Abstract:
Shoot and berry growth, sugar. titratable acidity, and anthocyanin contents of berries and crop yields of “Fujiminori” grapes (Vitis vinifera × V. labruscana) were determined in vines grafted 10 eight different rootstocks: 3309, 3306, 101-14. 5BB. 5C, 8B. SO4, and 420A. Three-year-old vines of 5BB stock and S-year-old vines of each of the other stocks grown in an unheated plastic house were used for this investigation. Shoot growth was more vigorous on vines grafted to 5BB compared to 3309, SO4, and 8B. The highest yield per unit area was observed in vines grafted to 3306. followed in order by 5BB, 3309, 101-14, SO4, 5C, 8B, and 420A. The largest berry size was observed in vines grafted to 3306, followed by 5BB, 101-14, 3309, 8B, 5C, SO4, and 420A. Berries of vines grafted to 420A and 5BB had the highest tota1 soluble solids, followed in descending order by 8B, 101-14. and 5C. Titratable acidity of berry juice was lowest in vines grafted to 420A. The anthocyanin content of berry skin was higher in vines grafted to 420A and 101-14 than in berries of other stocks. GA-treatment did not increase the percentage of seedless berries of this cultivar to a commercially acceptable level for any of the rootstocks used.

AbstractBackground and aimsNeuropathic pain is a significant medical problem needing more effective treatments with fewer side effects. Overactive glutamatergic transmission viaN-methyl-D-aspartate receptors (NMDARs) are known to play a role in central sensitization and neuropathic pain. Although ketamine, a NMDAR channel-blocking antagonist, is often used for neuropathic pain, its side-effect profile and abusive potential has prompted the search for a safer effective oral analgesic. A novel oral prodrug, AV-101 (L-4 chlorokynurenine), which, in the brain, is converted into one of the most potent and selective GlyB site antagonists of the NMDAR, has been demonstrated to be active in animal models of neuropathic pain. The two Phase 1 studies reported herein were designed to assess the safety and pharmacokinetics of AV-101, over a wide dose range, after daily dosing for 14-days. As secondary endpoints, AV-101 was evaluated in the capsaicin-induced pain model.MethodsThe Phase 1A study was a single-site, randomized, double-blind, placebo-controlled, single oral ascending dose (30–1800 mg) study involving 36 normal healthy volunteers. The Phase 1B study was a single-site randomized, double-blind, placebo-controlled, study of multiple ascending doses (360, 1080, and 1440mg/day) of AV-101 involving 50 normal healthy volunteers, to whom AV-101 or placebo were administered orally daily for 14 consecutive days. Subjects underwent PK blood analyses, laboratory assessments, physical examination, 12-lead ECG, ophthalmological examination, and various neurocognitive assessments. The effect of AV-101 was evaluated in the intradermally capsaicin-induced pain model (ClinicalTrials.gov Identifier: NCT01483846).ResultsTwo Phase 1, with an aggregate of 86 subjects, demonstrated that up to 14 days of oral AV-101, up to 1440mg per day, was safe and very well tolerated with AEs quantitively and qualitatively like those observed with placebo. Mean half-life values of AV-101 were consistent across doses, ranging with an average of 1.73 h, with the highestCmax(64.4 μg/mL) and AUC0-t(196 μgh/mL) values for AV-101 occurring in the 1440-mg dose group. In the capsaicin induce-pain model, there was no significant change in the area under the pain time curve (AUPC) for the spontaneous pain assessment between the treatment and the placebo groups on Day 1 or 14 (the primary endpoint). In contrast, there were consistent reductions at 60–180 min on Day 1 after dosing for allodynia, mechanical hyperalgesia, heat hyperalgesia, and spontaneous pain, and on Day 14 after dosing for heat hyperalgesia.ConclusionsAlthough, AV-101 did not reach statistical significance in reducing pain, there were consistent reductions, for allodynia pain and mechanical and heat hyperalgesia. Given the excellent safety profile and PK characteristics demonstrated by this study, future clinical trials of AV-101 in neuropathic pain are justified.ImplicationsThis article presents the safety and PK of AV-101, a novel oral prodrug producing a potent and selective GlyB site antagonist of the NMDA receptor. These data indicate that AV-101 has excellent safety and PK characteristics providing support for advancing AV-101 into Phase 2 studies in neuropathic pain, and even provides data suggesting that AV-101 may have a role in treating depression.

Abstract:
Abstract ZIO-101 is organic arsenic with similar anti-leukemia activity to arsenic trioxide (ATO) but much less toxicity. Consequently, ZIO-101 can be given at substantially higher doses than ATO and may be active in more diverse cancers than ATO. The precise anti-cancer mechanism of ZIO-101 is unknown: ZIO-101 increases intracellular production of reactive oxygen species (ROS) in dose dependent manner, resulting in apoptosis of leukemic cells. Modifying intracellular ROS levels alters the ability of ZIO-101 to induce apoptosis. We focused on the 2 most important intracellular ROS generators, mitochondria and NADPH oxidase. Cells were treated with ZIO-101 and NADPH oxidase expression levels, membrane translocation and interactions of subunits were studied. Effects of modified NADPH oxidase levels on ZIO-101-induced ROS production were studied (diphenyleneiodonium [DPI], a specific inhibitor of NADPH oxidase, and SiRNA technique were used to inhibit enzyme function, while bryostatin-1, a specific activator of NADPH oxidase, was used to increase enzyme function). Mitochondrial DNA-depleted HL60 Rho-0 cells were also used to evaluate mitochondrial input to ROS-generation. Our data show that ZIO-101 activates NADPH oxidase. A low concentration of ZIO-101 (1uM) requires 14 h to significantly increase intracellular ROS levels and kill leukemic cell. This effect is strongly inhibited by DPI-pretreatment and is also seen in P47-SiRNA transfected HL60 cells. On the other hand, this A higher dose of ZIO-101 (4uM) increases ROS levels more rapidly (2–6 h). This early increase is not inhibited by DPI-pretreatment or SiRNA transfection. A 14 h but not 2–6 h increase in ROS is detectable in Rho-0 HL60 cells. These data indicate dose-dependent mechanisms of ZIO-101 induction of ROS: 1uM ZIO-101 induces NADPH oxidase activity which results in ROS production detectable at 14 h; there is no mitochondrial component; 4uM ZIO-101 disrupts mitochondrial function resulting in an earlier increase in ROS levels and subsequent apoptosis. effect is significantly enhanced in cells that have increased enzyme function. ROS-production in Rho-0 HL60 cells is unaffected by these interventions.

