Abstract:
Harry Stewart Wragge (23 November 1929–31 July 2023), Director of the Telecom (later Telstra) Research Laboratories (TRL) in Melbourne from 1985 to 1992, was a leading Australian research engineer. He catalysed the evolution of the public switched telephone network in Australia from its analogue, electromechanical form in the 1950s and 1960s to the digital, computer-controlled circuit-switched network of the 1980s and 1990s. His own research work was famous for his IST (integrated switching and transmission) project, building the first computer-controlled, integrated digital switch in the world to handle commercial telephone traffic (from 1974 to 1978). He also provided major support to telecommunications research at several Australian universities. His many contributions were recognized by honours from the Australian Government, Melbourne and Monash universities, the Pearcey Foundation, the Telecommunications Society of Australia and the City of Frankston.

Abstract:
Abstract:

"Recent safety and efficacy findings from a phase 1b/2 open-label combination study of ASP-1929 photoimmunotherapy with anti-PD-1 therapy in EGFR-expressing advanced head and neck squamous cell carcinoma (HNSCC)." Journal of Clinical Oncology 42, no. 16_suppl (June 1, 2024): 6083. http://dx.doi.org/10.1200/jco.2024.42.16_suppl.6083.

Abstract:
6083 Background: In the challenging landscape of advanced HNSCC, patients (pts) have overall low survival and high locoregional (LR) recurrence rates. While LR progression is a key contributor to morbidity and mortality, LR control remains low despite aggressive therapy including surgery, radiation, chemotherapy, and immunotherapy. ASP-1929 photoimmunotherapy (PIT) combines EGFR-targeting cetuximab, with a light-activatable dye, IRDye 700DX, and a laser light for localized drug activation. This novel approach has demonstrated rapid and selective tumor necrosis in preclinical studies, and a manageable safety profile in HNSCC early clinical trials. Phase I/II and preclinical data indicate a potential synergy in anti-tumor activity of PIT when combined with anti-PD-1 therapy. Here we present updated interim evaluation findings based on a recent data cut from this study initially presented at the 2023 American Head and Neck Society meeting. Methods: A phase 1b/2 open-label study of pts with recurrent locally advanced (rLA) and/or metastatic (m) HNSCC evaluating the safety and efficacy of ASP-1929 PIT in combination with anti-PD-1 (pembrolizumab) therapy (NCT04305795). Pts with measurable disease by modified RECIST 1.1, at least 1 lesion accessible to light illumination, combined positive score (CPS) ≥1, and who were not candidate for LR therapy could be eligible for the study. Treatment included ASP-1929 PIT (infusion day 8, target tumors illuminated 24±4 hours later) plus anti-PD-1 (days 1&22) during a 6-week cycle for up to 24 months. Primary objectives: safety/tolerability; objective response rate (ORR). Secondary objectives: overall survival (OS); progression-free survival (PFS); duration of response (DOR). Planned sample size: 26 HNSCC pts. Results: Of the 19 r/m HNSCC pts enrolled, 18 pts received both study therapies (PIT-evaluable). Data cut-off was August 31, 2023 (previously reported data cut-off: October 4, 2022). Efficacy results updated for the 18 PIT-evaluable pts: ORR of 33.3% (95% CI 13.3-59.0), including 4 CRs (22.2%) and 2 PRs (11.1%). Median OS not reached; 24-month OS rate estimate was 52.4% (95%CI 25.9-73.4). No change in median PFS, 2.9 months (95%CI 1.4-14.6). Median DOR was not reached at data cut-off (range 2.8+ to 18.0+ months). Safety data remained consistent with the data previously presented. Most common serious adverse reactions were dysphagia (10.5%) and tongue edema (10.5%). There were two grade 4 events: laryngeal edema (PIT-related) and tumor hemorrhage due to advanced disease, and no fatal events. Conclusions: ASP-1929 PIT in combination with anti-PD-1 therapy was generally well tolerated. Initial data demonstrate promising overall survival and response rates with this combination therapy in pts with rLA and/or m HNSCC lacking LR treatment options. Clinical trial information: NCT04305795 .

