The aim of the thesis was to determine if the substrate treatment with temperature lower than 100°C could replace the sterilization at 120°C. Another task was to compare different types of substrates and supplements.
For the substrate treatment were selected temperatures of 90°C for 48 hours, 90°C for 20 hours, 121°C for 2 hours, 80°C for 20 hours and 70°C for 24 hours. The treatment with temperature lower than 100°C could replace the sterilization at 120°C.
In the experiments were used substrates compound of beech sawdust or straw pellets supplemented by wheat bran or wheat groats. The research was also focused to influence of substrate fermentation on growth characteristics of Hericium erinaceus. The most suitable substrate for mycelium growth and subsequent fructification of Hericium erinaceus were beech sawdust. Faster mycelial growth and higher yield of fruiting body were achieved in the case of supplemented substrate of sawdust beech by wheat bran or wheat groats. There were observed no effects of fermentation substrates on mycelium growth of Hericium erinaceus.
Another part of the research was dedicated to mycelium growth and fructification of Hericium coralloides. The best substrate for Hericium coralloides is beech sawdust without supplements.

The waste from agricultural and food industry are accessible in large quantity anywhere in the whole world nowadays. Most of these wastes include cellulose (30 - 40 %), hemicellulose (20 - 40 %) and lignine (10 - 20 %). Therefore these waste materials have wide use as the substrates for the microbial growth and the production of the enzymes. The microorganisms are able to use organic compounds from the wastes as the source of energy for the growth and carbon for synthesis of cellular biomass [24]. Wheat and rice straw are possible to use as the substrates for cultivation of the microorganisms and following production of the enzymes. In this thesis the utilization of the wastes from food industry for the production of the enzymes by the microorganisms was studied. We observed utilization of wheat straw as source of energy for growth of tested microorganisms and investigated their ability for the production of oxidoreductase (laccase). The optimalization of growth conditions of Aureobasidium pullulans was proceeded. Further the activity of laccase was studied. Milled wheat straw was used as the substrate. The cultivation was done in the thermoregulator at the temperature of 27°C. The activity of laccase was not found in this thesis. Petri dishes were contaminated by three unknown microoganisms during optimalization of growth of Aureobasidium pullulans. One of them produced laccase in cultivation with straw.

This master’s thesis applies utilization of alternative raw materials in thermal insulation system of the ETICS of new generation for houses. The theoretical part dealt with the explanation of the ETICS and the selection of suitable raw material for his innovating. In the following practical part were used selected materials to create a new thermal insulation made of straw and binding materials of cement, casein glue, bicomponent fibers and mushroom mycelium. At the end have been applied the variants of base coat of clay and mineral screed on new thermal insulation, from which was selected the optimal variant. From optimal thermal insulation and suitable base coat was conducted proposal of final version of new ETICS.

This diploma thesis is focused on a comparison of construction cost of the house made of classic materials with the equivalent made of natural building materials and consequently on comparison of both construction variants. In order to perform those tasks, the used natural building materials were defined. Furthermore, the specific options of usage of these natural building materials as well as the used composition structures were introduced. The thesis concludes with a price calculation of the constructions made of natural building materials which were not available in the common budgetary software. The appendix of the thesis contains budgets for the first variant, where the house is made of natural building materials, and also for the second variant, where the house is made of classic materials.

This diploma thesis deals with the topic of setting the poems of Josef Václav Sládek to music. The main source of the analyzed data is a bibliographic list of compositions, which was compiled after my own research in the databases and catalogues of the National Library of Czech Republic, the Municipal Library in Prague and Czech Museum of Music, as well as in specialised literature. The first chapter briefly summarizes the general principles of setting poetry to music and then it focuses on particular aspects of Sládek's poetics, which encourage composers to set his poems to music. The first chapter also outlines Sládek's poetics in different poetry collections. The second chapter gives a detailed analysis of the compiled bibliographic list of compositions in terms of the frequency in which particular poems have been set to music, the number of works composed by different authors, vocal and instrumental cast, and also time contexts. Sládek's American experience influenced all fields of his work. This thesi s analyzed Sládek a s a publisher, editor and translator, i t also focused on hi s correspondence and essays. Sládek admired America and strived for national confidence in our country a s well. The enthusiasm about America was j oined by Sládek' s de sire for hi s homeland 1 for freedom in Czech lands; generally1 for the improving of Czech situation. In his correspondence 1 Sládek wrote more about his physical problems and pains and he mentioned more negatives of American life. We can find there also more Sládek's own negative opinions. As far as the essays are concerned1 I focused on two of them: "Ústava Spojených obcí severoamerických" ("The Constitution of U ni ted States of America") and "První století severoamerické republiky" ("The First Century of the North-American Republic"). Also, I looked at Sládek's activity in Lumír (a Czech magazíne of 19th century) 1 where I was trying to find which translations Sládek published there. Finally1 Sládek as a translator is very sensitive to the relationships between poetical form and content. Though 1 we can say that his tendency is to nivelize the intensity of romantic dramatic expres s ion. On the other side 1 hi s translations of poems that resemble songs are perfect.

