Abstract Semi-automated segmentation algorithms hold promise for improving extraction and identification of objects in images such as tumors in medical images of human tissue, counting plants or flowers for crop yield prediction or other tasks where object numbers and appearance vary from image to image. By blending markup from human annotators to algorithmic classifiers, the accuracy and reproducability of image segmentation can be raised to very high levels. At least, that is the promise of this approach, but the reality is less than clear. In this paper, we review the state-of-the-art in semi-automated image segmentation performance assessment and demonstrate it to be lacking the level of experimental rigour needed to ensure that claims about algorithm accuracy and reproducability can be considered valid. We follow this review with two experiments that vary the type of markup that annotators make on images, either points or strokes, in tightly controlled experimental conditions in order to investigate the effect that this one particular source of variation has on the accuracy of these types of systems. In both experiments, we found that accuracy substantially increases when participants use a stroke-based interaction. In light of these results, the validity of claims about algorithm performance are brought into sharp focus, and we reflect on the need for a far more control on variables for benchmarking the impact of annotators and their context on these types of systems.

There are several methods of information convenient for natural science education, including press and television. The author checks the information which was published on the TV show and press by Rūta Janutienė, concerning the bread contamination with high stability toxin glyphosate. This herbicide is used to sprey the crop. The article provides information about the worldwide scientific research in order to investigate the effect of toxic glyphosate on humans and the environment. The conclusion is drawn about the negative influence of glyphosate on the reproducability. The author proves that journalist Rūta Janutienė has touched an essential contemporary ecological issue. Two commercial freezers were modified to provide an inexpensive chamber system to investigate frost effects on wild, lowbush blueberries (Vaccinium angustifolium) under field conditions. A computer control system was developed with software written in Visual Basic 6.0 for MSWindows, which precisely controlled temperature in the plant canopy when the chambers were placed over blueberry plants in the field. Frost events (with temperatures ranging from -2 to -15 °C (28.4 to 5.0 °F)) were simulated by user input to control the cooling and warming rates, and minimum temperatures. The system records temperature set points, and current temperature in the plant canopy, or elsewhere in the plant environment, and provides a graphical display of key parameters. Trials have verified the reproducability of temperature profiles and the chambers have been used to provide preliminary information on the effects of frost at bloom on fruit set and development.

The charge density of a hexapeptide was determined from high-resolution CCD area-detector experiments at 100 K. Two datasets, one from a rotating anode and a second one from synchrotron radiation, were measured and the results are compared. The data are interpreted in terms of the `rigid pseudoatom' model. The topology of the experimental density is analyzed and compared with the topology of the constituting amino acids, and shows good agreement. All critical points of the electron density at the covalent and hydrogen bonds, as well as those of the Laplacian, were located. With respect to the transferability of electronic and bond topological properties the six peptide bonds were compared with values given in the literature.

ObjectiveA Working Group (WG) of tobacco regulatory science experts identified measures for the tobacco environment domain.MethodsThis article describes the methods by which measures were identified, selected, approved and placed in the PhenX Toolkit.FindingsThe WG identified 20 initial elements relevant to tobacco regulatory science and determined whether they were already in the PhenX Toolkit or whether novel or improved measures existed. In addition to the 10 complementary measures already in the Toolkit, the WG recommended 13 additional measures: aided and confirmed awareness of televised antitobacco advertising, interpersonal communication about tobacco advertising, media use, perceived effectiveness of antitobacco advertising, exposure to smoking on television and in the movies, social norms about tobacco (for adults and for youth), worksite policies, youth cigarette purchase behaviours and experiences, compliance with cigarette packaging and labelling policies, local and state tobacco control public policies, and neighbourhood-level racial/ethnic composition. Supplemental measures included youth social capital and compliance with smoke-free air laws and with point of sale and internet tobacco marketing restrictions. Gaps were identified in the areas of policy environment (public and private), communications environment, community environment and social environment (ie, the norms/acceptability of tobacco use).ConclusionsConsistent use of these tobacco environment measures will enhance rigor and reproducability of tobacco research.

