In this article I argue that translated texts and translational paratexts invite readers to read translated texts as if they were the originals, a hitherto widely ignored premise of translations. Although translations are produced by many agents in collaboration (authors, publishers, copy-editors and translators), they are generally presented as texts produced predominantly by one agent, the author. I therefore claim that there is a ‘translation pact’ at work in translated literature, a rhetorical construction through which readers are invited to read translated texts as if they were the originals. A narratological implication of the pact is that individual readers who accept the pact will reconstruct only an ‘implied author’ and not an ‘implied translator’. This view differs from earlier works on the implied translator (e.g. Munday, 2008: 11; O’Sullivan, 2003; Schiavi, 1996). The translation pact is most often constructed implicitly, but sometimes translators draw attention to themselves and manifest their agency, for example by discussing translational decisions in prefaces and notes. Against what one would assume from previous claims on the translator’s ‘visibility’ (Venuti, 1995), I demonstrate that the translator’s presence does not necessarily work against the pact but can rather strengthen it. The translation pact explains why readers, including critics, literary scholars and other professional readers, often talk and write about translations as if they were originals composed solely by the author.

The anti-diabetic drug metformin is rapidly emerging as a potential anti-cancer agent. Metformin, effective in treating type 2 diabetes and the insulin resistance syndromes, improves insulin resistance by reducing hepatic gluconeogenesis and by enhancing glucose uptake by skeletal muscle. Epidemiological studies have consistently associated metformin use with decreased cancer incidence and cancer-related mortality. Furthermore, numerous preclinical and clinical studies have demonstrated anti-cancer effects of metformin, leading to an explosion of interest in evaluating this agent in human cancer. The effects of metformin on circulating insulin levels indicate a potential efficacy towards cancers associated with hyperinsulinaemia; however, metformin may also directly inhibit tumour growth. In this review, we describe the mechanism of action of metformin and summarise the epidemiological, clinical and preclinical evidence supporting a role for metformin in the treatment of cancer. In addition, the challenges associated with translating preclinical results into therapeutic benefit in the clinical setting will be discussed.

Abstract This paper takes the notion of translaboration as a stepping stone for an exploration of some of the recent debates about translational hermeneutics. In doing so, it aims to expand translaboration’s focus beyond concrete collaborations between multiple translators, or authors and translators, and to think about, and theorise, translaboration as a possible means of framing textual agents reading and writing each other within texts. The argument presented draws on both Hans-Georg Gadamer’s and Paul Ricoeur’s conceptions of the individual subject as interpretative agent, and of translation as an object of philosophical enquiry, and adopts the concept of a “hermeneutics of decipherment” (Maitland 2017, 38) as an alternative to dialogic models of understanding and translating. Similarly, the relationship between philosophical and translational hermeneutics is interrogated and recast as a translaborative endeavour rather than as an immediately reciprocal dialogue. Translaboration, this paper argues, thus also actively furthers the move away from what Blumczynski (2016, 29) calls “an arborescent epistemological paradigm” of interdisciplinarity and contributes to animating a transdisciplinarity that is fundamentally “rhizomatic” (ibid.; see Deleuze and Guattari 2004) in nature.

Monolingual Language behavior rests on three components: human agent, code and message. Translation processing requires three more constructs: translator, two codes, and a message in two texts. Equivalence theories attempted to supersede faithfulness and sameness of meaning in translation, but equivalence is a “convenience”, and is “always relative” (Baker 1992). Translational commensurability and semantic transportation thwart obtaining equivalence; therefore, the Interpretive Frame includes experience and identity among the elements necessary for any translation (Author, 2008). To explore poetic aesthetics, experience is related to personality observed in the Mental lexicon, while identity is related to phonic appeal observed in euphony. These relations are investigated in Arabic translations of English poems by Coleridge, Keats, Tennyson, Wordsworth, and Auden. Preliminary results show that: 1) the Mental Lexicon and euphony vary according to experience and identity, 2) contextualizing lexical appropriateness, euphony and metaphors contributes to poetic aesthetics.

