It revealed that chassis gear, telpher, and beam were the most suitable parts for the gantry crane. The technological analysis showed what kind of equipment was needed in order to product two shafts and the girder. The gantry crane’s economical indicators, the crane’s cost price and its possible sale-price were calculated.

Full textIn this thesis, we propose a new technic to identify a hybrid system for Data Acquisition,by using ad-hoc sensors on the vehicle, the sensors in the recent smartphones, MBED and CAN-BUS. The assumption is that the Smartphone's sensors will reduce the complexity and the high cost of these instrumentations. The objective is obtaining acceptable measurement accuracy of the collected trajectories and enable for a large-scale deployment of the system's instrumentation, such as a helpful system in the domain of transport. Weshow in this thesis how to build a hybrid system by depending on the properties of the used sensors in both the smartphones and in the vehicles to identify several situation like a failure sensor, accident situation and Rider's behaviour. This system is tested and evaluated on several real time on line - off- line including the used mode and method

Full textEsta investigación pretende reconstruir um perído de la historia de Israel los seis años del govierno de la Reina Atalia, en Judea (841-836 aC). Teniendo como marco el análisis exegético e histórico del 2Rs11,1-3.13-16. El cenario y el estilo del 2Rs 11 nos permite afirmar que el autor pertencía al círculo de la escribas de la corte o de la institución religiosa. La investigación recurre a la particular ocurrencia de los términos reina madre y gebirah, presentes en el Antiguo Testamento, así como las contribuciones de las tradiciones de los países vecinos con el fin de explicar la ascensión de Atalia al trono de Judea. Atalia ejerció la función de reina madre; dentro de esta función tenía como compentencia la importante missón de cuidar de la realeza y eventualmente ejercer la regencia hasta que la ocupación por derecho y viabilizar su acción. Atalia gobernó durante seis años en Judea; para mantenerse tanto tiempo en el poder contó con las fuerzas aliadas es decir con un grupo que la afianzó en el trono. Atalia es hija de la casa de Omri su madre israelita de fe yavista. La partida de Atalia para Jerusalén es el resultado de un periodo de paz entre los dos reinos hermanos. La durabilidad de su gobierno se debe no a los actos de beligerancia y sí a su ejemplar administración basada principalmente en la actividad comercial favorecida por la posición estratégica de Jerusalén, siendo el puonto geográfico donde convergían impotantes rutas comerciles de la época. Atalia se emcontraba preparada para liderar un país cuya meta era el comercio internacional.
Esta pesquisa busca reconstruir um período da história de Israel os seis anos de governo da Rainha Atalia, em Judá (841-836 aC) tendo como base a análise exegética e histórica de 2Rs11,1-3.13-16. O cenário e o estilo de 2Rs 11 permitem afirmar que o autor pertencia ao círculo dos escribas da corte ou da instituição religiosa. A pesquisa recorre às ocorrências dos termos rainha-mãe e gebirah, presentes no Antigo Testamento, e às contribuições das tradições dos países vizinhos, a fim de explicar a ascensão de Atalia ao trono de Judá. Atalia ocupou a função de rainha-mãe. Com isso competia-lhe a importante função de cuidar da estabilidade da realeza e, eventualmente, exercer a regência até que a ocupação por direito se tornasse viável. Atalia foi soberana por seis anos em Judá. Para manter-se tanto tempo no poder, Atalia contou com forças aliadas, ou seja, um grupo que a garantiu no trono. Atalia é filha da casa de Omri com mãe israelita de fé javista. A vinda de Atalia para Jerusalém é resultado de um período de paz entre os dois reinos irmãos. A durabilidade de seu governo deve-se, não a atos de beligerância, e provavelmente sim à sua exemplar administração, baseada principalmente na atividade comercial, favorecida pela posição estratégica de Jerusalém no cruzamento de importantes rotas comerciais da época. Atalia estava preparada para liderar um país com metas no comércio internacional.

