The decade-long revolution known as May ’68 is commonly framed as a political protest radiating out from European and North American universities. However, much is gained by instead viewing May ’68 within the context of both anticolonial struggle and the emergence of what Wallerstein terms “the world university system.” Understanding student protests within the context of anticolonial struggle, including within African universities, reveals the extent to which the neoliberal university we inhabit today is the product of a profound counterrevolution designed to undermine the promise of the university as a site of radical and anticolonial transformation.

Abstract This commentary on the preceding six articles identifies those elements that contributed to Baltimore '68: Riot and Rebirth's success as a public history program, even as it raises questions about the program's long-term impact. It pays particular attention to the way the oral history interviews conducted as part of the program created a more inclusive public conversation about the Baltimore riot. It also recognizes the importance of the University of Baltimore's commitment to what is often termed the scholarship of engagement by marshalling institution-wide resources for the program; and suggests commonalities between engaged scholarship and public history. Finally, this commentary suggests that while Baltimore '68 was enormously successful as a public humanities program, the depth and duration of its civic impact are less certain, and as a consequence, it raises issues simultaneously organizational, conceptual, and social.

AbstractStudy ObjectivesTo assess differences in cognitive impairment in a group of patients with schizophrenia receiving first-generation long-acting antipsychotics (FG-LAI) versus Aripiprazole long-actinginjectable (ALAI).MethodA descriptive, cross-sectional, multi-centerstudy.Study sample: 28 outpatients with stable schizophrenia (18 men and 10 women) with ages ranging from 22 to 64years.Inclusion criteria were: Clinically stable patients with a diagnosis of schizophrenia (according to DSM-5 criteria) and without any changes to their antipsychotic or antidepressant therapy in the last six months.Simple stratified sampling was performed to collect data from patients with schizophrenia receiving FG-LAI (n=14) versus patients with schizophrenia receiving Aripiprazole long-acting injectable (ALAI) (n=14)Groups were matched by age, gender, years of evolution of the disease, and years on formal education.Functionality in the different cognitive domains was evaluated based on the Brief Assessment of cognition in Schizophrenia (BACS.), a hetero-applied instrument, which Spanish version has been validated. BACS evaluates the following cognitive domains:Verbal memory (V.M): Word list testWorking memory (W.M): Digit sequencing taskMotor speed (M.S): Token motor taskVerbal fluency (V.F): Semantic or category fluencyAttention (A): Symbol codingExecutive function (E.F): Tower of LondonThe data obtained were analyzed using the SPSS 22.0 statistical package Differences between the means of quantitative variables were assessed using the independent-sample Student-t test. Individual test scores were converted into standardized (T and Z) scores and composite scores that were corrected for age and gender.Informed consent was obtained from all patients according to the Declaration of Helsinki (World Medical Association).ResultsIn the group receiving ALAI, the most severely impaired cognitive domain was attention, whereas motor speed was barely affected. In contrast, verbal memory was most impaired, whereas motor speed was the least impaired cognitive domain in the group receiving FG-LAI.Patients with schizophrenia taking ALAI showed a better cognitive function in all domains (except for motor speed and attention) than patients receiving FG-LAI.Summarized results in Table1.ConclusionsIn our study, patients with schizophrenia receiving Aripiprazole long-acting injectable have better cognitive function than patients receiving first-generation long-acting antipsychotics.

The article presents a methodology for examining a temporal sequence of synthetic aperture radar (SAR) images, as applied to the detection of the A-68 iceberg and its drifting trajectory. Using an improved image processing scheme, the analysis covers a period of eighteen months and makes use of a set of Sentinel-1 images. A-68 iceberg calved from the Larsen C ice shelf in July 2017 and is one of the largest icebergs observed by remote sensing on record. After the calving, there was only a modest decrease in the area (about 1%) in the first six months. It has been drifting along the east coast of the Antarctic Peninsula, and is expected to continue its path for more than a decade. It is important to track the huge A-68 iceberg to retrieve information on the physics of iceberg dynamics and for maritime security reasons. Two relevant problems are addressed by the image processing scheme presented here: (a) How to achieve quasi-automatic analysis using a fuzzy logic approach to image contrast enhancement, and (b) The use of ferromagnetic concepts to define a stochastic segmentation. The Ising equation is used to model the energy function of the process, and the segmentation is the result of a stochastic minimization.

