Abstract:
Abstract Blood group incompatibility causes transfusion reactions and neonatal isoerythrolysis in cats. We investigated the molecular nature of the blood group antigens from cats that had blood type A, B, and AB erythrocytes. Naturally occurring anti-type B antibodies, Triticum vulgaris lectin, monoclonal antibody (MoAb) 32–27, and MoAb R-24 were used in agglutination tests, Western blots, and thin-layer chromatography (TLC) enzyme immunostaining. Type A erythrocytes had NeuGc-NeuGc-Galactose-Glucose-Ceramide ([NeuGc]2GD3) where NeuGc represents N-glycolylneuraminic acid, and NeuAc-NeuGc-GD3, where NeuAc represents N-acetylneuraminic acid, and may have [NeuGc]2 disialylparagloboside and NeuAc-NeuGc-disialylparagloboside. Type B erythrocytes only had [NeuAc]2GD3. Type AB erythrocytes had [NeuGc]2GD3, NeuAc-NeuGc-GD3, and [NeuAc]2GD3. Blood group antigens were also found on a 50-Kd membrane protein. We conclude that type B erythrocytes are characterized by [NeuAc]2GD3 as the only form of this ganglioside and the presence of NeuAc on a 50-Kd membrane protein. NeuGc is the major determinant of the A antigen; specifically, [NeuGc]2GD3 is the major glycolipid form. The A antigen is also present on a 50-Kd membrane protein.

Abstract:
Small-molecule inhibition of hypoxia-inducible factor prolyl 4-hydroxylases (HIF-P4Hs) is being explored for the treatment of anemia. Previous studies have suggested that HIF-P4H-2 inhibition may also protect the heart from an ischemic insult.Hif-p4h-2gt/gtmice, which have 76 to 93% knockdown ofHif-p4h-2mRNA in endothelial cells, fibroblasts, and cardiomyocytes and normoxic stabilization of Hif-α, were subjected to ligation of the left anterior descending coronary artery (LAD). Hif-p4h-2 deficiency resulted in increased survival, better-preserved left ventricle (LV) systolic function, and a smaller infarct size. Surprisingly, a significantly larger area of the LV remained perfused during LAD ligation inHif-p4h-2gt/gthearts than in wild-type hearts. However, no difference was observed in collateral vessels, while the size of capillaries, but not their number, was significantly greater inHif-p4h-2gt/gthearts than in wild-type hearts.Hif-p4h-2gt/gtmice showed increased cardiac expression of endothelial Hif target genes for Tie-2, apelin, APJ, and endothelial nitric oxide (NO) synthase (eNOS) and increased serum NO concentrations. Remarkably, blockage of Tie-2 signaling was sufficient to normalize cardiac apelin and APJ expression and resulted in reversal of the enlarged-capillary phenotype and ischemic cardioprotection inHif-p4h-2gt/gthearts. Activation of the hypoxia response by HIF-P4H-2 inhibition in endothelial cells appears to be a major determinant of ischemic cardioprotection and justifies the exploration of systemic small-molecule HIF-P4H-2 inhibitors for ischemic heart disease.

Abstract:
Y2O3-stabilized ZrO2 (YSZ) has been the material of choice for thermal barrier coatings (TBCs) in the past decades, yet its phase decomposition limits its application above 1200 °C. In this study, Gd2O3 and Yb2O3 co-doped YSZ powders were produced, in which some amounts of monoclinic (m) phase were introduced into the cubic (c) phase matrix. XRD results showed that the fabricated powders obtained by a solid phase synthesis were composed of m and c phases, and hat the m phase content decreased in a sequence of 4Gd-2Yb-4Y, 2Gd-2Yb-6Y and 2Gd-4Yb-4Y powders. This indicated that Yb3+ is an excellent stabilizer in the ZrO2-based lattice, which could largely suppress the formation of the m phase. The m phase content in the powders was almost kept unchanged with heating at 1300 °C, which could provide a toughening effect to the ceramic. All the powders exhibited no obvious sintering at 1300 °C for 150 h. As compared to YSZ, the three fabricated ceramics had lower thermal conductivities, and they increased in a sequence of 4Gd-2Yb-4Y, 2Gd-4Yb-4Y and 2Gd-2Yb-6Y.

Abstract:
Simulation of CaMoO4-NaGd(MoO4)2 solid solutions by the method of interatomic potentials is carried out. It is shown that solid solutions exist in the entire range of compositions and are close to ideal. Dependences on the composition of the lattice parameters and volume of the unit cell, density, bulk modulus, enthalpy, vibrational entropy and heat capacity are obtained. Temperature dependences of heat capacity and vibrational entropy are constructed. Analysis of the local structure and its changes depending on the composition of the Ca1-xNax/2Gdx/2MoO4 solid solution showed that in solid solution the interatomic distances of Gd-O are on average 2.62% less, and Na-O is 3.15% greater than the distances of Ca-O. In general, this leads to an increase in the lattice parameters and volume of the unit cell during the formation of Ca1-xNax/2Gdx/2MoO4 solid solutions compared to CaMoO4. Keywords: simulation, molybdates, the solid solutions, local structure.

