Der volle Inhalt der QuelleExternal radiotherapy is widely used in the treatment of cancers of head and neck cancers. The main side effect is osteoradionecrosis, which can lead to a major deterioration in the quality of life. Low-intensity pulsed ultrasound (LIPUS) has been described in the stimulation of bone healing in traumatology and orthopaedics. They would have an effect on the early vascular phase of bone consolidation. We therefore wanted to study, through an animal study, their impact on the healing of irradiated maxillary bones. For this, we have developed a model of radiotherapy in rabbits, in order to study the healing after creation of a bone defect on an irradiated mandible. Irradiation was performed according to the following protocol : 8.5Gy/week, with one session per week, for 5 weeks, for a total dose of 42.5Gy. Mandibular surgery consisted of performing a standardized bone defect immediately after irradiation. A control group benefited from the same surgery, without prior irradiation. With this model we obtained an alteration in bone healing, confirmed by histology and microscanner. Once the radio-induced bone alterations were highlighted, we were able to renew the protocol by adding an additional step of applying LIPUS to the surgical sites. The protocol consisted of the postoperative application of LIPUS, for 10 sessions, during 20 minutes, with the following parameters : 30mW/cm², pulse 1:4, 20 min, 1MHz. Application of LIPUS on the irradiated rabbit model did not appear to have a beneficial effect on bone healing : in histology, healing did not appear to be improved after application of LIPUS ; on the microscan, some parameters even seemed to be negatively impacted by LIPUS. We can however assume an interesting effect on the general state of health of the animals, with in particular an increase in their weight and their water consumption. Our results should be interpreted with great caution. Indeed, the sample studied is very small, and there is a lot of heterogeneity between the different subjects. In addition, several limitations that could question the validity of this study should also be taken into consideration. In the end, even if the LIPUS technology does not seem beneficial on bone healing in irradiated terrain with regard to the results that we obtained, this work validates an animal radiotherapy model. This model could be used to test other possible therapies in the future

Der volle Inhalt der QuelleBone metastases are common complications of advanced breast cancer. They increase morbidity of patients and alter their quality of life. Bisphosphonates (BPs) stop the progression of osteolysis. However, BPs do not affect the tumor burden located inside the bone marrow cavity. Antitumoral effects have been shown but with high doses incompatibles with a clinical use. BPs bind also avidly to bone mineral which limits their bioavailability and reduce their antitumoral potential in vivo. This work is incorporated within the framework of the enhancement of antitumoral effects of BPs. BPs were combined with low intensity ultrasound (LIUS), which are known to induce a mechanical stimulation and a slight increase of temperature without involving cavitationnal effect. Initially, LIUS were found to increase the penetration of BPs inside several mammary tumor cell lines without affecting their viability by increasing endocytosis. In vivo, a daily repeated treatment of LIUS associated with a single and clinical dose of BPs lead to a decrease in osteolysis as well as tumor burden. The accumulation of unprenylated Rap1A form was found in bone marrow of mice suggesting that LIUS promote BPs penetration inside cells of the bone cavity. The effect of BPs and LIUS was evaluated in a subcutaneous mammary tumor xenograft. Tumor growth was slowed during the first days of LIUS treatment. A study was performed using doxorubicin and BPs leading to a better penetration of both compounds when LIUS were used. This last result allows increasing the field of application of LIUS for breast cancer treatment. In conclusion, this work showed that LIUS are an interesting method to enhance the penetration of drugs inside bone and mammary tumors leading to an increase of their antitumoral activity

Der volle Inhalt der QuelleThis work concerns the valorization of Lebanese apples by evaluating its potential to produce cider, specifically the fermentation stage of this process. For this purpose, the chemical composition of the Lebanese apples “Ace spur” was analyzed in comparison with a cider apple “Kermerrien” used industrially in France for cider production. The Lebanese apple variety “Ace spur” appears to be suitable for cider making. In addition, a strain responsible for the spontaneous fermentation of this variety of apples was isolated and identified. Its fermenting potential in these two varieties of apples has been studied. The isolated yeast Hanseniaspora sp. can ferment apple juices up to 4% ethanol (v,v). This study will provide first-time data on the chemical composition of the Lebanese apple, on the potential of a strain present during its fermentation. Also, this work evaluates the impact of emerging technologies like pulsed electric field and ultrasound on yeast fermentative activity. A continuous apple juice treatment that can be scaled from laboratory to industrial scale is proposed. The various results obtained showed the chemical potential of the Lebanese variety “Ace spur” in cider production. The application of those emerging technologies can accelerate growth kinetics while reducing ethanol contents