Abstract Where a run of pollbooks exists it is possible, by transcribing the material into a computer data‐base, to trace the voting history of individual voters over the entire period of their psephological history. Sandwich was eventually disenfranchised in 1881 for gross corruption, which had a long history in the borough, and the analysis indicates—though it can never prove—which groups of voters were most likely to have been influenced by cash considerations. A further consideration of those voters who split their votes suggests which party might have been the principal beneficiary of this relatively unusual voting choice.

Introduction/Objective. The use of bladder augmentation and/or continent urinary diversion has gained wide acceptance, particularly in children with small, abnormally developed bladder or high-pressure bladder that poses great risk for renal deterioration and incontinence. We discuss indications, results, and complications with various types of continent vesicostomy (CV) in children. Methods. Sixty-eight patients with CV are retrospectively reviewed (51 boys and 17 girls) 1987?2008. The median follow-up time was 17.8 years (3?22 years). CV included appendicovesicostomy in 31 (41.3%), vesicostomy with distal ureter in 27 (36.0%), and preputial CV in 10 (13.3%) patients. CV in patients with augmented bladder was in 18 (26.47%) children. The indications for performing CV were various types of neurogenic and myogenic dysfunctions of urine bladder with incontinence due to the following pathoanatomical substrates: anomalies of the brain?spine segment development (27), bladder exstrophy (10), posterior urethral valve (15), expansive processes (4), and other anatomical defects in 12 patients. Results. Continence was achieved in 94.64% of the cases, without statistically significant difference between particular types of the stoma (p = 0.065). Early complications included stoma necrosis, stoma bleeding, peristomal infection in 5/68 (7.35%) patients, and late complications included calculosis, in 20/68 (29.4%), stomal stenosis, in 8/68 (11.5%), and difficulties of catheterization, in 3/68 patients (4.08%). Calculosis was predominant in appendicovesicostomy (p = 0.012). Conclusion. CV is a safe procedure applied with the main purposes of achieving continence, preservation of renal function, and improvement of the quality of life, along with an acceptably low rate of complications.

The γ-herpesviruses are oncogenic B cell lymphotrophic viruses that establish life-long latency in the host. Murine γ-herpesvirus 68 (MHV-68) infection of mice represents a unique system for analyzing γ-herpesvirus latency in splenic B cells at different stages of infection. After intranasal infection with MHV-68 we analyzed the establishment of latency 14 days after infection, and the maintenance of latency 3 months after infection in different purified subpopulations of B cells in the spleen. The data show that MHV-68 latency is mainly established in germinal center B cells and that long-term latency is preferentially maintained in two different subsets of isotype-switched B cells, germinal center and memory B cells. Cell cycle analysis indicates that MHV-68 is located in both cycling and resting isotype-switched B cells. Analysis of viral gene expression showed that both lytic and latent viral transcripts were differentially expressed in germinal center and memory B cells during long-term latency. Together, these observations suggested that γ-herpesviruses exploit the B cell life cycle in the spleen.

Abstract. It is generally believed that the low-frequency variability of climatic parameters seems to be connected to solar cycles. The principal periodicities are: 11-year (Schwabe), 22-year (Hale), 33-year (Bruckner) and 80–100-year (Gleissberg) cycles. The main heliophysical factors acting on climate, the biosphere and the atmosphere are solar irradiance, the intensity of solar and galactic cosmic rays (relativistic charged particles with energies >500 MeV) changing the cloud cover of the atmosphere, and UV-B-radiation. The 11-year and 80–90-year solar cycles are apparent in solar radiation and galactic cosmic ray trends. At the same time the bidecadal Hale cycle, related to a reversal of the main solar magnetic field direction is practically absent in either solar radiation or galactic cosmic ray variations. Besides, nobody can identify any physical mechanisms by which a reversal in the solar magnetic field direction could influence climate. However, the 22-year cycle has been identified in rather many regional climatic (droughts, rainfall, tree growth near 68° N, 30° E) and temperature records all over the world. We discuss here three possible cause of the bidecadal periodicity in climatic records, one of which is associated with a variation of stardust flux inside the Solar System. The most recent observations by the DUST experiment on board the Ulysses spacecraft have shown that the solar magnetic field lost its protective power during the last change of its polarity (the most recent solar maximum), so that the stardust level inside of the Solar System has been enhanced by a factor of three. It is possible that the periodic increases of stardust in the Solar System may influence the amount of extraterrestrial material that falls to the Earth and consequently act on the Earth's atmosphere and climate through alteration of atmospheric transparency and albedo. This material (interstellar dust and/or cometary matter) may also provide nucleation sites and thereby influence precipitation.

