Three-dimensional models of the body parts were developed using the basic 3D modeling operations (rotation, extrusion, threading, etc.) of the T-FLEX CAD 3D CAD system, which made it possible to significantly simplify the work with assembly 3D models representing a complex structure consisting of a large number of parts. During the trial operation, the advantages of using the T-FLEX CAD 3D CAD system have been proven when working with complex 3D models. The advantages of using the program in the machine-building cluster are shown.

The article presents the experience of using the Russian computer-aided design program T-FLEX CAD to study 3D modeling and prototyping in pre-professional engineering classes as a course of choice for students. The advantages and limitations of the educational version of T-FLEX CAD are considered, a brief overview of the tasks for which the professional T-FLEX CAD program can be used is given.A practical work for beginners is proposed, in which the creation of a manual citrus juicer is described step by step. At the end of the practical work, tasks for independent work are provided, during which students receive their own simulation result, which can be used to create a prototype of the product.The material will be useful for teachers of informatics and technology working in engineering or information and technology classes, when teaching an advanced course in informatics or a course in 3D modeling and prototyping.

Object of research. The article presents a variant of improving the design of the working body of the tunneling shield, namely, its cutting part. Research aims to confirm the performance of the proposed design using the method of solid 3D modeling in T-Flex CAD. Methodology. The working body of the tunneling shield has been improved by making changes to the design of the working surface of the rotor. Theoretical studies have been carried out to determine the strength characteristics of the proposed rotor design. The proposed rotor model was visualized using the T-Flex CAD software product. The advantages of the solid-state method of 3D modeling are that the mathematical model is endowed with the real physical properties of the object (weight, volume, material, surface area, etc.), and the technology of rapid prototyping allows you to get layouts of photorealistic images in the shortest possible time. Results and discussion. Based on the results of the data obtained in the course of theoretical studies, a comparative analysis of the strength characteristics of the basic and modernized models of the tunneling shield cutting working body was carried out. The studies carried out in T-Flex CAD have confirmed the functionality of the modernized design of the tunneling shield working body. An algorithm for calculating the strength characteristics of the proposed rotor design is presented. The algorithm is universal and can be used to calculate other designs of the tunneling shield working body. Conclusions. As a result of the research carried out, the strength characteristics of the rotor have been determined. Maximum values: displacement modulus – 2.875 · 10–5 m, equivalent stresses – 29.47 MPa, and safety factor for an equivalent stress is 9.446 · 106.

The article presents a variant for improving the design of a working body of a tunneling shield, its cutting part to be more precise. The working body of the tunneling shield has been improved with the changes in the design of a rotor working surface. The theoretical studies aimed at determining the strength characteristics of the proposed rotor design have been carried out. The proposed rotor model has been visualized with the use of the T-Flex CAD software. On the basis of the results obtained in the course of the theoretical research, a comparative analysis of the strength characteristics of the main and improved model of the cutting working body of the tunneling shield have been made. The studies carried out at T-Flex CAD confirmed the working capacity of the improved design of the tunneling shield working body. The strength of the rotor has been determined as a result of the studies carried out. The maximum values of the displacement module is 2.875E-05 m, the equivalent stresses are 29.47 MPa, a reserve coefficient on equivalent stresses is 9.446E 06.

In this work, the calculation of Maag gear shaper cutter parameters is performed for spur gears with helical teeth in three variants – straight-tooth tool with machine offset, helical-tooth tool without machine offset and helical-tooth tool with machine offset. It is therefore a prerequisite that the manufactured involute gearing has helical teeth for each variant. The created CAD program is universal and can be used for construction in other combinations as well. The tool clamping angles on the tool holder, the cutting geometry of the cutter and the characteristic of the gearing are introduced into the calculation. The output of the work will be then calculated individual parameters of the Maag shaping cutter in the tooling system, necessary for its construction. The calculation are performed in program T-Flex CAD and the summary output is graphical 2D/3D representation of the rack-shaped cutter in its base, normal and side planes, always in a different design based on the change of the tool input data and gearing characteristic. The analysis of the tool´s involute profile was solved by vector calculus and matrices and by rotating the tool in the chosen coordinate systems. The specific calculation of the tool parameters will be solved using goniometric functions.

