Annotation:
Penna's bit-string model of biological ageing due to the accumulation of deleterious mutations is generalized to allow for more than one disease per year. The results remain qualitatively unchanged except for a more complicated non-monotonic approach to equilibrium. We also look at "mutational meltdown", the extinction of the whole population if all mutations are deleterious and heritable, and why the Penna model can escape this extinction. No dependence on population size is found for mutational meltdown, with up to 108 individuals.

Using the Penna bit-string model for biological ageing we compare two kinds of reproductive regimes: Sexual reproduction (SR) and meiotic parthenogenesis (MP). The last one is a common type of asexual reproduction with recombination, found in diploid organisms. We show that although both regimes present roughly the same survival rates, the diversity generated by SR is much larger, and can prevent the extinction of a population submitted to a natural disaster. The fixation of bad genes inside an MP population, after many generations, explains our results. We also study the consequences of cloning (simple copy) on population diversity.

The Penna model for biological aging was modified. The reproducibility of each individual was determined according to the number of mutations relevant at that time. The results of Monte-Carlo calculations using the modified model show that the ranges of the reproducible age are broadened as time goes by, thus showing self-organization in biological aging to the direction of maximum self-conservation. In addition, the population, survival rate, and average life span were calculated and analyzed by changing the number of new mutations at birth. It is observed that the more the number of new mutations at birth is considered, the shorter the average life span that is obtained.

The sexual version of the Penna model of biological aging, simulated since 1996, is compared here with alternative forms of reproduction as well as with models not involving aging. In particular we want to check how sexual forms of life could have evolved and won over earlier asexual forms hundreds of million years ago. This computer model is based on the mutation-accumulation theory of aging, using bits-strings to represent the genome. Its population dynamics is studied by Monte Carlo methods.

We studied different versions of the Penna bit-string model for biological ageing and found that, after many generations, the number of lineages N (maternal family names) always decays to one as a power-law N∝t-z with an exponent z roughly equal to one. Measuring the mean correlation between the ancestor genome and those of the actual population we obtained the result that it goes to zero much earlier before the number of families goes to one, the population keeping thus its biological diversity. Considering maternal and paternal family names (doubled names) we also finished with only one pair of common ancestors. Computing the number of families of a given size as a function of the size (number of individuals the family has had during its whole existence) again a power-law decay is obtained.

The Penna model for biological aging is modified so that the fertility of each individual is determined by means of the number of activated mutations at that time. A new concept of "good" mutation, which makes an individual to mature enough to reproduce, is introduced. It is assumed that each individual can reproduce only during adulthood, which is determined by the number of activated mutations. The results of Monte Carlo calculations using the modified model show that the ranges of the reproductive age are broadened as time goes by, thus showing self-organization in the biological aging to the direction of the maximum self-conservation. In addition, the population, the survival rate, and the average life span were calculated and analyzed by changing the number of new mutations at birth. It is observed that the higher is the considered number of new mutations at birth, the shorter is the obtained average life span. The mortality functions are also calculated and they showed the exponential increase in adulthood, satisfying the Gompertz law.

We present a model of evolution of the age structured population based on the Monte Carlo method. We have assumed that the health status of an individual is described by variance of its fluctuations. Each expressed deleterious mutation increases the fluctuations. Additionally, the fluctuations of the environment are superimposed on the fluctuations of individuals in the population. An individual dies if the combination of both stochastic processes trespass the limit (level of homeostasis) set as the model parameter. The genes are switched on chronologically, what leads to accumulating defective genes expressed during the late periods of life in the genetic pool of the population. That results in the specific age structured population, in accordance with the predictions of Medawar's hypothesis of ageing and the results of the Penna model simulations. A decrease of the variation of the environmental noise increases the average expected lifespan of individuals.

We have simulated the effect of the diversity of the late expressed genes in the genetic pool of population on the phenotypes of individuals in the late ages. Using Penna model based on the Monte Carlo method we have obtained for the oldest fractions of populations lower mortality rates than predicted by the exponential Gompertz function. Such deviations from the expected exponential increase of mortality are the characteristic for populations which are not in equilibrium with the environment, or if a relatively high probability of reversions was assumed, or if the population is heterogeneous. In such populations, the genes expressed in the late ages, are under the very weak selection pressure and thus, highly-polymorphic. As an effect, the probability of the genetically-determined death of the oldest organisms does not grow as fast as predicted by the Gompertz exponential curve describing mortality during earlier periods of life.

