Background: Relli caste group was chosen for the present population genetic study, as it is one of the scheduled caste groups. The present work was designed to be a descriptive study mainly aiming at unveiling a precise population genetic profile of Relli caste. Methods: A community-based cross-sectional study was done among 428 Relli families belonging to two subsets (Relli-I: 208; Relli-II: 220) residing in different localities of Visakhapatnam city. The demographic information regarding age, occupation, economic status, marriage patterns and reproductive events were collected. The data was processed and analyzed by using software SPSS V.19. Results: The age composition in both the subgroups of Rellis indicates a high proportion of children and a low proportion of older people which are the common characteristics of a rapidly growing population. The overall consanguinity is more among Relli-II (42.3%) than among the Relli-I (36.9). The highest mean marital distance is recorded in Relli-I (37.9 km). The mean ages at marriage, first conception, and menopause are found to be slightly higher among the Relli-I. Whereas, the mean ages of menarche and last conception were higher in Relli-II. Tubectomy records the highest frequency in Relli-II group and vasectomy registered the highest frequency among Relli-I group. Conclusions: The demographic and reproductive structure of the Rellis in the present study indicates that observations made with regard to most of the variables fall well within the ranges observed for other caste populations of Andhra Pradesh.

INTRODUCTION MCL is an incurable disease and treatment approach should be adapted to patient's characteristics: age, PS, co-morbidities, social conditions, presence of caregiver, etc. In this study we describe the experience of RELLI in real life about old MCL pts followed in Latium region and diagnosed and registered in our database between January 2013 to December 2017. MATHERIALS and METHODS Data were collected in a regional data base. All new diagnosis of lymphoproliferative disease were considered medical history, clinical characteristics and lymphoma related characteristics were registered starting from an existing data base or medical records of single Institutions. RESULTS In the database were registered 91 pts with MCL (70M/21F) older than 65 years with a median age of 74 yrs (range 66-87). At diagnosis 11/91 (12.1%) were in stage I-II and 80/91 (87.9%) in stage III-IV; only 6 (6.6%) pts presented systemic symptoms. High levels of LDH were present in 45.1% of pts, at least one extranodal localization was reported in 4.4% and Ki67 > 30% in 48.5%. Prognostic score was evaluated at diagnosis: MIPI (LR 46.5%, IR 19.7%, HR 33.8%) and MIPI-c (LR 36.6%, ILR 22.5%, IHR 22.5%, HR 18.4%). Treatment was evaluated according to the age of pts: 65 -70 and >70 years; in the first group immuno-chemotherapy (ICT) was: Benda containing regimen 43.5% (R-BAC 17.4%, R-B 26.1%) and CHOP-like regimen 36.1% (R-CHOP21 17.4%, R-COMP21 8.7%). In contrast in older pts the choice of ICT was: Benda containing regimen 64% (R-BAC 14.8%, RB49.2%), and CHOP-like regimen 13.1% (R-CHOP21 8.2%, R-COMP 4.9%).The overall response rate (ORR), progression free-survival (PFS) and overall survival (OS) were calculated from the start of treatment and evaluated in the two groups of pts: ORR was 100% in younger (CR 69.6%, PR 30.4%) and 68.8% in older (CR 50.8%, PR 18%). According to the type of ICT, as expected, pts treated with bendamustine containing regimens (+/- Cytarabine) have better response and longer survival. With a median follow-up of 34.5 months, media OS of the entire population isn't reached and PFS is projected at 50% at 60 months. CONCLUSIONS In the era before new biologic drugs the approach to treatment of MCL was sufficiently homogeneous in the Lazio region. In real life Ky67 was principal factor influencing OS. MIPI and MIPI-c score divided the entire population into two groups at high and low risk. Age not change the OS but only the response rate to treatment (Figure 1). Abstract Receptor Expressed in Lymphoid Tissues (RELT) is a human TNFR superfamily member expressed most prominently in cells and tissues of the hematopoietic system. Mouse knockout studies indicate that RELT functions in part by negatively regulating T-cell activation. RELT has two homologous binding partners, RELL1 and RELL2, and collectively, these proteins are described as RELT family members. This study sought to identify novel protein interactions with RELT family members and to compare the expression of RELT in normal and diseased tissues. A yeast two-hybrid screen utilizing RELL1 as bait identified the cytoskeletal protein Filamin A (FlnA); physical interaction between FlnA and RELT family members was confirmed by in vitro co-immunoprecipitation. A truncated mutant of FlnA disrupts the ability of RELT family members to activate the p38 pathway. Previous reports indicate that autoantibodies against RELT are associated with breast cancer and that RELL2 prevents migration and invasion of the metastatic breast cancer cell line MDA-MB-231 (231), while multiple studies suggest a link between FlnA and breast cancer. We therefore sought to examine the association of RELT family members with breast cancer. Western blotting revealed significant expression of endogenous RELT family members in the breast cancer cell lines 231 and MCF-7. Immunohistochemistry (IHC) revealed a higher intensity of RELT staining in breast cancer tissues versus normal mammary tissue. Additionally, overexpression of RELT family members enhanced apoptosis in 231 cells. Collectively, these results further our understanding of signal transduction pathways associated with RELT family members and provide evidence that RELT is upregulated in breast cancer.