Recently published studies have shown that microfluidic devices fabricated by in-house three-dimensional (3D) printing, computer numerical control (CNC) milling and laser engraving have a good quality of performance. The 3-in-1 3D printers, desktop machines that integrate the three primary functions in a single user-friendly set-up are now available for computer-controlled adaptable surface processing, for less than USD 1000. Here, we demonstrate that 3-in-1 3D printer-based micromachining is an effective strategy for creating microfluidic devices and an easier and more economical alternative to, for instance, conventional photolithography. Our aim was to produce plastic microfluidic chips with engraved microchannel structures or micro-structured plastic molds for casting polydimethylsiloxane (PDMS) chips with microchannel imprints. The reproducability and accuracy of fabrication of microfluidic chips with straight, crossed line and Y-shaped microchannel designs were assessed and their microfluidic performance checked by liquid stream tests. All three fabrication methods of the 3-in-1 3D printer produced functional microchannel devices with adequate solution flow. Accordingly, 3-in-1 3D printers are recommended as cheap, accessible and user-friendly tools that can be operated with minimal training and little starting knowledge to successfully fabricate basic microfluidic devices that are suitable for educational work or rapid prototyping.

AbstractN-nitrosamines are a new class of emerging nitrogenous drinking water disinfection by-products. These compounds are probably carcinogenic which could seriously affect the safety of drinking water consumers. The aim of this study is to develop a simple, fast, and specific analytical method for the routine determination of low part per trillion levels of N-nitrosamines in waters. An ultra high pressure liquid chromatography coupled with tandem mass spectrometry (UHPLC/MS/MS) method was developed for the qualitative and quantitative analysis of N-nitrosamines in waters. N-nitrosamines were extracted, purified and concentrated from water samples in one step using a solid-phase extraction (SPE). The compounds were detected in multiple reaction monitoring via electrospray ionisation source with positive ionisation mode. To achieve symmetrical peak shapes and a short chromatographic analysis time, the mobile phase consisting of acetonitrile, water and formic acid (60:40:0.1, v/v/v) was used in the experiment. Chromatographic separation of N-nitrosamines was done in less than two minutes. All calibration curves had good linearity (r2≥ 0.9989). The intra- and inter-day precision of the assay ranged from 0.59% to 3.11% and accuracy ranged from 99.66% to 104.1%. The mean recoveries of N-nitrosamines in spiked water were 98%-101%. The reproducability was acceptable with relative standard deviations of less than 3.53%. The proposed method yielded detection limits very low which ranges from 0.04 to 0.16 ng L−1. Finally, the developed analytical method was successfully applied to the analysis of N-nitrosamines in natural water sample