Изменение геометрической формы абразивного инструмента в результате износа рабочей поверхности (РП) абразивного инструмента и затупления вершин режущих зерен приводит к существенным изменениям статистических показателей их распределения в объеме рабочего слоя шлифовального круга. Исследование процесса взаимодействия абразивного инструмента с заготовкой и определение параметров процесса с учетом характеристик и текущих состояний рабочей поверхности инструмента позволили разработать расчетные методики для определения оптимальной прочности закрепления абразивных зерен на рабочей поверхности шлифовального круга и силовых показателей процесса шлифования. Описаны методика исследования и приведены результаты, содержащие данные о параметрах взаимодействия вершин активных абразивных зерен шлифовального круга с заготовкой в зоне их контакта. Исследования проводились с использованием разработанных компьютерных геометрических и статистико-вероятностных моделей рабочей поверхности инструмента и процесса обработки, созданных с использованием инженерной 3D САПР T-Flex CAD. Применение компьютерных моделей позволило определить форму и размеры сечения срезаемых стружек с учетом параметров режима шлифования и характеристик инструмента. Применение предложенной методики, разработанной при использовании базовых математических моделей, позволило определить дискретные и интегральные показатели микрорезания процесса абразивной обработки. В ходе исследований в расчетах использован комплексный параметр γ, характеризующий удельную производительность единицы площади рабочей поверхности инструмента. Проведенный регрессионный анализ в программном пакете STATISTICA10 с целью оценки влияния изменения исследуемых факторов на показатели взаимодействия инструмента с заготовкой позволил получить зависимости плотности вершин активных зерен в зоне контакта r и средних сил резания зернами Ррt от комплексного коэффициента g.

The article is devoted to the methods which help to achieve precision of manufacturing long parts by means of mathematical modeling and dimensional analysis. The possibility of three-dimensional modeling of the shaft of the beater combine harvester “Vector” is considered and the model of turning of a long shaft is presented. The solution of problems arising in the manufacture of long shafts is possible by selecting the best way to install the work piece on the machine and calculate the optimal allowances of the work piece. Method of ad-justing the milling machine, using a special complex is introduced. For the analysis of technological process we will use the method of graph theory. Graph functional relationships established technological dimensional chains and is made up of the original equation. The solution of these equations allows us to find the optimal operating allowances and dimensions in the axial direction. To simplify and improve the calculations, we use the T-Flex CAD 3D complex module, translating the obtained formulas into a parametric table and linking the variables with the 3D-model. Thus, we obtain a simulation dynamic model of dimensional connections of the technological process of manufacturing the shaft. Simulation of dimensional connections of the shaft manufacturing process in the radial direction also determined the operating allowances and dimensions. The proposed research methods are universal and can be applied to any details of this class.

Os fios de níquel-titânio são uma grande preocupação devido ao alto índice de níquel e sua possível liberação quando expostos principalmente ao flúor. Assim, o objetivo neste estudo foi avaliar a rugosidade de fios de niquel-titânio (NiTi) utilizados no tratamento ortodôntico quando expostos à solução fluoretada. Os fios foram divididos em grupos de acordo com sua composição (Flexy NiTi Super Elástico, Flexy NiTi Thermal 35°, Flexy NiTi Copper e Flexy Blue-Ti), tempo (0 dias – T0, 7 dias – T1, 14 dias – T2, e 28 dias – T3) e imersão (solução de saliva artificial ou saliva artificial mais solução fluoretada). Para o ensaio de rugosidade foram utilizadas seções retas (24 mm de comprimento) de cada fio, com velocidade constante de 0,25 mm/s, comprimento de 2,5 mm e cut-off de 0,25 mm. A rugosidade da superfície para cada fio ortodôntico foi composta pela média de três leituras. Os resultados foram submetidos ao teste de normalidade Kolmogorov-Smirnov, seguidos de análise de variância para comparação entre os diferentes tempos e teste t de Student para comparação da diferença dos tempos para cada grupo e na comparação entre os grupos. Não houve diferença estatisticamente significante da rugosidade intra e intergrupos entre diferentes composições, tempos ou imersão. O uso de flúor na concentração de 0,2% uma vez por semana não causou alterações significativas na rugosidade da superfície dos fios utilizados.

