Abstract:
Two-dimensional gel electrophoresis (2DE) is an important and well-established technical platform enabling extensive top-down proteomic analysis. However, the long-held but now largely outdated conventional concepts of 2DE have clearly impacted its application to in-depth investigations of proteomes at the level of protein species/proteoforms. It is time to popularize a new concept of 2DE for proteomics. With the development and enrichment of the proteome concept, any given “protein” is now recognized to consist of a series of proteoforms. Thus, it is the proteoform, rather than the canonical protein, that is the basic unit of a proteome, and each proteoform has a specific isoelectric point (pI) and relative mass (Mr). Accordingly, using 2DE, each proteoform can routinely be resolved and arrayed according to its different pI and Mr. Each detectable spot contains multiple proteoforms derived from the same gene, as well as from different genes. Proteoforms derived from the same gene are distributed into different spots in a 2DE pattern. High-resolution 2DE is thus actually an initial level of separation to address proteome complexity and is effectively a pre-fractionation method prior to analysis using mass spectrometry (MS). Furthermore, stable isotope-labeled 2DE coupled with high-sensitivity liquid chromatography-tandem MS (LC-MS/MS) has tremendous potential for the large-scale detection, identification, and quantification of the proteoforms that constitute proteomes.

Abstract:
Background: Myocardial contrast two-dimensional echocardiography (MC-2DE) is widely used to address alcohol septal ablation (ASA) in obstructive hypertrophic cardiomyopathy (HCM). Owing to its limited cut-planes, MC-2DE may inaccurately identify the contrast misplacement associated with an unsuccessful or complicated ASA outcome. Objective: The aim of this study was to assess the added value of myocardial contrast three-dimensional echocardiography (MC-3DE) compared with MC-2DE to identify the appropriate matching between the target septal zone (TSZ) and coronary artery branch for safe and long-term effective ASA in HCM patients. Methods: A consecutive series of 52 symptomatic obstructive HCM patients referred for isolated surgical myectomy (SM) was analyzed with MC-2DE and MC-3DE following injection of echocontrast into one or more septal branches. MC-2DE and MC-3DE patterns were categorized according to complete (Type 1) or incomplete (Type 2) TSZ covering, high-risk (Type 3) exceeding TSZ, or life-threatening outside TSZ distribution (Type 4). Results: MC-2DE per patient analysis showed a Type 1 pattern in 32 patients and Types 2–4 in the remaining 20 patients; subsequent MC-3DE analysis provided a re-phenotyping of MC-2DE findings in 22 of the 52 patients (42%), showing a high-risk Type 2 pattern in 17 of the 32 patients with Type 1, and a new life-threatening Type 4 in three patients with Type 2, respectively. All patients with MC-3DE Type 1 pattern underwent safe and effective ASA with a long-term uneventful follow-up, while the remaining patients underwent SM. Conclusions: Refining high risk or life-threatening contrast misplacement, MC-3DE is more accurate than conventional MC-2DE to target safe and long-term effective septal reduction with ASA in obstructive HCM patients referred for isolated SM.

Abstract:
Background.Left ventricular (LV) longitudinal deformation can be assessed with new echocardiographic techniques like triplane echocardiography (3PE) and four-dimensional echocardiography (4DE). We aimed to assess the feasibility, reproducibility, and agreement between these different speckle-tracking techniques for the assessment of longitudinal deformation.Methods. 101 consecutive subjects underwent echocardiographic examination. 2D cine loops from the apical views, a triplane view, and an LV 4D full volume were acquired in all subjects. LV longitudinal strain was obtained for each imaging modality.Results. 2DE analysis of LV strain was feasible in 90/101 subjects, 3PE strain in 89/101, and 4DE strain in 90/101. The mean value of 2DE and 3PE longitudinal strains was significantly higher with respect to 4DE. The relationship between 2DE and 3PE derived strains (r=0.782) was significantly higher (z=3.72,P<0.001) than that between 2DE and 4DE (r=0.429) and that between 3PE and 4DE (r=0.510;z=3.09,P=0.001). The mean bias between 2DE and 4DE strains was-6.61±7.31%while-6.42±6.81%between 3PE and 4DE strains; the bias between 2DE and 3PE strain was of0.21±4.16%. Intraobserver and interobserver variabilities were acceptable among the techniques.Conclusions. Echocardiographic techniques for the assessment of longitudinal deformation are not interchangeable, and further studies are needed to assess specific reference values.

