Energi Listrik adalah salah satu energi yang sudah menjadi kebutuhan primer bagi masyarakat. Dalam produksi energi listrik, dibutuhkan pembangkit tenaga listrik yang dalam produksinya tidak merugikan lingkungan yang ada di sekitar. Oleh karena itu teknologi pengembangan pembangkit listrik ramah lingkungan yang sering disebut dengan sebutan EBT ( Energi Baru Terbarukan ) sedang digencarkan, simulasi penggabungan antara PLTS dan PLTB menjadi satu kesatuan sistem hibrida dengan mode off-grid dapat menjadi salah satu solusi alternatif dari permasalahan diatas. Sistem dirancang dengan alur dari pembangkit tenaga surya dan angin dijadikan menjadi satu melalui controller hybrid 700 W, kemudian dari controller akan disalurkan menuju baterai dengan tegangan 12V dan dari baterai akan diteruskan ke beban pompa aerator kolam 25 W dengan pengkonversian tegangan dari 12 V ke 220 V menggunkan bantuan inverter. Dari Hasil monitoring data logger, energi rata – rata per hari yang dapat dihasilkan oleh PLTS adalah kisaran sebesar 316,84 Wh. Sedangkan energi rata – rata per hari yang dapat dihasilkan oleh PLTB adalah kisaran sebesar 67,77 Wh. Lalu pada Hasil monitoring IoT, didapatkan energi rata – rata yang dapat digunakan beban dalam kisaran waktu 14,2 jam adalah sebesar 242,25 Wh. Terdapat energi sisa sebesar 142,36 Wh yang dapat disimpan ke dalam baterai.

P′2-type Na0.67[Ni0.1Fe0.1Mn0.8]O2 is introduced as a promising new cathode material for sodium-ion batteries (SIBs) that exhibits remarkable structural stability during repetitive Na+ de/intercalation. The O-Ni-O-Mn-O-Fe-O bond in the octahedra of transition-metal layers is used to suppress the elongation of the Mn-O bond and to improve the electrochemical activity, leading to the highly reversible Na storage mechanism. A high discharge capacity of ≈220 mAh g−1 (≈605 Wh kg−1) is delivered at 0.05 C (13 mAg−1) with a high reversible capacity of ≈140 mAh g−1 at 3 C and excellent capacity retention of 80% over 200 cycles. This performance is associated with the reversible P′2–OP4 phase transition and small volume change upon charge and discharge (≈3%). The nature of the sodium storage mechanism in a full cell paired with a hard carbon anode reveals an unexpectedly high energy density of ≈542 Wh kg−1 at 0.2 C and good capacity retention of ≈81% for 500 cycles at 1 C (260 mAg−1).

Abstract INTRODUCTION Previous SEER-Medicare analyses reported lower receipt of guideline concordant care for American Indian (AI) compared with White (WH) patients across several cancers. While large scale assessments are informative, there is a need to assess cancer outcomes and develop interventions at a level closer to communities. To address this gap, we assessed measures of quality care in AI and WH breast, prostate, and colorectal cancer patients. METHODS We assembled novel cohorts of AI and WH patients from the North Carolina (NC) cancer registry, the state with the largest AI population east of the Mississippi River. Eligible patients were diagnosed with breast, prostate, and colorectal cancers who linked to public and private insurance claims with continuous enrollment (2003-2019). Measures were assessed within 1 year of diagnosis unless stated otherwise. Breast cancer measures included receipt of: (1) HER2 testing within 3 months of diagnosis [NQF #1878]; (2) Tamoxifen/Aromatase Inhibitors for Stage I-III ER/PR+ [NQF #220]; (3) radiation among stage I-III breast conserving surgery or mastectomy patients [NQF #0219]. Stage II-III colorectal cancer measures included receipt of: (1) carcinoembryonic antigen (CEA) testing; (2) surveillance colonoscopy; and (3) computed tomography (CT) among resected patients [NCCN 2.2023 COL-8]. We assessed receipt of prostate-specific antigen (PSA) testing in the first 15 months after prostate cancer diagnosis to monitor disease recurrence. Poisson regression adjusting for sex, urbanicity, insurance type, and age at diagnosis was used to estimate risk ratios (RR) and 95% confidence intervals (CI) comparing the receipt of care between AI and WH patients. RESULTS The sample sizes of AI patients differed across measures, ranging from 104 colorectal to 554 prostate cancer patients. Evidence of disparity was mixed for the breast cancer measures. AI breast cancer patients less frequently received HER2 testing within 3 months of diagnosis (AI=61% vs WH=71%), although adjustment attenuated this association (RR=0.90; CI: 0.81-1.01). Among stage I-III breast cancer patients, AIs more frequently received radiation within 1 year of surgery (AI=75% vs WH=71%) compared with WH patients (RR=1.11; CI: 1.01-1.21). Across colorectal cancer measures, AI patients performed as well or better than WH patients. Notably, AI patients more frequently received CEA testing (AI=71% vs WH=59%), a difference that persisted after adjustment (RR=1.17; CI: 1.03-1.32). AI prostate cancer patients received PSAs less often in the 15 months following diagnosis (AI=82% vs WH=88%) than WH patients (RR=0.96; CI: 0.93-1.00). CONCLUSIONS The magnitude and direction of AI/WH disparities in receipt of quality cancer care differed across measures. Racial misclassification is a challenge in AI health outcomes research, which may misrepresent the experience of AIs. Nevertheless, our findings provide insight into specific areas of breast, prostate, and colorectal cancer care where local tailored interventions can be focused to improve care equity. The microbial diversity of wastewater used for irrigation and fertilization was assessed using specific polymerase chain reaction (PCR) assays to detect and genotype several pathogenic protists including Cryptosporidium spp., Giardia duodenalis, Cyclospora spp., Eimeria spp. and Enterocytozoon bieneusi. A total of 220 wastewater samples (110 raw, 110 treated) and 12 sludge samples were collected from 2005 to 2008 from 18 treatment plants located throughout Tunisia. Except for Cyclospora, which was detected only once, E. bieneusi (61%), G. duodenalis (28%), Cryptosporidium spp. (27%) and Eimeria spp. (45%) were frequently observed in wastewater and sludge. Sequencing of PCR products showed that C. hominis, C. andersoni, G. duodenalis sub-assemblage A-II and E. bieneusi genotypes D and IV were the most prevalent. An analysis of the distribution of 209 internal transcribed spacer sequences of E. bieneusi originating from wastewater at the 18 treatment plants showed a similar genetic diversity, regardless of the geographical location. The identification of these parasite species and genotypes and of host-specific Eimeria species indicates that the microbial quality of wastewater was impacted by humans, livestock and rodents. Given the public health risks that some of these parasites represent, guidelines on wastewater usage are needed to minimize human exposure to these pathogens.

