In this paper, the purification and biochemical characterization of the endogenous oyster (Crassostrea gigas) trypsin inhibitor were researched. A oyster trypsin inhibitor(OTI)has been purified by successive ammonium sulfate precipitation, gel filtration, affinity chromatography and high performance reversed-phase liquid chromatography. OTI has a molecular weight of approximately 5036 Da estimated by high performance size exclusive liquid chromatography. OTI was heat-, acid- and basic-stable competitive trypsin inhibitor. And OTI was double-head inhibitor with the inhibition constant (Ki) value of 1.644×10-2 mmol L-1. OTI was composed of nine kinds of amino acid, and rich in cysteine, alanine and glutamic acid. ABSTRACT Strategies to prime CD8+ T cells against Murine gammaherpesvirus 68 (γHV68; MHV68) latency have, to date, resulted in only limited effects. While early forms of latency (<21 days) were significantly reduced, effects were not seen at later times, indicating loss of control by the primed CD8+ T cells. In the present study, we evaluated CD8+ T cells in an optimized system, consisting of OTI T-cell-receptor (TCR) transgenic mice, which generate clonal CD8+ T cells specific for Kb-SIINFEKL of OVA, and a recombinant γHV68 that expresses OVA (γHV68.OVA). Our aim was to test whether this optimized system would result in more effective control not only of acute infection but also of later forms of latent infection than was seen with previous strategies. First, we show that OTI CD8+ T cells effectively controlled acute replication of γHV68.OVA in liver, lung, and spleen at 8 and 16 days after infection of OTI/RAG mice, which lack expression of B and CD4+ T cells. However, we found that, despite eliminating detectable acute replication, the OTI CD8+ T cells did not prevent the establishment of latency in the OTI/RAG mice. We next evaluated the effectiveness of OTI T cells in OTI/B6 animals, which express B cells—a major site of latency in wild-type mice—and CD4+ T cells. In OTI/B6 mice OTI CD8+ T cells not only reduced the frequency of cells that reactivate from latency and the frequency of cells bearing the viral genome at 16 days after infection (similar to what has been reported before) but also were effective at reducing latency at 42 days after infection. Together, these data show that CD8+ T cells are sufficient, in the absence of B cells and CD4+ T cells, for effective control of acute replication. The data also demonstrate for the first time that a strong CD8+ T-cell response can limit long-term latent infection.

Purpose The purpose of this paper is to investigate the integrative effects of abusive supervision (AS), perceived injustice (PI) and ethical conflict (EC) on occupational turnover intentions (OTI) among frontline employees (FLEs). Furthermore, the mediating role of quiescent silence (QS) is examined. Design/methodology/approach The authors collected survey-based data from 320 FLEs working in different service sector organizations of Pakistan. The snow-ball sampling technique is employed to approach respondents of this study. The collected data are analyzed through structural equation modeling (SEM) technique using SmartPLS 3.0. Findings The AS, PI and EC are found to collectively predict OTI among FLEs with a mediating effect of QS. The EC is the strongest predictor of OTI. Practical implications The results imply that service managers must ensure that employees are treated fairly. Furthermore, relevant trainings are offered to FLEs in order to maintain the required ethical standards at work to reduce EC perceptions. Originality/value The study is pioneer in presenting the relationship between QS and occupational turnover among FLEs. Furthermore, the study advocates the mediating role of employee quiescent silence to enrich the relationship between socio-psychological factors (i.e. AS, EC and PI) and OTI.

Unmanned aerial vehicles (UAV), also known as drones, are gaining more and more practical application in modern society, particularly as the tools of implementation of the concepts of «smart city», «smart health care», «smart industries», Internet of Things, 3D mapping, digital transport. But currently it is impossible to use of UAV at certain objects, comprising objects of transport infrastructure (OTI), primarily, airports, because of existing restrictions due to security threats arising during the UAV flight.The authors of the present work have set a goal to offer a solution that allows to start operating UAV at transport infrastructure facilities that are currently prohibited for UAV flights. To achieve the objective of the work, using analysis and synthesis, comparison and generalization, factors and conditions for safe use of UAV at OTI have been formulated, a method for increasing security of UAV flight has been developed, followed by the suggestion on the UAV route control system.The proposed system makes it possible to safely use UAVs at OTI by restraining their flight area strictly to the designated corridor, which eliminates a threat of a collision of UAV with other vehicles operated at OTI, dangerous elements of OTI, as well as with people at the object.The system does not need electric power feeding, which makes it possible to implement the system without creating an auxiliary power supply infrastructure.The practical application of the proposed system and, as a consequence, implementation of a greater number of opportunities for the use of UAV, are capable to generate fundamentally new technological processes and structures at transport facilities, which is one of the directions for creating the next generation transport infrastructure based on IoT and artificial intelligence.

