The SGSN-MME node, which is important in wireless networks, handles many users and therefore the uptime requirements for it are very high. The goal at Ericsson is to reach 99.9999% uptime for their SGSN-MME nodes and to reach this a lot of testing is required. Therefore the test process during the SGSN-MME development is both resource expensive and time consuming. To optimize both resource utilization and test runtimes a common test node pool solution for their different test tools has been proposed. During this thesis a first exploratory investigation about how to optimize such a solution was made. During the investigation different aspects were evaluated and a first input about how an optimal solution can be implemented is proposed. By having a scheduling layer in the common node pool, which determines how many nodes each regression job will get, depending on current load, the number of test cases in the job and the current node utilization optimized solutions can be found. Future work in the area is still needed, but the exploratory research made during this thesis will give a good base to continue from.

The goal of this Thesis project is to implement a prototype  for Serving GPRS Support Node (SGSN) which is the central part of the General packet radio service (GPRS). SGSN is responsible for the transfer of data packets between the mobile stations covered in its geographical service area. The SGSN should be integrated into existing Mobile Arts products, open source projects and nanoBTS radio hardware to construct a minimal GPRS network that support sending and receiving Short Message Service (SMS). The prototype  is tested with radio hardware and mobile phones.

Dividing cells are constantly under threat from both endogenous and exogenous DNA damaging stresses that can lead to mutations and structural variations in DNA. One contributor to genome instability is three-stranded DNA:RNA hybrid structures called R-loops. Though R-loops are known to induce DNA damage and DNA replication stress, it is unclear whether they are recognized and processed by an established DNA repair pathway prior to inducing DNA breaks. Canonically, DNA repair proteins work downstream of R-loop-induced DNA damage to stimulate repair and suppress genome instability. Recently, the possibility that some DNA repair pathways actively destabilize R-loops, thus preventing unscheduled DNA damage has emerged. Here we identify the helicase SGS1 as a suppressor of R-loop stability. Our data reveals that SGS1 depleted cells accumulate R-loops. In addition, we define a role for transcription in genome instability of cells lacking SGS1, which is consistent with an R-loop based mechanism. Hyper-recombination in SGS1 mutants is dependent on transcript length, transcription rate, and active DNA replication. Also, rDNA instability in sgs1Δ can be suppressed by ectopic expression of RNaseH1, a protein that degrades DNA:RNA hybrids. Interestingly, R-loops are known to form at rDNA loci. We favour a model in which SGS1 contributes to the stabilization of stalled replication forks associated with transcription complexes, and unresolved DNA:RNA hybrids. Finally, we showed that knockdown of the human Sgs1 orthologue BLM in HCT116 cells also led to the accumulation of more R-loops than control HCT116 cells. In summary, our data supports the idea that some DNA repair proteins involved in replication fork stabilization might also prevent and process R-loops.

Genome instability is a hallmark of human cancers. Patients with Bloom’s syndrome, a rare chromosome breakage syndrome caused by inactivation of the RecQ helicase BLM, result in phenotypes associated with accelerated aging and develop cancer at a very young age. Patients with Bloom’s syndrome exhibit hyper-recombination, but the role of BLM and increased genomic instability is not fully characterized. Sgs1, the only member of the RecQ family of DNA helicases in Saccharomyces cerevisiae, is known to act both in early and late stages of homology-dependent repair of DNA damage. Exo1, a 5'–3' exonuclease, first discovered to play a role in mismatch repair has been shown to participate in parallel to Sgs1 in processing the ends of DNA double-strand breaks, an early step of homology-mediated repair. Here we have characterized the genetic interaction of SGS1 and EXO1 with other repair factors in homology-mediated repair as well as DNA damage checkpoints, and characterize the role of post-translational modifications, and protein-protein interactions in regulating their function in response to DNA damage. In S. cerevisiae cells lacking Sgs1, spontaneous translocations arise by homologous recombination in small regions of homology between three non-allelic, but related sequences in the genes CAN1, LYP1, and ALP1. We have found that these translocation events are inhibited if cells lack Mec1/ATR kinase while Tel1/ATM acts as a suppressor, and that they are dependent on Rad59, a protein known to function as one of two sub-pathways of Rad52 homology-directed repair.

Through a candidate screen of other DNA metabolic factors, we identified Exo1 as a strong suppressor of chromosomal rearrangements in the sgs1Δ mutant. The Exo1 enzymatic domain is located in the N-terminus while the C-terminus harbors mismatch repair protein binding sites as well as phosphorylation sites known to modulate its enzymatic function at uncapped telomeres. We have determined that the C-terminus is dispensable for Exo1’s roles in resistance to DNA-damaging agents and suppressing mutations and chromosomal rearrangements. Exo1 has been identified as a component of the error-free DNA damage tolerance pathway of template switching. Exo1 promotes template switching by extending the single strand gap behind stalled replication forks. Here, we show that the dysregulation of the phosphorylation of the C-terminus of Exo1 is detrimental in cells under replication stress whereas loss of Exo1 suppresses under the same conditions, suggesting that Exo1 function is tightly regulated by both phosphorylation and dephosphorylation and is important in properly modulating the DNA damage response at stalled forks.

It has previously been shown that the strand exchange factor Rad51 binds to the C-terminus of Sgs1 although the significance of this physical interaction has yet to be determined. To elucidate the function of the physical interaction of Sgs1 and Rad51, we have generated a separation of function allele of SGS1 with a single amino acid change (sgs1-FD) that ablates the physical interaction with Rad51. Alone, the loss of the interaction of Sgs1 and Rad51 in our sgs1-FD mutant did not cause any of the defects in response to DNA damaging agents or genome rearrangements that are observed in the sgs1 deletion mutant. However, when we assessed the sgs1-FD mutant in combination with the loss of Sae2, Mre11, Exo1, Srs2, Rrm3, and Pol32 we observed genetic interactions that distinguish the sgs1-FD mutant from the sgs1 deletion mutant. Negative and positive genetic interactions with SAE2, MRE11, EXO1, SRS2, RRM3, and POL32 suggest the role of the physical interaction of Sgs1 and Rad51 is in promoting homology-mediated repair possibly by competing with single-strand binding protein RPA for single-stranded DNA to promote Rad51 filament formation.

Together, these studies characterize additional roles for domains of Sgs1 and Exo1 that are not entirely understood as well as their roles in combination with DNA damage checkpoints, and repair pathways that are necessary for maintaining genome stability.

SGS1, a Saccharomyces cerevisiae 3 '-5' DNA helicase, is a homologue of the Escherichia coli RecQ gene. It is essential for genomic stability of both during mitosis and meiosis. The purpose of this thesis is to provide a better understanding of the role of this helicase during meiotic recombination. In meiosis, SGS1 mutant cells display a decrease in sporulation efficiency and spore viability. In addition, the unusual spore viability pattern observed in SGS1 mutants cannot be explained solely by meiosis I or meiosis II missegregations. These problems could be partially explained by defects in mitotic chromosome segregation or problems with meiotic S-phase. Cytological experiments demonstrating an increase in synapsis initiation complexes and axial associations in sgs1Delta could be explained by an early function of Sgs1p in meiosis, such as the unwinding of inappropriate strand invasion events. Consistent with this, we observe increased gene conversion, increased homeologous recombination and increased interaction between sister choromatids.;Recent observations have suggested that, Sgs1p and Top3p in S. cerevisiae, and the human orthologue protein BLM, in conjunction with the Top3alpha protein, can dissolve double Holliday junctions. Physical analyses of double-strand break repair in meiosis, combined the genetic analysis of this work, indicate a late function of the Sgs1 protein in the dissolution of double Holliday junctions. We have shown an unusual class of tetrads in which non-sister spores and recombinant spores are dead. We interpret this as a consequence of the failure to untangle intertwined chromatids. This defect in SGS1 mutant strains could be explained by either the presence of pre-meiotic S-phase catenates, a defect in crossover resolution and/or a defect in the dissolution of closely spaced double Holliday junctions.

