Dinosaurs have been major components of ecosystems for over 200 million years. Although different macroevolutionary scenarios exist to explain the Triassic origin and subsequent rise to dominance of dinosaurs and their closest relatives (dinosauromorphs), all lack critical support from a precise biostratigraphically independent temporal framework. The absence of robust geochronologic age control for comparing alternative scenarios makes it impossible to determine if observed faunal differences vary across time, space, or a combination of both. To better constrain the origin of dinosaurs, we produced radioisotopic ages for the Argentinian Chañares Formation, which preserves a quintessential assemblage of dinosaurian precursors (early dinosauromorphs) just before the first dinosaurs. Our new high-precision chemical abrasion thermal ionization mass spectrometry (CA-TIMS) U–Pb zircon ages reveal that the assemblage is early Carnian (early Late Triassic), 5- to 10-Ma younger than previously thought. Combined with other geochronologic data from the same basin, we constrain the rate of dinosaur origins, demonstrating their relatively rapid origin in a less than 5-Ma interval, thus halving the temporal gap between assemblages containing only dinosaur precursors and those with early dinosaurs. After their origin, dinosaurs only gradually dominated mid- to high-latitude terrestrial ecosystems millions of years later, closer to the Triassic–Jurassic boundary.

In the mammalian genome, early- and late-replicating domains are often separated by temporal transition regions (TTRs) with novel properties and unknown functions. We identified a TTR in the mouse immunoglobulin heavy chain (Igh) locus, which contains replication origins that are silent in embryonic stem cells but activated during B cell development. To investigate which factors contribute to origin activation during B cell development, we systematically modified the genetic and epigenetic status of the endogenous Igh TTR and used a single-molecule approach to analyze DNA replication. Introduction of a transcription unit into the Igh TTR, activation of gene transcription, and enhancement of local histone modifications characteristic of active chromatin did not lead to origin activation. Moreover, very few replication initiation events were observed when two ectopic replication origin sequences were inserted into the TTR. These findings indicate that the Igh TTR represents a repressive compartment that inhibits replication initiation, thus maintaining the boundaries between early and late replication domains.

AbstractThe argument of Rousseau's Essay on the Origin of Languages is intimately connected with that of his Discourse on the Origin and the Foundations of Inequality among Men. The origin of political society is inseparable from the origin of language, which, in turn, is inseparable from the origin of reason. That so much of the Essay is concerned with music leads us to wonder what music has to do with reason, politics, and language. These two books share what is a regular feature of Rousseau's manner of writing—presenting what seem to be logical foundations as temporal origins. In emphasizing the priority of melody to harmony in music, the Essay on the Origin of Languages articulates the necessarily melodic, and hence temporal, character of thinking, which proves to be the key to understanding both why Rousseau must write as he does and what it means for language to be musical.

ABSTRACT Aims . Hyperkinetic seizures are usually associated with frontal lobe epilepsy. However, some patients have hyperkinetic seizures of temporal lobe origin. The semiological differences in hyperkinetic seizures between frontal and temporal lobe epilepsy have not been well studied. Here, we retrospectively assessed ictal semiology in order to distinguish between hyperkinetic seizures of frontal lobe origin and those of temporal lobe origin. Methods . We retrospectively reviewed data on patients who had undergone surgery for hyperkinetic seizures of temporal or frontal lobe origin and achieved favourable seizure outcomes (Engel Class I) with a minimum postoperative follow‐up of 24 months. We reviewed seizure histories, imaging reports, video‐EEG monitoring data, operative records, and pathological findings. We analysed and compared the hyperkinetic semiology of video‐recorded seizures of temporal lobe origin and those of frontal lobe origin. Results . Forty hyperkinetic seizures in eight patients (seven adult patients and one 12‐year‐old patient) with temporal lobe epilepsy and 45 hyperkinetic seizures in nine patients (eight adult patients and one 16‐year‐old patient) with frontal lobe epilepsy were analysed. Emotional facial expressions (such as fear, laughing, or anger), bilateral forceful elbow flexion, bilateral forceful grasping, facial flushing, and bilateral facial contraction were observed significantly more frequently in seizures of frontal lobe origin. Oroalimentary automatisms, seizures during wakefulness, salivation, and bilateral drop of the corners of the mouth were observed significantly more frequently in seizures of temporal lobe origin. Conclusions . Observation of a number of signs during hyperkinetic manifestations may help to predict whether a seizure originates from the frontal lobe or the temporal lobe.

