A significant feature of spiking neural networks with varying connection delays, such as those in the brain, is the existence of strongly connected groups of neurons known as polychronous neural groups (PNGs). Polychronous groups are found in large numbers in these networks and are proposed by Izhikevich ( 2006a ) to provide a neural basis for representation and memory. When exposed to a familiar stimulus, spiking neural networks produce consistencies in the spiking output data that are the hallmarks of PNG activation. Previous methods for studying the PNG activation response to stimuli have been limited by the template-based methods used to identify PNG activation. In this letter, we outline a new method that overcomes these difficulties by establishing for the first time a probabilistic interpretation of PNG activation. We then demonstrate the use of this method by investigating the claim that PNGs might provide the foundation of a representational system.

This paper analyses the feasibility of using the cutaneous saphenous nerve (CSN) as a source of peripheral nerve graft (PNG). In 34 fresh dog cadavers, the PNG length (PNGL) was measured, pattern of collateralization was studied, fascicles diameter and number were assessed at multiple levels along the length of the CSN (levels of section A, B, C and D) and surgical approach was described. The PNGL was correlated to femoral length. The pattern of CSN collateralization was consistent for all but 2 of the dogs. The average cross section diameter and fascicle number of the CSN was 2.0 to 0.1 mm (1 to 7 fascicles). CSN provides a large-sized, easily accessible nerve graft that could be used to bridge nerve gaps in the dog.

We have previously identified and purified a novel β-glucosidase, designated PNGH (pyridoxine-5´-β-d-glucoside hydrolase), from the cytosolic fraction of pig intestinal mucosal. PNGH catalyses the hydrolysis of PNG (pyridoxine-5´-β-d-glucoside), a plant derivative of vitamin B6 that exhibits partial nutritional bioavailability in humans and animals. Preliminary amino acid sequence analysis indicated regions of close similarity of PNGH to the precursor form of LPH (lactase–phlorizin hydrolase), the β-glucosidase localized to the brush-border membrane. We report in the present study amino acid sequence data for PNGH and results of Northern blot analyses, upon which we propose a common genomic origin of PNGH and LPH. Internal Edman sequencing of the PNGH band isolated by SDS/PAGE yielded data for 16 peptides, averaging 10.8 amino acids in length. These peptides from PNGH (approx. 140 kDa) were highly similar to sequences existing over most of the length of the >200 kDa precursor of rabbit LPH; however, we found no PNGH sequences that corresponded to approx. 350 amino acids between positions 463 and 812 of the LPH precursor, a region encoded by exon 7 of the LPH precursor gene (amino acids 568–784), and no sequences that corresponded to regions near the N-terminus. MS analysis of tryptic peptides yielded 25 peptides, averaging 15 amino acids, with masses that matched segments of the rabbit LPH precursor. Northern blot analysis of pig and human small intestinal polyadenylated mRNA using a non-specific LPH cDNA probe showed an expected approx. 6 kb transcript of the LPH precursor, but also an approx. 4 kb transcript that was consistent with the size predicted from the PNGH protein mass. Using a probe specific to the region encoded by exon 7, hybridization occurred only with the 6 kb transcript. Based on these observations, we propose that both PNGH and LPH enzymes have the same genomic origin, but differ in transcriptional and, possibly, post-translational processing.

ABSTRACT We have analyzed changes to proviral Env gp120 sequences and the development of neutralizing antibodies (NAbs) during 1 year of simian/human immunodeficiency virus SHIV-89.6P infection in 11 Macaca nemestrina macaques. Seven macaques had significant env divergence from that of the inoculum, and macaques with greater divergence had higher titers of homologous NAbs. Substitutions in sequons encoding potential N-linked glycosylation sites (PNGs) were among the first to be established, although overall the total number of sequons did not increase significantly. The majority (19 of 23) of PNGs present in the inoculum were conserved in the sequences from all macaques. Statistically significant variations in PNGs occurred in multiple macaques within constrained regions we term “hot spots,” resulting in the selection of sequences more similar to the B consensus. These included additions on V1, the N-terminal side of V4, and the outer region of C2. Complex mutational patterns resulted in convergent PNG shifts in V2 and V5. Charge changes in Env V1V2, resulting in a net acidic charge, and a proline addition in V5 occurred in several macaques. Molecular modeling of the 89.6P sequence showed that the conserved glycans lie on the silent face of Env and that many are proximal to disulfide bonds, while PNG additions and shifts are proximal to the CD4 binding site. Nonsynonymous-to-synonymous substitution ratios suggest that these changes result from selective pressure. This longitudinal and cross-sectional study of mutations in human immunodeficiency virus (HIV) env in the SHIV background provides evidence that there are more constraints on the configuration of the glycan shield than were previously appreciated.