Abstract K-ras mutant lung adenocarcinoma (KM-LUAD) is a difficult-to-drug cancer subtype characterized by a chronic inflammatory tumor microenvironment (TME). Resistance to therapies, including immune checkpoint blockade (ICB), necessitates therapies that target this inflammatory TME. A major transcription factor that mediates chronic inflammation in KM-LUAD is signal transducer and activator of transcription 3 (STAT3). Inhibiting STAT3 may attenuate pro-tumor inflammation. Moreover, STAT3 modulates PD-L1 transcription, so STAT3 inhibitors in conjunction with ICB may increase ICB response rates. Here, we tested the anti-tumor ability of TTI-101, a selective STAT3 inhibitor, in a STAT3 addicted lung cancer cell line (MDA-F471) and in a transgenic mouse model of KM-LUAD (CCSPCre/LSL-KrasG12D, CC-LR). For in vivo experiments, CC-LR mice were treated daily with 50 mg/kg TTI-101 by oral gavage from 10 to 14 weeks of age to model a preventative regimen or from 14 to 18 weeks of age to survey the treatment effect on established tumors. TTI-101 was compared to anti-PD-1 ICB, with 200 μg injected intraperitoneally 3 times per week. In MDA-F471 cells, TTI-101 treatment decreased cell viability, with an IC50 of ~ 20 μM. In mice treated from 10 to 14 weeks of age, TTI-101 therapy significantly reduced the tumor burden compared to ICB. TTI-101 also reduced the number of proliferating cells within tumors. Mice in the 14-18-week group displayed similar trends, but these experiments are ongoing, as are combination TTI-101 and ICB treatment regimens. Our studies show that TTI-101 can reduce K-ras driven tumor cell proliferation in vitro and in vivo, suggesting STAT3 inhibition as an alternative preventive and therapeutic modality for KM-LUAD. The ongoing combination treatments and the 14-18-week cohorts will elucidate the timing of treatments as well as reveal if by targeting the inflammatory TME we are able to improve response to ICB. All possible grafting combinations of seven roostock s and six scion cultivars were made to evaluate the rootstock/scion compatibility in grapevine nursery production. Percentage of takes was variable from 20.5%(Uni Blanc/161-49 C.) to 87.3%(K 188-2/161-49 C). Among scion cultivars, mean percentage of takes varied from 82.5%(K 188-2) to 37.5%(Ugni Blanc) while roostocks with all scion cultivars varied from 69.9%(R 110) to 52.8%(101-14 Mgt.), indicating the greater effects of scion on percentage of takes. Variation in rooting index(0: none to 4:profuse rooting scale) was from 3.48(SV 5276/Rip. Gloire). 3.49(Neo Muscat/101-14 Mgt.) to 1.63(SV 5276/161-49 C). Mean rooting index of rootstocks with all scion cultivars varied from 3.10(101-14 Mgt.) to 1.95(161-49 C.) while that of scion cultivars varied from 3.07(SV 5276) to 2.44(Ugni Blanc). In rooted grafting, rootstock had a greater effect than scion cultivars.

ABSTRACT KBPA-101 is a human monoclonal antibody of the immunoglobulin M isotype, which is directed against the O-polysaccharide moiety of Pseudomonas aeruginosa serotype O11. This double-blind, dose escalation study evaluated the safety and pharmacokinetics of KBPA-101 in 32 healthy volunteers aged 19 to 46 years. Each subject received a single intravenous infusion of KBPA-101 at a dose of 0.1, 0.4, 1.2, or 4 mg/kg of body weight or placebo infused over 2 h. Plasma samples for pharmacokinetic assessments were taken before infusion as well as 0.25, 0.5, 1, 2, 2.5, 4, 6, 8, 12, 24, 36, and 48 h and 4, 7, 10, and 14 days after start of dosing. Plasma concentrations of KBPA-101 were detected with mean maximum concentrations of drug in plasma of 1,877, 7,571, 24,923, and 83,197 ng/ml following doses of 0.1, 0.4, 1.2, and 4.0 mg/kg body weight, respectively. The mean elimination half-life was between 70 and 95 h. The mean volume of distribution was between 4.76 and 5.47 liters. Clearance ranged between 0.039 and 0.120 liters/h. At the highest dose of 4.0 mg/kg, plasma KBPA-101 levels were greater than 5,000 ng/ml for 14 days. KBPA-101 exhibited linear kinetics across all doses. No anti-KBPA-101 antibodies were detected after dosing in any subject. Overall, the human monoclonal antibody KBPA-101 was well tolerated over the entire dose range in healthy volunteers, and no serious adverse events have been reported.

