Abstract:
Background: MALT1 (Mucosa-associated lymphoid tissue lymphoma translocation protein 1) is a component of the MALT1-BCL10-CARD11 complex downstream from the Bruton Tyrosine Kinase (BTK) on the B-cell receptor signaling pathway. MALT1 is a key mediator of nuclear factor kappa B (NF-κB) signaling, which is the main driver of a subset of B-cell lymphomas. MALT1 is considered a potential therapeutic target for several subtypes of non-Hodgkin B-cell lymphomas and chronic lymphocytic leukemia (CLL), including tumors with acquired BTK inhibitor (BTKi) resistance. Constitutive activation of the NF-κB is a molecular hallmark of activated B cell-like diffuse large B cell lymphoma (ABC-DLBCL), and MALT1 may have utility as a treatment option for ABC-DLBCL. Previously, we described the discovery of novel MALT1 inhibitors with anti-proliferative effects in non-Hodgkin B-cell lymphoma cells and the strong anti-tumor activity of our MALT1 inhibitors across multiple tumor models as well as combination potential with agents including standard-of-care (ref 1, 2). SGR-1505 is an oral potent small molecule allosteric inhibitor of MALT1 that inhibits MALT1 enzymatic activity and demonstrates anti-proliferative activity in ABC-DLBCL cell lines, both BTKi-sensitive (OCI-LY10) and BTKi-resistant (OCI-LY3). When administered as a single agent and in combination with the approved Bruton's tyrosine kinase (BTK) inhibitor, ibrutinib, SGR-1505 demonstrated tumorostatic and regressive antitumor activity in ABC-DLBCL cell line-derived xenograft and patient-derived xenograft models. These data suggest that SGR-1505-mediated MALT1 inhibition has therapeutic potential for patients with selected B-cell lymphomas. Here we further characterized SGR-1505, in a series of in vitro and ex vivo assays, as well as RNA-seq analysis to examine changes in gene expression from in vivo tumor samples. We also compared SGR-1505 with a competitor Phase I candidate, JNJ-67856633 (JNJ-6633) (ref 3, 4). Results: SGR1505 potency and downstream effects were evaluated in a series of biochemical and cell based assays. SGR-1505 showed excellent potency in the biochemical assay and strong anti-proliferative effects on ABC-DLBCL cells. SGR-1505 was more potent than JNJ-6633 in all assays tested (Table 1). These results are also consistent with the result from a human primary T-cell based assay, where SGR-1505 showed at least ten-fold better potency than JNJ-6633. RNA-seq analysis was conducted to examine changes in gene expression from in vivo tumor samples. Greater modulation of BIOCARTA NF-kB pathway genes was seen with SGR-1505 compared to JNJ-6633, as measured by mean absolute change in gene expression. At 6 hr and later timepoints, we also observed a trend of increases in genes related to cell cycle pathways, such as cell cycle, DNA damage, and apoptosis. SGR-1505 is being evaluated in the SGR-1505-102 phase 1 study, which is an ongoing first-in-human, single center, dose escalation study to evaluate the safety, tolerability, PK and PD of SGR-1505 tablets in healthy participants (ACTRN12623000358640p). Preliminary data showed changes in target engagement markers at concentrations predicted by the in vitro and ex vivo assays, consistent with MALT1 protease inhibition. Conclusions: SGR-1505, a MALT1 protease small molecule inhibitor, consistently demonstrated better potency in in vitro and ex vivo assays when compared to the clinical-stage JNJ-6633 compound and greater effects on NF-kB pathway gene expression in in vivo tumor samples based on RNA-seq analysis. Changes in biological pathways mediated by MALT1 were also observed at relevant doses in the ongoing SGR-1505 healthy volunteer study. Currently, a phase 1 clinical trial in patients with mature B cell neoplasms is also ongoing (NCT05544019). The data presented suggests SGR-1505 has a potential best-in-class profile and supports advancing the ongoing clinical development of SGR-1505.

Study Design and Methods: SGR-1505-101 (NCT05544019) is a phase 1, multicenter trial of SGR-1505 as monotherapy in subjects with mature B-cell malignancies (figure 1). At present, the study is open to accrual at multiple investigative sites in the US with plans to expand to Europe. The primary objective is to evaluate safety and tolerability of SGR-1505 as monotherapy and to identify the maximum tolerated dose (MTD) and/or recommended dose (RD). Secondary objectives are to evaluate the pharmacokinetic (PK) profile, food effect, drug-drug interaction, and preliminary anti-tumor activity of SGR-1505. Key inclusion criteria are: history of mature B-cell malignancy (including aggressive and indolent B-cell lymphomas, Waldenström macroglobulinemia, and CLL); measurable or detectable disease according to the applicable disease-specific classification system (Lugano, iwCLL, WWM6); Eastern Cooperative Oncology Group (ECOG) performance status of ≤ 2. Patients with indolent B-cell lymphomas or CLL must have an indication for treatment and not require immediate cytoreductive therapy. Subjects with symptomatic or active CNS involvement, and other conditions or laboratory findings placing them at increased risk to the use of an investigational drug are excluded. SGR-1505 is initially dose-escalated using an accelerated titration design in cohorts of 1-6 subjects, and at higher dose levels using a conventional 3+3 design.