ABSTRACT CD4 T cells are dispensable for acute control of murine gammaherpesvirus 68 (MHV-68) but are necessary for effective long-term control of the virus by CD8 T cells. In contrast, protein kinase C θ (PKCθ) is not essential for either acute or long-term viral control. However, we found that while either CD4 or CD8 T cells could mediate the clearance of MHV-68 from the lungs of PKCθ+/+ mice, PKCθ−/− mice depleted of either subset failed to clear the virus. These data suggest that there are two alternative pathways for MHV-68 clearance, one dependent on CD4 T cells and the other on PKCθ. Protection mediated by the latter appears to be short-lived. These observations may help to explain the differential requirement for PKCθ in various models of CD8 T-cell activation and differences in the costimulatory requirements for acute and long-term viral control.

ABSTRACT The murine gammaherpesvirus 68 (MHV-68) replicates in respiratory epithelial cells, where it establishes a persistent, latent infection limited predominantly to B lymphocytes. The virus-specific CD4+ T-cell response in C57BL/6 mice challenged intranasally with MHV-68 is detected first in the mediastinal lymph nodes and then in the cervical lymph nodes and the spleen. The numbers of MHV-68-specific CD4+ T cells generated in congenic mice homozygous for disruption of the β2-microglobulin gene tended to be higher, indicating that the absence of the CD8+ set in this group resulted in a compensatory response. The peak frequency within the splenic CD4+ T-cell population may reach 1:50 in the acute response; it then drops to 1:400 to 1:500 within 4 months and stays at that level in the very long term. Sorting for L-selectin (CD62L) expression established that all virus-specific CD4+ T cells were initially CD62Llow, with >80% maintaining that phenotype for the next 14 months. The overall conclusion is that MHV-68-specific CD4+ T cells remain activated (CD62Llow) and at a stable frequency in the face of persistent infection.

A total of 136 lower limbs from 68 human cadavers were dissected to determine the incidence of the long accessory flexor muscle and its origins and insertions. The muscle was present in 11 of 136 lower extremities (8%) and 9 of 68 cadavers (13%). Five long accessory flexor muscles originated from the tibia and fascia of the deep posterior compartment and six began on the fibula. All of the tendons were deep to the laciniate ligament and coursed within the tarsal tunnel. All five of the tendons originating from the tibia inserted on the quadratus plantae, with two of the tendons having an additional insertion on the flexor digitorum longus. Three of the tendons with fibular origin inserted on the quadratus plantae and three inserted on the flexor digitorum longus. The importance of the long accessory flexor muscle relates as a cause or association with tarsal tunnel.

ABSTRACT CD4 T cells are not essential for primary clearance of replicating murine gammaherpesvirus 68 (MHV-68) but are required for effective long-term control. The virus reactivates in the lungs of major histocompatibility complex class II-deficient (CII−/−) mice that lack functional CD4 T cells. CD40 ligand (CD40L) is upregulated on activated CD4 T cells, and it is thought that CD40-CD40L interactions are an important component of CD4 T-cell help. Our previous studies have shown that agonistic antibodies to CD40 can substitute for CD4 T-cell function in the long-term control of MHV-68. In the present study, we sought to identify the CD40-positive cell type mediating this effect. To address this question, we adoptively transferred MHV-68 peptide-pulsed CII−/− dendritic cells (DC) that had been treated with an agonistic antibody to CD40 into MHV-68-infected CII−/− recipients. Viral reactivation was significantly lower in mice injected with anti-CD40-treated DC than in those injected with control DC or in mice that did not receive any DC. However, in similar experiments with B cells, anti-CD40 treatment had no effect. We also investigated the requirement for CD40 expression on T cells by adoptive transfer of T cells from CD40+/+ or CD40−/− mice into T-cell-deficient recipients that were subsequently infected with MHV-68. The results showed that CD40 expression on T cells is not necessary for preventing viral reactivation. Taken together, our data suggest that CD40 engagement on DC, but not on T or B cells, is essential for effective long-term control of MHV-68.