OBJETIVO: Avaliar os efeitos da eletroestimulação por aparelhos de uso doméstico sobre o condicionamento neuromuscular. MÉTODOS: A amostra foi composta por 20 mulheres voluntárias, sedentárias, destras, com idades entre 18 a 25 anos em Maceió, estado de Alagoas, em 2006. As mulheres foram divididas aleatoriamente em dois grupos: as do grupo A foram submetidas a eletroestimulação passiva com aparelhos comerciais e as do grupo B, a exercício físico com resistência. O programa de treinamento dos grupos totalizou 16 sessões em dois meses, com duas sessões semanais. As comparações do peso corporal, da cirtometria, fleximetria, e da força muscular antes e após os exercícios, foram utilizadas utilizando-se o teste T pareado. Nas comparações entre os grupos A e B, foi utilizado o teste t de Student. O nível de significância adotado foi de 5%. RESULTADOS: A comparação da força muscular medida de forma subjetiva antes e após cada um dos procedimentos, mostrou que ocorreu aumento da força em ambos os grupos. Foram observados aumentos significantes na massa e na força muscular apenas nos indivíduos que realizaram exercício voluntário. O exercício físico resistido de flexo-extensão dos joelhos foi efetivo em aumentar massa e força muscular, ao contrário das sessões de eletroestimulação com correntes de freqüência de pulsos de 87 Hz, que não tiveram o mesmo efeito. CONCLUSÕES: Os resultados encontrados mostraram que os aparelhos de eletroestimulação para ganho passivo de condicionamento físico comercializados são menos eficientes do que a prática de exercício físico voluntário.