Full textOs tumores de mama são caracterizados pela sua alta heterogeneidade. O câncer de mama é uma doença complexa, que possui o seu desenvolvimento fortemente influenciado por fatores ambientais, combinada a uma progressiva acumulação de mutações genéticas e desregulação epigenética de vias críticas. Alterações nos padrões de expressão gênica podem ser resultado de uma desregulação no controle de eventos epigenéticos, assim como, na regulação pós-transcricional pelo mecanismo de RNA de interferência endógeno via microRNA (miRNA). Estes eventos são capazes de levar à iniciação, à promoção e à manutenção da carcinogênese, como também ter implicações no desenvolvimento da resistência à terapia Os miRNAs formam uma classe de RNAs não codificantes, que durante os últimos anos surgiram como um dos principais reguladores da expressão gênica, através da sua capacidade de regular negativamente a atividade de RNAs mensageiros (RNAms) portadores de uma seqüencia parcialmente complementar. A importância da regulação mediada por miRNAs foi observada pela capacidade destas moléculas em regular uma vasta gama de processos biológicos incluindo a proliferação celular, diferenciação e a apoptose. Para avaliar a expressão de miRNAs durante a progressão tumoral, utilizamos como modelo experimental a série 21T que compreende 5 linhagens celulares originárias da mesma paciente diagnosticada com um tumor primário de mama do tipo ErbB2 e uma posterior metástase pulmonar. Essa série é composta pela linhagem obtida a partir do tecido normal 16N, pelas linhagens correspondentes ao carcinoma primário 21PT e 21NT e pelas linhagens obtidas um ano após o diagnóstico inicial, a partir da efusão pleural no sítio metastatico 21MT1 e 21MT2. O miRNAoma da série 21T revelou uma redução significativa nos níveis de miR-205 e nos níveis da proteina e-caderina e um enriquecimento do fator pró-metastático ZEB-1 nas células 21MT. Considerando a importância dos miRNAs na regulação da apoptose, e que a irradiação em diferentes espectros é comumente usada em procedimentos de diagnóstico como mamografia e na radioterapia, avaliamos a expressão de miRNAs após irradiação de alta e baixa energia e do tratamento doxorrubicina. Para os ensaios foram utilizados as linhagens não tumorais MCF-10A e HB-2 e as linhagens de carcinoma da mama MCF-7 e T-47D. Observou-se que raios-X de baixa energia são capazes de promover quebras na molécula do DNA e apoptose assim como, alterar sensivelmente miRNAs envolvidos nessas vias como o let-7a, miR-34a e miR-29b. No que diz respeito à resposta a danos genotóxicos, uma regulação positiva sobre a expressão de miR-29b, o qual em condições normais é regulado negativamente foi observada uma regulação positiva sobre miR-29b expressão após todos os tratamentos em células tumorais. Nossos resultados indicam que miR-29b é um possível biomarcador de estresse genotóxico e que miR-205 pode participar no potencial metastático das células 21T.
Breast tumors are characterized by their high heterogeneity. It is a complex disease, which has its development strongly influenced by environmental factors, combined with a progressive accumulation of genetic mutations and epigenetic dysregulation of critical pathways. Changes in gene expression patterns may be a result of a deregulation in epigenetic events as well as in post-transcriptional regulation driven by RNA interference endogenously represented by microRNA (miRNA) these mechanisms are capable to promote the initiation, maintenance and progression of carcinogenesis; they are also implicated on the development of therapy resistance. miRNAs form a class of non-coding RNAs which have emerged in recent years as one of the major regulators of gene expression through its capacity to silence messenger RNAs (mRNAs) containing a partially complementary sequence. The importance of regulation mediated by miRNAs was observed on their ability to regulate a wide range of biological processes including cell proliferation, differentiation and apoptosis.To gain insights into the mechanisms involved in breast cancer initiation and progression conducted a miRNA global expression on 21T series that are an in vitro model of breast cancer progression comprising cell lines derived from the same patient which include a normal epithelia (16N), primary in situ ductal carcinoma (21PT and 21NT) and cells derived from pleural effusion of lung metastasis (21MT-1 and 21MT-2). Considering the importance of miRNAs in the regulation of apoptosis, and that irradiation in different spectra is commonly used in diagnostic procedures as mammography and on radiotherapy, we evaluate the miRNA expression after cell low and high energy irradiation and doxorubicin treatment to determine whether miRNAs are useful biomarkers to detect cell response after DNA damage. The experiments were done on the non-tumoral cell lines MCF-10A and HB-2 and on the breast carcinoma derived cell lines MCF-7 and T-47D. We observed that of low energy X-rays is able to promote DNA strand breaks and apoptosis and to slightly change the expression of miRNAs involved on this pathway such as let-7a, miR-34a and miR-29b. Regarding DNA stress response pathways an upregulation on miR-29b expression, that in normal conditions is downregulated in tumor cell lines could be observed after all treatments. The microRNAome of 21T series revealed a significant downregulation of miR-205, an enrichment of the prometastatic factor ZEB-1, potential target for miR-205 and the consequent reduction of ecadherin levels in 21MT cells checked by western blot. Our results indicate that miR-29b is biomarkers of genotoxic stress and that miR-205can participate on the metastatic potential of 21T cells.

