Središnja tema rada usmjerena je na oblike povezanosti iseljenika s otoka Hvara u hrvatskoj bratovštini sv. Jurja i Tripuna u Mlecima u razdoblju od 15. do 18. stoljeća. Rad se zasniva na obradi gradiva pohranjenoga u Državnome arhivu u Mlecima (oporuke) i Arhivu bratovštine sv. Jurja i Tripuna (spisi vezani za prihode i rashode bratovštine te popisi članova i dužnosnika u bratimskim vijećima i odborima). Uvidom u gradivo razvidno je da su Hvarani stoljećima predstavljali jednu od najvažnijih hrvatskih regionalnih skupina u gradu na lagunama, a u 17. stoljeću može se kazati da su, prema ugledu i učestalosti spominjanja, prednjačili nad drugim bratimima zavičajem od Dalmacije do Boke kotorske. U prilogu se donosi popis svih dosad istraženih Hvarana uključenih u bratovštinu sv. Jurja i Tripuna.

Annotation:
The author discusses a marble pluteus discovered in 1906 in the Kotor cathedral, where it was incorporated in the main altar as a spolium. Apparently, it originally belonged to the chancel screen in the sanctuary of St Tryphon, constructed in the early 9th century, of which only remnants of the foundation layer have been preserved. The pluteus has been analysed on several occasions, with the results published in scholarly literature, and most experts have justly dated it to the early period of pre-Romanesque sculpture in Dalmatia. Its front features a relief with geometric and vegetal ornaments. To the left, there is a panel with knotted triangles along the edge and rhombuses in the central section. The panel to the right contains two arcades with crosses underneath. Special attention has been paid to the relatively numerous ornaments on similar plutei in areas of Adrio-Byzantine tradition, in Roman cities of the Istrian and Dalmatian coastlines. Thereit combines with influences from Lombard and Carolingian centres, and in this combination it is also present in the area of the ancient Croatian state and other Sclavinias. The pluteus of Kotor is a telling element in the discussion on numerous similar relief arrangements and ornaments in the sacral art of the Adriatic cultural circle during the early medieval period.

U središtu istraživačkog zanimanja ovog rada jest prisutnost i djelovanje iseljenika iz Trogira u Mlecima, posebice s obzirom na njihovu uključenost u tamošnju hrvatsku nacionalnu bratovštinu sv. Jurja i Tripuna. Rad je zasnovan na istraživanju gradiva pohranjenog u Archivio di Stato di Venezia (fond: Notarile testamenti) i Archivio della Scuola Dalmata dei Santi Giorgio e Trifone (fondovi: Capitolar della Veneranda Scuola dei SS. Giorgio e Trifone della Nation Dalmata i Libri conti e spese). U radu se, uz osnovne podatke o prisutnosti Trogirana u Mlecima u širokom vremenskom rasponu od 15. do 18. stoljeća, raščlanjuju oporuke trogirskih iseljenika te konkretan udio te regionalne skupine hrvatskih iseljenika u nacionalnoj bratovštini. U tome dijelu rada osobita je pozornost posvećena kako onim Trogiranima koji su zabilježeni isključivo kao obični članovi bratovštine, ali i onima koji su – zahvaljujući svojem ugledu i priznatosti u hrvatskoj zajednici – obnašali dužnosti u pojedinim upravnim tijelima udruge. Naposljetku se zaključuje da su Trogirani, iako nisu u bratovštini sv. Jurja i Tripuna bili prisutni i djelatni kao Zadrani, Šibenčani, Splićani i Bokelji, činili jednu od nezaobilaznih sastavnica u sklopu proučavanja iseljavanja Hrvata u Mletke.

Tripuna prikazuju i analiziraju temeljne odrednice vezane uz demografska kretanja u Kotoru u vrijeme posljednjeg mletačko-osmanskog rata (1714. – 1718.). U mjeri u kojoj to dopuštaju izvori promatraju se promjene u kretanju broja rođenih, vjenčanih i umrlih u predratnome, ratnome i poslijeratnome razdoblju. Prikazuju se i veze broja rođenih, vjenčanih i umrlih s podrijetlom i zanimanjem aktera. Dobiveni kotorski rezultati uspoređuju se s kretanjima u Dubrovniku i Puli.