In this paper, we report the results of hydrogen adsorption properties of a new 2D carbon-based material, consisting of pentagons and octagons (Penta-Octa-Penta-graphene or POP-graphene), based on the Grand-Canonical Monte Carlo simulations. The new material exhibits a moderately higher gravimetric uptake at cryogenic temperatures (77 K), as compared to the regular graphene. We discuss the origin of the enhanced uptake of POP-graphene and offer a consistent explanation.

Since 2007, China has advanced several reforms to restrict and control the application of the death penalty. This is rather puzzling if we consider China’s historical use of the capital punishment, as well as its widespread public support. This article contributes to the debate on the causes of these reforms by testing the relevance of the hypothesis that the United Nations Human Rights System (UNHRS) successfully socialised China into a more lenient understanding of the death penalty, which in turn would have prompted those reforms. The results partially confirm this hypothesis. There is strong evidence that the UNHRS might have socialised China into a particular aspect of these reforms — the cancellation of some economic and non-violent crimes —, but it was not responsible for triggering these reforms and it failed to socialise China into conducting a number of other specific death penalty reforms. Despite not being responsible for the totality of the reforms, this study affirmed the relevance of the possibility that the UNHRS influenced part of them, which raises the necessity of considering this factor to arrive at a comprehensive explanatory model. ‘A penny saved is a penny earned’ as this phrase says it all, the key for increased profit lies in the elimination of non-value adding actions in any production process. This study concentrates mainly on the procedures to identify and to reduce the different types of wastes in a production process. Initially the data related to types of waste and their classification according lean techniques were analyzed and implemented on a production process. The data were recorded for a whole month before implementation and for a whole month after implementation. Later the influence of the lean techniques was analyzed on two different sections of waste, for 5% significance value using paired t-test. It was found that there is a positive impact of lean techniques on some areas of production flow.

Este artigo discute os dados qualitativos da pesquisa “Violência Doméstica perpetrada contra a mulher no município de Montes Claros: um recorte possível”, financiada pelo CNPq e pela FAPEMIG. O objetivo do estudo foi investigar quantitativamente e qualitativamente os atos violentos contra as mulheres na cidade de Montes Claros, Minas Gerais. A vertente qualitativa pretendia conhecer o sentido da violência para as mulheres envolvidas e seus reflexos na família, através de entrevistas semiestruturadas que não se efetivaram. A partir da Análise Institucional de René Lourau analisamos essa inviabilidade como dado qualitativo, examinando o cotidiano da delegacia após a lei Maria da Penha. Concluímos que a intervenção judicial não é suficiente para a inibição da violência contra a mulher, pois em certos casos afasta da delegacia mulheres com outras demandas acerca da violência, que sofrem e que não podem contar com a ajuda policial para a resolução dos seus conflitos.

The MO´Ã Foundation in Rio Grande do Sul / Brazil is a Non-Governmental Organization (NGO) of Studies and Research for Environmental Protection and Development and works through actions and projects aimed at Environmental Education. In 2007 the MO´Ã Foundation acquired an area of 24 hectares of Atlantic Forest that was classified as a Private Reserve of Natural Heritage (RPPN), in 2015, after a long and complex process. In Guimarães, Portugal, the Sisterhood of Nossa Senhora do Carmo da owns about 60 hectares of the area where it receives the devotees and promotes the management of green space and the conservation of tourist infrastructure and religious heritage. In 2019, the Municipality of Guimarães submitted the application of Monte da Penha to a locally protected area, in a process still under evaluation by the Institute for the Conservation of Nature and Forests. The work presented aims to infer the role of the private sector in the administration and shared management of protected areas, in a comparative analysis between the MO´Ã Foundation and the Sisterhood of Penha. Based on a systematic review of a database in journals (CAPES, EBSCO, SCIELO, Open Access Scientific Repositories of Portugal), documents, fieldwork and interviews, we conclude that private institutions assume a fundamental relevance in the governance of areas protected, ensuring the main objective of these: the conservation and / or protection of nature in a sustainable way.

Penja is an endemic fish in the Mandar river and is one of the small pelagic fish species that has been used by fishermen in Polewali Mandar for a long time. Penja fish is one of the water organisms that utilize plankton as its food. This study aims to determine the composition of the plankton species the fishing season in Mandar waters of Polewali Mandar Regency, West Sulawesi Province. This research was conducted in November-December 2017. The sampling location was conducted in two different water locations which are at estuary and body of Mandar River, Polewali Mandar Regency, West Sulawesi Province. And the identification of samples was done at the Biology Laboratory of Plankton and Natural Feed at the Research Center for Brackishwater Aquaculture Development and Fisheries Extension (BRPBAP3) of Maros Regency. The results of this study indicate that the highest abundance of plankton during the fishing season in the estuary waters and the month of December. Bacillariophyceae class is the most plankton class found during the study and there is 37 genus plankton consisting of 28 genus of phytoplankton and 9 genus of zooplankton. The range of diversity index value during fishing time, sampling location, research month and sampling time are 1 <H '<3 which means medium diversity, uniformity index value is 0.7-0.9 so that the plankton uniformity index is relatively the same or equal, and the Domination index value indicates that no domain genus in the community.