Abstract:
AbstractAn endoplasmic reticulum transmembrane prolyl 4-hydroxylase (P4H-TM) is able to hydroxylate the α subunit of the hypoxia-inducible factor (HIF) in vitro and in cultured cells, but nothing is known about its roles in mammalian erythropoiesis. We studied such roles here by administering a HIF-P4H inhibitor, FG-4497, to P4h-tm−/− mice. This caused larger increases in serum Epo concentration and kidney but not liver Hif-1α and Hif-2α protein and Epo mRNA levels than in wild-type mice, while the liver Hepcidin mRNA level was lower in the P4h-tm−/− mice than in the wild-type. Similar, but not identical, differences were also seen between FG-4497–treated Hif-p4h-2 hypomorphic (Hif-p4h-2gt/gt) and Hif-p4h-3−/− mice versus wild-type mice. FG-4497 administration increased hemoglobin and hematocrit values similarly in the P4h-tm−/− and wild-type mice, but caused higher increases in both values in the Hif-p4h-2gt/gt mice and in hematocrit value in the Hif-p4h-3−/− mice than in the wild-type. Hif-p4h-2gt/gt/P4h-tm−/− double gene-modified mice nevertheless had increased hemoglobin and hematocrit values without any FG-4497 administration, although no such abnormalities were seen in the Hif-p4h-2gt/gt or P4h-tm−/− mice. Our data thus indicate that P4H-TM plays a role in the regulation of EPO production, hepcidin expression, and erythropoiesis.

Abstract:
A newly developed Mg-2Gd-0.5Zr-xZn (x = 0.5, 1.0, 2.0, 3.0 wt %) alloy system exhibits significant strengthening by doping with Zn. In order to understand the strengthening mechanism, the microstructure, texture evolution, and mechanical properties of ultrahigh ductility Mg-2Gd-0.5Zr alloys with a Zn addition were systematically investigated. The addition of Zn results in the formation of Mg-Gd-Zn intermetallic compounds along grain boundaries, which encourages grain refinement during hot extrusion via the particle stimulated nucleation (PSN) mechanism. Furthermore, during texture sharpening the pole changes from <20 2 ¯ 1> to <01 1 ¯ 0>, which also occurred in the extruded alloys with Zn addition, which is unfavorable for the basal slip and tensile twinning. Mg-2Gd-0.5Zr-3Zn shows well-balanced strength and ductility with a tensile yield strength (YS) and ultimate tensile strength (UTS) of 285 and 314 MPa, accompanied by a high tensile elongation of 24%. They are superior to those of commercial AZ31. The enhanced strength is attributed to grain refinement, precipitation strengthening, and texture sharpening induced by alloying with Zn. The research result is also of great value to the development of low rare-earth, high strength, and high room temperature ductility magnesium alloy.

Abstract:
Abstract Erythrocytosis, an increase in absolute red cell mass, is mainly driven by erythropoietin, while hypoxia-inducible factor (HIF) regulates the expression of a number of genes involved in it, including erythropoietin. Mutations in HIF prolyl 4-hydroxylase 2 (HIF-P4H-2/PHD2/EGLN1), the major regulator of the stability of HIFα subunits, are found in familiar erythrocytosis, and large-spectrum conditional inactivation of HIF-P4H-2 in mice leads to severe erythrocytosis and premature death. Although bone marrow is the primary site for erythropoiesis, spleen retains a capability for extramedullary erythropoiesis. We studied HIF-P4H-2 hypomorphic mice (Hif-p4h-2gt/gt) which show slightly induced erythropoiesis only upon aging despite no increased erythropoietin levels. Spleen was identified as the site of extramedullary erythropoiesis in these mice. Hematopoietic stem cells (HSCs) from spleens of the Hif-p4h-2gt/gt mice showed increased growth of BFU-Es and the mice were protected against anemia by induced extramedullary erythropoiesis. HIF-1α and HIF-2α were stabilized in the spleens, while the Notch ligands and target Jag1, Jag2, Dll1 and Hes1 became downregulated upon aging dependent on HIF-2α. Inhibition of Notch signaling in wild-type spleen HSCs phenocopied the increased growth of BFU-Es in the Hif-p4h-2gt/gt mice. We conclude that HIFα stabilization can mediate non-erythropoietin-driven extramedullary erythropoiesis in the spleen via altered Notch signaling.
