AbstractNot all other-initiations of repair (OIR) are instantly followed by a functional self-repair that restores the progress of the conversation. Despite previous observations of OIRs generally leading to restored progress after one single-repair initiation, data from a multiperson conversational corpus of Norwegian Sign Language (NTS) show that 68% of 112 individual repair initiations occur in multiple OIR sequences. This article identifies three different trajectories of multiple OIR sequences in the NTS data, which are as follows: (1) a trouble source being targeted by more than one repair initiation, (2) the self-repair becomes a new trouble source, or (3) the repair initiation becomes a new trouble source. The high frequency of multiple OIR sequences provides an opportunity to quantitatively investigate how the various formats of repair initiation are distributed in single- and multiple-OIR sequences, how they occur as first or subsequent, and whether they restore the progress of the conversation or are followed by another repair initiation.

Complexities of heating mechanisms make it difficult to investigate the safety of a polymer bonded explosive (PBX) charge of earth-penetrating-weapons (EPWs) during penetration. In this paper, the dynamic damage and non-shock initiation of PBX1314 (60 wt % hexahydro-1, 3, 5-trinitro-1, 3, 5-s-triazine (RDX), 16 wt % aluminum, 24 wt % hydroxy-terminated polybutadiene (HTPB)) during penetration is investigated through experiments and simulations. In the experiments, steel projectiles filled with PBX1314 are launched to penetrate concrete targets. In the results, non-shock initiations occur on the tail surface of PBX1314 along with mechanical damage of the tail and middle part of PBX1314. A dynamic damage and initiation model is proposed to characterize the effects of microcracks on the mechanical and thermal responses of PBX1314. Investigation based on the model suggests that microcrack interfacial friction plays significant roles in damage, heat generation and localization in PBX1314. A non-shock initiation criterion is developed based on macroscale variables in PBX1314. Numerical simulations of the penetration experiments are performed by using the proposed model and criterion. The mechanical damage and non-shock initiation of PBX1314 in the experiments are successfully predicted. The simulation results indicate that the tail of PBX1314 impacts the projectile repeatedly during penetration. Finally, the initiation criterion is satisfied because of frictional heat localization near microcrack surfaces and initiation is activated in the tail of PBX1314.

Voevodsky (1917–1967) along with the history of the development of the instrumentation basis for this field of science in the Union of Soviet Socialist Republics (USSR). The design of the first EPR spectrometers for the chemical radio spectroscopy initiated the emergence of a new scientific instrumentation field in this country. Based on recollections shared by scientists and engineers and an examination of archive materials, the author reconstructs relevant events and identifies major participants in this process.

Abstract In all domains of life, the regulation of transcription by DNA-dependent RNA polymerases (RNAPs) is achieved at the level of initiation to a large extent. Whereas bacterial promoters are recognized by a σ-factor bound to the RNAP, a complex set of transcription factors that recognize specific promoter elements is employed by archaeal and eukaryotic RNAPs. These initiation factors are of particular interest since the regulation of transcription critically relies on initiation rates and thus formation of pre-initiation complexes. The most conserved initiation factor is the TATA-binding protein (TBP), which is of crucial importance for all archaeal-eukaryotic transcription initiation complexes and the only factor required to achieve full rates of initiation in all three eukaryotic and the archaeal transcription systems. Recent structural, biochemical and genome-wide mapping data that focused on the archaeal and specialized RNAP I and III transcription system showed that the involvement and functional importance of TBP is divergent from the canonical role TBP plays in RNAP II transcription. Here, we review the role of TBP in the different transcription systems including a TBP-centric discussion of archaeal and eukaryotic initiation complexes. We furthermore highlight questions concerning the function of TBP that arise from these findings.

ObjectivesGuidelines for the use of drugs for type 2 diabetes mellitus (T2DM) have changed since 2000, and new classes of drug have been introduced. Our aim was to describe how drug choice at initiation and first stage of intensification have changed over this period, and to what extent prescribing was in accord with clinical guidelines, including adherence to recommendations regarding kidney function.DesignRepeated cross-sectional study.SettingUK electronic primary care health records from the Clinical Practice Research Datalink.ParticipantsAdults initiating treatment with a drug for T2DM between January 2000 and July 2017.Primary and secondary outcome measuresThe primary outcomes were the proportion of each class of T2DM drug prescribed for initiation and first-stage intensification in each year. We also examined drug prescribing by kidney function and country within the UK.ResultsOf 280 241 people initiating treatment with T2DM drugs from 2000 to 2017, 73% (204 238/280 241) initiated metformin, 15% (42 288/280 241) a sulfonylurea, 5% (12 956/280 241) with metformin and sulfonylurea dual therapy and 7% (20 759/280 241) started other options. Clinicians have increasingly prescribed metformin at initiation: by 2017 this was 89% (2475/2778) of drug initiations. Among people with an estimated glomerular filtration rate of ≤30 mL/min/1.73 m2, the most common drug at initiation was a sulfonylurea, 58% (659/1135). In 2000, sulfonylureas were the predominant drug at the first stage of drug intensification (87%, 534/615) but by 2017 this fell to 30% (355/1183) as the use of newer drug classes increased. In 2017, new prescriptions for dipeptidyl peptidase-4 inhibitors (DPP4i) and sodium/glucose cotransporter-2 inhibitors (SGLT2i) accounted for 42% (502/1183) and 22% (256/1183) of intensification drugs, respectively. Uptake of new classes differs by country with DPP4is and SGLT2is prescribed more in Northern Ireland and Wales than England or Scotland.ConclusionsOur findings show markedly changing prescribing patterns for T2DM between 2000 and 2017, largely consistent with clinical guidelines.