Abstract Receptor Expressed in Lymphoid Tissues (RELT) is a human Tumor Necrosis Factor Receptor (TNFR) that is expressed most prominently in cells and tissues of the hematopoietic system. RELT has two identified homologous binding partners RELL1 and RELL2. This study sought to further elucidate the function of RELT by identifying novel protein interactions with RELT family members. Human I-mfa domain-containing protein (HIC), also known as MDFIC, was identified in a yeast two-hybrid genetic screen using RELL1 as bait. HIC is a transcription factor that was initially identified to differentially regulate HTLV and HIV gene expression. HIC is now known to regulate many other cellular processes and the hic gene is located proximally to regions of chromosome 7 (7q31.1) frequently lost in Acute Myeloid Leukemia (AML) patients. HIC physically interacts with both RELT and RELL1, as determined by in vitro co-immunoprecipitations. HIC co-localizes with RELL1 at the plasma membrane and co-localizes with RELT in intracellular compartments. Deletion mutants of RELT were utilized to determine regions of RELT required for both the activation of p38 and the induction of apoptosis in HEK-293 cells, two previously described functions of RELT. Co-immunoprecipitation experiments utilizing the deletion mutants indicate that regions of RELT proximal to the plasma membrane are sufficient for physical interaction with HIC. Overexpression of RELT induces cleavage of PARP and Caspase-3 as determined by western blotting. Interestingly, induction of apoptotic morphology by RELT overexpression was not prevented if signaling by FADD or Caspase-8 was blocked, indicating RELT induces apoptosis by a pathway distinct from death-domain containing TNFRs such as TNFR1.

Abstract Nanoscopic, millisecond-lasting membrane platforms are modeled to constitute main functional signaling units. However, multiple signaling events in cell growth induction, including receptor Tyr-kinase (RTK) stimulation, calcium signaling, cytoplasmic kinase recruitment, operate over much broader space-time frames. Through dynamic confocal/super-resolution/electron-microscopy on living cells, we discovered that signaling competence is acquired through growth factor-induced coalescence of activated growth inducers (RTK, CD9, CD81, CD98, Co-029, CD316, Trop-1, Trop-2, PI3K, PI4K), and downstream effectors (Grb-2, PKCα, ERK, Akt, Src, Syk, ILK, ezrin) into confined segments of the cell membrane (‘docks’), that recursively assemble over hundreds of seconds. Average size (25±5 μm; ~80,000 individual analyses) and lifetime (222±25 sec) of docks extend by orders of magnitude previously recognized space-time frames for cell signaling. Activated/phosphorylated kinases clustered at membrane docks (99% P-Erb-B4; 91% P-Ret; 90% P-PKCα; 90% P-Src). STED super-resolution microscopy revealed up to 10 µm2 signal transducer areas of effector colocalization. Docks >1.0 µm2 accounted for ~90% of colocalizations versus ≈1% of raft-sized domains (<0.005 µm2). FRET-FLIM subsequently indicated that inter-molecular signal transducer interactions are essentially confined at docks. Docks were induced by growth factors (FGF-1, PDGF, EGF>HGF, IGF-1>>SCF/VEGF), were inhibited by Tyr kinase and cytoskeleton inhibitors, and were shown to induce cancer cell proliferation. RELT has two identified homologous binding partners, RELL1 and RELL2. This study sought to further elucidate the function of RELT by identifying novel protein interactions with RELT family members and to study the localization of RELT in both normal and diseased tissues. A yeast two-hybrid screen identifed Phospholipid Scramblase 1 (PLSCR1) and MyoD family inhibitor domain-containing protein (MDFIC) as potential RELT-family member binding proteins that were confirmed by in vitro co-immunoprecipitations. PLSCR1 has been demonstrated to possess anti-leukemic properties, and RELT expression results in an altered cellular localization of PLSCR1 as determined by immunofluorescence (IF). The MDFIC gene encodes for a transcription factor and is located proximally to regions of chromosome 7 (7q31.1) frequently lost in AML patients. MDFIC was observed to co-localize with RELL1 at the plasma membrane and co-localize with RELT in intracellular compartments as determined by IF. Since RELT, PLSCR1 and MDFIC are prominently expressed in the hematopoietic system, we sought to characterize the expression of RELT in both normal lymph nodes and B cell lymphomas. Immunohistochemistry revealed a higher intensity of RELT staining in germinal centers in comparison to surrounding lymph node regions. Interestingly, the level of RELT staining increased progressively in low and high-grade B cell lymphomas versus normal lymph nodes. Collectively, these results further our understanding of proteins that interact with RELT and identify an association of RELT with B cell lymphomas.