Background: Temporal Lobe Epilepsy (TLE) comprises the most common form of symptomatic refractory focal epilepsy in adults. Accurate lateralization and localization of the epileptogenic focus are a significant prerequisite for determining surgical candidacy once the patient has been deemed medically intractable. Structural MR imaging, clinical, electrophysiological, and neurophysiological data have an established role in the localization of the epileptogenic foci. Nevertheless, hippocampal sclerosis cannot be detected on MR images in more than 30% of patients with TLE, and the presurgical assessment remains controversial. </P><P> Discussion: In the last years, advanced MR imaging techniques, such as 1H-MRS, DWI, DTI, DSCI, and fMRI, may provide valuable additional information regarding the physiological and metabolic characterization of brain tissue. MR imaging has shifted towards functional and molecular imaging, thus, promising to improve the accuracy regarding the lateralization and the localization of the epileptogenic focus. Additionally, nuclear medicine studies, such as SPECT and PET imaging modalities, have become an asset for the decoding of brain function and activity, and can be diagnostically helpful as well, since they provide valuable data regarding the altered metabolic activity of the seizure foci. Conclusion: Overall, advanced MRI, SPECT, and PET imaging techniques are increasingly becoming an essential part of TLE diagnostics, when the epileptogenic area is not identified on structural MRI or when structural MRI, clinical, and electrophysiological findings are not in concordance.

Purpose The purpose of this paper is to enrich country of origin (COO) effect in international marketing theory by adding the understanding of temporal dynamism into COO research. Design/methodology/approach Utilizing a qualitative and interdisciplinary phenomenological approach, this paper analyses historical and contemporary sources triangulated with contemporary primary interview data. The example of how perceptions of Italians about the values typical of the British Sixties varied over time periods is presented. Findings COO perceptions are both malleable and in evolution. Results show that values from earlier peak periods of appeal can be combined and recombined differently over time due to the varying historical and contemporary resonances of COO values. Research limitations/implications This study focuses on COO applied to two product areas, fashion and music, over a limited time period, in a two-country study and so the findings are not fully generalizable, but rather are transferable to similar contexts. Practical implications The fact that COO is neither static nor atemporal facilitates a segmented approach for international marketing managers to review and renew international brands. This enriched COO theory provides a rich and variable resource for developing and revitalizing brands. Originality/value The major contribution of this paper is that temporal dynamism, never before discussed in international marketing theory, renders COO theory more timeless; this addresses some critiques recently made about its relevance and practicality. The second contribution is the original research design that models interdisciplinary scholarship, enabling a thorough historical look at international marketing.

The large numbers of neurons that comprise the adult brain display an immense diversity. Repeated divisions of a relatively small pool of neural stem cells generate this neuronal diversity during development. To increase progress towards medical treatments for neurodegenerative diseases, it is of interest to understand both how neural stem cells generate the assortment of neurons and how these neurons come together to form a functional brain. Brain assembly occurs sequentially across time with early events laying the foundation for later events. Drosophila neural stem cells, neuroblasts (NBs), are an excellent model for investigating how neural diversity is generated and what roles early and late born neurons have in shaping the stereotypical adult brain structure. Generation of neural diversity, begins with specifying the diverse population of stem cells, called spatial patterning, and continues with diversifying neurons made from the diverse stem cells, called temporal patterning. Drosophila NBs exhibit both spatial and temporal patterning. Drosophila NBs have three types of division modes: type 0, type I and type II. Type II NBs expand the number of neurons made with progeny that exhibit a transit-amplifying division pattern, similar to that of mammalian outer subventricular zone (OSVZ) progenitors. Additionally, type II NBs exhibit temporal patterning across both the NB and their progeny to generate a large diversity of neurons that populate a conserved region of the brain responsible for many sensory and motor functions, called the central complex. Type II NBs have only been identified and studied during later stages in development, with nothing known about their origin or early divisions. In this dissertation, I describe the early lineages of the type II NBs within the Drosophila embryo. I show that type II NBs and lineages originate early in development, exhibit temporal patterning across both the NB and transit-amplifying progeny, and produce neurons that survive into the adult brain to innervate and potentially serve as a foundation within the adult central complex. Additionally, I explain how live imaging of the developing Drosophila brain can answer questions not easily addressed through other methods.