Hall's Bond Forming Initiation Theory, derived for [2 + 2] systems, was applied to [4 + 2] systems. Polymerizable electron-rich and electron-poor olefins were reacted to obtain evidence for reactive intermediates using polymerization as a trap. A diradical intermediate was successfully trapped in reactions of 1-methoxy-1,3-butadiene with methyl 2,2-dicyanoacrylate, dimethyl cyanofumarate, trimethyl ethylenetricarboxylate, maleic anhydride and acrylonitrile. Diradical intermediates were also trapped in reactions of isoprene, 1,3-butadiene, 2,3-dimethyl-1,3-butadiene, cis and trans-piperylene and 2,5-dimethyl-2,4-hexadiene with acrylonitrile. Copolymers were obtained in all cases. Copolymerization was accompanied by [4 + 2] cycloaddition. A zwitterionic intermediate was successfully trapped in the reactions of p-anisyl-1,3-butadiene and 1-phenyl-1,3-butadiene with various leaving groups in the β-position. Homopolymer of the arylbutadiene was obtained in all cases. Polymerization was initiated from a cationic species which arose from the elimination of the leaving group from a zwitterionic hexamethylene species. Polymerization was accompanied by [4 + 2] cycloaddition. Reactions of p-anisyl-1,3-butadiene and 1-phenyl-1,3-butadiene with methyl 2,2-dicyanoacrylate, dimethyl cyanofumarate, trimethyl ethylenetricarboxylate gave only the [4 + 2] cycloadduct. No polymer was formed in this reaction and no evidence for a reactive intermediate was obtained. The BFI theory was also applied to Group Transfer Polymerization reactions to determine whether the reaction mechanism involved a tetramethylene diradical intermediate. No styrene was incorporated into the methyl acrylate polymer, and so no evidence for a radical intermediate was obtained. LUMO energies calculated by AM1 were compared to reduction potential and UV data for tetrasubstituted and trisubstituted electrophilic olefins. The trends exhibited by the calculated LUMO energies agreed well with the experimental data, implying that calculations may be a reasonable method of predicting reactivity.

The evaluation of the dynamic fracture properties with the current impact testing techniques is subject of some controversy, because the crack initiation conditions cannot be established with accuracy. Besides, some of them are limited to low loading velocities and the evaluations are made following a static approach. In consequence, a fundamental study is required with two aims, namely, a development of a new impact testing technique and the analysis of the extension of the fracture mechanics concepts to crack initiation under dynamic loading. These are the objectives of this research project. The impact testing technique proposed is based on the shock tube technique and is capable to test the specimens, which are central cracked circular plates, under static and dynamic conditions. The findings show that the specimens are loaded uniformly with blast waves, which have an initial ramp that increases steadily without oscillations. The magnitude of the load is controllable and reproducible and the fracture analysis should be based on a dynamic calibration. This technique has been applied to the evaluation of the dynamic fracture toughness of PMMA following an experimental-numerical procedure. The crack initiation load and the strain rate were measured experimentally and the fracture toughness was calculated numerically with standard solutions and with numerical calculations of the J-Integral with the finite element code ABAQUS. The problem of crack initiation under pressure load has not been completely solved. Consequently, the significance of the static and dynamic test results was established with the findings of additional experimental and numerical work, which showed that the modulus of elasticity and fracture stress increases with the strain rate, that there is plastic deformation at the crack tip of those specimens tested under static conditions and that the dynamic calculations with ABAQUS involves the interaction of the stress waves with the boundaries. Furthermore, it has to be noticed that the crack tip is under bending stresses. Thus, this test is not a standard plane strain material dynamic test. Finally, conclusions are drawn on the use of the this experimental-numerical technique and its further application.

Aim – To identify any discrepancy between new graduate nurses’ perceptions of their professional roles before they begin practice and what they experience at the initiation of their duties.

Background – Turnover and attrition is a major issue within the nursing profession. Better understanding of new graduate perceptions will provide the opportunity to identify stressors that can be addressed to slow the rate of turnover.

Method – A case study research design was selected for data collection from a population of newly licensed Registered Nurses at the initiation of professional responsibilities.

Evaluation – Survey results were clustered thematically to look for common results among responses.

Implications for Nursing – Identifying discrepancies between expectations and reality can guide nursing leadership and educators to create strategies to better prepare new nurses for practice, reduce stress and anxiety, and limit the amount of turnover and attrition.