Due to challenges in manufacturing composite cathodes with oxide solid electrolytes, new cell concepts are emerging in which the infiltration of solid-polymer electrolyte (SPE) into 3D cathode pore structures improves capacity retention and cycling stability. However, the performance limitation and the resulting practical relevance of such a hybrid concept have not yet been analyzed and discussed. This study investigates the impact of laser-ablated geometric structures on the performance of hybrid solid-state batteries (SSBs). A Doyle–Fuller–Newman modeling approach is developed and parameterized for structured hybrid SSBs that incorporate a PEO/LiTFSI SPE and an LLZO ceramic separator, as well as NMC-811 and Li-metal for the positive- and negative-electrode active materials. Comparison between structured and planar cell designs reveals significant rate capability improvements in structured designs due to reduced diffusion and interfacial charge transfer polarization. A sensitivity analysis of geometric structure parameters shows further potential for performance improvement in terms of specific capacity and energy density. However, current constriction effects in the LLZO separator can deteriorate the rate capability. A more general perspective is then taken by analyzing the impact of changing SPE parameters. An energy density of 128 Wh kg−1 at 1C, and 220 Wh kg−1 at 1C with improved SPE parameters is achieved in the best case, approaching the target of 250 Wh kg−1, which is currently achieved for conventional Li-ion batteries.

Different body dimensions were compared between 16 normally muscled (NM) and 16 heterocygous double muscled (DM) Senepol heifers. Body weight (BW), withers height (WH), hip height (HH), thorarcic perimeter (TP), barrel (BA), shoulder to pin bone distance (SPD), and the internal pelvic width (PW), height (PH), and area (PA) were recorded in each heifer. Also included were the relationships of BW/body dimensions, except for dimensions PW and PH. Data were analyzed by the GLIMMIX and CORR procedures of SAS. The respective genotypes MN and DM did not differ (P>0.10) in BW (415 and 434 kg), WH (both 113 cm), HH (both 120 cm), TP (both 175 cm), B A (209 and 214 cm), B W/B A (1.99 and 2.03 kg/cm), SPD (both 147 cm), PW (both 13 cm), PA (212 and 220 cm2), and BW/PA (1.96 and 1.98 kg/cm2). However, DM heifers tended to present (P<0.10) higher values than NM heifers for BW/WH, BW/HH, BW/SPD, and PH by margins of 0.14 kg/cm, 0.15 kg/cm, 0.12 kg/cm, and 0.59 cm, respectively. Moreover, the BW/TP value was 0.09 kg/cm greater (P<0.05) in the DM than in NM heifers. The correlations between different pairs of body dimensions were lower in the DM than in the NM heifers. These results suggest that the enhanced musculature visually observed in DM heifers must be accompanied by hypotrophy of other body organs and tissues, but not including the pelvic region. This compensatory effect minimizes the posibility of observing differences in body dimensions in vivo between DM and NM heifers.

