La culture sur mais de la souche mrc 2654 du phomopsis leptostromiformis n'a pas conduit à la production de la phomopsine. Néanmoins, le résidu méthanolique du maïs contaminé présentait une toxicité aigue vis à vis des rats. Une méthodologie d'analyse et de purification associée à un contrôle des activités biologiques des fractions purifiées nous a permis, grâce en particulier a la clhp préparative sur silice, de localiser et d'isoler 2 toxines cristallisées a4 et a4' à des taux respectifs de 80 et 40 mg par kg de maïs contaminé. L’étude structurale des 2 toxines basée sur l'analyse spectroscopique par fab/ms et par rmn 1d et 2d, montre que les deux toxines appartiennent à la famille des chaetoglobosines. La toxine a4 a pu être identifiée à la chaetoglobosine m tandis que la toxine a4' s'avère être une nouvelle chaetoglobosine. L’étude de la cytotoxicite in vitro sur hépatocytes de rats des chaetoglobosines a permis d'estimer leur ci50 respectivement à 70 et 64 ug/ml. La dl50 déterminée in vivo sur rat est d'environ 20 mg par kg de rat pour les deux toxines. La bonne corrélation des résultats obtenus in vitro et in vivo montre que les 2 chaetoglobosines sont des produits hépatotoxiques. L’étude réalisée in vitro sur cellules ovariennes d'hamster montrent que les chaetoglobosines a4 et a4' inhibent le clivage cytoplasmique des cellules. Enfin un suivi par clhp de la production en milieu liquide au cours du temps des chaetoglobosines a4 et a4' a montré que l'obscurité est un facteur favorable a leur production dont l'optimum est atteint vers 10 jours d'incubation

in many of the vine-growing areas of the world. Four different Phomopsis spp. have
previously been associated with this disease. The present study investigates the
taxonomic significance of the different taxa found on grapevines in South Africa, as well
as the endophytic growth and fungicide sensitivity of Phomopsis viticola isolates. The
thesis is compiled of several different parts, which deal with specific, but related topics,
and hence some duplication has been unavoidable.
Understanding the epidemiology of a disease is important for the correct timing of
disease control. To investigate the endophytic growth of P. viticola, asymptomatic
shoots were collected at eight different growth stages. Nodes, internodes, leaf petioles,
leaves, tendrils and bunch peduncles were investigated. Two Phomopsis spp., taxon 1 and
2 were identified in this study. The Phomopsis viticola-complex had a relative
importance of 9% and accounted for 3% of the isolations. P. viticola (taxon 2) is mainly
isolated from the nodes and internodes. Inoculations of healthy, young vine tissue
confirmed taxon 2 to be a virulent pathogen, suggesting that it is a latent pathogen rather
than an endophyte. In contrast, taxon 1 appeared to be a true endophyte, and did not
seem to be an important pathogen on vines.
The true identity of the causal organism of Phomopsis cane and leaf spot disease
was investigated by collecting samples from 58 different vineyards in the grapevine
growing areas of the Western Cape. P. viiicola occurred in grapevine material collected
from Lutzville to Swellendam, but was not found in the Oudtshoorn and Orange River
grapevine areas. Diaporthe perjuncta (taxon 1), P. vutcola (taxon 2), taxon 3 and a
Phomopsis species commonly associated with shoot blight of peaches in the U.S.A., P.
amygdali, were identified among the South African grapevine isolates. Examination of
the Australian culture designated as taxon 4 found it to be a species of Libertella, thus
excluding it from the P. viticola-complex. An Italian isolate was found to represent a
species of Phomopsis not previously known from grapevines, and this was subsequently
described as taxon 5. Species delimitation was based on morphological and cultural
characteristics, stem inoculations and the formation of the teleomorph in vitro. The
identity of each morphological taxon was confirmed by means of phylogenetic analyses
of the nuclear ribosomal DNA internal transcribed spacers (ITS 1 and ITS2) and the 5'
end partial sequence of the mitochondrial small subunit (mtSSU). P. amygdali,
associated with peach shoot blight in the U.S.A., was isolated once only and appeared to
be of lesser importance in this disease complex. Furthermore, taxa 1 (Diaporthe
perjuncta) and 3 were also rarely encountered and proved to be non-pathogenic,
indicating their non-functional role in Phomopsis cane and leaf spot disease. Taxon 2
(Phomopsis viticolas was common and widely distributed in diseased vineyards. This
taxon was associated with the typical disease symptoms and proved to be pathogenic.
Morphologically taxon 2 corresponded best with P. viticola, which was also neotypified
in this study. Taxon 2 was mostly isolated from buds and nodes, indicating that these are
important sites in which the fungus survives during winter. Molecular data indicated that
taxon 3 and P. amygdali were not host specific to grapevine.
The currently used foliar fungicides were compared to the new strobilurin
fungicides. The effects of nine fungicides (azoxystrobin, flusilazole, folpet, fosetyl-
Al+mancozeb, kresoxim-methyl, mancozeb, penconazole, spiroxamine and
trifloxystrobin) were tested in vitro on inhibition of mycelial growth. The following EC50
(ug/ml) values were obtained: azoxystrobin (0.350), flusilazole (0.007), folpet (4.489),
fosetyl-Al+mancozeb (3.925), kresoxim-methyl (1.665), mancozeb (2.891), penconazole
(0.023), spiroxamine (0.321) and trifloxystrobin (0.051). Additionally, azoxystrobin,
folpet, kresoxim-methyl, mancozeb, propineb and trifloxystrobin were tested for their
ability to inhibit spore germination in vitro. The subsequent EC50 (ug/ml) values were
obtained: azoxystrobin 0.123), folpet (0.510), kresoxim-methyl (0.0037), mancozeb
(0.250), propineb (0.156) and trifloxystrobin (0.003). The results reported in part 4
showed that the strobilurin fungicides inhibited the mycelial growth and spore
germination of P. viticola. However, further trials need to be conducted to verify these
findings under field conditions. In the present study taxa 1, 3 and P. amygdali were
infrequently isolated, suggesting that they played a less prominent role in the P. viticolacomplex.<br>AFRIKAANSE OPSOMMING: Streepvleksiekte van wingerd is 'n ekonomies belangrike siekte wat in die meeste
wingerdproduserende gebiede van die wêreld voorkom. Vier Phomopsis spesies is in die
verlede met dié siekte geassosieer. Hierdie studie ondersoek die taksonomiese
belangrikheid van die verskillende taksa wat op wingerd in Suid Afrika gevind word,
asook die endofietiese groei en fungisiedsensitiwiteit van die Phomopsis vitico/a isolate.
Hierdie tesis bestaan uit verskeie dele met spesifieke, maar verwante onderwerpe wat tot
onafwendbare duplisering lei.
Dit is belangrik om die epidemiologie van 'n siekte te verstaan sodat korrekte en
tydsberekende siektebeheer toegepas kan word. Die endofietiese groei van P. vitico/a is
ondersoek deur simptoomlose lote by agt verskillende groei stadiums te versamel.
Nodusse, internodusse, blaarstele, blare, rankies en trosstele is ondersoek. Twee
Phomopsis spp., takson 1 en 2 is geïdentifiseer. Die Phomopsis vitico/a-kompleks het
3% van die isolasies uitgemaak en 'n relatiewe belangrikheid van 9% getoon. P. vitico/a
(takson 2) is meestal uit die nodus en internodus geïsoleer. lnokulasies van gesonde, jong
wingerdweefsel het bevestig dat takson 2 'n virulente patogeen is en dat die takson eerder
'n latente patogeen as 'n endofiet is. In teenstelling hiermee is takson 1 'n ware endofiet
en 'n onbelangrike patogeen op wingerd.
Die ware identiteit van die veroorsakende organisme van streepvlek is ondersoek
deur plantmateriaal vanaf 58 verskillende wingerde in die wingerproduserende gebiede
van die Wes-Kaap te versamel. P. vitico/a is in wingerdmateriaal vanaf Lutzville tot
Swellendam aangetref, maar nie in die Oudtshoorn en Oranjerivier wingerd produserende
gebiede nie. Diaporthe perjuncta (takson 1), P. vitico/a (takson 2), takson 3 en P.
amygdali is in die Suid Afrikaanse wingerdisolate geïdentifiseer. P. amygdali word met
lootverskroeiing van perske bome in die V.S.A. geassosieer. Die Australiese isolaat wat
benoem is as takson 4, is met die huidige ondersoek gevind om 'n spesie van Libertella
te wees. Takson 4 is daarvolgens uit die P. vitico/a-kompleks gelaat. 'n Italiaanse isolaat
het 'n nuwe spesie van Phomopsis op wingerd verteenwoordig en is vervolgens as takson
5 beskryf. Spesie-onderskeiding is op morfologiese en kulturele eienskappe,
staminokulasies en die vorming van die teleomorf in vitro gebaseer. Die identiteit vanelke morfologiese takson is met behulp van filogenetiese analises van die nukleêre
ribosomale DNS intern transkriberende spasieerders (ITS 1 en ITS2) en die 5' punt
gedeeltelike nukleotied volgorde van die mitochondriale klein subeenheid (mtSSU)
bevestig. P. amygdali is slegs een keer geïsoleer en blyk van minder belang in die
siektekompleks te wees. Takson 1 (Diaporthe perjuneta) en takson 3 het ook min
voorgekom en is nie-patogenies, wat hul nie-funksionele rol in streepvleksiekte aandui.
Takson 2 (P. viticola) is algemeen geïsoleer en kom wyd verspreid voor. Hierdie takson
is geassosieer met die tipiese siektesimptome en is ook patogenies. Morfologies stem
takson 2 met P. viiicola ooreen en is ook geneotipifiseer in hierdie studie. Takson 2 is
meestal vanaf die ogies en nodusse geïsoleer, wat daarop dui dat hierdie belangrike setels
is waar die swam tydens die winter oorleef. Die molekulêre data toon aan dat takson 3 en
P. amygdali nie gasheerspesifiek tot wingerd is nie.
Die swamdoders wat tans teen streepvlek gebruik word, is met die nuwe
strobilurin swamdoders vergelyk. Die effek van nege swamdoders (azoksistrobin,
flusilasool, folpet, fosetyl-Al + mancozeb, kresoxirn-metiel, mankozeb, penconasool,
spiroksamien en trifloksistrobin) is in vitro op die inhibisie van miseliumgroei getoets.
Die volgende EKso-waardes (g/ml) is verkry: azoxystrobin (0.350), flusilasool (0.007),
folpet (4.489), fosetiel-Al + mankozeb (3.925), kresoxirn-metiel (l.665), mankozeb
(2.891), penkonasool (0.023), spiroksamien (0.321) en trifloxystrobin (0.051).
Azoxystrobin, folpet, kresoxim-rnetiel, mankozeb, propineb en trifloksistrobin is ook in
vitro getoets vir hul inhibisie op spoorontkieming. Die volgende EKso-waardes is verkry:

taksa 'n minder belangrike rol in die P. viticola-kompleks speel.

They are considered an important source to the production of secondary metabolites with antimicrobial, antitumor activities, among others. The present work describes the chemical and biological study of the secondary metabolites produced by the endophytic fungus Phomopsis sp. isolated from the plant Passovia stelis. For that, the fungus at hand was cultivated in large scale using the liquid medium of dextrose potato. The ethylacetat crude extract was obtained through extraction from the broth and then submitted to larvicide assay against larvae of the mosquito Aedes aegypti and antimicrobial against the pathogenic bacteria Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli and Pseudomonas aeruginosa. It was also performed assays against the enzyme anticholinesterase, antioxidants and antifungal against Cladosporium cladosporioides e Cladosporium sphaerospermum. In the selective larvicide, 3 fractions showed bioactivity (100 ppm) in 24h. In the dosing assay larvicide, the extract of the fungus Phomopsis sp. showed bioactivity with LD50 of 84.7, 81.6 and 79.5 ppm and LD90 de 147.8, 140.2 and 137.7 ppm after 24, 48 e 72 h, respectively. The crude extract was analyzed using TLC, HPLC and NMR and then purified through column chromatography and HLPC-DAD. From F4, the 3-nitropropionic acid was isolated, which is used in neurological research. Succinic acid, which is considered one of the construction blocks in chemical synthesis industry, was isolated from the fraction F7. Other 3 esterified substances, derivatives of tyrosol, were isolated of the fraction F1, and one of them unpublished. In the antioxidant assay, two fractions (F5, F6) showed positive results and in the assay against the enzyme anticholinesterase four fraction (F2, F5, F6, F7) presented bioactivity. These results demonstrate the expressive biopotency of the fungus Phomopsis sp. and open a new perspective for further studies.<br>Fungos endofíticos são fungos que vivem no tecido interno das plantas sem que lhes causem prejuízo imediato. Esses microrganismos são considerados uma fonte importante para a produção de metabólitos secundários com ação antimicrobiana, antitumoral, entre outras. Este trabalho descreve o estudo químico e biológico dos metabólitos secundários produzidos pelo fungo endofítico Phomopsis sp. isolado da planta Passovia stelis. Para tanto, o fungo em questão foi cultivado em escala ampliada, utilizando meio líquido de batata dextrose. O caldo obtido foi extraído com acetato de etila fornecendo o extrato bruto. Este foi submetido à ensaio larvicida, frente as larvas do mosquito Aedes aegypti, antimicrobiano frente às bactérias patogênicas Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli e Pseudomonas aeruginosa. Também foram realizados ensaios anticolinesterase, antioxidante e antifúngico frente aos fungos Cladosporium cladosporioides e Cladosporium sphaerospermum. No ensaio larvicida de dose, o extrato do fungo Phomopsis sp. exibiu bioatividade com valores de DL50 de 84,7; 81,6 e 79,5 ppm e DL90 de 147,8; 140,2 e 137,7 ppm respectivamente, nas leituras realizadas após 24, 48 e 72 h. No ensaio larvicida seletivo, três frações obtiveram bioatividade em 100 ppm nas primeiras 24 horas. Após análise por CCD, CLAE e RMN, o extrato bruto foi fracionado por cromatografia em coluna e por CLAE-DAD. Da fração F4 foi isolado o ácido 3-nitropropiônico que é muito utilizado em pesquisas neurológicas. Da fração F7 foi isolado o ácido succínico tido como um dos principais blocos de construção na indústria da síntese química. Outras três substâncias da classe dos ésteres, derivados do tirosol foram isolados da fração F1 sendo uma delas inédita. No ensaio antioxidante duas frações (F5, F6) deram resultado positivo e no ensaio anticolinesterásico quatro frações (F2, F5, F6, F7) mostraram atividade. Estes resultados mostraram a expressiva biopotência do fungo Phomopsis sp, abrindo novas perspectivas atrativas para estudos futuros.

Esses micro- organismos são considerados uma fonte importante para a produção de metabólitos secundários com ação antimicrobiana, antitumoral, entre outras. Este trabalho descreve o estudo químico e biológico do fungo endofítico Phomopsis sp. associado à espécie vegetal do cerrado Senna spectabilis. Phomopsis sp. foi cultivado em diferentes meios de cultura para avaliar a variação metabólica em relação a composição dos nutrientes. Foram usados 5 meios líquidos comerciais (MBD, YM, Extrato de Malte, Nutrient e Czapek) e 2 meios sólidos (Milho e Arroz). Os extratos brutos Acetato e Acetonitrila obtidos foram avaliados por Cromatografia Líquida de Alta Eficiência com detector de arranjo de diodos e por Ressonância Magnética Nuclear de 1 H, e submetidos aos ensaios para a avaliação da potencialidade antifúngica, antioxidante e anticolinesterásica. Os resultados evidenciaram extratos com uma variação metabólica intensa e potencialmente bioativos. Destes, os meios MDB e Milho foram selecionados para obtenção dos extratos brutos em escala ampliada. O extrato bruto acetato obtido em MDB foi submetido a fracionamento cromatográfico e levou à identificação da uracila (1), ácido nitropropanóico (2), citocalasina H (3), tirosol (4), ciclo(L- Pro-L-Leu), (5), ciclo(L-Pro-L-Ile) (6), ciclo(L-Pro-L-Val) (7), ciclo(L-Pro-L-Tyr) (8) e ciclo(L-Pro-L-Phe) (9). Este é o primeiro relato de dicetopiperazinas obtidos de fungos do gênero Phomopsis. Esta classe de substâncias apresenta inúmeras bioatividades, principalmente antimicrobiana. Microorganisms are regarded as an important source for the production of secondary metabolites with antimicrobial, antitumoral actions, among others. This work describes the chemical and biological study of endophytic fungus Phomopsis sp. associated with plant species of Cerrado Senna spectabilis. Phomopsis sp. was cultived in different culture media to assess the metabolic variation in relation to nutrient composition. Were used 5 commercial liquid media (PDB, YM, Malt extract, nutrient and Czapek) and 2 solid mediums (Corn and Rice). The acetate and acetonitrile crude extracts were evaluated by High Performance Liquid Chromatography with Diode Array Detector and 1 H Nuclear Magnetic Resonance and subjected to assays for the evaluation of potential antifungal, antioxidant and anticholinesterase. The results showed extracts with an intense metabolic variation and potentially bioactive. From these, the mediums PDB and Corn were selected to obtain the crude extracts on a large scale. The crude acetate extract obtained in PDB was submitted to chromatographic separation and led to the identification of uracil (1), 3-nitropropionic acid (2), cytochalasin H (3), tyrosol (4), cyclo (L- Pro-L-Leu) (5), cyclo (L-Pro-L-Ile) (6), cyclo (L-Pro-L-Val) (7), cyclo (L-Pro-L-Tyr) (8) and cyclo (L-Pro-L-Phe) (9). Phomopsis convolvulus Ormeno, a foliar pathogen of field bindweed, is a good candidate to be developed as a bioherbicide. Large numbers of infective propagules were produced in shake-flask liquid fermentation with modified Richard's (V-8) medium and in solid-substrate fermentation with pearl barley grains. In complex liquid media, pycnidium-like structures were observed. Most conidia stored at $-$70$ sp circ$C remained viable and virulent for at least six months.<br>In controlled environment studies, a minimum of 18 hr of dew was required for severe disease development on inoculated plants. The addition of gelatin, Sorbo $ sp{ rm TM}$, or BOND$ sp{ rm TM}$ to the inoculum did not enhance the disease under various leaf wetness periods. A continuous dew period of 18 hr was superior to the cumulative effect of three interrupted 6 hr dew periods. Secondary inoculum was produced on diseased plants placed under moist conditions for 48 hr or more.<br>In greenhouse experiments, seedlings at the cotyledon and 3- to 5- leaf stage were severely diseased and killed when inoculated with 10$ sp9$ conidia/m$ sp2$. This inoculum density adversely affected the regenerative ability of 4 wk old seedlings and established plants, but few plants were killed. Inoculation of the healthy regrowth from plants previously inoculated with the fungus resulted in much less disease symptoms than expected.