A yeast two-hybrid screen utilizing RELL1 as bait identified the cytoskeletal protein Filamin A (FLNA) as a prospective binding partner and physical interaction between FLNA and RELT family members was confirmed by in vitro co-immunoprecipitation. A truncated mutant of FLNA disrupts the ability of RELT family members to activate the p38 pathway and blocks the ability of RELT to induce death in human epithelial cells. Western blotting revealed endogenous RELT family members are most significantly upregulated in the breast cancer cell lines MDA-MB-231 and MCF-7, as well as the lung cancer cell line H358. Preliminary immunohistochemistry results reveal a higher intensity of RELT staining in both breast cancer and lung cancer human biopsies versus non-malignant tissue. Additionally, overexpression of RELT family members enhanced apoptosis in MDA-MB-231 and H358 cells as measured by X-gal morphology and luciferase assays. Collectively, these results further our understanding of signal transduction pathways associated with RELT family members and report the novel finding that RELT is upregulated in human breast and lung cancers. BACKGROUND: In 2014 we identified a new subset of DLBCL, defined as "IgM-secreting" (Cox MC & Di Napoli A , PLOS One 2014). This was characterised by poor prognostic features and outcome as well as frequent central nervous (CNS) system localizations. Furthermore, IgM-secretion, was an independent prognostic factor in multivariate analysis. Here we report on the largest series of IgM-secreting-DLBCL, from a multicentre Italian study. METHODS: The observational and biological study was approved by the Ethical Committee of the AUO Sant'Andrea, Italy. Enrolment criteria were: DLBCL with an associated IgM paraprotein diagnosed between 1st January 2010 and 31st December 2018 (IgM-secreting). Data were collected both prospectively and retrospectively from 17 Centres participating in the study. In addition, histopathology samples were centrally revised for immunohistochemistry (IHC) and FISH analyses. The control group (CTRL) consisted in a series of consecutive DLBCL, without an associated IgM-paraprotein (diagnosed between 01/01/2013 and 30/06/2016, enrolled in the Lymphoma Registry of the Lazio region (ReLLi Network). Last follow-up was carried out on 31st December 2019. RESULTS: 569 DLBCL cases were enrolled: 102 (17.9%) were IgM-secreting; 48 (8.4%) had a non-IgM paraprotein (IgA, IgG, or other), and 414 (72.7%) had no associated paraprotein (CTRL). IgM-secreting cases within the consecutive DLBCL patients enrolled in the ReLLi Registry were 41/466 (8.8%, 95CI 6.4-11.7%) while non IgM-paraprotein DLBCL cases were 11/466 (2.4%, 95CI 1.2-4.2%). The median level of IgM paraprotein was 17gr/L (range: <1-84gr/L); 83/102 (81.3%) were IgMk and 23/102 (22.5%) IgML respectively. The IgM-secreting group differed from the CTRL because the following characteristics were significantly more frequent: 1] age>60 (p=.001); 2] advanced stage (p<.001); 3] PS≥2 (p=.001); 4] LDH>UNL (p=.008) ; 5] ≥2 Extra-nodal sites involved (p<.001) ; 6] IPI 3-5 (p<.001); 7] central nervous system (CNS) involvement at diagnosis or relapse (p<.001); 8] lower rate of complete remission(CR) at the end of induction immunochemotherapy (p<.001). Conversely, no differences were observed for: sex, B-symptoms, HCV and HBV status, bulky disease, age≥80 years, and for transformation from low-grade lymphoma. PATHOLOGICAL AND MOLECULAR FEATURES: Paraffin tissue from 74 CTRL and 69 IgM-secreting was suitable for immunohistochemistry (IHC). The non-GCB subtype, based on Hans algorithm, was prevalent in the IgM-secreting (p=.005). No difference in BCL2 expression alone or in MYC and BCL2 double expression was observed within groups. In 48/63(76%; 95CI: 64-86%) IgM-secreting cases, both the IgM heavy and the corresponding kappa or lambda light chain protein expression were detected in the cytoplasm of the neoplastic clone. FISH analyses for MYC, BCL2 and BCL6 genes rearrangements performed in 25 IgM-secreting cases with either expression of MYC protein or a GC-phenotype showed no evidence of double or triple hits (DH/TH). TREATMENT: in the IgM-secreting group more patients were treated with RCOMP and with less intensive approach than the CTRL (p<.001). SURVIVAL: The median follow-up time was 46 months (95CI= 44-49; range 18-101) with 130 events and an incidence rate x100 person/year of 7.22 (95%CI 6.08-8.58) and a 5-yr OS of 76% (95CI 72-79%). The 5-PFS was 61% (95CI 57-65%). In univariable analysis age>60, B-symptoms, bulky disease, IPI >low risk and IgM-secreting IgM showed a worse survival (all with p<0.001). Also, the IgM-secreting group, showed a worse survival compared to the DLBCL with an associated IgG/IgA paraprotein (p<0.001). Adjusting in multiple Cox regression, IgM-secreting with IPI, gender, bulky and B-symptoms, maintain a higher risk of death either in the all cohort (HR 1.93, 95CI 1.34-2.78, p<0.001) or in patients with age<80 (HR 1.71, 95CI 1.16-2.54, p=0.007). Noteworthy, a survival sub-analysis showed that the 12/69 (17.4%) IgM-secreting with a GC-type had a better OS (9=0.008) and PFS (p=0.002) compared to the 57/69 (82.6%) IgM-secreting with a non-GC-type. CONCLUSION: Our data confirm that IgM-secreting DLBCL: 1) represents a sizable proportion of non-DH DLBCL; 2) have poor prognostic features and 3) have mostly a non-GC phenotype. Furthermore, IgM secretion appears to be an independent prognostic factor for both PFS and OS. Studies to define the biological features of this new subset are ongoing. This review highlights Receptor Expressed in Lymphoid Tissues (RELT), a Tumor Necrosis Factor Superfamily member, and its two paralogs, RELL1 and RELL2. Collectively, these three proteins are referred to as RELTfms and have gained much interest in recent years due to their association with cancer and other human diseases. A thorough knowledge of their physiological functions, including the ligand for RELT, is lacking, yet emerging evidence implicates RELTfms in a variety of processes including cytokine signaling and pathways that either promote cell death or survival. T cells from mice lacking RELT exhibit increased responses against tumors and increased inflammatory cytokine production, and multiple lines of evidence indicate that RELT may promote an immunosuppressive environment for tumors. The relationship of individual RELTfms in different cancers is not universal however, as evidence indicates that individual RELTfms may be risk factors in certain cancers yet appear to be protective in other cancers. RELTfms are important for a variety of additional processes related to human health including microbial pathogenesis, inflammation, behavior, reproduction, and development. All three proteins have been strongly conserved in all vertebrates, and this review aims to provide a clearer understanding of the current knowledge regarding these interesting proteins.