Peningkatan konsumsi bahan bakar minyak terjadi setiap tahunnya, hal ini disebabkan oleh peningkatan jumlah penduduk. Untuk mengatasi permasalahan tersebut, dikembangkan bahan bakar alternatif seperti bioetanol. Penelitian ini dilakukan dengan menggunakan metode eksperimen untuk menganalisis unjuk kerja motor SI silinder tunggal berbahan bakar campuran bioetanol. Bioetanol dihasilkan dari nira tebu melalui fermentasi dengan bantuan bakteri Saccharomyces cerevisiae. Komposisi campuran bahan bakar yang digunakan adalah 5% bioetanol+95% Premium (Ron 88), 5% bioetanol+95% Pertalite (Ron 90) dan 5% bioetanol+95% Pertamax (Ron 92) yang kemudian dianalisis kinerja genset motor bensin. Hasil penelitian menunjukkan bahwa Premium sebesar 240,9 W memperoleh daya tertinggi dari campuran bioetanol. Konsumsi bahan bakar spesifik berkurang, diperoleh untuk Premium sebesar 1,417 kg/Wh, dan daya campuran bioetanol tertinggi diperoleh untuk Pertalite. Hasilnya menunjukkan daya tertinggi 218,67 W dan torsi 0,803 Nm. Efisiensi termal diperoleh sebesar 38,711% untuk Pertalite. Campuran bioetanol Pertamax memiliki tenaga dan torsi masing-masing 220 W dan 0,875 Nm. Sedangkan nilai konsumsi bahan bakar yang diperoleh adalah 1,573 kg/Wh, dan efisiensi termal campuran adalah 42,524%. Hasil dari penelitian ini akan memberikan wawasan tentang potensi penggunaan campuran bioetanol untuk aplikasi mesin generator dan menawarkan jalur untuk penelitian selanjutnya dalam rasio bioetanol yang berbeda untuk mewujudkan manfaat signifikan dari mesin generator untuk mesin pembakaran dalam.

Carbon nanotubes (CNTs) were doped by ammonium borate as the sources of nitrogen and boron. Under the protection of Ar gas, boron-nitrogen doped CNTs were prepared through nitriding and boronization at high temperature. It is a conductive additive. Then, the obtained CNTs were mixed with activated carbon (AC), SP, sodium dodecyl sulfate (SDS), and cellulose fiber to prepare electrodes. With all the materials, a symmetric electric double-layer supercapacitor (EDLC) was assembled. Next, the materials and electrodes were also characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM), and transmission electron microscopy (TEM). The factors, chemical connections, and specific surface area of the CNTs were analyzed by X-ray energy spectrum analysis (EDS), X-ray photoelectron spectroscopy (XPS), as well as a specific surface area and porosimetry analyzer (BET). In addition, the electrochemical performances of electric double-layer capacitors were tested with the help of cyclic voltammetry, constant-current charging and discharging, and so on. From the results, we can make a conclusion, that is, both B and N atoms were added into the CNTs and formed bonds successfully with carbon atoms mutually. Besides, the specific surface area is about 1.5 times than that of the CNT. When the charge/discharge current density reaches 50[Formula: see text]mA/g, we can find that the mass specific capacitance of the capacitor can run up to 32.19[Formula: see text]F/g. Also, we observe that the maximum power density is close to 220[Formula: see text]W/kg (700[Formula: see text]mA/g), and the energy density can arrive 9.31[Formula: see text]Wh/kg (50[Formula: see text]mA/g). Based on the impedance test, the electrodes are characterized with low impedance. After 2000 cycles, the boron-nitrogen doped double-layer capacitors maintain a capacitance retention ratio of above 95%. Its power density can still achieve 220[Formula: see text]W/kg when the energy density keeps at 3.46[Formula: see text]Wh/kg. In other words, the electrochemical performance functions of the electric double-layer capacitors are enhanced while the CNTs serve as the electrodes.

The primary concern of this paper is to investigatethe extent to which three variables (i.e. personality traits, demographic variables, and job satisfaction) are interrelated with each other and what effect they have on each other in relation to the UK retail sector. The four different types of retail stores i.e. Tesco, Primark, Ikea and WH Smith were selected for survey purpose to minimise the class biasness.Total 300 close-ended questionnaires were distributed and 220 responses were obtained.The findings reveal that ‘Neuroticism’ is negatively associated with job satisfaction as well as with ‘Extraversion’. However, it is positively correlated with other three personality groups including ‘Agreeableness’, ‘Conscientious’, and ‘Openness’. On the other hand, ‘Openness to experience’ has a negative relationship with ‘Agreeableness’.It is also found from the ranking analysis that employees with ‘Agreeableness’ and ‘Conscientiousness’ personalities are more successful in their career and consequently they are more satisfied with their jobs.

Bibliometric analysis studies of researchers about the use of rainwater as electrical energy are still very few. There is still no utilization of rainwater which is directly used for electrical energy. Analysis of the capacity of the electrical energy requirements of electronic devices shows that the electrical energy requirements for several types of laptops and mobile phones range from (3 – 18) milliwatts per second mWs or (11.02 – 63.13) watt-hour Wh. So that this mini hydropower can be used to turn on LED lights 220 V AC, laptops, and cellphones. Gutters at home that drain water when it rains have a much larger water discharge capacity than a miniature hydropower plant, of course, have enormous potential in independently supplying household electrical energy needs. In which the electrical energy can be used directly or stored in a power saver energy such as Accu, or power bank.

