15% vs 21%, p=0.043), MLL -PTD (11% vs 21%, p=0.025), NPM1 (13% vs 21%, p=0.018) and KRAS codon 61 (3% vs 20%, p<0.001). Conversely, patients with mutated SF3B1 had more RS than patients with wild type (27% vs 19%, p=0.054). However, the number of RS did not translate into an increase in the mutational burden of SF3B1. Given the limited degree of overlap between mutations of the four most frequently mutated genes, we hierarchically separated the patients into 5 groups: TP53 mut, ASXL1 mut, NPM1 mut, SF3B1 mut and patients without any of these mutations. Interestingly, the percentage of RS was very similar in the two groups, TP53 mut and SF3B1 mut, and significantly higher as compared to all other groups (TP53 mut/SF3B 1mut: 28% vs ASXL1 mut/NPM1 mut/others 14%, p<0.001). The number of RS did not have an impact on the overall survival (OS) and event free survival (EFS) of patients. Conclusion 1. Ring sideroblasts ≥ 15% are present in 10% of de novo and t-AML. 2. The blast count correlates inversely with the number of RS. 3. Patients with ≥ 15% RS more frequently carry TP53 mutations and adverse cytogenetics. 4. Although patients with ≥ 15% RS have worse molecular (TP53 mut) and cytogenetic features, there is no statistically significant impact on survival when compared to patients with <15% RS. 5. Background:Circulating anti-drug antibodies (ADAs) are detectable approximatively in 33% of adalimumab treated rheumatoid arthritis (RA) patients, often within the first 6 months of therapy1-2. Classically, circulating ADAs associate with their specific drugs to form immune complexes, increasing drug clearance, and by this mechanism reducing therapeutic effect3. B cell involvement leading to ADAs production is not yet well established.Objectives:To study early ADAs formation according to clinical response to an adalimumab therapy in RA patients and the relationship between ADAs and circulating B cell subsets.Methods:28 RA patients and 13 healthy controls were included. Patients all presented inadequately controlled RA under conventional treatment, were naive of biotherapies, and started an adalimumab treatment at baseline (M0). Responder status was determined according to the DAS28CRP score (<or>3.2) at 3 (M3) and 6 months (M6). ADAs plasma concentration >10pg/mL at M3 defined the immunized patient group. Circulating B cell subsets were quantified by flow cytometry at M0 and M3.Results:11 (42.3%) patients were immunized at M3. Among them, 4 (36.4%) were responders at M6 and 7 (63.6%) were non-responders. Presence and concentration of ADAs at M3 was associated to non-responder status at M6 (p=0.043; p=0.042). Immunized patients had lower transitional B cells count at M0 compared to non-immunized patients (p=0.031).Conclusion:A high but classical proportion of RA patients developed ADAs after only 3 months of adalimumab treatment. This immunization was associated to non-responder status at M6 and to a low blood transitional B cells count at baseline. Our results suggest transitional B cells implication inRA activity and biotherapy resistance due to immunization. The intrapulmonary pharmacokinetics of oral azithromycin were studied in 25 healthy volunteers, each of whom received an initial dose of 500 mg and then 250 mg once daily for four additional doses. Bronchoscopy, bronchoalveolar lavage, and venipuncture were performed 4, 28, 76, 124, 172, 244, 340, and 508 h after the first dose was administered. Azithromycin concentrations in epithelial lining fluid (ELF), alveolar macrophages, peripheral blood monocytes, and serum were measured by high-performance liquid chromatography. Azithromycin was extensively concentrated in cells and ELF. Drug concentrations in AMs (peak mean +/- standard deviation, 464 +/- 65 micrograms/ml) exceeded 80 micrograms/ml up to 508 h (21 days) following the first dose, while concentrations in PBMs (peak, 124 +/- 28 micrograms/ml) exceeded 20 micrograms/ml up to 340 h (14 days). Azithromycin concentrations in ELF peaked at 124 h (3.12 +/- 0.93 micrograms/ml) and were detectable up to 172 h (7 days), when they were 20 times the concurrent serum concentrations. Although the clinical significance of antibiotic concentrations in these compartments is nuclear, the sustained lung tissue penetration and extensive phagocytic accumulation demonstrated in this study support the proven efficacy of azithromycin administered on a 5-day dosage schedule in the treatment of extracellular or intracellular pulmonary infections.