Abstract:
About two-thirds of the population in Iceland lives in the Reykjavik capital area (RCA), which is close to active volcanoes and seismic zones. In the period 1900–2019, a total of 53 earthquakes of Mw≥5.0 struck in these zones. The two largest events in the Reykjanes Peninsula, Mw6.36 and Mw6.12, occurred in 1929 and 1968, respectively. Both events were less than 20 km from the outskirts of the RCA. Late in the year 2020, the seismicity on the peninsula greatly increased due to magma intrusion and volcanic activity, which has so far resulted in three eruptions, in 2021, 2022, and 2023, and six earthquakes of Mw≥5.0. Based on historical and geological data, the ongoing activity is probably the initial phase of an active period ahead that could continue for many decades, and has the potential to trigger larger earthquakes like those in 1929 and 1968. Further east, in the South Iceland Seismic Zone, two earthquakes of Mw6.52 and 6.44 struck in June 2000, and in May 2008, a Mw6.31 earthquake occurred. In both cases, around 5000 buildings were affected. Insurance loss data from these events have been used to develop empirical vulnerability models for low-rise buildings. In this study, the loss data are used to calibrate seismic vulnerability models in terms of the source-site distance. For a given magnitude scenario, this provides a simpler representation of seismic vulnerability and is useful for emergency planning and disaster management. These models are also used to compute different types of scenario risk maps for the RCA for a repeat of the 1929 earthquake.

Abstract:
BackgroundIn contrast to ORAL Surveillance data, reports from real world data did not show difference in rates of major adverse cardiovascular events (MACE) and malignancy between tofacitinib and biologic disease modifying anti-rheumatic drugs (b-DMARDs) [1-3].ObjectivesInvestigate the rate of MACE and malignancy post janus kinase inhibitors (JAK-i) initiation in Qatar population.MethodsAll JAK-i prescriptions were captured electronically from the database of the pharmacy of Hamad Medical Corporation (HMC) from January 2018 to November 2022. Patients with non-dispensed prescriptions were excluded from the analysis. Electronic medical records of patients who received any class of JAK-i were reviewed retrospectively to identify demographic and clinical characteristics, starting and stopping date of JAK-i, MACE and malignancy events post JAK-i initiation.Results219 JAK-i course therapies (tofacitinib 193, upadacitinib 14 and baricitinib 12) were identified from the database of the pharmacy of HMC. Six patients were excluded from the analysis as they were prescribed with JAK-i but their prescriptions were not dispensed. Five patients received two classes of JAK-i due to treatment failure. Mean (standard deviation [SD]) time of JAK-i exposure was 25 (17) months.A total of 208 patients included in the analysis with mean (SD) age at JAK-i initiation was 45.9 (14) years. Females were 72% of the cohort. JAK-i was prescribed for the following indications: rheumatoid arthritis 129 (60%), psoriatic arthritis 28 (13.1%), alopecia 29 (13.6%), spondyloarthropathy (SPA) 16 (7.5%), inflammatory bowel disease (IBD) 8 (3.8%), other rheumatic diseases 5 (2.3%). Two patients were having IBD with peripheral and axial SPA involvement. In patients with rheumatic diseases, JAK-i was received in 102 (55%) b-DMARDs naïve patients, post one b-DMARDs in 38 (20%) and post ≥ 2 in 46 (25%) patients. The comorbidity profile (hypertension, dyslipidemia, diabetes mellitus, and coronary artery disease) was 27%,25%, 23%, and 5.6%, respectively.New cancer diagnosis (follicular thyroid) was confirmed in one patient post tofacitinib and one patient developed lymphoma relapse after tofacitinib initiation. One ischemic cerebrovascular accident (CVA) identified post tofacitinib initiation. This patient was in atrial fibrillation when CVA occurred. No venous thromboembolism events reported from this cohort.ConclusionMACE and malignancy rates are very low in JAK-i users in Qatar population. Comparator group is needed for further evaluation the association of the use of JAK-i and risk of MACE and malignancy.References[1]Ytterberg SR, Bhatt DL, Mikuls TR, Koch GG, Fleischmann R, Rivas JL, et al. Abstract:
Finding a long-term solution to the world's growing dependence on conventional energy sources and the depletion of fossil fuels is what worries scientists the most right now. An alternative that appears promise is biodiesel. It was investigated how to increase the generation of biodiesel from leftover fish processors' oil using sodium methoxide and sulphonated eggshells as a catalyst, based on Box-Behnken design. The greatest yield of 96.81% was achieved at 1:12 oil to methanol ratio, 65 °C reaction temperature, 90 minutes, and 0.5 w/w% catalysts loading. The variables constant, methanol to oil ratio, reaction temperature, reaction time, reaction temperature* reaction temperature, and methanol to oil ratio*catalyst load all had a significant effect on the biodiesel production, according to the response surface regression. The model does a good job of accounting for the link between biodiesel and process variables. Thus, residual fish processor oil might be effectively converted into more advantageous and environmentally benign biodiesel fuel.