A Leishamaniose Visceral (LV) é transmitida pela picada de insetos da espécie vetora Lutzomyialongipalpis através da inoculação dos parasitas juntamente com a saliva na pele do hospedeiro durante a alimentação sanguínea. A saliva deste vetor desempenha um papel importante na obtenção do repasto, sendo também capaz de modular o sistema imune do hospedeiro. Algumas proteínas salivares são imunogênicas e podem induzir a produção de anticorpos específicos. Neste sentido, a presença de anticorpos anti-saliva pode ser utilizada como marcador de exposição ao vetor. Na LV, a presença de galinhas em área endêmica é considerada um fator importante na manutenção da fauna flebotomínica, além de ser citada como um importante fator de risco para a transmissão da doença. Entretanto, o papel da galinha na cadeia epidemiológica da LV ainda não é bem compreendido. Nosso objetivo neste estudo foi detectar nestes animais a presença de anticorpos contra o sonicado de glândula salivar (SGS) a serem utilizados como marcadores de exposição a L. longipalpis e investigar o potencial papel destas aves como sentinela para a presença do vetor em área endêmica. Para isso, quarenta galinhas foram distribuídas em cinco residências em Cavunge, área endêmica para LV na Bahia e naturalmente expostas a picadas de flebotomíneos durante oito meses. Para avaliar a densidade vetorial armadilhas luminosas foram distribuídas e monitoradas mensalmente nas mesmas residências. Amostras de sangue destas aves foram coletadas a cada dois meses e monitorada a soroconversão para SGS. O teste de ELISA foi realizado para avaliar o reconhecimento de SGS onde, após quatro meses do início da exposição 26% das aves se tornaram positivas e com seis meses de exposição detectamos uma soroconversão de 100% das galinhas. Após oito meses, todas apresentavam altos títulos de anticorpos anti-SGS, aumentado 2,7 vezes sobre o valor do cut-off. Além do SGS foram utilizadas as proteínas recombinantes LJM17 e LJM11 da saliva da L. longipalpisque apresentaram desempenho semelhante ao SGS. O teste de correlação aplicado entre a densidade flebotomínica e os níveis de anticorpos específicos após oito meses de exposição não foi positivo, porém pôde-se observar que independente da variação na densidade de vetores, após a soroconversão, galinhas são capazes de manter os níveis de anticorpos IgY anti-SGS constantes. Para avaliar a reatividade cruzada para o reconhecimento a saliva da L. longipalpis, foram utilizados soros de galinhas expostas a picadas de barbeiros e Aedes aegypti, onde identificamos o reconhecimento de duas proteínas da saliva da L. longipalpis com peso de 61 e 79kDa. A proteína recombinante LJM11 não apresentou reatividade cruzada quando testada. Estes resultados demonstram que a exposição natural de galinhas ao vetor L. longipalpis resulta na produção de anticorpos IgY específicos anti-saliva sugerindo o uso destes animais como indicadores da presença do vetor, levantando a possibilidade da utilização de galinhas como um animal sentinela para a detecção da L. longiplapisem áreas endêmicas.
Visceral Leishmaniasis (VL) is transmitted by Lutzomyia longipalpis , when saliva and parasites are injected into the host skin during blood feeding. Saliva has an important role not only in the blood meal process but it is also able to modulate the host’s immune response. Some salivary proteins are immunogenic inducing production of anti-saliva antibodies in the host that can be used as markers of exposure to the vector. In VL, the presence of chicken in the endemic area is indicated as an important source for a blood meal and is often considered as a risk factor for transmission. However the role of the chicken in the VL epidemiology has not been defined. Here we investigate if the detection of antibodies against salivary gland sonicate (SGS) from L. longipalpis could be used as exposure markers to L. longipalpis and the potential role of the chicken as a sentinel animal. Forty chickens were distributed in five houses in Cavunge, a VL endemic area in Bahia, where they were naturally exposed to sand flies during eight months. To evaluate vector density, light traps were distributed and monitored monthly in the same houses. Blood samples were collected every two months to monitor the development of anti-SGS antibodies. ELISA was performed for the recognition of SGS and, after 4 months 26% of the exposed chickens became positive and 6 months later 100% were positive. After 8 months all of them were positive with high antibodies titers against SGS, 2.7x above cut-off value. Beside SGS we also used LJM17 and LJM11 and these recombinants proteins showed similar performance as SGS. There was no correlation between specific anti-saliva antibody levels and sandfly density. Although a cross-reactivity test using sera from chicken exposed to triatomines and Aedes aegypti demonstrated that the proteins of 61 and 79 kDa salivary proteins were only detected in animals exposed to triatomines, LJM11 recombinant protein was shown to be specific to L. longipalpis exposure. Altogether these results show that chicken naturally exposed to the vector develop high titers of anti-saliva antibodies and may be used as important indicators of vector presence, raising the possibility of using chicken as a sentinel animal in endemics areas.

O presente relatório visa descrever as competências desenvolvidas durante o estágio de seis meses na empresa SGS Portugal, no âmbito do Mestrado em Tradução em Serviços Linguísticos. Em primeiro lugar é apresentada uma breve introdução à empresa em questão, e em particular ao departamento onde decorreu o estágio, o Consumer Testing Services. Em segundo lugar inclui-se uma descrição do ambiente de trabalho e da relação com os colegas de departamento com quem houve a oportunidade de trabalhar. Após este ponto foi introduzido um enquadramento teórico e académico do estágio, que visa exemplificar de que forma os conhecimentos teóricos foram úteis durante o estágio e também de que modo este teve importância na inserção no mercado de trabalho, estando também incluída uma reflexão teórica sobre as diversas teorias da tradução, particularmente aquelas que se adequaram à realidade do estágio. Segue-se a apresentação de todas as tarefas pelas quais fui responsável no departamento, com destaque para quatro fases distintas: Gestão de Processos de Tradução, Tradução e Revisão de Manuais Técnicos, Tradução de Conteúdos da Empresa e Outras Competências. Nas duas fases intermédias encontram-se também integrados cinco casos práticos de documentos traduzidos ou revistos, de forma a justificar as técnicas adoptadas em cada uma das situações. Conclui-se com uma breve reflexão sobre a elaboração do mesmo e sobre a importância do Mestrado em Tradução e Serviços Linguísticos para a formação na área. O presente relatório visa descrever as competências desenvolvidas durante o estágio de seis meses na empresa SGS Portugal, no âmbito do Mestrado em Tradução em Serviços Linguísticos. Em primeiro lugar é apresentada uma breve introdução à empresa em questão, e em particular ao departamento onde decorreu o estágio, o Consumer Testing Services. Em segundo lugar inclui-se uma descrição do ambiente de trabalho e da relação com os colegas de departamento com quem houve a oportunidade de trabalhar. Após este ponto foi introduzido um enquadramento teórico e académico do estágio, que visa exemplificar de que forma os conhecimentos teóricos foram úteis durante o estágio e também de que modo este teve importância na inserção no mercado de trabalho, estando também incluída uma reflexão teórica sobre as diversas teorias da tradução, particularmente aquelas que se adequaram à realidade do estágio. Segue-se a apresentação de todas as tarefas pelas quais fui responsável no departamento, com destaque para quatro fases distintas: Gestão de Processos de Tradução, Tradução e Revisão de Manuais Técnicos, Tradução de Conteúdos da Empresa e Outras Competências. Nas duas fases intermédias encontram-se também integrados cinco casos práticos de documentos traduzidos ou revistos, de forma a justificar as técnicas adoptadas em cada uma das situações. Conclui-se com uma breve reflexão sobre a elaboração do mesmo e sobre a importância do Mestrado em Tradução e Serviços Linguísticos para a formação na área. A importância deste documento incide sobre o contributo do mesmo para a compreensão das dificuldades com que um estagiário se depara ao entrar no mercado de trabalho e para avaliar de que forma este aplica os conhecimentos adquiridos ao longo da vida académica.