In a vulnerable population of critically ill patients after cardiac surgery, however, it is unclear whether haloperidol administration affects the QTc interval. Thus, the aim of this study is to explore the effect of haloperidol in low doses on this interval.MethodThis retrospective cohort study was performed on a cardio-surgical intensive care unit (ICU), screened 2,216 patients and eventually included 68 patients with delirium managed with oral and intravenous haloperidol. In this retrospective analysis, electrocardiograms were taken prior and within 24 h after haloperidol administration. The effect of haloperidol on QTc was determined with a Person correlation, and inter-group differences were measured with new long QT comparisons.ResultsIn total, 68 patients were included, the median age was 71 (64–79) years and predominantly male (77%). Haloperidol administration followed ICU admission by three days and the cumulative dose was 4 (2–9) mg. As a result, haloperidol administration did not affect the QTc (r = 0.144, p = 0.23). In total, 31% (21/68 patients) had a long QT before and 27.9% (19/68 patients) after haloperidol administration. Only 12% (8/68 patients) developed a newly onset long QT. These patients were not different in the route of administration, cumulative haloperidol doses, comorbidities, laboratory findings, or medications.Significance of resultsThese results indicated that low-dose intravenous haloperidol was safe and not clinically relevant for the development of a newly onset long QT syndrome or adverse outcomes and support recent findings inside and outside the ICU setting.

ABSTRACT The chemokine IP-10 (CXCL10) and its cellular receptor CXCR3 are upregulated in the lung during murine gammaherpesvirus 68 (MHV-68) infection. In order to determine the role of the CXCR3 chemokine receptor in the immune response to MHV-68, CXCR3−/− mice were infected with the virus. CXCR3−/− mice showed delayed clearance of replicating MHV-68 from the lungs. This correlated with delayed T-cell recruitment to the lungs and reduced cytolytic activity prior to viral clearance. Splenomegaly and the numbers of latently infected cells per spleen were transiently increased. Ηowever, CXCR3−/− mice showed normal virus-specific antibody titers and effective long-term control of MHV-68 infection.

Objective: Hydrocephalus is a major cause of morbidity in the pediatric population, with potentially severe consequences if left untreated hydrocephalus. Endoscopic third ventriculostomy (ETV) has become the method of choice in the treatment of obstructive hydrocephalus. However, age, ventricular anatomy, and etiology could determine success rates (SR) of ETV. The aim of this study was to identify putative factors that could influence the outcome of ETV in children. Methods: Clinical data of 148 consecutive patients under 18 years of age who underwent 150 consecutive ETVs from January 2000 to January 2019 were reviewed. Data were analyzed with regards to clinical and radiological SR. The actual long-term SR was compared to that predicted by the ETV Success Score (ETVSS) model. Results/Discussion: The study group included 63 males (43%) and 85 females (57%) ranging in age from 11 days to 18 years (mean age of, 6.17 ± 1.02 years; median age 4.5 years) at surgery. Concerning the etiology of hydrocephalus, tumors and aqueductal stenosis (AS) were the most frequently observed, with each occurring in 43 cases (29%), followed by malformations in 31 (21%), cystic lesions in 7 (5%) and other etiologies in 67 patients (16%). The overall SR at the first ETV attempt was 80% (118/148), compared to 74.8% (variance 14.35, 95% CI 69.37–78.22) predicted by the ETVSS. Regarding age, SR was 58% in patients under 6 months of age, 65% in children between 6 months and 1 year, and 86% in children older than 1 year. SR for AS and hydrocephalus associated with posterior fossa tumors were 88 and 90%, respectively. Unsatisfactory results were related to previous intraventricular hemorrhage and infection. The overall complication rate in this series was 13%. The lower SR after ETV in young children may be explained by the poor CSF reabsorption capacity of newborn infants due to immaturity of the arachnoid granulations; Univariate and multivariate analysis showed in our series that both the etiology of hydrocephalus and patient´s age group were relevant factors predicting success. A literature review showed that SR in children under 6 months of age ranged from 32% to 45%, and in children older than one year it ranged from 56% to 71%. A few studies have analyzed the postoperative course and SR after a period of 3 years. Conclusion: ETV is safe and effective in children. In this series, the age of the patient and etiology of hydrocephalus were related to SR. Also, the ETVSS was accurate to predict outcome. Intraventricular hemorrhage, previous CNS infection, myelomeningocele, prematurity and recurrent shunt revisions have been associated with high failure rates of ETV in all age groups, particularly in infants under 6 months of age. In a long-term follow-up, surgical experience was statistically significant in reducing complications.

At the same time, it tries to show the importance of diversifying the analyses on the long ’68 – focusing on different countries, regions and cities – in order to grasp its transnational nature.