Resumo: A presente dissertação visa contribuir para a área de diagnóstico de falhas em produtos de fabricação em massa após terem sido vendidos ao consumidor final. Para alcançar este objetivo, foi proposta a metodologia 2RP (Rapid Response For Production), a qual integra as seguintes ferramentas: Diagrama de Pareto, Diagrama de Afinidades, Diagrama FAST e Diagrama Matriz, sendo esta última auxiliada pela lógica difusa, a fim de superar as limitações ao pontuar as variáveis correlacionadas. Para validar a metodologia, foi feita uma aplicação em um caso prático, utilizando a base de dados da empresa Alpha, extraídas da plataforma virtual Reclame Aqui. Na primeira etapa, a metodologia serviu para identificar o produto e suas principais reclamações por meio do uso das ferramentas Diagramas de Pareto e Diagrama de Afinidade. Na segunda etapa, o produto foi caracterizado e classificado em suas funções básicas e secundárias, mediante o Diagrama FAST. Já na terceira etapa, utilizou-se o Diagrama Matriz para relacionar os principais problemas com os componentes que afetam o desempenho do produto para, assim, identificar os componentes que causaram a falha. Finalmente, o resultado obtido após a aplicação da metodologia 2RP permite que as empresas possam tomar ações para corrigir problemas de produção a fim de evitar reincidências de falhas e, consequentemente, de reclamações. Abstract: The following master thesis aims to contribute to the area of diagnosis of failures in mass-produced products after being sold to the final consumer. To reach this objective, the Rapid Response For Production (2RP) methodology was proposed, which integrates the following tools: Pareto Diagram, Affinity Diagram, FAST Diagram and Matrix Diagram, this last tool being aided by diffuse logic to overcome the limitations when punctuating the correlated variables. Methodology validation was applied in a practical case, using the Alpha company database, extracted from the virtual platform Reclame Aqui. In the first step, the methodology served to identify the product and its main complaints through the use of the Pareto Diagrams and Affinity Diagram tools. In the second step, the product was characterized and classified in its essential and secondary functions, through the FAST Diagram. In the third step, the Matrix Diagram was used to relate the main problems with the components that affect the performance of the product to identify which ones caused the failure. Finally, the result obtained after applying the 2RP methodology allows companies to take actions to correct production problems to avoid recurrence of failures and, consequently, complaints. Full textThe SNAP (SuperNovae Acceleration Probe) mission is designed to measure very precisely the cosmological parameters and to determine the nature of the Dark energy. The mission is based on the measurement of some thousands supernovae of type Ia up a redshift of z=1. 7 and on weak gravitational lensing measurements of more than one thousand square degrees of the sky. The SNAP experiment consists in a 2-meter telescope with a one square-degree imager and an integral field spectrograph. We present in this thesis study on hybrid detector H2RG (produce by Teledyne) to improve performances of the SNAP spectrograph. The H2RG 40 detector was characterized and used in the spectrograph demonstrator. The way of sampling has been optimized to decrease the readnoise and detect cosmic ray and the impact on the spectrograph performances have been evaluated

Full textAccording to the WHO, major depressive disorder (MDD) will be the second leading cause of disability in the world in 2020 and will become the first in 2030. Conventional antidepressant drugs have delayed therapeutic effects and many patients are resistant. Ketamine, an N-methyl-D-aspartate (NMDA-R) receptor antagonist of L-glutamate, exerts a rapid antidepressant effect in patients who are resistant to standard therapy. The mechanism of this amazing activity is not well understood. By coupling intracerebral microdialysis to a predictive behavioral test of antidepressant activity in a BALB/cJ mouse model with an anxious phenotype, we show that this ketamine activity is dependent on the excitation-inhibition balance between glutamate/NMDA-R and AMPA-R, GABA/GABAA-R, serotonin systems in the prefrontal cortex/raphe nucleus circuit. Our results also suggest that it would be the combination [ketamine-(2R,6R)-hydroxynorketamine, its main brain metabolite] that would carry the antidepressant effect. My thesis work pave the way for the development of new fast-acting antidepressant drugs

