This chapter discusses the dynamics of bimolecular collisions within the framework of (quasi-)classical mechanics as well as quantum mechanics. The relation between the cross-section and the reaction probability, which can be calculated theoretically from a (quasi-)classical or quantum mechanical description of the collision, is described in terms of classical trajectories and wave packets, respectively. As an introduction to reactive scattering, classical two-body scattering is described and used to formulate simple models for chemical reactions, based on reasonable assumptions for the reaction probability. Three-body (and many-body) quasi-classical scattering is formulated and the numerical evaluation of the reaction probability is described. The relation between scattering angles and differential cross-sections in various frames is emphasized. The chapter concludes with a brief description of non-adiabatic dynamics, that is, situations beyond the Born–Oppenheimer approximation where more than one electronic state is in play. A discussion of the so-called Landau–Zener model is included.

This chapter discusses bimolecular reactions from both a microscopic and macroscopic point of view. The outcome of an isolated reactive scattering event can be specified in terms of an intrinsic fundamental quantity, the reaction cross-section that can be measured in a molecular beam experiment. It depends on the quantum states of the molecules as well as the relative velocity of reactants and products. The relation between the cross-section and the macroscopic rate constant is derived. The rate constant is a weighted average of the product between the relative speed of the reactants and the reaction cross-section. The chapter concludes with the special case of thermal equilibrium, where the velocity distributions for the molecules are the Maxwell–Boltzmann distribution. The expression for the rate constant at temperature T is reduced to a one-dimensional integral over the relative speed of the reactants.

Direct approaches to the calculation of the rate constant that bypasses the detailed state-to-state reaction cross-sections are presented, including transition-state theory, which plays an important role in practice. The second part gives a comprehensive discussion of basic theories of reaction dynamics in condensed phases, including Kramers and Grote–Hynes theory for dynamical solvent effects. Examples and end-of-chapter problems are included in order to illustrate the theory and its connection to chemical problems. In a chemical system with many chemical species several questions can be asked: what species react with other species: in what temporal order: and with what results? These questions have been asked for over one hundred years about simple and complex chemical systems, and the answers constitute the macroscopic reaction mechanism. In Determination of Complex Reaction Mechanisms authors John Ross, Igor Schreiber, and Marcel Vlad present several systematic approaches for obtaining information on the causal connectivity of chemical species, on correlations of chemical species, on the reaction pathway, and on the reaction mechanism. Basic pulse theory is demonstrated and tested in an experiment on glycolysis. In a second approach, measurements on time series of concentrations are used to construct correlation functions and a theory is developed which shows that from these functions information may be inferred on the reaction pathway, the reaction mechanism, and the centers of control in that mechanism. A third approach is based on application of genetic algorithm methods to the study of the evolutionary development of a reaction mechanism, to the attainment given goals in a mechanism, and to the determination of a reaction mechanism and rate coefficients by comparison with experiment. Responses of non-linear systems to pulses or other perturbations are analyzed, and mechanisms of oscillatory reactions are presented in detail. The concluding chapters give an introduction to bioinformatics and statistical methods for determining reaction mechanisms.

All other things being equal, physiological reaction rate increases roughly exponentially with temperature. Organisms that have adapted over evolutionary time to live at different temperatures can have enzyme variants that exhibit similar kinetics at the temperatures to which they have adapted to operate. Within species whose distribution covers a range of temperatures, there may be differential expression of enzyme variants with different kinetics across the distribution. Enzymes adapted to different optimum temperatures differ in their amino acid sequence and thermal stability. The Gibbs energy of activation tends to be slightly lower in enzyme variants adapted to lower temperatures, but the big change is a decrease in the enthalpy of activation, with a corresponding change in the entropy of activation, both associated with a more open, flexible structure. Despite evolutionary adjustments to individual enzymes involved in intermediary metabolism (ATP regeneration), many whole-organism processes operate faster in tropical ectotherms compared with temperate or polar ectotherms. Examples include locomotion (muscle power output), ATP regeneration (mitochondrial function), nervous conduction and growth.