OBJECTIVES/GOALS: Chronic or new symptoms after infection with severe-acute-respiratory-coronavirus-2 (SARS-CoV-2) has been termed post-acute sequelae of Covid-19 (PASC) or Long Covid. Our objective is to present results from COVID-OUT, a phase 3 double-blind, randomized controlled trial of early outpatient treatment of Covid-19 with repurposed medications. METHODS/STUDY POPULATION: COVID-OUT enrolled adults age 30 to 85 with overweight or obesity who had proof of SARS-CoV-2 infection and fewer than 7 days of symptoms. In this 2 by 3 factorial design trial of metformin, ivermectin, fluvoxamine, or exact-matching placebo of each medication, participants were randomized 1:1:1:1:1:1 to the 6 treatment allocations. This abstract focuses on whether early treatment with metformin prevented Long Covid. Immediate release metformin was titrated to 1500mg daily over the first 6 days. We assessed the incidence of clinician-diagnosed Long Covid with follow up through 10 months after enrollment. We also assessed where participants were diagnosed with Long Covid, and where they received Long Covid treatment. RESULTS/ANTICIPATED RESULTS: Of 1124 participants, 98 (8.7%) report having a healthcare provider make a diagnosis of long covid. By arm, 6.9% (39/564) of metformin participants report having a diagnosis for long covid as compared with 10.5% (59/560) of matched placebo controls. The absolute reduction attributable to metformin was 3.6% (95%CI, 0.3% to 7.0%; P=0.031) with a relative risk reduction of 34% (95%CI, 3% to 55%). The metformin cost per long covid case averted was $28 (95%CI, $15 to $306). 10-month follow-up data will be available at the time of presentation as well as an analysis of baseline factors associated with the development of Long-Covid, independent of treatment allocation in the trial. DISCUSSION/SIGNIFICANCE: Metformin reduced the incidence of clinician-diagnosed long covid by 34% in a double-blind randomized placebo-controlled trial, and previous research published in-vitro activity by metformin against SARS-CoV-2 and other RNA viruses. Further investigation of metformin as early treatment for SARS-CoV-2 is warranted.

ObjectiveTo report multicentred use of the heavy silicone oil Densiron 68 for anatomical reattachment following rhegmatogenous retinal detachment (RRD) repair and its associated complications.Methods and analysisPatients from seven vitreoretinal units within the UK that underwent RRD repair with Densiron 68 between January 2015 and December 2019 were identified. Primary outcome measures were primary and final reattachment rate, retained Densiron and failure rate. Secondary outcome measures were duration of tamponade, final visual acuity (VA) and complications of heavy silicone oil.Results134 eyes of 134 patients were involved in the study. Primary surgical success was achieved in 48.5%, while a final reattachment rate of 73.4% was observed. The mean duration of Densiron 68 tamponade was 139.5 days. Mean final VA was 1.01 (range 0–2.9). 8 eyes (6.0%) required long-term topical steroids for anterior uveitis, whereas none of the eyes required long-term pressure-lowering treatment. Emulsification rate was 10.7% (6 eyes).ConclusionThis is the largest real-world study on Densiron 68 in the UK. Densiron 68 facilitates tamponade of inferior retinal pathology and may be considered as an option for tamponade of inferior retinal pathologies.

Abstract Background Standard performance parameters measured during a laboratory test have been shown to be related to mountain ultra-marathon performance up to a competition length of 75 km. It is not known if a similar relationship exists for longer races. Objective This study aimed to investigate the association between laboratory-based performance parameters and performance times in a short (68 km) and a long (121 km) mountain ultra-marathon. Materials and Methods Eleven male finishers of the short race and seven male finishers of the long race were investigated. Participants performed an incremental exercise test to exhaustion in the 2 weeks prior to the event. During the event, the heart rate was monitored and finishing times were registered. Results The maximal oxygen consumption and the oxygen uptake at the ventilatory thresholds 1 and 2 were related to performance time during the short run (~12h; r = –0.764 up to r = –0.782; p < 0.05), but there was no correlation during the long race (~28h; r = –0.107 to 0.357; p > 0.05). Conclusions This study shows that physical fitness parameters established in a laboratory setting determine competition completion times in ultra-mountain marathon events lasting for ~12 h. During longer races, i. e. ~28 h, other factors not established in the present investigation, such as experience, race strategy, coping with pain and fatigue resistance, may be important for performance.