Abstract:
Nociceptin/orphanin FQ (N/OFQ) and its receptor (NOP) are widely distributed in the central nervous system (CNS), where they modulate several functions such as pain, anxiety, stress, learning, memory, food intake and drug addiction. On this basis, the purpose of the present research was to investigate, in the rat, the role of NOP ligands in: a) modulation on nociception in paracetamol-induced analgesia; b) anxiety-related behaviours after development of tolerance to hypolocomotor effects and c) exposure to chronic stressful conditions which cause depression. As regards point a), we have demonstrated that both the antinociceptive effect of paracetamol (400 mg/kg, i.p.), evaluated by means of the hot-plate test (1) and the changes in central serotonin content were completely abolished by administration of N/OFQ (10 nmol/rat, i.c.v.) and restored by a pre-treatment with the NOP antagonist UFP-101 (20 nmol/rat, i.c.v.). In anxiety experiment (b), a double i.c.v. injection on N/OFQ dose-dependently decreased the expression of anxiety-related behaviour in both the elevated plus maze and the conditioned defensive burying tests without affecting locomotor activity. UFP-101 significantly reduced the effects of N/OFQ to control values in either test (2). In the stress paradigm (c), rats were exposed to chronic mild stress (CMS) (3) for a period of at least 6 weeks to induce a condition of anhedonia, measured as reduction of 1% sucrose solution intake. The stressed groups were treated, once a day, with UFP-101 (5, 10 and 20 nmol/rat, i.c.v.), or imipramine (IMI, 15 mg/kg, i.p.), or saline for 21 days. UFP-101, reinstated sucrose solution intake within the 1st week of treatment following the highest dose; at 10 and 5 nmol/rat it abolished the reduction in sucrose intake from the 2nd and 3rd week treatment, respectively. The restoration of sucrose consumption, once induced, remained stable up to the end of the experiment for all treatments. In the FST, 24 h after the last administration, all UFP-101 treatments decreased the time of immobility to that of non stressed controls. IMI produced similar effects on sucrose intake and on the FST. Pre-treatment with either UFP-101 at the higher doses or with IMI completely abolished the increase in CORT induced by CMS. 5-HT turnover was increased by CMS in the frontal cortex and decreased in the pons; UFP-101, as well as IMI, were able to revert these changes to values comparable to those of non stressed controls. Repeated co-administration of N/OFQ (5 nmol/rat, from day 12 to day 21) completely prevented the behavioural and biochemical effects of UFP-101 (10 nmol/rat). Our results showed that UFP-101 reversed the CMS-induced changes in behaviour, HPA axis control and central 5-HT turnover. It also abolished increase in serum corticosterone induced by CMS and reverted changes in central 5-HT/5-HIAA ratio. Brain-derived neurotrophic factor mRNA and protein in hippocampus were not reduced by CMS nor did UFP-101 modify these parameters. On the whole, the present findings support the view that the N/OFQ-NOP system represents an important candidate target for the development of innovative therapeutics for several neurological conditions, including nociception, and psychiatric diseases, chiefly involving anxiety and depression. La Nocicettina/Orfanina FQ (N/OFQ) è un peptide endogeno non oppioide agonista dei recettori ORL1, attualmente chiamati recettori NOP. N/OFQ e il NOP sono altamente espressi nel sistema nervoso centrale (SNC), in particolare nelle strutture coinvolte nei processi emozionali (ponte, corteccia frontale). La Nocicettina è coinvolta in molti sistemi e funzioni biologici integrati a livello periferico, spinale e sopraspinale; nel SNC, questo peptide ha infatti un'importante influenza sulla risposta ad ansia e stress, apprendimento e memoria, locomozione, alimentazione, ricompensa e dolore. Su questa base, lo scopo della mia tesi di dottorato è stato quello di: a) studiare l'azione della nocicettina nell'analgesia indotta da paracetamolo, b) analizzare il comportamento ansioso dopo sviluppo di tolleranza all'effetto locomotorio e c) indagare il possibile coinvolgimento del sistema dopo esposizione a stress cronico moderato. In riferimento al punto a) abbiamo dimostrato che sia l'effetto analgesico del paracetamolo (400 mg/kg, i.p.), valutato con il test della piastra calda (1), che i cambiamenti dei livelli centrali di serotonina nel ratto, sono completamente revertiti dalla somministrazione di N/OFQ (10 nmol/rat, i.c.v.) e riportati ai livelli dei ratti trattati con paracetamolo dal pre-trattamento con l'antagonista selettivo del recettore NOP, [Nphe1,Arg14,Lys15]N/OFQ-NH2 (UFP-101) (20 nmol/rat, i.c.v.). Per quanto riguarda gli esperimenti sull'ansia (b), una doppia somministrazione intracerebroventricolare ha ridotto l'effetto sulla locomozione della nocicettina, favorendo lo studio del suo ruolo ansiolitico nel ratto, utilizzando due test specifici sui comportamenti legati all'ansia: elevated plus maze e conditioned difensive burying. UFP-101 ha ridotto in modo significativo l'effetto della N/OFQ in entrambi i test (2). Infine, per gli studi del punto c), i ratti sono stati sottoposti allo stress cronico moderato (SCM), un'esposizione cronica giornaliera dell'animale ad una sequenza inevitabile di lievi stimoli stressanti che determina, dopo alcune settimane, un comportamento simil-depresso (anedonia) caratterizzato da una diminuzione del consumo di una soluzione "gratificante" contenente 1% di saccarosio (3). L'UFP-101, somministrato i.c.v. ripetutamente per 21 giorni (5, 10 and 20 nmol/rat, i.c.v.), è in grado di revertire, già a partire dalla prima settimana di trattamento e in modo dose-dipendente, la riduzione del consumo di saccarosio indotta dal SCM. Questo