Seventeen microsatellite loci from the non-recombining region of the human Y chromosome were typed using AmpFlSTR® Yfiler® PCR amplification systems in 404 male subjects belonging to the three largest ethnic groups in Bangladesh. A total of 150 haplotypes from the Chakma, 144 from the Tripura, and 110 from the Khasia were detected with a corresponding discrimination capacity of 73.885%, 65.563%, and 81.250%, respectively. The highest allele frequency of 0.828 was detected in DYS391 locus in the Tripuras, while the lowest allele frequency of 0.009 was detected at the same locus for the Chakma population. The highest gene diversity (0.964) was observed at DYS385a/b locus in the Khasias, while the lowest gene diversity (0.301) was detected at DYS391 locus in Tripuras population. The overall haplotype diversity for the studied populations was 0.986141. Both the Neighbour-Joining tree and pairwise genetic distances showed that Chakma lies closer to a clade consisting of Tripuras (Khagrachari, Bangladesh) and Tripuri (Tripura, India). In contrast, the Khasias demonstrated a close affinity with the Oraon (Chhattisgarh, India), followed by the Santals. The Y-STR haplotype matching probabilities within and between populations demonstrated that the Chakma, Tripura, and Khasia were 100% genetically distinct. The studied ethnic populations exhibited higher frequency for haplogroups L and Q as opposed to haplogroups R1a, H, and L found in the mainstream Bengali population. The Median-joining networking showed haplogroups L and R1a have the most compact clustering within populations, followed by haplogroups Q and H. The presence of haplogroup R1a suggests that Bengali may have originated through west-to-east migration, whereas haplogroups L and Q distribution in the studied tribes reveal a very significant affinity with the South-East Asian populations and may have shared a common ancestral origin with the Mongoloid stock populations. The Tripuri tribe from the state of Tripura constitutes around 50 percent of the total tribal population and can be found in all eight districts of the state. The tribe follows its own culture and tradition in terms of marriage and other customary practices. This study investigates the role of gender in inheritance of property among the Tripuri tribe and how Tripuri women are excluded from ownership of property. It also attempts to discover how property ownership affects their income and position in the household. The study has been conducted in the districts of West Tripura and Dhalai. Focus Group Discussion and interview schedules are employed as methods for collection of data. Results show that while 20 out of 54 married women from rural areas of West Tripura have inherited property, only 2 out of 13 married women have inherited property in the urban area. In comparison with West Tripura, Dhalai features a low ratio among women in inheriting property (only 4 out of 38 married women). A few causes include low level of literacy, slow urbanization and less inter-community marriages. The reasons for not inheriting property include: a woman failing to live up to the concept of a ‘good sister’ in the brother’s eyes, son needs property to care for parents, cost of marriage is borne by brother or parents so no right to claim, and to avoid unnecessary conflict in the family. In this manner, societal perceptions prevent women from claiming the legitimate share of their ancestral property.

Introduction. Russian-Indian Anthropological expedition was organized in 2019 by Paleoethnology Research Center (Russia), Timiryazev State Museum of Biology (Russia) and Department of Anthropology University of Calcutta (India). The goal was to examine the peoples of Tripura state, North-East India. Data and methods. About 350 individuals from eight different locations next to Agartala were examined. Currently there are 19 tribes in Tripura, Tripuri being one of them. The most common Tripuri’s surname is Debbarma. During the research 254 Debbarmas were examined, among them 106 individuals were children and teenagers (58 male and 48 female), and 148 individuals were older than 17 years (76 male and 72 female). The Russian-Indian Anthropological expedition research consisted of a complex study of Tripuri including morphology of body, head and face, dermatoglyphics, and dental anthropology. Social questionnaire and anthropological photo were also obligatory. All data was hold according to bioethics and the Privacy law. For further examination the data was depersonificated. Results. During the survey, basic information about the individual, including tribal affiliation, was clarified. An extended questionnaire made it possible to study many aspects of social relationships and build a population-genetic network. According to somatometric characteristics, tripuri males from the Debbarma family have an average body length (162.1 cm), relatively broad shoulders and an average pelvic width. A cephalometric study of 19 features was carried out. The studied group can be characterized as brachycephalic, leptoprosopic according to the morphological facial index and mesorrhine according to the nasal index. The zygomatic diameter is large, and the mandibular diameter is above average (on the Indian scale of variability). A detailed fixation of cephaloscopic features in children and adults was carried out, which made it possible to describe tripuri as representatives of a large Asian-American branch (race) of humanity. According to the odontological program, 293 people were examined, 221 of which were tripuri. Anthropological photo-fixation was carried out, and based on these materials the first composite tripuri portraits were compiled, published in this article. Conclusion. In the second season of the Russian-Indian anthropological expedition, data were collected on one of the insufficiently studied peoples of Northeast India – the Tripuri tribe. The group was investigated according to the maximum number of anthropological methods available in the field. For the first time, somatometric, cephalometric and cephaloscopic materials have been obtained and preliminary presented.

