Abstract:
Delsoline, a major alkaloid of Delphinium anthriscifolium Hance, has both a curare-like effect and a ganglion-blocking effect and is used to relieve muscle tension or hyperkinesia. A ultraperformance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method was established for the determination of delsoline in mouse blood, and the pharmacokinetics of delsoline after intravenous administration (1 mg/kg) and intragastric administration (9, 6, and 3 mg/kg) were studied. Gelsenicine served as an internal standard, and a UPLC BEH C18 chromatographic column was used. The mobile phase consisted of acetonitrile and 0.1% formic acid; the gradient elution flow rate was 0.4 mL/min. The MRM model was used for the quantitative analysis of delsoline m/z 468.3⟶108.1 and the internal standard m/z 327.1⟶296.1. Mouse blood samples were treated with acetonitrile precipitation to remove proteins. In the concentration range of 0.1–1000 ng/mL, delsoline in mouse blood showed a good linearity (r2 > 0.995), and the lower limit of quantitation was 0.1 ng/mL. The intraday precision relative standard deviation (RSD) was below 14%, and the interday precision RSD was below 15%. The accuracy ranged between 94.3% and 110.1%, the average recovery was above 90.8%, and the matrix effect ranged between 97.0% and 102.5%. The UPLC-MS/MS method was sensitive, rapid, and selective in the study of pharmacokinetics of delsoline. The absolute bioavailability of delsoline was 20.9%.

Abstract:
In health, food carbohydrate is stored as glycogen in muscle and liver, preventing a deleterious rise in osmotically active plasma glucose after eating. Glycogen concentrations increase sequentially after each meal to peak in the evening, and fall to fasting levels thereafter. Skeletal muscle accounts for the larger part of this diurnal buffering capacity with liver also contributing. The effectiveness of this diurnal mechanism has not been previously studied in Type 2 diabetes. We have quantified the changes in muscle and liver glycogen concentration with 13C magnetic resonance spectroscopy at 3.0 T before and after three meals consumed at 4 h intervals. We studied 40 (25 males; 15 females) well-controlled Type 2 diabetes subjects on metformin only (HbA1c (glycated haemoglobin) 6.4±0.07% or 47±0.8 mmol/mol) and 14 (8 males; 6 females) glucose-tolerant controls matched for age, weight and body mass index (BMI). Muscle glycogen concentration increased by 17% after day-long eating in the control group (68.1±4.8 to 79.7±4.2 mmol/l; P=0.006), and this change inversely correlated with homoeostatic model assessment of insulin resistance [HOMA-IR] (r=−0.56; P=0.02). There was no change in muscle glycogen in the Type 2 diabetes group after day-long eating (68.3±2.6 to 67.1±2.0 mmol/mol; P=0.62). Liver glycogen rose similarly in normal control (325.9±25.0 to 388.1±30.3 mmol/l; P=0.005) and Type 2 diabetes groups (296.1±16.0 to 350.5±6.7 mmol/l; P<0.0001). In early Type 2 diabetes, the major physiological mechanism for skeletal muscle postprandial glycogen storage is completely inactive. This is directly related to insulin resistance, although liver glycogen storage is normal.

Abstract:
In a double-blind comparative study, 135 depressed patients were treated in 20 centers. Inclusion diagnoses were typical depressions with single episode (296.2), several episodes (296.3), depressive neurosis (300.4), and adjustment disorder with depressed mood (309.0) in accordance with DSM-III-R. The dosage was 3 x 300 mg hypericum extract LI 160 or 3 x 25 mg imipramine daily. The treatment lasted for 6 weeks. Main assessment criteria were the Hamilton Depression Scale (HAMD), the Depression Scale according to von Zerssen (D-S) and the Clinical Global Impressions (CGI). In both treatment groups, a parallel reduction of the Hamilton score from 20.2 to 8.8 (LI 160, n = 67) or from 19.4 to 10.7 (imipramine, n = 68), and the transformed D-S point values from 39.6 to 27.2 (LI 160) and 39.0 to 29.2 (imipramine) were found. The analysis of CGI revealed comparable results in both treatment groups. Clinically relevant changes of the safety parameters were not found. In the LI 160 group fewer and milder side effects were found as compared to imipramine.

Telah berhasil dibuat Dye Sensitized Solar Cell dengan menggunakan pewarna dari ekstrak biji kakao dengan variasi lama perendaman semikonduktor dalam pewarna. Hasil pengujian XRD menunjukkan bahwa Kristal TiO2 berada difase anatase dengan ukuran Kristal 25.99228 nm, 12.94122 nm dan 12.94136 nm yang dihasilkan dengan metode Debye Scherrer. Karakterisasi UV-Vis dye pada panjang gelombang 300 nm - 800 nm menunjukkan bahwa pewarna dapat bekerja pada cahaya UV dan cahaya tampak sedangkan untuk hasil UV-Vis pada semikonduktor TiO2 hanya memiliki puncak absorbansi pada cahaya UV pada panjang gelombang 296.0 nm yaitu 2.5938 a.u. Nilai tegangan dan arus yang diperoleh akan diakumulasi untuk mengetahui efisiensi dari rangkaian DSSC. Masing –masing efisiensi() yang dihasilkan untuk lama perendaman 6 Jam, 12 Jam, 18 Jam dan 24 Jam secara berurut adalah 0,0197%; 0.0258%; 0.0397% dan 0.0426%.