We describe a new Typhlops species from mid-altitude rainforest (ca. 950 m elevation) of the Andasibe region in central eastern Madagascar. Typhlops andasibensis sp. nov. is a medium-sized species (up to 340 mm total length) and can be separated from all other Malagasy typhlopids by the combination of 26 midbody scale rows, less than 400 middorsals, and a T–V supralabial imbrication pattern. Typhlops microcephalus is confirmed as a valid species, T. boettgeri is resurrected from the synonymy of T. arenarius, and T. capensis is synonymized with Ramphotyphlops exocoeti. Keys to the three genera and 14 described species of Madagascar and Comoro typhlopids are provided.

Irradiation of 3-methyl-2-phenyl-2H-azirine (1) at 254 nm in argon matrices results in ylide 6. Similarly, laser flash photolysis (λ = 266 nm) of azirine 1 in acetonitrile yields ylide 6, which has a transient absorption with λmax at ~340 nm and a lifetime of 14 μs. Density functional theory calculations were preformed to support the characterisation of ylide 6 in solution and argon matrices. Irradiation of azirine 1 above 300 nm has previously been reported (J. Org. Chem. 2014, 79, 653) to yield triplet vinylnitrene in solution and ketenimine in cryogenic argon matrices. Thus, the photochemistry of azirine 1 is dependent on the irradiation wavelength.

Abstract:
A 12-year pregnant Thoroughbred mare at ~ 340 days of pregnancy was presented for evaluation of lethargy, anorexia, and colicsigns. Reproductive examination was unremarkable; however, uroperitoneum was diagnosed via transabdominal ultrasonography,serum biochemistry, and peritoneal fluid analysis. Mare was medically stabilized for 24 hours. To promote fetal maturation, 80mg dexamethasone was given prior to Caesarean section and exploratory celiotomy. A ventral urinary bladder wall rupture andmultiple descending colon mesenteric rents were identified and repaired. Foal was delivered alive, treated for neonatal maladjustmentsyndrome, and discharged after 14 days. Seven days after surgery, severe complications of the mare led to its abrupt declinein health and sudden death.

Abstract:
Introduction The innovative Pratt pouch could optimize dispensing nevirapine prophylaxis to HIV-exposed infants in pre-measured single dose pouches to increase completion of the full 6 week infant nevirapine regimen. Materials and methods Nineteen health facilities with highest HIV positivity rates among pregnant women across 9 districts in southwest and central Uganda were assigned to control and intervention groups. HIV-positive women enrolled at intervention facilities received pouches filled with premeasured single doses of nevirapine using Uganda national guidelines, which were integrated into the existing drug distribution system. During antenatal care (ANC) women received 14 pouches to cover time until the 6 day postpartum visit, with an additional 8 pouches if women were delayed in returning to the facility, and 28 pouches after delivery. Women enrolled at control facilities received standard nevirapine syrup following delivery for postnatal infant prophylaxis. In a select number of intervention facilities, during ANC, women received all 42 pouches needed to complete the 6 weeks regimen. Medical record data from enrolled women were extracted; interviews with HIV-positive women during postnatal care visits were conducted. Data were collected January to August 2018 (control sites) and October 2019 to February 2020 (intervention sites). Unadjusted and adjusted logistic regression models were used to identify factors associated with facility delivery, postnatal care follow-up visit, and completion of the full 6 weeks infant nevirapine regimen. Results Significantly more women in the intervention (n = 320) versus control (n = 340) group had facility delivery (292/316, 92.4% versus 169/340, 49.7%, p<0.0001), postnatal visits within 2 weeks postpartum (295/297, 99.3% versus 133/340, 39.1%, p<0.0001) and reported their infants completing the full 6 weeks infant prophylaxis regimen (299/313, 95.5% versus 210/242, 86.8%, p = 0.0002). Dispensing 42 versus 14 pouches during ANC did not have negative effects on these outcomes. Among out-of-facility deliveries, a higher proportion of infants received nevirapine within 72 hours of birth in the intervention versus control group, 95.8% versus 77.9%. In multivariate models, the intervention group was the only significant factor associated with facility delivery or completion of the full 6 weeks infant prophylaxis. Conclusions Use of the Pratt pouch resulted in an increase in HIV-exposed infants completing the full 6weeks prophylaxis regimen and associated benefits including increasing facility delivery and women’s adherence to postnatal care services.