Les lymphocytes T CD4 (LT CD4) sont nécessaires à l'établissement d'une réponse anticancéreuse efficace en contribuant à l’induction des LT cytotoxiques spécifiques de la tumeur et à leur persistance. Cependant, le pronostic des patients peut être affecté différemment selon le type de LT CD4 infiltrant la tumeur (TIL). L'épigénétique joue un rôle important dans la régulation de la polarisation des LT CD4, de leur plasticité vers d'autres sous-ensembles et de leur maturation. Il a été démontré que l'utilisation de modulateurs épigénétiques pouvait réguler la différenciation des LT CD4. Ainsi, la manipulation des LT CD4 par des thérapies épigénétiques peut être utilisée comme une stratégie pour améliorer l'immunothérapie. Nos résultats ont confirmé la présence de LTh17 parmi les LT expandus provenant de métastases hépatiques du cancer colorectal. Le nombre des LTh17 présents dans les TIL expandus était corrélé à la survie des patients. Nous avons ensuite tenté d'évaluer l'effet des régulateurs épigénétiques sur la différenciation et la fonction des LT CD4. Le criblage d'une banque de régulateurs épigénétiques nous a permis d'identifier un activateur de Sirtuine 1, l’Agrimol B, capable de réduire la prolifération, la sécrétion de cytokines et l'expression de CCR6 à la surface des LT CD4. L’Agrimol B a ainsi permis de réguler la migration et la fonctionnalité des LTh17 provenant de donneurs sains. L'inhibition des caspases par notre molécule a empêché le clivage de Sirtuine 1, permettant ainsi le maintien de son activité dans les LT CD4 activés. Dans l'ensemble, ces travaux visent à mettre en évidence le rôle de l'épigénétique dans la régulation des différentes populations de LT CD4 afin de potentialiser l'efficacité de l'immunothérapie anticancéreuse
CD4 T-cells are necessary for the establishment of an efficient anti-cancer response by providing help for the priming and persistence of tumor-specific cytotoxic T lymphocytes. However, the prognosis of patients can be differently affected, depending on CD4 T-cell subtypes infiltrating the tumor (TIL). Epigenetics take an important part in : i) the regulation of CD4 T-cells polarization, ii) plasticity towards other subsets and iii) maturation. It has been shown before that epigenetic modulators could regulate CD4 T-cells differentiation. Thus, the treatment of CD4 T-cells by epigenetic therapies can be used as a strategy to improve immunotherapy. Our results confirmed the presence of Th17-cells in expanded T-cells issued from liver metastases of colorectal cancer. Moreover, the number of expanded Th17-cells among total TIL was inversely correlated with patients' survival. We then attempt to evaluate the effect of epigenetic regulators on both CD4 T-cells differentiation and function. A screening of a bank of epidrugs allowed us to identify a Sirtuin 1 activator, Agrimol B, that might downregulate proliferation, cytokine secretion and CCR6 expression on CD4 T-cells. Our results thus indicated that Agrimol B might regulate the migration and the functionality of Th17-cells from healthy donors. Mechanistically, the inhibition of caspases by our molecule could prevent the cleavage of Sirtuin 1 and thus maintain its activity compared to control conditions. Together, this work aims to uncover the role of epigenetics in the regulation of CD4 T-cell subsets in order to potentiate the effectiveness of cancer immunotherapy

HIV, syphilis, malaria, and anaemia are major causes of adverse pregnancy outcomes in sub-Saharan Africa (SSA). Despite global and national policies advocating for screening of these conditions, only HIV testing has achieved good coverage, precluding early detection and appropriate management in pregnancy. Rapid pointof-care tests (POCTs) provide an opportunity to integrate diagnosis and provide timely treatment of these conditions in rural antenatal care (ANC) settings. After an introductory chapter, a review of the literature on these four conditions in pregnancy is presented with a focus on SSA. The thesis then shifts attention to Kenya, a country that embodies many of the disease challenges and health system characteristics of the region. Kenyan ANC policy recommends testing for HIV, syphilis and anaemia and preventive strategies for malaria. The following chapters are comprised of three linked studies conducted in western Kenya, that use different methods to progressively investigate the implementation success of integrated point-of-care testing (POCT) for HIV, syphilis, malaria and anaemia at seven peripheral dispensaries. Baseline data confirmed that testing requirements for syphilis, malaria and anaemia are not currently met at dispensary level. We implemented an intervention where test kits were supplied and training plus supervision were provided to enable healthcare workers to conduct integrated POCT for pregnant women. Adoption and fidelity were measured quantitatively using exit interviews, antenatal registers and proficiency scores (Study 1: Integrating point-of-care testing (POCT) for HIV, syphilis, malaria and anaemia in antenatal care at dispensary level in western Kenya: an implementation study) while acceptability, appropriateness and feasibility were assessed qualitatively (Study 2: Exploring healthcare workers and pregnant women’s perspectives on appropriateness, acceptability and feasibility of integrating point-of care testing: A qualitative study). Our findings show that the innovation was highly adopted, meaning almost all pregnant women received the essential tests. This was supported by the qualitative findings where healthcare workers and pregnant women found the innovation acceptable and appropriate. However, fidelity to clinical management guidelines can still be improved. Our qualitative findings provide some explanation for these gaps. One common sentiment among interviews with healthcare workers was that workload was perceived to be a barrier to providing quality care. We explored this further with discrete-event simulation modelling (Study 3: Investigating the operational impact of integrating HIV, syphilis, malaria and anaemia point-of-care testing in antenatal care clinics in western Kenya: a discrete event simulation model) and found the healthcare workers were actually under-utilized. This suggests that nurses should, in theory, have sufficient time to deliver essential ANC services. While integrating POCT addresses one gap, additional interventions to support and supervise healthcare workers are needed to ensure appropriate and high quality of care. An integrated approach to health systems strengthening and more investment in implementation and translation research using multi-methods are needed.