RecQ-like proteins are a family of DNA helicases that are evolutionary conserved from prokaryotes to eukaryotes. A large amount of experimental evidence suggests these proteins have a major role in the maintenance of genome stability. In humans five RecQ like helicase have been identified (RecQL1, BLM, WRN, RecQL4, and RecQL5), three of which are associated with rare genetic disorders with sever chromosomal and developmental abnormalities, and an elevated predisposition to cancer. Among the disease associated RecQ-like helicases, BLM and WRN have been subject to extensive research, while our collective knowledge about the function of RecQL4 is still very limited. Similarly, little is known about the role of RecQL1 and RecQL5 in maintenance of genome integrity. In the past studies of Sgs1, the Saccharomyces cerevisiae homolog of RecQ, have been very informative regarding BLM function. Thus, here we sought to further investigate BLM, RecQL1, and RecQL4 by using yeast as a model. By constructing humanized yeast strains we evaluated the ability of these genes to complement defects observed in sgs1∆. In Chapter 2, our investigation led to the development of a novel chimeric system, which was able to complement some defects of the sgs1∆ strain. In Chapter 3, by taking advantage of this chimeric system, we evaluated 41 known BLM variants identified in the general human population. This study resulted in identification of six novel variants that completely impaired BLM function and three novel variants that partially impaired BLM function. In Chapter 4 we conducted multiple yeast 2-hybrid screens in search for novel protein-protein interaction for RecQL1 and RecQL4. We have identified two new putatively interacting partners for RecQL1 and three putatively interacting partners for RecQL4. In Chapter 5, functional characterization of RecQL1, BLM, RecQL4 and RecQL5 in yeast suggested genetic interaction between BLM and RecQL4 and RecQL5. Finally, in Chapter 6 a random mutagenesis screen of BLM has led to identification a mutation that impairs BLM function by disrupting the HRDC domain. This mutant suggests that the HRDC domain of BLM has an important role in proper functionality of this helicase.

Homeologous recombination is the genetic exchange that occurs between diverged DNA sequences. This type of recombination can be detrimental to the cell, as it could lead to deletions, duplications and even chromosome loss. Therefore, it must be suppressed in order to maintain the integrity of the genome. The mismatch repair (MMR) complex, along with the 3’-to-5’ helicase Sgs1, has been implicated in this early role in meiosis. We propose a model in which the MMR complex scans the genome searching for single end invasion (SEI) events occurring between diverged sequences. On finding such events, the MMR complex binds to them and impedes their progression. The MMR complex then recruits the Sgs1 helicase which unwinds the heteroduplex DNA, allowing the invading strand to continue its homology search. To test this hypothesis, we investigated whether the interaction between Sgs1 and one member of this MMR complex, Mlh1, affected the ability of Sgs1 to suppress homeologous recombination in the baker’s yeast Saccharomyces cerevisiae. In addition, we investigated which domains of Sgs1 were required for this suppression. The data presented here show that the interaction between Sgs1 and Mlh1 may be important in the suppression of homeologous recombination at the SEI stage. In addition to this, we present data suggesting that the interaction between Sgs1 and the type IA topoisomerase Top3 may also be important in the resolution of recombination intermediates formed between diverged sequences. We suggest that there may still be additional factors that are utilised by the cell in order to maintain the barrier to inter-species recombination. Sgs1 has also been suggested to function at a later stage of meiosis, in the decatenation of Holliday junctions. This process was proposed to be carried out in concert with Top3. We present data that conflict with this hypothesis. We propose that interactions between Sgs1 and the type II topoisomerase Top2 are required in the decatenation of pre-existing replication errors prior to the onset of meiosis. The data implicate Sgs1 in the pre-meiotic replication checkpoint to aid in the repair of errors caused during DNA replication prior to meiosis. We also hypothesise an additional role of Sgs1 in the activation of this pre-meiotic replication checkpoint for the process of sporulation. This investigation therefore emphasises the importance of Sgs1 in the early stages of meiosis.

The RecQ family of helicases has been termed the “Caretakers of the Genome,” and rightfully so. These proteins are highly conserved from bacteria to humans and have been implicated in functions from homologous recombinatorial repair to damage checkpoint response to telomere maintenance and more. Mutant genes of three of the human RecQ helicases lead to syndromes characterized by a high incidence of cancer, premature aging and early death. Despite their implications in several biological functions and importance to the integrity of the human genome and suppression of cancer, many aspects of the RecQ family structure and function remain unknown. To date, much is known about the catalytic function of the helicase domain and accompanying domains, but considerably less is known about the non-catalytic N-terminus in these proteins, which, in many cases, including those human orthologs involved in disease, can make up about half of the total protein length. While experiments have been able to identify protein partners that interact with the N-terminal region, few are able to narrow the binding sites to minimally functional parts and fewer still describe any detail regarding the structural features of these binding areas. In fact, some reviews have generally described the N-terminus as “featureless,” a concept we challenge in our studies. Many of the N-termini of these RecQs have long been known to contain large stretches of acidic residues, a feature of intrinsically disordered regions. These regions/proteins are rich in charged and polar residues, lack compactness that makes crystallography possible, and have flexible and dynamic conformations that are prevalent in “high specificity, low affinity” interactions. Disordered proteins are well-known to be hot spots for protein/protein interactions and post-translational modifications, amongst other functions. Considering these facts, and recognizing the ties between these and what we know about the N-termini of the RecQs, we hypothesized that these proteins likely have long disordered termini. In Chapter 3, we confirm the presence of disorder at the Top3/Rmi1 binding site on Sgs1, the Saccharomyces cerevisiae RecQ helicase. We show that even in a disordered state, this binding region is not “featureless,” but in fact contains a transient alpha-helical molecular recognition element that is necessary to facilitate complex formation between Sgs1, Top3 and Rmi1. Loss of helical structure at this site leads to increased genomic instability and sensitivity to DNA damaging agents. Based on these results, we suggest that there are likely many more such elements in the N-terminus that that are important for other Sgs1 protein/protein interactions and provide an estimate for the number of interactions in this region. In Chapter 4, we evaluate the prevalence of disorder in a set of Chromatin Processes proteins in an effort to establish a role for disorder with regards to maintaining chromatin integrity. In our bioinformatics study, we found that disorder is overrepresented in the Chromatin Processes proteins, and that a major driving force for disorder in these proteins is protein/protein interaction and post-translational modification. We also show a biological connection to disorder and increased protein/protein interaction by investigating these parameters in the context of the DNA damage checkpoint response and in complex formations. Mediators between highly structured kinases in the checkpoint were the most interactive proteins and over half of all predicted interaction sites occurred in disordered areas. Complexed proteins often contained one protein with a high number of disordered sites and a high number of predicted interactions, while the rest were considerably more ordered. Chapter 5 explores a Sgs1 interaction partner, Rmi1 and uses bioinformatics to design structurally-based point mutations in an effort to further elucidate Rmi1 function in yeast, which remains largely unknown outside of its enhancement of Top3/Sgs1 catalytic function. Using AGADIR, which predicts alpha-helical structure and is particularly useful in our hands for guided-mutagenesis in disordered regions, we identified several point mutations that lead to Δrmi1 phenotypes or intermediate growth on hydroxyurea. We hypothesize that these mutants are important in maintaining Rmi1 stability. Together, these studies suggest an important change in how the field approaches further studies into the RecQ helicases; traditional methods of primary sequence comparisons and crystal structures limit the study of disordered regions that are still functionally important. Future care should be given to consider the conservation of structure or structural elements in the RecQs over strict alignments when comparing functional regions between orthologs. Our studies also suggest that it is highly likely that structural motifs for important protein interactions in RecQs are being overlooked because they are not readily obvious using traditional methods. By understanding these motifs and the interactions they facilitate, we may be able to more easily identify polymorphisms in patients with genomically unstable conditions like cancer and, having better understood the biological process these structures facilitate, design drugs to counteract detrimental effects.