A phased array Doppler Mini-Sodar has been installed at Anupuram near Kalpakkam nuclear site during the Coastal Atmospheric Boundary Layer Experiments (CABLE) carried out on the east coast of India. Mini-Sodar was used to measure the wind profile and investigate the coastal boundary layer development due to land-sea breeze circulation for the study of coastal dispersion of pollutants. The atmospheric boundary layer, its variability during SW (August 13, 14 : ARMEX I and September 3, 7, 11, 14) and pre-monsoon period (April 27, 28 : ARMEX II) 2003 has been studied. Large wind direction change and high vertical velocity variance were observed during the onset of sea breeze. The Thermal Internal Boundary Layer (TIBL) height has been found to increase progressively with season: summer - 170 m, SW monsoon - 210 m and NE monsoon - 325 m whereas the Nocturnal Boundary Layer (NBL) height decrease progressively during the corresponding period: summer - 370 m, SW monsoon - 340 m and NE monsoon - 210 m.

The occurrence of polyprenols in leaves of over 340 species of dendroflora in natural habitats in the regions of Hanoi and Hue in Vietnam was studied. Plant material was collected in the late autumn (October/November) during the end of a vegetation season. Leaves of about 200 plant species did not contain detectable amounts of polyprenols in contrast to few systematic families, e.g. Moraceae, Euphorbiaceae, where polyprenols were highly abundant and their pattern could be used as a chemotaxonomic criterion. Most often dominating polyprenols were prenol-11 and prenol-12. In several angiosperm species prenol-13 and detectable amounts of prenol-14 were also found. The incidence of prenol-13 and -14 was not restricted to a specific taxonomic group since species exhibiting domination of such longer chain polyprenols belonged to various systematic families. In some plants (e.g. Ceiba pentandra) alpha-cis polyprenols were accompanied by alpha-trans counterparts. This report describes several new plant species that may serve as natural sources of long chain polyprenols.

The advantages of straight-run diesel fuel hydrotreated at pressure of 80 atm on a nickel-molybdenum catalyst over hydrocracking diesel fuel at the use of isodewaxing process as a feedstock to obtain hydrocarbon base for drilling fluids. Based on the results of the laboratory assessment, a test run was carried out at process unit. It was possible to obtain a hydrocarbon base for drilling fluids at the dewaxing unit of the diesel fuel hydrotreatment unit in the amount of 340 tons/day (14 t/h) without compromising quality. The dewaxing unit is idle in the summer time due to the lack of demand for winter diesel fuel, so its use at this time significantly increases the production margin.

Extracellular vesicles (EVs) are involved in intercellular signalling through the transfer of molecules during physiological and pathological conditions, such as ischaemic disease. EVs might therefore play a role in ischaemic stroke (IS) and myocardial infarction (MI). In the present study, we analysed the similarities and differences in the content of circulating EVs in patients with IS and MI. This prospective observational study enrolled 140 participants (81 patients with IS, 37 with MI and 22 healthy controls [HCs]). We analysed the protein and microRNA content from EVs using proteomics and reverse transcription quantitative real-time polymerase chain reaction and compared it between the groups. In the patients with IS and MI, we identified 14 common proteins. When comparing IS and MI, we found differences in the protein profiles (apolipoprotein B, alpha-2-macroglobulin, fibronectin). We also found lower levels of miR-340 and miR-424 and higher levels of miR-29b in the patients with IS and MI compared with the HCs. Lastly, we found higher miR-340 levels in IS than in MI. In conclusion, proteomic and miRNA analyses suggest a relationship between circulating EV content and the patient’s disease state. Although IS and MI affect different organs (brain and heart) with distinct histological characteristics, certain EV proteins and miRNAs appear to participate in both diseases, while others are present only in patients with IS.

The data of 340 issues of PJSIR was downloaded and collected to tabulate from the website of electronic journal: (http: www.pjsir.org/arc.php) during January-July, 2018. This study expressed that n=4929; 14.4% articles were published in 340 issues of PJSIR during the period of 1958-2007. Total 4417 (1790; 36.3%, 1651; 33.5%, and 976; 19.8%) articles published under the caption of Physical, Biological, and Technology out of 4929 articles. Remaining 512; 10.3% articles were short communications. Maximum articles n=1375; 28% were published in the fourth decade and n=694; 14% articles in the first decade as a minimum. The short communications n=208; 4.2% related to biological science take a position with the slight margin to other disciplines. PJSIR published regularly from 1958 to this day. It is counted a teamwork of the management of Journal and supported by Pakistan Council of Scientific and Industrial Research (PCSIR) Government-owned body. There are few examples in the world to publish a scientific journal which covers three major disciplines of science.