Pada makalah ini dilakukan evaluasi kinerja teknik pengodean konvolusi pada sistem komunikasi kooperatif dengan protokol amplify-and-forward (AF) dan menerapkan strategi seleksi relai dengan simulasi. Di sisi pemancar (sumber) diterapkan gabungan teknik reduksi peak to average power ratio (PAPR) skema selective mapping (SLM) dan predistorsi model Hammerstein. Di relai diterapkan juga teknik predistorsi model inverse Rapp. Di sisi kanal digunakan relai sebagai antena virtual. Penggunaan relai dalam sistem komunikasi kooperatif dapat diimplementasikan untuk jaringan 4G atau 5G pada penelitian selanjutnya, tetapi tentu membutuhkan bandwidth yang besar. Penerapan strategi seleksi relai mampu meningkatkan efisiensi bandwidth karena hanya relai terbaik saja yang akan meneruskan informasi dari pengirim ke penerima. Strategi seleksi relai jenis konvensional digunakan untuk mengevaluasi kinerja teknik pengodean konvolusi pada skema multirelai dengan memilih sebuah relai terbaik yang mempertimbangkan nilai signal to noise ratio (SNR) pada kanal sumber ke relai dan relai ke tujuan. Relai terbaik tersebut akan meneruskan sinyal dari sumber ke tujuan dengan menggunakan protokol AF. Kinerja sistem dinyatakan dalam nilai probabilitas bit error rate (BER). Dari hasil simulasi, dapat ditunjukkan bahwa teknik pengodean konvolusi mampu memperbaiki kinerja sistem hingga mencapai 16,59%, baik dengan menggunakan teknik predistorsi maupun tanpa teknik predistorsi. Kemudian, penerapan teknik predistorsi di sisi sumber dan relai mampu menghasilkan kinerja paling baik, yaitu kinerja sistem dapat meningkat hingga mencapai 34%. Selain itu, penerapan strategi seleksi relai jenis konvensional menunjukkan bahwa skema dengan relai terbanyak, yaitu enam relai, mampu menghasilkan kinerja yang paling baik disebabkan semakin banyaknya pilihan jalur yang tersedia.

Relai Arus Lebih merupakan peralatan yang mensinyalir adanya arus lebih, baik yang disebabkan oleh adanya gangguan hubung singkat yang dapat merusak peralatan sistem tenaga listrik yang berada dalam wilayah proteksinya. Relai Arus Lebih digunakan hampir pada seluruh pola pengamanan sistem tenaga listrik, dan dapat juga digunakan sebagai pengaman utama ataupun pengaman cadangan. Penelitian dilaksanakan pada Penyulang Beo Transformator-1 30 MVA 70/20 kV Gardu Induk Seduduk Putih Palembang. Hasil penelitian adalah saat gangguan hubung singkat 3 fasa, waktu kerja relai outgoing lebih cepat dibandingkan dengan waktu kerja relai incoming, dengan selisih waktu 0,7568. Kemudian saat gangguan hubung singkat 2 fasa, waktu kerja relai outgoing lebih cepat dibandingkan dengan waktu kerja relai incoming, dengan selisih waktu 0,9223. Besar kecilnya selisih waktu kerja tersebut dipengaruhi jauh dekatnya lokasi gangguan. Dimana semakin jauh lokasi gangguan, maka akan semakin besar selisih waktu kerja relai outgoing dengan waktu kerja relai incoming, dan apabila semakin dekat lokasi gangguan maka semakin kecil selisih waktu kerja relai outgoing dengan waktu kerja relai incoming. Hal ini bertujuan memberi kesempatan pada relai outgoing untuk bekerja terlebih dahulu sebagai pengaman utama (main protection) apabila terjadi gangguan hubung singkat di penyulang, dan relai incoming akan bekerja sebagai pengaman cadangan (back-up protection) apabila relai outgoing tidak bekerja. Hasil perhitungan dengan data yang ada dilapangan masih dalam kondisi yang sesuai dengan perbedaannya tidak terlalu jauh, sehingga dapat disimpulkan bahwa secara keseluruhan kinerja Relai Arus Lebih yang ada dilapangan masih dalam kondisi baik.