The induction of cachectin/tumor necrosis factor (TNF) synthesis by bacterial endotoxins is a process that entails activation at several levels. Cachectin/TNF gene transcription is accelerated, leading to rapid accumulation of mRNA within the macrophage cytosol. In addition, translational derepression occurs, leading to far more efficient message utilization. Through the use of posttranscriptional reporter constructs, we now demonstrate that certain agents capable of inhibiting cachectin/TNF biosynthesis operate through different mechanisms. In RAW 264.7 macrophages, pentoxifylline blocks cachectin/TNF mRNA accumulation but has no effect upon the efficiency of reporter mRNA translation. Dexamethasone, on the other hand, has only a modest effect on cachectin/TNF mRNA accumulation, but strongly impedes translational derepression. Combined application of dexamethasone and pentoxifylline to macrophages causes a greater suppression of cachectin/TNF biosynthesis that can be achieved by either agent alone. These findings suggest that the signaling pathway activated by endotoxin is branched, and that selective inhibition of different parts of the pathway may be achieved through the use of distinct agents.

This work develops and implements a multi-agent time-based path-planning method using A*. The purpose of this work is to create methods in which multi-agent systems can coordinate actions and complete them at the same time. We utilized A* with constraints defined by a dynamic model of each agent. The model for each agent is updated during each time step and the resulting control is determined. This results in a translational path that each of the agents is physically capable of completing in synchrony. The resulting path is given to the agents as a sequence of waypoints. Periodic updates of the path are calculated, utilizing real-world position and velocity information, as the agents complete the task to account for external disturbances. Our methodology is tested in a dynamic simulation environment as well as on real-world lighter-than-air robotic agents.

ENGLISH ABSTRACT: In this dissertation two relatively under-researched areas within the scope of translation studies are investigated: the production process of translations and the sociological and cognitive aspects involved in this production process (as opposed to only studying the translation product). Process and sociological studies are currently on the forefront of research in translation studies and this dissertation attempts to contribute to these fields. The current study investigates the role of the various agents (among others the publisher, reviser and editor) involved in the production process of translations to illustrate how this process involves more agents than only the translator. Translation is proven to be a process involving a network of agents who work together to produce the final product, i.e. the target text. In this dissertation this interaction between agents is examined by making use of Latour's Actor-Network-Theory (ANT), a sociological theory that focuses on the network and links between agents within a production process. The production processes of literary translations that are analysed in this dissertation are Niggie – To hell with Cronjé (Ingrid Winterbach), Vaselinetjie – My name is Vaselinetjie (Anoeschka von Meck) and a selection of the "Stoffel tales" – In bushveld and dessert: A game ranger's life (Christiaan Bakkes). These texts were all translated by the literary translator Elsa Silke and published by NB Publishers in 2007, 2009 and 2008, respectively. The versions of the texts analysed are the source text, the concept translation, any other preliminary versions (manuscripts) and the completed published target text. These texts are analysed along with all available correspondence between agents to reconstruct the production processes in order to determine how translations are produced by means of interaction between different agents. This study is therefore greatly exploratory by nature and aimed at revealing the "invisible" of the production process of literary translations in order to contribute to and promote research within this under-researched field of translation studies. The main focus of this study is to analyse how each agent's inputs have a sociological impact on other agents and the production processes, as well as how their interaction and decisions impact the target text. The results are predominantly presented descriptively (within the framework of Descriptive Translation Studies [DTS]) and concepts of Nord's Functionalism, Even-Zohar's Polysystem Theory, Bourdieu's theory on the field of cultural production, Sharpe and Gunther's editing principles and Mossop's theory on editing and revision of translated texts are used to analyse and discuss the production processes of the three literary translations mentioned above. In conclusion a model is presented, based on these analyses, depicting a possible optimal production process for literary translations. A possible chronology of steps in the production process and means of interaction between agents are depicted by this model. With this proposed model this dissertation contributes to theory development within the field of translation studies and editing and revision theory. This model can be applied to production processes of literary translations to test if it is viable to apply in practice to ensure a more effective production process as well as a successful product (target text). If this model proves to be effective, it can eventually serve as a guideline for literary translation practice.

