Abstract:
The genetic control of resistance to Cucurbit aphid-borne yellows virus (CABYV; genus Polerovirus, family Luteoviridae) in the TGR-1551 melon accession was studied through agroinoculation of a genetic family obtained from the cross between this accession and the susceptible Spanish cultivar ‘Bola de Oro’. Segregation analyses were consistent with the hypothesis that one dominant gene and at least two more modifier genes confer resistance; one of these additional genes is likely present in the susceptible parent ‘Bola de Oro’. Local and systemic accumulation of the virus was analyzed in a time course experiment, showing that TGR-1551 resistance was expressed systemically as a significant reduction of virus accumulation compared with susceptible controls, but not locally in agroinoculated cotyledons. In aphid transmission experiments, CABYV inoculation by aphids was significantly reduced in TGR-1551 plants, although the virus was acquired at a similar rate from TGR-1551 as from susceptible plants. Results of feeding behavior studies using the DC electrical penetration graph technique suggested that viruliferous aphids can salivate and feed from the phloem of TGR-1551 plants and that the observed reduction in virus transmission efficiency is not related to reduced salivation by Aphis gossypii in phloem sieve elements. Since the virus is able to accumulate to normal levels in agroinoculated tissues, our results suggest that resistance of TGR-1551 plants to CABYV is related to impairment of virus movement or translocation after it reaches the phloem sieve elements.

Abstract:
ABSTRACTAtypical enteropathogenicEscherichia coli(aEPEC) strains are diarrheal pathogens that lack bundle-forming pilus production but possess the virulence-associated locus of enterocyte effacement. aEPEC strain 1551-2 produces localized adherence (LA) on HeLa cells; however, its isogenic intimin (eae) mutant produces a diffuse-adherence (DA) pattern. In this study, we aimed to identify the DA-associated adhesin of the 1551-2eaemutant. Electron microscopy of 1551-2 identified rigid rod-like pili composed of an 18-kDa protein, which was identified as the major pilin subunit of type 1 pilus (T1P) by mass spectrometry analysis. Deletion offimAin 1551-2 affected biofilm formation but had no effect on adherence properties. Analysis of secreted proteins in supernatants of this strain identified a 150-kDa protein corresponding to SslE, a type 2 secreted protein that was recently reported to be involved in biofilm formation of rabbit and human EPEC strains. However, neither adherence nor biofilm formation was affected in a 1551-2sslEmutant. We then investigated the role of the EspA filament associated with the type 3 secretion system (T3SS) in DA by generating a doubleeae espAmutant. This strain was no longer adherent, strongly suggesting that the T3SS translocon is the DA adhesin. In agreement with these results, specific anti-EspA antibodies blocked adherence of the 1551-2eaemutant. Our data support a role for intimin in LA, for the T3SS translocon in DA, and for T1P in biofilm formation, all of which may act in concert to facilitate host intestinal colonization by aEPEC strains.

Abstract:
Forty-four accessions of Cucumis melo L. and related wild species were tested for reaction to a yellowing disease, incited by the recently identified cucurbit yellowing stunting disorder virus (CYSDV), under natural and controlled-inoculation conditions. The C. melo TGR-1551 accession and one Cucumis metuliferus Naud. accession were asymptomatic. The segregation ratios obtained following controlled inoculations of the family produced by crossing TGR-1551 with the susceptible Spanish cv. Piel de Sapo revealed that the resistance to CYSDV in TGR-1551 is conditioned by a dominant allele at one locus. The name Cucurbit yellow stunting and symbol Cys is proposed for this locus.

