Dinosaurs have been major components of ecosystems for over 200 million years. Although different macroevolutionary scenarios exist to explain the Triassic origin and subsequent rise to dominance of dinosaurs and their closest relatives (dinosauromorphs), all lack critical support from a precise biostratigraphically independent temporal framework. The absence of robust geochronologic age control for comparing alternative scenarios makes it impossible to determine if observed faunal differences vary across time, space, or a combination of both. To better constrain the origin of dinosaurs, we produced radioisotopic ages for the Argentinian Chañares Formation, which preserves a quintessential assemblage of dinosaurian precursors (early dinosauromorphs) just before the first dinosaurs. Our new high-precision chemical abrasion thermal ionization mass spectrometry (CA-TIMS) U–Pb zircon ages reveal that the assemblage is early Carnian (early Late Triassic), 5- to 10-Ma younger than previously thought. Combined with other geochronologic data from the same basin, we constrain the rate of dinosaur origins, demonstrating their relatively rapid origin in a less than 5-Ma interval, thus halving the temporal gap between assemblages containing only dinosaur precursors and those with early dinosaurs. After their origin, dinosaurs only gradually dominated mid- to high-latitude terrestrial ecosystems millions of years later, closer to the Triassic–Jurassic boundary.

In the mammalian genome, early- and late-replicating domains are often separated by temporal transition regions (TTRs) with novel properties and unknown functions. We identified a TTR in the mouse immunoglobulin heavy chain (Igh) locus, which contains replication origins that are silent in embryonic stem cells but activated during B cell development. To investigate which factors contribute to origin activation during B cell development, we systematically modified the genetic and epigenetic status of the endogenous Igh TTR and used a single-molecule approach to analyze DNA replication. Introduction of a transcription unit into the Igh TTR, activation of gene transcription, and enhancement of local histone modifications characteristic of active chromatin did not lead to origin activation. Moreover, very few replication initiation events were observed when two ectopic replication origin sequences were inserted into the TTR. These findings indicate that the Igh TTR represents a repressive compartment that inhibits replication initiation, thus maintaining the boundaries between early and late replication domains.

AbstractThe argument of Rousseau's Essay on the Origin of Languages is intimately connected with that of his Discourse on the Origin and the Foundations of Inequality among Men. The origin of political society is inseparable from the origin of language, which, in turn, is inseparable from the origin of reason. That so much of the Essay is concerned with music leads us to wonder what music has to do with reason, politics, and language. These two books share what is a regular feature of Rousseau's manner of writing—presenting what seem to be logical foundations as temporal origins. In emphasizing the priority of melody to harmony in music, the Essay on the Origin of Languages articulates the necessarily melodic, and hence temporal, character of thinking, which proves to be the key to understanding both why Rousseau must write as he does and what it means for language to be musical.

ABSTRACT Aims . Hyperkinetic seizures are usually associated with frontal lobe epilepsy. However, some patients have hyperkinetic seizures of temporal lobe origin. The semiological differences in hyperkinetic seizures between frontal and temporal lobe epilepsy have not been well studied. Here, we retrospectively assessed ictal semiology in order to distinguish between hyperkinetic seizures of frontal lobe origin and those of temporal lobe origin. Methods . We retrospectively reviewed data on patients who had undergone surgery for hyperkinetic seizures of temporal or frontal lobe origin and achieved favourable seizure outcomes (Engel Class I) with a minimum postoperative follow‐up of 24 months. We reviewed seizure histories, imaging reports, video‐EEG monitoring data, operative records, and pathological findings. We analysed and compared the hyperkinetic semiology of video‐recorded seizures of temporal lobe origin and those of frontal lobe origin. Results . Forty hyperkinetic seizures in eight patients (seven adult patients and one 12‐year‐old patient) with temporal lobe epilepsy and 45 hyperkinetic seizures in nine patients (eight adult patients and one 16‐year‐old patient) with frontal lobe epilepsy were analysed. Emotional facial expressions (such as fear, laughing, or anger), bilateral forceful elbow flexion, bilateral forceful grasping, facial flushing, and bilateral facial contraction were observed significantly more frequently in seizures of frontal lobe origin. Oroalimentary automatisms, seizures during wakefulness, salivation, and bilateral drop of the corners of the mouth were observed significantly more frequently in seizures of temporal lobe origin. Conclusions . Observation of a number of signs during hyperkinetic manifestations may help to predict whether a seizure originates from the frontal lobe or the temporal lobe.