Intra-axonal localization of mRNAs and protein synthesis machinery (PSM) endows neurons with the capacity to generate proteins locally, allowing precise spatiotemporal regulation of the axonal response to extracellular stimuli. A number of studies suggest that this local translation is a promising target to enhance the regenerative capacity of damaged axons. Using a model of central nervous system (CNS) axons regenerating into intraspinal peripheral nerve grafts (PNGs) we established that adult regenerating CNS axons contain several different mRNAs and protein synthetic machinery (PSM) components in vivo. After lower thoracic level spinal cord transection, ascending sensory axons regenerate into intraspinal PNGs but axon growth is stalled when they reach the distal end of the PNG (3 versus 7 weeks after grafting, resp.). By immunofluorescence with optical sectioning of axons by confocal microscopy, the total and phosphorylated forms of PSMs are significantly lower in stalled compared with actively regenerating axons. Reinjury of these stalled axons increased axonal localization of the PSM proteins, indicative of possible priming for a subcellular response to axotomy. These results suggest that axons downregulate protein synthetic capacity as they cease growing, yet they retain the ability to upregulate PSM after a second injury.

ABSTRACT Three Saccharomyces cerevisiae proteins (Yng1/YOR064c, Yng2/YHR090c, and Pho23) and two Schizosaccharomyces pombeproteins (Png1/CAA15917 and Png2/CAA21250) share significant sequence identity with the human candidate tumor suppressor p33ING1in their C-terminal regions. The homologous regions contain PHD finger domains which have been implicated in chromatin-mediated transcriptional regulation. We show that GFP-Yng2, like human Ing1, is localized in the nucleus. Deletion of YNG2 results in several phenotypes, including an abnormal multibudded morphology, an inability to utilize nonfermentable carbon sources, heat shock sensitivity, slow growth, temperature sensitivity, and sensitivity to caffeine. These phenotypes are suppressed by expression of either human Ing1 or S. pombe Png1, suggesting that the yeast and human proteins are functionally conserved. Yng1- and Pho23-deficient cells also share some of these phenotypes. We demonstrated by yeast two-hybrid and coimmunoprecipitation tests that Yng2 interacts with Tra1, a component of histone acetyltransferase (HAT) complexes. We further demonstrated by coimmunoprecipitation that HA-Yng1, HA-Yng2, HA-Pho23, and HA-Ing1 are associated with HAT activities in yeast. Genetic and biochemical evidence indicate that the Yng2-associated HAT is Esa1, suggesting that Yng2 is a component of the NuA4 HAT complex. These studies suggest that the yeast Ing1-related proteins are involved in chromatin remodeling. They further suggest that these functions may be conserved in mammals and provide a possible mechanism for the human Ing1 candidate tumor suppressor.

Object Facial nerve injury results in facial palsy that has great impact on the psychosocial conditions of affected patients. Reconstruction of the facial nerve to restore facial symmetry and expression is still a significant surgical challenge. In this study, the authors assessed a hypoglossal-facial nerve anastomosis method combined with neurotrophic factor gene therapy to treat facial palsy in adult rats after facial nerve injury. Methods Surgery consisted of the interposition of a predegenerated nerve graft (PNG) that was anastomosed with the hypoglossal and facial nerves at each of its extremities. The hypoglossal nerve was cut approximately 50% for this anastomosis to conserve partial hypoglossal function. Before their transplantation, the PNGs were genetically engineered using lentiviral vectors to induce overexpression of the neurotrophic factor neurotrophin-3 (NT-3) to improve axonal regrowth in the reconstructed nerve pathway. Reconstruction was performed after facial nerve injury, either immediately or after a delay of 9 weeks. The rats were followed up for 4 months postoperatively, and treatment outcomes were then assessed. Results Compared with the functional innervation in control rats that underwent facial nerve injury without subsequent treatment, functional innervation of the paralyzed whisker pad by hypoglossal motoneurons in rats treated 4 months after nerve reconstruction was evidenced by the retrograde transport of neuronal tracers, the recording of muscle action potentials conducted by the PNG, and the recovery of facial symmetry. Although a better outcome was observed when reconstruction was performed immediately after facial nerve injury, reconstruction with NT3-treated PNGs significantly improved functional reinnervation of the paralyzed whisker pad even when implantation occurred 9 weeks posttrauma. Conclusions Results demonstrated that hypoglossal-facial nerve anastomosis facilitates innervation of paralyzed facial muscle via hypoglossal motoneurons without sacrificing ipsilateral hemitongue function. Neurotrophin-3 treatment through gene therapy could effectively improve such innervation, even after delayed reconstruction. These findings suggest that the combination of surgical reconstruction and NT-3 gene therapy is promising for its potential application in treating facial palsy in humans.