Relai Rate of Change of Frequency (ROCOF) bekerja berdasarkan kecepatan perubahan frekuensi (df/dt) yang merupakan indikasi langsung dari ketidakseimbangan beban dengan pembangkit pada sistem. Dengan karakteristik ini pemanfaatan relai ROCOF sebagai relai pelepasan beban sangat menjanjikan. Pada penelitian ini bertujuan untuk menganalisa kemampuan skema relai ROCOF untuk pemanfaatan sebagai relai pelepasan beban frekuensi rendah. Skema relai ROCOF yang diusulkan terdiri dari lima tahap pelepasan dan disupervisi oleh relai frekuensi untuk mencegah maloperasi pelepasan beban saat kejadian hubung singkat. Skema relai ROCOF diuji melalui simulasi pelepasan pembangkit dengan kontingensi N-1, N-2 dan N-2-1. Simulasi dilakukan menggunakan Digsilent Powerfactory dan dibandingkan dengan skema UFR. Untuk kontingensi N-1 dan N-2, hasil simulasi menunjukkan skema relai ROCOF dapat mengembalikan frekuensi ke nilai yang lebih baik dan dalam waktu yang lebih cepat dari pada skema UFR. Namun terdapat permasalahan dalam pengaturan sensitifitas relai ROCOF melalui setting delay waktu. Untuk kondisi kekurangan pembangkitan ringan dan sedang dibutuhkan setting delay yang cukup besar untuk memperoleh nilai df/dt yang sebenarnya dari nilai df/dt yang cenderung berosilasi. Namun untuk kekurangan pembangkitan yang parah, setting delay yang besar menyebabkan relai menjadi kurang sensitif sehingga sebagian kecil relai tidak bekerja pada semua tahap yang diharapkan.

Background. Receptor expressed in lymphoid tissues-like 2 (RELL2), which is a member of RELT family, is closely associated with the plasma membrane and acts as a modulator for RELT signaling. Overexpression of RELL2 induces the activation of MAPK14/p38 cascade and apoptosis. However, whether RELL2 contributes to cancers remains unclear. Here, we examined its role in cancer patient prognosis and various tumors. Methods. We used several bioinformatics methods, specifically gene set enrichment analysis (GSEA), ScanNeo, and ESTIMATE, to analyze the CCLE dataset, GTEx dataset, and TCGA dataset. We investigated the possible association of RELL2 with the microsatellite instability (MSI) of various tumors, tumor mutational burden (TMB), immune checkpoint, immune neoantigens, immune microenvironment, and patient prognosis. Result. RELL2 is highly expressed in cancer compared with normal tissues. RELL2 expression is linked with worse progression-free interval and overall survival in numerous cancers. In most cancers, high RELL2 expression was related to a poor prognosis. RELL2 expression was significantly associated with the tumor microenvironment, MSI, and TMB. RELL2 expression is strongly associated with phenotypes that are of major clinical significance, particularly those associated with immune neoantigens and the expression profiles of immune checkpoint genes in pan-cancer. RELL2 expression strongly linked with the expressions of methyltransferases and DNA repair genes. It also significantly correlated with multiple signaling pathways through gene set enrichment analysis. Conclusion. RELL2 may be a prognostic biomarker in pan-cancer and may have an important function in tumorigenesis and progression.

Karena relai diferensial merupakan proteksi utama pada transformator maka dari itu penulis melakukan studi penggunaan relai diferensial sebagai sistem proteksi transformator agar settingan relai tersebut tetap dalam keadaan normal tanpa gangguan. Hasil penelitian ini menunjukkan sistem kerja relai diferensial pada Pembangkit Listrik Tenaga Uap (PLTU) Barru yaitu berdasarkan pada besaran arus gangguan di luar zona. Apabila berdasarkan pada besaran nominal maka arus besar yang melewati sekunder transformator akan merusak peralatan serta setting slope yang tepat untuk relai diferensial agar dalam keadaan normal pada sistem proteksi Pembangkit Listrik Tenaga Uap (PLTU) Barru sesuai pada besaran arus gangguan di luar zona yaitu sebesar k = 0,533 sampai k = 2.

Oleh karena itu digunakan relai gangguan tanah atau ground fault relay pada suatu peralatan, salah satunya pada feeder. Perhitungan setting kerja relai gangguan tanah di dapatkan dari nilai setting yang di terapkan dan karakteristik waktu yang digunakan. Setelah dilakukan perhitungan dan menganalisa didapatkan perbedaan atau perselisihan waktu kerja relai. Intron retention plays an important role in cancer progression and chemotherapy resistance and seems to be essential for the maintenance of genome stability in cancer. Here, our goal was to analyze the role of receptor expressed in lymphoid tissue (Relt)-like 2 (RELL2) intron 4 retention in promoting pancreatic ductal adenocarcinoma (PDAC) progression. Our results showed that intron retention (IR) occurs at the fourth intron of RELL2 transcript in gemcitabine resistant PDAC cells, however, the regulatory mechanism and the clinical implications of IR of RELL2 are unclear. Firstly, we found that RELL2 plays an anti-oncogenic role in PDAC by performing in vitro functional assays including cell proliferation, GEM cytotoxicity assay and apoptosis. Subsequently, we identified the upstream gene of RELL2, DEAH-Box Helicase 38 (DHX38), and demonstrated the direct interaction between DHX38 and RELL2 by RIP-qPCR. We also found that altered expression of DHX38 resulted in corresponding changes in intron 4 retention of RELL2. Importantly, we unveiled that overexpression of DHX38 on the basis of knocking down of the fourth intron of RELL2 resulted in an impaired intron 4 intention. Overall, our study identified a new IR site in PDAC, which could be a possible target for PDAC therapy.