Background: Temporal Lobe Epilepsy (TLE) comprises the most common form of symptomatic refractory focal epilepsy in adults. Accurate lateralization and localization of the epileptogenic focus are a significant prerequisite for determining surgical candidacy once the patient has been deemed medically intractable. Structural MR imaging, clinical, electrophysiological, and neurophysiological data have an established role in the localization of the epileptogenic foci. Nevertheless, hippocampal sclerosis cannot be detected on MR images in more than 30% of patients with TLE, and the presurgical assessment remains controversial. </P><P> Discussion: In the last years, advanced MR imaging techniques, such as 1H-MRS, DWI, DTI, DSCI, and fMRI, may provide valuable additional information regarding the physiological and metabolic characterization of brain tissue. MR imaging has shifted towards functional and molecular imaging, thus, promising to improve the accuracy regarding the lateralization and the localization of the epileptogenic focus. Additionally, nuclear medicine studies, such as SPECT and PET imaging modalities, have become an asset for the decoding of brain function and activity, and can be diagnostically helpful as well, since they provide valuable data regarding the altered metabolic activity of the seizure foci. Conclusion: Overall, advanced MRI, SPECT, and PET imaging techniques are increasingly becoming an essential part of TLE diagnostics, when the epileptogenic area is not identified on structural MRI or when structural MRI, clinical, and electrophysiological findings are not in concordance.

Purpose The purpose of this paper is to enrich country of origin (COO) effect in international marketing theory by adding the understanding of temporal dynamism into COO research. Design/methodology/approach Utilizing a qualitative and interdisciplinary phenomenological approach, this paper analyses historical and contemporary sources triangulated with contemporary primary interview data. The example of how perceptions of Italians about the values typical of the British Sixties varied over time periods is presented. Findings COO perceptions are both malleable and in evolution. Results show that values from earlier peak periods of appeal can be combined and recombined differently over time due to the varying historical and contemporary resonances of COO values. Research limitations/implications This study focuses on COO applied to two product areas, fashion and music, over a limited time period, in a two-country study and so the findings are not fully generalizable, but rather are transferable to similar contexts. Practical implications The fact that COO is neither static nor atemporal facilitates a segmented approach for international marketing managers to review and renew international brands. This enriched COO theory provides a rich and variable resource for developing and revitalizing brands. Originality/value The major contribution of this paper is that temporal dynamism, never before discussed in international marketing theory, renders COO theory more timeless; this addresses some critiques recently made about its relevance and practicality. The second contribution is the original research design that models interdisciplinary scholarship, enabling a thorough historical look at international marketing.

In behavior, action and perception are inherently interdependent. However, the actual mechanistic contributions of the motor system to sensory processing are unknown. We present neurophysiological evidence that the motor system is involved in predictive timing, a brain function that aligns temporal fluctuations of attention with the timing of events in a task-relevant stream, thus facilitating sensory selection and optimizing behavior. In a magnetoencephalography experiment involving auditory temporal attention, participants had to disentangle two streams of sound on the unique basis of endogenous temporal cues. We show that temporal predictions are encoded by interdependent delta and beta neural oscillations originating from the left sensorimotor cortex, and directed toward auditory regions. We also found that overt rhythmic movements improved the quality of temporal predictions and sharpened the temporal selection of relevant auditory information. This latter behavioral and functional benefit was associated with increased signaling of temporal predictions in right-lateralized frontoparietal associative regions. In sum, this study points at a covert form of auditory active sensing. Our results emphasize the key role of motor brain areas in providing contextual temporal information to sensory regions, driving perceptual and behavioral selection.

This research investigates the feasibility of adapting CONTRAM to the problem of determining the temporal distribution of traffic demand in an urban transport network. It combines CONTRAM with a second model which determines departure time selection as a function of the output of CONTRAM. The two worked iteratively together. The research considères the single-origin destination and the more realistic network with multiple origins and destinations and multiple routes. The resulting behaviours of demand and of traffic in the system were compared for a variety of changes to base networks. Although some of the single origin–destination pair networks reached an equilibrium solution, demand for the multiple origin–destination problem never converged and showed variations from iteration to iteration within a band. Comparisons of the results of average demand distributions showed that CONTRAM could be applied to this problem, and used to compare alternative network plans for the multiple origin–destination problem, as it provided reasonable and logical results. CONTRAM permits the modelling of a maximum of 13 time intervals. Therefore, as the time frame of study increases, a coarser approximation of continuous demand will result, and will limit the application. ABSTRACT Hydroxyurea (HU) is a DNA replication inhibitor that negatively affects both the elongation and initiation phases of replication and triggers the “intra-S phase checkpoint.” Previous work with budding yeast has shown that, during a short exposure to HU, MEC1/RAD53 prevent initiation at some late S phase origins. In this study, we have performed microarray experiments to follow the fate of all origins over an extended exposure to HU. We show that the genome-wide progression of DNA synthesis, including origin activation, follows the same pattern in the presence of HU as in its absence, although the time frames are very different. We find no evidence for a specific effect that excludes initiation from late origins. Rather, HU causes S phase to proceed in slow motion; all temporal classes of origins are affected, but the order in which they become active is maintained. We propose a revised model for the checkpoint response to HU that accounts for the continued but slowed pace of the temporal program of origin activation.