Resumo: Os inibidores foram isolados de TOI em sementes de Copaifera langsorffii, conhecida vulgarmente por Copaíba, uma árvore nativa do Brasil da subfamília Caesalpinoideae da família Leguminosae. A fração TOI foi purificada a partir de precipitação com sulfato de amônio, cromatografia em uma coluna de troca-iônica em OEAE-Sepharose e coluna de afinidade em Sepharose- Trypsina. TOI apresentou-se homogêneo em gel de poliacrilamida-SOS 12,5%, com uma massa molecular aparente de 24 kOa em condições não reduzidas, e 12 kOa em condições reduzidas com OIT. A massa molecular relativa estimada por filtração em gel em uma coluna Superdex-75, sistema FPLC, confirmou os resultados obtidos em PAGE-SOS. TOI tem um Ki de 1,2nM para a enzima tripsina bovina mostrando uma alta afinidade. No entanto, TOI não mostrou atividade inibitória para quimotripsina. O estudo da estequiometria do complexo TOI-tripsina revelou que este inibe tripsina na razão molar de 1: 1. O complexo TripsinaTOI foi eluído na cromatografia de filtração em gel com um tempo de retenção equivalente a uma massa molecular relativa de 39 kOa. Evidenciou-se também que TOI é ativo na sua forma monomérica e não na sua forma dimérica. A Focalização Isoelétrica de TOI revelou a presença de duas bandas de proteínas, ambas coradas para inibidor de tripsina com pls de 6,45 e 7,35, indicando dois inibidores em TO!. Os inibidores de TOI, designadas por TOI-I e TOI-II, foram isoladas em coluna de fase reversa C18 /lBondapack, em Sistema HPLC Walters. Foi observado em gel de Tricina 16,5% uma diferença mínima na massa molecular aparente, sendo aproximadamente 12 e 10 kOa para TOI-I e TOI-1I respectivamente. O sequenciamento N-terminal dos inibidores de TOI revelou a presença da sequência consenso observada nos inibidores de tripsina tipo Kunitz para TOI-I, contudo, pesquisas nos banco de dados de proteínas não revelou nenhuma homologia para TOI-II. TOI mostrou alta estabilidade para diferentes condições de pH, temperatura e concentração de OTT, indicando um possível número elevado de pontes de dissulfeto na estrutura molecular. TOI não foi degradado proteoliticamente na digestibilidade in vitro pelas proteases encontradas no intestino médio do gorgulho Callosobruchus maculatus, e demonstrou um efeito deletério no desenvolvimento dos insetos, sugerindo que TOI atua como um fator antinutritivo e prejudica o desenvolvimento de Callosobruchus maculatus
Abstract: Two inhibitors have been isolated de TOI fraction in seeds of Copaifera langsorffii, ordinarily known as Copaíba, one Brazilian native tree of the Caesalpinoideae subfamily of Leguminosae family. The fraction TOI was purified by ammonium sulfate preeipitation, ion-exehange ehromatography on OEAE-Sepharose and affinity ehromatography on trypsin-Sepharose. TOI presented homogeneity in PAGE-SOS geI12,5%, with apparent moleeular mass of 24 and 12 kOa in non-and reduetion condition, respeetively. The relative moleeular mass determined by FPLC gel filtration on a Superdex 75 eolumn, eonfirmed on the results in PAGE-SOS. TOI have Ki of 1,2nM to bovine trypsin enzyme showing high affinity. However, no inhibition was found for quimotrypsin. The stoiehiometrie study of the trypsin-TOI eomplex evideneed that inhibitor is aetive for trypsin at 1: 1 molar ratio. The trypsin- TOI eomplex was eluted on a gel filtration ehromatography with retention time equivalent a relative moleeular mass of the 39kOa. Evidenced too, that TOI is active on enzyme trypsin on its monomerie form and not dimerie one. The isoelectrie foeusing of TOI demonstrated the presenee of two proteie bands, with isoelectrie point between 6.45 e 7,35, indicating two inhibitors in TOI. These inhibitors of TOI, designated for TOI-I and TOI-II, was isolated by reversed - phase HPLC on a J.L-Bondapaek column (Waters System) and was observed in Trieine-gel 16,5% one minimal differenee on apparent moleeular mass, of approximately 12 and 10 kOa for TOI-I and TOI-1I respectively. N-terminal sequeneing of the inhibitors of TOI, demonstrated the presenee of the eonsensus sequenee present in the soybean Kunitz-Type trypsin inhibitor for TOI-I, however, protein database searehes did not reveal any homology to TOI-II. The TOI showed high stability to pH different eonditions, temperature and amount of OTT, indicating one elevated number of disulfides bonds in a moleeular strueture. The TOI was not proteolytically degraded on a in vitro digestibility by midgut proteinases of the gorgulho Callosobruchus maculatus and demonstrated one deleterious effect on a insect's development, suggesting that TOI acts like toxie factor and deereased survival of Callosobruchus maculatus

