Dislocation interactions were investigated in near-alpha titanium alloy Ti6242Si after low cycle fatigue. Samples from the gauge section and the crack initiation site on the fracture surface were studied. Grain pairs with different crystallographic orientations were analysed from the gauge section to understand the dislocation interactions near the boundary. Deformation was primarily caused by planar slip, localized into slip bands in primary alpha (αp) grains. Direct slip transfer was observed within grains having similar orientations. In contrast, slip transfer resulted in a different kind of dislocation nucleation in the neighboring hard grain where there was misorientation between the grains, with the observation of cross-slip. Strain transfer was observed between highly misoriented grain pairs. Crack nucleation occurred on an αp grain by basal plane splitting, due to the large tensile stress developed by a double ended pile-up. This kind of pile-up is suggested to result from the incomplete reversibility of dislocation motion during load reversals. The observation of superjogs on the basal dislocations in the crack nucleated αp grain provides a rationale for why cracks nucleate near, rather than on, the basal plane.

Site-directed polyclonal antibodies were generated against four oligopeptides derived from the rat alpha-isoforms of Na(+)-K(+)-adenosinetriphosphatase (i.e., Na(+)-K+ pump) and were used to assess the phylogenetic conservation of the target regions. The first antibody was directed against a region near the phosphorylation site that is conserved among alpha-subunits from numerous species. The remaining antibodies were specific for the isoforms of alpha and recognized a region near the fluorescein isothiocyanate-reactive lysine of the adenine binding site. Immunoblots of protein from a number of species probed with these antibodies revealed dramatic differences in the conservation of the target sequences. The region derived from alpha 1 was found in four of the six mammals screened. In contrast, the analogous region derived from alpha 2 was found in all the screened mammals. Most conserved was the region derived from alpha 3, which was found in all the screened mammals as well as in chickens, catfish, dogfish, and sea slugs. Conservation of the region from alpha 3 suggests functional significance and implies that strong selective pressures have discouraged mutational change. Results from the immunoblots further suggest that isoforms are present in all vertebrates.

Deformation of Titanium alloys close to optimal superplastic condition i.e. near superplastic regime of deformation leads to significant change in microstructures.VT-9 titanium alloy was used in order to find out those parameters of microstructure which are varying significantly during near superplastic regime of deformation. Tensile tests were carried out at 930°C up to fracture with a constant strain rate of 5*10-4 s-1 and a jump wise varying strain rate of 1*10-4 s-1 & 5*10-4 s-1 .The microstructural parameters of both air-cooled and water quenched portion i.e. size of alpha phase, percentages of alpha phase and parameter of non-uniaxiality of alpha phase were found to change significantly during near superplastic regime of deformation. It has been found that in the near superplastic regime of deformation percentage of α-phase decreased from 90% to 13%. As the β-transus temperature of this alloy is 970°C, this significant change in percentage of α-phase is attributed to deformation induced phase transformation. Optical microscopes, micro Vickers hardness test, XRD, FESEM have been used to characterize the microstructure of the material.

Near-u.v. and far-u.v. c.d. spectra of human alpha-calcitonin-gene-related peptide (h alpha CGRP), analogues and fragments of CGRP and amylin were recorded in aqueous solution and in trifluoroethanol (TFE)/water mixtures. All peptides contained significant amounts of alpha-helix in aqueous solution, and this amount increased on adding TFE. The helical content was unaffected by pH and salt. However, amylin contained much less helix than CGRP and the c.d. spectrum was more temperature-sensitive. A band in the near-u.v. c.d. spectrum of CGRP (but not present in the spectrum of amylin) was attributed to the disulphide bond in CGRP. The intensity of this band was pH-dependent and titrated with a pKa of 6.5, suggesting the involvement of histidine ionization.

Dynamic recrystallization in Ti-1100 was investigated. Ti-1100 is one of near α titanium alloys and contains Si for improving high temperature mechanical properties. Ti-1100 exhibits martensitic transformation by quenching into iced brine after solid solution treatment. Hereafter specimens subjected to quenching into iced brine and to cooling in air after solid solution treatment are called IBQ specimen and AC specimen, respectively. After tensile test at high temperature, IBQ specimen exhibits morphological change from lath structure to equiaxed structure, but AC specimen does not. It is indicated that dynamic recrystallization occurs during the tensile test of IBQ specimen. Effect of silicide on the dynamic recrystallization was investigated using two specimens: one included more silicide precipitates and the other less. The former specimen shows smaller recrystallized grains than the latter. It is indicated that the specimen including more silicides exhibits smaller recrystallized grains.

A near-alpha type titanium alloy Ti-4Al-1.5Mn tube was analyzed after extrusion deformation and annealing treatment. Electron backscattered diffraction is employed to explore the Microstructure and texture characteristic of the recrystallization alloy. The alloy is consisted of equiaxed α grains and fine β phase with {1 0-1 2} twins .It was found that the (10-10) fiber texture along the extrusion direction was the dominate texture in this alloy and the volume fraction was 40.7%.