In Saccharomyces cerevisiae, the forkhead (Fkh) transcription factor Fkh1 (forkhead homolog) enhances the activity of many DNA replication origins that act in early S-phase (early origins). Current models posit that Fkh1 acts directly to promote these origins’ activity by binding to origin-adjacent Fkh1 binding sites (FKH sites). However, the post-DNA binding functions that Fkh1 uses to promote early origin activity are poorly understood. Fkh1 contains a conserved FHA (forkhead associated) domain, a protein-binding module with specificity for phosphothreonine (pT)-containing partner proteins. At a small subset of yeast origins, the Fkh1-FHA domain enhances the ORC (origin recognition complex)-origin binding step, the G1-phase event that initiates the origin cycle. However, the importance of the Fkh1-FHA domain to either chromosomal replication or ORC-origin interactions at genome scale is unclear. Here, S-phase SortSeq experiments were used to compare genome replication in proliferating FKH1 and fkh1-R80A mutant cells. The Fkh1-FHA domain promoted the activity of ≈ 100 origins that act in early to mid- S-phase, including the majority of centromere-associated origins, while simultaneously inhibiting ≈ 100 late origins. Thus, in the absence of a functional Fkh1-FHA domain, the temporal landscape of the yeast genome was flattened. Origins are associated with a positioned nucleosome array that frames a nucleosome depleted region (NDR) over the origin, and ORC-origin binding is necessary but not sufficient for this chromatin organization. To ask whether the Fkh1-FHA domain had an impact on this chromatin architecture at origins, ORC ChIPSeq data generated from proliferating cells and MNaseSeq data generated from G1-arrested and proliferating cell populations were assessed. Origin groups that were differentially regulated by the Fkh1-FHA domain were characterized by distinct effects of this domain on ORC-origin binding and G1-phase chromatin. Thus, the Fkh1-FHA domain controlled the distinct chromatin architecture at early origins in G1-phase and regulated origin activity in S-phase.

However, subgroup analysis of the seizures of temporal lobe origin suggests a role for motor automatisms in discriminating seizures of neocortical versus mesial temporal lobe origin.Methods:Video-EEG of a patient with well-defined neocortical temporal lobe epilepsy was reviewed to assess the localizing value of motor automatisms.Results:We report a patient with left upper extremity motor automatisms and clonic movements of the proximal left lower extremity with altered awareness as the sole manifestations of right temporal neocortical seizures.Conclusion:Early onset unilateral motor automatisms without dystonic posturing can localize the seizure origin to the contralateral temporal lobe neocortex.

Abstract A patient with medically intractable status epilepticus of temporal lobe origin is presented. A computed tomogram showed a low density area adjacent to the midbrain, possibly related to atrophy of the medial temporal lobe. Cerebral angiography revealed early filling veins and an anterior temporal blush. Magnetic resonance (MR) scanning (T2 weighted images) showed increased signal intensity in the region of the amygdala and anterolateral left temporal lobe. Ictal activity was recorded from scalp electrodes over the left temporal area, and many paroxysms were recorded from cortical surface electrodes. An anterior temporal lobectomy revealed only gliosis. The cerebral blood flow changes accompanying status epilepticus of focal origin are reviewed, and a possible relation of electroencephalographic, angiographic, and MR findings is discussed.

The tumour consisted of a main mass with the appearance of an acoustic neuroma but close by and not connected were some nests of tumour cells inside the vestibule. This very unusual finding raises questions of the aetiology of this tumour which may have a bearing on the aetiology of other tumours of the VIIIth. nerve.

