Abstract:
Clostridioides difficile is an opportunistic gut pathogen which causes severe colitis, leading to significant morbidity and mortality due to its toxins, TcdA and TcdB. Two intra-muscular toxoid vaccines entered Phase III trials and strongly induced toxin-neutralising antibodies systemically but failed to provide local protection in the colon from primary C. difficile infection (CDI). Alternatively, by immunising orally, the ileum (main immune inductive site) can be directly targeted to confer protection in the large intestine. The gut commensal, non-toxigenic C. difficile (NTCD) was previously tested in animal models as an oral vaccine for natural delivery of an engineered toxin chimera to the small intestine and successfully induced toxin-neutralising antibodies. We investigated whether NTCD could be further exploited to induce antibodies that block the adherence of C. difficile to epithelial cells to target the first stage of pathogenesis. In NTCD strain T7, the colonisation factor, CD0873, and a domain of TcdB were overexpressed. Following oral immunisation of hamsters with spores of recombinant strain, T7-0873 or T7-TcdB, intestinal and systemic responses were investigated. Vaccination with T7-0873 successfully induced intestinal antibodies that significantly reduced adhesion of toxigenic C. difficile to Caco-2 cells, and these responses were mirrored in sera. Additional engineering of NTCD is now warranted to further develop this vaccine.

Abstract Introduction Cases of depressive symptoms are highly prevalent among older adults. Moreover, depression among older adults is strongly associated with cognitive impairment, with a prevalence of approximately 8% to 16% of aging adults (Blazer, 2003). Therefore, this study sought to investigate and examine the interplay between physical activity, insomnia, and depressive symptoms among cognitively impaired older adults without dementia using data from the Health and Retirement Study. Methods Depressive symptoms and insomnia levels are based on self-reports. We used a negative binomial to assess these relationships and present the incident rate ratios (IRRs) and 95% confidence intervals from our analysis of depressive symptoms associated with insomnia and moderate physical activity. Results We observed that insomnia (IRR: 1.18, 95% CI: 1.13-1.24) and moderate physical activity (IRR: 1.13, 95% CI: 1.07 – 1.19) are positively associated with an increase in depressive symptom levels among older adults. Model Two included an interaction term for insomnia and physical activity; results indicate that the interaction term is negative (IRR: .97 95% CI: .94 - .99). Model three tested gender to moderate the relationship between physical activity and depressive symptoms. Conclusion The results from the non-significant interaction term suggest that this relationship is non-significant. Similarly, the interaction term (gender x physical activity) results show that the link between physical activity and depressive symptoms did not vary by gender. Support (if any)

Abstract:
Over fifty years after its original inception, the idea for a European unitary patent system has not yet been realised. Each attempt to introduce such a system has been met with fierce criticism. For the most recent proposal, the so-called unitary patent package, this tradition has continued. One of the most significant issues at the core of a majority of criticisms is that the existing European patent system is fragmented and that the proposed system will further increase this fragmentation. The almost universal assumption of commentators has been that this is a shortcoming of the system. However, to date no proper consideration of the legitimacy of this assumption has been attempted. Commentators have not asked what might the consequences be for the European patent system if fragmentation is a significant feature of the proposed system. This thesis considers the legitimacy of the assumption regarding the dangers of fragmentation for the European patent system, including substantive patent law. The conclusion reached has been that while fragmentation is a central feature of the existing European patent system, and is moreover likely to be a central feature of the proposed European unitary patent system, this is cause for neither criticism nor concern. On the contrary, fragmentation is not only an inherent feature of any European patent system, but also a desirable one. It is an inherent feature because no system has replaced the last, and rather than unifying the area, indirect law making has ensured a level of fragmentation. It is a desirable feature because the goals of European harmonisation and integration are not pursued entirely at the expense of national diversity. The fragmentation that exists within the current and future European patent systems should not be perceived as a negative feature of those systems, but rather embraced as an inherent and positive feature of them.