We examined the effect of early exercise training (Ex) on glucose kinetics, basal, and insulin-stimulated skeletal muscle (SKM) plasma membrane (PM) GLUT4 in pre- and/or postnatal nutrient-restricted adult rat offspring compared with sedentary (Sed) state. Pregestational control female (Ex CON vs. Sed CON) and offspring exposed to prenatal (Ex IUGR vs. Sed IUGR), postnatal (Ex PNGR vs. Sed PNGR), or pre- and postnatal (Ex IUGR + PNGR vs. Sed IUGR + PNGR) nutrient restriction were studied. The combined effect of exercise and pre/postnatal nutrition in the Ex IUGR demonstrated positive effects on basal and glucose-stimulated plasma insulin response (GSIR) with suppression of endogenous hepatic glucose production (HGP) compared with sedentary state. Ex PNGR was hyperglycemic after glucose challenge with no change in glucose-stimulated insulin production or HGP compared with sedentary state. Ex IUGR + PNGR remained glucose tolerant with unchanged glucose-stimulated insulin production but increased endogenous HGP compared with sedentary state. Basal SKM PM-associated GLUT4 was unchanged by exercise in all four groups. Whereas Ex PNGR and Ex IUGR + PNGR insulin responsiveness was similar to that of Ex CON, Ex IUGR remained nonresponsive to insulin. Early introduction of regular Ex in the pregestational female offspring had a positive effect on hepatic adaptation to GSIR and HGP in IUGR and IUGR + PNGR, with no effect in PNGR. Change in insulin responsiveness of SKM GLUT4 translocation was observed in exercised IUGR + PNGR and PNGR but not in exercised IUGR.

This study is an attempt to understand how PNGO’s are impacted as a result of the Israel- Palestine conflict today, while touching upon issues such as the Gaza blockade, relief aid, political participation, Hamas and the PA, and the occupation. I have had the opportunity of coming into contact with four different PNGO workers based in the Gaza Strip, and conducted qualitative structured interviews with each of them. They possess key insights as they exist at the center of the research problem. Through the theoretical lens of Paul Lederach (1997) and the utilization of the Directed Content Analysis (DCA) method, the structured interviews were analyzed in order to explore both the factors that Gazan PNGO workers identified as having an effect on their organization, as well as how it affects their organizational capacity and peace work.

Papua New Guinea has the highest infant and maternal morbidity and mortality rates in the Western Pacific Regions and 50% of hospital admissions are from vaccine preventable diseases. About 85% of 6 million inhabitants live in rural PNG where basic services are lacking or non-existent. Difficulties such as lack of infrastructures like road network and communication, geographical barriers like big mountain ranges, large rivers and swamps, shortage of skilled health professionals and higher concentration of health workers in cities pose great challenges in delivering health services effectively across the country. Telehealth may play an important role in reaching health services to the underserved population in PNG. As part of this study, it is essential to assess the potential of telehealth to enhance the delivery of health services. Specifically, this research aims to report the knowledge, attitudes and skills towards information and communication technology of health care providers in PNG. The study used a cross sectional method of health professionals working at the time of the survey. It collected 174 questionnaires from four hospitals and used SPSS (version17.0) to analyse the data. The results showed that younger male physicians, paramedics, those with gadgets, higher educational qualifications and exposed to ICT resources possess better knowledge and skills than female, older age group and those without gadgets or under exposed to ICT. Currently, the health care professionals in general have leap frogged the technology by focusing on handheld devices such as cell phones rather than landlines. This represents scope for growth and willingness by health workers to adopt and expand telehealth in PNG.