A elaboração deste trabalho final enquadra-se no âmbito do mestrado em turismo, vertente de gestão estratégica de destinos turísticos, nos termos do regulamento do ciclo de estudos conducente à obtenção do grau de mestre pela Escola Superior de Hotelaria e Turismo do Estoril. Este documento apresenta sob a forma de revisão bibliográfica a compilação da pesquisa efectuada sobre os temas desenvolvidos e aprofundados para melhor cumprir os objectivos definidos pela administração da empresa que acolheu o estágio. É o caso da análise do desenvolvimento turístico actual, da qualidade, do processo de normalização em geral e o que especificamente diz respeito à actividade turística, os sistemas de gestão, a certificação e ainda aos rótulos (nomeadamente de distinção ecológica ou de prestação de serviço) existentes aplicáveis ao Turismo. No núcleo do documento é apresentada e caracterizada a unidade de acolhimento, são descritas as actividades executadas, os principais projectos desenvolvidos, sendo ainda feita uma abordagem sucinta a outros subsectores turísticos com manifesto potencial para a implementação da certificação. No relatório são ainda apresentados os dados referentes a um estudo exploratório, realizado voluntariamente através da aplicação de um inquérito uma selecção de clientes da SGS, ICS cujas actividades se relacionam com o sector turístico, de forma a perceber quais as motivações que levaram estas empresas a certificarem-se e qual o seu interesse nos projectos mais recentes de normalização específica para o sector. O relatório termina com as considerações finais relativas à avaliação de desempenho das tarefas atribuídas durante o estágio, bem como das competências adquiridas e desenvolvidas de acordo com os objectivos propostos.
The main objective of this report is to present a description and a reflective analysis of the trainee development in the certification body SGS, ICS, Lda. This work falls under the scope of the master's degree in tourism, specialization in strategic management of tourism destinations pursuant to the rules of the course leading to the achievement of the degree of Master's by Escola Superior de Hotelaria e Turismo do Estoril. This document is presented with the form of a bibliographic compilation, the deep research done on the subjects developed in order to fulfill the objectives set by the management of the host enterprise. It includes, also, the analysis of current developments in tourism, the concept of quality, the standardization process in general and the one specific to the tourism market, management systems, certification and also the labels (including distinctive ecological or service) applicable to tourism. The hosting unit is presented and characterized at the core of the document, where the activities carried out are described together with the main projects and also a brief approach to other tourism sub-sectors with clear potential for the implementation of the certification. Furthermore, in this report it is also presented data related with an exploratory study, conducted voluntarily through the application of a survey to a selection of SGS ICS customers. Their activities are related to the tourism sector in order to understand the motivations that lead these companies to the certification and their interest in recent standards projects specific to the sector. The document concludes with final remarks concerning the evaluation of the overall performance on the assigned tasks during the training and skills obtained and developed in accordance with the proposed objectives.

This thesis aims to investigate a method and tool for emulation of the General Packet Radio Service (GPRS) core network needed as an environment to test the routing functionality. GPRS is the most widely adopted mobile packet data delivery technology in the world. It utilizes an Intranet Protocol (IP)-based core network and involves significant changes to the way the Global System for Mobile communications (GSM) air interface is structured. It also forms the basis of the future structure of mobile network transmission and switching.

The Serving GPRS Support Node (SGSN) is the most fundamental node in GPRS. Ericsson produces and manages an increasing number of SGSN nodes in the world. One of main functionalities of SGSN node is to forward IP packets according to the destination address in the IP header on IP core network.

In each new release of SGSN, or when implementation or upgrades have been done on routing application on SGSN, design and test engineers at Ericsson need to emulate the IP core network. This must be done with use of many routers to generate huge amounts of data that can simulate the real world IP core network.

The major goal of this thesis was to analyze and verifying the use of a suitable and economical solution to emulating IP Core Network of the GPRS system for testing of different functionality of the routing application running in SGSN , instead of building up a physical Core Network with different infrastructure and many routers.

The method chosen for emulating the IP core network with many routers, and investigated in the thesis, is based on a Cisco simulator called “Dynamips”, which runs many actual Cisco Internetwork Operating Systems (IOS) with many different models of Cisco products in a virtual environment on Windows or Linux platforms. With this simulator, engineers at Ericsson will be able to use this simulator to emulate IP core network easily and efficiently to accomplish system test cases.

A conclusion of this work is that Dynamips could be used to emulate many complicated IP core network scenarios, with many routers to generate huge amounts of data to simulate the real world IP core network. The emulated system fulfils its purpose for testing of the routing application of SGSN regarding different functionality and characteristics. This is done to ensure and verify that SGSN routing application meets its functional and technical requirements, and also helps to find undiscovered errors as well as helps to ensure that the individual components of routing application on SGSN are working correctly.

Sub-grid scale (SGS) models are required in order to model the influence of the unresolved small scales on the resolved scales in large-eddy simulations (LES), the flow at the smallest scales of turbulence. In the following work two SGS models are presented and deeply analyzed in terms of accuracy through several LESs with different spatial resolutions, i.e. grid spacings. The first part of this thesis focuses on the basic theory of turbulence, the governing equations of fluid dynamics and their adaptation to LES. Furthermore, two important SGS models are presented: one is the Dynamic eddy-viscosity model (DEVM), developed by \cite{germano1991dynamic}, while the other is the Explicit Algebraic SGS model (EASSM), by \cite{marstorp2009explicit}. In addition, some details about the implementation of the EASSM in a Pseudo-Spectral Navier-Stokes code \cite{chevalier2007simson} are presented. The performance of the two aforementioned models will be investigated in the following chapters, by means of LES of a channel flow, with friction Reynolds numbers $Re_\tau=590$ up to $Re_\tau=5200$, with relatively coarse resolutions. Data from each simulation will be compared to baseline DNS data. Results have shown that, in contrast to the DEVM, the EASSM has promising potentials for flow predictions at high friction Reynolds numbers: the higher the friction Reynolds number is the better the EASSM will behave and the worse the performances of the DEVM will be. The better performance of the EASSM is contributed to the ability to capture flow anisotropy at the small scales through a correct formulation for the SGS stresses. Moreover, a considerable reduction in the required computational resources can be achieved using the EASSM compared to DEVM. Therefore, the EASSM combines accuracy and computational efficiency, implying that it has a clear potential for industrial CFD usage.

The IMS(In-vehicle Monitoring Subsystem) of VMS(Vehicle Monitoring System) is the multifunctional and complex integrate embedded system, which sends the data of various in-vehicle devices to MC(Monitoring Center) and accepts commands and schedules from there. Using GPRS platform in this system make it possible for real-time and effective data transmission. This paper proposes some new insights on IMS applied to public traffic, including its software and hardware composition, and its realization method.

Double-strand breaks and stalled forks arise when the replication machinery encounters damage from exogenous sources like DNA damaging agents or ionizing radiation, and require specific DNA helicases to resolve these structures. Sgs1 of Saccharomyces cerevisiae is a member of the RecQ family of DNA helicases and has a role in DNA repair and recombination. The RecQ family includes human genes BLM, WRN, RECQL4, RECQL1, and RECQL5. Mutations in BLM, WRN, and RECQL4 result in genetic disorders characterized by developmental abnormalities and a predisposition to cancer. All RecQ helicases have common features including a helicase domain, an RQC domain, and a HRDC domain. In order to elucidate the role of these domains and to identify additional regions in Sgs1 that are required for the maintenance of genome integrity, a series of systematic truncations to the C terminus of Sgs1 were created. We found that ablating the HRDC domain does not cause an increase in accumulating gross chromosomal rearrangements (GCRs). But deleting the RQC domain and leaving the helicase domain intact resulted in a rate similar to that of a helicase-defective mutant. Additionally, we exposed these truncation mutants to HU and MMS and demonstrated that losing up to 200 amino acids from the C terminus did not increase sensitivity to HU or MMS, whereas losing 300 amino acids or more led to sensitivity similar to that of an sgs1∆ cell. These results suggest that the RQC domain, believed to mediate protein-protein interactions and required for DNA recognition, is important for Sgs1’s role in suppressing GCRs and sensitivity to HU and MMS, whereas the HRDC domain that is important for DNA binding is not necessary. RecQL5 is a RecQ-like helicase that is distinct from the other members through its three different isoforms, RecQL5α, RecQL5β, and RecQL5ɣ. It has a helicase domain and an RQC domain, but lacks the HRDC domain that other RecQ-like helicases possess. In contrast to Blm, Wrn, and RecQL4, no human disorder has been associated with defects in RecQL5. For this reason the role of RecQL5 in the cell has remained largely unknown. To try to elucidate the pathways RecQL5 may be involved in we performed a yeast two hybrid to identify RecQL5-interacting proteins. We found that RecQL5 interacts with Hlp2, an ATP-dependent RNA helicase, and Ube2I, a SUMO-conjugating enzyme. These novel interactions shed light on a potential role of RecQL5 in the cell as a transcriptional regulator. Saccharomyces cerevisiae, Rrm3, is a 5’-3’ DNA helicase that is part of the Pif1 family of DNA helicases and is conserved from yeast to humans. It was initially discovered as a suppressor of recombination between tandem arrays and ribosomal DNA (rDNA) repeats. In its absence there are increased rates of extra-chromosomal rDNA circles, and cells accumulate X-shaped intermediates at stalled forks. Rrm3 may be involved in displacing DNA-protein blocks and unwinding DNA to facilitate fork progression. We used stable isotope labeling by amino acids in cell culture (SILAC)- based quantitative mass spectrometry in order to determine proteins that deal with the stalled fork in the absence of Rrm3. We found that in the absence of Rrm3 and increased replication fork pausing, there is a requirement for the error-free DNA damage bypass factor Rad5 and the homologous recombination factor Rdh54 for fork recovery. We also report a novel role for Rrm3 in controlling DNA synthesis upon exposure to replication stress and that this requirement is due to interaction with Orc5, a subunit of the origin recognition complex. Interaction of Orc5 was found to be located within a 26-residue region in the unstructured N-terminal tail of Rrm3 and loss of this interaction resulted in lethality with cells devoid of the replication checkpoint mediator Mrc1, and DNA damage sensitivity with cells lacking Tof1. In this study we describe two independent roles of Rrm3, a helicase-dependent role that requires Rad5 and Rdh54 for fork recovery, and a helicase-independent role that requires Orc5 interaction to control DNA synthesis. Our data provides novel insight into the role of DNA helicases and their role in protecting the genome. Through yeast genetics it was possible to determine the importance of the C terminus of Sgs1 and elucidate new RecQL5 interacting partners that shed light onto roles for RecQL5 distinct from other RecQ like helicases. Quantitative mass spectrometry allowed us to take on a more global view of the cell and determine how it responds to replication fork pausing in the absence of Rrm3. Using both proteomics and yeast genetics we were able to better understand how these DNA helicases contribute to maintaining genome stability.