Based on analysis of characters we try to show how Pavel Brycz portrays various aspects of identity (national, personal, social, professional, gender). Structurally, the thesis is divided into two parts: the first one is devoted to the basic characteristics of identity and represents the generalization of various concepts of identity from the point of view of philosophy, psychology, sociology and literary theory; the second part deals with the analysis of the characters of Brycz's novel The Long Lost Glory of the Patriarchy, which shows that each character carries a certain distinctive identity.

Smrt Zikmunda Lucemburského symbolicky uzavřela jednu etapu českých dějin. Husitská revoluce přinesla do života společnosti mnohé změny a byla odrazovým můstkem ke stavovskému státu. Téměř dvě a půl století jsme sledovali osudy rodu erbu ostrve, Ronovců, kteří se stejně jako ostatní šlechtické rody snažili vybojovat své místo na slunci. Z původních severočeských sídel se Ronovci rozšířili skoro do všech regionů Čech a Moravy, když ne přímo získáním držav, tak alespoň sňatkovou politikou.K tomu je nutno připočítat držbu lén, nejčastěji v Horní Lužici nebo v Kladsku. Snad žádný český ani moravský rod se nemohl vykázat takovou migrací. Ne všechny ronovské odnože Lucemburky přežily. Již v roce 1351 vymřeli po meči Žlebští z Lichtenburka, v roce 1410 Pyknové z Lichtenburka. Odnož Škopků z Dubé vyhasla přibližně ve stejnou dobu jako lucemburská dynastie a mnoho času už nezbývalo ani pánům z Pirkštejna. Slavný Hynek Ptáček z Pirkštejna zemřel bez mužských dědiců v roce 1444. Ostatní větve prokázaly větší životaschopnost, Krušinové a Bítovští z Lichtenburka žili ještě v 2.polovině 16.století, Berkové z Dubé a páni z Lipé dokonce ještě o půl století déle.

The aim of the diploma thesis was to determine heat loss and energy demand of a particular family house in a village. The thesis is focused on finding technical measures to reduce the heat loss. The measures are assessed from the perspective of reconstruction of the house (additional insulation, replacing of windows) and from the perspective of technical equipment of the house (change of a heat source). According to the amount of a fuel per one heating season there were determined areas of agricultural and forest land needed to ensure the heating energy self-sufficiency. As a fuel, there was proposed firewood and straw pellets and it was compared with the alternative fuel, natural gas. Using of fuels from agricultural and forest lands is a suitable way to ensure the energy self-sufficiency of the house.

Many agricultural biogas stations process predominantly phytomass specifically grown for power engineering purposes as a primary raw material. This results in gradual increase of maize proportion in sowing sequences at the exclusion of traditional area-specific crops, which presents various potential negative effects. The aim of study is to evaluate the substitution of maize with wheat straw in the anaerobic fermentation process from the biotechnological and economic viewpoint. For this purpose, wheat straw pellets were pretreated using the high-pressure extruder in compliance with UV CZ 21314 at different runtime parameters. Cumulative methane (CH4) production in quarter-mode operation simulation, based on the extrusion pressure, cycle duration and fermentation temperature, was the main evaluation criteria. Optimal run time parameters were derived from the approximation ? extrusion pressure 1,35 MPa, reaction time 210 s, in order to achieve maximal CH4 production at mesophilic fermentation temperature (38 °C). For fermentation temperature 48 °C, optimal pressure 1,25 MPa and cycle duration 180 s were approximated. The discount rate of Net Present Value (NPV) was established as a main comparative criterion considering overall executive, legislative and technological aspects of analyzed biological effects. Based on these findings suggest, wheat straw extrusion is interesting, unconventional and profitable technology in the field of biogas production.