We performed experiments to determine the photoperiodic response of Ceratostigma plumbaginoides Bunge., or leadwort, which is a low-growing hardy herbaceous perennial native to China with deep gentian-blue flowers. Tip cuttings were rooted in 72-cell trays and grown under a 24-hour photoperiod for 2 weeks and then transplanted into 11.4-cm pots and grown for one more week. Plants were then placed under different primary photoperiods (10, 16, or 24 hours) for 4, 6, or 8 weeks, then transferred to secondary photoperiods (10, 14, 16, or 24 hours) at a constant 20 °C. Pots were also placed under continuous 10, 14, 16, or 24 hours. Nearly all plants flowered under all treatments except under continuous 10- or 24-hour photoperiods, in which no plants flowered. Plants grown under 14 hours flowered earliest (50 days), followed by plants under the 16-hour primary treatment. The 10-hour primary treatment delayed flowering for as long as its duration, whereas the 16-hour primary photoperiod initiated rapid flowering, regardless of duration and subsequent secondary photoperiod. Flowering was also delayed when the primary photoperiod was 24 hours. Collectively, these responses indicate that Ceratostigma is an intermediate-day plant.

Abstract Objective This pilot study examines the effects of yoga therapy on anxiety, depression symptoms, inattention, and seizure frequency in children diagnosed with Epilepsy. Data Selection The sample consisted of 5 participants (4 male, 1 female), ages 7-12 years old, with diagnoses of generalized epilepsy, absence epilepsy, and complex partial epilepsy. Participants all had average cognitive abilities (IQ SS = 100.7; VCI SS = 106.3; PRI SS = 108.3), a diagnosis of Attention Deficit Hyperactivity Disorder (ADHD), and clinically significant levels of anxiety and depression. Participants participated in 30-45-minute-long yoga sessions over a 6-week period with 2-3 home sessions each week. Differences in hyperactivity, anxiety, depression, and inattention were measured via parent rating Likert scales (0-poor, 10-best) at 1 month, 3 months, and 6 months. Participants also completed weekly self-report measures to assess anxiety and depression (Revised Children’s Manifest Anxiety, Second Edition (RCMAS-2) and Children’s Depression Inventory, Second Edition (CDI-2)). Data Synthesis No significant reliable change differences were found between sessions; however, a downward trend was visible in reduction of anxiety (RCMAS-2; first session M = 16.4, last session M = 9.4) and depression symptoms (CDI-2; first session M = 52.8, last session M = 49.2). Further, parents reported a decrease in anxiety (first session M = 4.8, last session M = 6.4). No seizures were reported during the study. Conclusions There is no previous research using pediatric populations and yoga therapy effects on reducing seizure frequency, anxiety, depression, and attention problems. This pilot study showed promising results in reducing anxiety, and depression symptoms. Limitations of study include small sample size and attrition, which are both important to consider when conducting another study.

By elaborating on these extensions, the article considers 1968 as a global moment in which diverse histories converge and then diverge again, while the synchronicity of events generates an enhanced consciousness of the world, making the outlines of larger structures and connections more visible. Seeing 1968 in this way, the article argues, can provide a more adequate grasp on eastern Europe's ’68 and its variegations.

ABSTRACT The role of CD28-dependent costimulatory interactions in the development and maintenance of antiviral immune responses was investigated in a mouse model of gammaherpesvirus infection. CD28−/− mice could clear a productive infection with murine gammaherpesvirus 68 (MHV-68), although early lung viral titers were significantly increased. Both CD28−/− and CD28+/+ mice maintained effective long-term control of MHV-68. Gamma interferon responses appeared to develop more slowly in CD28−/− mice, while cytotoxic T-cell activity was similar to that in wild-type mice. Splenomegaly developed normally in CD28−/− mice, whereas virus-specific antibody responses were significantly reduced and aberrant class switching was observed. This work demonstrates that costimulatory interactions involving CD28 are not an absolute requirement for the control of infection with MHV-68.