Les télomères sont des structures nucléo-protéiques présentes à l'extrémité des chromosomes. Ils sont un des facteurs garant de la stabilité génomique. Ils assurent la protection des extrémités des chromosomes et leur entière réplication. Les dysfonctionnements du télomère sont impliqués dans la tumorigénèse et le vieillissement.Un des rôles majeurs des télomères est d'éviter que les extrémités des chromosomes ne soient reconnues comme des cassures double brin de l'ADN et traitées comme telles par la machinerie de réparation. Cependant, de nombreuses protéines impliquées dans la reconnaissance et le métabolisme des cassures double brin, comme la protéine Tel1 et le complexe MRX par exemple, sont présentes au niveau des télomères et participent au maintien de leur taille par la télomérase. En s'appuyant sur cette analogie, j'ai étudié le rôle télomérique de l'endonucléase Sae2 et de l'hélicase Sgs1, impliquées dans l'étape de dégradation du brin 5' qui précède la réparation des cassures double brin de l'ADN par recombinaison.Les rôles des protéines Sae2 et Sgs1 ont été étudiés sur les télomères natifs et sur les télomères érodés lors de la sénescence réplicative. L'ensemble de mes résultats suggèrent que, bien que les télomères érodés en absence de télomérase soient reconnus comme une cassure double brin de l'ADN et traités comme tels par les nucléases et hélicases, le rôle majeur de Sae2 et Sgs1 au niveau des télomères natifs serait de les protéger contre des recombinaisons illégitimes au cours de leur réplication
Telomeres are nucleoprotein complexes that protect the extremities of linear chromosomes, avoiding end-to-end fusions and nucleolytic degradation of chromosome ends. The failure of cells to properly maintain telomeres can be an important source of chromosome instability involved in cancer progression and aging.A major role of telomeres is to prevent chromosome ends from being recognized as damage-induced double-strand DNA breaks (DSBs). However, many proteins involved in recognition and processing of DSBs are also involved in telomeres maintenance, like Tel1 and MRX. Based on this analogy, I have studied the role at telomeres of the role of the endonuclease Sae2 and the helicase Sgs1, two proteins that have a key function in the processing of DSBs through nucleolytic degradation of their 5' end.The role of protein Sae2 and Sgs1 has been studied at native and eroded telomeres. My results showed that eroded telomeres, in telomerase deficient cells, are recognized and resected as a double-strand break DNA by a set of nucleases and helicases including Sae2 and sgs1. In contrast, the main role of Sae2 and Sgs1 at native telomeres would be to protect telomeres against illegitimate recombination during replication

Although numerous spider fibroins have been reported, no known silk coating peptides have been discovered. We provide the first biochemical evidence for a spider coating peptide, called SCP-2, found on gumfooted lines, scaffolding joints and egg cases. The presence of this spider coating peptide on the fibers is supported by MS/MS analysis. Using quantitative real-time PCR analysis, we also demonstrate that SCP-2 has a flagelliform-restricted mRNA pattern of expression. Molecular modeling of the SCP-2 amino acid sequence predicts it adopts an alpha-helical structure that is amphipathic in nature. SCP-2, which can be extracted from fibers using water, is hypothesized to influence the mechanical properties of the silk fibers as well as serve a protective function for the threads. Based upon the restricted pattern of expression of SCP-2, our findings reveal novel insight regarding the glandular function of the flagelliform gland in . cob weaving spiders, suggesting it produces aqueous coating materials that are deposited on a wide range of different silk types. In addition, in an attempt to advance our understanding regarding silk gene transcription, our lab has developed the first antibody against the bHLH factor SGSF. SGSF has been implicated as a potential transcriptional regulator of silk gene transcription in spiders. Development of the anti-SGSF antibody was accomplished via the overexpression and purification of a fusion protein in bacteria, which consisted of the C-terminal region of SGSF fused to thioredoxin. Purified SGSF fusion proteins were injected into rabbits and the polyclonal antiserum was collected and tested by western blot analysis to determine the specificity of the immunological reagent. Western blot analyses revealed the anti-SGSF antiserum was capable of recognizing bacterially expressed SGSF in an efficient manner. Collectively, these studies lay the groundwork for future investigations involving the use of the antibody to determine the role of SGSF in silk transcription.

A actual posição dos mercados e da sociedade em termos de Segurança Sanitária Alimentar vieram impor um elevado grau de exigência a todos os intervenientes na cadeia alimentar, as quais quiseram responder em conformidade e demonstrar as suas capacidades para identificar e controlar potenciais perigos associados aos alimentos. Este trabalho descreve as fases inicias de desenvolvimento de um sistema de gestão de segurança sanitária alimentar segundo a ISO 22000:2005, numa empresa dedicada à exportação de produtos da pesca frescos. A empresa opera a nível internacional, num segmento alto do mercado, e por isso foi verificada a necessidade de certificar o sistema de gestão de segurança sanitária alimentar para aumentar a sua competitividade e garantir o cumprimento dos requisitos de segurança sanitária alimentar a nível global. Numa fase inicial de desenvolvimento do projecto realizou-se uma auditoria interna de diagnóstico. Com base na análise dos resultados foi planeado o sistema de gestão da segurança sanitária alimentar e preparada a sua plena implementação sem ser definido um prazo para a conclusão. As necessidades de planeamento verificadas foram inicialmente de preparação de documentação. No decorrer do projecto foi dedicada especial atenção à análise dos critérios internos de classificação individual dos produtos da pesca frescos, por métodos sensoriais, uma vez que estes critérios sustentam a política interna de qualidade. Devido às limitações de tempo curriculares este trabalho não acompanhou o projecto até à sua completa implementação e certificação.
ABSTRACT - Development of a food safety management system according to ISO 22000:2005 in an industrial export unit of fresh fishery products - The current position and demands of the market and society in terms of food safety imposed a high standard to all stakeholders in the food chain, which wanted to answer accordingly and demonstrate their abilities to identify and control potential hazards associated with food. This thesis describes the early stages of development of a food safety management system, according to ISO 22000:2005, for a company dedicated to the export of fresh fishery products. The company operates internationally in the top segment of the market witch required the certification of the food safety management system, to increase its competitiveness and ensure compliance with food safety requirements worldwide. In an initial phase of the project development took place an internal audit of diagnosis. Based on the analysis of the results, the food safety management system was planned and prepared for its full implementation without a set deadline for its conclusion. The planning needs were initially the preparation of documentation. During the project design, a special attention was devoted to the analysis of internal criteria for classification of individual fresh fishery products, by sensory methods, since these criteria support the internal policy of quality. Due to curriculum time constraints this study did not follow the full implementation and certification.