IntroductionLymphopenia is a known adverse event of dimethyl fumarate (DMF) with treatment dis- continuation recommended for patients with severe prolonged (≥6 months) lymphopenia. Because of the COVID-19 pandemic burden on healthcare systems, we retrospectively examined the impact of absolute lymphocyte count (ALC) monitoring frequency (every-3-months vs every-6-months) on lym- phopenia detection.MethodsSamples from patients enrolled in phase 3 trials (DEFINE/CONFIRM) and the extension study (ENDORSE) were retrospectively analysed using 3-month or 6-month intervals. Lymphopenia was defined as ALC <0.91x109/L and severe lymphopenia as ALC <0.5×109/L. Times to the first lymphopenia event and first severe lymphopenia event were estimated using Kaplan-Meier methods.ResultsThe analysis included 741 patients. There were 355 lymphopenia cases (76 severe) with 3-month monitoring, and 314 cases (70 severe) with 6-month monitoring. Over 120 months, incidence of first lympho- penia event was significantly different for 3-month vs 6-month monitoring (difference range: 2.64%–6.54%; P=0.0088). Incidence of first severe lymphopenia event was not significantly different for the two intervals (P=0.5866). Proportions of patients with severe prolonged lymphopenia with 3-month and 6-month moni- toring were similar (4.0% vs 4.2%).ConclusionsThe results of this study may be informative to clinicians managing pandemic-related health- care resource burdens. Abstract OBJECTIVE AND IMPORTANCE Desmoplastic infantile gangliogliomas (DIGs) are extremely rare tumors that respond well to treatment. However, their biological behavior remains to be clarified. We describe two patients whose DIGs spontaneously regressed after surgery, without adjuvant therapy. CLINICAL PRESENTATION A 9-month-old girl presented with left hemiparesis, and a 6-month-old boy presented with increasing head circumference. For both patients, neuroimaging demonstrated a huge cystic tumor that included a solid portion and was widely attached to the dura. Gadolinium-diethylenetriamine penta-acetic acid produced strong enhancement. INTERVENTION One patient underwent partial and the other subtotal tumor removal. Histologically, both tumors were diagnosed as DIGs. Postoperatively, the residual tumors were monitored without adjuvant therapy, and both regressed in several months. CONCLUSION Our experience suggests that DIGs may include a subgroup of tumors with a tendency for spontaneous regression, possibly attributable to the induction of apoptosis.

O presente texto tem por objetivo identificar traços do conservadorismo, pedagógico e teórico, que é preponderante na história do ensino de música nas escolas brasileiras. O recorte observado tem início com as práticas jesuíticas, ainda no período colonial, passando por 1854, quando foi publicada a primeira norma institucionalizando o ensino de música nas escolas brasileiras, pelo período do Canto Orfeônico – primeira grande política pública de educação musical –, chegando até 1971, quando foi promulgada a Lei de Diretrizes e Bases 5.692, que implementa a disciplina Educação Artística nos currículos escolares. Tendo como referência obras de Fonterrada (2008), Monti (2015), Penna (2013), Jardim (2009, 2012) entre outros, são apresentados elementos metodológicos e históricos do ensino de música no Brasil, a fim de se identificar seu caráter conservador, por vezes marcado pelo eurocentrismo e outras por um ufanismo nacionalista, que flerta com o fascismo. Por fim, a reflexão de que, embora tenha sido alvo de diferentes e importantes iniciativas, por vezes bem-intencionadas e conectadas a propostas mais criadoras, o ensino de música nas escolas brasileiras, no recorte temporal escolhido, seguiu marcado por seu caráter conservador.

Designing the future energy supply in accordance with ambitious climate change mitigation goals is a challenging issue. Common tools for planning and calculating future investments in renewable and sustainable technologies are often linear energy system models based on cost optimization. However, input data and the underlying assumptions of future developments are subject to uncertainties that negatively affect the robustness of results. This paper introduces a quadratic programming approach to modifying linear, bottom-up energy system optimization models to take cost uncertainties into account. This is accomplished by implementing specific investment costs as a function of the installed capacity of each technology. In contrast to established approaches such as stochastic programming or Monte Carlo simulation, the computation time of the quadratic programming approach is only slightly higher than that of linear programming. The model’s outcomes were found to show a wider range as well as a more robust allocation of the considered technologies than the linear model equivalent.