Abstract Abstract 1921 Poster Board I-944 Background: Janus kinase 1 (JAK1) plays a critical role in lymphocyte proliferation and differentiation. Somatic JAK1 mutations are found in 18% of adult precursor T acute lymphoblastic leukemias (T-ALL). Some of the mutations were shown to induce the phosphorylation of JAK1 and STAT5 and lead to cytokine-independent proliferation. These data suggest that dysregulation of JAK1 can be involved in the development or progression of T-ALL (Flex et al. J Exp Med. 2008;205:751-758). Adult T-cell leukemia/lymphoma (ATLL) is a type of T-cell neoplasm, and the activation of JAK/STAT is sometimes observed in the tumor cells. Therefore, we investigated JAK1 mutations in ATLL patients. Patients and methods: Twenty Japanese ATLL patients whose percentage of peripheral abnormal lymphocytes was greater than 30% total cell count were sequentially enrolled into the study from 2000 to 2007. Diagnosis of ATLL was made on the basis of clinical features and laboratory characteristics. All cases tested positive for the serum anti-HTLV-1 antibody. The diagnosis was confirmed by observing monoclonal insertion of the HTLV-1 viral genome into leukemia cells by Southern blot hybridization. Peripheral blood mononuclear cells (PBMCs) were isolated and cryopreserved at -80°C. These PBMCs were thawed and genomic DNA was isolated using standard protocol. The entire coding sequence of the JAK1 gene (exons 2 through 25) was amplified by the polymerase chain reaction (PCR) method. The sequence of PCR primers were kindly provided by Dr. Marco Tartaglia (Istituto Superiore di Sanità, Roma, PhD). The nucleotide sequences were determined by fluorescent dye chemistry sequencing and analyzed by sequencing analysis software. By referencing the assembled sequence in the Ensembl genome database, the presence of homozygous mutations was first checked and then candidates for heterozygous mutations or single nucleotide polypeptides (SNPs) on each allele were screened by comparing the ratio of different bases calculated with the height of the peaks seen from sequencing to the reference genome when the ratio was between 0.15 and 1.0. Result: The percentage of abnormal lymphocytes ranged from 30-90%, and the mean value was 55.4%. The mean value of WBC and lymphocyte number was 40.5×109/L and 33.4×109/L, respectively. The mean value of LDH, Ca2+ or sIL-2R was 609 IU/L, 11.4 mg/dL, or 54748 U/mL, respectively. According to Shimoyama criteria (Shimoyama et al. Br J Haematol. 1991;79:428-437), 19 cases were diagnosed as acute-type ATLL, and one case was diagnosed as chronic-type ATLL. The surface markers of all but one abnormal PBMC were CD3+CD4+CD8-CD25+. In that one exception, loss of CD4 expression was observed. We examined the entire coding sequence of the JAK1 gene in 20 ATLL patients and identified no nonsynonymous or nonsense mutations and five types of silent substitutions in 12 cases. All silent substitutions were synonymous SNPs, as determined from referencing the base sequence in the Ensembl genome database. In the ATLL patients examined, the genotype frequency (%) is c546-AA/AG/GG, 97.5/2.5/0; c1590-CC/CT/TT, 97.5/2.5/0; c2049-CC/CT/TT, 50/50/0; c2097-CC/CG/GG, 95/5/0; c2199-AA/AG/GG, 60/40/0. There is no statistical difference in genotype frequency pattern of these SNPs, between the Japanese ATLL patients examined and the general Asian population on the Ensembl database. Conclusion: Mutations in the coding region of JAK1 do not associate with either activation of the JAK/STAT pathway or leukemogenesis in ATLL. We only examined the coding region of JAK1, and the regulatory region of JAK1 remains to be investigated. Further investigation including downstream signaling molecules and inhibitory molecules in the JAK/STAT signaling pathway is necessary to clarify the mechanism contributing to the leukemogenesis of ATLL. The aim of the work is to create a computer program in the 3D CAD system T Flex CAD allowing the creation of parametric models for the design and construction of a modular hob cutter for the selected gearing. The thesis includes the analysis of the problem of hob cutters for the production of spur gearing with an involute profile, the analysis of the initial surface and the shape of the tool cutting edge for the involute surface, the determination of the basic profile of the cutting edge of the hob cutter (geometry of the basic worm, geometry of the cutting edge) and the calculation of the design parameters of the tool. The individual calculations will be programmed in the 3D CAD system T Flex CAD, which will allow to generate a specific 3D model of the hob cutter based on the change of input requirements for the production of a given spur gear (module, engagement angle, tooth inclination angle), with the possibility of displaying the required tool in a 2D view in its base, normal and side planes, which will be in the form of a standard technical drawing. This paper presents the preparation (unification calculations, introduction) before the final version of the computer program for the creation of the parametric tool models.

ABSTRACT Matrix Gla protein (MGP) is an extracellular protein responsible for inhibiting mineralization. MGP inhibits osteoblast mineralization and bone formation by regulating the deposition of bone matrix. However, Mgp–/– mice display an osteopenic phenotype. To explain this contradiction, we investigated the role of MGP in osteoclastogenesis, the other side of bone remodeling. We found that MGP expression is markedly increased by osteoclastic commitment. Osteoclast differentiation and bone resorption are accelerated by MGP depletion while suppressed by MGP overexpression. The in vivo results confirmed its inhibitory role in osteoclastogenesis by the administration of Cre-dependent FLEX-On recombinant MGP-AAV to LysM Cre mice. Furthermore, we found that the expression and nuclear translocation of nuclear factor of activated T cells, cytoplasmic 1 (NFATc1), are under the control of MGP. MGP loss results in elevation of intracellular Ca2+ flux. Vitronectin-induced activation of Src/Rac1 is magnified in the absence of MGP but reduced when MGP is overexpressed. Inhibition of Src activation or NFATc1 nuclear import rescues the increased osteoclastogenesis induced by MGP deficiency. These observations (i) establish, for the first time to our knowledge, that MGP plays an essential role in osteoclast differentiation and function, (ii) enrich the current knowledge of MGP function, and (iii) indicate the potential of MGP as a therapeutic target for low-bone-mass disorders.