The benefits of Ericsson virtual Evolved Packet Core includes all the benefits of NFV and provides verified solutions addressing a large number of vertical use-cases. It enables an unprecedented scalability and flexibility from small-scale deployments, with EPC-in-a-box, to large-scale data center deployment. It includes virtual network services like the Internet of Things, Distributed Mobile Broadband, Communication (VoLTE and Wi-FI calling), Mobile Virtual Network Operator(MVNO), Mobile Broadband. Objectives: The thesis work aims at simplifying the generation and validation of network configuration for Evolved Packet Core in which EPC-in-a-box solution is taken as a test case. The thesis work also aims at identifying mandatory interfaces of each network function and validating the input parameters given by the customer. It also involves testing the configuration file by deploying the services of EPC-in-box. Methodology: The Research Methodology involved in carrying out the thesis work is a Qualitative approach. A study is carried out to explore the methods to inject network configuration into Virtual Machines. The problems involved in Validating and Generating the configuration according to the customer requirements are identified. A suitable method is developed to simplify the process. Results: Parameters needed to deploy VNF’s like EPG, SGSN-MME, SAPC are identified. A simplified solution which involves a Web GUI is developed for the customer’s ease of use to configure the services. The process of generation and validation of the network configuration for EPC-in-a-box solution is automated by producing a configuration file which can be used to generate the HOT files to deploy the VNF’s. Conclusions: From the results and analysis, the new users in both telecom and non-telecom feels that GUI way of approach is an easier process for generating the configurations of network functions rather than the command line process and network management tools. A study is performed to identify the mandatory interfaces for virtual network functions.

Context: Data compression is the technique that is used for the fast transmission of the data and also to reduce the storage size of the transmitted data. Data compression is the massive and ubiquitous technology where almost every communication company make use of data compression. Data compression is categorized mainly into lossy and lossless data compression. Ericsson is the telecommunication company that deals with millions of user data and, all these data get compressed using the Deflate compression algorithm. Due to the compression ratio and compression speed, the deflate algorithm is not optimal for the present use case(compress twice and decompress once) of Ericsson. This research is all about finding the best alternate algorithm which suits the current use case so that the deflate algorithm can replace it. Objectives: The objective of the research is to replace the Deflate algorithm with the algorithm, which is useful for compressing the Serving GPRS Support Node-Mobility Management Entity(SGSN-MME) user data effectively. The main objectives to achieve this goal are: Investigating the better algorithm which fits the SGSN-MME compression patterns, investigating the few alternate algorithms for Deflate algorithm, the SGSN- MME dataset used to perform experimentation, the experiment should perform by using all selected algorithms on the dataset, the results of the experiment were compared based on the compression factors, based on the performance of algorithm the Deflate algorithm will get replaced with the suitable algorithm. Methods: In this research, a literature review performed to investigate the alternate algorithms for the Deflate algorithm. After selecting the algorithm, an experiment conducted on the data which was provided by Ericsson AB, Gothenburg and based on the compression factors like compression ratio, compression speed the performance of the algorithm evaluated. Results: By analyzing the results of the experiment, Z-standard is the better performance algorithm with the optimal compression sizes, compression ratio, and compression speed. Conclusions: This research concludes by identifying an alternate algorithm that can replace the Deflate algorithm and also which is suitable for the present Use case.

Notre equipe etudie les relations entre la structure chromatinienne et l'expression des genes, avec comme modele le gene sgs3 chez drosophila melanogaster. L'utilisation de larves transgeniques a revele, en amont du site d'initiation, deux zones regulatrices: la zone a, de 130 a +1, responsable des specificites tissulaire et temporelle d'expression du gene ainsi que de la reponse a l'ecdysone; la zone b, de 1300 a 130, responsable du niveau eleve de l'expression du gene. La cartographie des sites hypersensibles a la dnase i (dhs) a revele dans la zone b un complexe de trois dhs dont celui a 600 est etroitement correle a l'activite du gene dans les glandes salivaires au troisieme stade larvaire et possede certaines caracteristiques des elements cis-activateurs (enhancers). Par l'etude in vivo de la structure chromatinienne de cette region nous avons directement montre l'existence d'un nucleosome positionne sur la region 600 lorsque sgs3 est inactif et que l'activation du gene s'accompagne du deplacement de cette structure. Parallelement, une proteine gebf-i, dont la presence suit l'activite du gene a ete caracterisee. Elle se lie simultanement a deux sites d'une sequence de 63 pb centree a 600. L'utilisation de mutants dans le locus 2b5, inductible par l'ecdysone, montre que la presence de gebf-i est liee a l'expression de ce locus. Grace aux larves hybrides issues du croisement entre d. Melanogaster et d. Simulans, des modifications post-traductionnelles importantes pour la liaison de gebf-i ont ete mises en evidence. Avec des larves transgeniques dans lesquelles les sites de liaison de gebf-i ont ete mutes, nous avons montre, in vivo, l'implication directe de cette proteine dans la regulation du niveau d'expression de sgs3 et dans le fonctionnement de l'enhancer par deplacement d'un nucleosome positionne. L'ensemble de ces resultats conduit a penser que le role des activateurs de la transcription ne peut etre etudie independamment de l'engagement de l'adn dans la structure chromatinienne

Porcine reproductive and respiratory syndrome virus (PRRSV) is arguably the most economically devastating pathogen affecting the global swine industry. Since the emergence of the virus in the late 1980s, vaccination strategies aimed to control the virus have not been very effective. Current commercial vaccines are generally protective against homologous or closely-related strains but ineffective at conferring heterologous protection against genetically-diverse strains of the virus. Consequently, emergence of variant and sometime more pathogenic strains of PRRSV continues in global swine herds. As such, there is a need for better understanding of the molecular mechanisms involved in the replication of the virus. In order to better understand the molecular mechanisms of host responses to PRRSV replication, we first sought to evaluate the ability of the virus to induce stress granules (SGs) during PRRSV infection. SGs are intracellular, cytoplasmic aggregates of RNA-binding proteins (RBPs) and mRNA. Formation of SGs is observed upon cellular stress and ultimately function to arrest cellular translation to promote cellular survival until the stress has been remedied. Indeed, several viruses have been shown to modulate the SG pathways to facilitate viral replication and even suppress the host's immune response. However, it is currently unknown whether PRRSV modulates the SG response. First, we used confocal microscopy and fluorescent in situ hybridization (FISH) to determine the distribution of known SG marker proteins and cellular mRNAs. Our findings revealed that PRRSV induces a potent SG response at late time points post-infection, and that SGs were closely associated with viral replication complexes (VRCs). Subsequently, we demonstrated that SGs are dispensable for viral replication, as short hairpin RNA (shRNA)-mediated knockdown of critical SG components (G3BP1 and G3BP2) did not affect viral replication. Interestingly, we found that the PRRSV-induced SGs are formed in a PERK-dependent manner. PERK is an important sensor of ER stress and activator of the unfolded protein response (UPR). Further investigation into the PERK signaling pathway revealed that PRRSV induces a significant amount of ER stress upon the cell during viral infection, and that exogenous stress significantly impaired the ability of the virus to replicate in MARC145 cells. We also showed that PRRSV potently induces all three signaling branches of the UPR, including PERK. While PERK knockdown had no effect on cell viability or viral replication, it significantly upregulated the mRNA expression of interferon-β and interferon stimulated genes (ISGs). The results from our studies suggest a critical role for PERK in regulating the host innate immune response to PRRSV infection. Only with a better understanding of the underlying molecular mechanisms of PRRSV replication will we be able to rationally design more effective vaccines against the virus.