Full textIt is thought a depth psychosis literature review, that we have, in a first time, highlighted various positions within the psychiatric nosography, in an historic and then, in a current context. We cross a perspective called objective to an approach more subjective, however very rewarding: the metapsychological model, whose milestones were thrown by Freud (1911, 1920, 1924, 1940). In addition, we highlighted an opposition between Freud and Bleuler thought their epistolary exchanges published in 2016 in French. This about, question of autism that Freud (1911) designated as well as a retreat of the libido on the Ego; contrarily to Bleuler (1911). To us, that epistemological and theoretical cleavages, have influenced the current nosography (DSM-5, A.P.A., 2013). The empirical research developed in this thesis, aimed at the study of dissociated psychosis through a clinical and projective compared approaches. Indeed, this research is composed by many studies: the first was aimed at the statistical comparisons of Rorschach’s indicators (Nina Rausch de Traubenberg, 1990) between non-clinical population ([n= 310], (de) Tychey et al., 2012) and clinical population, psychiatrist, and more specifically: psychosis dissociated ([in]patient [n= 81]). Within the first study, in a second time, we proceeded to the statistical comparisons between the three subgroups of the sample, constitute of: 25 subjects diagnosed with paranoia, 30 subjects diagnosed with schizophrenia and 26 diagnosed with paranoid schizophrenia. All diagnostics were realized in double bind et and according to a double approach (psychoanalytic and psychiatrist). The second study is a qualitative analysis contrasted, devoted to the deeply psychic investigation of exemplary clinical cases, representatively of three subgroups from the population’s study (paranoia; schizophrenia paranoid; pure schizophrenia). The sample is itself constituted of limits’ subgroups suffering from psychotic decompensations and hospitalized on a long court. In the qualitative study of clinical contrasted cases, we presented firstly, a subject from a pure paranoia structure: Catherine, an autopunitive feminine paranoia case.Secondly, we presented the analysis of a clinical picture from schizophrenic structure: Joseph, from which megalomaniac Ego and invasive delirium will surely arrested the lector; and finally, we analysed a pure schizophrenia case: Doug, which psychic desert particularly challenges us in conter-transferential’s dynamic. Then, to illustrated closer that we understand by sliding of an a-structure to a dissociated psychosis (and not to a melancholic proceed, as well as that is permitted by Bergeret [1974]), we analysed the cases of Jordan and Ali. Full textBreast tumors are characterized by their high heterogeneity. Breast cancer is a complex disease, which has its development strongly influenced by environmental factors, combined with a progressive accumulation of genetic mutations and epigenetic dysregulation of critical pathways. Changes in gene expression patterns may be a result of a deregulation in epigenetic events as well as in post-transcriptional regulation driven by RNA interference endogenously represented by microRNA (miRNA). These mechanisms are capable to promote the initiation, maintenance and progression of carcinogenesis and are also implicated on the development of therapy resistance. miRNAs form a class of non-coding RNAs, which have emerged in recent years as one of the major regulators of gene expression through its capacity to silence messenger RNAs (mRNAs) containing a partially complementary sequence. The importance of regulation mediated by miRNAs was observed on their ability to regulate a wide range of biological