Books dealing with the mechanisms of enzymatic reactions were written a generation ago. They included volumes entitled Bioorganic Mechanisms, I and II by T.C. Bruice and S.J. Benkovic, published in 1965, the volume entitled Catalysis in Chemistry and Enzymology by W.P. Jencks in 1969, and the volume entitled Enzymatic Reaction Mechanisms by C.T. Walsh in 1979. The Walsh book was based on the course taught by W.P. Jencks and R.H. Abeles at Brandeis University in the 1960's and 1970's. By the late 1970's, much more could be included about the structures of enzymes and the kinetics and mechanisms of enzymatic reactions themselves, and less emphasis was placed on chemical models. Walshs book was widely used in courses on enzymatic mechanisms for many years. Much has happened in the field of mechanistic enzymology in the past 15 to 20 years. Walshs book is both out-of-date and out-of-focus in todays world of enzymatic mechanisms. There is no longer a single volume or a small collection of volumes to which students can be directed to obtain a clear understanding of the state of knowledge regarding the chemicals mechanisms by which enzymes catalyze biological reactions. There is no single volume to which medicinal chemists and biotechnologists can refer on the subject of enzymatic mechanisms. Practitioners in the field have recognized a need for a new book on enzymatic mechanisms for more than ten years, and several, including Walsh, have considered undertaking to modernize Walshs book. However, these good intentions have been abandoned for one reason or another. The great size of the knowledge base in mechanistic enzymology has been a deterrent. It seems too large a subject for a single author, and it is difficult for several authors to coordinate their work to mutual satisfaction. This text by Perry A. Frey and Adrian D. Hegeman accomplishes this feat, producing the long-awaited replacement for Walshs classic text.

Adverse drug reactions are implicated in more than 40% of anaesthesia-related deaths. Undoubtedly, many more patients experience morbidity from adverse drug reactions. Widely cited definitions of adverse drug reactions encompass common side-effects but this chapter focuses on those that are unexpected reactions to drugs administered by anaesthetists and that occur at normal drug doses. The chapter includes a comprehensive account of malignant hyperthermia, which remains a major contributor to anaesthesia related to deaths. Malignant hyperthermia is a pharmacogenetic condition triggered by potent inhalational anaesthetics and possibly also suxamethonium. The genetic basis, pathophysiology, clinical presentations, and management of malignant hyperthermia are discussed. The chapter covers two other pharmacogenetic conditions: the acute porphyrias and butyrylcholinesterase (plasma cholinesterase) deficiency. Drug-induced anaphylaxis is another cause of perioperative morbidity and mortality. Recent data indicate that anaphylaxis during anaesthesia may be much more common than previously thought. The other type of adverse drug reaction covered in the chapter is serotonin syndrome. Perioperative serotonin syndrome is most likely to occur when patients who are already taking serotonergic drugs, such as selective serotonin reuptake inhibitor antidepressants or ‘triptan’ antimigraine treatments, are given a second drug with serotonergic properties, such as tramadol.

This chapter brings together the strands of theory and research discussed previously to introduce our theory on inequality and morality. The general argument is that in societies with greater economic inequality, the negative sanctioning-based moral emotions of anger, contempt, disgust, shame, and the like will be more frequent and severe. Societies with lower levels of inequality will conversely normalize and exhibit the more positive moral emotions of self-transcendence (compassion, praise, and empathy). Inequality thus begets negative moral emotions. These various emotional reactions to moral events not only affect everyday interaction, but also overlap with criminal justice systems’ reactions to those who offend societies’ moral codes. The more negative the moral reaction in a society, the more likely events are to prompt feelings associated with condemning others, the more the criminal justice system will be similarly focused on sanctioning as opposed to rehabilitation.

Topics covered include: (i) the development of ab initio potential-energy surfaces (PES) for complex multichannel systems using modified novelty sampling and feedforward NNs; (ii) methods for sampling the configuration space of critical importance, such as trajectory and novelty sampling methods and gradient fitting methods; (iii) parametrization of interatomic potential functions using a genetic algorithm accelerated with a NN; (iv) parametrization of analytic interatomic potential functions using NNs; (v) self-starting methods for obtaining analytic PES from ab inito electronic structure calculations using direct dynamics; (vi) development of a novel method, namely, combined function derivative approximation (CFDA) for simultaneous fitting of a PES and its corresponding force fields using feedforward neural networks; (vii) development of generalized PES using many-body expansions, NNs, and moiety energy approximations; (viii) NN methods for data analysis, reaction probabilities, and statistical error reduction in chemical reaction dynamics; (ix) accurate prediction of higher-level electronic structure energies (e.g. MP4 or higher) for large databases using NNs, lower-level (Hartree-Fock) energies, and small subsets of the higher-energy database; and finally (x) illustrative examples of NN applications to chemical reaction dynamics of increasing complexity starting from simple near equilibrium structures (vibrational state studies) to more complex non-adiabatic reactions. The monograph is prepared by an interdisciplinary group of researchers working as a team for nearly two decades at Oklahoma State University, Stillwater, OK with expertise in gas phase reaction dynamics; neural networks; various aspects of MD and Monte Carlo (MC) simulations of nanometric cutting, tribology, and material properties at nanoscale; scaling laws from atomistic to continuum; and neural networks applications to chemical reaction dynamics. It is anticipated that this emerging field of NN in chemical reaction dynamics will play an increasingly important role in MD, MC, and quantum mechanical studies in the years to come.