Abstract:
ABSTRACT Staphylococcus saprophyticus is a common cause of uncomplicated urinary tract infections in women. S. saprophyticus strain ATCC 15305 carries two staphylococcal cassette chromosome genetic elements, SCC15305RM and SCC15305cap. The SCC15305cap element carries 13 open reading frames (ORFs) involved in capsular polysaccharide (CP) biosynthesis, and its G+C content (26.7%) is lower than the average G+C content (33.2%) for the whole genome. S. saprophyticus strain ATCC 15305 capD, capL, and capK (capDSsp , capLSsp , and capKSsp ) are homologous to genes encoding UDP-FucNAc biosynthesis, and gtaB and capISsp show homology to genes involved in UDP-glucuronic acid synthesis. S. saprophyticus ATCC 15305 CP, visualized by immunoelectron microscopy, was extracted and purified using anionic-exchange and size exclusion chromatography. Analysis of the purified CP by 1H and 13C nuclear magnetic resonance (NMR) spectroscopy and gas-liquid chromatography revealed two types of branched tetrasaccharide repeating units composed of the following: \batchmode \documentclass[fleqn,10pt,legalpaper]{article} \usepackage{amssymb} \usepackage{amsfonts} \usepackage{amsmath} \pagestyle{empty} \begin{document} \[\begin{array}{c}\mathbf{-4)-{\beta}-Glc}-\mathbf{(1-3)-Sug}-\mathbf{(1-4)-{\beta}-GlcA}-(\mathbf{1-}\\\mathbf{{\vert}}\\\mathbf{{\beta}-GlcNAc}-\mathbf{(1-2)}\end{array}\] \end{document} Sug represents two stereoisomers of 2-acetamido-2,6-dideoxy-hexos-4-ulose residues, one of which has an arabino configuration. The encapsulated ATCC 15305 strain was resistant to complement-mediated opsonophagocytic killing by human neutrophils, whereas the acapsular mutant C1 was susceptible. None of 14 clinical isolates reacted with antibodies to the ATCC 15305 CP. However, 11 of the 14 S. saprophyticus isolates were phenotypically encapsulated based on their resistance to complement-mediated opsonophagocytic killing and their failure to hemagglutinate when cultivated aerobically. Ten of the 14 clinical strains carried homologues of the conserved staphylococcal capD gene or the S. saprophyticus gtaB gene, or both. Our results suggest that some strains of S. saprophyticus are encapsulated and that more than one capsular serotype exists.

Abstract:
A cold-tolerant strain of the mycoparasite Fusarium proliferatum was isolated following UV mutagenesis of the G6 strain, which is a biocontrol agent of grape downy mildew. The isolated strain (designated 1505) exhibited radial growth two to threefold that of the parent strain when grown at 13°C, which is generally suboptimal for growth of Fusarium spp., but desirable for its host, Plasmopara viticola. This rapid growth was correlated with improved biological control of P. viticola, determined by a detached-leaf assay. Even though radial growth of strain 1505 at higher temperatures was slower than that of G6 and the strain failed to conidiate, there was no reduction in biocontrol efficacy. Significantly higher levels of extracellular β-glucosidase and endo-1,4-β-glucanase activity were measured in the culture filtrate of strain 1505 relative to that of strain G6. A DNA-mediated transformation procedure that included the introduction of antibiotic resistance and a GUS reporter gene system was adapted for F. proliferatum. Using the GUS-engineered strains, we demonstrated that both G6 and 1505 exhibit the characteristic coiling and penetration of host structures.