Current works of bike demand prediction mainly focus on cluster level and perform poorly on predicting demands of a single station. In the first task, we introduce a contextual based bike demand prediction model, which predicts bike demands for per station by combining spatio-temporal network and environment contexts synergistically. Furthermore, since people's movement information is an important factor, which influences the bike demands of each station. To have a better understanding of people's movements, we need to analyze the relationship between different places. In the second task, we propose an origin-destination model to learn place representations by using large scale movement data. Then based on the people's movement information, we incorporate the place embedding into our bike demand prediction model, which is built by using multi-source large scale datasets: New York Citi bike data, New York taxi trip records, and New York POI data. Finally, as deep learning methods have been successfully applied to many fields such as image recognition and natural language processing, it inspires us to incorporate the complex deep learning method into the bike demand prediction problem. So in this task, we propose a deep spatial-temporal (DST) model, which contains three major components: spatial dependencies, temporal dependencies, and external influence. Experiments on the NYC Citi Bike system show the effectiveness and efficiency of our model when compared with the state-of-the-art methods.

At the Castle Creek study area a remarkably well exposed section in the Isaac Formation, Windermere Supergroup, was measured to document potential lateral and vertical changes in facies. Here, a consistent lateral succession of facies was observed consisting of matrix-poor sandstone to clayey sandstone to bipartite bed to sandy claystone, collectively overlain by a thin-bedded turbidite and mudstone cap. These changes are interpreted to reflect particle settling in a negligibly-sheared sand-mud suspension following detachment from the margins of an avulsion jet. Vertically, similar facies stack to form packages 2-7 beds thick implying temporary stabilization of jet-margin dynamics. At a larger scale facies stack to form three stratal assemblages. SA-1: intercalated matrix-rich and matrix-poor strata and SA-2: matrix-rich strata stack vertically and laterally but then are sharply overlain everywhere by classical turbidites of SA-3, indicating a dramatic change from deposition immediately downflow of an avulsion node to conventional levee deposition.

Annotation:
In the dentate gyrus (DG) of the hippocampus, the generation of dentate granule neurons(DGNs) starts during late embryogenesis, peaks around birth and continues at low levels during adulthood. This continuous neurogenesis makes the DG a unique structure, composed of DGNs from distinct temporal origins, which form subpopulations potentially bearing unique anatomical characteristics and functional roles in hippocampal physiology. Surprisingly, this hypothesis has received limited attention. In this context, our research aimed to elucidate the morphological, electrophysiological, and behavioral characteristics of DGNs subpopulations based on their temporal origin. Building on prior findings from our team that high lighted dendritic differences between these populations, we focused on examining the features of their axons, called mossy fibers (MFs). Using sparse labeling strategies — electroporation to targetembryonically-born (E14.5) and neonatally-born (P0) DGNs, and retroviral injections foradolescent-born (P21) and adult-born (P84) DGNs — we uncovered that DGNs generated laterin life develop larger MF boutons with more filopodia, and exhibit a shorter axon initialsegment. Additionally, using the Osteocalcin-Cre and Ascl1CreERT2 mouse lines to selectivelylabel large cohorts of embryonically-born and adult-born DGNs, respectively, we found thatearlier-born neurons project further onto the CA2 compared to later-born neurons. Following these morphological findings, we further investigated the functional characteristics of temporally distinct DGNs at both the electrophysiological and behavioral levels. The electrophysiological studies revealed similar intrinsic properties between neonatally- and adult born DGNs, and higher basal transmission in neonatally-born DGNs, potentially reflecting alarger number of active sites. Finally, we examined the role of embryonic-born DGNs in socialbehavior, and showed that acute inhibition of these neurons delayed the expression of social preference. However, these functional data remain preliminary and need further investigation.Altogether, this PhD work highlights the significant impact of the birthdate of DGNs on their anatomical and potentially functional characteristics, and emphasizes the importance of considering their precise temporal origin in any structural or functional analysis of the DG

The monograph presents the author's concept of information modeling of the world based on navigation — orientation and movement. The concept establishes links between the spatial and temporal dynamics of the Earth's nature, human evolution and the formation of geocultural space. It is shown that the construction of ancient labyrinths, petroglyphs, and stone sculptures allows them to perform the functions of a gnomon and a sundial, and the mythopoeic plots reflect elements of the celestial sphere. The theoretical foundations, methods and typical examples of studies of prehistoric objects of navigation purpose are considered. It is intended for specialists and students in the fields of cultural geography and tourism, archeology, anthropology, ethnography, semiotics, philology and philosophy, as well as for a wide range of readers interested in new research results on the prehistoric origins of modern culture.