Abstract:
Background Standard apical four-chamber and two-chamber views often maximize the long-axis of the left ventricle, resulting in artifactitious foreshortening of the left atrium (LA), which may overestimate LA longitudinal reservoir strain (LALS). We compared LALS values between 2D echocardiography (2DE) and 3D echocardiography (3DE) in healthy subjects to determine whether 2DE speckle tracking analysis overestimates the reference value of LALS. Methods and results In this study, 4 types of cohorts were included: 1. 105 normal subjects (retrospectively), 2. 53 patients with cardiovascular diseases (retrospectively), 3. 15 patients who received cardiac magnetic resonance (prospectively), and 4. 20 normal subjects (prospectively). LALS and LA length were measured using both 2DE and 3DE in 105 healthy subjects (median age: 42 years). Biplane LALS was measured in apical four- and two-chamber views using 2DE speckle tracking software, and 3DE LALS was measured using new 3DE LA strain software. To determine sensitivity, we also performed the same analysis in 53 patients with cardiovascular disease. The mean value of biplane LALS was 39.6%. LA length at both end-diastole (r = -0.43) and end-systole (r = -0.54) was negatively correlated with biplane LALS. Multivariate regression analysis revealed that both end-diastolic and end-systolic LA length had significant negative relationships with biplane LALS after adjusting for anthropometric and echocardiographic image quality parameters. 3DE LALS (23.7±7.6%) gave significantly lower values than 2DE LALS (39.5±12.0%, p<0.001) with a weak correlation (r = 0.33). LA length measured by 2DE was significantly shorter than that measured by 3DE. The same trend was observed in diseased patients. Conclusions Our results revealed that in 2DE, the LA cavity consistently appears longitudinally foreshortened in apical views, potentially overestimating LALS. 3DE may overcome this limitation.

Abstract:
This study intends to use 3D cardiac anatomical model automatic quantitative technique Heart Model (HM) to assess left ventricular systolic function in patients with myocardial infarction. Thirty patients with myocardial infarction (case group) and 32 healthy individuals (control group) were enrolled in this study.M-mode echocardiography (MME), two-dimensional echocardiography (2DE), 3D cardiac anatomical model (HM) and real-time 3D echocardiography (3DE) were used to collect echocardiographic images. Left ventricular systolic function parameters (LVEDV), left ventricular end-systolic volume (LVESV), and left ventricular ejection fraction (LVEF) were obtained from two post-surgical analysis of more than 5 years of experience. Compared to control group, MME, 2DE, HM, and 3DE were significantly higher in the LVEDV and LVESV cases and lower in the LVEF (P < 0 05). MME, 2DE, HM and 3DE were correlated and best correlation as found between HM and 3DE (P < 0 05). For the consistency of MME, 2DE, HM and 3DE, the best consistency was observed in the LVEF of HM and 3DE, and case group was lower than control (P < 0 05). In terms of repeatability of MME, 2DE, HM and 3DE, HM showed the best repeatability followed by MME. In comparison of the time of MME, 2DE, HM and 3DE, HM had shortest time (< 0 05). HM automatic quantitative technique can evaluate left ventricular systolic function in myocardial infarction patients, and it is simple, rapid and reproducible.