processes, including cell proliferation, differentiation and apoptosis.To gain insights into the mechanisms involved in breast cancer initiation and progression we conducted a miRNA global expression on 21T series that are an in vitro model of breast cancer progression, comprising cell lines derived from the same patient, which include a normal epithelia (16N), primary in situ ductal carcinoma (21PT and 21NT) and cells derived from pleural effusion of lung metastasis (21MT-1 and 21MT-2). Considering the importance of miRNAs in the regulation of apoptosis, and that irradiation in different spectra is commonly used in diagnostic procedures, as mammography and on radiotherapy, we evaluated the miRNA expression after cell low and high energy irradiation and doxorubicin treatment to determine whether miRNAs are useful biomarkers to detect cell response after DNA damage. The experiments were done on the non-tumoral cell lines MCF-10A and HB-2 and on the breast carcinoma derived cell lines MCF-7 and T-47D. We observed that low energy X-rays is able to promote DNA strand breaks and apoptosis and to slightly change the expression of miRNAs involved on this pathway, such as let-7a, miR-34a and miR-29b. Regarding DNA stress response pathways, an upregulation on miR-29b expression, that in normal conditions is downregulated in tumor cell lines could be observed after all treatments. The microRNAome of 21T series revealed a significant downregulation of miR-205, an enrichment of the pro-metastatic factor ZEB-1, potential target for miR-205 and the consequent reduction of e-cadherin levels in 21MT cells checked by western blot. Our results indicate that miR-29b is a possible biomarker of genotoxic stress and that miR-205 can participate on the metastatic potential of 21T cells

Full textAldosterone is involved in the progression of renal disease in experimental models and clinical trials. Considering that the model of renovascular hypertension 2 kidney-1 clip (2K-1C) is characterized by a high activity of the renin-angiotensin-aldosterone system, the objective of this study was to assess the role of aldosterone in renal injury in this model by blocking the mineralocorticoid receptor (MR) with spironolactone. Male Wistar rats were submitted to surgery to place a silver clip on the left renal artery and after two weeks of the development of 2K-1C renovascular hypertension they were divided into three groups. The first group was administered no drug (n=8), in the second group spironolactone 20 mg/kg/day was given orally (n=10) and the third group received amiloride 7 mg/kg/day (n=12). Body weight and systolic blood pressure were monitored weekly. Samples of urine (collected for 24 h) and blood were performed in 3 distinct periods: before surgery; 2 weeks after surgery (before treatment) and 6 weeks after surgery (after treatment). The samples were used to analyze creatinine, osmolality, sodium, potassium and albuminuria. At the end of the experiment, the kidneys were perfused and weighed. Histological analysis to assess the extent of tubulointerstitial changes was performed. Additionally, markers of inflammation (ED-1 and p-JNK), production of extracellular matrix (fibronectin), myofibroblasts (-smooth muscle actin) and tubular transdifferentiation (vimentin) in the area of tubulointerstitial cortical and injury in podocytes (desmin) were evaluated. Treatment with spironolactone was able to attenuate the increase in albumin excretion, the increase in plasma concentration of creatinine and the reduction in creatinine clearance. In the clipped kidney, tubulointerstitial and podocytes injuries, demonstrated by markers studied, were discrete and therapy with spironolactone or amiloride was not able to minimize them. However, in the non-clipped kidney, administration of spironolactone attenuated the increase of extracellular matrix and podocyte injury. The beneficial effects of spironolactone occurred independent of the reduction blood pressure and can be partly dependent of epithelial sodium channel (ENaC) blockade. These effects bring perspective to espironolactone as a therapeutic tool to be explored in patients with renal artery stenosis.