424 Background: Capecitabine (cape), an oral chemotherapy, is the treatment backbone for many GI cancers. Its complex dosing and narrow therapeutic index make medication adherence and toxicity management crucial for quality patient care. Methods: We conducted a feasibility study of “Penny,” an augmented intelligence mobile phone chatbot that leverages algorithmic surveys and natural language processing (NLP) to engage with patients in conversational, bi-directional text messages. Penny provides patients with medication reminders tailored to their prescribed doses and schedules, sends weekly check-in messages, manages low-grade symptoms in real time, and escalates high-grade symptoms for resolution by the clinical team. Patients ≥18 years old receiving cape for the treatment of a GI cancer were accrued in sequential cohorts of 20 for participation over a three-month period. Feasibility was assessed during planned interim analyses and was predefined as the completion of a 20-patient cohort without a safety event, defined as the communication of incorrect medication or symptom management recommendations as ascertained by two independent clinician reviewers (Κ = 0.89). Secondary outcomes included patient-reported adherence and engagement with the chatbot’s weekly check-in messages. At study completion, all patients were invited to participate in structured interviews to provide feedback on the platform. Results: The first cohort of 20 patients was enrolled from 8/2021 to 4/2022; the median age was 57 years, and patients were primarily female (55%), white (65%), commercially insured (55%), and had colorectal cancer (55%). Chemotherapy regimens included cape with oxaliplatin (50%), concurrent RT (30%), temozolomide (5%), and monotherapy (15%). A total of 2,149 text messaging exchanges were reviewed with 150 (7%) medication-related and 9 (0.4%) symptom-related safety events identified. Most medication-related safety events were due to misalignment with prescribed chemotherapy schedules (55%) and doses (32%). Symptom-related safety events were primarily due to the misinterpretation of patient messages by Penny’s NLP functionality (89%). Average patient-reported adherence was 67% (SD 27%), and patients engaged with 27% (SD 24%) of the chatbot’s weekly check-in messages. In post-study interviews with 12 patients, participants reported that the medication reminders were reliable and user-friendly, whereas the symptom management tool was too simplistic to be helpful. Conclusions: Although Penny has not yet met its feasibility endpoint, the lessons learned from this first cohort have informed further refinements to the platform. Ongoing efforts aim to integrate Penny with the electronic health record and further train the chatbot’s NLP functionality to minimize medication- and symptom-related safety events, respectively. ABSTRACTSurveillance of gonococcal antimicrobial resistance and the molecular characterization of the mechanisms underlying these resistance phenotypes are essential in order to establish correct empirical therapies, as well as to describe the emergence of new mechanisms in local bacterial populations. To address these goals, 149 isolates were collected over a 1-month period (October-November 2008) at the Ontario Public Health Laboratory, Toronto, Canada, and susceptibility profiles (8 antibiotics) were examined. Mutations in previously identified targets or the presence of some enzymes related to resistance (r), nonsusceptibility (ns) (resistant plus intermediate categories), or reduced susceptibility (rs) to the antibiotics tested were also studied. A significant proportion of nonsusceptibility to penicillin (PEN) (89.2%), tetracycline (TET) (72.3%), ciprofloxacin (CIP) (29%), and macrolides (erythromycin [ERY] and azithromycin; 22.3%) was found in these strains. Multidrug resistance was observed in 18.8% of the collection. Although all the strains were susceptible to spectinomycin and extended-spectrum cephalosporins (ESC) (ceftriaxone and cefixime), 9.4% of them displayed reduced susceptibility to extended-spectrum cephalosporins. PBP 2 mosaic structures were found in all of these ESCrsisolates. Alterations in themtrRpromoter, MtrR repressor (TETr, PENns, ESCrs, and ERYns), porin PIB (TETrand PENns), and ribosomal protein S10 (TETr) and double mutations ingyrAandparCquinolone resistance-determining regions (QRDRs) (CIPr) were associated with and presumably responsible for the resistance phenotypes observed. This is the first description of ESCrsin Canada. The detection of this phenotype indicates a change in the epidemiology of this resistance and highlights the importance of continued surveillance to preserve the last antimicrobial options available.