Ce travail présente l'approche UML/PNO (Unified Modelling Language with Petri Net Objects) pour la spécification de systèmes temps réel. La méthode propose d'enrichir la description semi-formelle UML du système par une modélisation formelle basée sur les réseaux de Petri. Les concepts UML de sous-système et d'interface ont été étendus afin d'améliorer la description de la vue système en termes de tructuration, gestion de projet et organisation de la modélisation. L'objectif est également d'adapter la méthode de modélisation système à une approche " orientée composant " pour le temps réel. Le concept " d'objet composé " permet d'intégrer des spécificités temps réel au sein du composant (protocole de communication, contrainte temporelle, effet observable). Le comportement des objets est spécifié à l'aide des réseaux de Petri à places et transitions stochastiques temporelles afin de permettre la validation et la vérification du système en cours de spécification. L'approche de validation propose des traductions semi-automatiques des diagrammes UML en réseaux de Petri. Les techniques classiques de simulation et d'évaluation de performances peuvent alors être appliquées. Ces traductions présentent l'avantage de rassembler, sur un même modèle à réseau de Petri, tous les aspects dynamiques du composant apparaissant dans différents diagrammes UML et de vérifier ainsi la cohérence de son comportement et de son utilisation. La vérification utilise les techniques d'analyse formelle, basées sur l'utilisation conjointe du graphe de classes et de la logique linéaire. Une approche de vérification quantitative des contraintes temporelles est proposée au niveau de l'analyse des besoins et des exigences du système. Une partie de ce travail a été concrétisée par l'intégration d'un module (ArgoPNO) dans l'atelier de génie logiciel ArgoUML. A ce jour, une première automatisation de la démarche de vérification des contraintes temporelles est opérationnelle sur cet outil.

In this thesis I examine the contemporary nexus between White women and the raced and classed institution of White womanhood. More specifically, I focus on White Australian women who are middle class, rich in cultural capital, and generally consider themselves to be progressive; that is race privileged women but women who are not usually associated with overt racism. My analysis unfolds White Australian women in the discursive context of the ideologies of feminism and feminist-influenced anti-racist politics, as well as the ideologies of femininity. The thesis shows how this nexus is enacted through a vision of White women as Good as expressed in the political commitments, mentalities, relationships, narratives and corporeality of such women. The research problem that I identified and worked through in the thesis is as follows: for middle class White women, (who can be seen and see themselves as generic 'women'), Whiteness has been seen and played out as Goodness. Further, in the playing out of this Goodness White women accumulate and defend the prestige and privileges of Whiteness. Specifically, I argue that Whiteness is reproduced in some of the discourses and practices of White feminism, by the progressive White women involved in anti-racist politics, and in the femininity industry and the ways it is taken up. The nub of the problem I identify is that White women's involvement in the structures and narratives that support Whiteness is often grounded in the very qualities of character and conduct that emerge from the colonial and class-constructed ideal of White womanhood and which have historically distinguished them from denigrated others. These qualities- notably virtue, innocence and self-restraint- whilst differently nuanced in other contexts are an ongoing expression of the uses made of White womanhood as the visible sign of race and class superiority. The work examines four key periods: the Australian colony of PNG during the decolonising 1960's and 1970's; the high years of 1970's and 1980's feminism; the race debates of the 1990's; and the bodily practices of present day White women gripped by fears of fat and aging. I explore the ways in which White women's Whiteness is played out in benevolent Black/White relationships, the over-reach of difference feminism, particular kinds of anti-racist identities and activism, and body-improvement practices. In all these cultural sites, White women's Whiteness is often represented as a kind of moral being and deployed as moral authority in ways that are consonant with the raced and classed construction of White women as moral texts. My research approach was determined by the research problem I identified. Given my argument that White women mis-recognise Whiteness as Goodness in a race-structured society, then the collecting of data through interviews or surveys would have yielded material subject to this blindness. Instead, I explored sites and material where moral claims were being pressed, and case studies where 'women' were enacting themselves or being represented or interpellated as moral texts. My selection of primary source material ranges from feminist newsletters, women's and other magazines, literature, film, event programs and flyers, radio and television broadcasts, newspapers and websites, as well as reflections on my own experiences. Secondary source material includes feminist theoretical texts as well as texts drawn from a range of other disciplines, and other historical background materials. I lay out and support my arguments using a technique not dissimilar to collage, aiming to construct a picture that is compelling in its detail as well as coherent in its overall effect. This thesis is a contribution to the de-naturalisation of Whiteness. Navigating a course between the opposing hazards of essentialising Whiteness and understating its effects in contemporary Australian society, I have brought into clearer view some of the strategies which maintain the authority of Whiteness.