The literary translator in Iran is the key link in the chain of translation that contributes to the creation and development of new forms and themes in national literature. For the most part, it is the translator who begins the process of producing a translation by selecting the source text. However, this choice is not insignificant: it is affected by the whole trajectory of the translator's social life, his habitus according to Bourdieu and his horizon according to Berman; it is subject to the position occupied by the translator, and depends on the level of his symbolic, social, and economic capital which the previous expression of his standpoints, in this case his translations, have accumulated. Once the source text has been selected, the choice of the translation strategy follows the same pattern. It is the habitus and the position of the translator, which can orient him towards a particular technique or approach. Furthermore, the aim of the translator is important: does he/she want to earn money or recognition from his/her peers? Does he/she choose to subvert the established rules or to respect them in his/her translation? By analyzing the case of (re)translations into Persian of Albert Camus' Caligula, using a multidisciplinary approach at the crossroads of sociology and translation studies, the present study attempts to provide an overview of the field of literary translation in Iran and to identify the role of the agents who play an active role in it such as the translator, the publisher, the controlling authorities and agencies such as cultural ministries and academies, as well as the booksellers whose influence on the reception of (re)translations has often been ignored in translation studies of the Iranian context

In response to DNA damage, cells initiate a complex DNA-damage response (DDR) that includes mechanisms of DNA repair, cell cycle arrest, or in the cases of irreparable damage, apoptosis. Although regulation of the DDR has been extensively studied at the level of post-translational modifications and transcription, less is known about the role played by translational regulation. The data presented in this study describes the translational response and genome-wide translational profiles of human cells exposed to the DNA-damaging agents, ethyl methanesulphonate (EMS) and ionising radiation (IR). Exposure of HeLa cells to EMS caused a 60% decrease in global protein synthesis, mediated by phosphorylation of the a-subunit of eukaryotic initiation factor 2 (eIF2), whereas IR modulated global protein synthesis through changes in 4E-BP phosphorylation. Polysome profiling by cDNA micro array was used to identify on a genome-wide scale the differential translation of mRNAs following exposure to EMS or IR. Functional analysis of the data revealed translational regulation of distinct subsets of genes that, in the most part, were unique to EMS or IR exposure. An enrichment of genes involved in the ER stress response was identified following exposure to EMS, whereas genes involved in the mitochondrial oxidative phosphorylation pathway and ubiquitin-proteasome pathway were identified following exposure to IR. These data suggest that translation of distinct subsets of mRNAs may be necessary to initiate appropriate stress responses following exposure to different types of DNA-damaging agents, and in agreement with previous studies (powley et al., 2009) implicates translational control as an important mechanism of the DRR.