Advanced age or preexisting comorbidities have been characterized as risk factors for severe coronavirus disease 2019 (COVID-19) cases requiring hospitalization and intensive care. In recent years, clonal hematopoiesis (CH) of indeterminate potential (CHIP) has emerged as a risk factor for chronic inflammatory background and subsequent aging-associated diseases. The purpose of this study was to identify biological factors (particularly leukocyte subtypes and inflammatory markers) associated with a risk of clinical deterioration (i.e., orotracheal intubation (OTI)) and to determine whether CH was likely to influence clinical and biological behavior in patients with severe COVID-19 requiring hospitalization. Here, we describe clinical and biological features, including the screening of CHIP mutants in a well-annotated cohort of 122 hospitalized patients with a laboratory-confirmed diagnosis of COVID-19 (55% requiring OTI). We showed that elevated white blood cell counts, especially neutrophils and high C-reactive protein (CRP) levels at admission, were associated with an increased requirement of OTI. We noticed a high prevalence of CH (25%, 38%, 56%, and 82% of patients aged <60 years, 60–70 years, 70–80 years, and >80 years) compared to a retrospective cohort of patients free of hematological malignancy explored with the same pipelines (10%, 21%, 37%, and 44%). However, the existence of CH did not significantly impact clinical outcome, including OTI or death, and did not correlate with other laboratory findings.

Rubble islands are dynamic sedimentary features present on reef platforms that evolve under a variety of morphodynamic processes and controlling mechanisms. They provide valuable inhabitable land for small island nations, critical habitat for numerous species, and are threatened by climate change. Aiming to investigate the controlling mechanisms dictating the evolution of One Tree Island (OTI), a rubble island in the Southern Great Barrier Reef, we combined different remotely-sensed data across varying timescales with wave data extracted from satellite altimetry and cyclone activity. Our findings show that (1) OTI had expanded by 7% between 1978 and 2019, (2) significant gross planform decadal adjustments were governed by the amount, intensity, proximity, and relative position of cyclones as well as El Niño Southern Oscillation (ENSO) phases, and (3) the mechanisms of island growth involve rubble spits delivering and redistributing rubble to the island through alongshore sediment transport and wave overtopping. Frequent short-term monitoring of the island and further research coupling variations in the different factors driving island change (i.e., sediment availability, reef-wave interactions, and extreme events) are needed to shed light on the future trajectory of OTI and other rubble islands under a climate change scenario.

Abstract This study evaluated the effects of a pyramidal training approach that used an expert trainer who taught teachers how to train their paraeducators. Three special education teachers were taught to train four paraeducators to provide students with intellectual and developmental disabilities opportunities to initiate (OTI). A multiple baseline design across participants was used to evaluate the rate and fidelity that paraeducators provided OTI and least to most prompting strategies with students. Rates increased from 0 to an average of .58 per minute. Fidelity of implementation increased from 0% to an average of 94.5%. Maintenance data were recorded for three paraeducators. Schools should consider using this cost- and time-effective training model with staff.

Abstract Background Scabies is a neglected tropical disease. In resource-poor settings, scabies and other skin infections are often unreported to a health centre, or misdiagnosed. Dermatological expertise and training are often lacking. Little is known about patient healthcare-seeking behaviour. This study reviewed diagnosed skin infections reported to urban (Greater Accra) and rural (Oti region) study health centres in Ghana over six months in 2019. Methods Study staff received classroom and clinical dermatology training. Skin infection diagnoses and anonymised patient information were recorded. Descriptive statistics and spatial analysis described patient demographics, and distance travelled to clinic, noting bypassing of their nearest centre. Results Overall, 385 cases of skin infections were reported across the Greater Accra and Oti study clinics, with 45 scabies cases (11.6%). For scabies, 29 (64.4%) cases were in males. Scabies was the third most common diagnosis, behind bacterial dermatitis (102, 26.5%) and tinea (75, 19.5%). In the rural Oti region, 48.4% of patients bypassed their nearest clinic, travelling a mean 6.2 km further than they theoretically needed to. Females travelled further in comparison to males. Conclusions There must be greater public and professional awareness of scabies and skin infections as high-burden but treatable conditions, along with assessment of their community burden.