Este trabalho descreve o estudo químico e biológico do fungo endofítico Phomopsis sp. associado à espécie vegetal do cerrado Senna spectabilis. Phomopsis sp. foi cultivado em diferentes meios de cultura para avaliar a variação metabólica em relação a composição dos nutrientes. Foram usados 5 meios líquidos comerciais (MBD, YM, Extrato de Malte, Nutrient e Czapek) e 2 meios sólidos (Milho e Arroz). Os extratos brutos Acetato e Acetonitrila obtidos foram avaliados por Cromatografia Líquida de Alta Eficiência com detector de arranjo de diodos e por Ressonância Magnética Nuclear de 1 H, e submetidos aos ensaios para a avaliação da potencialidade antifúngica, antioxidante e anticolinesterásica. Os resultados evidenciaram extratos com uma variação metabólica intensa e potencialmente bioativos. Destes, os meios MDB e Milho foram selecionados para obtenção dos extratos brutos em escala ampliada. Le phomopsis du a phomopsis helianthi / diaporthe helianthi munt. -cvet. Est une des principales maladies du tournesol en france, pouvant causer de fortes baisses de rendements sur les cultures. L'amelioration varietale constitue l'element essentiel de lutte. Les resultats suivants ressortent de cette etude : 1) les isolats francais de p. Helianthi / d. Helianthi presentent une forte variabilite phenotypique, du point de vue de leurs croissances lineaire et ponderale, de leur morphologie et de leur agressivite sur tournesol. Les isolats yougoslaves ne montrent pas d'agressivite particuliere par rapport aux isolats francais. 2) des interactions entre isolats et hybrides ont ete mises en evidence lors d'infections artificielles mesurant la croissance mycelienne sur feuille, petiole/tige, et capitule mais elles ne changent le classement que pour des genotypes intermediaires. Cette etude confirme la necessite de maintenir, a cote des tests artificiels, des essais en attaques semi-naturelles qui font intervenir l'ensemble des genotypes de l'agent pathogene. 3) la resistance du tournesol au phomopsis suit un modele quantitatif. Sur les 54 hybrides f1 etudies sur 3 annees en conditions d'attaque semi-naturelle multisites et d'infection artificielle, l'aptitude generale a la combinaison apparait plus importante que l'aptitude specifique. Les interactions entre parents sont probablement causees par des effets environnementaux car elles n'apparaissent pas lorsque tous les essais sont regroupes en une seule analyse. Des facteurs de resistance differents ont ete mesures par les tests mycelium sur feuille et petiole. Par ailleurs, les resultats de ces tests refletent generalement ceux obtenus en attaques semi-naturelles. Il existe plusieurs sources de resistance au phomopsis parmi le materiel etudie et il serait utile de mieux connaitre les facteurs de resistance pour determiner les meilleures combinaisons donnant les plus hauts niveaux de resistance sous diverses conditions environnementales.

Os fungos endofíticos são microrganismos que residem nos espaços intercelulares das plantas e estabelecem uma interação não-patogênica com a planta. Portanto, o estudo dos endófitos parece ser uma estratégia promissora na obtenção ou identificação e substâncias potencialmente bioativas. Phomopsis stipata isolado como endófito da espécie vegetal Styrax camporum foi cultivado em diferentes meios de cultivo para se avaliar a produção metabólica e a bioatividade dos extratos brutos obtidos. Com esta avaliação, realizada por RMN de 1H e CLAE-DAD, os meios MDB e Milho foram escolhidos para o cultivo de P. stipata em escala ampliada. O extrato bruto obtido em MDB foi fracionado utilizando técnicas cromatográficas adequadas fornecendo nove frações, sendo que as frações foram submetidas à desreplicação utilizando a técnica de simulação de espectro de RMN de 1H e 13C com o auxílio do software MestreNova®, e posteriormente a EM. Through experiments in vitro were studied, initially, the best conditions of temperature and photoperiod on the development of the pathogen. Later, under the conditions found, the fungus was subjected to different substances: plant essential oils (eucalyptus, clove, oregano, peppermint, ginger, lemongrass, citronella, rosemary and chamomile); different products extraction plant Calea hispida Baker (quercitrin (CHAM - Acetate methanol), 6-acetyl-7-hydroxy-2,2-dimethylchromene (HCRC - crystals), aqueous extract (CHEA) and essential oil (CHOE)); synthetic fungicides (captan, cyproconazol, tebuconazole, triadimenol, mancozeb and sodium hypochlorite); an inductive resistance (Ascophyllum nodosum) and an insecticide (Bacillus thuringiensis) in order to evaluate the fungistatic effect of these in vitro development of the pathogen. The assessments were made by mean of two measurements, diametrically opposite the colonies daily until seven days after the onset of the experiment. The temperature better favored the development of the pathogen in vitro was 20°C. Have to photoperiod, there was no statistical difference between the treatments tested. It was observed that treatments with essential oils of oregano and lemon grass showed higher antifungal activity against the pathogen when compared to other oils used, highlighting the oregano oil, which completely inhibited growth of the pathogen. The compounds isolated from C. hispida showed low percentage of inhibition of P. amygdali, which did not exceed 21.88%. The fungicides captan, tebuconazole cyproconazol and showed a high level of inhibition relative to the other substances tested. From the concentration of 500 ppm, these fungicides reached 100% of control, showing the most efficient at the fungus control.<br>A produção de pêssegos no Brasil vem sofrendo danos na produção devido à queima dos ramos causado pelo fungo Phomopsis amygdali. Através de experimentos in vitro, estudaram-se, inicialmente, as melhores condições de temperatura e fotoperíodo no desenvolvimento do patógeno. Posteriormente, sob as condições encontradas, o fungo foi submetido a diferentes substâncias: óleos essenciais vegetais (eucalipto, cravo, orégano, menta, gengibre, capim-limão, citronela, alecrim e camomila); diferentes produtos da extração da planta Calea hispida Baker (quercitrina (CHAM - Acetato metanólico), 6-acetil-7-hidroxi-2,2-dimetilcromeno (CHCR - cristais), extrato aquoso (CHEA) e óleo essencial (CHOE)); fungicidas sintéticos (captan, ciproconazol, tebuconazol, triadimenol, mancozeb e hipoclorito de sódio); indutor de resistência (Ascophyllum nodosum) e um inseticida (Bacillus thuringiensis), a fim de se avaliar o efeito fungistático destes no desenvolvimento in vitro do patógeno. As avaliações foram feitas pela média de duas medidas, diametralmente opostas das colônias, diariamente, até os sete dias após a instalação do experimento. A temperatura que melhor favoreceu o desenvolvimento do patógeno in vitro foi a de 20°C. Já para o fotoperíodo, não houve diferença estatística entre os tratamentos testados. Observou-se que os tratamentos com os óleos essenciais de orégano e capim-limão apresentaram maior atividade antifúngica sobre o fitopatógeno, quando comparados aos demais óleos utilizados, destacando o óleo de orégano, que inibiu completamente o desenvolvimento do fitopatógeno. As substâncias isoladas de C. hispida apresentaram baixa porcentagem de inibição de P. amygdali, a qual não ultrapassou 21,88%. Os fungicidas captan, ciproconazol e tebuconazol apresentaram elevado nível de inibição em relação às outras substâncias testadas. A partir da concentração de 500 ppm, esses fungicidas atingiram 100% de controle, se mostrando os mais eficientes no controle do fungo.