And the final commercial product is a mixture of nucleotidic fractions which have formed by the ability of the molecule to form intra-molecular bonding., with random sequences such as:P1-5, (dAp)12–24, (dGp) 10–20, (dTp)13–26, (dCp)10–20, Wherein, P=phosphoric radical; dAp= deoxyadenylic monomer; dGp= deoxyguanylic monomer; dTp= deoxythymidylic monomer; dCp= deoxycytidilic monomer.(US Patent # 4,985,552,Crinos). This report aims to investigate for the first time the sequence spesificity, and the reproducability of the nucleotidic fractions of DF. and to define a structural base for DF as a global modulator of the signalling network. Method:.Materials from two commercial forms of DF, Prociclide and Noravid in solution and capsule forms, were separated in each case by HPLC on a Vydac, C8 column using 0.1% trifluoroacetate in water. 513 mass spectra peaks were collected, freeze dried, preserved on ice and subjected to Voyager Elite Biospectrometry Workstation using the technique of delayed extraction matrix assisted laser desorption ionization time of flight (MALDI-TOF) mass spectrometry..A blank run representing DNA matrix was done in each case on MALDI-TOF to detect the low MW particles that were not related to the sample. The peaks were superimposed on the HPLC profiles of mono-, di-, tri-phosphates of T,C,G, A via crossmatching the MW, together with retention times as described above.260,000 crossmatches were done. Statistics: The min, max, mean and median mass values were stratified into four respective batches of DF, capsule and liquid forms, and two commercial labels The standard deviations (SD) were calculated in each of the above stratifications with the “n-1” method. Results: HPLC profiles contained more than one molecular species with MWs ranging from <300 to <1500 Da. and were not superimposable. A representative panel would include deoxyadenylic and deoxycytidylic monomers, guanine and other nucleotides of 3–25 bases, such as dC,dA, G, dGMP, AMP, oligonucleotides with 3–5 bases such as UTP, dTTP, CTP, ATP, dGTP, cTMP, CMP, cGMP, and dAMP. Since the study was only directed at bases, deoxy-bases and mono-and di-, and tri-nucleotides of </= 560 Da (the calculated MW of alkali sodium salt of ATP), statistical calculations were carried out only for these fractions. When compared to capsules, liquid DF showed double number of peaks of ATP, ADP, AMP, GTP, GDP, and GMP. The standard deviations (STD) on the nucleotides in the various liquid forms of Defibrotide were much narrower, ranging between (+ / −) 0–1.4, capsule forms showing less consistency with STD’s of (+ / −)0–2.1 Conclusion: The sequence analysis of the nucleotidic fractions of <560 D MW displayed presence of cAMP, ATP, ADP, GTP, GDP,,GMP, CTP,CMP,CDP, TTP,TDP,TMP reproducably between batches. DF maybe a direct and indirect supplier of messenger nucleotides. This structural analysis may support this hypothesis generating report presenting DF as the prototype global modulator of the signalling network as a mechanism of its polypharmacology.

AbstractA kinetic model is presented to explain GaN growth. The model is based on established values for the N and Ga desorption kinetics and well founded assumptions on the adsorption and decomposition of the N and Ga containing precursors. When grown on similar nucleation layers, it is shown that high quality GaN films are achieved when the V/III ratio is chosen to be slightly larger than the Ga and N desorption rates. The model is verified by comparing the structural, optical, and electrical properties of the GaN to the growth temperature and V/III ratio. The model explains several features of GaN growth including, growth conditions for smooth surface morphology, growth conditions for highly resistive GaN, and a possible explanation for the origin of Ga and N vacancies in GaN. Based on the growth model, ordering of the GaN during growth is achieved via an adsorption/desorption cycle where Ga and N containing species are exchanged between the gas phase boundary layer and the solid surface. Consequences of the model on establishing growth conditions and run-to-run reproducability are also discussed.

Propionic acid bacteria, including Propionibacterium acidipropionici, are widely used in the chemical industry to produce propionic acid and also for food and feed preservation. However, the efficiency of the industrial production of these bacteria is limited by their sensitivity to high concentrations of propionic acid excreted into the cultivation medium. Therefore, the development of new biotechnological processes and strains able to overcome this limitation and to improve the profitability of the microbiological production remains a relevant problem. A new P. acidipropionici FL-48 strain characterized by an increased resistance to 10 g/L of propionic acid (the number of viable cells after 24-h cultivation reached 1.05 × 106) was developed by a two-step induced mutagenesis using UV and diethyl sulphate from the P. acidipropionici VKPM B-5723 strain. The mutant strain exceeded the parental strain in the biomass accumulation rate and the amount of produced propionic and acetic acids by 35%, 20%, and 16%, respectively. The stability of such important characteristics as the biomass accumulation rate and the viability on media containing heightened concentrations of propionic acid was confirmed by three sequential monoclonal subculturings on a medium supplemented with 10 g/L of propionic acid. The optimization of the cultivation technology made it possible to determine the optimum seed inoculum dose (10% of the fermentation medium volume) and the best pH level for the active growth stage (6.1 ± 0.1). The scaling up of the fermentation to a 100-L bioreactor under observance of optimum cultivation conditions demonstrated a high biomass growth rate with a sufficient reproducability; after 20 h of fermentation, the number of viable cells in the culture broth exceeded 1 × 1010 CFU/mL. The new strain could be interesting as the component of silage and haylage biopreservatives and also could be used as an efficient producer of propionic acid.