Pada sistem distribusi biasanya terjadi black out (sistem kelistrikan mati) diakibatkan karena kesalahan koordinasi antara pengaman penyulang masuk sebagai pengaman cadangan dengan salah satu penyulang keluar sebagai pengaman utama, sehingga jika di salah satu penyulang mengalami gangguan maka penyulang yang lainnya juga ikut terganggu. Untuk itu diperlukan koordinasi proteksi yang sensitif, cepat dan selektif pada penyulang masuk dan penyulang keluar. Pada penelitian ini, dilakukan perhitungan arus hubung singkat untuk melihat pengaruh panjang jaringan terhadap arus hubung singkat, serta menentukan setelan relai untuk mengetahui kondisi koordinasi antara pengaman penyulang keluar dengan penyulang masuk berdasarkan waktu kerja relai terhadap arus gangguan yang terjadi pada penyulang Gardu Induk 20 kV Marisa Dari hasil penelitian didapat bahwa arus gangguan hubung singkat terbesar adalah arus gangguan 3 fasa di penyulang keluar MR 4 jika dibandingkan dengan penyulang keluar lainya. Kondisi koordinasi antara relai arus lebih (OCR) penyulang masuk MR 2.1 dengan relai arus lebih (OCR) penyulang keluar MR 1 dan MR 6 tidak begitu bagus, karena penyulang MR 1 dan MR 6 nilai arus gangguan di tengah-tengah jaringan lebih kecil dari setelan arus (Iset) OCR MR 2.1 sehingga membuat relai OCR MR 2.1 sebagai pengaman cadangan tidak dapat berkerja jika pengaman utama gagal berkerja pada saat terjadi gangguan hubung singkat di ujung jaringan. Sedangkan untuk koordinasi MR 2.1 dengan MR 3 dan MR 4 sudah bagus karena relai penyulang masuk dapat berkerja pada saat gangguan di ujung jaringan dan relainya tidak berkerja secara bersamaan. Adapun kondisi koordinasi antara relai gangguan tanah (GFR) penyulang masuk MR 2.1 dengan relai gangguan tanah (GFR) penyulang keluar MR 1, MR 3, MR 4, MR 6 sudah bagus karena gambar kurvanya tidak saling berpotongan sehingga relai tidak akan berkerja secara bersamaan dan memiliki waktu kerja relai yang cepat untuk memerintahkan pemutus tenaga (PMT) berkerja.

Makalah ini bertujuan melakukan investigasi terhadap hasil pengukuran impedansi gangguan lingkar tertutup oleh algoritma relai jarak konvensional (tipe distance relay SEL-421) ketika digunakan sebagai alat proteksi pada saluran transmisi dengan kompensator seri dan sejumlah parameter ketidakpastian (faktor). Sejumlah faktor dari sistem dapat muncul secara bersamaan dan keberadaan kompensator seri dapat memengaruhi kinerja algoritma relai, terutama pada gangguan fase ke tanah. Sejumlah investigasi terhadap kinerja relai telah dilakukan. Namun, metode pengujian yang dikembangkan hanya mengubah satu faktor, dengan faktor lain dijaga konstan pada waktu bersamaan. Teknik ini tidak lagi relevan ketika tidak mempertimbangkan sejumlah faktor secara bersamaan, sehingga dapat memengaruhi kinerja relai saat terjadi gangguan. Untuk keperluan investigasi algoritma seperti dalam kondisi nyata, sejumlah skenario gangguan perlu dilakukan pada titik gangguan sebelum dan setelah kompensator seri, sambil sekaligus melakukan perubahan nilai dari sejumlah faktor pada model sistem secara bersamaan melalui simulasi gangguan. Pada penelitian ini, perangkat lunak DIgSILENT PowerFactory digunakan dalam pemodelan sistem daya dan simulasi gangguan. Rangkaian ekivalen Thevenin dengan dua sumber dan kompensator seri sebesar 42% diletakkan di tengah saluran transmisi sepanjang 300 km dengan tegangan sistem 400 kV untuk simulasi pengujian gangguan. Sejumlah skenario gangguan dan perhitungan impedansi gangguan sebagai fungsi perubahan nilai dari sejumlah faktor dilakukan secara otomatis. Simulasi pengujian otomatis dikembangkan melalui DIgSILENT Programming Language (DPL) untuk melakukan pembacaan sampel data yang dibangkitkan melalui perangkat lunak SIMLAB untuk sejumlah faktor. Kinerja algoritma relai untuk perhitungan impedansi gangguan tidak dipengaruhi oleh keberadaan kompensator seri ketika gangguan terjadi sebelum kompensator, tetapi sebaliknya ketika gangguan terjadi setelah kompensator. Pengubahan sejumlah faktor secara bersamaan memengaruhi akurasi relai dan menyebabkan kinerja relai memburuk untuk gangguan setelah kompensator, yaitu terjadinya kegagalan operasi relai berupa underreaching dan overreaching. Teknik pengujian yang dikembangkan diharapkan dapat digunakan sebagai alat bantu pengujian terkini untuk pengembangaan dan implementasi relai secara tepat dan seperti pada kondisi nyata.