O Brasil tem uma posição de destaque mundial na produção de frutas e alguns grãos, os quais são comercializados in natura ou na forma de produtos processados. O processamento têm várias vantagens, porém gera grandes quantidades de resíduos, os quais têm sido aproveitados para a alimentação animal e geração de energia. Entretanto, muito tem sido relatado sobre o potencial bioativo desses materiais, dentre eles a ação fungitóxica de alguns de seus compostos sobre fungos fitopatogênicos. Assim, o objetivo desse trabalho foi avaliar o potencial de extratos de resíduos agroindustriais de abacate, uva, café, manga, abacaxi, maracujá e caju na inibição do crescimento micelial in vitro dos fungos Fusarium pallidoroseum, Colletotrichum dematium, Rhizoctonia solani e Phomopsis sp., bem como a composição química do resíduo mais promissor. Os resíduos foram liofilizados, moídos e submetidos a extração com etanol 80% (etanol:água; 8:2, v/v) em banho de ultrassom (extrato denominado bruto ou EB). O EB também foi tratado com a resina Amberlite XAD-2, visando à eliminação de açúcares e interferentes, dando assim origem ao extrato semi-purificado (EP). Os extratos que apresentaram alto rendimento foram avaliados quanto à atividade antifúngica in vitro e teor de compostos fenólicos totais. A amostra que apresentou os melhores índices de inibição foi selecionada para dar continuidade ao estudo, sendo a mesma fracionada em coluna de gel Sephadex LH-20. Os extratos e as frações ativas, detectadas pelo ensaio de bioautografia, foram analisadas quanto a composição química pelas técnicas de HPLC e GC/MS. Na análise de fenólicos totais os maiores teores encontrados foram para a casca de abacate, enquanto que para o ensaio de inibição de crescimento micelial in vitro o melhor resultado foi para a semente de abacate, tanto para o EB quanto para o EP. Desta maneira, a semente de abacate foi selecionada para as etapas posteriores. Para o fracionamento em gel Sephadex LH-20 foi eleito o EP para F. pallidoroseum enquanto que para os demais fungos foi eleito o EB. No fracionamento do EP obtiveram-se 13 frações, sendo que as frações 3 e 4 foram ativas, enquanto que no do EB obtiveram-se nove, sendo as frações 3, 4 e 5 ativas. Pela técnica de HPLC foram detectados em comum nas frações 3, 4 e 5 do EB dois compostos majoritários, e nas frações 3 e 4 do EP, sete compostos, os quais não puderam ser identificados pelos padrões comerciais disponíveis. Já pela técnica de GC/MS foi possível a identificação de quatro compostos em comum nas frações ativas do EB e de 11 nas frações do EP. Dentre os compostos presentes nas frações ativas, foram identificados ácidos graxos, os quais têm sido reconhecidos por apresentarem ação antifúngica. Assim, pode-se concluir que os resíduos agroindustriais estudados são fontes de compostos com atividade antifúngica, podendo assim ser uma alternativa para o controle de fungos fitopatogênicos à cultura da soja<br>Brazil holds an important position in the world production of fruit and some grains, which are commercialized in natura or in processed products. The processing has several advantages, but generates large amounts of by-products which have been used for animal feed and power generation. However, bioactive potential of these materials have been reported, including the fungitoxic effect. The goal of this study was to evaluate the potential of agro-industrial by-products extracts of avocado, grape, coffee, mango, pineapple, passion fruit and cashew to inhibit in vitro mycelial growth of fungi Fusarium pallidoroseum, Colletotrichum dematium, Rhizoctonia solani and Phomopsis sp. as well as the chemical composition of the most promising by-products. The by-products were freeze-dried, milled and the extraction was done with ethanol 80% (ethanol:water, 8:2, v/v) in an ultrasound bath (called crude extract or CE). The CE was also treated with Amberlite XAD-2 to eliminate sugars and interfering compounds so that the semi-purified extract (PE) could be obtained. Extracts with high yield were evaluated to in vitro antifungal activity and phenolic compounds content. Samples with the best inhibition rates were selected to continue this study, and it was fractioned on gel Sephadex LH-20. The extracts and the active fractions, detected by bioautography, were analyzed by HPLC and GC/MS. The highest phenolic content was found in the avocado peel, meanwhile for the mycelial growth inhibition in vitro, the best result was found in the avocado seeds for both extracts (CE and PE). Thus, avocado seed was selected for subsequent steps. For fractionation on Sephadex LH-20 gel, PE was chosen for F. pallidoroseum and CE for the other fungi. For PE, 13 fractions were obtained in which fractions 3 and 4 were active. For CE, 9 fractions were obtained and the actives were 3, 4 and 5. In CE, HPLC technique detected two major compounds in common in fractions 3, 4 and 5. In PE, seven major compounds were detected in fractions 3 and 4, which could not be identified by commercial available standards. By the GC/MS technique was possible to identify four compounds in common in the active CE fractions and 11 in PE. Among the compounds presented in the active fractions, fatty acids were identified. It has been reported that antifungal action has been found in these compounds. Therefore, the studied agroindustrial by-products are sources of compounds with antifungal activity and they can be used as an alternative to control phytopathogenic fungi on soybean

Turnera subulata L. is a plant belonging to the Turneraceae family and is popularly known in Brazil as chanana. The genus Turnera has about 120 species distributed in the Americas and Africa, being the most representative of the family Turneraceae. Some species of Turnera are highly used in popular medicine for different types of inflammatory diseases. The association between living beings is a vital condition for species unable to achieve alone, as a means of survival for obtaining nutrients and defenses against predatory species. Among the microorganisms, the fungi are those who are most often associated with plants. Endophytic are microorganisms that inhabit the bodies of plants that, at some period of their life cycle, colonize internal tissues of the plant, without causing any apparent damage to its host. From the fragments of leaves from Turnera subulata L. could be isolated a filamentous fungus that was identified as the fungus Phomopsis sp. through traditional microscopy and molecular techniques. In a statistical evaluation it was noted that the endophytic Phomopsis sp. presents a higher growth in BDA and BSA media, as well as in the presence of light. The steroid ergosterol could be isolated from the hexane extract of this fungus and identified by the NMR techniques. In tests of antagonism of the endophytic Phomopsis sp. against phytopathogens, it was observed halum of inhibition against Rhizoctonia sp., Colletotrichum sp. and Pestalotiopsis sp. Concerning the antioxidant activity it was observed that the chloroform extract was more effective than the hexane one. On the other hand all the extracts from the mycelium of Phomopsis sp. and its ethylacetate extract from the cultured filtrate showed no antimicrobial activity against strains of Escherichia coli, Staphylococcus aureus, Candida albicans, Candida tropicalis and Candida glabrata. Therefore it was concluded that Phomopsis sp. may act as endophytic of T. subulata. it has shown to grow best in BDA and BSA medium, as well as under continuous lightining. A associação entre os seres vivos é uma condição vital para espécies incapazes de conseguirem, sozinhas, meios de sobrevivência como obtenção de nutrientes e defesas contra espécies predadoras. Entre os microrganismos, os fungos são os que se encontram mais frequentemente associados às plantas. Microrganismos endofíticos são os organismos que habitam órgãos de plantas sem causar dano aparente a seu hospedeiro. A partir dos fragmentos de folhas de Turnera subulata L. isolou-se um fungo filamentoso que foi identificado através de características morfológicas (morfoculturais) e moleculares como sendo Phomopsis sp. O isolado obtido apresentou um crescimento superior nos meios BDA (Batata-Dextrose-Ágar) e BSA (Batata-Sacarose-Ágar), com relações à cinco meios testados, sendo a presença de luz constante um fator de estímulo. Foi identificada por RMN a presença de ergosterol no extrato hexânico das céluas do Phomopsis sp. cultivado em meio líquido BD (Batata-dextrose). Em testes de antagonismo do endofítico Phomopsis sp. contra patógenos de plantas, observou-se halo de inibição deste frente a Rhizoctonia sp., Colletotrichum sp. e Pestalotiopsis sp. Nos resultados da avaliação do percentual de atividade antioxidante dos extratos do micélio em hexano e clorofórmio pôde-se observar que extrato em hexano teve menor atividade que o extrato em clorofórmio. Os extratos hexânico, clorofórmico, acetato etílico e metanólico do micélio de Phomopsis sp. e o extrato acetato etílico do filtrado de cultura desse fungo isento de células não apresentaram atividade antimicrobiana contra as cepas de Escherichia coli, Staphylococcus aureus, Candida albicans, Candida tropicalis e Candida glabrata. Natural products have, historically, played an important role in drug discovery. Nevertheless, drug resistance, pathogen evolution, and global climate change threaten human health and nearly all current anti-infective treatments on the market today. It is undeniable that new drug discovery efforts are needed with increasing urgency.
Bolstered by a rich history of discovering treatments in the world around us, natural products chemists continue to look to the environment with increasing understanding and emerging technologies that allow efficient, effective isolation of new chemical entities. This thesis will describe one such endeavor.
Focusing on fungal natural products, herein is described the isolation and structure elucidation of new, bio-active natural products. Further, the development and implementation of a large fungal screening program will be discussed, the results of which stand to advance microbial drug discovery in the Baker lab for years to come.