Previous studies have established that strain 68-1–derived rhesus cytomegalovirus (RhCMV) vectors expressing simian immunodeficiency virus (SIV) proteins (RhCMV/SIV) are able to elicit and maintain cellular immune responses that provide protection against mucosal challenge of highly pathogenic SIV in rhesus monkeys (RMs). However, these efficacious RhCMV/SIV vectors were replication and spread competent and therefore have the potential to cause disease in immunocompromised subjects. To develop a safer CMV-based vaccine for clinical use, we attenuated 68-1 RhCMV/SIV vectors by deletion of the Rh110 gene encoding the pp71 tegument protein (ΔRh110), allowing for suppression of lytic gene expression. ΔRh110 RhCMV/SIV vectors are highly spread deficient in vivo (~1000-fold compared to the parent vector) yet are still able to superinfect RhCMV+ RMs and generate high-frequency effector-memory–biased T cell responses. Here, we demonstrate that ΔRh110 68-1 RhCMV/SIV–expressing homologous or heterologous SIV antigens are highly efficacious against intravaginal (IVag) SIVmac239 challenge, providing control and progressive clearance of SIV infection in 59% of vaccinated RMs. Moreover, among 12 ΔRh110 RhCMV/SIV–vaccinated RMs that controlled and progressively cleared an initial SIV challenge, 9 were able to stringently control a second SIV challenge ~3 years after last vaccination, demonstrating the durability of this vaccine. Thus, ΔRh110 RhCMV/SIV vectors have a safety and efficacy profile that warrants adaptation and clinical evaluation of corresponding HCMV vectors as a prophylactic HIV/AIDS vaccine.

Caloric restriction (CR) slows the aging process, extends lifespan, and exerts neuroprotective effects. It is widely accepted that CR attenuates β-amyloid (Aβ) neuropathology in models of Alzheimer’s disease (AD) by so-far unknown mechanisms. One promising process induced by CR is autophagy, which is known to degrade aggregated proteins such as amyloids. In addition, autophagy positively regulates glucose uptake and may improve cerebral hypometabolism—a hallmark of AD—and, consequently, neural activity. To evaluate this hypothesis, APPswe/PS1delta9 (tg) mice and their littermates (wild-type, wt) underwent CR for either 16 or 68 weeks. Whereas short-term CR for 16 weeks revealed no noteworthy changes of AD phenotype in tg mice, long-term CR for 68 weeks showed beneficial effects. Thus, cerebral glucose metabolism and neuronal integrity were markedly increased upon 68 weeks CR in tg mice, indicated by an elevated hippocampal fluorodeoxyglucose [18F] ([18F]FDG) uptake and increased N-acetylaspartate-to-creatine ratio using positron emission tomography/computer tomography (PET/CT) imaging and magnet resonance spectroscopy (MRS). Improved neuronal activity and integrity resulted in a better cognitive performance within the Morris Water Maze. Moreover, CR for 68 weeks caused a significant increase of LC3BII and p62 protein expression, showing enhanced autophagy. Additionally, a significant decrease of Aβ plaques in tg mice in the hippocampus was observed, accompanied by reduced microgliosis as indicated by significantly decreased numbers of iba1-positive cells. In summary, long-term CR revealed an overall neuroprotective effect in tg mice. Further, this study shows, for the first time, that CR-induced autophagy in tg mice accompanies the observed attenuation of Aβ pathology.

68 Background: High-dose conformal proton radiation for localized prostate cancer results in favorable clinical outcomes and low toxicity rates. Here, we report long-term quality of life (QOL) outcome for men treated with conformal protons. Methods: Serial QOL questionnaires were administered to men who received proton radiation. Long-term questionnaires (minimum 2 years) were completed by 72 patients. Men were stratified into functional groups from their baseline questionnaires (normal, intermediate or poor function) for each symptom domain. Symptom scores were calculated at baseline and long-term follow-up and expressed as mean values. QOL changes were assessed overall and within functional groups in a paired fashion using the Student's t-test. Results: The median age at treatment and follow-up were 66 years and 44 months, respectively. The median dose was 82 GyE (range 74-82 GyE). For all 72 patients, there were increased scores for incontinence (ID) (3.2 baseline vs. 9.9 long-term, p=<0.001), obstructive/irritative voiding (OID; 20 vs. 24, p=0.028), bowel (BD; 4.4 vs. 8.0, p=0.001) and sexual dysfunction (SD; 25 vs. 48, p<0.001). When stratified by functional category, more specific estimates were possible. For ID, only normal function was associated with a significant increased score (0 vs. 8.4 at baseline and long-term, p=<0.001); for OID, only the group with normal baseline function showed a significant increased score (12 vs. 17, p=0.01); similarly for BD, only men with normal function had a significant increased score (0 vs. 5.4, p<0.001). For SD, the score increased in men with normal (1.4 vs. 30, p<0.001) and intermediate function (17 vs. 46, p<0.001). Conclusions: Patient reported outcomes are sensitive indicators of treatment related sequelae and here quantitate, for the first time, the long-term consequences of proton monotherapy for prostate cancer. Analysis by baseline functional category is a useful means of predicting long-term QOL scores for an individual patient. High-dose proton radiation was associated with small increases in bowel dysfunction, obstructive/irritative voiding dysfunction and incontinence. With long term follow-up, sexual dysfunction increased more than any other symptom domain. No significant financial relationships to disclose.