Abstract:
Abstract Two case studies were conducted in the Shennan mining area of Shaanxi Province, China to evaluate the surrounding rock deformation and stress evolution in pre-driven longwall recovery rooms. These studies mainly monitored the surrounding rock deformation and coal pillar stress in the recovery rooms of the N1206 panel of 2−2 coal seam at Ningtiaota Coal Mine and the 15205 panel of 5−2 coal seam at Hongliulin Coal Mine. The monitoring results showed that the surrounding rock deformation of the main recovery room and the coal pillar stress in the N1206 and 15205 panels began to increase significantly when the face was 36 m and 42 m away from the terminal line, respectively. After the face entered the main recovery room, the maximum roof-to-floor convergence in the N1206 and 15205 panels was 348.03 mm and 771.24 mm, respectively, and the coal pillar stresses increased more than 5 MPa and 7 MPa, respectively. In addition, analysis of the periodic weighting data showed that the main roof break position of the N1206 and 15205 panels after the longwall face entered the main recovery room was − 3.8 m and − 8.2 m, respectively. This research shows that when the main roof breaks above the coal pillar, the surrounding rock deformation of the main recovery room and the coal pillar stress increase sharply. The last weighting is the key factor affecting the stability of the main recovery room and the coal pillar; main roof breaks at disadvantageous positions are the main cause of the support crushing accidents.

Abstract:
The aim of this paper is to offer as full a discussion as possible of the best-known portrait of Henry VII. Information from a variety of sources elucidates the context in which the portrait was painted and the nature of its iconography; in particular, evidence is presented to show that the red rose was a symbol of great personal significance to Henry. With regard to the portrayal of Henry himself it is suggested that his face conforms essentially to an approved ‘pattern’ that had been recently reworked and updated, and that the frequently expressed modern view of the portrait as representing a mean and crafty character results from a misinterpretation of the artist's intentions. In a final section attention is given to the question of the identity of the artist, whose name remains unknown.

Sleman Regency has many attractions that trigger urbanization which can be seen from the phenomenon of land-use change. This phenomenon occurs in Sleman Regency particularly in the area of food security and sustainability which is spreading in various districts such as Moyudan, Minggir, Sayegan, Godean, Mlati, and Tempel. The phenomenon of land change must be managed and controlled because the areas have an important role for food security in Sleman Regency. The method used for the research is descriptive method. Survey is a part of descriptive method which used to determine the existing conditions of land use in the research object areas. Whereas for land-use changes in the area of food security and sustainability in Sleman Regency from 2012 to 2018 was obtained with the method of land-use map overlay obtained from Quickbird imagery in those years.Land-use change in the area of food security and sustainability of Sleman Regency reaches 57.33 km2 or 33.93% from the total of research study area. Land-use change is dominated by the diversions of rice fields to forests that reach 15.05 km2. The driving factor for this phenomenon is the decreasing annual productivity of rice field.

Although the Wellcome Collection of Hebraica does not constitute one of the larger and more distinguished collections of oriental material in the Wellcome Institute, it nevertheless comprises a number of important manuscripts along with early printed books representative of several sixteenth and seventeenth-century Hebrew presses. One of these is a fragment of a larger work printed in Constantinople in 1505 at the press of David and Samuel Nahmias. It is the second earliest example of printing in Turkey, the first also coming from the press of the Nahmias brothers.

Abstract:
IntroductionAging and menopause as a medico-social determinant of female gender influence on mental health. Female gender in the period of menopause is associated with the end of fertility that affects adaptation to changed conditions, decreases intellectual and physical possibilities, narrows role positions, leads to non-psychotic mental disorders (NPMD).ObjectiveTo identify influence of the age, menopause, female sex, sociodemographic parameters on hysterical symptoms in structure of NPMD.MaterialIn the borderline states department, 93 female patients with hysterical manifestations in structure of NPMD were treated.MethodsPsychopathological, clinical-dynamic, clinical-catamnestic, psychometric, psychological and statistical.ResultsIn 100%, the hysterical symptoms in the puberty leveled at reproductive age, intensified in climax: 72.04% - pre-menopause (P < 0.05), meno-, postmenopause - 13.98% each. Sociodemographic characteristics (marital status, social status, place of living, education, family composition, family relations) were not interrelated with frequency of hysterical manifestations in the structure of NPMD (P > 0.05). There were more married (68.82%) than divorced (15.05%), widowed (10.75%), single (5.38%); more working (70.97%) than pensioners (22.58%), unemployed (6.45%); more living with husband (51.61%) than with husband and children (17.21%), living alone (15.05%), with adult children (16.13%); more townswomen (80.65%) than villagers (19.35%); education more often secondary (51.61%), high (45.16%) than elementary (3.23%); disharmonic family relations more often (70.97%) than harmonic (13.98%), indifferent (15.05%).ConclusionsAge, menopause, female sex are reliably (P < 0.05) connected with intensification of hysterical symptoms in the structure of NPMD in difference from sociodemographic parameters not associated with frequency of hysterical manifestations.Disclosure of interestThe authors have not supplied their declaration of competing interest.

Abstract:
Niniejszy artykuł stanowi próbę odtworzenia struktury dworu Elżbiety Jagiellonki (1482–1517), najmłodszej córki Kazimierza IV Jagiellończyka i Elżbiety Rakuszanki, funkcjonującego w latach 1505–1515.