Assembly of neuronal networks with distinct patterns of connectivity during nervous system development involves the growth, extension and branching of axons and dendrites. Over the years genetic and biochemical studies in model organisms have contributed significantly in identifying mechanisms regulating axon growth and extension. However the molecular mechanisms underlying axon branching remain unclear. The egg-laying neuronal circuitry in C. elegans has proven to be a robust system for identifying and characterizing novel genes involved in neuronal morphology. This circuitry which mediates egg-laying behavior in nematodes is composed of two families of motorneurons, HSNs and VCs, which are among the most branched neurons in C. elegans. A genetic screen for axon branch defects in the egg-laying neurons identified png-1 to disrupt neuronal morphology including axon branching. png-1 encodes a Peptide: N-glycanase (PNGase), a conserved cytosolic enzyme that removes N-linked sugar moieties from glycoproteins. In this thesis I present my work characterizing and examining png-1 and its role in mediating axon branching. Mutations in png-1 resulted in excessive ectopic axon branching in the VC4 and VC5 egg-laying neurons as well as branching in the normally unbranched AVL and DVB neurons. Behavioral analysis in these mutants revealed defects in egg-laying behavior and mild in-utero egg retention phenotypes. Cellular characterization shows ubiquitous expression of png-1 in many tissues including vulva cells, muscles, gonads, and neurons. My analysis also shows that png-1 acts both cell-autonomously and cell non-autonomously from neurons and epithelial cells to restrict axon branching around the vulva. Using a candidate gene approach I identified a deletion allele of the DNA repair gene, rad-23, to display axon branching defects and interact with png-1 within a common pathway to regulate axon branching. Additionally, through a genetic modifier screen for enhancers and suppressors of VC4-5 branching defects in png-1, I identified a new allele of sax-2 as an enhancer mutation. sax-2 encodes a scaffolding protein that regulates the activity and localization of sax-1, an NDR kinase. Examination of neuronal phenotypes in sax-1 and sax-2 mutants revealed similar png-1 like defects in VC4-5. Genetic analysis of the double mutants png-1;sax-1 and png-1;sax-2 revealed strong synergistic phenotypes suggesting that png-1 and sax-1/sax-2 function in parallel pathways to regulate axon branching. In summary, this thesis reveals novel components and pathways in the regulation of neuronal branching.

This thesis studies the effect of using delta encoding to compress archivesof images. The results are compared with two types of lossless PNGcompression. The tests show that the most sophisticated PNG methodtested compresses the archives around 2-12% higher than the delta encodingmethods, but at the cost of taking more than 10 times as long.The conclusion is therefore drawn that delta encoding could be a usefulmethod for compressing image archives in environments where speed ismore important than storage.

Abstract ExxonMobil PNG Limited (EMPNG) operates the Papua New Guinea Liquefied Natural Gas Project (PNG LNG), an integrated LNG project comprising wellpads, gathering lines, gas conditioning plant, onshore and offshore export pipelines, liquefaction plant and marine terminal in Papua New Guinea (PNG). The PNG LNG project is a joint venture with participation by ExxonMobil, Oil Search Limited (OSL), Kumul Petroleum, Santos, JX Nippon Oil and Gas Exploration and Mineral Resources Development Company, and began production in 2014. The highlands of PNG presents a challenging physical environment, with high rainfall, steep terrain, active tectonics and seismicity, and ongoing landsliding and erosion. The PNG LNG onshore gas and condensate pipelines confront these physical challenges by having to traverse approximately 150 km of steep volcanic, mudstone and Karstic highlands along the Papuan Fold and Thrust Belt, the modern leading edge of active mountain-building, plus an additional 150 km in Karstic lowlands. During design, construction and operations of the pipelines, ExxonMobil has addressed these challenges in partnership with the engineering, construction and specialist consulting communities. On February 25th, 2018 (UTC) a Magnitude 7.5 earthquake struck the PNG highlands. The event, along with its approximately 300 aftershocks, caused widespread community impact, landsliding and damage to over 1000s of km2, and was centered directly under the highlands portion of the PNG LNG pipelines. The pipelines however, did not lose containment or pressure, and, following inspections and repairs to the PNG LNG gas conditioning plant, PNG LNG production was restored within seven weeks of the main shock. This technical paper and companion oral presentation discuss the key factors of this successful outcome, in particular the sustained condition of the gas and condensate pipelines. Contributing factors to the pipeline’s success include route selection, pipe material specification, early commitment to field studies, careful assessment of geohazards, high awareness of off-ROW community impacts, micro-routing during construction, and active geohazard management during startup and operations. The paper demonstrates that, with respect for the host community, thoughtful engineering, careful construction and ongoing surveillance, pipelines can be safely and successfully designed, constructed and operated in remote and extreme geohazardous environments.