Nous avons produit des CART-cells et des LT-mock ont été produits ex-vivo. L'efficacité de transduction est mesuré par l'expression CD3+/CD19+ (82% en moyenne, n=S), en cytométrie en flux. L'expression protéique de CAR a été vérifiée par western blot et en cytométrie en flux attestant de son expression membranaire. Nous avons testé la fonctionnalité de la cassette suicide iCaspase9/ AP1903 sur CART-cells après 48h de traitement à I' AP1903. Nous avons montré que plus de 90% des CART-cells entrent en apoptose (Marquage 7-AAD en cytométrie en flux). Nous avons ensuite réalisé des tests fonctionnels in-vitro des CART-cells contre des lignées LMC exprimant naturellement IL-lRAP ou contre des lignées transfectées pour exprimer la forme membranaire d'IL-lRAP. Après co­culture des CART-cells en présence des cellules cibles IL-lRAP+ nous avons montré qu'ils prolifèrent (test CFSE), qu'ils sécrètent de l'interféron y et des cytokines inflammatoires. Enfin, par marquage 7-AAD, nous avons démontré la cytotoxicité des CART-cells contre les cellules ILl-RAP+. Finalement, nous avons montré l'efficacité des CART-cells in vivo, dans un modèle de xénogreffe tumorales dans des souris immunodéficiences NSG greffées par la lignée K562-IL1RAP+ / Luciférase +. On constate une diminution de la masse tumorale 4 jours après injection des CART-cells, jusqu'à disparition totale. Cette preuve de concept laisse entrevoir des perspectives thérapeutiques alternatives dans le traitement de la LMC ou LAM, qui devront être optimiser dans des modèles murins (séquence et nombres d'injection, nombres de cellules, associations avec ITKS ou autres chimiothérapies) afin de conduire un essai clinique de phase I, pour démontrer la faisabilité et la sécurité de l'approche pour envisager une démonstration d'efficacité chez l'homme. La sécurisation par la cassette suicide permet d'envisager l'utilisation des CART-cells en situation autologue ou allogénique (Donor Lymphocytes infusion, DU)
Chronic myeloid leukemia (CML) is a myeloproliferative disorder and is characterized by the presence of a Philadelphia chromosome (Phl) encoding the BCR-ABL fusion protein with constitutive tyrosine kinase activity. The treatment by Tyrosine Kinase lnhibitors (ITK) is not curative. The problem today is ta target leukemic stem cells (HSCs) to prevent relapse of patients after stopping treatment with TKI and permanently eradicate the disease. Studies have identified the interleukin-1 accessory protein receptor (IL-lRAP) as a potential target for killing CSL (BCR-ABL1 positive) in CML or in acute myeloid leukemia (AML). We therefore propose to develop a cell therapy approach targeting IL-lRAP, using T cells (LT) redirected by a CAR (Chimeric Antigen Receptor) in the treatment of CML and AML. We produced a specific anti-lL-1RAP murine monoclonal antibody (mAb) called # A3C3. We tested the specificity of the antibody # A3C3 in flow cytometry, in Western blot, by immunohistochemistry or confocal microcopy, on positive human cell lines IL-1RAP (KU812) or IL-1RAP negative (Raji). Moreover, by using this antibody, we have demonstrated by immunohistochemistry, on 20 different normal tissues that our antibody does not recognize or very few non-specific (off-target) targets. Finally, we demonstrated that this antibody is able to detect positive IL-1RAP CSLs in primary bone marrow or peripheral blood samples of LMC patients at diagnosis or after ITK treatment. By molecular biology, we have sequenced and identified rearrangements of the heavy (lgH) and light {lgL) chains coding region for hypervariable regions of# A3C3. The coding sequence for the "single chain variable fragment" (scFv), lin king the 2 lgH and lgl chains, was cloned into a 3rd generation lentiviral skeleton securised by a suicide gene, inducible caspase 9 (iCASP9). We produced CART-cells and LT-mock ex-vivo. The transduction efficiency is measured by CD3 + / CD19 + expression (82% on average, n = 5), in flow cytometry. The functionality of the suicide cassette iCaspase9/AP1903 on CART-cells after 48h treatment with AP1903 was tested. We have shown the death of more than 90% of CART-cells (7-AAD labeling in flow cytometry). We then performed in-vitro functional tests of CART-cells against LMC lines naturally expressing IL-1RAP or against transfected lines to express the membrane form of IL-1RAP. After co-culture of CART-cells in the presence of IL-1RAP + target cells, we have shown that they proliferate (CFSE test), that they secrete interferon y and inflammatory cytokines (intracellular labeling and Cytokines Bead Array assay). They are able to degranulate (CD107a & b labeling). Finally, by 7 AAD labeling, we demonstrated the cytotoxicity of CART-cells against IL1-RAP + cells. Finally, we showed the effectiveness of CART-cells in vivo, in a tumor xenograft model in NSG immunodeficiency mice engrafted by the K562-IL1RAP+/Luciferase+ line. There is a decrease in the tumor load 4 days after injection of CART-cells, until complete disappearance.This proof of concept suggests alternative therapeutic perspectives in the treatment of CML or AML, which should be optimized in murine models (sequence and injection numbers, cell numbers, associations with ITKS or other chemotherapies) in order to Phase I clinical trial, to demonstrate the feasibility and safety of the approach to consider a demonstration of efficacy in human. Securing by the suicide cassette makes it possible to consider the use of CART-cells in an autologous or allogeneic situation (Donor Lymphocytes infusion, DU)

