Abstract:
Abstract Interphotoreceptor retinoid-binding protein (IRBP), a retinal-specific Ag, induces experimental autoimmune uveitis in a variety of animals. We have previously shown that sequence 1169-1191 of bovine IRBP is the immunodominant epitope of this protein in Lewis rats and is highly immunogenic and uveitogenic in these rats. The active site of peptide 1169-1191 was determined by testing its truncated forms. The shortest peptide to be immunologically active was found to be 1182-1190 (WEGVGVVPD). To determine the role of individual residues of this sequence, we have tested the immunologic activities of nine analogs of peptide 1181-1191, in which each of residues 1182-1190 was substituted with alanine (A). The tested activities included the capacity to induce experimental autoimmune uveitis and cellular responses in immunized rats, as well as the capability to stimulate lymphocytes sensitized against IRBP or the parent peptide 1181-1191. Analogs that did not stimulate these lymphocytes were also tested for their capacity to competitively inhibit the proliferative response to 1181-1191. Analogs A(1184), A(1186), and A(1187) resembled 1181-1191 in their activities, whereas the other analogs exhibited remarkably reduced activities, with several patterns being noticed. Analog A(1182) was inactive in all tests. Analog A(1190) was very weakly uveitogenic and non-immunogenic, but it stimulated lymphocytes sensitized against IRBP or 1181-1191 when added at exceedingly high concentrations. Analogs A(1183) and A(1185) resembled A(1190) in being weakly uveitogenic and A(1185) was also found to be poorly immunogenic. In addition, relatively high concentrations of A(1183) and A(1185) were needed to stimulate lymphocytes sensitized against IRBP or 1181-1191. However, a different pattern of activities was exhibited by analogs A(1188) and A(1189). These peptides were uveitogenic and immunogenic, but failed to stimulate lymphocytes sensitized to IRBP or 1181-1191. Furthermore, A(1188) and A(1189), but not A(1182), also inhibited the response to 1181-1191 of a cell line specific toward this parent peptide. The data are interpreted to show that residues 1188 and 1189 are involved in the interaction of the peptide with the TCR, whereas residues 1182 and 1190 and, perhaps, 1183 and 1185, are pivotal for the binding of peptide 1181-1190 to the MHC molecules on APC.

Nascido em Assis, 1181 ou 1182, filho de Pietro di Bernadone e Jeanne di Bernardone, o jovem Giovanni di Pietro di Bernadone, apelidado de Francesco por seu pai, notabilizou-se, entre outras coisas, por sua forma radical de vida. Sua trajetória se encontra, principalmente, em seus testamentos e nas fontes franciscanas. Francesco é conhecido por revolucionar a Igreja, sem, no entanto, afastar-se dela. Munido de um pensamento de unidade e fraternidade, conseguiu, de forma rara, transcender o tempo, até os dias atuais. O pensamento de Francisco colaborou visão de mundo voltada à unidade dos povos, da diplomacia, meio ambiente e cultura de paz. Para escrita deste artigo foram utilizados os pressupostos teóricos-metodológicos da História Cultural, e para contribuir ao entendimento do homem de Assis, a bibliografia conta com autores como Foucault (1989), Le Goff (2005a – 2005b), Duby (1978, 2017), Franco (2001), além de Fassini (2004), entre outros.

Abstract:
Summary The Law of Beaumont, established by a charter issued in 1182, was granted over a period of about 150 years for up to 700 villages in present-day France, Belgium, Luxembourg and Germany. It seemingly was to provide incentives to settle or stay in such villages. Based on an analysis of the Latin text (provided with a German translation) the paper questions that the focus of the charter was accurately described in older literature and common handbook entries. In those, the impression is given that the focus was on liberation from serfdom, tax relief and municipal self-government. The paper concludes that a high degree of self-government was indeed an outstanding feature, although it remains unclear whether the citizens originally had the right to elect mayor and jurors. Particularly favourable tax burdens could not be substantiated. There is no mentioning at all of any liberation from serfdom and burdens associated with it. The focus of the charter is on a precise definition of tax burdens and of penalties for crimes and their distribution, and on rules of legal process. Legal certainty in these regards must have been the core of its attractiveness both for citizens and lords and explain its popularity.