effetto è confermato anche dai dati ottenuti nel test del nuoto forzato (FST). L'imipramina (IMI, 15 mg/kg), farmaco antidepressivo di riferimento, è stata somministrata i.p. per 21 giorni, ed ha prodotto effetti comportamentali simili a quelli dell'UFP-101. Il trattamento con UFP-101 o IMI, inoltre, ha ripristinato, fino ai livelli basali, anche le azioni biochimiche prodotte dallo SCM quali l'incremento dei livelli di corticosterone sierico, le alterazioni del rapporto tra 5-HIAA e 5-HT a livello della corteccia frontale e del ponte. Infine, ripetute co-somministrazioni di N/OFQ (5 nmoli/ratto, dal 12° al 21° giorno) prevengono completamente gli effetti comportamentali e biochimici dell'UFP-101 (10 nmoli/ratto). L'esposizione allo SCM non comporterebbe, invece, una riduzione dei livelli di mRNA o del peptide del fattore neurotrofico brain-derived neurotrophic factor (BDNF) nell'ippocampo in confronto con l'animale non stressato. Questi risultati forniscono un supporto all'ipotesi secondo la quale il recettore NOP possa rappresentare un candidato target per lo sviluppo di nuove terapie in campo neurologico, come nocicezione, ansia e depressione. This thesis deals with the question as to whether the public and the private enforcements of European competition law, as currently shaped, are able to generate a sufficient and effective deterrence level. Proposals aimed at addressing certain possible gaps and shortages in this respect, in particular the criminalization of cartel conducts, are eventually put forward. More in details, sections I and II carry out a preliminary assessment of the real level of deterrence stemming from public and private enforcement tools with respect to violations of European competition law. Section III poses the question as to whether a possible criminalization of certain anticompetitive conducts in breach of European competition law, in particular cartels, along the line of what already occurred in many jurisdictions over the last decades, might contribute to enhance the current level of deterrence in this context. Section IV analyzes specific legal issues connected to a possible criminalization of cartel conducts in breach of art. 101 TFUE through directive, such as the identification of an appropriate legal basis pursuant to the Treaties of the European Union, the definition of the criminal offence, the penalties that could be provided, the interplay between criminal sanctions and leniency programs, the coordination between competition authorities and criminal judges, and certain further issues.

Abstract:
Pour contribuer a resoudre le probleme du manque d'acidite de certains vins, nous avons etudie, en culture hydroponique, l'influence de differents equilibres k-ca sur la nutrition cationique et la qualite des mouts et des vins de negrette. Dans un premier essai, une serie d'equilibres k-ca a ete testee. Les sommes cationiques etant constantes, nous avons fait varier les teneurs relatives en potassium et en calcium (respectivement de 0,4 a 8,7 et de 0,1 a 8,4 meq. L#-#1). Cet essai a montre que la negrette est sensible aux variations de composition cationique de la solution nutritive, que ses besoins en potassium sont faibles et qu'elle absorbe facilement cet element. En essai de longue duree, trois des solutions precedentes ont ete etudiees: une solution temoin (k:3,9-ca:4,8 meq. L#-#1), une solution faible en potassium (k:0,3-ca:8,4 meq. L#-#1), une solution riche en potassium (k:7,1-ca:1,6 meq. L#-#1), ainsi que la solution temoin surchargee en calcium pour simuler un chaulage (k:3,9-ca:13,8 meq. L#-#1). Ce deuxieme essai a confirme les resultats de la premiere experimentation sur la partie vegetative. Sur les mouts, l'augmentation de la concentration du potassium dans la solution (et la baisse du calcium) entraine celle de la teneur en cet element (+3,7 g. L#-#1), du ph (+0,4 unite) et de la concentration en acide malique (+7 g. L#-#1) ; d'autre part, la surcharge en calcium entraine une diminution de ces trois parametres (respectivement de 0,7 g. L#-#1 ; 0,1 unite et 1,9 g. L#-#1). Des microvinifications ont montre que les variations du ph sont amplifiees dans les vins (+0,8 unite entre les solutions (k:0,3-ca:8,4 meq. L#-#1) et (k:7,1-ca:1,6 meq. L#-#1) ; -0,2 unite entre les solutions (k:3,9-ca:4,8 meq. L#-#1) et (k:3,9-ca:13,8 meq. More than 80% of vineyards around the world use grafted plants: a scion of Vitis vinifera grafted onto a rootstock of single or interspecific hybrids of American Vitis species, resistant or partially resistant to Phylloxera (Daktulosphaira vitifoliae (Fitch, 1856)). The genetic variability of grapevine rootstocks plays a fundamental role in their adaptation to the environment (Serra et al., 2013). In the climate change scenario, predicting an increase of aridity in the near future (Dai, 2013), the more frequent and severe drought events may represent the major constrain for the future of viticulture (IPCC, 2018; Schultz, 2000). Therefore, the selection of new rootstocks able to cope with unfavourable environmental condition is a key asset, as well as a strategy to improve crop yield/vegetative growth balance on scion behaviour (Corso and Bonghi, 2014). So far, the influence of rootstock on scion physiological performance during water stress has always aroused great interest. On the contrary, the scion impact on rootstock response is still less debated. Therefore, the effect of grafting on rootstock behaviour have been investigated. Phenotypical and large-scale whole transcriptome analyses on two genotypes, a drought-susceptible (101-14) and a drought-tolerant (1103 P), own-rooted and grafted with Cabernet Sauvignon, subjected to a gradual water shortage in semi-controlled environmental conditions have been performed. The ungrafted condition affected