L’histoire naturelle du cancer implique des interactions entre la tumeur et les mécanismes de défense de l’hôte, tout particulièrement avec le système immunitaire adaptatif. Ainsi la transformation de cellules normales en cellules malignes peut engendrer l’expression d’antigènes tumoraux reconnus par les lymphocytes T. Plusieurs sous-populations de lymphocytes T (LT) CD4 contrôlent les réponses antitumorales, parmi elles, les LT CD4 helper de type-1 (Th1) jouent un rôle activateur majeur de l’immunité à médiation cellulaire antitumorale. Ils deviennent actifs grâce à la reconnaissance des peptides de 15 à 20 acides aminés dérivés d’antigènes tumoraux et présentés par les molécules HLA de classe II. Ils sont nécessaires à l’induction et la fonction des cellules effectrices dirigées contre les tumeurs notamment les lymphocytes T CD8 cytotoxiques (CTL). De plus la présence de lymphocytes CD4 Th1 infiltrant les tumeurs est souvent associée à un bon pronostic chez les patients. A l’aide d’un modèle in vitro chez l’homme et in vivo chez des souris transgéniques HLA, nous avons identifié quatre nouveaux peptides CD4 dérivés de la télomérase (TERT) un antigène de tumeur exprimé dans la majorité des cancers humains. Ces peptides appelés «Universal Cancer Peptide, UCP» se lient à la majorité des allèles HLA-DR et sont capables d’activer spécifiquement les LT CD4 de type-1. Des LT CD4 circulants spécifiques des UCP sont naturellement détectables dans plusieurs cancers humains mais absents chez des individus sains. Des clones T CD4 spécifiques des UCP générés à partir des lymphocytes de patients, produisent de forts taux d’IFN, TNF, et d’IL-2, cytokines associées à la polarisation Th1. L’analyse par ELISPOT IFN, de LT CD4 anti-UCP circulants au sein d’une cohorte de 84 patients atteints de cancers bronchiques métastatiques a montré la présence naturelle de ces lymphocytes chez 38 % des patients. De plus un effet bénéfique de la présence de cette réponse sur la survie globale a été observé chez les patients ayant une réponse clinique objective après chimiothérapie (13 vs 10 mois, P< 003). In vivo, l’immunisation de souris transgéniques HLA-A2/HLA-DR1 (Tg A2/DR1) avec les peptides UCP stimule des réponses T CD4 spécifiques caractérisées par une polarisation Th1. Nous avons montré que la présence in vivo de LT CD4 anti-UCP est nécessaire pour l’induction de réponses CTL antitumorales efficaces. Ainsi chez des souris co-immunisées en présence d’un peptide UCP, on observe un accroissement en nombre et de la qualité des réponses CTL proportionnellement à l’aide délivrée par les LT CD4 anti-UCP. L’induction de LT CD4 anti-UCP s’accompagne également d’une activation des cellules dendritiques in vivo via un mécanisme impliquant CD40L, IFN et GM-CSF. Dans un modèle de mélanome transplantable chez les souris Tg A2/DR1 nos résultats ont montré qu’une vaccination thérapeutique comportant un peptide UCP favorise un meilleur recrutement de CTL fonctionnels dans les tumeurs et améliore ainsi l’efficacité antitumorale du vaccin. Ces résultats confirment le rôle antitumoral majeur des lymphocytes CD4 Th1 et soulignent l’intérêt clinique de stimuler des réponses T CD4 spécifiques d’antigènes tumoraux de relevance clinique comme TERT
Recent advances in immunology have now validated the concept of cancer immunosurveillance and the leading role of adaptative T cell immunity. Until a few years ago, antitumor CD8 T cell responses have been the most studied due to their direct cytotoxic activity on tumor cells. On the other hand, study of antitumor CD4 T cell responses are even more challenging because of the heterogeneity and plasticity of the various CD4 T cells subpopulations described. Among them, CD4 T helper type-1 cells (Th1), mainly characterized by the production of IFN, control the activation of antitumor cellular immunity. Thus, stimulation of specific CD4 Th1 cells may have a major interest for the development of anticancer immunotherapies. During this research thesis, we characterized novel HLA class II epitopes derived from a relevant tumor antigen, telomerase (TERT), and studied their capacities to stimulate specific CD4 Th1 cell responses. Using a method based on predictive immunology, we identified 4 peptides derived from TERT, referred as « Universal Cancer Peptides » (UCPs), enable to bind the most commonly found HLA-DR alleles in human. Using HLA-A2/HLA-DR1 transgenic mouse model, we first evaluated the in vivo immunogenicity of these peptides. Immunization of mice with UCPs induces high avidity specific CD4 T cells. The study of their polarization showed that UCP-specific CD4 T cells do not produce IL-4, -5, -10 or -17 cytokines, excluding a Th1, Treg or Th17 differentiation. In contrast, we measured high amount of IFN and IL-2 which characterize a Th1 pattern. The study of helper role allow us to demonstrate that CD8 peptide-based vaccinations in presence of UCPs enhance the efficacy of tumor specific CTL responses. Indeed, the intensity of these responses is strongly correlated with that of UCP-specific CD4 T cells induced in vivo. Furthermore, the stimulation of UCP-specific CD4 T cells promotes activation and IL-12 release by dendritic cells through a mechanism that involves IFN, GM-CSF and CD40L. We also demonstrated the antitumor efficacy of UCPs during a therapeutic vaccination in mice, as well as their capacity to foster the recruitment of specific CD8 T cells at the tumor site. In addition, the presence of naturally occurring UCP-specific CD4 T cell responses was found in different types of cancers such as leukemia, lung, colorectal or renal cancers. A study conducted in a cohort of 84 metastatic lung cancer patients revealed a synergistic effect of spontaneous UCP-specific CD4 Th1 and chemotherapy-treatment. Altogether, this study provides further evidences that stimulation of antitumor CD4 Th1 cells is a powerful method to improve cancer vaccines and also highlights the interest of TERT-derived UCPs for the innovative monitoring of antitumor CD4 T cell responses