La commande artistique et littéraire de Charles le Chauve est la commande princière la plus importante de l’époque carolingienne. Les œuvres reçues par le roi témoignent tant de sa grande piété que de sa volonté de gouverner selon l’idéal du roi « philosophe » , le souverain cherchant à se gouverner lui-même pour mieux gouverner son peuple et le mener au salut. Les œuvres témoignent en outre des ambitions politiques de Charles qui souhaite accéder à l’Empire. Proclamant la continuité avec l’Empire romain, l’iconographie et les thèmes développés par les sources littéraires et figurées sont extrêmement inspirés de l’époque paléochrétienne. Ceux-ci s’inscrivent par ailleurs dans le sillon des œuvres commandées par Charlemagne, empruntant aussi plus ponctuellement aux œuvres de l’Empire byzantin. Le croisement des sources textuelles et des images permet de mesurer l’influence de Jean Scot et d’Hincmar de Reims sur la production artistique du royaume ; il permet aussi de mieux comprendre l’élaboration de l’image royale (dont les codes se mettent alors en place) et l’utilisation que Charles fit des œuvres pour renforcer son pouvoir, donnant de lui l’image du souverain chrétien idéal
The literary and artistic patronage of Charles the Bald is by far the richest of the carolingian age. The king received works that were either commissioned, or inspired by him. They actually reflect not only his piety, but also his ambition to rule as an enlightened king and to become an Emperor. The works contain the carolingian ideal of continuity with Roman Empire : iconography and themes of the literary sources and the pictures are extremely inspired by the paleochristian age. The artistic and literary patronage reveals the influence of works commisionned by Charlemagne and byzantine emperors too. The study of both texts and images bring to light the influence of John Scotus and Hincmar of Reims on the artistic production of the realm. It allows as well a better understanding of the making of the royal image (whose codes are defined in this period) and the use of the works by Charles the Bald to strengthhen his power, giving of himself the image of an ideal christian king

Cette thèse est consacrée à l'étude des diplômes royaux de Louis le Germanique (t 876), roi en Francie orientale (833-876). Il s'agissait avant tout d'explorer les linéaments d'une évolution fondamentale pour l'histoire européenne: l'individualisation de la Francie orientale. Mais poser cette question supposait de s'appuyer sur une documentation dont deux des traits principaux ont guidé la démarche de cette thèse: le rôle éminent de l'écrit dans l'expression du pouvoir royal au IXe siècle, tout d'abord, et donc l'attention accordée par ses acteurs aux formes et aux conséquences de l'instrumentation des décisions; la longue chaîne ensuite qui relie ces traces écrites du pouvoir aux historiens qui les examinent aujourd'hui. Les diplômes ont ainsi été étudiés sous trois angles différents: historique, diplomatique et historiographique. Lorsque cela s'avérait pertinent, ils ont été comparés avec les actes des autres rois carolingiens, particulièrement ceux de Charles le Chauve (840-877). Pour comprendre l'évolution de la documentation diplomatique du IXe siècle, il fallut d'abord retracer celle des conditions de production des actes (partie I). Il importa ensuite de retourner à la source, c'est-à-dire d'examiner attentivement les diplômes royaux (partie II). Enfin, une analyse des éditions critiques des actes, publiées aux XIXe-XXe siècles et qui sont les intermédiaires privilégiés entre la source et 1 'historien, vint compléter ce travail (partie III). Notre étude permet de conclure à une évolution distincte des diplômes royaux qui est le reflet d'un projet politique cohérent: la création d'un royaume de Francie orientale par Louis le Germanique

Abstract:
An eleven-year series of radio timing observations of 0833- 45 (Vela) and PSR 1641- 45 is presented. During this time, five large spin-ups ("glitches") were observed in 0833- 45 and one in 1641-45. The stellar response to these events is investigated, and the three relat ively long complete inter-glitch intervals in 0833-45 are modeled. The results are of relevance to studies of the interiors of neutron stars. The initial aim of the project - to obtain good observational coverage of large glitches in the Vela pulsar - was successfully achieved, and high quality observations of the periods between glitches were obtained as a by-product. The results of the analysis presented here provide support for the existence of both linear and non-linear coupling in the Vela pulsar, and put a limit on the former in PSR 1641- 45. The recently observed existence of a rapidly recovering component of part of a glitch in Vela was verified in the subsequent glitch, although there is now evidence to contradict the suggestion that this component involves a particular region of the star that is implicated in every glitch. Observations of a recent glitch in the same pulsar have resolved a small component of the spin-up; such a component has not been reported for any other large glitch.