photosynthesis and transpiration, meaning the decisive role of scion in modulation of gas exchanges and in general in plant adaptation. Molecular evidence highlighted that the scion delays the stimulus perception and rootstock reactivity to drought. Since 1985, the DiSAA research group operating at the University of Milan is carrying on a rootstock crossbreeding program which has led to the release of four genotypes: M1, M2, M3 and M4. They show from moderate to high tolerance to drought (M4 > M1 = M3 > M2). In order to characterize their performance during water stress, their physiological (gas exchanges and stem water potential) and transcriptome response (genes involved in ABA-synthesis and ABA-mediated responses to drought) under well-watered and water stress conditions were examined. The behaviour of M-rootstocks (M1, M2 and M3) was compared with that of other commercial genotypes largely used in viticulture, either tolerant (140 Ru, 41 B, 110 R, 1103 P), less tolerant (SO 4, K 5BB) and susceptible (420 A and Schwarzman). Discriminant analysis (DA) showed that when water availability starts to decrease, rootstocks firstly perceives the stress activating a transcriptome response, consequently physiological changes have been observed. It also demonstrated that the three M-rootstocks were clearly discriminated: M4 was grouped with the most tolerant genotypes while M3 with the less tolerant or susceptible ones from a physiological standpoint, confirming their different attitude to tolerate water stress. M4 has proven to be a promising rootstock due to its ability to adapt to drought conditions. Considering the constant great demand for vine planting materials, the obtainment of genetically homogeneous populations (i.e. clones) from elite individuals through micropropagation represents a rapid alternative to conventional multiplication. For this reason, an efficient high-throughput protocol for M4 in vitro propagation was set up. Its attitude to shooting, root development and callus proliferation was compared to that of other rootstocks largely used in viticulture (K5BB, 1103P, 101-14 and 3309C). Moreover, pro-embryogenic and embryogenic callus from bud explants were also produced, representing a cellular material manipulable with the genetic engineering techniques. In water scarcity condition, among the mechanisms activated by M4, the great ability to scavenge ROS, related to the increased accumulation of stilbenes and flavonoids, may be such as to give it tolerance to the stress. In particular, the higher levels of trans-resveratrol were correlated with the up-regulation of some stilbene synthase genes, mainly VvSTS16, VvSTS18, VvSTS27 and VvSTS29. The over expression of these genes was linked to a structural variation in their promoter region. To confirm that VvSTSs genes may be considered putative factors of M4 better adaptation to water stress, a genome editing protocol based on the CRISPR/Cas9 system, aimed at knock-out the genes, was performed. For testing the gRNAs functionality, a transient assay on in vitro micropropagated plantlets of M4 and 101-14 was performed. The positive results obtained by this experiment will lead to the transformation of somatic embryos and regeneration of whole-edited plants using the vectors developed.

La presente ricerca si propone di analizzare la Direttiva 2012/13/UE sul diritto all'informazione nei procedimenti penali ed il suo impatto sul sistema processuale italiano. L'analisi prende le mosse da un primo capitolo dedicato al sistema multilivello delle fonti: sul panorama nazionale e sovranazionale, infatti, la direttiva è solo l'ultima norma, in ordine di tempo, a disciplinare il diritto fondamentale alla conoscenza dell'indagato e dell'imputato. Necessario quindi apprestare una panoramica delle fonti che garantiscono la protezione multilevel dei diritti, e descrivere le loro reciproche interazioni. Imprescindibile, poi, un approfondimento sulla tutela dei diritti nello Spazio di Libertà Sicurezza e Giustizia dell'UE, con un'attenzione particolare all'era post-Lisbona ed al valore aggiunto che le direttive ex art. 82 co. 2 TFUE possono portare sul sistema multilevel. Il secondo ed il terzo capitolo sono dedicati all'analisi normativa della fonte europea. La trattazione si muove lungo le tre visuali prospettiche che la norma europea attribuisce al diritto all'informazione: diritto alla conoscenza dei propri diritti; diritto alla conoscenza dell'accusa; diritto alla conoscenza degli atti di indagine. Le disposizioni europee vengono continuamente integrate con la giurisprudenza della Corte EDU, che inietta di significato le norme della direttiva e fornisce gli standards di tutela laddove non specificati. Vengono messe in rilievo le disposizioni più innovative, che consentono alla direttiva di non essere solo “codificazione” del case law di Strasburgo, ma fonte autonoma e progredita di diritti. Il capitolo finale è infine focalizzato sull'impatto che la direttiva ha prodotto sul sistema processuale interno. La trattazione è suddivisa tra l'analisi delle modifiche apportate dalla normativa di attuazione italiana, d. lgs. 101/2014, e la disamina delle sue lacune: il legislatore ha dato luogo ad un intervento minimalista, omettendo di dare esecuzione proprio alle disposizioni europee più innovative che avrebbero permesso al nostro sistema di essere in linea con i dettami sovranazionali. Particolare attenzione è data al tema delle modifiche all'imputazione e al principio Iura novit curia, sulla scorta dei punti saldi elaborati dalla Corte EDU nel noto caso Drassich. In conclusione, vengono proposti gli scenari futuri che potrebbero conseguire all'efficacia diretta della direttiva e alla penetrazione, per il suo tramite, delle norme CEDU nell'ordinamento giuridico nazionale.
