Der volle Inhalt der QuelleInflammatory bowel diseases (IBD) are gastrointestinal tract pathologies characterized by recurrent episodes of inflammation with periods of remission, and symptoms that negatively affect patients’ quality of life. Treatment of these complex diseases currently relies mainly on the oral administration of chemical molecules. Nevertheless, the therapeutic arsenal remains limited, and surgical intervention often remains inevitable for the most severe forms. In recent years, polysaccharides (PS) have aroused major interest for their ability to modulate intestinal inflammation and extend the crisis-free periods. Their combination with active substances has opened new prospects for the development of innovative and targeted treatments for IBD. In this context, the overall objective of the present thesis was to develop a PS-based medical form of a pro-resolving drug candidate to promote the healing and local tissue repair of damaged intestinal tissues. The first part of this thesis was devoted to the development of a new in vitro preclinical tool designed to study the effects of bioactive excipients on intestinal inflammation. The second part involved identifying PS compounds with anti-inflammatory activity for inclusion in the future drug formulation. The third focuses on the design and evaluation, in vitro and in vivo, of the final form of the drug candidate. These results highlighted the potential of hypromellose (HPMC) as bioactive excipient and its additive anti-inflammatory effect in the biological activity of the drug candidate. Although further investigations are required to better understand its mechanism of action, all this work underlines the interest of HPMC as a potential drug or adjuvant in the prevention and treatment of IBD