Abstract Long non-coding RNA FAM230B and microRNA (miR-1182) have been characterized as critical players in cancer biology, while their roles in colorectal cancer (CRC) are unclear. We predicted that they could interact with each other and therefore explored the interaction between them in CRC. CRC and paired non-tumor tissue samples were collected from 60 CRC patients, and the expression of FAM230B and miR-1182 (premature and mature) in these samples was analyzed with RT-qPCR. The direct interaction between FAM230B and premature miR-1182 was analyzed with RNA-RNA pull-down assay, and the subcellular location of FAM230B was detected with subcellular fractionation assay. The interaction between FAM230B and miR-1182 was explored with overexpression assay, and their roles in regulating CRC cell proliferation, viability, and colony formation were assessed by BrdU assay, MTT assay, and colony formation assay, respectively. We found that FAM230B and premature miR-1182 were highly upregulated in CRC, while mature miR-1182 was downregulated in CRC. FAM230B was detected in both nucleus and cytoplasm, and it directly interacted with miR-1182. FAM230B overexpression increased the expression levels of premature miR-1182 but decreased the expression levels of mature miR-1182 in CRC cells. FAM230B promoted CRC cell proliferation, increased cell viability, accelerated colony formation, and suppressed the role of miR-1182 in inhibiting CRC cell proliferation. In conclusion, FAM230B is upregulated in CRC and it suppresses the maturation of miR-1182 to promote tumor growth.

ABSTRACT In the recent years, incidence of the leafhopper, Amrasca devastans (Distant) in cotton has been prevalent from vegetative to reproductive phase of crop growth. The loss in seed cotton yield due to leaf hopper is accounted to 390 kg ha-1 and 330 kg ha-1. In India 45 per cent of the pesticides are applied to cotton. Hence, development of a resistant / tolerant cultivar is the need of the hour to to reduce three ‘ R ’ viz., Resistance, Resurgence and Residues in biota. Screening of twenty six cotton accessions against the leafhopper, A. devastans (Distant) was carried out by following ICCC grades. The susceptiblity of cotton entries varied significantly. Based on the resistance index the entries were grouped under five categories as, Highly resistant- KC 2, SVPR 2; Resistant-TKH 1128: Intermediate- MCU 5, MCU 10, NISD 2, TKH 1143,TKH 1175: Susceptible- TKH 1789, TKH 1173, TKH 1174, TKH 1178, TKH 1179, TKH 1185, TKH 1186, TKH 1209, TKH 1225, TKH 1233 and Highly susceptible- ICMF 20, LRA 5166, TKH 1133, TKH 1172, TKH 1176, TKH 1182, TKH 1197, TKH 1198. Studies on the mechanism of resistance revealed that leafhoppers discriminated cotton entries for orientation and settling and feeding. Highly susceptible entiries were preferred for settling and feeding. Varieties less preferred for settling were less preferred for oviposition too. Since antixenosis and antibiosis were considered for mechanism of resistance various plant parameters viz., Plant height, internodal length, petiole length, leaf area, thickness of leaf, trichome density on the ventral surface of the leaves, hair length, hair density on mid-vein, distance between bases of hairs in mid vein, chlorophyll content, moisture content and protein content were estimated and correlated with leafhopper ovipostion and damage caused. Plant height, internodal length, trichome density on the ventral surface of the leaves, hair length, hair density on mid-vein had negative association with leafhopper damage and oviposition by leafhopper; leaf area, leaf thickness and protein content had no significant association. Susceptible entries had less chlorophyll and more moisture.

The Drosophila melanogaster cell line 1182-4, which constitutively lacks centrioles, was established many years ago from haploid embryos laid by females homozygous for the maternal haploid (mh) mutation. This was the first clear example of animal cells regularly dividing in the absence of this organelle. However, the cause of the acentriolar nature of the 1182-4 cell line remained unclear and could not be clearly assigned to a particular genetic event. Here, we detail historically the longstanding mystery of the lack of centrioles in this Drosophila cell line. Recent advances, such as the characterization of the mh gene and the genomic analysis of 1182-4 cells, allow now a better understanding of the physiology of these cells. By combining these new data, we propose three reasonable hypotheses of the genesis of this remarkable phenotype.

В статье исследуются взаимосвязи между тремя памятниками историописания: «Деяниями датчан» Саксона Грамматика (ок. 1208/1206 г.), анонимной «Историей похода датчан в Иерусалим» (до 1202 г.) и привилегией Лундскому кафедральному собору (1186 г.). Текстологический анализ показал, что во всех этих источниках датский король Кнут VI (1182–1202) сопоставляется со своими предками-соименниками. Каждое такое сопоставление служит прославлению датского короля и легитимации его власти. Статья призвана показать, что на первый взгляд независимые друг от друга тексты в действительности образуют единое идеологическое поле.