Abstract Introduction: Left ventricle (LV) volumes and ejection fraction (EF) determined with two-dimensional echocardiography (2DE) and three-dimensional echocardiography (3DE) show significant differences.The aim of this study is to determine the agreement of the measurements of LV volumes and EF with 2DE and 3DE in the general adult population, with preserved LV systolic function. Material and methods: In 52 subjects, older than 65 years, LV end-diastolic volume index (EDVi), end-systolic volume index (ESVi) and EF were measured with 2DE and 3DE according to the official recommendations, and reproducibility of both methods and their agreement were determined. Results: Intraclass correlation coefficient for intra-observer reproducibility in the measurement of EDVi, ESVi and EF with 2DE was 0.861, 0.891 and 0.917 respectively, whereas with 3DE 0.854, 0.893 and 0.913, respectively. The difference in the measurement of EDVi and ESVi was significant (p<0.001) whereas the measurement of EF was insignificant (p=0.153). The mean difference value EDVi and ESVi determined with 2DE and 3DE was 5.6+/-5.21 and 3.01+/-2.69 ml/m2 (p<0.001), and of EF 0.306+/-1.475%. Spearman’s correlation coefficient for EDV was 0.693, for ESV 0.763 and for EF 0.97. Conclusion: Larger LV volumes were measured in the adult population using 3DE compared to 2DE, but identical values for EF were obtained. This difference in the measured values could not be attributed to the largeness of the LV volume and EF itself.. 3DE demonstrated better intra-observer reproducibility for LV volumes and EF as a major parameter in many clinical decisions.

Abstract Assessing hemodynamics, especially central venous pressure (CVP), is essential in heart failure (HF). Right heart catheterization (RHC) is the gold-standard, but non-invasive methods are also needed. However, the role of 2-dimensional echocardiography (2DE) remains uncertain, and 3-dimensional echocardiography (3DE) is not always available. This study investigated standardized and breathing-corrected assessment of inferior vena cava (IVC) volume using echocardiography (2DE and 3DE) versus CVP determined invasively using RHC. Sixty consecutive HF patients were included (82% male, age 54 ± 11 years, New York Heart Association class 2.23 ± 0.8, ejection fraction 46 ± 18.4%, brain natriuretic peptide 696.93 ± 773.53 pg/mL). All patients underwent Swan-Ganz RHC followed by 2DE and 3DE, and IVC volume assessment. On 2DE, mean IVC size was 18.3 ± 5.5 mm and 13.8 ± 6 mm in the largest deflection and shortest distention, respectively. Mean CVP from RHC was 9.3 ± 5.3 mmHg. Neither 2DE nor 3DE showed acceptable correlation with invasively measured CVP; IVC volume acquisition showed optimal correlation with RHC CVP (0.64; 95% confidence interval 0.46–0.77), with better correlation when mitral valve early diastole E wave and right ventricular end-diastolic diameter were added. Using a CVP cut-point of 10 mmHg, receiver operating characteristic curve showed true positivity (specificity) of 0.90 and sensitivity of 62% for predicting CVP. A validation study confirmed these findings and verified the high predictive value of IVC volume assessment. Neither 2DE nor 3DE alone can reliably mirror CVP, but IVC volume acquisition using echocardiography allows non-invasive and adequate approximation of CVP. Correlation with invasively measured pressure was strongest when CVP is > 10 mmHg.

Context: Subclinical hypothyroidism (SHT) is associated with left ventricular (LV) remodeling. The LV mechanics has not been previously assessed by two- and three-dimensional (2DE and 3DE) speckle tracking imaging in the SHT patients. Objectives: The objective of the study was to investigate LV mechanics by 2DE and 3DE speckle tracking in the SHT patients and evaluate the influence of levothyroxine therapy on LV remodeling. Design: We conducted a prospective study. All SHT patients received levothyroxine therapy and were followed up for 1 year after the euthyroid state had been achieved. Setting: The study was performed at a university hospital. Patients: We included 54 untreated women with SHT and 40 healthy control women who were of similar age. Main Outcome Measures: The 2DE strain and strain rates, 3DE volumes, 3DE strain, and thyroid hormones levels were assessed. Results: The 2DE LV longitudinal and circumferential strain and systolic and early diastolic strain rates were significantly decreased in the SHT patients before therapy in comparison with the controls or the SHT patients after therapy. The 3DE LV cardiac output and ejection fraction were significantly reduced in the SHT patients at baseline compared with the controls or patients after 1 year of treatment. The 3DE LV longitudinal and radial strains were significantly lower in the SHT group before treatment in comparison with the controls or patients after therapy, whereas the 3DE LV circumferential and area strains gradually increased from untreated SHT patients, among the treated SHT patients, to the controls. Conclusion: SHT significantly affects LV deformation assessed by 2DE and 3DE speckle tracking. The improvement of LV mechanics after 1 year of levothyroxine treatment is significant but incomplete.