The reports of the students indicate the Erasmus as a 'bubble of experience' and the book explores these inner experiences through a sociological approach, illustrating the vast potential in terms of the moulding of a 'homo novus Europaeus'. The data gathered prompt a reflection on the redefinition of the role of the student when he or she directly experiences the comparison with a context different and distant from that of origin, to which he or she is nevertheless destined to return. From this perspective, the Erasmus experience assumes the significance of a sort of temporary upheaval of status open to forms of 'experimentation of identity'.

The analysis of origin-destination traffic flows may be useful in many contexts of application (e.g., urban planning, tourism economics) and have been commonly studied through the gravity model, which states that flows are proportional to ''masses" of both origin and destination, and inversely proportional to distance between them. Using data on the flow of mobile phone SIM among different aree di censimento, recorded hourly basis for several months and provided by FasterNet in the context of MoSoRe project, in this work we characterize and model the dynamic of such flows over the time in the strongly urbanized and flood-prone area of the Mandolossa (western outskirts of Brescia, northern Italy), with the aim of predicting the traffic flow during flood episodes. Whereas a traditional ”static” mass explanatory variable is represented by residential population (Pop), or by gross domestic product (GDP), here we propose to use a most accurate set of explanatory variables in order to better account for the dynamic over the time. First, we employ a time-varying mass variable represented by the number of city-users by area and by time period, which has been estimated from mobile phone data (provided by TIM) using functional data approach and already adopted to derive crowding maps for flood exposure. Secondly, we include in the model a proper set of factors such as areal and time dummies, and a novel set of indices related to (e.g.) the number and the type of streets, the number of offices, restaurants or cinemas, which may be retrieved from OpenStreetMap. The joint use of these two novel sets of explanatory variables should allow us to obtain a better linear fitting of the gravity model and a better traffic flow prediction for the flood risk evaluation.

AbstractIndigenous peoples the world over are speaking out for their rights in former colonial societies. The term Indigenous, derived from Latin, means within, originating where it is found, or belonging to a particular place by birth or origin, a temporal claim to a place. In an archaeological sense, the San can claim to be the true Indigenous people in all of southern Africa, having lived in the region for thousands of years, before any migrations, and well before any colonial onslaught. Yet in the Northern Cape, South Africa, well-known for a significant concentration of rock engravings and archaeological sites, the current San inhabitants are the most recent arrivals, with no record of an Indigenous population since at least the mid-nineteenth century. In 1999 the South African government resettled some 400 formerly military !Xu and Khwe families of different origins, language backgrounds, and histories in Platfontein without any deliberations about their relation to local boundaries, history or heritage. Indigeneity here is far more complicated and vexing. In this chapter I probe the quest for an authentic Indigenous past of ancient images, to show that the complex history of postcolonial locales demands that archaeologists attend to the dislocations and violence of global forces of the past hundreds of years. The insistence on ancient roots of Indigenous people in a place can effectively deprive them of a role in global history, and of agency in political events. Contested spaces, centuries of conflict, truce, and temporary agreements that fester and erupt with unsurprising regularity are all a part of the context that frames ancient images. We should account for this context when studying them, in order to avoid one-dimensional, simplistic notions of Indigenous heritage.

While quadratic solitons had been predicted to exist theoretically and have been observed recently in planar waveguides and bulk quadratic crystals,[1-3] temporal soliton formation, or compensation for the refractive index dispersion by the quadratic nonlinearity, is difficult to be achieved in single path configurations because of the lack of materials with large enough dispersion. In other words, while the nonlinearities can afford parametric gain and phase front distortions, dispersion lengths are in the best case scenarios in the order of 10 cm which are difficult to be achieved experimentally with currently known quadratic materials.[4]

A near-global framework for automated training data generation and land cover classification using shallow machine learning with low-density time series imagery does not exist. This study presents a methodology to map nine-class, six-class, and five-class land cover using two dates of a Sentinel-2 granule across seven international sites. The approach uses a series of spectral, textural, and distance decision functions combined with modified ancillary layers to create binary masks from which to generate a balanced set of training data applied to a random forest classifier. For the land cover masks, stepwise threshold adjustments were applied to reflectance, spectral index values, and Euclidean distance layers, with 62 combinations evaluated. Global and regional adaptive thresholds were computed. An annual 95th and 5th percentile NDVI composite was used to provide temporal corrections to the decision functions, and these corrections were compared against the original model. The accuracy assessment found that the regional adaptive thresholds for both the two-date land cover and the temporally corrected land cover could accurately map land cover type within nine-class, six-class, and five-class schemes. Lastly, the five-class and six-class models were compared with a manually labeled deep learning model (Esri), where they performed with similar accuracies. The results highlight performance in line with an intensive deep learning approach, and reasonably accurate models created without a full annual time series of imagery.