Translational efficiency is dictated in part by the availability of charged transfer RNA. Depletion of aminoacylated tRNAs (e.g. during recombinant protein expression) can increase translational errors and associated stress responses. Here, the role of tRNAs as regulators of gene expression was explored through development of synthetic, tRNA-regulated gene circuits, and through an investigation of the impact of tRNA aminoacylation on endogenous gene expression. Synthetic gene circuits initially explored the use of dominant negative alleles of the release factor eRF1 to modulate stop codon readthrough and translationally regulate gene expression. Mutant eRF1 proteins exhibited only a six-fold stimulatory effect on stop codon readthrough. The dominant negative phenotype was rescued partially by overexpression of eRF1, but not eRF3. Ultimately the severity of growth inhibition by these eRF1 alleles limited their utility in synthetic gene circuit design. A novel synthetic circuit was then implemented that utilised TetR interaction with a TetR-inducing peptide in order to control the expression of a suppressor tRNA, and thus a luciferase reporter gene. Using a parameterised mathematical model, the promoter configuration of the circuit was successfully optimised, allowing suppressor tRNAs to regulate the production of luciferase in both feedforward and positive feedback modes of operation. The effects of charged tRNA levels on the global translation network were dissected by regulating the S.cerevisiae glutamine tRNA synthetase gene GLN4 using a tet-off doxycyclineregulated promoter. tRNA synthetase depletion caused the activation of the Gcn4 amino acid starvation response due to accumulation of uncharged glutamine tRNAs. Doxycycline GLN4 shut-off caused increased amino acid production, and decreased ribosome biosynthesis at the transcriptomic and proteomic level, and further physiological changes proposed to result from compromised translation of glutamine-rich regulatory proteins. tRNA overexpression in the GLN4 depletion strain successfully caused altered competition between different isoacceptor tRNA types for their cognate synthetase resource. Together, these results support a growing understanding of tRNA as a key modulator of translation and gene expression in synthetic and natural systems.

Iberian Studies have developed in the last quarter of a century to the point of making one speak of a real Iberian Turn. Starting from the rejection of the classic scheme that places the two states (Portugal and Spain) as privileged agents of the representation of the Iberian space, the proposal of the Iberian Studies is to work on the system of historical exchanges and interferences that have shaped the cultural fabric of the peninsula, investigating both the points of connection as much as those of the fracture between its different realities (such as the Basque, Catalan and Galician ones, as well as the Castilian and Lusitanian ones). Accompanied by a “Reasoned Bibliography on Iberian Studies and Iberian Studies from Italy”, this volume examines the state of the art, with particular attention to the Italian context, in which these researches show a still unequal rooting and diffusion. A first section, dedicated to a general framework of the discipline and the exposition of theoretical issues and method problems, is followed by a second that presents critical contributions that address individual case studies. Born in part as a reaction to the so-called “crisis of Hispanism”, Iberian Studies offer themselves as an alternative to the traditional model of peninsular Hispanism, to its uninational and monolingual paradigm. They also place the emphasis on diversity and the relational aspect, looking with suspicion at every hegemonic design aimed at establishing a “centre” within a heterogeneous cultural landscape. Attentive to the phenomena of immigration and linguistic minorities, to the colonial past and relations with the Latin American world, but also to the themes of comparativism, translation, theory and the rethinking of criticism, Iberian Studies are a field in which not only debates about literature and the arts are included, but also about ideology.

Abstract Many multi-agent systems in nature are comprised of agents that interact with, and respond to, the dynamics of their environment. In this paper, we approach the study of such agent environment interactions through the study of passively compliant vehicles coupled to their environment via simple nonholonomic constraints. We first consider a single passively compliant Chaplygin beanie atop a platform having translational compliance, introduce the reduced equations for the system using the notion of nonholonomic momentum, and provide proof for its stability under arbitrary deformations of the elastic element modeling its compliance. We then direct our focus to results concerning the frequency response and control of passive Chaplygin beanies under actuation of the platform, discuss rich dynamical features arising from periodic actuation, and develop rules by which control can be exerted to collect and disperse multiple passive vehicles. We then discuss how the latter of these results clarifies the extent to which stable behavior can be excited in the system through exogenous control.