Manganese (Mn) is widely used in industry. However, its extensive applications have generated a great amount of manganese waste, which has become an ecological problem and has led to a decrease in natural resources. The use of microorganisms capable of accumulating Mn ions from contaminated ecosystems offers a potential alternative for the removal and recovery of this metal. The main aim of this work was an investigation of removal potential of Mn from soil by isolated bacterial. For this purpose, eleven bacterial strains were isolated from the soil from metallurgical waste heap in Upper Silesia, Poland. Strain named 2De with the highest Mn removal potential was selected and characterized taking into account its ability for Mn sorption and bioaccumulation from soil and medium containing manganese dioxide. Moreover, the protein profile of 2De strain before and after exposition to Mn was analyzed using SDS/PAGE technique. The 2De strain was identified as a Pseudomonas sp. The results revealed that this strain has an ability to grow at high Mn concentration and possesses an enhanced ability to remove it from the solution enriched with the soil or manganese dioxide via a biosorption mechanism. Moreover, changes in cellular protein expression of the isolated strain were observed. This study demonstrated that autochthonous 2De strain can be an effective tool to remove and recover Mn from contaminated soil.

The thesis presents the investigation on how to increase the sensitivity of the narrowed sensors of radiation. Also the thesis also deals with the attempts to analyze the influence of the microwave radiation on to the narrowed semiconductor formations and to reveal the physical nature of the observed effects as well as to determine the influence of structure of the samples on the detected signal magnitude. The work solves the following major tasks: the characteristics of the narrowed semiconductor heterostructures depending on the quality of the modulation layers and on the parameters of the semiconductor materials as well as the characteristics, depending on the selectively doped structure, on the conductivity of the highly doped semiconductor layer, and on the thickness of the separating layer, and the type of metallization of the gate. In order to achieve the goal there were produced and investigated narrowed different heterostructures (AlGaAs/GaAs, AlGaAs/InGaAs/GaAs) and n-GaAs structures. The thesis consists of four chapters, the final one is the generalization of the results. Taip pat disertacijoje siekiama ištirti mikrobangų spinduliuotės poveikį susiaurintiems puslaidininkiniams dariniams ir atskleisti stebimų efektų fizinę prigimtį bei nustatyti bandinių struktūros įtaką detektuojamo signalo dydžiui. Darbe sprendžiami tokie pagrindiniai uždaviniai: tiriamos įvairialyčių susiaurintų puslaidininkinių darinių savybės, priklausančios nuo darinių sluoksnių kokybės ir puslaidininkinių medžiagų parametrų bei analizuojamos savybės, priklausančios nuo stipriai legiruoto puslaidininkinio sluoksnio laidumo, nuo skiriamojo sluoksnio storio ir nuo sklendės pobūdžio metalizacijos. Abstract:
The polar lobe of Ilyanassa is asymmetrically partitioned into the D lineage of cells. Two of these cells, 3D and 4d, induce proper axial cell fate patterning in the embryo. Based on known embryological data in Ilyanassa, I hypothesized that Notch signaling would be required for this induction. I found that Notch signaling is required for cell fates induced by 4d and is temporally required well after 4d induction. Based on these results, I hypothesize that Notch signaling is involved in a reciprocal induction between the micromeres and the macromeres (endoderm) resulting in the maintenance of micromere fate induction and endoderm specification.Loss of the polar lobe results in the loss of cell fate induction by 3D/4d. Therefore, I hypothesized that proteins are asymmetrically bequeathed to the inducing D lineage cells by the polar lobe. To test this hypothesis, I compared global protein differences between two cell stage intact embryos, lobeless embryos, and isolated polar lobes by 2-Dimensional Electrophoresis analysis. I found several (12) quantitative differences between these samples including four spots enriched in the polar lobe isolates. I identified voltage-dependent anion-selective channel (VDAC) as one of the candidate proteins enriched in polar lobe isolates. I propose that VDAC is asymmetrically distributed by the polar lobe to the D cell and that it may function in D cell induction and mesendoderm fate specification.Lastly, I identify an acetylated tubulin antigen as a marker for cilia. I describe the pattern of cilia differentiation in the developing larvae that results in the formation of two ciliary bands, the prototroch and the metatroch, required for locomotion and feeding. These ciliary bands are conserved among annelid and mollusc larvae. Interestingly, the metatroch is derived from third quartet derivatives in the annelid Polygordius and from second quartet derivatives in the mollusc Crepidula. I provide evidence that the metatroch is derived from the first quartet derivatives in the mollusc Ilyanassa. Thus while the larval metatroch is conserved, its clonal origin is not. Based on these results, I provide support for the hypothesis that the metatroch is not homologous between annelids and molluscs or even among molluscs.