Annotation:
Arabidopsis mutants that we have isolated, encode for fertilization-independent endosperm (fie), fertilization-independent seed2 (fis2) and medea (mea) genes, act in the female gametophyte and allow endosperm to develop without fertilization when mutated. We cloned the FIE and MEA genes and showed that they encode WD and SET domain polycomb (Pc G) proteins, respectively. Homologous proteins of FIE and MEA in other organisms are known to regulate gene transcription by modulating chromatin structure. Based on our results, we proposed a model whereby both FIE and MEA interact to suppress transcription of regulatory genes. These genes are transcribed only at proper developmental stages, as in the central cell of the female gametophyte after fertilization, thus activating endosperm development. To test our model, the following questions were addressed: What is the Composition and Function of the Polycomb Complex? Molecular, biochemical, genetic and genomic approaches were offered to identify members of the complex, analyze their interactions, and understand their function. What is the Temporal and Spatial Pattern of Polycomb Proteins Accumulation? The use of transgenic plants expressing tagged FIE and MEA polypeptides as well as specific antibodies were proposed to localize the endogenous polycomb complex. How is Polycomb Protein Activity Controlled? To understand the molecular mechanism controlling the accumulation of FIE protein, transgenic plants as well as molecular approaches were proposed to determine whether FIE is regulated at the translational or posttranslational levels. The objectives of our research program have been accomplished and the results obtained exceeded our expectation. Our results reveal that fie and mea mutations cause parent-of-origin effects on seed development by distinct mechanisms (Publication 1). Moreover our data show that FIE has additional functions besides controlling the development of the female gametophyte. Using transgenic lines in which FIE was not expressed or the protein level was reduced during different developmental stages enabled us for the first time to explore FIE function during sporophyte development (Publication 2 and 3). Our results are consistent with the hypothesis that FIE, a single copy gene in the Arabidopsis genome, represses multiple developmental pathways (i.e., endosperm, embryogenesis, shot formation and flowering). Furthermore, we identified FIE target genes, including key transcription factors known to promote flowering (AG and LFY) as well as shoot and leaf formation (KNAT1) (Publication 2 and 3), thus demonstrating that in plants, as in mammals and insects, PcG proteins control expression of homeobox genes. Using the Yeast two hybrid system and pull-down assays we demonstrated that FIE protein interact with MEA via the N-terminal region (Publication 1). Moreover, CURLY LEAF protein, an additional member of the SET domain family interacts with FIE as well. The overlapping expression patterns of FIE, with ether MEA or CLF and their common mutant phenotypes, demonstrate the versatility of FIE function. FIE association with different SET domain polycomb proteins, results in differential regulation of gene expression throughout the plant life cycle (Publication 3). In vitro interaction assays we have recently performed demonstrated that FIE interacts with the cell cycle regulatory component Retinobalsoma protein (pRb) (Publication 4). These results illuminate the potential mechanism by which FIE may restrain embryo sac central cell division, at least partly, through interaction with, and suppression of pRb-regulated genes. The results of this program generated new information about the initiation of reproductive development and expanded our understanding of how PcG proteins regulate developmental programs along the plant life cycle. The tools and information obtained in this program will lead to novel strategies which will allow to mange crop plants and to increase crop production.

Frozen soil foundations are prone to frost heave and thawing under freeze-thaw cycles, which can cause substantial damage to road structures in permafrost areas. The performance of the thermal insulation of the foamed mixture soil subgrade of the S301 highway in Xinjiang was examined as a case study, and the temporal and spatial variation in temperature and strain of the foamed mixture soil subgrade analyzed and compared with those of the original gravel subgrade. The results show that (1) the thermal insulation performance of the foamed mixture soil subgrade is excellent, temperatures tend to stabilize quickly, and changes in amplitude are small—only 8–40% of that of the gravel subgrade; (2) the thermal energy is converted into elastic strain energy for release, which supports the thermal stability of the frozen soil foundation; and (3) a physical model of heat conduction in the foamed mixture soil subgrade structure was established, revealing the mechanism of thermal insulation. This provides a possible solution to help mitigate against frost expansion, thawing, and subsiding of frozen soil foundations.