Full textThe grave challenges to future of traditional memories (flash and DRAM) at 1X nm regime has resulted in increased quest for new physical state variables (other than charge or voltage), new devices and architectures offering memory and logic functions beyond traditional transistors. Many thin film devices with resistance change phenomena have been extensively reported as ’promising candidates’. Among them, Ox- ide Resistive Memory (OxRRAM) and Conductive Bridge Resistive Memory (CBRAM) are leading contenders for the next generation high density memories. In this work, we focus on the role of Resistive Memories in embedded memories and their impact on FPGAs in particular. We begin with the discussion on the compact modeling of resistive memory devices for design enabling, we have designed novel circuits of non- volatile flip-flop (NVFF), non-volatile look-up table (NVLUT), non-volatile 2x2 switch and non-volatile SRAM (NVSRAM) using Resistive Memories. We simulated the impact of these design structures on the FPGA system assessing the performance parameters of area, delay and power. By using the novel 1T-2R memory element concept of CBRAMs in FPGAs to implement Look-up Tables (NVLUT), we would scale down the area impact by 5%, enhance speed by 24% and reduce the power by 18% compared to SRAM based FPGAs. The thesis addresses aspects of compact modeling, circuit design and system evaluation using resistive memories

Full textThe pre-competitive state anxiety and it´s possible relationship with the sport performance has become a focus of investigation for many researchers, especially in sport psychology. However in the sport futsal, this relationship is still inconclusive. Thus, the aim of this study was to investigate the relationship between pre-competitive anxiety and performance of professional athletes of futsal during the playoffs of a championship. Furthermore, in this study the possible influence of the phase and game condition on the anxiety and performance responses was analyzed, additionally, the Catastrophe Theory was used to see the interaction amongst cognitive anxiety, arousal and performance responses. To develop this study, 16 male professional futsal athletes were analyzed, belonging to an elite club of São Paulo, Brazil. The study was conducted throughout the playoffs of an important championship on high level for the team. The samples were collected in home and away venue in phases of quarterfinals, semifinals and finals. The data were collected about 15 to 30 minutes before the warm-up in each game, in which the anxiety by CSAI-2R and the physiological arousal by HRV were evaluated. Performance was analyzed subjectively (Likert Scale questionnaire) and objectively (match analysis). The used statistics treatments were: the ANOVA 2x3 for analyzing the influence of phase and game condition; the Pearson\'s correlation for testing the correlation among the variables (state anxiety, physiological arousal and performance) and for the Catastrophe Theory followed specific statistical tests. For all analyzes the alpha was set on 5%. Among the main results we identified a significant influence of the phase of competition for the pre-competitive physiological arousal and the subjective performance. Significant correlations were found among the variables, however, the majority of them was classified as \"moderate\". The Catastrophe Theory presented a positive application on the interaction among anxiety, physiological arousal by HRV indices and performance. Concerning the findings, we can conclude that the pre-competitive state of anxiety and the performance showed a negative relationship in futsal athletes. This relationship can be observed in the Catastrophe Theory, when under high levels of cognitive anxiety, decrease their performance as a function of the increase of physiological arousal. Beyond that, we can conclude that the phase of the game can influence the athlete\'s behavior. When the phase of championship increases, the athletes show a higher level of physical arousal, plus the great subjective performance

Full textDentre os diversos tipos de câncer agressivos, o câncer de mama é o mais comum em mulheres. Mutações hereditárias e adquiridas, assim como alterações epigenéticas atuam em sinergia na carcinogênese mamária e na progressão tumoral. A proteína P53 é uma supressora de tumor e possui uma atuação fundamental na integridade genômica. Apesar do vasto conhecimento sobre o controle da P53 a nível de proteína, ainda pouco se sabe sobre o controle transcricional do gene TP53. A série 21T, uma série de 4 linhagens celulares originadas da mama da mesma paciente, representando diferentes estágios de progressão tumoral mamária, é um eficiente modelo para investigação das alterações epigenéticas e suas influências na expressão gênica ao longo da progressão do câncer de mama. Nós analisamos a organização do domínio do gene TP53 através da técnica de arranjo de DNA, em diversas linhagens celulares de câncer de mama e linhagens controle, e realizamos uma tentativa de caracterizar estes elementos de DNA nas linhagens controle não-tumorais