Darbe sprendžiami tokie pagrindiniai uždaviniai: tiriamos įvairialyčių susiaurintų puslaidininkinių darinių savybės, priklausančios nuo darinių sluoksnių kokybės ir puslaidininkinių medžiagų parametrų bei analizuojamos savybės, priklausančios nuo stipriai legiruoto puslaidininkinio sluoksnio laidumo, nuo skiriamojo sluoksnio storio ir nuo sklendės pobūdžio metalizacijos. Siekiant užsibrėžto tikslo, buvo gaminami ir tiriami susiaurinti skirtingi įvairialyčiai dariniai (AlGaAs/GaAs, AlGaAs/InGaAs/GaAs) ir n-GaAs dariniai. Disertaciją sudaro penki skyriai, kurių paskutinis – rezultatų apibendrinimas. Pirmajame (įvadiniame) skyriuje nagrinėjamas problemos aktualumas, formuluojamas darbo tikslas bei uždaviniai, aprašomas mokslinis darbo naujumas, pristatomi autoriaus pranešimai, disertacijos struktūra. Antrasis skyrius skirtas literatūros apžvalgai. Jame apžvelgiami elektromagnetinės spinduliuotės detektavimo principai, aptariamos šiluminės ir bigradientinės elektrovaros susidarymo priežastys, AlGaAs/GaAs įvairialytė sandūra, selektyvusis legiravimas bei puslaidininkinių prietaisų fizikinės galimybės. The thesis presents the investigation on how to increase the sensitivity of the narrowed sensors of radiation. Also the thesis also deals with the attempts to analyze the influence of the microwave radiation on to the narrowed semiconductor formations and to reveal the physical nature of the observed effects as well as to determine the influence of structure of the samples on the detected signal magnitude. The work solves the following major tasks: the characteristics of the narrowed semiconductor heterostructures depending on the quality of the modulation layers and on the parameters of the semiconductor materials as well as the characteristics, depending on the selectively doped structure, on the conductivity of the highly doped semiconductor layer, and on the thickness of the separating layer, and the type of metallization of the gate. In order to achieve the goal there were produced and investigated narrowed different heterostructures (AlGaAs/GaAs, AlGaAs/InGaAs/GaAs) and n-GaAs structures. Normally present in water, soil and waste water, Acinetobacter baumannii has become an important nosocomial pathogen, as causal agent of pneumonias, septicemias and urinary tract infections, among other complications in compromised patients from hospital’s intensive care units. One of its last acquired abilities is the resistance to colistin (polymixin E), the last therapeutic option for its infections. In this thesis, descriptive and quantitative differential expression proteomics is used in the study of acquired colistin resistance. As result of this research, 1,097 proteins belonging to the Acinetobacter genus have been identified by combined application of bidimensional gel electrophoresis (2DE), differential gel electrophoresis (DIGE), and peptide labeling with stable isobaric isotopes tags (iTRAQ). Analyses have been performed on the global expressed proteome of a reference, colistin-sensible strain (A. baumannii ATCC 19606) and, for comparative purposes, on a derived strain on which colistin resistance has been induced in vitro. The resistant phenotype shows reduced fitness, with significant differences in expression found in outer membrane proteins, membrane active transporters, diverse metabolic enzymes (fatty acids, citrate, phenylacetate, piruvate and nitrogen), proteins involved in stress response and biofilm formation, as well as in protein synthesis and folding pathways. The work has allowed to assess the strengths and weaknesses of the different techniques currently used in this type of proteomic analysis.