Topics covered include: (i) the development of ab initio potential-energy surfaces (PES) for complex multichannel systems using modified novelty sampling and feedforward NNs; (ii) methods for sampling the configuration space of critical importance, such as trajectory and novelty sampling methods and gradient fitting methods; (iii) parametrization of interatomic potential functions using a genetic algorithm accelerated with a NN; (iv) parametrization of analytic interatomic potential functions using NNs; (v) self-starting methods for obtaining analytic PES from ab inito electronic structure calculations using direct dynamics; (vi) development of a novel method, namely, combined function derivative approximation (CFDA) for simultaneous fitting of a PES and its corresponding force fields using feedforward neural networks; (vii) development of generalized PES using many-body expansions, NNs, and moiety energy approximations; (viii) NN methods for data analysis, reaction probabilities, and statistical error reduction in chemical reaction dynamics; (ix) accurate prediction of higher-level electronic structure energies (e.g. MP4 or higher) for large databases using NNs, lower-level (Hartree-Fock) energies, and small subsets of the higher-energy database; and finally (x) illustrative examples of NN applications to chemical reaction dynamics of increasing complexity starting from simple near equilibrium structures (vibrational state studies) to more complex non-adiabatic reactions. The monograph is prepared by an interdisciplinary group of researchers working as a team for nearly two decades at Oklahoma State University, Stillwater, OK with expertise in gas phase reaction dynamics; neural networks; various aspects of MD and Monte Carlo (MC) simulations of nanometric cutting, tribology, and material properties at nanoscale; scaling laws from atomistic to continuum; and neural networks applications to chemical reaction dynamics. It is anticipated that this emerging field of NN in chemical reaction dynamics will play an increasingly important role in MD, MC, and quantum mechanical studies in the years to come.

Food allergy is an immune response to food that can be classified as immunoglobulin (Ig)-E and non-IgE mediated. Milk, egg, peanut, tree nuts, and fish are among the most prevalent causes of food allergy. A detailed clinical history is essential when making a diagnosis, and skin prick testing and quantitative measurement of food-specific IgE antibodies can be helpful. Cow milk protein allergy causes a plethora of symptoms and frequently resolves spontaneously over the first 2 years of life; diagnosis is based mainly on clinical history. Food challenges have a pivotal role in the diagnosis of food allergy. Introduction of ‘allergic’ foods at 3–6 months alongside continuing breastfeeding may prevent allergy.

The 1973 paper by Jacquet and Lajtha, was a pivotal report in understanding the effects of opioids on nociception, as it investigated the effects of morphine on different brain structures. To do this, the authors used a microinjection technique that allowed them to specifically target subcortical sites. The animals used in this study were rats, and evaluation of the nociceptive threshold was based on the behavioural reaction to electrical shock. Two reactions were evaluated: flinch or jump responses. The main result was that, depending on the dose and the site of injection, morphine produced either an increase in nociceptive threshold (analgesia), or a decrease in the nociceptive threshold (hyperalgesia). The authors also reported severe effects characterized by hyper-reactivity and violent uncontrolled jumping in stereotyped circular leaps when morphine was administered in the periaqueductal grey matter.

Depressive symptoms are common in many dementia syndromes, and depressive disorders are much more common in older adults with dementia than in cognitively intact older adults. Depression may be a risk factor for, or a prodromal feature of, subsequent dementia. Several neuropathological mechanisms have been suggested to explain these relationships, including the role of underlying cerebrovascular risk factors for depression and cognitive impairment. Depression also may be present in dementia as an emotional reaction to cognitive decline, or as a recurrence of early and midlife depression. Differential diagnosis between depression and dementia is essential, but complicated by problems in assessment, overlapping symptoms between the two conditions, and other medical co-morbidities. Pharmacological treatment of depression in dementia may also be complicated by medical co-morbidity, and can run the risk for adverse reactions or interactions between medications. Psychotherapy and psychosocial interventions, however, hold some promise for effective reduction of depressive symptoms.

The static equilibrium structure of the solvent will modify the potential energy surface for the chemical reaction. This effect is analyzed within the framework of transition-state theory. The rate constant is expressed in terms of the potential of mean force at the activated complex. Various definitions of this potential and their relations to n-particle- and pair-distribution functions are considered. The potential of mean force may, for example, be defined such that the gradient of the potential gives the average force on an atom in the activated complex, Boltzmann averaged over all configurations of the solvent. It concludes with a discussion of a relation between the rate constants in the gas phase and in solution.

This chapter describes the American Protestant reactions to the Catholic understanding of sacramental confession. That reaction is analyzed within the context of the heritage of the Protestant Reformation’s understandings of sin, repentance, and confession. The chapter demonstrates how the Protestant Episcopal Church in the late eighteenth century and American Lutherans in the early nineteenth century transformed the inherited Anglican and Lutheran traditions on the confession of sins to a priest or pastor. In the nineteenth century, sacramental confession became a central polemic issue, because for American Protestants that doctrine seemed to violate the Protestant understanding of justification by faith alone. The Protestant polemic was based on biblical, theological, legal, disciplinary, and historical issues. But, in some cases, the polemic made sensational charges on the immoral and evil political and social consequences of the practice of sacramental confession. Salacious accounts of confessional practice became a part of the polemical record.