HB2 e MCF10A e nas tumorais MCF-7, MDA-MB-231, T47D, através dos marcadores epigenéticos de eucromatina, H4Ac, e heterocromatina, H3K9me3. Ainda analisamos a ligação de proteínas à região associada à matriz nuclear (MAR), denominada MAR 2, e a possível ligação da proteína ligante à matriz nuclear (MARBP), PARP-1, através de ensaios de gel shift (EMSA). Detectamos que na linhagem controle epitelial mamária, HB2, o gene TP53 está posicionado num domínio de DNA relativamente pequeno, aproximadamente 50 kb, delimitado por dois sítios de fixação à matriz nuclear. Interessantemente, esta estrutura de domínio se apresentou radicalmente diferente nas linhagens de câncer de mama estudadas, MCF7, T47D, MDA-MB-231 e BT474, nos quais o tamanho do domínio estudado estava aumentado e a transcrição do TP53 diminuída. Os enriquecimentos com os marcadores epigenéticos de cromatina H4Ac e H3K9me3 estão diferentemente distribuídos nas MARs nas linhagens celulares. Surpreendentemente, a MAR 2 apresentou uma ligação altamente específica, o que poderia representar a atuação de fatores transcricionais envolvidos na organização da cromatina. Através de programas de bioinformática, detectamos putativos sítios para interessantes fatores de transcrição, tais como o c/EBP-beta e c-myb, que poderiam atuar em cis regulando a expressão do gene TP53 e outros flanqueadores. Nós propusemos um modelo para a organização da cromatina na região de domínio do gene TP53 com os genes flanqueadores. Através da série 21T, detectamos uma hipometilação global genômica, nas células cancerosas 21NT e 21MT1. Uma importante diminuição da expressão global do marcador H4Ac nas células metastáticas 21MT1, foi detectada em relação às outras linhagens. Os níveis de RNAm das principais enzimas relacionadas as modificações epigenéticas são consistentes com as observadas hipometilação genômica e hipoacetilação. Através de microscopia confocal, verificamos que o marcador H4Ac está localizado, na maior parte na periferia e o marcador H3K9me3, pericêntrico nos núcleos tumorais. Por fim, verificamos que o promotor P1 do gene TP53 apresenta um estado de cromatina aberta, e a expressão do gene TP53 é similar em todas as células da série 21T.
Breast cancer is the most common aggressive cancer type in women. Inherited and acquired mutations as well as epigenetic alterations act together in breast carcinogenesis and tumor progression. P53 is a tumor suppressor protein critical for genome integrity. Although its control at the protein level is well known, the transcriptional regulation of the TP53 gene is still unclear. The 21T series, a series of 4 breast cell lines originating from the same patient and representative of the breast tumor progression stages, is a suitable model to investigate epigenetic alterations and their influences upon gene expression during breast tumor progression. We have analyzed the organization of the TP53 gene domain using DNA arrays in several breast cancer and control cell lines and we made an attempt to characterize these DNA elements in breast non-cancerous cell lines HB2 and MCF-10, and cancerous MCF-7, MDA-MB-231 and T47D, through the determination of epigenetic markers of euchromatin, H4Ac, and heterochromatin, H3K9me3. We further analyzed the matrix attachment region (MAR), named MAR 2, protein binding, and possible MAR 2 binding of the important MAR binding protein (MARBP), PARP-1, by Electrophoretic mobility Shift Assay (EMSA). We have found that in the control breast epithelial cell line, HB2, the TP53 gene is positioned within a relatively small DNA domain, encompassing 50 kb, delimited by two nuclear matrix attachment sites. Interestingly, this domain structure was found to be radically different in the studied breast cancer cell lines, MCF7, T47D, MDA-MB-231 and BT474, in which the domain size was increased and TP53 transcription was decreased. H4Ac and H3K9me3, chromatin epigenetic markers enrichment are differentially distributed through MARs in cell lines. Surprinsingly, MAR 2 presented a defined band-shift, which could represent trans-acting factor(s), involved in chromatin organization. By bioinformatics software, we found interesting transcription factors putative binding sites, such as for c/EBP-beta and c-myb, which could be cis-acting elements regulating TP53 and neighboring genes expression. We propose a model for the chromatin organization of the TP53 gene domain with neighboring genes. Through 21T cell line series, we detected a global genomic hypomethylation profile in the cancerous 21NT and 21MT1. An important global decrease of the active chromatin mark H4Ac in the metastatic 21MT1 relative to other cell lines was detected. mRNA levels of key enzymes linked to epigenetic modifications are consistent with the observed genomic hypomethylation and hypoacetylation. By confocal immunofluorescent assay we observed that H4Ac is mostly located at periphery, and the repressive mark H3K9Me3 located pericentric, in tumorigenic cells nuclei. TP53 P1 promoter in 21T series was found to be in an open state and TP53 transcription level was found to be similar in all 21T cell lines.