Acinetobacter baumannii, normalmente aislado en suelos y aguas (corrientes o residuales), se ha convertido en importante patógeno nosocomial, siendo agente causal de, entre otras complicaciones, neumonías, septicemias e infecciones del tracto urinario de pacientes comprometidos en unidades hospitalarias de cuidados intensivos. La más reciente de sus capacidades adquiridas es la resistencia a colistina (polimixina E), antibiótico peptídico considerado la última opción terapéutica en contextos clínicos. Esta tesis doctoral emplea la proteómica descriptiva y de expresión diferencial cuantitativa para investigar la resistencia adquirida por A. baumannii a dicho antibiótico. Los resultados han supuesto la identificación de 1.097 proteínas de Acinetobacter mediante el empleo combinado de electroforesis bidimensional convencional (2DE), 2DE diferencial (DIGE) y marcaje peptídico mediante isótopos isobáricos estables (iTRAQ). Los análisis se han realizado en el proteoma expresado por una cepa de referencia sensible a colistina (A. baumannii ATCC 19606), así como en una cepa derivada de ésta en la que se ha inducido, a efectos comparativos, resistencia a colistina in vitro. El fenotipo resistente manifestó reducida adaptabilidad biológica, encontrándose las principales diferencias en la estructura de la membrana externa, en la expresión de transportadores activos de membrana, en diversos enzimas metabólicos (ácidos grasos, citrato, fenilacetato, piruvato, nitrógeno) y de respuesta a condiciones de estrés, así como en la expresión de proteínas participantes en la formación de biopelículas y en el proceso de síntesis y plegamiento de proteínas. Además, el trabajo ha permitido evaluar los puntos fuertes y débiles de las técnicas empleadas actualmente en este tipo de análisis proteómicos.

Contemporary astrology, a thousand-year-old divinatory system, is the fruit of a mental state made of borrowings and adjustments. It is a practice of wich the legitimacy is contested, but also an alternative resort sometimes institutionaly integrated into our society. Astrology generates different discourses, customs and behaviours, and through these, it is a social reality which makes sens. The objectives of the present study is an analysis of close anthropology as appreciated from a distant and external point of view. Essentially, this research concentrates on aspects of astrology termed scholarly and its environment. It particularly focuses on the process of conversion to astrology - its nature, practices and effects. La réflexion, nourrie d’expériences personnelles et de plusieurs années de pratique professionnelle en France, émet l’hypothèse que vivre sur un campus est une expérience spatiale qui contribue à l’éducation de l’individu et qu’en conséquence, elle doit être appréhendée à l’échelle de l’homme. La recherche développe une vision de la conception de l’espace universitaire au XXIe siècle fondée sur la notion de milieu(x) et à partir de la dynamique du territoire. L’analyse de cas de campus japonais est support de cette réflexion. Within the field of Architecture and Environmental Design, and by applying Foucault’s “heterotopia” concept, this work examines the campus as a space that is different from other spaces. According to its defining principles, the American campus is a reference for highlighting major themes: community engagement, questions of Town and Gown, and nature’s role in university life. This reflection, nourished by personal experiences and many years of professional practice in France, leads to the hypothesis that living on a campus is a spatial experience that contributes to an individual’s education, and therefore, must be fully understood on a human scale. The research develops a vision of university space design in the 21st century, based on the notion of environmental surroundings and beginning with territorial dynamics. An analysis of the Japanese campus helps support this reflection. History serves as a privileged tool for understanding how places were shaped at the time of Heian (794-1185), Meiji (1868-1912), and Heisei (1989-)