A intenção de melhorar o conhecimento sobre alguns aspectos da epidemiologia e meios de luta contra Botryosphaeriaceae e Phomopsis viticola estiveram na base do presente trabalho. No âmbito dos estudos epidemiológicos estudou-se a relação entre a captura de esporos em lâminas-armadilha e os dados meteorológicos. Para tal procedeu-se à captura de esporos em lâminas-armadilha e efectuou-se uma amostragem quinzenal. Constatou-se que o valor máximo de recolha de esporos coincidiu com períodos durante ou após a ocorrência de alguma pluviosidade. Relativamente aos meios de luta, foi feita uma avaliação da eficácia de alguns fungicidas (ciprodinil+fludioxonil, azoxistrobina, oxicloreto de cobre, tebuconazol e difenoconazol) e das respectivas épocas de aplicação (poda e/ou estado fenológico D/E) contra as doenças em causa. Avaliou-se a incidência e a severidade das doenças e, no final de dois anos de ensaio, verificou-se que difenoconazol, tebuconazol e o oxicloreto de cobre tiveram um efeito positivo significativo na redução da incidência das doenças. Em relação à época, o tratamento no estado fenológico D/E foi, tendencialmente, o que provocou maior redução na incidência das doenças. No que diz respeito à severidade das doenças não se verificou, neste ensaio, nenhuma variação positiva com a realização de dois tratamentos à vinha. Nesse contexto, o emprego de substâncias naturais com ação fungicida surge como uma opção de baixo impacto ecológico e ambiental. Diversas espécies vegetais já mostraram efeito positivo no controle de doenças de plantas, sugerindo sua potencialidade também no tratamento de sementes. Para ser uma prática viável, o emprego de substâncias naturais no tratamento de sementes, além de reduzir ou controlar a população microbiana das sementes, não pode ter efeito negativo sobre a fisiologia das sementes, prejudicando sua germinação e vigor. Diante do exposto, o objetivo da presente pesquisa foi verificar a possibilidade de emprego de óleos essenciais de diferentes espécies vegetais no tratamento de sementes, visando o controle de Phomopsis sojae, e avaliar também o efeito destes produtos sobre a qualidade fisiológica das sementes tratadas. Uma das mais importantes estratégias para acessar tal produção metabólica é o uso de cultivos em co-cultura, onde a interação entre dois ou mais microrganismos pode mimetizar, de certa maneira, as relações tróficas existentes na natureza. A relação de competição por nutrientes e espaço leva a produção de enzimas e compostos relativos a um determinado tipo de interação que pode estar relacionado a reprodução, inibição de crescimento ou estimulação. Neste trabalho, foi realizado o co-cultivo entre os fungos Fusarium solani e Phomopsis cassiae, ambos isolados da Senna spectabilis. A investigação foi direcionada ao encontro de novos produtos naturais biossitentizados pelas espécies em co-cultivo. Para isso, os fungos foram cultivados em meio sólido de batata dextrose ágar. O acompanhamento do crescimento dos fungos visou a observação de possíveis distinções morfológicas de acordo com nível de interação. Os perfis metabólicos dos microrganismos foram obtidos através do uso de técnicas do estado da arte em CLAE-DAD, CG-EM e RMN. Para os fungos em co-cultivo foi possível determinar a presença de cinco substâncias quando comparado aos cultivos isoladamente pela técnica de CLAE-DAD com o auxílio das ferramentas quimiometricas. Os espectros obtidos por RMN de 1H propiciaram a visualização da existência de sinais únicos presentes no experimento de co-cultivo, porém, sua proporção minoritária impossibilitou a correlação ou identificação dos sinais. A técnica de CG-EM propiciou a sugestão de 30 substâncias através da comparação com o banco de dados NIST.<br>In recent years, the search for new bioactive compounds has increased the need to develop several methods of analysis, aiming to activate metabolic routes silenced in microorganisms. An important strategy to access the production of distinct secondary mebatolites is co-culture, where the interaction between two or more microorganisms occurs, mimicking, to a certain extent, the relationships existing in nature. The competition for nutrients and space leads to the production of enzymes and compounds, which are related to the type of interaction (reproduction, inhibition of growth or stimulation). In this work, co-cultivation was carried out between the fungi Fusarium solani and Phomopsis cassiae, both isolated from Senna spectabilis’s. The research was directed to the encounter of new natural products biossentetized by the species in co-cultivation. For this, the fungi were cultivated in the solid batata dextrose ágar medium, being monitored their growth in order to observe possible distinctions in their morphologies when the interaction occurs. The metabolites profiles of the microorganisms were observed through the LC-DAD, GC-MS and NMR techniques. For the fungi in co-culture it was possible to determine the presence of five substances when compared to the cultures alone by the CLAE-DAD technique with the help of chemometric tools. The spectra obtained by 1H NMR allowed the visualization of the existence of unique signals present in the co-cultivation experiment, however, its minority proportion made it impossible to correlate or identify the signals. The research was directed to the encounter of new natural products biossentetized by the species in co-cultivation. For this, the fungi were cultivated in the solid batata dextrose ágar medium, being monitored their growth in order to observe possible distinctions in their morphologies when the interaction occurs. The metabolites profiles of the microorganisms were observed through the LC-DAD, GC-MS and NMR techniques. For the fungi in co-culture it was possible to determine the presence of five substances when compared to the cultures alone by the CLAE-DAD technique with the help of chemometric tools. Endophytic microorganisms are important sources of bioactive compounds and have been widely studied due to the properties that their metabolites have. Compared to the host plants are easier handling in the laboratory. From the Phomopsis sp fungus genus have been isolated compounds with different biological activities. The investigation of the metabolites, whether primary or secondary, present in the filtrate of culture is of utmost importance. The metabolomic study of Phomopsis sp can lead to understanding of the production pathways of the metabolites of interest. The objective of the present work was to trace the metabolic profile of the filtrate from the endophytic fungus Phomopsis sp., An isolate of Syzygium jambolanum DC, through the consumption of dextrose as a source of carbon and energy for 6 weeks of culture using the 1H NMR With water signal suppression by NOESY 1D. In addition, liquid column chromatography and thin layer chromatography were performed in order to isolate compounds from organic mycelium extract. The analyzes had the help of MatLab® software
for data normalization. Thus, it was possible to trace the metabolic profile of Phomopsis sp., cultivated under conditions of hypoxia, and verify the occurrence of reductive routes such as alcoholic fermentation and reductive Tricarboxylic Acid Cycle. It was also possible to observe the intensification of yellow pigmentation, characteristic of this genus of fungus, from the fourth week of cultivation, which is the period most of the primary metabolites are no longer present. By means of NMR experiments it was possible to elucidate a structure of a substance isolated from the chloroform extract of the fungus dry mycelium, 5-methylmethane. Based on these results, it can be inferred that, under these conditions, the microorganism studied performs alcoholic fermentation throughout the culture. It was also verified the occurrence of the reductive Tricarboxylic Acid Cycle in the cytosol until the third week of culture and, after this period, from the fourth week the secondary metabolism is intensified. It is the first time that the
secondary metabolite 5-methylmeleina is isolated from this genus of endophytic.<br>Coordenação de Aperfeiçoamento de Pessoal de Nível Superior<br>Microrganismos endofíticos, que são importantes fontes de compostos bioativos, vêm sendo amplamente estudados devido às propriedades que seus metabólitos apresentam e, em comparação com as plantas hospedeiras, são de mais fácil manipulação em laboratório. Visto que do gênero de fungo endofítico Phomopsis já foram isolados compostos com diferentes atividades biológicas, a investigação dos metabólitos, sejam eles primários ou secundários, presentes no filtrado de sua cultura é de extrema importância. O estudo metabolômico de Phomopsis sp. pode levar ao entendimento das vias de produção dos metabólitos de interesse. O presente trabalho teve como objetivo traçar o perfil metabólico do filtrado da cultura do fungo endofítico Phomopsis sp., um isolado de Syzygium jambolanum DC, através do consumo da dextrose como fonte de carbono e energia durante 6 semanas de cultivo utilizando a técnica de RMN 1H com supressão do sinal da água por NOESY 1D. O enfoque do estudo é. Além disso, foram realizadas cromatografia líquida em coluna e cromatografia em camada delgada com o intuito de isolar compostos a partir de extrato orgânico do micélio. As análises estatísticas tiveram o auxílio dos softwares MatLab® para normalização e quantificação relativa dos dados. Assim, foi possível traçar o perfil metabólico de Phomopsis sp, cultivado em condições de
hipóxia, e constatar a ocorrência de rotas redutivas, tais como fermentação alcoólica e Ciclo dos Ácidos Tricarboxílicos redutivo. Também foi possível observar a intensificação da pigmentação amarela, característica desse gênero de fungo, a partir da quarta semana de cultivo, período este que a maioria dos metabólitos primários já não se fazem mais presentes. Através de experimentos de RMN foi possível elucidar a estrutura de uma substância isolada do extrato em clorofórmio do micélio seco do fungo, a 5-metilmeleina. Com base nesses resultados, pode-se inferir que, sob estas condições, o microrganismo estudado realiza fermentação alcoólica durante todo o cultivo. Também foi verificada a ocorrência do Ciclo dos Ácidos Tricarboxílicos redutivo no citosol até a terceira semana de cultivo e, após esse período, a partir da quarta semana o metabolismo secundário é intensificado. É a primeira vez que o metabólito secundário 5-metilmeleina é isolado desse gênero de endofítico.