Abstract:
Abstract Background Influenza and respiratory syncytial virus (RSV) are recognized as important causes of hospital-acquired infection. Increased use of multiplex molecular diagnostic testing is shedding light on the incidence of other hospital-associated respiratory virus infections (HA-RVI). However, the incidence and clinical impact of HA-RVI are not well understood. Methods We identified hospitalized patients admitted between July 1, 2017 and June 30, 2018 who were clinically tested to diagnose respiratory virus infections. HA-RVI were defined as respiratory virus positivity beginning more than 48 hours after hospital admission. The clinical outcomes of HA-RVI were compared with respiratory virus infections that were not considered hospital-associated (non-HA-RVI). Results Respiratory virus testing was performed on 4,690 individuals during 5,942 inpatient encounters. At least 1 virus was identified in 1,871 (31%) encounters, and 229 (12%) were defined as HA-RVI (median hours from admission to positivity [IQR]: 154 [79, 308]). Among the patients with a respiratory virus infection, 56% were adults, 52% were male, 77% were non-Hispanic white, and the median Charlson score was 2 (IQR: 1, (4); HA-RVI patients were more likely to be male (59% vs. 51%, P = 0.01) and had higher median Charlson scores (3 vs. 2, P < 0.001). All 14 respiratory viruses in the diagnostic panel were positive for at least one HA-RVI (Figure 1), but rhino/enterovirus (99), influenza A (27), human metapneumovirus (22) and respiratory syncytial virus (20) were most common. Compared with non-HA-RVI patients, those with HA-RVI had longer post-infection lengths of stay (median: 9 vs. 4 days, P < 0.001) and were more likely to die during hospitalization (odds ratio [95% confidence interval]: 3.4 [2.0, 5.7]) (Table 1). Conclusion A substantial number of HA-RVI were identified during the 2017–2018 respiratory virus season, and they were associated with a striking number of severe outcomes. More in depth analyses are required to determine whether severe outcomes are a direct result of HA-RVI or whether HA-RVI are more common in critically ill patients and serve as a marker for severe morbidity. A broader understanding of HA-RVI transmission and prevention strategies is needed. Disclosures All authors: No reported disclosures.

Four wheat variety/genotype (BARI Gom-26, BAW-1202, BAW-1182 and BARI Gom-27) were tested under three heat stress regimes (normal, moderate and severe) to evaluate the effect of late seeding warmer condition on phenology and yield of wheat, as well as to identify suitable cultivars to develop heat-tolerant genotypes at Hajee Mohammad Danesh Science and Technology University (HSTU), Dinajpur during November, 2016 to April, 2017. Results revealed that genotype BAW-1182 and BAW-1202 showed greater thermostability of cell membrane with acceptable yield performance under heat stress condition. The order of tolerance based on heat susceptibility index (based on grain yield) was BAW-1182>BAW-1202>BARI Gom-27>BARI Gom-26 under heat stress conditions. Thus, BAW-1182 and BAW-1202 have the greatest potential to be used as high-yielding wheat genotypes under warm to hot environments and could be used in a breeding programme to develop heat-tolerant wheat. The amphipod genus Hirondellea Chevreux, 1889 is a cosmopolitan largely bathyal genus. This paper adds a new species from the bathyal Arabian Sea, Indian Ocean, based on material collected by baited traps at 1182, 1184 and 1864 m off the coast of Pakistan. The species can be distinguished from the most closely allied species, Hirondellea brevicaudata (Chevreux, 1910) by the subacute lateral head lobes, the very weakly cleft telson, and characters of the mouthparts. A key to the ten species in the genus is provided.

An experiment was conducted to evaluate seedling growth of wheat under saline condition. Thirty wheat genotypes were grown in trays containing different levels of salinity (Control, 6 and 12 dSm-1) during October to December, 2013. Seedling emergence index, shoot and root length, shoot and root dry weight were found to be reduced with the increases of soil salinity level but the degree of reduction were not similar for all wheat genotypes. Salt tolerance index (STI) also indicated a wide difference in salt tolerance among the wheat genotypes. Sourav, Gourav, Shatabdi, BAW 1185, BAW 1186, BAW 1187, BAW 1189 and BAW 1193 were more salt tolerance while BAW 1177, BAW 1190 and BAW 1198 showed greater salt sensitivity than the other wheat genotypes at 6 dS m-1. However, at 12 dS m-1, Sourav, Gourav, Shatabdi, Sufi and BAW 1184 showed more salt tolerance and BAW 1183, BAW 1190, BAW 1192, BAW 1194 and BAW 97 provided greater stress sensitivity among the testing wheat genotypes. Considering both saline stress Sourav, Gourav and Shatabdi were found to be salt tolerant and BAW 1190 was saline sensitive wheat genotypes.Bangladesh Agron. J. 2017, 20(2): 53-66