Cette thèse analyse l’émergence d’une science de la catalyse en France dans la deuxième moitié du XXe siècle. Suite à la mise au point de nombreux procédés catalytiques durant les décennies 1930 à 1950, en lien avec la chimie du pétrole, la recherche académique est fortement encouragée par le Centre national de la recherche scientifique (CNRS). Jusqu’au milieu des années 1980, trois écoles de recherche fortement typées oeuvrent à la promotion de la catalyse comme discipline académique. L’Institut de recherches sur la catalyse (IRC), créé en 1958, dirigé par Marcel Prettre, et richement doté d’un équipement d’avant-garde, est reconnu comme leader sur les scènes nationale et internationale. Malgré l’hégémonie de l’IRC, l’équipe de l’Institut français du pétrole (IFP) et celle formée autour de Jean-Eugène Germain à Lille développent des approches alternatives et complémentaires. À partir des années 1970, les problématiques énergétiques d’abord, environnementales ensuite, contribuent à effacer les barrières entre les trois écoles. Derrière Raymond Maurel, et autour du mythe fondateur de Paul Sabatier, la communauté française dans son ensemble s’unit pour répondre aux mots d’ordre de « développement durable ». Un nouveau profil de la catalyse se dessine dans cette nouvelle phase. Au lieu d’une consolidation disciplinaire la catalyse se développe plutôt au carrefour des disciplines, au plus près des logiques industrielles et tournée vers les enjeux majeurs de la société. This dissertation describes the emergence of a science of catalysis in France in the second half of the twentieth century. Following various advances in catalytic technology between 1930 and 1950, and in relation with oil industries, academic research on catalysis was strongly supported by the Centre National de la Recherche Scientifique from 1944 onwards. Until the mid 1980s, three distinct research schools promoted catalysis as an academic discipline in France. The Institut de recherches sur la catalyse headed by Marcel Prettre, founded in 1958 and equipped with sophisticated, soon gained national and international recognitions. In spite of this hegemony, the Institut français du pétrole and the research team conducted by Jean-Eugène Germain in Lille developed alternative and complementary research programs. In the 1970s, the focus on energetic and later environmental issues contributed to blur the boundaries between the three research schools. Under the leadership of Raymond Maurel, a single French community gradually emerged, reunified around the issue of “sustainable development” and cemented by the founder myth of Paul Sabatier. A new profile of catalysis emerged over the past decades, which no longer belongs to the disciplinary regime of academic science. Abstract:
Pancreatic Ductal Adenocarcinoma (PDA), the most common pancreatic cancer, remains one of the most lethal tumor types, with extremely low survival rates due to late diagnosis and resistance to standard therapies. Expression of tumor-associated carbohydrate antigens such as SLeX enhances several tumor features like cell migration or metastasis, representing a good source of new therapeutic targets and biomarkers. In this work, it has been studied the effect of the inhibition of the expression of the main genes that code for the enzymes responsible for the final steps of SLeX biosynthesis (the α2,3-sialyltransferases). Silencing of ST3GAL3 and ST3GAL4 in two PDA cell lines led to a significant reduction in SLeX associated with a decrease in cell migration, invasion and adhesion to E-selectin, decreasing PDA metastatic potential. Also, using glycoproteomic techniques, we identified the glycoprotein MFAP4 carrying SLeX as potential PDA biomarker since it is only present in PDA tissue samples
El adenocarcinoma ductal pancreático o PDA, es el tipo más frecuente de cáncer de páncreas. El PDA es uno de los tumores más letales, con tasas de supervivencia extremadamente bajas, debido principalmente al diagnóstico tardío y su amplia resistencia a las terapias actuales. La expresión de antígenos carbohidratos asociados a tumores como el SLeX potencia varias características tumorales como la migración celular o metástasis, por lo cual representa una buena fuente de nuevas dianas terapéuticas y biomarcadores. En este trabajo, se ha estudiado el efecto de la inhibición de la expresión de los principales genes que codifican para las enzimas responsables de la biosíntesis de SLeX en sus etapas finales (las α2,3-sialiltransferasas). El silenciamiento de ST3GAL3 y ST3GAL4 en dos líneas celulares de PDA produjo a una reducción significativa en los niveles de SLeX. Este descenso se asoció con una disminución en la migración, invasión y adhesión celular a E-selectina, disminuyendo el potencial metastásico del PDA. Además, mediante técnicas glico-proteómicas, pudimos identificar la glicoproteína MFAP4 portadora de SLeX como potencial biomarcador de PDA, ya que esta glicoforma solo fue detectada en la cohorte de muestras de tejido de PDA

