Abstract:
Diversos estudios han explicado los orígenes del sistema neoliberal y sus expresiones en el contexto urbano global. El presente artículo, además de discutir los orígenes y aplicaciones del neoliberalismo en América Latina, busca explicar el papel de la reestructuración urbana en la aplicación de políticas neoliberales en ciudades portuarias latinoamericanas. En este sentido, fue desarrollado un análisis bibliográfico sobre los orígenes del neoliberalismo y su comportamiento en Latinoamérica. También fueron realizadas entrevistas con informantes clave que proporcionaron explicar el rol de la reestructuración urbana como parte importante del sostenimiento del sistema neoliberal en ciudades latinoamericanas. Los resultados de la investigación apuntan distintas expresiones del neoliberalismo en las ciudades portuarias de Rosario (Argentina) y Valparaíso (Chile).

Abstract:
Salt-inducible kinase 2 (SIK2) has previously been identified as a potential therapeutic target in high-grade serous ovarian cancer. Depletion of SIK2 sensitises ovarian cancers to paclitaxel treatment both in vitro and in xenograft models. In patients, increased tumour SIK2 expression is associated with paclitaxel resistance and correlates with increased risk of cancer progression following paclitaxel treatment. Although high-grade serous tumours are highly chemosensitive, 80-90% of patients with advanced stage disease eventually relapse due to recurrent chemotherapy-resistant disease, so there is a pressing need for novel therapeutic targets. Inhibition of SIK2 is a plausible strategy to augment existing chemotherapy in ovarian cancer, but signalling downstream of SIK2 has not been well studied. Here, I present the optimisation and results of a chemical genetic screen to identify novel SIK2 substrates to facilitate its development as a therapeutic target. The screen used a SIK2 gatekeeper mutant engineered to utilise a 'bulky' A*TP?S analogue to specifically label its substrates in lysates from an ovarian cancer cell line. A covalent capture approach was then used to isolate labelled peptides for identification by tandem mass spectrometry. Between four independent screen experiments, 73 putative substrate proteins were identified, 18 of which were present in multiple replicates. SIK2 was found to phosphorylate the p85a regulatory subunit of PI3 kinase at S154, resulting in activation of the PI3K/AKT signalling pathway. Furthermore, PI3K/AKT signalling was stimulated in ovarian cancer cells upon incubation with primary adipocytes extracted from the omentum, a key site of ovarian cancer metastasis, in a SIK2 dependent manner. Finally, overexpression of wild-type SIK2, but not a kinase-inactive mutant, in ovarian cancer cell lines promoted tumour growth and metastasis in a xenograft model. Together, this evidence suggests that in addition to its role in chemotherapy resistance, SIK2 also promotes the establishment of omental metastases in ovarian cancer.

Abstract:
This thesis introduces an innovative method for the analysis of the player character (PC) in offline computer role-playing games (cRPGs). It derives from the assumption that the character constitutes the focal point of the game, around which all the other elements revolve. This underlying observation became the foundation of the Pivot Player Character Model, the framework illustrating the experience of gameplay as perceived through the PC's eyes. Although VG characters have been scrutinised from many different perspectives, a uniform methodology has not been formed yet. This study aims to fill that methodological void by systematising the hitherto research and providing a method replicable across the cRPG genre. The proposed methodology builds upon the research of characters performed in video games, fiction, film, and drama. It has been largely inspired by Anne Ubersfeld's semiological dramatic character research implemented in Reading Theatre I (1999). The developed theoretical model is applied to three selected cRPGs, which form an accurate methodological sample: The Witcher (CD Projekt RED 2007), Fallout 3 (Bethesda Game Studios 2008), and Vampire: The Masquerade - Bloodlines (Troika Games 2004). The choice of the game genre has been incited by the degree of attention it draws to the player character's persona. No other genre features such a complex character development system as a computer role-playing game.