Introduction: Frenum attachments are fold of mucous membrane attaching the lips to alveolar mucosa and underlying periosteum. Aberrant location of attachment of maxillary labial frenum poses many clinical problems such as mucogingival problems and midline diastema. Objective: The study was performed to assess the variations in morphology of maxillary labial frenum in a sample of Newari children of Bhaktapur, Nepal. Methods: A descriptive cross-sectional study was performed enrolling 340 Newari children residing in Bhaktapur from August 14, 2020 to March 30, 2021. The study comprised both genders within age group of 6 to 16 years. The frenal attachment was recorded according to Mirko and Sewerin classification. The data was analysed with Statistical Package for the Social Sciences version 16 using descriptive statistics. Results: In total, 340 Newari children participated in this study. The mucosal type was observed in 204 (60%) of the participants followed by gingival type 101 (29.7%). More than 80% of the participants had simple frenum (283, 83.2%). The frenum with nodule was present in 25 (7.4%) and frenum with appendix in 22 (6.5%) of study participants. Frenum with appendix was mostly present in males 18 (10.1%) than females 4 (2.5%). Conclusion: Within the Newari children, the mucosal type of maxillary labial frenum was more prevalent followed by gingival type. The simple frenum was the most common type on basis of morphology.

Leukotrienes are potent inflammatory and lipid mediators that participate in atherosclerosis. We analyzed the association of Leukotriene gene (ALOX5, ALOX5AP, LTA4H, and LTC4S) polymorphisms and plasma Leukotriene B4 (LTB4) levels with coronary artery disease (CAD) in a representative cohort of Asian Indians. In all, 136 functional single nucleotide polymorphisms (SNPs) were selected using in silico tools. Forty-five polymorphic SNPs were ranked for predicted functional effect using FastSNP. Finally, 14 functional SNPs along with 10 SNPs identified from the literature were genotyped in 340 CAD patients and 340 controls. Plasma LTB4 levels were measured in 150 cases and 150 controls. None of the 24 SNPs showed significant association with CAD. Plasma LTB4 levels were higher in cases than in controls (76.42 ± 4.46 pg/mL versus 60.89 ± 2.61 pg/mL) (P=0.003), with greater risk being associated with the top quartile as compared to the bottom quartile after adjusting for potential confounders (OR 8.94, 95% CI 2.56–31.95; P=0.001). Four SNPs in the LTA4H gene showed significant association with LTB4 levels (P<0.05) of which rs1978331 (P=0.035) remained significant after correction for multiple testing. LTB4 showed strong correlation with lipids (r=0.24–34) only in cases. Our pilot study suggests that the association between Leukotrienes gene polymorphisms and CAD risk may be modulated through plasma LTB4 levels.

Objective: To evaluate the action of ascorbic acid on the healing of malnourished rats' cutaneous wounds compared with normal weight rats. Methods: We used 92 adult, male Wistar rats divided into four groups: 24 normal weight rats given only water and chow; 24 normal weight rats given vitamin C by gavage (340 mg/kg 12/12 hours); 22 malnourished rats given only water and chow; and 22 malnourished rats given vitamin C by gavage (340 mg/kg 12/12 hours). Malnutrition was induced by feeding the animals half of their daily energy requirement for 30 days. Two incisions were made, one sutured (healing by primary intention) and one left unsutured (healing by secondary intention). The rats were euthanized on the third, seventh, and fourteenth days of the experiment. Results: The following parameters differed significantly between the groups (p>0.05): granulation of the wound edge in the primary and secondary intention; extent of injuries on day 7 for primary intention and on day 3 for secondary intention; reepithelialization on day 7 for primary intention; fibrin-leukocyte scab on day 14 for primary intention; amount of neovascularization and concentration of macrophages, fibroblasts, and collagen fibers for primary and secondary intention. Conclusion: The use of vitamin C in malnourished and normal weight rats increases fibroblast proliferation and collagen deposition in the tissue, which helps to improve healing both by primary and secondary intention.

The article presents the results of research on the impact of various foliar fertilizers on the formation of grain productivity of maize in the Western Forest-Steppe. The research has established the best parameters for the use of fertilizers for foliar application in maize cultivation technologies. It was found that among the corn hybrids studied, the highest yield was recorded by RAGT Reakksion from FAO 320. According to the results of research in 2023, its grain productivity was 10.79 t/ha in terms of basic moisture content 14 %. A slightly lower yield was observed in the RAGT Zanetikks hybrid (FAO 340) – 10.73 t/ha. The dependence of harvesting grain moisture content on the ripeness group of the studied hybrid crop was proved. The highest moisture content was recorded in the hybrids RAGT Zanetikks (FAO 340) – 24.8 % and RAGT Reakksion (FAO 320) – 24.3 %. At the same time, it was the lowest in the hybrid RAGT Alikks (FAO 220) – 21.7 %. Among the options for foliar application of microfertilisers, the most effective was the use of YaraVita Gramitrel and Vuksal Grain. Depending on the maize hybrid, the grain yield in these experimental variants was 9.56-11.01 and 9.80-11.12 t/ha, respectively. The harvesting moisture content of the grown products was determined by the biological characteristics of the studied hybrid, while foliar fertilisation had almost no effect on this indicator