With increasing numbers of international flights and air travelers arriving in the US annually, the rapid spread of communicable diseases has grown. Epidemics of novel infectious diseases have emerged and rapidly spread globally in association with air travel, including the severe acute respiratory syndrome (SARS) outbreak in 2003 and H1N1 in 2009. In order to anticipate and mitigate the consequences of future rapid disease spread, the MITRE Corporation, in collaboration with the (US) Centers for Disease Control and Prevention, developed a risk assessment tool using a Susceptible-Exposed-Infectious-Recovered model and detailed flight and population data. The emergence and spread of prototypic pandemic influenza was simulated based on a theoretical geographical point of origin and its communicability. More than 50 international metropolitan areas were analyzed as potential points of origin to simulate the rapidity of spread to the US. The basic reproduction number (Ro), defined as the average number of persons to whom one infected individual transmits disease in an immune naive population, was varied from 1.4 to 1.9. The starting numbers of infectious persons at each origin also were varied (100 or 500 persons, 5% infectious may travel). Waves were computed as aggregate across metropolitan areas modeled in the US. The visualization of the first pandemic wave was most apparent in simulations of Ro = 1.9, resulting from 500 infectious persons at each origin. More than 50% of origins indicated that aggregate waves peaked around Day 125, while 30% of origins peaked around Day 90. Additionally, the time, in days, from its origin in six continents into the US was compared, and a two-week delay was found from South America compared with other continents. This simulation tool better equips policy makers and public health officials to quickly assess risk and leverage resources efficiently via targeted and scalable border mitigation measures during a rapid global outbreak.

Abstract. Self-similarity may stem from two origins: the process increments infinite variance and/or process memory. The b-value of the Gutenberg-Richter law comes from the first origin. In the frame of natural time analysis of earthquake data, a fall of the b-value observed before large earthquakes reflects an increase of the order parameter fluctuations upon approaching the critical point (mainshock). The increase of these fluctuations, however, is also influenced from the second origin of self-similarity, i.e., temporal correlations between earthquake magnitudes. This is supported by observations and simulations of an earthquake model.

We have examined the distribution of early replicating origins on stretched DNA fibers when nuclei from CHO cells synchronized at different times during G1 phase initiate DNA replication in Xenopus egg extracts. Origins were differentially labeled in vivo versus in vitro to allow a comparison of their relative positions and spacing. With nuclei isolated in the first hour of G1 phase, in vitro origins were distributed throughout a larger number of DNA fibers and did not coincide with in vivo origins. With nuclei isolated 1 h later, a similar total number of in vitro origins were clustered within a smaller number of DNA fibers but still did not coincide with in vivo origins. However, with nuclei isolated later in G1 phase, the positions of many in vitro origins coincided with in vivo origin sites without further change in origin number or density. These results highlight two distinct G1 steps that establish a spatial and temporal program for replication.

Encephaloceles are pathological herniations of brain parenchyma through congenital or acquired osseus-dural defects of the skull base or cranial vault. Although encephaloceles are known as rare conditions, several surgical reports and clinical series focusing on spontaneous encephaloceles of the temporal lobe may be found in the otological, maxillofacial, radiological, and neurosurgical literature. A variety of symptoms such as occult or symptomatic CSF fistulas, recurrent meningitis, middle ear effusions or infections, conductive hearing loss, and medically intractable epilepsy have been described in patients harboring spontaneous encephaloceles of middle cranial fossa origin. Both open procedures and endoscopic techniques have been advocated for the treatment of such conditions. The authors discuss the pathogenesis, diagnostic assessment, and therapeutic management of spontaneous temporal lobe encephaloceles. Although diagnosis and treatment may differ on a case-by-case basis, review of the available literature suggests that spontaneous encephaloceles of middle cranial fossa origin are a more common pathology than previously believed. In particular, spontaneous cases of posteroinferior encephaloceles involving the tegmen tympani and the middle ear have been very well described in the medical literature.

Recent cladistic analyses have all suggested a diapsid origin of ichthyosaurs. However, an intermediate evolutionary stage of the lower temporal region of ichthyosaurian skull between basal diapsids and derived ichthyosaurs has been absent from the fossil record. Here we describe the cranial skeleton of a new mixosaurid ichthyosaur specimen with a well-preserved lower temporal region from the Anisian Guanling Formation of eastern Yunnan. It is characterized by the most primitive lower temporal region within known ichthyosaurs. The primitive characters of the lower temporal region include both external and internal separation between the jugal and the quadratojugal, an anterior process of the quadratojugal, an apparent posteroventral process of the jugal, and a large lower temporal opening surrounded by the jugal, the postorbital, the squamosal, and the quadratojugal. The lower temporal region of this specimen provides the most direct evidence to the diapsid origin of ichthyosaurs. It also suggests that the disappearance of the lower temporal fenestra is caused initially by the reduction of the lower temporal arcade rather than the enlargement of the surrounding bones.