Resumo: Algumas espécies vegetais podem apresentar em suas sementes, agentes bioquímicos que atuam no mecanismo de defesa da planta. Uma grande variedade destes são, na maioria, constituídos de proteínas e carboidratos. Dentre os agentes de origem protéica estão os inibidores de proteinases, que além da sua atuação em plantas, podem ter aplicações no tratamento de patologias, como inflamações, hemorragia e até mesmo o câncer. Neste trabalho, um inibidor de tripsina presente nas sementes de Dimorphandra mollis (Leguminosae-Mimosoideae), conhecida comumente por Faveiro-Doce, foi isolado e purificado a partir de precipitação com sulfato de amônio (30-60%), cromatografias em coluna de exclusão molecular (Sephadex G-75), troca iônica (DEAE-Sepharose) e de afinidade (Sepharose- Tripsina). A pureza do inibidor de D. mollis (DMTI-II) foi verificada por eletroforese em gel de poliacrilamida, que revelou uma única banda de proteína, mesmo em condições redutoras (O,IM DTT), indicando que DMTI-II é constituído por uma única cadeia polipeptídica. A massa molecular relativa de 25 kDa foi estimada por gel filtração em coluna de Superdex-75, em sistema FPLC. Uma constate de inibição de Ki de 1,7 x IO-IOM foi obtida para DMTI-II com a enzima tripsina bovina, mostrando portanto uma alta afinidade entre enzima e inibidor. Estudos estequiométricos revelaram que DMTI-II inibiu a tripsina na razão molar de 1:1, sendo a massa molecular do complexo formado de 46 kDa, determinada por cromatografia de filtração em gel, em coluna Superdex-75 (sistema FPLC). A atividade inibitória de DMTI-II mostrou ser altamente estável para diferentes condições de pH e concentrações do agente redutor DTT. A determinação da sequência N-terminal do inibidor que apresentou alto grau de homologia com outros inibi dores da família Kunitz, permitiu confnmar as características e propriedades mostradas durante o decorrer do trabalho. DMTI-II foi também submetido a ensaios de digestibilidade in vitro por pepsina e papaína e por enzimas presentes no intestino médio de larvas de Callosobruchus macu/atus com o intuito de verificar sua provável ação contra esta espécie de inseto. Os resultados mostraram que DMTI-II foi refratário à ação dessas enzimas, sugerindo uma possível aplicação na biotecnologia vegetal
Abstract: Some plants species can have in their seeds biochemical agents that act on their defense mechanism. A great number of them are of proteins or carbohydrates nature. Among the ones with protein origin are the proteinase inhibitors, that can also have a role on the treatment of pathologies, such as inflammation, hemorrhage and even on cancer. In this work, a trypsin inhibitor from Dimorphandra mol/is (Leguminosae Mimosoideae) seeds, commonly known as "Faveiro-Doce", was isolated and purified by ammnium sulfate precipitation (30-60%), gel filtration chromatography (Sephadex G-75), ionexchange chromatography (DEAE-Sepharose) and affinity chromatography (SepharoseTrypsin). D. mollis inhibitor's (DMTI-II) was assayed by polyacrylamide gel electrophoresis, yielding a single protein band, indicating that DMTI-II is constituted by one polypeptide chain. A molecular weight of25 kDa was estimated by gel filtration on Superdex G-75 column (FPLC system). An inhibition constant value of 1.7 x IO-IOM was obtained for DMTI-II with bovine trypsin, showing a high affmity between enzyme and inhibitor. Stoichiometric studies showed that DMTI-II inhibited trypsin in a I: I molar ratio. The molecular weight of the complex was determined as 46 kDa, by gel filtration chromatography in the Superdex G75 column. DMTI-II inhibitory activity was stable in the different pH conditions and reducing agent (DTT) concentration. Determination ofthe inhibitor N-terminal sequence showed a high degree of homology in relation to other inhibitors from the Kunitz fami1y, reinforcing the characteristics and properties found during this work. DMTI-II inhibitor was submitted to digestion assays in vitro by pepsin and papain and by enzymes present in the larvae midgut from Callosobruchus maculatus, to verify its probable _on against this insect specie. The results showed that DMTI-II was refractory to the action of those enzymes, suggesting a possible application on plant biotechnology