Background: Irritable Bowel Syndrome (IBS) is a chronic and common functional bowel disorder that currently has no definite treatment. Depending on the type and severity of the individual's symptoms, medication, diet, and/or lifestyle changes are recommended. Aims: This study aimed to determine the relationship between the quality of life, perceived stress levels, and nutrition of individuals diagnosed with IBS. Subjects and Methods: Between March 2020 and March 2021, 340 patients with IBS volunteered for this study. The demographic information questionnaire, Irritable Bowel Syndrome Quality of Life (IBS-QOL) scale, Perceived Stress Scale-14 (PSS-14), and Food Frequency Questionnaire were applied via an online form. Results: From the 340 patients, 230 were eligible. Out of them, 27% followed a special diet for IBS. When the IBS-QOL scores of men were compared according to their educational status, the scores of those with a high school or higher education level (109.7±32.8) were found to be higher than those with a lower education level than high school (95.4±17.5) (p=0.008), while the educational status of women did not affect the IBS-QOL scores (p>0.05). The mean IBS-QOL score of men was 44.8 ± 20.1 and lower than women (50.9±21.7) (p=0.030). The mean IBS-QOL score of women following an IBS-specific diet was 57.6 ± 22.6 and higher than women who did not follow an IBS-specific diet (48.2±20.9) (p=0.023). The IBS-QOL scores of women with gluten or lactose intolerance were lower than women without any intolerances (p=0.004). The mean IBS-QOL score for women with lactose intolerance was higher than those with gluten intolerance (p=0.004). When the IBS-QOL scores were evaluated according to the frequency of food consumption, no significant differences were found in the food groups (p>0.05). In this study, no significant correlation was found between IBS-QOL scores and PSS-14 scores (p>0.05). Conclusions: The prediction equations developed for healthy populations are not accurate enough to determine the energy requirements in SCD. In the past, Nitobegiku (the herb of Tithonia diversifolia (Hemsl) A. Gray) has been used as a medicinal plant for diabetes. Antidiabetic effect of the water extract of Nitobegiku (NG) was investigated in KK-Ay-mice — one of the animal models of type 2 diabetes. NG (1500 mg/kg body weight) reduced the blood glucose of KK-Ay mice from 509 ± 22 mg/dl to 340 ± 14 mg/dl (p < 0.001) and also lowered the plasma insulin (p < 0.05) 7 hours after single oral administration. No change in blood glucose of NG-treated normal mice (ddY) was seen. These results support that NG improve glucose metabolism by reducing insulin resistance. Therefore, NG may be useful for treatment of type 2 diabetes.

A simple plasma emission spectroscopic method for the determination of iron in liver and cardiac tissue is described. Using this technique, iron was extracted quantitatively from liver tissue of mass 14·2 to 65·4 mg wet weight, and heart tissue of mass 5·9 to 27·4 mg wet weight. Iron added to liver as aqueous ferric nitrate was recovered in the range 93 to 108%. Reference ranges for liver and myocardial iron on post mortem tissue gave respectively mean values of 0·841 mg/g dry weight (Range 0·310 to 1·600, n = 37) and 0·340 mg/g dry weight (Range 0·290 to 0·470, n = 8). Data on patients with haemochromatosis and transfusion siderosis are also presented.

Abstract:
Field studies indicated thiazopyr at 220 g ha−1injured soybeans planted in no-till, and 340 g ha−1injured soybean in 1 of 2 yr planted into tilled soil. Thiazopyr half-lives in three field studies ranged from 12 to 14 d, indicating minimal potential for carryover injury to sensitive rotational crops. Thiazopyr dissipation under controlled conditions was slower than under field conditions. Half-lives at 15 and 30 C were 133 and 43 d, respectively. This indicates that microbial degradation is an important mechanism in thiazopyr dissipation, but that other loss mechanisms also play a role. A method using methanol extraction of moist soil samples followed by gas chromatography using a nitrogen-phosphorous detector had > 90% recovery of thiazopyr.