Once considered a partial exception to the recent diffusion of populism worldwide, Romania saw Radical Right populism return to Parliament in 2020. The Alliance for the Union of Romanians (AUR) successfully campaigned on a platform of defending the Christian faith, freedom, the traditional family, and the nation. Although the party was initially considered the result of individual entrepreneurship linked to its founding leaders, it has successfully built on diffused networks of societal activism whose origins could be traced back to the early 2000s. However, the AUR’s track record of discourse aligned with Kremlin rhetoric calling for Western economic, political and cultural hegemony to be resisted and rolled back saw a temporary decline in voters’ support for the party. However, the party managed to rebuild consensus strategically by drawing on voters’ increased anxiety regarding the economic effects of the war. This report offers a cogent analysis of the political performance of the AUR, examining the party’s formative phase as well as its evolution since 2020, alongside a discussion of the impact of the war in Ukraine on Romanian party politics.

Navigation channels are periodically dredged to maintain safe depths. Dredged sediment was historically placed in upland management areas or in offshore disposal areas. Florida state law prohibits placement of beach fill sediment that contains more than 10% by weight of silt and clay, which is typically a characteristic of dredged material. An alternative is placement in a nearshore berm. Some potential benefits of nearshore berms include wave energy dissipation, reduced cost of dredging and shore protection, and possible onshore movement of the berm material. This study considers sediment distribution, morphological evolution, sediment transport, and shoreline trends along Fort Myers Beach, Florida, related to the nearshore berm constructed in August 2016. Due to timing of the field study, this report also includes information on the influence of a major hurricane that impacted the area. The overall conclusion of this study is that the dredge-sourced sediment in the berm performed as expected. Within 2 years, the berm adjusted to the shoreface environment, maintained a large part of its original volume, and contributed to protection of the beach and shoreline. The impact of Hurricane Irma included a shift in sediment textures and a large but temporary increase in shoreface sediment volumes.

Department of Aquatic Resources, Swedish University of Agricultural Sciences, 2024. http://dx.doi.org/10.54612/a.4iseib7eup.