In this thesis, "The Evaluation of the Quality of Hops from Traditional and Non-traditional Growing Areas", the quality of three varieties of hop seed cones (Saaz, Sladek and Kazbek) from a non-traditional hop-growing region was analyzed and the data used for comparison. The primary aim of the study was to determine if there is a possibility of growing quality hops in a region that is not considered a hop-growing region. The hops that were tested were harvested in 2015 from a private hopfield located in Obora near Tábor. This hopfield is owned and managed by Ing. Martin Novák. Mechanical and chemical analysis was carried out on samples from Obora. The main goal of the analysis was to determine the alpha and beta acid content of the hop seed cones harvested there. Samples were analyzed with EBC 7.7 (HPLC). Alpha acid content of the tested samples in Saaz hops was 4.06 % w/w, in Sladek 7.14 % w/w and in Kazbek 4.67 % w/w. The content of beta acids in Saaz hops was 3.16 % w/w, in Sladek 3.93 % w/w and in Kazbek 4.51 % w/w. The collected data was then compared to published data for hops grown in traditional hop-growing regions. Giuseppe Tartini is one of the most important personalities in the history of violin playing and literature. His rich heritage has contributed to the development of both, and his influence has crossed afar the Italian borders. This diploma thesis brings a complex view into the life and creation of this 18th century violin virtuoso. His composing activity uprose from his master violin skills, and therefore he composed especially for this instrument. Tartini transmitted his knowledge to the students of paduan School of nations, which he himself had founded. His treatise on ornaments Traité des agréments, compiled of the practical comments on the ornament interpretation, is of great significance. No small attention deserves also his letter to the pupil Maddalena Lombardini. As a matter of fact, it is a violin lesson in a letter form, and the value of the letter is by no means just historical. Another area of Tartini's interest was music theory, in which he invented so called difference tones. His contribution to the development of violin bow is also praiseworthy; he made several adaptations which brought the bow nearer to its current shape.

The master thesis Development of czech extreme right-wing after 1989 deals with three the most significant far right-wing parties in Czech republic after 1989. These parties are Association for republic - Czechoslovak Republican Party (SPR-RSČ), National Party (NS) and Workers' Party (DS). The only far right-wing body shall, which was able to enforce on Czech political scene and obtain representation in the Parliament was SPR-RSČ. Other two parties were unsuccessful in elections. Thesis has an ambition to make clear, why SPR-RSČ was that successful and other parties were not. But at the beginning of the thesis is explained concept of far right-wing. After this, is a chapter about SPR-RSČ, which was significant political party in the 90's. Nevertheless after unsuccessful elections in 1998 became piece by piece minor political force. Next part refers about NS, which was very unsuccessful in elections, but was able to attract attention of Czech media by its actions. Thesis also include chapter about DS, which was, as the only one after 1989, dissolved by the Supreme Administrative Court. However the party works under the name DSSS and nowadays is the most successful far right-wing party in Czech Republic. The aim of this diploma thesis is to point out how personal experience and memories on the drug violence that was common in Colombia during 80's - 90's reflect in the novelistic works of a few contemporary Colombian writers. The thesis is based on the following five novels: Noticia de un secuestro (Gabriel García Márquez), El olvido que seremos (Hector Abad Faciolince), El ruido de las cosas al caer, Reputaciones (Juan Gabriel Vásquez) a La luz difícil (Tomás González). Aforementioned books were published after the year 2000 (Noticia de un secuestro makes an exception), thus they come from the writers that represent same generation. Almost all of the analyzed books are connected with the topic of terrorism of drug mafia and they take interest in the life of ordinary people. By analyzing novels, the thesis tries to point out also another topics, such as death or power of the fame and public opinion in the country. In each novel are analyzed also autobiographical features, language style and the process of the creation of the text. The last chapter contains memories and opinions of ten Colombian respondents which we can compare with the memories of the writers.