A artrite reumatoide é uma doença autoimune que em seu curso pode causar inatividade do paciente e levá-lo a óbito. Acredita-se que 1,5% da população mundial seja afligida pela doença, que não tem cura e os tratamentos disponíveis nos dias de hoje além de terem alto custo, podem causar efeitos adversos graves, limitando o tratamento da doença. A pesquisa em busca de medicamentos com menos efeitos adversos e de custo acessível faz-se então necessária. O presente estudo mostrou que a tripsina administrada em ratas pela via oral na dose de 2,95mg/Kg foi capaz de diminuir a dor e a inflamação articular em um modelo de artrite induzida por CFA, e que uma única administração foi capaz de manter o efeito antiartrítico durante os 7 dias de avaliação. Animais vagotomizados não tiveram melhora do quadro artrítico após a administração da tripsina na mesma dose anterior, indicando um papel vagal na atividade da enzima. Da mesma forma, a tripsina não teve efeito quando administrada em animais que receberam injeção intratecal das neurotoxinas 6-OHDA e 5,7-DHT. Esse resultado sugere o papel das vias descendentes inibitórias noradrenérgicas e serotoninérgicas, respectivamente, na ação da tripsina. As vias descendentes inibitórias monoaminérgicas não somente podem diminuir a nocicepção, como também, a partir da inibição do reflexo da raiz dorsal, seria capaz de controlar a inflamação articular. O presente trabalho propõe, então, que a ativação vagal seguida da liberação de monoaminas na medula pela via descendente inibitória compõem o mecanismo de ação da tripsina via oral, que pode ser considerada como uma alternativa de tratamento para doenças inflamatórias crônicas como a artrite reumatoide. Por extensão, esse trabalho sugere que ativadores orais de aferências do nervo vago subdiafragmático, e não absorvíveis, podem ser um novo ramo para o desenvolvimento de fármacos com menos efeitos colaterais.

O desdobramento no equilíbrio para a β-tripsina, induzido por desnaturação térmica, foi caracterizado termodinamicamente. O desdobramento foi acompanhado através de absorção no ultravioleta (UV) e espectroscopia de dicroísmo circular (DC) no UV próximo e distante. Além disso, experimentos utilizando a técnica de calorimetria diferencial de varredura (DSC) foram realizados. As curvas de transição térmica foram reversíveis mostrando um desdobramento altamente cooperativo, e os dados experimentais foram bem ajustados à um modelo de transição de dois-estados, para o desdobramento dessa proteína. O gráfico da fração desnaturada versus temperatura, calculado a partir das curvas de desnaturação térmica, em diferentes comprimentos de onda, mostra coincidência dessas curvas. Além disso, a razão entre a entalpia de desnaturação calorimétrica e a entalpia de van ́t Hoff, obtidas por DSC, é próxima à unidade, o que suporta o modelo de dois-estados. O espectro de DC da enzima nativa foi registrado na faixa espectral de 184 a 260 nm e utilizado para se estimar as porcentagens dos elementos da estrutura secundária, pelos métodos VARSCL e SELCON. A estimativa da estrutura secundária, em solução, correspondeu aos resultados de difração de raios-X. As mudanças na estrutura da β-tripsina, resultante de desnaturação por calor e por uréia foram monitoradas por meio de espectroscopia de DC no UV próximo e distante. A quantidade dos vários elementos de estrutura secundária foi estimada para essas mudanças espectrais, através dos espectros no UV distante, utilizando o método CCA (“Convex Constraint Algorithm”). De acordo com as estimativas, os estados desdobrados para essa proteína, ou seja, a 70 o C e em uréia 2,6 M, apresentaram quantidades consideráveis de estrutura secundária regular. Os espectros de fluorescência intrínsica dos resíduos de triptofano, registrados em diferentes concentrações de uréia, mostram a presença de um estado intermediário no equilíbrio, que apresenta grande afinidade pela sonda bis-ANS. Além disso, a ausência de contatos terciários e a presença de estrutura secundária para esse estado, verificados por meio de espectros de DC no UV próximo e distante, são consistentes com as características de um molten globule. As mudanças na atividade da β-tripsina foram observadas em concentrações de uréia nas quais não foram visualizadas nenhuma alteração espectroscópica da estrutura terciária da proteína. Os parâmetros cinéticos, k cat e K M , determinados na ausência e na presença de uréia, mostraram que o decréscimo na atividade da enzima, em baixas concentrações de uréia, foi provavelmente devido à formação do complexo EI, formado entre a β-tripsina e a uréia, um inibidor competitivo dessa enzima.