Annotation:
For decades, the population of the European eel has been in severe decline. In 2007, the European Union decided on a Regulation establishing measures for the recovery of the stock, which obliged Member States to implement a national Eel Management Plan by 2009. Sweden submitted its plan in 2008. According to the Regulation, Member States shall report regularly to the EU-Commission, on the implementation of their Eel Management Plans and the progress achieved in protection and restoration. The current report provides an assessment of the eel stock in Sweden as of spring 2024, intending to feed into the national reporting to the EU in August this year. This report updates and extends previous evaluation reports by Dekker (2012, 2015) and Dekker et al. (2018, 2021). In this report, the impacts on the stock - of fishing, restocking and mortality related to hydropower generation - are assessed. Other anthropogenic impacts (climate change, pollution, increased impacts of predators, spread of parasites, disruption of migration due to disorientation after transport, and so forth) probably have an impact on the stock too, but these factors are hardly quantifiable, and no management targets have been set. For that reason, and because most factors were not included in the EU Eel Regulation, these other factors are not included in this report. Our focus is on the quantification of silver eel biomass escaping from continental waters towards the ocean (current, current potential and pristine) and mortality risks endured by those eels during their whole lifetime. The assessment is broken down on a geographical basis, with different impacts dominating in different areas (west coast, inland waters, Baltic coast). In the last decade, a break in the downward trend in glass eel recruitment has been observed, with recruitment no longer declining consistently. Whether that relates to recent protective actions, or is due to other factors, is yet unclear. Nevertheless, recruitment levels remain at historically low levels. This report contributes to the required international assessment, but does not discuss the causing factors behind the recent recruitment trend and the overall status of the stock across Europe. For the different assessment areas, results summarise as follows: On the west coast, a commercial fyke net fishery on yellow eel was exploiting the stock, until this fishery was completely closed in spring 2012. A fishery-based assessment no longer being achievable, we present trends from research surveys (fyke nets). Insufficient information is currently available to assess the recovery of the stock in absolute terms. Obviously, current fishing mortality is zero (disregarding the currently unquantifiable effect of illegal fishing), but none of the other requested stock indicators (current, current potential and pristine biomass) can be presented. The formerly exploited size-classes of the stock show a recovery in abundance after the closure of the commercial fishery, and the smaller size classes show a break in their decline in line with the recent global trend of glass eel recruitment. In order to support the recovery of the stock, or to compensate for anthropogenic mortality in inland waters, young eel has been restocked on the Swedish west coast since 2010. Noting the quantity of restocking involved, the expected effect (ca. 50 t silver eel) is relatively small, and hard to verify – in comparison to the potential natural stock on the west coast (an order of 1000 t). However, for the currently depleted stock, the contribution will likely constitute a larger share of silver eel escapement. For inland waters, this report updates the 2021 assessment, with substantial changes in methodology being the use of a new natural recruitment model, and the full separation of Trap & Transport catches from the fisheries statistics. The assessment for the inland waters relies on a reconstruction of the stock from information on the youngest eels in our waters (natural recruits, assisted migration, restocking). Based on 78 years of data on natural recruitment into 22 rivers, a statistical model is applied which relates the number of immigrating young eel caught in traps to the location and size of each river, the distance from the trap to the river mouth, and the year in which those eels recruited to continental waters as a glass eel (year class). The further into the Baltic, the larger and less numerous recruits generally are. Distance upstream comes with less numerous recruits. Using the results from the above recruitment analysis, in combination with historical data on assisted migration (young eels transported upstream within a drainage area, across barriers) and restocking (young eels imported into a river system), we have a complete overview of how many young eels recruited to Swedish inland waters. From this, the production of fully grown silver eel is estimated for every lake and year separately, based on best estimates of growth and natural mortality rates. Subtracting the catch made by the fishery (as recorded) and down-sizing for the mortality incurred when passing hydropower stations (percentwise, as recorded or using a default percentage), an estimate of the biomass of silver eel escaping from each river towards the sea is derived. Results indicate, that since 1960, the production of silver eel in inland waters has declined from over 700 to below 300 tonnes per year (t/yr). The production of naturally recruited eels is still falling; following the increase in restocking since 2010, an increase in restocking-based production is expected to be starting right around now. Gradually, restocking has replaced natural recruitment (assisted and fully natural), now making up over 90 % of the inland stock. Fisheries have taken 20-30 % of the silver eel (since the mid-1980s), while the impact of hydropower has ranged from 25 % to 60 %, depending on the year. Escapement is estimated to have varied from 72 t in the late 1990s, to 175 t in the early 2000s. The biomass of current escapement (including eels of restocked origin) is approximately 15 % of the pristine level (incl. restocked), or almost 30 % of the current potential biomass (incl. restocked). This is below the 40 % biomass limit of the Eel Regulation, and anthropogenic mortality (70 % over the entire life span in continental waters) exceeds the limit implied in the Eel Regulation (60 % mortality, the complement of 40 % survival). Mortality being that high, Swedish inland waters currently do not contribute to the recovery of the stock. The temporal variation (in production, impacts and escapement) is partly the consequence of a differential spatial distribution of the restocking of eel over the years. The original natural (not assisted) recruits were far less impacted by hydropower, since they could not climb the hydropower dams when immigrating. Since 2010, inland restocking is increasingly concentrated to drainage areas falling to the Kattegat-Skagerrak, also including obstructed lakes (primarily Lake Vänern, and many smaller ones). Even though Trap & Transport of silver eel - from above barriers towards the sea - has contributed to reducing the hydropower impact, hydropower mortality remains the largest estimated contributor to silver eel mortality in inland waters. Without restocking, the biomass affected by fishery and/or hydropower would be only 5-10 % of the currently impacted biomass, but the stock abundance would reduce from 15 % to less than 3 % of the pristine biomass. In summary: the inland eel stock biomass is below the minimum target, anthropogenic impacts exceed the minimum limit that would allow recovery, and those impacts have been increasing. It is therefore recommended to reconsider the current action plans on inland waters, taking into account the results of the current, comprehensive assessment. For the Baltic coast, the 2021 assessment has been updated without major changes in methodology. Results indicate that the impact of the fishery continues to decline over the decades. The current impact of the Swedish silver eel fishery on the escapement of silver eel along the Baltic Sea coast is estimated at 0.3 %. However, this fishery is just one of the anthropogenic impacts (in other areas/countries) affecting the eel stock in the Baltic, including all types of impacts, on all life stages and all habitats anywhere in the Baltic. Integration with the assessments in other countries has not been achieved. Current estimates of the abundance of silver eel (biomass) indicates an order of several thousand tonnes, but those estimates are extremely uncertain, due to the low impact of the fishery (near-zero statistics). Moreover, these do not take into account the origin of those silver eels, from other countries. An integrated assessment for the whole Baltic will be required to ground-truth these estimates. This would also bring the eel assessments in line with the policy to regionalise stock assessments for other (commercial) fish species (see https://ec.europa.eu/oceans-and-fisheries/fisheries/rules/multiannual-plans_en). It is recommended to develop an integrated assessment for the entire Baltic Sea eel stock, and to coordinate protective measures with other range states.