Le personnage de Merlin est né il y a fort longtemps à partir de chroniques galloises. Ses origines littéraires remontent à des textes du 12e siècle, d'abord en latin, puis en français, dans lesquels il devient le célèbre conseiller d'Arthur et acquiert une notoriété considérable auprès des lecteurs. Aujourd'hui, neuf siècles plus tard, le Merlin traditionnel est en pleine renaissance, alors que le Romantisme avait laissé paraître l'avoir oublié pour toujours. Le héros fait toujours rêver et devient le personnage principal de nombre d'oeuvres de la production culturelle francophone actuelle. Ce travail se propose d'analyser les rémanences contemporaines du personnage littéraire médiéval de Merlin autour de deux axes principaux : les origines et les pouvoirs. Dans un premier temps, un tableau représentant le portrait médiéval du héros est constitué d'après l'étude d'un corpus d'oeuvres faisant apparaître Merlin au Moyen Âge. Puis, sur la base de ce tableau dont la structure compose le plan de l'étude, les rémanences contemporaines du héros sont analysées de manière détaillée à l'intérieur d'un corpus d'éléments à la fois littéraires, cinématographiques, théâtraux, appartenant à l'univers de la bande dessinée ou à celui d'Internet. L'étude des origines (qui regroupe les thèmes de la conception et de la tradition sylvestre) révèle principalement un changement de statut du concepteur médiéval de Merlin, le diable, qui se trouve rationalisé, paganisé ou même supprimé. Cette modification se fait au profit d'une véritable mise en lumière de la tradition sylvestre et du motif de l'homme sauvage, qui apparaissaient déjà au Moyen Âge mais deviennent primordiaux dans le corpus contemporain. Une analyse détaillée de la dévalorisation du christianisme permet de mieux comprendre ces bouleversements narratologiques qui touchent également le thème des pouvoirs (regroupant la clairvoyance, l'emprise sur le temps et l'emprise sur l'espace). En plus d'être déchristianisés, ils se modernisent considérablement grâce aux techniques proposées par les différents médias et sont ainsi démystifiés, ce qui permet une identification efficace avec le lecteur-spectateur. Ainsi, Merlin devient accessible, il n'effraye plus comme au Moyen Âge où son origine diabolique en faisait un personnage douteux. Il se présente comme le porte-parole d'un retour à la nature, comme il l'avait déjà été dans les premiers textes médiévaux avant que son message ne se perde sous le poids de la domination chrétienne. Or, aujourd'hui plus que jamais, le monde a besoin d'exemples, de modèles en ce qui concerne le respect et l'amour de la nature dont l'avenir semble menacé. Merlin constitue donc le messager idéal d'une conception " naturiste " de la vie et du monde, ce que les auteurs contemporains ont bien compris. Par son appartenance au monde médiéval et son portrait, revisité par la production culturelle contemporaine, il nous attire, nous fait rêver, tout en nous rappelant notre propre être dans un rapport d'identification à lui-même

Abstract:
Este segundo Seminario Internacional de evaluación de la implementación del Acuerdo de Paz entre el Gobierno Nacional y las FARC se realizó en medio de una movilización de estudiantes y profesores de las universidades públicas que paralizó durante más de dos meses la educación superior estatal y que finalmente permitió un acuerdo sin antecedentes. Como estaba previsto, este Seminario se ocupó de atender, esencialmente el análisis de los avances y dificultades de la implementación de dicho Acuerdo que en 2018 sobrevivió en medio de aguas turbulentas. En efecto, durante el año de referencia, al tiempo que se realizaron elecciones parlamentarias y dos vueltas para la elección presidencial, en el país se realizó la consulta popular contra la corrupción. Aunque los once millones y medio de ciudadanos que participaron en dicha jornada democrática no fueron suficientes para superar el umbral, quedó claro que en la agenda pública nacional el tema es de prioridad inequívoca.