Daya listrik yang disalurkan melalui sistem distribusi harus memiliki mutu dan keandalan yang tinggi. Dalam penyaluran energi listrik sering terjadi gangguan hubung singkat. Gangguan hubung singkat merupakan suatu hubungan abnormal pada impedansi antara dua titik yang mempunyai potensial yang berbeda. Tujuan dari penelitian ini adalah menghitung arus hubung singkat, menentukan koordinasi kerja relai terhadap gangguan hubung singkat. Metode penelitian dilakukan dengan berbagai cara yaitu studi literature, obsevasi, dan wawancara. Hasil penelitian ini besarnya arus gangguan hubung singkat dipengaruhi jarak titik gangguan,semakin jauh jarak titik gangguan maka semakin kecil arus gangguan dan begitupun sebaliknya. Hasil perhitungan manual dan simulasi software ETAP 12.6 di dapatkan hasil perhitungan gangguan hubung singkat tiga fasa, dua fasa dan satu fasa sebesar 1147,99 A, 994,19 A dan 613,34 A dan hasil simulasi 1111 A, 962 A, dan 646 A. Penyetelan Over Current Relay (OCR) dan Ground Fault Relay (GFR) pada penyulang golf yaitu, TMS = 0,25 s untuk relai OCR sisi incoming 20 kV, TMS = 0,18 s untuk relai Over Current Relay (OCR) sisi penyulang 20 k, TMS = 0,23 s untuk relai Ground Fault Relay (GFR) sisi incoming 20 kV, TMS = 0,17 s untuk relai GFR sisi incoming 20 kV dan hasil perhitungan manual maupun simulasi memperlihatkan setting OCR GFR masih dalam kondisi yang sesuai dimana relai di penyulang sebagai main protection dan relai di incoming sebagai backup protection.

Pekerjaan rekonduktoring Tower 26 – 46 Ungaran Mranggen line 2 yang menyebabkan perubahan konduktor dari tipe ACSR (Alumunium Conductor Steel Reinforced) 240/30mm2 menjadi TACSR ( Thermal Alumunium Conductor Steel Reinforced) 410/67 mm2, sehingga terjadi peningkatan kemampuan hantar kawat penghantar menjadi 1600 A. Tujuan dari penelitian ini yaitu resetting ulang relai GFR (Ground Fault Relay) dan OCR (Over Current Relay) merupakan suatu relai yang bekerjanya berdasarkan adanya kenaikan arus yang melebihi suatu nilai pengaman tertentu dalam jangka waktu tertentu, sehingga relai ini dapat dipakai sebagai pola pengaman arus lebih. Perubahan ini berpengaruh dalam setting arus lebih dan relai gangguan fasa ke tanah sehingga nilai setting nya menjadi 1920 A dan 320A, nilai impedansi ekuivalen juga berubah untuk nilai total urutan positifnya menjadi 1.6625+12.29656j W , impedansi urutan negatif 1.6625+12.29656j W, dan untuk urutan nol 1.65842+11.75236j W, dari perubahan impedansi ini mempengaruhi nilai arus hubung singkatnya yang menjadi menyebabkan waktu bekerja relai juga berubah menjadi lebih lambat oleh karena itu dilakukan seting ulang relai pada Bay Mranggen 2 di GI Ungaran.

Salah satu tindakan kriminalitas yang masih sering terjadi adalah pencurian kendaraan sepeda motor. Teknik utama yang dilakukan adalah dengan merusak bagian kunci kontak. Maka dari itu, solusi pengamanan sepeda motor dapat dilakukan dengan mengganti kunci kontak sepeda motor dengan menggunakan modul relai yang dapat dikontrol secara jarak jauh serta memiliki fitur pelacakan. Pada penelitian ini, direalisasikan sebuah sistem yang dapat mengamankan dan mengontrol kelistrikan sepeda motor dari jarak jauh serta melacak keberadaan sepeda motor. Sistem ini dibuat dengan menggunakan mikrokontroler Arduino Uno sebagai pengontrol sistem yang dapat mengatur relai sebagai saklar ON-OFF pada jalur ignition key sepeda motor. Sistem juga dapat mengirimkan data lokasi sepeda motor serta terhubung dengan aplikasi Android untuk melakukan kontrol relai dan pelacakan menggunakan global positioning system (GPS). Pengujian kontrol relai dilakukan 10 kali pengujian dengan masing-masing jarak pengujian yang berbeda. Alat ini dapat mengontrol relai dari jarak jauh tanpa batasan jarak dengan tingkat keberhasilan hingga 70%. Pada pengujian pembacaan GPS, didapatkan nilai error pembacaan rata-rata sebesar 2,45 meter dan sistem dapat mengirim data serta mengakses data dari Firebase. Aplikasi dapat melakukan akses data lokasi dan menampilkannya dalam bentuk maps serta mengirimkan perintah ON-OFF ke Firebase.

E/L Repli-seq is a powerful tool for detecting cell type-specific replication landscapes in mammalian cells, but its potential to monitor DNA replication under replication stress awaits better understanding. Here, we used E/L Repli-seq to examine the temporal order of DNA replication in human retinal pigment epithelium cells treated with the topoisomerase I inhibitor camptothecin. We found that the replication profiles by E/L Repli-seq exhibit characteristic patterns after replication-stress induction, including the loss of specific initiation zones within individual early replication timing domains. We also observed global disappearance of the replication timing domain structures in the profiles, which can be explained by checkpoint-dependent suppression of replication initiation. Thus, our results demonstrate the effectiveness of E/L Repli-seq at identifying cells with replication-stress-induced altered DNA replication programs.