The unfolding equilibrium of β-trypsin induced by thermal denaturation was thermodynamically characterized. Thermal unfolding equilibrium were monitored using UV absorption and both far- and near-UV CD spectroscopy. In addition, experiments using differential scanning calorimetry (DSC), were performed. Thermal transition curves showed to be reversible and cooperative, and the data could be reasonably fitted using a two-state model for the unfolding of this protein. Plots of the fraction denatured, calculated from thermal denaturation curves at different wavelengths, versus temperature were coincident. In addition, the ratio of the enthalpy of denaturation obtained by scanning calorimetry to the van ́t Hoff enthalpy was close to one, which supports the two-state model. The dichroism circular (CD) spectrum of the native protein was taken over the wavelength range of 184- 260 nm, and the amount of various secondary structures were estimated using VARSLC and SELCON methods. The CD estimation of secondary strucutre, showed excellent agreement with X-ray results. The CD spectra of β-trypsin, as a function of temperature and urea, were monitored by far- and near-UV CD, and the relative proportions of secondary structures calculated by CCA method. According to CD estimation, although the secondary structure changed upon denaturation, its denatured state still has considerable amounts of ordered structure. The intrinsic fluorescence emission spectra of β-trypsin, at different urea concentration, showed an intermediate equilibrium state, with great affinity by bis-ANS. Besides, the absence of terciary contacts and the presence of secondary structure for this state, are consistent with the molten globule characteristics. Changes on the activity of β-trypsin were observed at urea concentrations in which terciary structure changes were not detected. Kinetic parameters, k cat and K M , determined in the presence and absence of urea, showed that the decreases in enzyme activity, at low concentrations of urea, were probably due to a development of the enzyme-inhibitor complex between β-trypsin and urea, a competitive inhibitor of this enzyme.

Uma nova isoforma tripsina bovina foi purificada a partir de amostras comerciais por cromatografia de troca iônica por Sephadex SP C50®. A nova isoforma contém, além da perda do hexapeptídeo N-terminal (como encontrado na molécula progenitora β-tripsina) uma clivagem intra-cadeia entre o Lys-155 e Ser-156. A nova enzima denominada -tripsina mostrou propriedades similares com a isoforma α-tripsina que contém o mesmo número de cadeia polipeptídica (duas cadeias), massa molecular (23312 Da), estrutura secundária, volume hidrodinâmico e outros. Apesar das semelhanças estruturais e enzimáticas dessas duas isoformas, -tripsina tem uma taxa de formação inferior a partir da β-tripsina, carga superficial inferior, mas a -tripsina tem estabilidade térmica mais elevada do que a α-tripsina. Devido à facilidade de purificação das isoformas a partir de tripsina bovina de fonte comercial, e as propriedades descritas acima, esta enzima tornar-se uma alternativa interessante para a indústria de alimentos e de detergente, bem como para pesquisa de biocatalisadores.