Abstract:
Abstract Handling of hazardous materials in occupational settings can lead to worker exposure by inhalation. Dustiness testing, such as the EN 15051-2 rotating drum tester, provides suppliers and users of bulk materials with information on the potential for dust emission from hazardous substances when handled or processed in the workplace. This tester agitates powder samples in a rotating drum and the resulting dust is collected and sized by drawing it through two reticulated metal foams. Two approaches were used to evaluate the performance of the metal foams. Firstly, the performance of the metal foams was measured in a calm air chamber using a polydisperse aerosol of glass particles and assessed against the EN 481 respirable convention. Secondly, the performance of the metal foam for the respirable fraction was compared using the rotating drum dustiness test, with that of a cyclone set-up, using four polydisperse glass powders of different size distribution and dustiness potential. In general, for the respirable fraction, the tests demonstrated a conservative oversampling by the current EN 15051-2 metal foam set-up in comparison with the EN 481 convention. This study also showed the importance of sealing the circumference of the metal foams when testing highly dusty powders. A direct comparison of the respirable dustiness fraction measured by the current EN 15051-2 metal foams set-up and by a cyclone set-up, showed broad agreement. However, the cyclone gave lower values except where foam clogging occurred. The research also highlighted suggested improvements to the EN 15051-2 standard. © Crown Copyright (2022)

Abstract:
Resistance to Watermelon mosaic virus (WMV) in melon (Cucumis melo L.) accession TGR-1551 is characterized by a significant reduction in virus titer, and is inherited as a recessive, loss-of-susceptibility allele. We measured virus RNA accumulation in TGR-1551 plants and a susceptible control (‘Tendral’) by real-time quantitative polymerase chain reaction, and also profiled the expression of 17,443 unigenes represented on a melon microarray over a 15-day time course. The virus accumulated to higher levels in cotyledons of the resistant variety up to 9 days postinoculation (dpi) but, thereafter, levels increased in the susceptible variety while those in the resistant variety declined. Microarray experiments looking at the early response to infection (1 and 3 dpi), as well as responses after 7 and 15 dpi, revealed more profound transcriptomic changes in resistant plants than susceptible ones. The gene expression profiles revealed deep and extensive transcriptome remodeling in TGR-1551 plants, often involving genes with pathogen response functions. Overall, our data suggested that resistance to WMV in TGR-1551 melon plants is associated with a defense response, which contrasts with the recessive nature of the resistance trait.

Abstract:
A three-leaf fragment of an unidentified London pewterer's business records, covering some three months of the year 1551, has been found among the archives of the Worshipful Company of Pewterers and is here transcribed together with a discussion of its contents. It provides an inventory of the pewterer's stock, describes the alloying and casting of over a ton of tin to produce a wide range of pewter flatware, and itemizes the sales and loans of pewterware to named customers. These include other pewterers and Alderman Richard Lambert. The manuscript provides a unique insight into a sixteenth century pewterer's workshop practices. Speculation into the identity of the pewterer has proved inconclusive.

Abstract:
The paper investigates the effects on building indoor air quality (IAQ) resulting from the choice of different design ventilation rates. A reference residential building was analysed by means of the multizone modelling software CONTAM, by monitoring the concentration of two pollutants: occupant-generated carbon dioxide (CO2) and total volatile organic compounds (TVOC) from indoor sources. A demand-controlled ventilation strategy based on building occupancy was implemented and users' presence schedules were defined. Specifically, the evolution of indoor pollutant concentrations was investigated when the design ventilation rates, recommended by two IAQ-related standards (the Italian UNI 10339 and the European EN 15251), were implemented through a mechanical ventilation system. Different results regarding the IAQ level were achieved accordingly to the dissimilar ventilation rates. After a statistical analysis on the distributions of CO2 and TVOC concentrations, EN 15251 outcomes showed overall better results of the analysed statistical metrics, i.e. prevalence, sensitivity and accuracy. Indeed, the EN 15251 design airflows led to indoor conditions that were more often classified either in the correct air quality class or in a higher class. Finally, a better alignment between the national and European IAQ standards is recommended, especially in terms of airflows, air quality classes and pollutant limit values.