Experiments have been carried out in a 36-m long, 10-cm diameter multiphase horizontal flow system to examine the effect of drag reducing agents (DRA) on average pressure drop, maximum pressure drop and slug characteristics with the presence of water. Superficial liquid velocities between 0.5 and 1.5 m/s and superficial gas velocities between 2 and 14 m/s were investigated. Oil with a viscosity of 2.5 cP at 25 °C was used for the study. ASTM salt was used as a substitute for seawater and carbon dioxide was used as the gas. Water cut was 50%. Temperature and pressure were maintained at 25 °C and 0.13 MPa. The DRA concentrations of 0, 20 and 50 ppm were used in this study. The results show that the average pressure drop in both slug flow and annular flow decreased significantly with addition of DRA. Under special conditions, it was found that DRA changed the flow pattern from pseudo-slug to annular resulting in a 74% reduction in pressure drop. For annular flow, the average pressure drop reduction of up to 53% was achieved. The maximum pressure drop across the slug also decreased with the presence of DRA. The average and maximum pressure drops at a DRA concentration of 50 ppm were more effective than 20 ppm for all cases. The slug frequency and effective height of the liquid film decreased significantly when DRA concentrations were added. This led to a decrease in the average pressure drop. However, the slug translational velocity did not change significantly with addition of DRA.

Project Title: "Development of a plasmid-based reverse genetics system for the Bluetongue and Epizootic Hemorrhagic Disease viruses to allow comparative characterization of the function of the NS3 viroporin in viral egress". Project details: No - IS-4192-09; Participants – Ehrlich M. (Tel Aviv University), Parker J.S. (Cornell University), DermodyT.S. (Vanderbilt University); Period - 2009-2013. Orbiviruses are insect-borne infectious agents of ruminants that cause diseases with considerable economical impact in Israel and the United States. The recent outbreaks of BTV in Europe and of Epizootic Hemorrhagic Disease Virus (EHDV) in Israel, underscore the need for: (i) a better comprehension of the infection process of orbiviruses, (ii) the identification of unique vs. common traits among different orbiviruses, (iii) the development of novel diagnosis and treatment techniques and approaches; all aimed at the achievement of more effective control and treatment measures. It is the context of these broad goals that the present project was carried out. To fulfill our long-term goal of identifying specific viral determinants of virulence, growth, and transmission of the orbiviruses, we proposed to: (i) develop reverse genetics systems for BTV and EHDV2-Ibaraki; and (ii) identify the molecular determinants of the NS3 nonstructural protein related to viroporin/viral egress activities. The first objective was pursued with a two-pronged approach: (i) development of a plasmid-based reverse genetics system for BTV-17, and (ii) development of an "in-vitro" transcription-based reverse genetics system for EHDV2-Ibaraki. Both approaches encountered technical problems that hampered their achievement. However, dissection of the possible causes of the failure to achieve viral spread of EHDV2-Ibaraki, following the transfection of in-vitro transcribed genomic segments of the virus, revealed a novel characteristic of EHDV2-Ibaraki infection: an uncharacteristically low fold increase in titer upon infection of different cell models. To address the function and regulation of NS3 we employed the following approaches: (i) development (together with Anima Cell Metrology) of a novel technique (based on the transfection of fluorescently-labeledtRNAs) that allows for the detection of the levels of synthesis of individual viral proteins (i.e. NS3) in single cells; (ii) development of a siRNA-mediated knockdown approach for the reduction in levels of expression of NS3 in EHDV2-Ibaraki infected cells; (iii) biochemical and microscopy-based analysis of the localization, levels and post-translational modifications of NS3 in infected cells. In addition, we identified the altered regulation and spatial compartmentalization of protein synthesis in cells infected with EHDV2-Ibaraki or the mammalian reovirus. In EHDV2-Ibaraki-infected cells such altered regulation in protein synthesis occurs in the context of a cell stress reponse that includes the induction of apoptosis, autophagy and activation of the stressrelated kinase c-Jun N-terminal Kinase (JNK). Interestingly, inhibition of such stress-related cellular processes diminishes the production of infectious virions, suggesting that EHDV usurps these responses for the benefit of efficient infection. Taken together, while the present project fell short of the generation of novel reverse genetics systems for orbiviruses, the development of novel experimental approaches and techniques, and their employment in the analysis of EHDV-infected cells, yielded novel insights in the interactions of orbiviruses with mammalian cells.