Original Objectives and revisions: (1) "To determine the biosynthetic pathway of JHB3 in the adult C. capitata CA in order to establish parameters for the future choice and synthesis of suitable inhibitors". Modified: to determine the pattern of FR-7 biosynthesis during normal reproductive maturation, and identify enzymes potentially involved in its synthesis. (2) "To correlate allatal epoxidase activity to the biosynthesis of JHB3 at different stages of reproductive maturation/vitellogenesis and evaluate the hypothesis that a specific JH-epoxidase may be rate limiting". Modified: to study the effects of epoxidase inhibitors on the pattern of allatal JH biosynthesis in vitro and on female reproduction in vive. (3) "To probe and clone the gene homologous to ap from C. capitata, determine its exon-intron organization, sequence it and demonstrate its spatial and temporal expression in larvae, pupae and adults." The "Medfly" (Ceratitis capitata) is a serious polyphagous fruit pest, widely distributed in subtropical regions. Damage is caused by oviposition and subsequent development of larvae. JH's are dominant gonadotropic factors in insects. In the higher Diptera, to which the Medfly belongs, JHB3 is a major homolog. It comprises 95% of the total JH produced in vitro in D. melanogaster, with JH-III found as a minor component. The biosynthesis of both JH-III and JHB3 is dependent on epoxidation of double bonds in the JH molecule. The specificity of such epoxidases is unknown. The male accessory gland D. melanogaster produces a Sex Peptide, transferred to the female during copulation. SP reduces female receptivity while activating specific JH biosynthesis in vitro and inducing oviposition in vive. It also reduces pheromone production and activates CA of the moth Helicoverpa armigera. In a previous study, mutants of the apterous (ap) gene of D. melanogaster were analyzed. This gene induces previteilogenic arrest which can be rescued by external application of JH. Considerable progress has been made in recombinant DNA technology of the Medfly. When fully operative, it might be possible to effectively transfer D. melanogaster endocrine gene-lesions into the Medfly as a strategy for their genetic control. A marked heterogeneity in the pattern of JH homologs produced by Medfly CA was observed. Contrary to the anticipated biosynthesis of JHB;, significant amounts of an unknown JH-like compound, of unknown structure and provisionally termed FR-7, were produced, in addition to significant amounts of JH-III and JHB3. Inhibitors of monooxygenases, devised for their effects on ecdysteroid biosynthesis, affect Medfly JH biosynthesis but do not reduce egg deposition. FR-7 was isolated from incubation media of Medfly CA and examined by various MS procedures, but its structure is not yet resolved. MS analysis is being done in collaboration with Professor R.R.W. Rickards of the Australian National University in Canberra, Australia. A homologue of the ap gene of D. melanogaster exists in the Medfly. LIM domains and the homeo-domain, important for the function of the D. melanogaster ap gene, are conserved here too. Attempts to clone the complete gene were unsuccessful. Due to the complexity of JH homologs, presence of related FR-7 in the biosynthetic products of Medfly CA and lack of reduction in eggs deposited in the presence of monooxygenase inhibitors, inhibition of epoxidases is not a feasible alternative to control Medfly reproduction, and raises questions which cannot be resolved within the current dogma of hormonal control of reproduction in Diptera. The Medfly ap gene has similar domains to the D. melanogaster ap gene. Although mutant ap genes are involved in JH deficiency, ap is a questionable candidate for an endocrine lesion, especially since the D. melanogoster gene functions is a transcription factor.