With the rise of the online ride-hailing market, taxi demand prediction has received more and more research interest in intelligent transportation. However, most traditional research methods mainly focused on the demand based on the original point and ignored the intention of the passenger’s destination. At the same time, many forecasting methods need sufficient investigation and data processing, which undoubtedly increases the complexity and operability of forecasting problems. Therefore, we regard the current taxi demand prediction as an origin–destination problem in order to provide more accurate predictions for the taxi demand problem. By combining a spatial network based on graph convolutional network (GCN) and a temporal network of convolutional long short-term memory (Conv-LSTM), we propose a new spatial-temporal model of Conv-LSTM two-dimensional bidirectional GCN (CTBGCN) to uncover the potential correlation between origin and destination. We utilize the temporal network for effective temporal information and the spatial network of multi-layers to get the implicit origin–destination correlation. Numerical results suggest that the proposed method outperforms the state-of-the-art baseline and other traditional methods.

Replication origins in eukaryotic cells never fire more than once in a given S phase. Here, we summarize the role of cyclin–dependent kinases in limiting DNA replication origin usage to once per cell cycle in the budding yeast Saccharomyces cerevisiae . We have examined the role of different cyclins in the phosphorylation and regulation of several replication/regulatory factors including Cdc6, Sic1, ORC and DNA polymerase α–primase. In addition to being regulated by the cell cycle machinery, replication origins are also regulated by the genome integrity checkpoint kinases, Mec1 and Rad53. In response to DNA damage or drugs which interfere with the progression of replication forks, the activation of late–firing replication origins is inhibited. There is evidence indicating that the temporal programme of origin firing depends upon the local histone acetylation state. We have attempted to test the possibility that checkpoint regulation of late–origin firing operates through the regulation of the acetylation state. We found that overexpression of the essential histone acetylase, Esa1, cannot override checkpoint regulation of origin firing. We have also constructed a temperature–sensitive esa1 mutant. This mutant is unable to resume cell cycle progression after α–factor arrest. This can be overcome by overexpression of the G1 cyclin, Cln2, revealing a novel role for Esa1 in regulating Start.

ABSTRACT To clarify the pathogenesis of a typical case of musicogenic epilepsy, we examined interictal spikes using the dipole tracing method (DTM). The patient was a 49‐year‐old, right‐handed Japanese man. He first experienced seizures at the age of 32 years; listening to his favorite piece of music frequently triggered them. His seizure type is partial (often complex, but sometimes simple). An interictal EEG examination revealed many focal spikes in F8 and T4. We estimated equivalent current dipoles (ECDs) using DTM. We performed one‐dipole analyses on the peaks of the spikes using an EEG analyzer with a three‐layer head model called the scalp‐skull‐brain (SSB) model. We analyzed the interictal EEG because there were no spikes during the seizure. The ECDs were located in the posterior transverse temporal gyrus. The characteristics in this patient not only bolstered arguments in favor of the role of the right temporal lobe in musicogenic epilepsy, but also showed that transverse temporal gyri, which are included in the auditory area, could play an important role in musicogenic epilepsy. Initiation of DNA replication in eukaryotic cells is regulated through the ordered assembly of replication complexes at origins of replication. Association of Cdc45 with the origins is a crucial step in assembly of the replication machinery, hence can be considered a target for the regulation of origin activation. To examine the process required for SpCdc45 loading, we isolated fission yeast SpSld3, a counterpart of budding yeast Sld3 that interacts with Cdc45. SpSld3 associates with the replication origin during G1–S phases and this association depends on Dbf4-dependent (DDK) kinase activity. In the corresponding period, SpSld3 interacts with minichromosome maintenance (MCM) proteins and then with SpCdc45. A temperature-sensitive sld3-10 mutation suppressed by the multicopy of the sna41+encoding SpCdc45 impairs loading of SpCdc45 onto chromatin. In addition, this mutation leads to dissociation of preloaded Cdc45 from chromatin in the hydroxyurea-arrested S phase, and DNA replication upon removal of hydroxyurea is retarded. Thus, we conclude that SpSld3 is required for stable association of Cdc45 with chromatin both in initiation and elongation of DNA replication. The DDK-dependent origin association suggests that SpSld3 is involved in temporal regulation of origin firing.
