Der volle Inhalt der QuelleColor is a key biological trait that allows individuals to adapt to their biotic and abiotic environment, and many genes have been shown to play an essential role in color acquisition in vertebrates. In this work, the consequences on this specifie set of genes of the whole-genome duplications that occurred during vertebrate evolution were investigated. Whereas most genes a re lost after a w hole- genome duplication event, this work shows that pigmentation genes and other genes within their families have been more retained compared to the average observed genome retention. This was particularly the case for the teleost-specific whole-genome duplications . Hence, this high retention rate could be related to the high pigmentation diversity observed in the teleost fish lineage, both in terms of patterns and of pigment cell types . In addition, during this PhD thesis, pigmentation in an emerging model organism, the clownfish Amphiprion ocellaris, was investigated. The white color in clownfish was shawn to be due to specifie pigment cells, iridophores, and new iridophore genetic markers were detected . One such marker is almost exclusively restricted to ray-finned fish and could have been acquired through a horizontal gene transfer event that occurred at the onset of the evolution of this lineage. Last, in this work, the genetic basis of pigmentation acquisition was scrutinized during the development of clownfish A. ocellar is, and especially during metamorphosis - a major life history transition in coral reef fish . In particular, the deployment during development of thyroid hormone signaling which is a key pathway in metamorphosis regulation in vertebrates was examined

Der volle Inhalt der QuelleMost of the studies conducted on teleost fish metamorphosis have used freshwater fish species or marine fish species of commercial interest. Even though those studies allowed better understanding the role of THs in the regulation of metamorphosis and how they control the expression of several target genes, there is little information on the role of the environment on the regulation of this important post-embryonic development step. However, environment is able to regulate metamorphosis as it has been revealed in the coral reef fish A.triostegus. This species is an excellent ecological model but as we cannot have access to its entire larval life cycle, it is difficult to study how environmental factors interact with THs signaling pathway. By contrast, the clownfish A. ocellaris could be an emerging model species as its entire life cycle can be obtained under laboratory conditions. This species has the potential to tackle questions regarding ecology, evolution and developmental biology. The objective of this thesis was to develop the clownfish A. ocellaris as a model species to study metamorphosis in coral reef fishes and gain insight on the role of environmental factors in this process. For that purpose, A. ocellaris metamorphosis has been characterized at the morphological, endocrinological and molecular levels by tackling the following questions: What are the morphological changes occurring during the post-embyronic development of A. ocellaris ? What are the molecular players of A. ocellaris metamorphosis ? Is the metamorphosis of A. ocellaris under the control of THs and which changes (morphological, physiological, behavioral and molecular) are under their control ?

Der volle Inhalt der QuelleThe evolutionary potential of wild populations to respond to environmental change will determine their capacity to adapt and survive. However, in order to achieve evolutionary change, variation in the contribution of an individual to the next generation — its fitness — must have a genetic basis and be heritable. The study of the evolutionary potential of populations requires longitudinal and relatedness data with different environments to partition the contribution of genes, maternal effects and environment on fitness. Estimates of genetic heritability of fitness traits are extremely rare, especially for marine species, where inter-generational relationships are rarely known. Here, we present the first multi-generational pedigree for a marine fish population by repeatedly genotyping all individuals in a population of the orange clownfish (Amphiprion percula) at Kimbe Island (Papua New Guinea) over a 10-year period. Based on 2927 individuals, our pedigree analysis revealed that longitudinal philopatry was recurrent over five generations. We show that local reproductive success has a very low (<1%) but significant heritability. We also show that additive genetic variation and maternal effects on local fitness are both extremely low (<1%). Habitat is the major driver that explain differences in the contribution of individuals to the next generation in the local population (until 96.5%). Together these results suggest that while the environment imposes strong selection pressures on the local fitness, the low heritability indicates the orange clownfish population has little evolutionary potential to adapt to local environmental changes