Chapter 7 explains why NATO’s institutional memory continues to develop in the way that it does – despite formal learning processes being underutilized. Findings in this chapter draw on the author’s survey-based interviews with 120 NATO elites. The chapter begins by arguing that NATO’s organizational culture locks-in elites’ preference for relying on informal processes and avoiding formal processes. Key characteristics of NATO’s culture posed challenges for identifying and reporting strategic errors. The organization’s norm of consensus made formal agreements on past strategic errors difficult. Moreover, NATO’s focus on reaction over retrospection and a broader culture of blame aversion provided elites with little incentive to break the tradition of reliance on informal processes for memory development. Elites described feeling continuous pressure to react to the crisis at hand and treat past crises as unique – leaving little reason to invest in learning from past failures.

Widely derided by popular psychologists as a destructive response, blame has many defenders among contemporary philosophers. The chapter pushes against their defenses of blame by distinguishing between blame as a reactive attitude and blaming as a speech act, arguing that some disagreement over blame’s value can be explained by the fact that blaming, as a speech act, takes several different forms. Critiques of blame properly target judgmental or strongly verdictive blaming, which treats the wrongdoer as deserving of the blamer’s hostile reactions. This tends to foreclose engagement in further moral dialogue with wrongdoers—an effect particularly destructive in therapeutic contexts; here, it is often more appropriate and constructive to hold others answerable without blaming them in the strongly verdictive sense. The chapter argues that such blame may be similarly destructive outside of straightforwardly therapeutic contexts, and challenges the existence of a sharp divide between therapeutic and nontherapeutic responses to wrongdoers.

This is a mistake that obscures the rational interconnections among the reactive attitudes. Visitors to the park—one that is known for celebrating difference and diversity—observe behaviour or utterances by an actor or a speaker in their presence that, they feel goes too far in violating their sense of acceptable norms. What do they do? How do they sanction the offender? The analysis in this book presents the reader with a series of anecdotes—events or episodes observed or statements overheard by the researcher that audiences, judging by their reactions, consider untoward. This chapter explores reactions to the Jansenist anti-casuist and anti-Jesuit writings among Restoration religious thinkers and pamphleteers. These ranged from a straightforward assimilation of the material into existing structures of anti-Catholic thinking, to a more nuanced reception, which took account of the positive theological case argued by the Port-Royalists as well as the colourful evidence of Catholic iniquity which their polemics comprised. The latter added little that was absolutely new for English thinkers already conversant with Catholic theology and casuistry, but their style and power was striking, and their impact is visible across English religious discourse in the period. The manner in which they were used, furthermore, points to divisions within English theology. Their focus on morality, and their technical criticisms of probabilism, it is suggested, were of greatest relevance to thinkers reacting against a theological framework which prioritized faith over obedience, and denigrated the authority of reason.

This chapter evaluates the evidence that hyper-reactive noradrenergic responses during trauma contribute to hyperarousal symptoms in PTSD, including disturbances in sleep. Some genetic vulnerability for PTSD involves the adrenergic system, and a hyperactive central noradrenergic system might serve to over-consolidate and sustain the affective component of fear memories. Reduced moderation of noradrenergic reactions during low hormone phases of the menstrual cycle could also lead to increased susceptibility to PTSD. This chapter considers a mechanism by which hyperactivity in the noradrenergic system during sleep would impair REM sleep theta and non-REM sleep spindles in the limbic system, both of which are implicated in the consolidation of new safety memories, thereby compromising extinction recall and setting into motion a positive feedback loop in PTSD pathophysiology, involving hyperarousal, failure to integrate contextual information, and biased attention to threat. If so, novel pharmacotherapeutic interventions inhibiting the noradrenergic system during sensitive periods in sleep should be considered.

Whereas most accounts of the reactive attitudes and of responsibility focus on norms of action, we must also consider norms of character: norms that govern the kind of person we can or must be. We are bound by norms of character and so responsible to fellow members for who we are in the same way as for norms of action: via the interpersonal rational patterns in our reactive attitudes. Accordingly, this Chapter develops an account of pride, shame, esteem, and contempt as character-oriented reactive attitudes, clarifies the sense in which these are globalist emotions, distinguishes between personal and social forms of pride and shame, and provides a partial defense of the value of shame and contempt. The result both illuminates the distinction between guilt and shame and, more fundamentally, provides a unified account of responsibility without dividing it into aretaic and accountability faces as Gary Watson does.