Para o estudo da emergência com disponibilidade hídrica, foram considerados três tratamentos: 60% (H1), 40% (H2) e 20% (H3) de sua capacidade de retenção de água. As sementes foram inoculadas artificialmente com Phomopsis sojae por contato dessas com o fungo por um período de 32 horas, posteriormente foram semeadas em caixas plásticas para a condução do teste de emergência com disponibilidade hídrica. A semeadura das quatro variedades (MSoy 8400, MSoy 8200, Embrapa 48 e Paraíso) combinadas com os três tratamentos de disponibilidade hídrica e o fungo, constituíram 24 tratamentos com quatro repetições. Cada caixa recebeu 50 sementes, essas foram mantidas em ambiente de laboratório. A avaliação da emergência das plântulas foi feita aos 10 dias após a semeadura e, ao final desse período, procedeu-se a avaliação da massa seca de plântulas de soja. Le phoma (Phoma macdonaldii / Leptosphaeria linquistii) et le phomopsis (Phomopsis / Diaporthe helianthi) sont deux champignons pathogènes majeurs du tournesol. Dans un contexte d’objectif de réduction de la lutte chimique, une meilleure compréhension des interactions peuplement-agents pathogènes-environnement-conduite de culture est nécessaire. Des essais menés au champ et en serre à Auzeville en 2010 et 2011 ont permis de décomposer les effets de la conduite de culture du tournesol sur l’apparition et le développement des deux maladies. Les variables clés de l’architecture des plantes en peuplement et du microclimat, modifiées par la conduite de culture (choix variétal, densité de semis, fertilisation azoté, irrigation) ont été décrites et mises en relation avec les composantes de la maladie. Ainsi, pour le phomopsis, le rôle déterminant du microclimat résultant du niveau de développement de la couverture foliaire lors des phases initiales d’infection a été confirmé. La taille des feuilles et le diamètre des tiges sont des caractères déterminants de la progression des dégâts sur feuille et tige. Les conséquences des attaques de phomopsis sur feuille en termes de mécanismes de nuisibilité ont été analysées finement et modélisées. Pour le phoma, le rôle du statut nutritionnel azoté dans la progression des attaques sur tige et collet a été confirmé. La fonction régulatrice du phoma vis-à-vis des attaques de phomopsis sur feuille a été quantifiée. Sur le plan de la modélisation, plusieurs approches ont été menées : (i) Evaluation de la qualité prédictive d’Asphodel, modèle épidémiologique prévoyant les émissions de ascospores de Phomopsis ; application à différentes conduites de culture et extension au phoma pour la partie inoculum primaire ; (ii) développement d’un modèle prédictif de l’incidence du phoma prenant en compte les effets climatiques, microclimatiques et agronomiques ; (iii) proposition d’une modélisation conceptuelle des interactions plante-agent(s) pathogène(s)-environnement-conduite de culture. Ce travail pourra, à terme, contribuer à la conception de stratégies de gestion des principales maladies du tournesol, à l’échelle de la parcelle ou du territoire<br>Phoma (Phoma macdonaldii / Leptosphaeria linquistii) and phomopsis (Phomopsis / Diaporthe helianthi) are two major sunflower fungal diseases. In a context of reduction of pesticides, a better understanding of the interactions between crop canopy, pathogens, environment and cropping practices is needed. Field and greenhouse experiments set up at Auzeville in 2010 and 2011 allowed to dissect the impact of sunflower crop management on disease appearance and development. Key variables of plant architecture and microclimate, modified by cropping practices (variety choice, sowing density, nitrogen fertilization, irrigation) were described and related to disease components. Thereby, for phomopsis, the key role of microclimate resulting from the level of development of leaf area during the initial stages of infection was confirmed. Leaf length and stem diameter are key characters for necrosis progression on leaf and stem. The impact of phomopsis leaf infection in term of damage mechanism was analysed and modelled. For phoma, the effect of nitrogen nutrition status on necrosis progression on stem and stem base was confirmed. The regulatory function of phoma towards phomopsis leaf infection was quantified. In terms of modelling, several approaches were developed: (i) evaluation of the predictive quality of Asphodel, an epidemiological model predicting Phomopsis ascospore release; application to different cropping practices and extension to phoma for primary inoculum; (ii) development of a predictive model for phoma incidence taking into account the climatic, microclimatic and agronomic effects; (iii) proposal of a conceptual model of the interactions between crop canopy, pathogens, environment and cropping practices. This work describes the isolation of endophytic fungi from plant species Hancornia speciosa and led to the isolation of 14 pure strains. Of which 4 were selected for the chemical study. The chemical study allowed identied 12 substances isolated from fungi Phomopsis sp., Guignardia sp., Aspergillus niger e Aspergillus sp., grown in different culture medium. The crude extracts of fungus Phomopsis sp., grown in PDB
medium led to isolation of five compounds, 5- methylmelein, nectriapyrone, succnic acid, 5-hydroxylmethylmelein and a diketopiperazine. Already when the same fungus was grown in ME medium, led to isolation of two compounds, tyrosol and
tryptofol. The crude extract of Guignardia sp grown in ME medium led to isolation of compound salicylic acid, which has a plant for ecological importance.The crude extract of Aspergillus niger, grown in PDB medium led to isolation of compounds pyrophen and nigragillin. Already when the same fungus was grown in ME medium, led to isolation of compound itaconic acid. And of crude extract of Aspergillus sp., grown in ME medium was identification of their major compound kojic acid, which was quantified by HPLC in the crude extract (0.7 mg/g of crude extract). O crude extract was tested for its antioxidant activity with DPPH, NO. and H2O2, and exhibited
significantly potent inhibition of NO (IC50 de 150 μg/mL) production, showed taht this fungi it owns big biotchenology potential. The structures of these substances were
established by spectroscopic methods, including the application of bidimensional NMR techniques, mass spectrum and comparison with published data.<br>Este trabalho descreve o isolamento de fungos endofíticos da espécie vegetal Hancornia speciosa, o qual conduziu a 14 linhagens puras, dos quais quatro foram selecionados para o estudo químico. O estudo químico possibilitou foi identicar 12
substâncias, isoladas dos fungos Phomopsis sp., Guignardia sp., Aspergillus níger e Aspergillus sp., cultivados em diferentes meios de culturas. O extrato bruto do fungo Phomopsis sp cultivado no meio liquido PBD levou ao isolamento de cinco compostos, 5- metilmeleína, nectapirona, ácido succínico, 5-hidroxilmetilmeleína e uma dicetopiperazina. Já o mesmo fungo quando cultivado no meio líquido-ME,
levou ao isolamento de dois compostos, tirosol e triptofol. O estudo do extrato bruto de Guignardia sp., cultivado no meio liquido ME conduziu ao isolamento do composto ácido salicílico, o qual tem um importancia ecológica para planta. O
extrato bruto do fungo Aspergillus niger cultivado no meio liquido PBD levou ao isolamento dos compostos pirofen e nigragillin. Já o mesmo fungo quando cultivado em extrato de malte, levou ao isolamento do ácido itacônico. E do extrato bruto do fungo Aspergillus sp., cultivado no meio liquido PBD foi identificada a substância majoritária ácido kójico, a qual foi quantificada por CLAE no extrato bruto (0,7g/ g de
extrato bruto). O extrato desse fungo foi testado quanto a sua atividade antioxidante com DPPH, NO. , e H2O2, e mostrou uma forte atividade de inibição da produção de
NO (IC50 de 150 μg/mL), indicando que este fungo possui grande potencial biotecnológico. As estruturas das substâncias foram determinadas por métodos espectrométricos, incluindo RMN bi-dimensional, espectros de massas e comparação com a literatura.

A diversidade de organismos oriundos do ambiente marinho constitui uma fonte significativa de substâncias estruturalmente inéditas e biologicamente ativas, dentre as quais, diversas inspiraram o desenvolvimento de novas classes de agentes terapêuticos. Neste contexto, macroalgas vermelhas do gênero Bostrychia (Rhodomelaceae) foram coletadas em praias do litoral norte do estado de São Paulo e têm sido objeto de estudos químicos e biológicos, no Laboratório de Química Orgânica do Ambiente Marinho (LQOAM - NPPNS) da FCFRPUSP, sob a supervisão da Profa. Dra. Hosana M. Debonsi. As algas da espécie Bostrychia radicans demonstraram potencial quando avaliadas as atividade citotóxica, tripanocida, leishmanicida e antimicrobiana; além de um perfil químico interessante, evidenciado pelo isolamento de substâncias inéditas na literatura. Neste contexto, o presente trabalho descreve a continuidade do estudo químico da espécie B. radicans, coletada no Manguezal do Rio Escuro, em Ubatuba-SP; bem como o potencial biológico desta espécie, além da avaliação da atividade de enzimas fenolsulfatases na espécie. Ainda, no sentido de explorar novas fontes promissoras para o isolamento de substâncias bioativas, este trabalho descreve o isolamento de micro-organismos endofíticos associados à espécie B. radicans. Foram isoladas 45 linhagens de micro-organismos; dentre as quais foram selecionadas nove linhagens para obtenção de extratos e realização de triagens química e biológica. A partir desta triagem inicial, foi realizado o estudo químico dos fungos Xylaria sp., Penicillium brevicompactum e Phomopsis longicolla. A partir da Xylaria sp. foram isoladas as seguintes substâncias: ácido 2,5-diidroxibenzóico, 8-diidroxinaftol 1-O-a-glucopiranosídeo, 8-metóxi-3-metil-1- isocromanona e ácido pilifórmico. O estudo químico de Penicillium brevicompactum resultou no isolamento das substâncias: ácido micofenólico, asperfenamato, brevianamida A, brevianamida C e brevianamida oxindol, substância inédita como produto natural. A partir de Phomopsis longicolla foi isolado o dicerandrol C. Este trabalho descreve ainda o potencial biológico de algumas destas substâncias isoladas. O estudo químico e biológico de microorganismos realizado no LQOAM estimulou a consolidação de uma colaboração com o Prof. Dr. Isidro C. Gonzalez, através da realização de um estágio de 12 meses no Laboratório Botrytis (Departamento de Química Orgânica, Universidade de Cádiz). Durante este período foi realizado o estudo químico do fitopatógeno Botrytis cinerea, visando o isolamento de novos metabólitos ou toxinas; além do estudo da biogênese destas substâncias, através de ensaios utilizando precursores isotopicamente marcados.<br>The diversity of organisms from the marine environment is a significant source of structurally novel and biologically active substances, several of which have inspired the development of new classes of therapeutic agents. In this context, red macroalgae belonging to Bostrychia genus (Rhodomelaceae) were collected on beaches of the north coast of São Paulo State and have been studied chemically and biologically in the Laboratory of Organic Chemistry of the Marine Environment - (LQOAM - NPPNS) at FCFRP-USP, under Prof. Hosana M. Debonsi supervision. Algae Bostrychia radicans species showed cytotoxic, trypanocidal, antileishmanial and antimicrobial potential, besides a interesting chemical profile, evidenced by the isolation of new compounds in the literature. In this context, this work describes the sequential chemical study of B. radicans species, collected at the Rio Escuro Mangrove, Ubatuba-SP, as well as the biological potential of this species. Also, the phenolsulphatases enzyme activity was evaluated in this species. Still, in order to explore new promising sources for the isolation of bioactive substances, this study describes the isolation of endophytic microorganisms associated to B. radicans. In this way, 45 strains of microorganisms were isolated and nine strains were selected for extracts preparation; and subsequently chemical and biological screenings. Based on the biological screening and chemical profile analyses, the large-scale fermentation of the endophytic fungi Xylaria sp., Penicillium brevicompactum and Phomopsis longicolla was carried out. The chromatographic purification of the bioactive acethyl acetate extract from Xylaria sp. allowed the isolation of 2,5-dihydroxybenzoic acid, 8- dihydroxynaphtol 1-O-a-glucopyranoside, 8-methoxy-3-methyl-1-isochromanone and piliformic acid. Chemical studies of Penicillium brevicompactum resulted in the isolation of: mycophenolic acid, asperphenamate, brevianamide A, brevianamide C and brevianamide oxindole, isolated for the first time as a natural product. From Phomopsis longicolla was isolated dicerandrol C. This thesis also describes the potential biological of some of these isolated compounds. The chemical and biological studies of microorganisms achieved in LQOAM encouraged the consolidation of a collaboration work with Prof. Dr. Isidro C. Gonzalez, through the completion of a 12-month internship at the Laboratory Botrytis (Department of Organic Chemistry, University of Cádiz). During this period, was conducted the chemical study of plant pathogen Botrytis cinerea, aiming the isolation of new metabolites or toxins, in addition to studying the biogenesis of these substances, through experiments using isotopically labeled precursors.