A novel bovine trypsin isoform was purified from commercial sample by ion exchange chromatography by Sephadex SP C50®. New isoform contain in addition of loss of N- terminus hexapeptide (as found in parent molecule β-trypsin) an intra-chain split between Lys-155 and Ser-156. The novel enzyme denominate gamma-trypsin showed similar properties with α-trypsin isoform in: polypeptide number chain (two chain), molecular masses (23,312 Da), secondary structure, hydrodynamic radius and others. In spite of enzymatic and structural similarities of both isoforms, -trypsin preferably has a lower rate formation from β-trypsin, a lower surface charge, but the gamma-trypsin has a higher thermal stability than α-trypsin. Due to obtaining facility of purification of bovine trypsin isoforms from commercial font, and properties described above, become this enzyme an interesting alternative for the food industry, detergent and biocatalysis research.

Este estudo analisa a viabilidade da produção de painéis de partículas utilizando resíduos de descaroçamento de algodão gerados em plantas de descaroçamento no nordeste argentino. Esses resíduos, que representam 40% da produção do algodão colhido no país, oferecem uma matéria-prima barata para a produção de painéis de aglomerado que atendem aos padrões internacionais. Foram avaliados painéis de camada simples e camada tripla, incorporando dois diferentes tamanhos de partícula do resíduo e teores de resina ureia-formaldeído entre 11,9 e 19,3%. Foram testadas a densidade, a resistência à flexão e a resistência à água. Verificou-se melhor comportamento nos painéis de camada tripa, o que por menores teores de resina permite obter maior resistência à flexão e melhor comportamento frente à água. Esses painéis poderiam oferecer uma alternativa de baixo custo para aplicação em móveis e construção civil, reduzindo o impacto ambiental da produção de algodão.

Abstract Objectives / Scope In a lifespan of a gas well multiphase flow regime keeps changing. When gas rate falls below critical rate, gas well loads up, significant reduction in gas rate occurs and eventually well ceases to flow. Tripura Asset is operating nearly 100 wells and most of them experience liquid loading in their production life, leading to loss of recovery. Many wells have experienced a sharp decline in production due to liquid loading. The paper highlights integrated production modelling approach adopted to identify liquid loading in wells and successful application of gas well deliquification techniques for optimising production and improving recoveries in Tripura. Methods, Procedures, Process All the 100 plus wells have been modelled in PIPESIM, an integrated production modelling software to identify propensity of liquid loading. Flowing tubing head pressure (FTHP) and casing head pressure (CHP) trends were captured in well monitoring sheets. Flowing gradient survey, variation in FTHP and CHP in union with the results of PIPESIM were used to identify liquid loading tendency in wells. A network model was created for determining the reserve gains on application of deliquification technique. Based on analysis wells were categorised as free from liquid loading, loaded in casing not in tubing and fully loaded. A multidisciplinary approach was adopted to identify and evaluate long and short-term deliquification measures to be adopted for deliquification and get maximum ultimate gas recovery from the wells – use of nitrogen, activation, smaller tubing, plunger, sucker rod pump (SRP) and progressive cavity pump (PCP). Results, Observations, Conclusions System analysis indicated about 25 wells, which required frequent activation, were completely liquid loaded. Only 5-6 wells were free from liquid loading. Implementation of short-term measures as activation by beaning up the well and injecting of nitrogen in identified wells has prevented well from ceasing. After an extensive study one well was identified and completed on pilot basis on 2 3/8″ tubing, with shoe run upto perforation. The technique has lifted the well out of liquid loading region. Positive displacement pump, with pump placed below the perforation, have been used for reviving 5 ceased liquid loaded wells and for increasing production from 1 liquid loaded well. Of the 6 wells – 5 have been completed on SRP and 1 on PCP, which has ensured revival of 41 MMSCM of gas. Plunger lift, a system that removes liquid slugs in gas wells using wells own energy has resulted in revival of 65 MMSCM of gas. The system has been implemented in 25 wells with substantial gains in 15 wells. Plunger lift optimisation being done on regular basis, based on changing liquid gas ratio, changes in THP and CHP has ensured optimal plunger performance. The pilot approach of putting in place continuous deliquification techniques has not only helped in enhancing production from the existing liquid loaded well but has also opened up new avenues to revive wells ceased due to liquid loading. During initial assessment of 100wells, ~70 wells are potential deliq candidates that can add potentially ~312 MMSCM of reserves. Novel/Additive Information The adopted approach has been useful in formulating a novel strategy to minimize the effect of liquid loading in wells of Tripura. Results of pilot implementation has helped Asset to understand and devise deliquification workflow to be adopted specific to the reservoir condition and well production parameters.