Long term coastal erosion in beaches induced by long-shore sediment transport is an important aspect to be taken account in beach behavior. If anthropogenic impacts in coastal stretch (harbor, detached breakwaters, lack of sediments from streams and gullies, and urban pressure) interrupting long-shore currents and transport and decreasing sediment supply, the coastal equilibrium disappears. In this paper a case study is shown in order to analyze long term beach behavior in a coastal stretch affected by erosion. After a brief description of morphological aspects, causes and effects that have incidence on the case study area will be shown. General background and coastal works are detailed. The ideas based on a classical analysis of beach behavior are exposed and results obtained from bathymetric analysis and numerical models are showed. Morphology, wave climate, morphodynamics and sediment budget are evaluated and morphodynamic beach behavior is proposed. A diagnosis is made and mitigation proposal for coastal erosion will be proposed.

An elevated heart rate at admission or discharge is known to be associated with poor cardiovascular outcomes in patients with acute myocardial infarction (AMI). The association between post-discharge average office-visit heart rate and cardiovascular outcomes in patients with AMI has rarely been studied. We analyzed data for 7840 patients from the COREA-AMI registry who had their heart rates measured at least three times after hospital discharge. The office-visit heart rates were averaged and categorized into four groups by quartiles (<68, 68–74, 74–80, and >80 beats per minute). The primary end point was a composite of cardiovascular death, myocardial infarction, and ischemic stroke. During a median of 5.7 years of follow-up, major adverse cardiovascular events (MACE) affected 1357 (17.3%) patients. An average heart rate higher than 80 bpm was associated with an increased incidence of MACE compared to the reference average heart rate of 68–74 bpm. When dichotomized into <74 or ≥74 bpm, a lower average heart rate was not associated with MACE in patients with LV systolic dysfunction, in contrast to those without LV systolic dysfunction. An elevated average heart rate at office visits after AMI was associated with an increased risk of cardiovascular outcomes. Heart rate monitoring at office visits after discharge provides an important predictor related to cardiovascular events.

AbstractUncontrolled scaling for furs and oil began on South Georgia about 1786 and continued sporadically until the early 20th century. From 1909 onward legislation covered licensed killing of male elephant seals Mirounga leonina. This provided for a successful oiling industry that lasted almost 60 years, operated by whalers primarily in early spring before the whaling season began. Biological research ensured that quotas in the last decade remained within the carrying capacity of the stocks. High-grade seal oil enhanced revenues from whaling, providing overall 16% of the volume and 19% of the value of total annual catches, with much higher proportions in some seasons. The seal oil industry did not long survive the demise of whaling on South Georgia, finally ending in 1968.

The aim of this study was to evaluate nitrogen (N) efficiency from different organic and mineral fertilizers applied to the spring barley. Dry matter yield, N content and N uptake of spring barley from 16 years of experiments at two sites in the Czech Republic with different soil and climatic conditions were analyzed. For assessing of nitrogen efficiency nitrogen utilization efficiency (NUtE, kg/kg), recovery efficiency of applied N (%), agronomic efficiency of applied N (kg/kg) and summary N balances (&Sigma;&Delta;N, kg/ha) were observed. Six fertilization treatments were utilized: no fertilization (control); sewage sludge; farmyard manure (FYM); N in mineral fertilizers (N); NPK in mineral fertilizers (NPK) and N in mineral fertilizers + straw (N + ST). Yields were about 68% higher at NPK (S1 site) and 55% at N + ST (S2 site) in comparison with control. The highest NUtE was recorded at both locations after application of FYM. Higher NUtE from mineral fertilizers was obtained at low productive S1 site. At both sites a trend of decreasing &Sigma;&Delta;N over time was observed. At both sites a trend of decreasing negative N balance was observed. At lower productive site the decline of N balance was minimized for mineral fertilizers treatments in last experimental years. At higher productive site the differences between treatments with mineral nitrogen and control were lower and the decline of N balance continued over all 16 years of experiment duration.