Assim, a otimização de culturas microbianas, pode ter efeitos significativos quando se quer aumentar o rendimento de um determinado metabólito. Este trabalho foi realizado para avaliar a influência de alguns fatores nutricionais e ambientais sobre a produção de compostos fenólicos (flavonoides e polifenóis), assim como foi conduzida a otimização das condições experimentais que resultassem em uma atividade antioxidante mais elevada dos extratos dos metabólitos produzidos pelo fungo Phomopsis sp., isolado a partir de folhas da Bauhinia variegata. Para a otimização da produção de polifenóis totais, flavonoides e da atividade antioxidante dos extratos (variáveis dependentes – respostas preditas), foi realizado um delineamento experimental fatorial 23 no modelo de faces centradas (DFC), com três variáveis independentes denominadas pH inicial do meio, temperatura de incubação (°C) e concentração de peptona no meio (g/L). As variáveis independentes foram codificadas em três níveis (-1, 0, +1). Utilizando-se a ferramenta de planejamento experimental com a metodologia de superfície de resposta (MSR) foi possível investigar a influência das variáveis independentes, assim como a forma de interação entre elas. O comportamento da produção de flavonoides e polifenóis, assim como da atividade antioxidante quando em diferentes condições ambientais foi eficientemente compreendido através de métodos estatísticos. Modelos polinomiais de segunda ordem foram utilizados para prever cada uma das três respostas. A análise de regressão da resposta atividade antioxidante mostrou que 93,9% da variação foi explicada pelo modelo. Estes resultados indicam que fatores nutricionais e ambientais desempenham um papel importante na produção de metabólitos secundários com atividade antioxidante pelo fungo Phomopsis sp. _______________________________________________________________________________________________ ABSTRACT<br>Variations in microbial growth environment can have significant impacts on the quantity and diversity of secondary metabolites produced. Thus, the optimization of microbial cultures may have significant effects when one wants to increase throughput of a particular metabolite. This study was conducted to evaluate the influence of nutrition and environmental factors on the production of phenolic compounds (polyphenols and flavonoids), as well as optimization of the experimental conditions that would result in a higher antioxidant activity on the extracts of the metabolites produced by the fungus Phomopsis sp. isolated from Bauhinia variegata leaves. For optimization of the production of total polyphenols, flavonoids as well as extracts with high antioxidant activity (dependent variables - predicted responses) a factorial experimental design 23 was performed using a face-centered cubic design (FCD) of experiments, with three independent variables, namely initial pH, incubation temperature (°C) and concentration of peptone (g/L). The independent variable were coded at three levels (-1, 0, +1). Using the experimental design tool with the response surface methodology (RSM) was possible to investigate the influence of the independent variables, as well as the form of interaction between them. Second order polynomial models were used to predict each of the three answers. The regression analysis of the antioxidant activity response showed that 93.9% of the variation was explained by the model. These results indicate that nutritional and environmental factors play an important role in production of secondary metabolites with antioxidant activity, which can be better understood and studied using mathematical and statistical methods. The production profile of flavonoids and polyphenols, as well as antioxidant activity of the extracts produced by Phomopsis sp. in different environmental conditions were efficiently predicted using statistical methods.

Ce travail concerne l'étude de nouvelles chaetoglobosines naturelles ou de synthèse. Les chaetoglobosines sont des toxines fongiques appartenant au groupe plus large des cytochalasanes (du grec cytos : cellule et chalasis : Relaxation) connues et étudiées surtout pour leur propriété d'inhiber la division de cellules de végétaux ou de mammifères. Le premier chapitre de cette thèse porte sur l'optimisation de la production de ce genre de toxines par différentes souches de champignon, notamment de phomopsis leptostromiformis, ainsi que sur leur isolement et leur purification chromatographique. Leur caractérisation spectroscopique à surtout été fondée sur les données de la rmn 1d, les expériences d'effet overhauser nucléaire (noe), la rmn 2d ainsi que la modélisation moléculaire. Deux nouvelles chaetoglobosines naturelles ont ainsi été isolées et caractérisées : la chaetoglobosine O et une chaetoglobosine pontée entre les positions c21 et c17 du macro cycle de la chaetoglobosine m. Dans le second chapitre traite de l'obtention et de la caractérisation de cinq chaetoglobosines non-naturelles préparées par des modifications simples de chaetoglobosines naturelles en vue d'en modifier éventuellement les propriétés biologiques. Tout d'abord, il est montré que la soude n'affecte pas le site époxydique de la chaetoglobosine m, mais qu'en revanche, elle en modifie son macro cycle par pontage entre les positions c21 - c17 deux chaetoglobosines pontées à jonction trans. Dénommées composes i et s ont ainsi été obtenues. Elles résultent d'une réaction de Michael intramoléculaire portant sur le site enonique c17 = c18 - c19 = o et présentant une remarquable regioselectivité puisque seul le site enolisable c20 = o - ch21 est implique. Le compose s a par ailleurs été identifie à la chaetoglobosine naturelle pontée entre les positions c21 et c17 du macro cycle. L'évaluation de la résitance au Phomopsis de différents génotypes de tournesol (Helianthus spp) nous a permis de déterminer quatre groupes de résistance: un groupe à résistance complète, un groupe à résistance partielle qui réagit tardivemnt à l'infection sur feuille, un groupe sensible présentant une résistance partielle à la pénétration du mycellium du champignon, et un groupe très sensible. Par infection artificielle avec le mycélium de Phomopsis déposé sur une feuille, nous avons d'une part confirmé que la résistance est de type horizontal et d'autre part caractérisé trois composantes de la résistance: vitesse maximale et finale de croissance des nécroses sur feuille, et vitesse de croissance des nécroses sur pétiole. Par infection semi-naturelle, nous avons révélé une composante indépendante ce celles évaluées pa infection artificielle, les taux d'attaque totaux sur tiges et feuilles à floraison. Nous avons alors construit une carte génétique de 309 marqueurs couvrant 2168,6 cM avec une distance moyenne entre marqueurs de l'ordre de 6. 1 cM, sur 171 lignées recombinantes issues du croisement intraspécifique HA89 (Sensible) X LR4 (Résistant), en utilisant des PCR spécifiques de gènes de résistance, des DALP (amplifications directes de polymorphismes de longueurs de fragments de restrictions). . .

L'objectif de cette etude est de contribuer a la mise au point des methodes de diagnostic sur semences par des techniques immunoenzymatiques. Deux modeles de parasites fongiques transmissibles par les semences ont ete etudies: le complexe diaporthe, phomopsis sur soja et peronospora pisi sur pois. Des serums ont ete produits contre les filtrats et les extraits myceliens de 5 souches (phomopsis longicolla, p. Phaseoli, diaporthe phaseolorum var. Batatatis, diaporthe p. Var. Caulivora, diaporthe p. Var sojae) et un isolat de phomopsis phaseoli. Aucun des douze serums ne s'est revele suffisamment specifique pour differencier les 5 especes entre elles. Les serums anti-filtrats reconnaissent bien les extraits myceliens. Les variations des profils, reveles apres immunotransfert, sont comparees avec ceux d'extraits proteiques d'autres champignons isoles du soja (alternaria sp. , aspergillus sp. Fusarium roseum, tricothecium sp. ) et phomopsis citri. Deux serums anti-peronospora pisi ont ete prepares (anti-conidies et anticonidiophores). Le serum anti-conidiophores (1cph) donne le meilleur niveau de reponse en elisa et en dot. Les plantes saines et les extraits de 16 champignons saprophytes et pathogenes du pois ne donnent pas de reaction croisee. En revanche, le serum reconnait plasmopara halstedii. Des essais pour la detection, en dot, de p. Pisi dans des semences de pois broyees ne sont pas concluants. No caso do teste de condutividade elétrica (CE) tem-se constatado efeito do patógeno sobre os resultados do mesmo. Assim, o trabalho teve como objetivo estudar a interferência dos fungos Phomopsis sojae e Colletotrichum dematium var. truncata nos resultados do teste em semente de soja. O trabalho foi conduzido usando-se sementes produzidas na safra 2005/2006. Estas foram inoculadas artificialmente com os fungos P. sojae e C. dematium var. truncata, utilizando-se meio de cultura BDA (batata, ágar e dextrose) acrescido de manitol (-1,0 MPa), por período de incubação de 20 horas, à temperatura de 25º C. Determinou-se o diâmetro de colônias, o índice de crescimento micelial; o teor de água da semente; a incidência de microorganismos (antes e após a inoculação) pelo teste de sanidade; o potencial fisiológico da semente pelos testes de germinação e de vigor (índice de velocidade de emergência, de frio, envelhecimento acelerado e condutividade elétrica) e a emergência de plântulas em campo. Determinou-se também a composição química da solução de embebição lixiviados (K+, CA+, Mg+ e Na+) e das sementes (K+, CA+, Mg+, Na+, P, C e N). Regarding the electrical conductivity test (EC) it has been verified the effect of some seedborn pathogens on the EC results. This research work was carried out in order to study the influence of the presence of Phomopsis sojae and Colletotrichum dematium var. truncata fungi on the EC results. The work was running using seeds produced in the agricultural year 2005/2006. Also, the composition of the soaked seed solution leachates (K+, Ca2+, Mg2+, Na+) and the seed (K+, Ca2+, Mg2+, Na+, P, C and N) were determined. A completely design in factorial 2 x 4 (two seed size 5.5 and 6.5 mm x four seed treatments purity seeds, seeds incubated in BDA without fungi, seeds incubated in BDA with Phomopsis and seeds incubated in BDA with Colletotrichum) arrangement, with eigth (5.5 mm) and six (6.5 mm) replications was used. The infection of soybean seeds with Phomopsis resulted on physiological seed quality reduction, but it did not xiii happen when the seeds were inoculated with Colletotrichum. There is no effect of seed inoculation in function of seed size. Based on the results is can be concluded that the occurrence of Phomopsis sojae fungi can interfere on the electrical conductivity results, but the same did not happen for the Colletotrichum dematium var. truncata fungi.