Abstract:
Abstract. The Budyko framework is a simple and effective tool for estimating the water balance of watersheds. Quantification of the watershed-characteristic-related parameter (Pw) is critical for accurate water balance simulations with the Budyko framework. However, there is no universal method for calculating Pw as the interactions between hydrologic, climatic, and watershed characteristic factors differ greatly across watersheds. To fill this research gap, this study introduced the principle of hydrologically similar groups into the Budyko framework for quantifying the Pw of watersheds in similar environments. We first classified the 366 selected watersheds worldwide into six hydrologically similar groups based on watershed attributes, including climate, soil, and vegetation. Results show that soil moisture (SM) and fractional vegetation cover (FVC) are two controlling factors of the Pw in each group. The SM exhibits a power-law relationship with the Pw values, with increasing SM leading to higher Pw values in dry watersheds (SM ≤ 20 mm) and lower Pw values in humid watersheds (SM > 20 mm). Additionally, the FVC shows to be linearly correlated with the Pw values in most hydrologically similar groups, except in that group with moist soil and no strong rainfall seasonality (SM >20 mm and seasonal index (SI) ≤ 0.4). Multiple non-linear regression models between Pw and the controlling factors (SM and FVC) were developed to individually estimate the Pw of six hydrologically similar groups. Cross-validations using the bootstrap sampling method (R2=0.63) and validations of time-series Global Runoff Data Centre (GRDC) data (R2=0.89) both indicate that the proposed models perform satisfactorily in estimating the Pw parameter in the Budyko framework. Overall, this study is a new attempt to quantify the unknown Pw in the Budyko framework using the method for hydrologically similar groups. The results will be helpful in improving the applicability of the Budyko framework for estimating the annual runoff of watersheds in diverse climates and with different characteristics.

Abstract:
Francis Haxo has been known to us as a thoughtful, patient, and dedicated scientist of the highest order. As a genuine scholar, he meticulously mulled over ideas for years before publishing his research. One of us (GG) knew him through three other friends: Barbara B. Prézelin, 1948-2021; David (Dave) C. Fork, 1929-2020; Beatrice (Beazy) M. Sweeney, 1914-1989; additionally, Anitra Thorhaug (AT) knew him as she was the last student of Larry Blinks, with whom Haxo had worked with. Francis Haxo was generous to his students, but he took a long time to finalize and submit their papers—to ensure that all was well. We provide here a glimpse of his research after a brief note on his personal life—remembering him more than a decade after his death. He was an unsung hero and he deserves to be remembered by algal, marine, and plant biologists throughout the world. A thorough and detailed memorial to Francis Haxo and his research, will soon be published by Anitra Thorhaug, Graeme Berlyn, and Synnøve Liaaen-Jensen (2023).