Western Himalayas (WH) have experienced a two-fold temperature increase compared to the Indian sub-continent post-2000, strongly linked to global warming with significant implications for precipitation patterns. Using ERA5 reanalysis, we examine seasonal precipitation changes in the WH between recent (2001–2020) and past decades (1961–2000). Mean summer precipitation has increased over foothills but declined at higher elevations, while winter precipitation has increased region-wide except in certain parts of Jammu-Kashmir (JK), Uttarakhand (UK), and Punjab. In summer, light precipitation has increased in JK, while moderate precipitation has decreased over foothills but enhanced at higher altitudes. Moreover, extreme precipitation has significantly increased in the UK and Himachal Pradesh. During winter, light and extreme precipitation has increased, while moderate and heavy precipitation declined. Maximum one and five-day precipitation extremes (Rx1day, Rx5day) have increased in the foothills with more consecutive wet days. Winter extremes have increased in the northern region, while consecutive dry and wet days have declined, except for specific areas in eastern Ladakh and JK. Furthermore, rising sea surface temperatures, enhanced moisture transport, increased precipitable water and cloud cover in WH are associated with increasing mean and extreme precipitation, emphasizing the impacts of global warming on temperature and precipitation transitions in the region.

Recent exacerbation of extreme precipitation events (EPEs) and related massive disasters in western Himalayas (WH) underpins the influence of climate change. Such events introduce significant losses to life, infrastructure, agriculture, in turn the country’s economy. This chapter provides an assessment of long-term (1979–2020) as well as recent changes (2000–2020) in precipitation extremes over WH for summer (JJAS) and winter (DJF) seasons. Different high-resolution multi-source climate datasets have been utilized to compute the spatiotemporal trends in intensity and frequency of EPEs. The hotspots of rising extremes over the region have been quantified using the percentile approach where daily precipitation exceeds the 95th percentile threshold at a given grid. The findings reveal geographically heterogeneous trends among different datasets; however, precipitation intensity and frequency show enhancement both spatially and temporally (though insignificant). For both seasons, dynamic and thermodynamic parameters highlight the role of increased air temperatures and, as a result, available moisture in the atmosphere, signifying the consequences of global warming. Rising precipitation extremes in summer are sustained by enhanced moisture supply combined with increased instability and updraft, due to orography, in the atmosphere whereas winter atmosphere is observing an increase in baroclinicity, available kinetic energy, vertical shear and instability, contributing to a rise in precipitation extremes.