Abstract Receptor Expressed in Lymphoid Tissues (RELT) is a Tumor Necrosis Factor Receptor family member implicated in several cancers. RELL1 and RELL2 (RELT-Like 1 and 2) are RELT homologs that bind RELT; the three proteins are collectively referred to as RELT family members (RELTfms). We sought to evaluate the role of the OXSR1 kinase and actin-binding protein Filamin A (FLNA) in modulating RELT-induced apoptosis of cancer cells. We also assessed RELTfm expression in human breast and lung cancer biopsies and their ability to sensitize breast cancer cells to chemotherapeutic agents. Methods: Empty vector or expression plasmids for RELTfms were transiently transfected into MDA-MB-231 (231) breast cancer cells with TransfeX reagent. Cell death was assessed by morphology assays and ATP luciferase assays. Apoptosis of 231 cells was assessed by Annexin V/PI staining using Flow cytometry (FC). Immunohistochemistry (IHC) was used to assess RELT expression in human breast and lung cancer biopsies. IHC and western blotting were utilized to determine RELT cellular localization. Results: All three RELLfms were co-immunoprecipitated by a C-terminal fragment of FLNA. RELT overexpression induced death in 231 cells, and this effect was increased with co-transfection of a FLNA S215A mutant, an important phosphorylation site that protects FLNA from cleavage. 231s transfected with RELT and RELL2 demonstrated increased Annexin V/PI signal in comparison to vector control, demonstrating that these RELTfms induce death via an apoptotic pathway. Co-transfection of a mutated plasmid for the OXSR1 kinase did not abrogate RELT-induced apoptosis. A RELT construct with a mutated binding site for the OXSR1 kinase induced apoptosis to a similar extent as wild-type RELT. An increase in doxorubicin-induced apoptosis was observed when 231 cells were transfected with RELL2 versus vector control. IHC results showed increased RELT expression in malignant breast cancer biopsies compared to patient-matched benign tissue. Interestingly, RELT was localized in the nucleus of some malignant lung cancer biopsies, and western blot analysis indicates that RELT translocates to the nucleus and interacts with chromatin in 231 cells. Conclusions: Collectively, these results demonstrate that RELTfms induce apoptosis of breast cancer cells, and potentially increase the sensitivity of breast cancer cells to chemotherapeutic agents. Furthermore, this work enhances our understanding of the interaction between RELT and FLN, a protein that is implicated in many cancers, including breast cancer. Phosphorylation by OXSR1 was previously shown to be required for RELT to activate the p38 pathway, yet the OXSR1 kinase is not required for RELT-induced death. RELT expression is enhanced in breast cancer biopsies, and interestingly, RELT can localize to the nucleus in cancerous cells. Collectively, these findings significantly enhance our understanding of RELTfms and cancer. Introducción. En la leishmaniasis visceral (LV), del viejo mundo causada por Leishmania donovani, el factor de elongación-1α (EF-1α), es un antígeno-activador de la fosfatasa SHP-1 que inhibe la actividad leishmanicida de macrófagos. Sin embargo, la relevancia del EF-1α de L. infantum en la LV del nuevo mundo aún es desconocida. Objetivo. Obtener la proteína recombinante del EF-1α de L. infantum (rEFLi). Material y métodos. Se clonó, secuenció y expresó como proteína recombinante el gen EFLi en un sistema procarionte. rEFLi fue purificada y se verificó su inmunogenicidad. Resultados. El gen EF-Li posee 1350 pb, con 25 diferencias nucleotídicas, 10 diferencias aminoacídicas y sin diferencias estructurales (3D), en comparación con EFLd. rEFLi (50kDa) fue reconocida en sueros de ratones inmunizados por su construcción genética homóloga. Conclusión. Nuestras evidencias sugieren que rEFLi puede evaluarse como vacuna biotecnológica sola o en combinación, para la prevención de la LV.

One of the major problems in the U.S. and global cotton production is the damage caused by the reniform nematode, Rotylenchulus reniformis. Amplification of DNA from single nematodes for further molecular analysis can be challenging sometimes. In this research, two whole-genome amplification (WGA) methods were evaluated for their efficiencies in DNA amplification from a single reniform nematode. The WGA was carried out using both REPLI-g Mini and Midi kits, and the GenomePlex single cell whole-genome amplification kit. Sequence analysis produced 4 Mb and 12 Mb of genomic sequences for the reniform nematode using REPLI-g and SIGMA libraries. These sequences were assembled into 28,784 and 24,508 contigs, respectively, for REPLI-g and SIGMA libraries. The highest repeats in both libraries were of low complexity, and the lowest for the REPLI-g library were for satellites and for the SIGMA library, RTE/BOV-B. The same kind of repeats were observed for both libraries; however, the SIGMA library had four other repeat elements (Penelope (long interspersed nucleotide element (LINE)), RTE/BOV-B (LINE), PiggyBac, and Mirage/P-element/Transib), which were not seen in the REPLI-g library. DNA transposons were also found in both libraries. Both reniform nematode 18S rRNA variants (RN_VAR1 and RN_VAR2) could easily be identified in both libraries. This research has therefore demonstrated the ability of using both WGA methods, in amplification of gDNA isolated from single reniform nematodes.

Se proponen algunas lecturas nuevas e integraciones en sellos reales hititas de Boğazköy. Probablemente no existió la reina * Malnigal, Malni sería más bien un término de augurio del léxico hitita-luvio. La impronta del sello SBo I 36 = HGB I 219 debe ser atribuida a Šuppiluliuma. En el sello Maṣat 75.10 el nombre de la reina que acompaña al monograma de Tudḫaliya III es Taduḫepa y no *Šatatuḫepa. Su nombre es Taduḫepa en la leyenda jeroglífica y es confirmado por los trazos cunei­formes en la corona circular.