Full textDC plasma spraying is a coating technology that utilizes the energy of a plasma flow to accelerate, heat up, melt and deposit the coating powder on a substrate. For a high reproducibility in coating production, the plasma flow should have low fluctuations of enthalpy and velocity, and the erosion of electrodes should be limited. Plasma torches with a cascaded anode yield a more stabilized arc length and could achieve the first goal. However, a stabilized arc does not guarantee a low erosion rate of electrodes and in particular anode. An insight into the electromagnetic and thermal processes inside the plasma torch could help to control the properties of the plasma flow issuing from the torch and extend the lifetime of the torch parts. This work dealt with the simulation of the commercial plasma torch SinplexPro™ manufactured by Oerlikon Metco operated in argon under atmospheric pressure. It involved two steps. The first consisted in developing a LTE model with the inclusion of the electrodes in the computational domain. The arc voltage and cooling loss predicted by this model were in good agreement with the experimental values for a low gas flow rate and high electric current. For such operation conditions, the model predicted a constricted anode arc attachment. Then, two methods were tested to rotate the constricted anode arc attachment: gas swirling injection and axial external magnetic field. The swirling gas injection was found to be more efficient for a gas injection angle of 45° than for an angle of 25°, which is the angle used in the actual plasma torch. The second step of this study was about the development of a two-temperature model. The latter considered two formulations for the enthalpy of electrons and heavy species differing in the attribution of the ionization energy to electrons or heavy species. The 2T model with both formulations projected a diffuse anode arc attachment and arc voltage that agreed with the experimental values for a wider range of arc current and gas flow rate. The mode of anode arc attachment was confirmed by the observation of tested anodes. The formulation of the energy equation with the ionization energy assigned to the electrons had an acceptable computational cost and yielded reasonable results in terms of electron and heavy species temperature. It will be used to develop further the model and apply it to diatomic gases

Full textAprès une présentation des généralités sur les systèmes de transmission optique et des principales dégradations subies par le signal, nous abordons la régénération optique. Nous mettons ensuite en évidence la difficulté de démontrer l'efficacité d'un régénérateur. Nous montrons notamment que pour démontrer son efficacité, un régénérateur optique doit être caractérisé dans une liaison.
L'étude numérique et expérimentale d'une liaison comportant une cascade de régénérateurs optiques 3R, nous permet ensuite, dans le cas où l'accumulation de bruit d'amplitude limite la transmission, de connaître l'évolution des densités de probabilité depuissance des symboles et donc du taux d'erreur binaire (TEB).
Le cas d'une régénération 2R est ensuite abordé. Un dispositif original constitué d'un absorbant saturable suivi d'un amplificateur optique à semiconducteur (SOA) est caractérisé avant d'être cascadé dans une ligne de transmission à 10 Gbit/s. L'efficacitédu dispositif est démontrée puisqu'un facteur d'amélioration de la distance de propagation supérieur à 9,5 est obtenu pour un TEB de 10-8 permettant une propagation sur 20 000 km. L'accordabilité du dispositif est également démontrée sur une bande spectrale de 13 nm, ce qui est prometteur pour une transmission WDM.
Ce dispositif 2R nous permet finalement d'aborder de manière plus générale, l'effet d'une cascade de régénérateurs 2R dans une liaison. Une étude numérique clarifie le phénomène d'accumulation de gigue temporelle en présence de régénérateurs 2R. Une boucle à recirculation à pas de régénération variable permet d'étudier expérimentalemen l'impact du pas entre régénérateurs. Il existe un pas de régénération optimal correspondant au meilleur compromis entre accumulation de gigue temporelle et de bruit d'amplitude. L'étude met également en évidence l'intérêt d'espacer largement les régénérateurs (distance doublée pour 6 régénérateurs cascadés à un TEB de 10-8 dans le cas considéré). Enfin la limitation de la transmission par la gigue temporelle est mise en évidence expérimentalement.