Porcine reproductive and respiratory syndrome virus (PRRSV) causes an economically-devastating disease in the global swine industry. Annually, PRRSV is estimated to cause more than $600 million in economic losses to the swine industry in the United States alone. Current commercial vaccines against the virus are not effective against the diverse field strains largely due to the extreme heterogeneity of the virus. PRRSV is also able to potently suppress several aspects of the host's immune response and therefore establish a persistent infection. The underlying mechanisms of PRRSV-mediated immune suppression are not well understood. Therefore, in this dissertation we decided to investigate the molecular mechanisms of host responses to PRRSV infection. We first investigated the ability of the virus to induce stress granules (SGs). SGs are important intracellular regulatory components that modulate many aspects of the host's cellular processes, and have even been shown to play roles in regulating viral replication and controlling immune responses to viral infection. We demonstrate that PRRSV not only induces SGs, but that the PRRSV-induced SGs are closely associated with viral replication complexes (VRCs) within infected cells. The PRRSV-induced SGs were dispensable for viral replication. PRRSV-induced SGs were previously shown to form in a PERK dependent manner. Therefore, in the second part of this dissertation research, we decided to investigate the PERK signaling pathway during PRRSV infection. PERK is an important sensor of ER stress and activator of the unfolded protein response (UPR). Our results showed that PRRSV potently induces ER stress and all three signaling branches of the UPR, including PERK. Furthermore, we revealed that PERK may play an important role in regulating the type I interferon response to PRRSV infection. The results from our studies will aid in understanding the underlying molecular mechanism of PRRSV replication which will help rationally design the next generation of more effective vaccines against this devastating swine pathogen.

Subglottic stenosis (SGS) is a narrowing of the airway in the region of the cricoid cartilage below the vocal folds and above the tracheal rings. Individuals with SGS experience difficulty breathing at rest and during exertion both of which become increasingly difficult with the level of stenosis severity. Some individuals also experience negative voice changes. Individuals whose stenoses significantly impact breathing generally require medical procedures or surgery, either balloon dilation or cricotracheal resection (CTR). CTR has been shown to improve patients' ability to breathe, but it can also result in permanent vocal changes. Alternatively, balloon dilation results in similar breathing improvements but for a relatively short period of time. Many studies have been published on the effectiveness of CTR; however, only a few have examined the effects of CTR on vocal production. The purpose of this study is to quantify the acoustic and auditory-perceptual features of subglottic stenosis and examine possible acoustic and auditory-perceptual changes in voice production following a revised CTR aimed to minimize voice impact in a group of women. A retrospective chart review identified women with idiopathic SGS who received revised CTR at The University of Utah Voice Disorders Center between 2008 and 2014. Presurgical and postsurgical groups included patients with both pre and post recordings (n = 11) as well as patients with only pre (n = 6) or post (n = 9) recordings. Acoustic quantification of voice signal periodicity, as well as cepstral, spectral, and fundamental frequency (F0) analyses were performed. Auditory-perceptual ratings of overall quality and monotonicity were performed. Cross-sectional and pre-post surgery analyses were completed. Aggregate analyses revealed that both pre and posttreatment SGS patients demonstrated voice disorders in the mild to moderate severity range. Pre-post comparisons indicated no significant voice change after surgery. Mean fundamental frequency decreased from 215 Hz (SD = 40 Hz) to 201 Hz (SD = 65 Hz). Voice disorder severity based on the cepstral spectral index of dysphoniaTM for sustained vowels decreased (i.e., improved) from 41 (SD = 41) to 25 (SD = 21) points. Semitone standard deviation (2.2 semitones) was equivalent from pretreatment to posttreatment. Auditory-perceptual ratings demonstrated similar results. These preliminary results indicate that the revised CTR procedure is promising in minimizing adverse voice effects. Future research is needed to determine causative factors for pretreatment voice disorders, as well as to optimize treatments in this population.

Computational fluid dynamics (CFD) is a significant tool routinely used indesign and optimization in aerospace industry. Often cases with unsteadyflows must be computed, and the long compute times of standard methods hasmotivated the present work on new implicit methods to replace the standardexplicit schemes. The implementation and numerical experiments were donewith the Swedish national flow solver Edge, developed by FOI,universities, and collaboration partners.The work is concentrated on a Lower-Upper Symmetric Gauss-Seidel (LU-SGS)type of time stepping. For the very anisotropic grids needed forReynolds-Averaged Navier-Stokes (RANS) computations of turbulent boundary layers,LU-SGS is combined with a line-implicit technique.  The inviscid flux Jacobians which contribute to the diagonalblocks of the system matrix are based on a flux splitting method with upwind type dissipation giving  control over diagonal dominance and artificial dissipation.The method is  controlled by several parameters, and comprehensivenumerical experiments were carried out to identify their influence andinteraction so that close to optimal values can be suggested. As an example,the optimal number of iterations carried out in a time-step increases with increased resolution of the computational grid.The numbering of the unknowns is important, and the numberings produced by mesh generators of Delaunay- and advancing front-type wereamong the best.The solver has been parallelized with the Message Passing Interface (MPI) for runs on multi-processor hardware,and its performance scales with the number of processors at least asefficiently as the explicit methods. The new method saves typicallybetween 50 and 80 percent of the runtime, depending on the case, andthe largest computations have reached 110M grid nodes. Theclassical multigrid acceleration for 3D RANS simulations was foundineffective in the cases tested in combination with the LU-SGS solverusing optimal parameters. Finally, preliminary time-accurate simulations for unsteady flows have shown promising results.

Les anticorps neutralisants exercent une pression de sélection sur le VIH-1 et sont un des moteurs de son évolution. Nous nous sommes intéressés à la relation entre la diversité génétique du virus et la neutralisation par les anticorps, à l’échelle individuelle et à l’échelle populationnelle. À l’échelle de l’individu. nous avons étudié le phénomène de compartimentation du VIH-1 au niveau du système nerveux central. Nos résultats suggèrent que la pression de sélection par les anticorps neutralisants n’est pas responsable de l’évolution des variants neurotropes, observée au niveau du LCR. Nous avons également mis en évidence des spécificités de ces variants neurotropes. Dans une seconde partie, nous avons étudié la sensibilité à différents anticorps monoclonaux largement neutralisants de virus de clade CRF02_AG récemment transmis en France. Nous avons mis en évidence une augmentation de résistance à plusieurs anticorps, positivement corrélée à la diversification génétique des virus sur une période de quinze ans. Ceci est en faveur d’une répercussion de la pression de sélection exercée au niveau individuel sur la résistance croissante du VIH-1 à la neutralisation, à l’échelle populationnelle
Neutralizing antibodies exert a selection pressure that drives HIV-1 evolution, playing a major role in its diversification. We were interested in the relationship between the genetic diversity of the virus and its neutralization by antibodies at both the individual and the populational levels. At the individual level, viral populations with distinct genetic properties have been observed in the central nervous system in about half of the subjects infected with HIV-1. We studied this phenomenon, termed compartmentalization. Our results suggest that the selection pressure by neutralizing antibodies is not responsible for the evolution pathway of neurotropic variants in the CSF. We also observed specific features for these neurotropic variants. In the second part of this work, we evaluated the sensitivity to broadly neutralizing antibodies of recently transmitted CRF02_AG viruses isolated in France. We observed an increased resistance to some monoclonal antibodies, which was positively correlated with the diversification of the viruses during the study period. This is in line with the hypothesis of the repercussion of the selection pressure within individuals towards a greater resistance of HIV-1 at the populational level