Les objectifs de ce travail sont de contribuer à la connaissance du déterminisme génétique de la résistance à Phompsis/Diaporthe helianthi Munt-Cvet et al. Chez le tournesol et de mettre au point une méthode de sélection alternative aux méthodes classiques. La culture in vitro de cellules et de tissus peut être utilisée comme outil de sélection rapide, simple et fiable. Les techniques in vitro mises en oeuvre dans ce travail sont basées sur la formation de cals et sur la viabilité des protoplastes, en réponse à des filtrats de culture partiellement purifiés de P. Helianthi, utilisés comme agents sélectifs. Le déterminisme génétique de la résistance au phomopsis en conditions de contamination naturelle, de la formation des cals et de la viabilité des protoplastes chez le tournesol, ainsi que la modification de ces deux derniers paramètres par les filtrats du pathogène, ont été étudiés dans un dispositif de plan factoriel. Nos résultats montrent que la résistance des plantes adultes de tournesol au phomopsis en conditions naturelles est controlée essentiellement par des gènes à effets additifs avec cependant des effets de dominance partielle. D'autre part, nous avons établi que la formation de cals est un paramètre à forte variabilité génétique, qui semble controlé principalement par des gènes à effet additifs, bien que les interactions inter-alléliques soient également significatives. L'apport de filtrats de culture à la dose de 5% a globalement entraîné une augmentation de la formation de cals. L'étude du niveau de mortalité des protoplastes après 48 h de culture montre que celle-ci varie en fonction du génotype et augmente significativement en présence du filtrat de culture. Le contrôle génétique de la mortalité des protoplastes, aussi bien sur le milieu témoin que sur le milieu sélectif, est apparu simplement additif. Il ressort également de cette étude que la présence de filtrats du pathogène dans le milieu de culture modifie l'expression des gènes impliqués aussi bien dans la formation des cals que dans la viabilité des protoplastes, ce qui se traduit par des valeurs différentes des aptitudes générales à la combinaison et des aptitudes spécifiques à la combinaison de plusieurs lignées et hybrides, par rapport au milieu témoin.

La tubuline est une protéine essentielle de la cellule. En polymérisant sous forme de microtubules, elle crée notamment le fuseau mitotique le long duquel migrent les chromosomes pendant la mitose. Les médicaments qui inhibent la polymérisation et/ou la dépolymérisation de la tubuline sont des composés majeurs de la thérapie anticancéreuse. Les vinca-alcaloïdes en sont des représentants importants. Ils induisent la mort des cellules par apoptose, en inhibant la dynamique des microtubules. D'autres molécules d'origine naturelle, comme la phomopsine A, se fixent sur la tubuline à proximité ou dans le même site de fixation que celui des vinca-alcaloïdes. C'est la raison pour laquelle nous avons envisagé d'élaborer des composés antimitotiques hybrides entre la vinblastine et la phomopsine A. Dans ce contexte, deux séries de composés ont été conçues. La première série d'hybrides correspondant à des dérivés de l'anhydrovinblastine fonctionnalisés en position 7'. Cependant, aucune des trois stratégies étudiées n'a permis d'accéder à ces composés. La deuxième série d'hybrides, dérivés de la 7'-homo-anhydrovinblastine a pu être synthétisée grâce à une réaction originale d'insertion d'acétylènes activés au niveau du pont gramine de la vinorelbine, suivie d'une réduction avec un contrôle totale de la régio- et stéréoselectivité. Dans un premier temps, les réactions d'insertion et de réduction ont été mise au point. Ensuite, deux familles d'hybrides portant la chaîne latérale de l'octahydrophomopsine en position 8' ou 7' ont été synthétisés. La plupart des composés ainsi obtenus possédent une excellente activité sur la tubuline et sont très cytotoxique.

Lupinosis is a mycotoxicosis caused by the ingestion of phomopsin toxins produced by the fungus Phomopsis leptostromiformis Kuhn (Bubak) which colonises lupin plants. Lupinosis is recognised primarily as an hepatic disease of sheep, however it is also associated with the occurrence of a myopathy. The myopathy is termed lupinosis associated myopathy (LAM) and is histologically indistinguishable from vitamin E/selenium responsive nutritional myopathy. Despite repeated attempts by others, LAM is nonresponsive to conventional selenium and vitamin E therapy. The main aim of this work was to investigate the relationship between LAM, selenium and vitamin E.
In order to correctly assess and stage the hepatic pathology associated with exposure to phomopsins, the biochemical indicators of hepatocellular damage and hepatic function were examined. BSP clearance, plasma bilirubin concentration, plasma AST activity and feed intake were the best indicators of concurrent hepatic pathology of individual sheep exposed to phomopsins. BSP clearance, bile acids and plasma bilirubin concentration provided the most accurate indication of the concurrent hepatic status of flocks of sheep grazing lupin stubble. Although plasma AST activity was previously thought to be an adequate indicator of the degree of active hepatocellular damage in a flock, its usefulness was dependent on the duration and the degree of intoxication.
The effect of lupinosis on selenium metabolism in sheep was examined. Phomopsin A intoxication significantly affected the storage and metabolism of selenium in sheep. This was a specific effect of phomopsins. Exposure to phomopsins significantly altered the whole blood and plasma pharmacokinetics, tissue distribution and excretion patterns of 75 Se-selenite and 75 Se-selenomethionine when given orally or intravenously. Furthermore these alterations affected the liver's capacity to produce selenoproteins, thereby changing the supply and metabolism of the body's selenium pool. Consequently the profound change in selenium metabolism resulted in muscle pathology. In contrast to this, the bioavailability of selenium from an intramuscular dose of either 75 Se-selenomethionine and 75 Se-selenite was unaffected by exposure to phomopsin toxin. This work was the first to monitor and relate alterations in selenium kinetics and metabolism to a specific pathological process.
A field trial was carried out to determine the effectiveness of intramuscular selenite and selenomethionine with and without vitamin E supplementation in preventing LAM. Pre-treatment of sheep with both selenomethionine and vitamin E prior to exposure to toxic lupin stubble significantly reduced the incidence of LAM. This was the first effective treatment of LAM. Selenite and vitamin E supplementation, either alone or in combination was not effective. Selenomethionine supplementation alone was also unsuccessful. The apparent necessity for both selenomethionine and vitamin E indicates that there is a significant metabolic and functional interaction between the two nutrients during the pathogenesis of LAM.

Sulla base di quanto premesso, lo scopo principale di questa tesi è stato quello di identificare e caratterizzare l'agente causale/i dei difetti del nocciolo e di definire un protocollo di gestione del problema, secondo i principi IPM. È stata quantificata l'incidenza dei generi fungini isolati da nocciole provenienti da diverse aree geografiche (Caucaso e Turchia), raccolte in diversi anni e in diverse fasi di crescita della coltura. Diaporthe è stato identificato come il principale genere fungino associato ai difetti della nocciola, e la caratterizzazione molecolare ha confermato il ruolo predominante della specie D. eres. Sono state studiate le caratteristiche ecologiche di questo fungo e l'efficacia in vitro di fungicidi chimici e biologici per controllarne la crescita miceliale, lo sviluppo di picnidi e la formazione di cirri.

The incidence of fungal genera in hazelnuts coming from different geographical areas (Caucasus and Turkey), collected in different years at different crop growth stages, was quantified. Diaporthe was identified as the main genus involved in hazelnut defects, and the molecular characterization confirmed the predominant role of D. eres. Its ecological needs were investigated, and the in vitro efficacy of chemical and biological fungicide were screened to control D. eres growth, pycnidial conidiomata development and cirrhi occurrence. This would be an essential tool for a sustainable crop protection approach in this economically important crop