Microbial methane from subsurface organic-rich units such as coals and shale support approximately 5% of the United States and Canada's energy needs. In the deep subsurface, microbial methane is formed by the metabolism of primarily CO2, H2, and acetate by methanogens. These metabolites are the by-products of multi-step biodegradation of complex organic matter by microbial consortia. This study investigates microbial methane in the Illinois Basin, which is present in organic-rich shallow glacial sediments (surficial), Pennsylvanian coals (up to 600 m depth), and the Upper Devonian New Albany Shale (up to 900 m depth). Findings from the study show that hydrogeochemical conditions are favorable for methanogenesis in each reservoir, with a decrease in groundwater flushing rates corresponding to a decrease in average reservoir depth and an increase in carbon isotopic fractionation. The deeper reservoirs (coals and shale) were paleopasteurized, necessitating re-inoculation by methanogens. The microbes were likely advectively transported from shallow sediments into the coals and shale, where areas of microbial methanogenesis correlate with freshwater recharge. The recharge in the shale was primarily sourced from paleoprecipitation with minor contributions from glacial meltwater during the Pleistocene (4He ages). All areas sampled in the shale were affected by Pleistocene recharge, however groundwater ages in areas of microbial methanogenesis are younger (average 0.33 Ma) than areas with thermogenic methane (average 1.0 Ma). Estimates of in-situ microbial methane production rates for the shale (10-1000 TCF/Ma) are 104-106 times slower than laboratory rates. Only limited biodegradation is observed in the shale. In-situ stimulation of methane production may be most effective if aimed at increasing production of the supporting microbial consortia as well as methanogens. Trace metal concentrations in the shale are below known levels of inhibition or enhancement, with the exception of Fe, suggesting that microbial methanogenesis is not repressed by any of the measured trace metals and may be improved with the addition of Ag, Co, Cr, Ni, and Zn.

Vesicular stomatitis virus (VSV) infection causes inhibition of host protein synthesis, in part by sequestering initiation factors required for mRNA cap recognition. The viral mRNAs share a common mRNA structure to those of the host cell, with a 5' cap and 3' polyadenylate tail, but continue to be efficiently translated despite host translational shutoff. This observation suggests that a non-canonical translation pathway is utilized for viral protein synthesis. To investigate this pathway, we performed an RNA interference screen to identify genes required for VSV replication. In contrast to bulk cellular translation, viral translation is hypersensitive to knockdown of a protein constituent of the 60S ribosomal subunit, rpL40. Depletion of rpL40 diminishes VSV protein synthesis by >90% and is restored through complementation with an siRNA-resistant mutant of rpL40. To delineate the mechanism by which rpL40 is required for viral protein synthesis, we reconstituted translation of VSV mRNA in yeast extracts in vitro. In the absence of rpL40, we show that the two ribosomal subunits fail to associate on VSV mRNA, and the small subunit does not scan to the initiation codon. Regulation by rpL40 occurs in context of the large subunit, providing direct evidence for translational control by the ribosome itself. This rpL40- dependent mechanism of translation initiation is broadly conserved within eukaryotes, governed solely through an RNA determinant, and is utilized by several viruses within the order Mononegavirales. To determine whether a subset of cellular transcripts also require rpL40 for translation, we identified polysome-associated mRNAs in yeast by deep sequencing. We demonstrate that in vitro and in vivo translation of candidate mRNAs, including factors involved in stress responses, are inhibited in the absence of rpL40. This finding suggests that rpL40 plays a critical role in transcript-specific translation during cellular stress. Collectively, our work identifies an alternative translation pathway that is specifically dependent on rpL40, revealing a previously unappreciated mechanism of protein synthesis regulation by the ribosome.

Background: T2DM patients can be treated with both Insulin or/and non-insulin antidiabetic drugs. Biguanide-derivate is the first-line antidiabetic that is cost-effective and safe. DPP-4Is are antidiabetic drugs that can be used to treat old patients and those with kidney failure. While SGLT2-inhibitors and GLP-1analogues can be used by T2DM patients with cardiovascular problems and those with overweight. The initiation of antidiabetic drug is influenced by several factors such as patient characteristic, physician’s knowledge and guidelines. Aim: The aim of this study was to study the type of antidiabetic medication initiated to naïve T2DM patients, how age and sex affect the choice of medication and difference in prescribing between Primary healthcare centers in Region Uppsala. Methods: The study was a cross-sectional based on health record data from Region Uppsala. The study included 2473 T2DM patients that were initiated antidiabetic medication between January 2019 and December 2020. Results: Non-insulins accounted for 88 % and insulins for 21.1% of the prescriptions. The proportion of men that received both insulin and non-insulins were higher than women. The proportion of patients that received insulins was higher for the age group 80 years and older. The proportion of men that received SGLT2- inhibitors was higher than for women, while women received more GLP-1- analogues. There was a variation between PHCs, but non-insulins were mostly prescribed at all PHCs. Conclusions: Initiation of antidiabetic drugs to naïve T2DM patients in region Uppsala fallowed the guidelines. There are some differences in prescription pattern between the two sexes and the age groups.