The present research examines the European Directive on the right to information in criminal proceedings (Directive 2012/13/EU, hereinafter ‘the Directive’), assessing the impact that it is likely to have on the Italian legal system. Before analyzing the legislation, the thesis provides an historical overview of the status of human rights safeguards in the EU and a description of its multi-layered system of protection. Starting from the early ECJ case law setting out a ‘human rights theory’, the research moves on to consider the Charter of Nice and the development of a European Area of Criminal Justice, until the Stockholm Program and the entry into force of the Lisbon Treaty. In addition, it addresses the question as to whether and to what extent the directives ‘of new generation’ based on art. 82 par. 2 TFEU bring an added value to the aforementioned human rights protection system. Chapters 2 and 3 of the research focus on the analysis of the legislation and on the three meanings that the Directive attaches to the right to information in criminal proceedings, namely, the right to information about rights, the right to information about accusation, and the right to information about case file. The effort is shedding some light on the most innovative prescriptions, while at the same time highlighting how much the EU legislation owes to the ECtHR case law, which is used as a yardstick for the evaluation and interpretation of the Directive. Finally, Chapter 4 addresses the Italian implementing legislation (d. lgs. 101/2014) and the impact of the Directive on our legal system. It finds that the NIM is highly unsatisfactory, as the Italian legislator has failed to comply with the most innovative EU standards. In this regard, the research illustrates the impact of EU prescriptions on the jurisdiction of national judges, in particular, the impact of the ‘new’ right to information about accusation. It concludes that Italian judges can (in)directly apply ECtHR case law standards due the direct effect of the Directive (which can be regarded as an ‘ECtHR case-law codification’).

Programming is one of the core and indispensable strategies in the way of successfulness that should be taken into account either by teachers or students. Owing to this fact teaching and learning of programming has been widely under investigation which shows how its effect could empower the student's achievements. In the other word, it is a very useful skill and can be a rewarding career not only for on-campus educations but also for distance and electronic learning systems. Therefore, recently the demand for programmers and student's interest in programming has rapidly grown and introductory programming courses have become increasingly more popular; however, learning to program is still tedious. Programming courses are generally regarded as difficult and often have highest dropout rates. In addition to that beginners in programming are basically suffered from a wide range of difficulties and deficits. It can draw more attentions to intend providing the most effective learning environment and experience for the students. Constructivist learning theory has shaped our thinking in this research. Fundamentally, constructivism says that people construct their own understanding and knowledge of the world through experiencing things and reflecting on those experiences. Consequently the researches are being attracted to understanding the processes of learning and teaching a first programming language. This study and research tries to point out the answer for questions such as: Why is programming hard to learn? What are the cognitive requirements of the task? Water quality impairment due to anthropogenic activities affects fishery resources. Surface water bodies in the Niger Delta Region of Nigeria are being adversely impacted by the effects of pollution and the destruction of its fishery resources. This study was carried out to assess the impacts of anthropogenic activities on the fish compositions and diversity of Okerenkoko Estuarine, Delta State, Nigeria. Okerenkoko Estuarine (62.79 Km) was spatially stratified into five stations (Z1, Z2, Z3, Z4 and Z5) based on nearness to major anthropogenic activities. Temporal stratification covered June to December. Fish samples were collected for 7 months from each station at the study area with prior arrangement with the fishers. Fishing gears used were Mid – Water Drift Gill Net of mesh sizes 101 mm and 127 mm; Bottom Set Gill Net (50 – 100 m) of mesh size 76 mm, 101 mm and 127 mm; Hook and line (2 – 6 m) of hook sizes No. 5, No. 7, No. 8, No. 9, No. 12 and No. 15; Long Line (300 – 1000 m) of hook sizes No. 7, No. 8. Parameters determined were Numbers of Species, Abundance (%), Simpson and Shannon by using standard procedures. Data were analysed by using Excel (Window 10) and Palaeontological Statistics (Past Version 3.6). A total number of 1261 of fishes and 45 species belonging to 14 families were identified. Highest 383 (30.4 %) and least 152 (12.1 %) number of fishes were obtained in Z5 and Z4, respectively. The most abundant fish species recorded was Sardinella aurita 192 (15.2 %), while the least was Sphyraena afra 1 (0.1 %), respectively. Temporally, the highest 331 (26.2 %) and least 45 (3.6 %) fish species Occurred in December and September. Simpson index ranged from 0.69 to 0.89 in Z1 and Z3, Shannon (1.97, 2.98) occurred in Z4 and Z2, respectively. Fish biodiversity richness (0.69) of Okerenkoko Estuarine could be threatened. Abstract:
Pulse jet mixed (PJM) process vessels are being developed for storing, blending, and chemical processing of nuclear waste slurries at the Waste Treatment and Immobilization Plant (WTP) to be built at Hanford, Washington. These waste slurries exhibit variable process feed characteristics including Newtonian to non-Newtonian rheologies over a range of solids loadings. Waste feed to the WTP from the Hanford Tank Farms will be accomplished via the Waste Feed Delivery (WFD) system which includes million-gallon underground storage double-shell tanks (DSTs) with dual-opposed jet mixer pumps. Experience using WFD type jet mixer pumps to mobilize actual Hanford waste in DSTs may be used to establish design threshold criteria of interest to pulse jet mixed process vessel operation. This paper describes a method to evaluate the pulse jet mixed vessel capability to process waste based on information obtained during mobilizing and suspending waste by the WFD system jet mixer pumps in a DST. Calculations of jet velocity and wall shear stress in a specific pulse jet mixed process vessel were performed using a commercial computational fluid dynamics (CFD) code. The CFD-modelled process vessel consists of a 4.9-m- (16-ft-) diameter tank with a 2:1 semi-elliptical head, a single, 10-cm (4-in.) downward facing 60-degree conical nozzle, and a 0.61-m (24-in.) inside diameter PJM. The PJM is located at 70% of the vessel radius with the nozzle stand-off-distance 14 cm (6 in.) above the vessel head. The CFD modeled fluid velocity and wall shear stress can be used to estimate vessel waste-processing performance by comparison to available actual WFD system process data. Test data from the operation of jet mixer pumps in the 23-m (75-ft) diameter DSTs have demonstrated mobilization, solid particles in a sediment matrix were moved from their initial location, and suspension, mobilized solid particles were moved to a higher elevation in the vessel than their initial location, of waste solids. Jet mixer pumps were used in Hanford waste tank 241-AZ-101, and at least 95% of the 0.46-m (18-in.) deep sediment, with a shear strength of 1,500 to 4,200 Pa, was mobilized. Solids with a median particle size of 43 μm, 90th percentile of 94 μm, were suspended in tank 241-AZ-101 to at least 5.5 m (216 in.) above the vessel bottom. Analytical calculations for this jet mixer pump test were used to estimate the velocities and wall shear stress that mobilized and suspended the waste. These velocities and wall shear stresses provide design threshold criteria which are metrics for system performance that can be evaluated via testing. If the fluid motion in a specific pulse jet mixed process vessel meets or exceeds the fluid motion of the demonstrated performance in the WFD system, confidence is provided that that vessel will similarly mobilize and suspend those solids if they were within the WTP. The single PJM CFD-calculated jet velocity and wall shear stress compare favorably with the design threshold criterion estimated for the tank 241-AZ-101 process data. Therefore, for both mobilization and suspension, the performance data evaluated from the WFD system testing increases confidence that the performance of the pulse jet mixed process vessels will be sufficient to process that waste even if that waste is not fully characterized.

Abstract:
This paper describes the evolution of an ultrasonic sensor to non-invasively measure slurry density and solids concentration. Three generations of probe are discussed: 1) density sensor, 2) densimeter, and 3) fluid and solids monitor. The initial application was to measure slurry density during radioactive waste transport. The probe uses ultrasonic signal reflection at the fluid-pipe wall interface to quantify density in situ in real time and signal attenuation to evaluate solid concentration. In the latest configuration, the transducers are mounted on the wall of the pipe spool piece. This instrument was selected for monitoring radioactive waste transport based on several characteristic features: the sensing surface is non-intrusive and does not disrupt the slurry flow, instrument performance is not affected by slight amounts of entrained air that could be present during waste retrieval and transfer; nor is it affected by electromagnetic noise from nearby pumps and other equipment; and the instrument is compact. The densimeter has been deployed at Hanford tank SY-101 in the prefabricated pump pit process manifold to monitor slurry properties during radioactive waste transfers. To qualify densimeter performance prior to manifold installation in the radioactive pipeline, the probe was installed in the process manifold and performance was evaluated during tests using non-radioactive waste simulants over the density range from 1000 to 1500 kg/m3. The probe predicted density within ± 2%. The sensor is installed in a nominal 5-cm-diameter (2-in.) pipe spool piece; the design pressure is 2.8 MPa (400 psi). To ensure operability during prolonged contact with radioactive waste, the probe wedge in contact with the slurry was selected to operate up to pH 14, and the probe components were radiation tested at exposures of 1×106 R.

Abstract:
In plasma exchange donation (PED), several liters of fresh plasma are removed fran a donor with a pheresis instrument as a source of cryoprecipitate, and replaced with autologous cryoprecipitate-supernatant from the previous donation. Repetitive PED can produce large quantities of factor VIII from individual donors over time, with a favorable impact on donor exposure for factor VIII recipients. To clarify the implications for donor safety, we report our experience with several donors who have undergone multiple PEDs. Detailed observations are presented for one donor who has undergone PED 101 times between 5/83 and 1/87, and has provided all the factor VIII needed by his son (now age 14) with severe hemophilia A during this period. Exchange volume was gradually increased while donation frequency was gradually decreased. There were 23 exchanges of 2 L, 52 of 2.5 L, and 26 of 3 L for a total of 254 L plasma exchanged. Desmopressin (20 meg tV) was given before 45 more recent donations to augment factor VIII yield. A total of 343,274 IU factor VIII have been collected; the mean (±SD) yield from a 3 L, desmopressin- stimulated PED is 5598 ± 899 IU. The donor has remained in good health; he has noted no adverse effects fran any PED, and none have been found in laboratory monitoring. Prior to the 100th donation the following were within normal limits: CBC,platelet count, urinalysis, SMA-18, protein electrophoresis, IgG, IgA, IgM, hemolytic complement, C3, C4, fibronectin, prothrombin time, partial thromboplastin time, thranbin time, factor VIII:C (140%), factor VIII:Ag (134%), von Willebrand factor (86%) and fibrinogen (215 mg/dL). In another family, the father has donated 40 times since 1981 and the paternal grandmother has donated 31 times since 1984 with no untoward effects detected in clinical or laboratory monitoring. They have supported two moderately affected patients now ages 7 and 9. Extensive experience with these donors suggests that repeated PED is safe, and that a highly motivated donor can sometimes provide single donor support, even for a severe hemophiliac.