Abstract:
Jan Orzelski was actively involved in the political life of the Republic of the Nobles as a member of a regional assembly, a deputy and next a senator. He recorded the history of his family in the work entitled Annales domus Orzelsciae. It was edited by Adam Tytus Działyński on the basis of a manuscript stored in the Kórnik Library together with a family diary Kopia pobożnej pamięci imci Elżbiety Orzelskiej. Annales… consists of two parts: a comprehensive introduction and a chronicle containing annual entries regarding the most important events in Jan Orzelski’s family in the years 1589–1611 (that period being extended to 1618 by adding the diary Kopia pobożnej pamięci…). Much focus in the first part of the Annales... was placed on Stefan Batory’s military campaigns to conquer Polotsk (1579), Velikiye Luki (1580) and Pskov (1581), in which Jan Orzelski took part as a cavalry captain. The author’s intention, however, was not to describe those military campaigns in detail but to present “only some memorable issues”. The Annales... depicts, first of all, the origins of the “family from Orle”, the history and the characteristics of the family members in the male line. The author included his biography as well.

Abstract:

Abstract:
ABSTRACT Multiwavelength observations of pulsar emission properties are powerful means to constrain their magnetospheric activity and magnetic topology. Usually a star centred magnetic dipole model is invoked to explain the main characteristics of this radiation. However, in some particular pulsars where observational constraints exist, such simplified models are unable to predict salient features of their multiwavelength emission. This paper aims to carefully model the radio and X-ray emission of PSR J1136+1551 with an off-centred magnetic dipole to reconcile both wavelength measurements. We simultaneously fit the radio pulse profile with its polarization and the thermal X-ray emission from the polar cap hotspots of PSR J1136+1551. We are able to pin down the parameters of the non-dipolar geometry (which we have assumed to be an offset dipole) and the viewing angle, meanwhile accounting for the time lag between X-ray and radio emission. Our model fits the data if the off-centred magnetic dipole lies about 20 per cent below the neutron star surface. We also expect very asymmetric polar cap shapes and sizes, implying non-antipodal and non-identical thermal emission from the hotspots. We conclude that a non-dipolar surface magnetic field is an essential feature to explain the multiwavelength aspects of PSR J1136+1551 and other similar pulsars.

Abstract:
Abstract The present contribution focuses on three previously unedited letters from the Louvain professor of Latin, Petrus Nannius (Nanninck; 1496–1557), to his patron Antoine Perrenot de Granvelle (1517–1586). The letters, now kept in Madrid at the Biblioteca Nacional de España and Real Biblioteca del Palacio Real, date from November 1545, April 1546, and January 1551. They reveal novel insights with regard to the professor’s biography, most notably that by 1551, through his patron’s mediation, he enjoyed a prebend from the Dendermonde Chapter and was most likely appointed canon there. By means of these new snippets of information, I reassess and further define the patronus-cliens relationship between Nannius and Granvelle, after which I edit the letters critically for the first time.

Abstract:
Transcriptional regulation requires co-ordinated action of transcription factors, co-activator complexes and general transcription factors to access specific loci in the dense chromatin structure. In the present study we demonstrate that the transcriptional co-regulator SPBP [stromelysin-1 PDGF (platelet-derived growth factor)-responsive element binding protein] contains two independent chromatin-binding domains, the SPBP-(1551–1666) region and the C-terminal extended PHD [ePHD/ADD (extended plant homeodomain/ATRX-DNMT3-DNMT3L)] domain. The region 1551–1666 is a novel core nucleosome-interaction domain located adjacent to the AT-hook motif in the DNA-binding domain. This novel nucleosome-binding region is critically important for proper localization of SPBP in the cell nucleus. The ePHD/ADD domain associates with nucleosomes in a histone tail-dependent manner, and has significant impact on the dynamic interaction between SPBP and chromatin. Furthermore, SPBP and its homologue RAI1 (retinoic-acid-inducible protein 1), are strongly enriched on chromatin in interphase HeLa cells, and both proteins display low nuclear mobility. RAI1 contains a region with homology to the novel nucleosome-binding region SPBP-(1551–1666) and an ePHD/ADD domain with ability to bind nucleosomes. These results indicate that the transcriptional co-regulator SPBP and its homologue RAI1 implicated in Smith–Magenis syndrome and Potocki–Lupski syndrome both belong to the expanding family of chromatin-binding proteins containing several domains involved in specific chromatin interactions.