Abstract Abstract 3235 T cell development relies on the interaction between the T-cell receptor (TCR) on thymocytes and the self-major histocompatibility complex (MHC) expressed on thymic epithelial cells in the thymus. This process, called positive selection, rescues developing thymocytes from cell death while leading to their differentiation into mature T cells. Since it is believed that the proper development of CD8 T cells requires an intact thymus, several groups studied their development using fetal or reaggregation thymus organ cultures in vitro. Unfortunately, these models were shown to be cumbersome requiring a complicated set-up while generating limited cellular yield. Thus, we sought of developing a novel in vitro system using bone marrow-derived stromal cells to support CD8 T cell development and maturation in vitro. We selected the OTI system as a working model due to the availability of previously identified positively selecting peptides. Non-selected T-cell-committed double-positive (DP) OTI thymocytes (CD4+CD8+CD69−) were first fractionated based on the surface expression intensity of both TCR and CD5. These 3 subsets designated as TCRloCD5lo (DP1), TCRintCD5hi (DP2), and TCRhiCD5int (DP3) express different levels of ZAP70. Following fractionation, the DP subsets were co-cultured with bone marrow-derived stromal cells presenting OTI-selecting peptides. In the absence of cytokines, no CD8+ OTI cell development occurred in vitro. When repeated in the presence of γc-cytokines (IL2, IL4, IL7, IL9, IL15 and IL21) only rIL4 and rIL7 were able to induce CD8 T cell development. Supplementing the co-culture system with rIL4 led to the generation of 50–60% single-positive (SP) CD8 T cells only from the DP3 fraction whereas rIL7 induced the development of a minor fraction of CD8 T cells from DP2s (3–4%) and a major population from DP3 (50–76%). Furthermore, we found that rIL4 treatment triggers the development of 2 distinct populations of SP OTI cells (based on their CD8 expression intensity) which we termed CD8int and CD8hi. When analyzed by flow-cytometry, ex vivo generated CD8int, but not CD8hi, expressed high levels of CD69, PD-L1 and CD44. In contrast, SP CD8 T cells developed in the presence of rIL7 did not upregulate these markers. Since IL7 promotes survival and proliferation of TCR-triggered DPs while IL4 affects their differentiation, we admixed both cytokines during the co-culture and found a dominant rIL4 effect: the phenotype of SP CD8 T cells was similar to that induced by rIL4 alone. Taken together, our findings demonstrate that some DP thymocytes are efficiently selected in our system by OTI-specific positively selecting peptides. Notably, the addition of rIL7 leads to the development and maturation of classic CD8 T cells whereas rIL4 induces both classic and innate CD8 T cells. This work was supported by grant a from CIHR. The present paper investigates the use of oti, na and negation in wh-question production in L1 Greek. Children’s preference is explored for use of oti and na, and for use of the negation markers ðen and min. These elements have been extensively studied from a theoretical perspective, yet they remain poorly investigated from an acquisition perspective, hence the present study. In long-distance wh-questions na is predicted to be preferred over oti due to its stronger entrenchment as clause-introducing element and as mood marker; in short-distance questions, however, na is predicted to be less preferred than the indicative due to the enriched modal semantics it carries in matrix clause environments. In negative matrix questions ðen is expected to be the preferred choice, since min occurs with na, which carries an extra semantic/pragmatic load. To test these predictions, a group of ninety four-to-seven-year-old Greek children participated in elicited production tasks designed mainly along the methodological principles of Crain and Thornton (1998). The results were generally in line with initial expectations. Children resorted mostly to na in long-distance contexts and to the semantically simpler indicative questions in short-distance contexts. With negative questions, higher accurate use rates were attested for target ðen than for target min, reflecting the simpler semantics associated with the former. Overall, these findings provide evidence that children opt for economy, with semantic factors contributing to their economy-based choices.

Abstract Understanding how extreme rainfall is changing locally is a useful step in the implementation of efficient adaptation strategies to negative impacts of climate change. This study aims to analyze extreme rainfall over the middle Oti River Basin. Ten moderate extreme precipitation indices as well as heavy rainfall of higher return periods (25, 50, 75, and 100 years) were calculated using observed daily data from 1921 to 2018. In addition, Mann–Kendall and Sen's slope tests were used for trend analysis. The results showed decreasing trends in most of the heavy rainfall indices while the dry spell index exhibited a rising trend in a large portion of the study area. The occurrence of heavy rainfall of higher return periods has slightly decreased in a large part of the study area. Also, analysis of the annual maximum rainfall revealed that the generalized extreme value is the most appropriate three-parameter frequency distribution for predicting extreme rainfall in the Oti River Basin. The novelty of this study lies in the combination of both descriptive indices and extreme value theory in the analysis of extreme rainfall in a data-scarce river basin. The results are useful for water resources management in this area.