Assim, a segurança de informação torna-se crítica ao proteger os ativos de informação, pelo que a estratégia de segurança organizacional tem vindo a alinhar-se com os seus objetivos de negócio. Por outro lado, as recentes alterações legais, tais como a Diretiva Segurança das Redes e da Informação e o Regulamento Geral de Proteção de Dados, vêm impor regras relativamente à privacidade e à segurança da informação, permitindo às organizações um redesenho ou ajuste dos seus processos de forma a garantir que a informação se encontra efetivamente segura. Neste contexto, o Chief Information Security Officer assume um papel de destaque na coordenação da confidencialidade, integridade e disponibilidade da informação na organização. Este trabalho pretende estudar o ambiente geral da segurança de informação nas organizações, analisar o papel do CISO, e compreender onde este deverá estar integrado na estrutura organizacional. Para tal, foram realizadas entrevistas a consultores especialistas e a pessoas com cargos diretivos nas áreas de sistemas de informação e de segurança da informação, que permitiram concluir que ainda é necessário um grande amadurecimento a nível das organizações em Portugal relativamente ao tema, e que tal poderá dever-se à ausência de uma cultura de segurança estabelecida no país. Por outro lado, o papel do CISO tem assumido uma maior relevância, sendo que é uma opinião geral que o mesmo deverá ter uma relação próxima com a administração das organizações.
In an increasingly connected and digital world, information is seen as a business enabler and a source of sustained competitive advantage. Thus, information security is becoming critical so to protect these information assets, which is why the concern with organizations’ security strategy has been aligning with their strategic objectives. On the other hand, recent changes in regulation, as Network and Information Security (NIS) directive and the General Data Protection Regulation (GDPR), come to regulate and create rules when it comes to information security, and allow organizations to redesign or adjust these processes in order to ensure that information is, in fact, safe. In this context, the Chief Information Security Officer (CISO) comes to play an important role in coordinating confidentiality, integrity, and availability of information in the organization. This paper aims to study organizations’ information security environment in general, analyse the CISO’s role inside them, and understand where they should be integrated in the corporate structure. To do so, interviews were conducted on experienced information security consultants and information systems and information security directors, which allowed to conclude that organizations in Portugal still need a great amount of maturing when it comes to information security, and that this may eventually be due to the absence of an established security culture in the country. On the other hand, the CISO’s role has been increasing in relevance, being a general opinion that their relationship with organizations’ boards should be close.

Ce travail de thèse part d’un constat : d’importantes pertes thermiques sont observées lors de l’élaboration des matériaux du génie civil. Motivés par l’ajout d’une double enveloppe, les travaux contenus dans ce manuscrit visent à caractériser un échangeur de chaleur appliqué en paroi des fours rotatifs. Tout d’abord, un modèle intégré est développé, puis validé sur une centrale d’enrobage dont la paroi intérieure du four est munie de releveurs nécessaires au mélange des matériaux. Leur distribution dans la section transversale est estimée à partir d’une loi de déchargement granulaire. Les expérimentations numériques montrent que les transferts Gaz / Solide et Solide / Paroi dominent, ce dernier phénomène limitant les performances globales du procédé. L’ajout de l’échangeur sur paroi mobile est quant à lui exploré sur un banc d’essai instrumenté de type rotor-stator, avec entrée tangentielle. Développant un écoulement de type Taylor-Couette-Poiseuille, les transferts thermiques pariétaux sont caractérisés expérimentalement. Les résultats sont sans équivoque dans la gamme de nombres de Reynolds imposés : la contribution du mouvement axial surpasse le mouvement rotationnel turbulent. Une corrélation adimensionnelle basée sur le nombre de Nusselt est proposée afin d’estimer numériquement l’effet de l’échangeur sur les profils de température internes dans le four. Enfin, les structures tourbillonnaires de l’écoulement dans l’espace annulaire sont étudiées à partir d’un code de mécanique des fluides numériques utilisant la Simulation aux Grandes Echelles. Les simulations permettent de décrire les cellules contrarotatives au sein de la couche limite turbulente dont l’amplitude et la fréquence de passages sont reliées aux paramètres de fonctionnement de l’échangeur (débit axial et vitesse de rotation du four)
This work results on a finding: the heat loss from rotary kiln represents a significant energy amount during materials processing in civil engineering domain. Motivated by traditional energy recovery methods from heat exchanger, this thesis is aimed at providing their rigorous thermodynamic diagnostics. Firstly, a thermal-granular model is developed, and then validated in asphalt plant whose the rotary kiln is composed of flights to ensure the materials mixing. Their cross-section distribution is calculated from a granular discharge law. The numerical experiments showed an increase of heat transfer phenomena between gases and solids, and those between the solids and the wall, this latter phenomenon limiting the process performances. Heat recovery exchanger applied to the rotary kiln is studied from a semi-industrial pilot based on a rotor stator configuration including a tangential inlet. Developing a Taylor-Couette-Poiseuille flow, the heat transfer results are undoubted in the range of the studied Reynolds numbers: the axial motion contribution is larger than the rotational turbulent motion. A dimension less criterion is proposed in order to be applied to the aforementioned integrated model including the heat exchanger applied to the rotary kiln, its effect being assessed upon the internal thermalprofiles. Finally, the vortices flow structure within the annular gap exchanger is studied from Large Eddy simulation. The amplitude and frequency passage of the contrarotatives cells located in the turbulent boundary layer are connected to the process parameters (the axial flowrate and the kiln shell rotation)

Notre travail consiste à prendre en considération les incertitudes dans la planification et l'ordonnancement de projets et résoudre les problèmes Rough Cut Capacity Planning, Resource Leveling Problem et Resource Constraint Project Scheduling Problem sous incertitudes. L'incertitude est modélisée avec l'approche floue/possibiliste au lieu de l'approche stochastique ce qui est plus adéquat avec notre cas d'étude. Trois types de problèmes ont été définis dans cette étude à savoir le Fuzzy Rough Cut Capacity Problem (FRCCP), le Fuzzy Resource Leveling Problem (FRLP) et le Fuzzy Resource Constraint Project Scheduling Problem (RCPSP). Un Algorithme Génétique et un Algorithme "Parallel SGS" sont proposés pour résoudre respectivement le FRLP et le FRCPSP et un Recuit Simulé est proposé pour résoudre le problème FRCCP.

L’invecchiamento delle installazioni industriali è stato oggetto di una crescente attenzione nell’ultimo decennio, sia da parte delle autorità pubbliche che dall’industria, legata al fatto che molte delle installazioni industriali attualmente in esercizio hanno raggiunto o superato la fine della loro vita nominale di progetto. Tale constatazione solleva la questione di come gestire in modo sicuro gli impianti datati e come rallentare l’invecchiamento di quelli più recenti. È compito del gestore dello stabilimento sincerarsi delle condizioni degli elementi presenti e mettere in atto un adeguato piano ispettivo e manutentivo, per ridurre al minimo la possibilità che si verifichi un incidente rilevante imputabile all’invecchiamento degli elementi stessi. Questo elaborato di tesi si propone di valutare l’adeguatezza del programma di gestione dell’invecchiamento degli elementi critici di uno stabilimento a rischio di incidente rilevante, applicando un metodo ad indici descritto all’interno di una Linea Guida messa a punto da un gruppo di lavoro inter-istituzionale coordinato dall’Istituto Nazionale Assicurazione contro gli Infortuni sul Lavoro (INAIL). Il presente lavoro di tesi si compone di 6 capitoli. Il Capitolo 1 è di carattere introduttivo. Il Capitolo 2 tratta il fenomeno dell’invecchiamento, spiegando come questo si manifesti, da dove si origini e quali apparecchiature ne sono maggiormente interessate. Il Capitolo 3 descrive il metodo INAIL per la valutazione della gestione dell’invecchiamento delle apparecchiature. Il Capitolo 4 descrive lo stabilimento presso il quale è stato sviluppato il lavoro di tesi, che rappresenta il caso di studio, a cui il metodo INAIL è stato applicato. Il Capitolo 5 presenta l’applicazione del metodo INAIL allo stabilimento assunto come caso di studio. Infine, il Capitolo 6 contiene alcune considerazioni conclusive.

This work deals with spatially-evolving supersonic turbulent boundary layers over adiabatic and cold walls at M∞ = 2 and up to Re0 ≈ 2600 using 3 different SGS models. The numerical methodology is based on high-order split-centered scheme to discretize the convective fluxes of the Navier-Stokes equations . For the adiabatic case, it is demonstrated that all SGS models require a comparable minimum grid-refinement in order to capture accurately the near-wall-turbulence. Overall, the models exhibit correct behavior when predictiong the dynamic properties, but show different performances for the temperature distribution in the near-wall region. For the isothermal case, it is found that the compressibility effects are not enhanced due to the wall cooling. As expected, the total temperature fluctuations are not negligible in the near-wall region. The study shows that the anti-correlation linking both velocity and temperature fields, derived from the Morkovin's hypothesis, is not satisfied.