Abstract:
Chronic autoimmune thyroiditis (CAT) is the most common thyroid disorder in the pediatric population. Ultrasound evaluation may suggest the diagnosis. Additionally, shear-wave elastography (SWE) proved to be a valuable additional diagnosis tool in adults with CAT by assessing thyroid stiffness (TS). This study aims to assess its use also in detecting children with CAT. The study group consisted of 50 children with confirmed diagnosis of CAT, who were compared to the control group, consisting of 50 children with no thyroid pathology and with an adult group of 50 subjects with CAT. The evaluation included, besides bioimmunochemical evaluation, also thyroid ultrasound evaluation and elastography measurements in the same session (Aixplorer Mach 30, Supersonic imagine, France). The mean TS values were significantly lower for children in the CAT group compared to adults with CAT (15.51 ± 4.76 kPa vs. 20.96 ± 6.31 kPa; p < 0.0001) and higher compared to the healthy aged matched controls (15.51 ± 4.76 kPa vs. 10.41 ± 2.01 kPa; p < 0.0001). SWE elastography definitely seems a promising technique in the evaluation of children with autoimmune thyroid pathology.

Abstract:
Introduction: The World Health Organization (WHO) strategy for leprosy control from 2021 to 2030 focuses on interrupting transmission, reducing autochthonous cases to zero, and using a safe and effective vaccine and chemoprophylaxis. In 2020, 127,396 new cases were registered in the world, 19,195 new cases in the Americas, and, of these, 17,979 cases in Brazil, about 93.66% of the total in the Americas. Brazil is classified as a country with a high burden of the disease, occupying the 2nd place in the world, behind only India (WHO, 2020). Análise do período de 2010-2015 em publicação recente9, apresenta as seguintes cidades no estado do Pará nas quais se observou maiores taxas de incidência (detecção): Marituba, Belém, Marabá, Parauapebas e Altamira [9]. Material and method: This is an analytical retrospective study carried out in a database - Epi-Info resulting from records of Surveillance and Seroprevalence actions in five endemic municipalities for leprosy located in the Southeast and West of Pará. The following variables were analyzed: age, sex, Clinical Classification, vaccination status with BCG, and the result of the search for IgM antibodies against PGL-1 of Mycobacterium leprae by the “In house” ELISA technique. Results: We evaluated 1551 records examined in the laboratory from 2014 to 2016, which were classified into 123 Multibacillary -MB patients (123/1551 = 7.93%); 71 Paucibacillary-PB patients (71/1551 = 4.57%); 451 Intradomicilliary Consanguineous Contacts - CCOSI (451/1551 = 29.07%) and 906 Non Consanguineous Contacts - CNCOS (906/1551 = 58.41%). 57 MB patients (13.47%), 13 PB patients (3.07%), 133 CCOSI (31.44%) and 220 CNCOS (52.00%) were positive for PGL-1. The correlation of the Classification with the vaccination status showed 57 MB patients without any BCG (57/125 = 45.6%) and only 3 patients with two doses of BCG (3/125 = 2.4%); 17 PB patients without any dose of BCG (17/69 = 24.63%); 80 CCOSI without any BCG (80/455 = 17.58%) and 171 CNCOS (171/906 = 18.87%). The odds ratio (OR) in the analysis between unvaccinated MB patients compared to CCOSI was statistically significant (OR = 14.25; p ˂ 0.0001). The study shows the importance of using the BCG vaccine in healthy contacts of patients with leprosy, as it shows the probability of unvaccinated individuals being 14.25 times more likely to become ill with Multibacillary forms compared to CCOSI. In addition, the BCG vaccine has been in use for 80 years and is the only vaccine that we can use in leprosy control programs. Conclusion: Although the leprosy epidemiological data analyzed recently (2010 - 2015) show a downward trend in the main indicators in Pará, such as the detection of new cases and prevalence, the endemic municipalities are still classified as hyperendemic for the population under 15 years of age and This proves that Surveillance is essential, as well as BCG vaccination according to the Ministry of Health Standards.