O objetivo deste estudo é avaliar o desempenho térmico de um sistema de aquecimento e resfriamento passivo com baixo consumo de energia, composto por um módulo de aquecimento e resfriamento radiante capacitivo, o qual é alimentado por um coletor solar/radiador noturno. São apresentados os resultados preliminares de uma avaliação experimental do sistema, realizado com células-teste em Curitiba, PR. São analisadas as temperaturas características, o potencial de resfriamento médio do sistema e sua capacidade de reduzir a demanda por resfriamento no verão. O potencial de resfriamento radiante noturno da localidade é representado pela depressão da temperatura do céu (ou seja, a diferença entre a temperatura ambiente e a temperatura do céu). A capacidade de resfriamento do sistema foi confirmada, atingindo-se valores médios de até 220 Wh/m2.dia. O sistema foi capaz de reduzir o sobreaquecimento em até 97% do tempo, com um consumo de energia muito baixo.

This paper presents findings from a current research project titled “Quantifying the Effect of Prestressing Steel and Concrete Variables on the Transfer Length in Pretensioned Concrete Crossties” that is funded by the Federal Railroad Administration (FRA). Specifically, the paper focuses on the effect of concrete properties on the resulting transfer lengths. These properties include concrete consistency (slump), compressive strength at the time of prestress transfer, the water-to-cementitious (W/C) ratio, the aggregate type, and the use of a viscosity-modifying admixture (VMA). Pre-tensioned concrete prisms were cast in the laboratory and transfer lengths were determined from surface strain measurements that were obtained prior-to and immediately after prestress transfer (de-tensioning). The concrete compressive strength at de-tensioning was determined using cylindrical concrete test specimens that were match-cured to the temperature of the pre-tensioned concrete members. The release strengths investigated were 3500 psi (24.13 Mpa), 4500 psi (31.03 Mpa), and 6000 psi (41.37 Mpa). The effect of concrete consistency on the transfer length was evaluated by varying the slump between 3″ (76.20 mm) and 9″ (228.60 mm) while maintaining release strength of 4500 psi (31.03 Mpa) and a W/C ratio of 0.32. The effect of W/C ratio on transfer length was evaluated by maintaining release strength of 4500 ±220 psi (31.03 ± 1.52 Mpa) and a slump of 6 ± 1/2″ (152.40 ± 12.7 mm) while varying the W/C between 0.27 and 0.42. These values represent the extreme values used in the North American concrete tie industry that were noted by the authors during research that was conducted in 2010–2011[15]. Results for each parameter type will be compared and discussed in this paper. Transfer length results obtained during earlier work [4] conducted by the authors at a W/C ratio of 0.32 will be compared. Finally, results will be presented from transfer length measurements that were obtained on identical-sized prisms that were manufactured with concrete mixtures that used different aggregate sources and also the use of a viscosity-modifying admixture. Work presented in this paper was conducted on two wire samples with generic labels [WG] and [WH]. Laboratory prism specimens of size 3 ½″ (88.9 mm) × 3 ½″ (88.9 mm) each with 4 wires were cast to measure transfer lengths. These proportions were chosen to replicate the original concrete crosstie wire-to-concrete proportions. Potential usage of these prisms in estimating transfer lengths was validated in another phase [5] and is not discussed here. Essential information obtained from results allowed researchers to discuss attributed influence of each concrete property on transfer length. The research knowledge acquired from this study will give proper insight about transfer length and will be helpful to manufacture a better product by adjusting concrete mix design.

Duas formas de análise da taxa de desempenho de sistemas fotovoltaicos são o cálculo convencional e a utilização de "Photovoltaic for Utility Systems Applications" (PVUSA). Portanto, este estudo aborda uma análise de quatro painéis fotovoltaicos de quatro tecnologias distintas instalados em Ponta Grossa- PR por meio destas duas metodologias. Para a realização das análises, foram coletados dados de geração, de irradiação no plano inclinado, velocidade do vento, e temperatura ambiente. Primeiramente, os dados de geração mensal e irradiação média mensal foram utilizados para o cálculo da taxa de desempenho convencional. Em um segundo momento, para a utilização da metodologia PVUSA nas condições "PVUSA Test Conditions"(PTC), os dados horários foram separados em valores de irradiação no plano inclinado superiores a 700 Wh/m² (PVUSA 700) e superiores a 800 Wh/m² (PVUSA 800). Como resultado, os valores obtidos por PVUSA 800 obtiveram menores diferenças percentuais em relação a metodologia de cálculo convencional do que os com PVUSA 700. Além disto, no caso da utilização da metodologia convencional e da PVUSA 700 a tecnologia com melhor taxa de desempenho foi telureto de cádmio (CdTe), e a tecnologia com menor taxa de desempenho em dois meses foi policristalina (p-Si).