Abstract:
In a prospective, randomized study patients undergoing hysterectomy were treated either by the low molecular weight heparin Fragmin or by the combination of unfractionated sodium heparin + dihydroergotamin (HDHE). The dosage in the Fragmin group was 2× 2500 anti Xa-U on day 1 = day of surgery, from day 2-8: 1× 5000 anti Xa-U, in the HDHE-group from day 1-8: 2× 5000 IU heparin + 0.5 mg DHE. 99 patients were randomly allocated to prophylaxis with Fragmin, 101 to HDHE prophylaxis. 95 and 96 respectively were evaluated, the others excluded for different reasons. The 2 groups were comparable for general data and risk factors. Duration of surgery, intraoperative blood loss, transfusion rates and postoperative hemoglobin levels were identical. Blood volumes in subcutaneous and subfascial drainages were slightly but not significantly higher in the Fragmin group. In patients with an additional Marshall-Marchetti-operation, blood volumes in the drainages of the spatium retzii were significantly higher in patients on Fragmin. No differences were observed in the incidence of minor and major wound hematoma. Painful injections and sugillations at the injection sites were more frequently observed in the HDHE-group. The thermographic DeVeTherm test, which was carried out daily for diagnosis of DVT, gave positiv results (= temperature difference 1°C) on one day only in 14 patients of each group. The test was positive on 2 or more consecutive days in 4 patients on Fragmin and 2 patients on HDHE. Phlebography, which was carried out in the latter patients, gave a positive result in 1 patient of each group. Localization of DVT was mainly the lower limb. Plasma anti-Xa activity (S-2222) 4 hrs. after injection of 5000 anti-Xa IU Fragmin was 0.45 IU/ml being 10 fold higher than after HDHE. aPTT was slightly prolonged in both groups, thrombin time and thrombelastogramm gave even more pronounced changes in the Fragmin group. The present data indicate that Fragmin dosage should be further decreased to avoid bleeding complications.

Denna rapport sammanställer information om projektet Ålen i Fardume träsk, som startade 1980 på Gotland med syftet att undersöka både ekologiska och ekonomiska resultat av ålutsättningar. Då projektet varit igång sedan 1980 har flera aktörer varit inblandade, och data har lagrats i pärmar och digitalt på olika hårddiskar, servrar och databaser genom åren. Syftet med denna rapport är därför att redogöra vad som gjorts inom projektet sedan 1980 fram till idag (2022), för att ge en bakgrund till det arbete som gjorts, vilka studier som publicerats med data från platsen och vilket data som idag finns tillgängligt. Denna syntes är nödvändig för att möjliggöra framtida användning av insamlad data från ålprojektet vid Fardume träsk. Totalt har ca 101 000 ålindivider satts ut i Fardume träsk, 1980 sattes det ut ca 53 000 individer och 1989 sattes det ut ytterligare ca 48 000 individer. Det introducerade ålbeståndets utveckling har sedan följts upp kontinuerligt via en fälla som fångar utvandrande ål från Fardume träsk i Bångån. Fällan har varit drift sedan 1980 och det samlas fortfarande in ål här vilket idag finansieras via SLU Aqua och länsstyrelsen Gotland. Fällan i Bångån har genererat utvandringsdata (antal fångade ålar per år) samt individdata då ål samlats in från fällan för individprovtagning i laboratorium. Totalt har det publicerats åtta vetenskapliga artiklar baserat på data från Fardume träsk, den första publicerades 1986 och den sista 2003. Sedan 2003 har inga vetenskapliga artiklar publicerats med data från Fardume träsk. Otoliter från ål från Fardume träsk har använts för att utveckla och validera åldersläsning av ål då de har en känd ålder (utsatta antingen 1980 eller 1989). Data från Fardume träsk gicks igenom av personal på SLU Aqua under 2020 och 2021 då det fanns flera frågetecken kring datakvalitén och var data lagrades. Sammanställningarna visade att fångstdata från fällan i Bångån behöver ses över och jämföras med fältprotokoll (främst efter 1995) men det går att använda data idag för att säga att det bör ha fångats minst 14 914 individer i fällan under 1980-2022. Individdata finns idag sparade i två separata Excel-filer samt i SLU Aquas databas Sötebasen, dessa filer behöver ses över, kombineras, kontrolleras så att inte samma individ förekommer flera gånger och sedan lagras på en beständig plats innan det kan användas i forskningssyfte. Det finns idag 26 år med data från Fardume träsk som inte använts i någon (för oss) känd vetenskaplig artikel eller rapport. Data från fällan i Bångån skulle kunna användas för att utvärdera hur stor andel av utsättningen som återfångats vilket kan användas för att skatta hur mycket ål som producerats på en känd sjöareal, möjliggöra nya studier gällande utvandringsstorlekar, kondition, tillväxt, parasitering med mera. Individdata skulle kunna användas för att följa tillväxtmönster, överlevnad, könsfördelning, konditionsfaktorer och storleksstrukturen för den ål som vandrat ut från Fardume träsk via Bångån samt för att utvärdera hur länge utsatt ål stannar i ett system efter en utsättning.