An efficient and regioselective synthetic reaction friendly to the environment has been described to synthesize various derivatives of pyrazolo[1,5-a]quinozolinone. Condensation of aminopyrazole (4a-m) with formylated dimedone (3) in the presence of KHSO4, under ultrasonic irradiation furnished 2/3-substituted 8,8-dimethyl-8,9-dihydropyrazolo[1,5-a]quinazolin-6(7H)-one (6a-m). This is a clean reaction, giving excellent yields with short reaction time. The structures were elucidated with the help of spectral and analytical data. X-ray crystallographic studies of a model compound 6a ascertained its structural configuration, crystal data for C12H12BrN3O (M =294.152 g/mol): Triclinic, space group P-1 (no. 2), a = 5.872(4) Å, b = 10.870(8) Å, c = 19.523(15) Å, α = 90.013(10)°, β = 90.009(11)°, γ = 93.838(11)°, V = 1243.3(16) Å3, Z = 4, T = 296.15 K, μ(Mo Kα) = 3.293 mm-1, Dcalc = 1.571 g/cm3, 37271 reflections measured (4.18° ≤ 2Θ ≤ 52.7°), 5073 unique (Rint = 0.2404, Rsigma = 0.2366) which were used in all calculations. The final R1 was 0.0596 (I≥2σ(I)) and wR2 was 0.1759 (all data).

Abstract:
Background: Crescentic IgA nephropathy (CreIgAN) has a poor prognosis despite aggressive immunosuppressive therapy. The efficacy of plasma exchange (PE) in CreIgAN is not well defined. Methods: Twelve patients with severe CreIgAN who received PE as addition to routine immunosuppressive therapy, followed for more than 6 months, were involved. Twelve matched historical controls who received immunosuppressive therapy alone were selected by propensity score matching. Renal survival, plasma IgA-IgG complex and active complement products were assessed. Results: Nine men and 3 women received a median of 7 PE courses (range 5-10). Their baseline urine protein excretion rate was 5.8 (4.5-8.7) g/day, and their serum creatinine level was 705.3 ± 296.4 μmol/l. During a mean follow-up of 15.6 months (6-51 months), 6 of the 12 PE group patients were free of dialysis, while all the control patients were dialysis dependent (6 of 12 vs. 0 of 12, p = 0.014). In the PE group, dialysis had to be restarted for 1 patient owing to the development of severe pneumonia and pulmonary failure. PE was associated with a higher kidney survival rate (log rank test, p = 0.026) during follow-up. It also significantly decreased plasma IgA-IgG complex levels (pre-PE: 85.3 ± 25.9% vs. post-PE: 38.4 ± 12.4%, p < 0.001) and plasma and urinary active complement product levels, including C3a, C5a and soluble C5b-9. The latter levels remained low until the last follow-up. Conclusion: This study indicated that PE could increase renal recovery rates in severe CreIgAN.

The final R1 was 0.0423 (I >2σ(I)) and wR2 was 0.1145 (all data). The Density functional theory optimized molecular geometries in TMQ agree closely with those obtained from crystallographic studies. The Highest Occupied Molecular Orbital (HOMO) and Lowest Unoccupied Molecular Orbital (LUMO) energy levels and energy gap were calculated by experimental (UV absorption & Cyclic voltammetry) and theoretical studies in two different solvents. The natural bond orbital analysis was performed to understand the molecular interaction on the basis of stability of molecule arising from hyper-conjugative interaction and charge delocalization. Hirshfeld surface and their related fingerprint plots enabled the identification of significant intermolecular interaction. The molecular electrostatic potential analysis provides the visual image of the chemically active sites and comparable reaction of atoms.

Abstract:
The storage experiment examined the effects of storage temperatures, seed moisture contents (SMC), and storage durations (SD) on the viability and vigour of sunflower seeds. Certified sunflower seeds were evaluated for seed germination, seedling vigour, and fungal infection. A 3x3x6 factorial experiment in Completely Randomized Design was set out in four replicates. The three factors were temperature (5, 20, 35 ºC), SMC (6,8,10%), and SD (15, 30, 45, 60, 75, 90Days After Storage, DAS). Before storage (0 DAS), the initial germination percentage (GP=94%), Germination index (GI=322), Seedling vigour index (SVI=2118), and fungal infection incidence (FII=52%) were determined and used as control. The lowest GP of 67%, GI (269.2), SVI (1295), and FII (78.0%), was found in seeds stored at a temperature of 35 ºC and SMC of 10% at 90DAS. Upon the interaction effect at the end of 90DAS, the temperature of 5 ºC with SMC of 6% followed by the temperature of 20 ºC with SMC of 6% recorded the greatest GP of 87.5 and 87.0%, GI of 304.3 and 296.8 SVI of 1910 and 1877 and fewer fungi incidence (%) of 61.5 and 63 respectively. The germination percentage was above the minimum certification standards of 85%, as per the Tanzania Official Seed Certification Institute. Conclusively, our results discovered that the optimum storage conditions-Temperature of 5 and 20 ºC with SMC of 6 and 8% could be suitable for maintaining the viability and vigour of sunflower seeds throughout the storage duration (90 DAS).