This work focuses on developing near-alpha Ti alloys via the selective addition of small concentrations of low-cost eutectoid [Formula: see text]-stabilizers like Cu and Mn. In particular, these newly designed near-alpha Ti alloys are manufactured via the cheapest powder metallurgy route of cold pressing plus sintering. Moreover, thermomechanical deformation of the sintered alloys via hot forging in the [Formula: see text] and [Formula: see text] field was also investigated aiming to enhance the mechanical properties through reduction of the residual porosity and microstructural control. It is found that the initial addition of a small amount of eutectoid [Formula: see text]-stabilizers leads to higher tensile properties with comprision to pure Ti produced by powder metallurgy, and this is due to the formation of a coarse lamellar structure due to the presence of [Formula: see text]-stabilizers. Further enhancement of the strength is achieved by means of hot thermomechanical processing thanks to sealing of the residual pores, texturing, and refinement of the microstructural features.

The article considers the influence of carbon in the range from 0.008 to 0.18 mass. % at the temperature of complete polymorphic transformation, microstructure and mechanical properties of near-alphatitanium alloy systems Ti–6.2Al–Sn–Zr–(2.19–3.53) Moeq–(0.18–0.28)Si–(0.008–0.18)С. Alloying with carbon is possible due to special titanium sponge or industrial ligatures. Six experimental forgings performed on bimodal microstructured ingots allowed to establish a rational level of carbon. It is shown that carbon alloying at solubility limit (up to 0.08% by weight) increases the heat resistance of the material, while a similar alloying with silicon gives a greater effect. The carbon effect on the strength at room temperature is negligible. Negative effect of carbon on the impact toughness of the alloys is marked. For the selected carbon alloying level at strength of 1165–1180 MPa, the impact viscosity remains at an acceptable level of KCU = 330–381 kJ/m2 . The alloys of the investigated system allow hot deformation in heavy conditions under hammer forging at relatively low temperatures of the two-phase region.

The recrystallization behaviour of near-alpha titanium alloy IMI834, which is primarily used for high temperature aerospace compressor disc applications, has been investigated at hot working temperatures. The latest results of a finite element model, developed using the commercial code DEFORM-3D with constitutive equations adapted from available literature, will be presented. Model development and validation involved the hot compression of specimens with an initial bimodal alpha+beta microstructure at temperatures of 1000°C-1100°C, strain rates of 0.01s 1-1s 1, and varied post-deformation annealing times. The characterization of microstructure through quantitative metallography revealed beta grain refinement achieved primarily through static/metadynamic recrystallization. The beta recrystallization kinetics were subsequently predicted through an Avrami-type relationship.

TA32 is a heat-resistant titanium alloy developed for superplastic forming in fabrication of near-space supersonic aerocraft. Clarification of superplastic deformation behavior is important to the optimization of forming parameters. Superplastic tensile test was conducted in vacuum to eliminate the effect of surface oxidation on experimental data, the test temperature and strain rate varied from 900oC to 960oC and 5.32×10-4 to 2.08×10-2s-1, respectively. It was observed that the size of equiaxed α grains exhibited a trend of coarsening with the increase of temperature and decrease of strain rate. Textures of deformed specimens exhibited random distribution with a decreased texture intensity compared with the as-received materials. The superplastic deformation mechanism of TA32 alloy was dominated by grain boundary sliding, which is accommodated by grain rotation and dynamic recrystallization.

Immunoglobulin (Ig) A serves as the first line of humoral defense at all mucosal surfaces and is present in large quantities of blood. In playing its role in humoral immunity, IgA interacts with a variety of effector molecules present both in serum and on the surfaces of immune and inflammatory cells. To study these interactions, we previously established expression of human IgA1 in insect cells using recombinant baculoviruses and showed that the expressed antibody is a structurally and functionally intact polypeptide useful for examining the molecular properties of IgA. Indeed, since the C alpha 2 N-linked glycosylation site lies near the Fab-distal pole of C alpha 2, the inability of a mutant IgA1 lacking C alpha 2 N-glycosylation to bind its cognate receptor suggested that the monocyte Fc alpha receptor (mFcalphaR) recognizes IgA at a hinge-distal site encompassing the boundary between the C alpha 2 and C alpha 3 domains. In this report, we utilize both domain-swapped IgA/IgG and point-mutated IgA chimeras to verify the above hypothesis. Using an antigen-specific rosetting assay and a mFc alpha R-expressing cell line, we show that (a) C alpha 2 and C alpha 3 together are necessary and sufficient for binding; (b) neither the IgA hinge nor the tailpiece is necessary for binding; (c) mutations away from the interdomain boundary do not affect binding; and (d) mutations located near the three-dimensional boundary between C alpha 2 and C alpha 3 completely disrupt binding. Taken together, these results localize the mFc alpha R recognition site on IgA to the boundary region between the second and third constant domains--a site analogous to that recognized by Staphylococcus aureus protein A on IgG. The use of this hinge-distal site is, to date, unique among Fc receptors of the Ig superfamily.