A ficção de Machado de Assis possui vocação filosófica. O objetivo deste artigo é rastrear, a partir de sua obra poética, os elementos filosóficos de uma visão estética de mundo que se torna, na maturidade, cética e pessimista. Na primeira parte, analisam-se as poesias da juventude de Machado que apresentam temas marcadamente cristãos, cujos fundamentos teóricos partem da concepção teológica agostiniana que congregava as duas perspectivas adotadas pelo poeta: o ecletismo romântico e a escatologia soteriológica. Essa concepção teológica é construída de maneira indireta por meio da recepção dos Pensamentos de Pascal. A hipótese é de que a Machado pensa a poesia como uma estratégia metafísica de acesso à verdade. Na segunda parte, considera-se o desenvolvimento deste pensamento ficcional no contexto da filosofia brasileira oitocentista. A filosofia pascaliana é o eixo ao redor do qual os intelectuais brasileiros respondem a questões de ordem política, religiosa e econômica. Discute-se, ao fim, como Machado de Assis reconfigura, em sua obra de juventude, as suas três principais influências: Chateaubriand, Pelletan e Monte Alverne.

IL TEATRO DI TAORMINA: UNA NUOVA CRONOLOGIAIl teatro di Taormina, nonostante la sua importanza, non era mai stato oggetto di nessun dettagliato rilevamento, fino al 1992 quando un team australiano cominciò lo studio delle sue fasi architettoniche. Tale lavoro ha confermato l'esistenza di tre principali fasi costruttive: una fase ellenistica (III secolo a.C.); una ricostruzione romana (inizio del secondo secolo d.C.); e la trasformazione del teatro in un'arena (inizio III secolo d.C.). Comunque il rilevamento ha anche messo in luce il fatto che molte delle strutture tipiche dell'edificio che si credeva appartenessero al periodo del teatro, di fatto vanno associate alla fase in cui esso operava da anfiteatro. La ricostruzione del teatro quale area designata per l'incontro di gladiatori evenationestrasformò radicalmente l'edificio, modificandone molte delle sue parti. L'aspetto dell'edificio nel periodo in cui funzionava da teatro era molto diverso da quello che comunemente si pensa, ed è di particolare interesse poiché il progetto dell'edificio per rappresentazioni teatrali fu influenzato dalla ricostruzione delscaenadel teatro di Pompeo a Roma, probabilmente all'epoca di Domiziano. Ci sono anche indizi che alcune delle caratteristiche architettoniche del teatro possano essere associate alle province orientali e dell'Asia Minore.

We studied the effects of a hydrophobized reversed-phase feed adsorbent in the form of a polyoctylated polysilicate hydrogel (POPSH) on productivity indicators, metabolic adaptation, and on the level of polychlorinated biphenyls (PCBs) in the blood of growing Holsteinized black-and-white heifers during the transition period. Two groups of two-month-old heifers of 20 head each were used. The experimental group received POPSH in addition to the main diet. The use of the adsorbent led to an increase in daily weight gain by 19.9% and to a decrease in the total concentration of PCB congeners found in whole blood by 40%. The greatest decrease in concentration (35–52%) was observed for tetra-, penta- and hexachlorobiphenyls. These results demonstrate the possibilities of effective protection of calves from lipophilic toxins of feed and their active decontamination.

The aim of this study was to evaluate the alveolus-capillary permeability by the lung clearance rate of 99mTc-DTPA(Technetium99m-diethylene triamine penta-acetate), (LCR-DTPA), both in normal and in asymptomatic HIV seroreactive patients. Thirty individuals were studied, 21 seronegative normal volunteers and 9 HIV seroreactive patients presenting normal chest radiography and no respiratory infection symptoms. LCR-DTPA was determined by inhaling 99mTc-DTPA and obtaining images in a gamma camera. The 99mTc-DTPA clearance rate in normal individuals was 0.99±0.15%.min-1 and in patients 2.31±1.25%.min-1. There was a significant statistical difference between the two groups (p< 0.05). Two patients who presented LCR-DTPA higher than 4.3 %.min-1 presented pneumocystis pneumonia one month later. Seroreactive AIDS patients, previously asymptomatic that presented pneumonia later showed higher LCR-DTPA than the seroreactive who did not develop the illness. These results suggested that LCR-DTPA could be a predictive method for the clinical development of pneumocystis pneumonia in asymptomatic HIV seroreactive patients.

e20002 Background: While BCMA-targeted CAR T therapies have yielded unprecedented efficacy in patients with multiple myeloma, most patients (pts) experience disease relapse. Treatment after progression on CAR T has become an unmet need given the limited novel treatment options available. We evaluated clinical outcomes for pts relapsing after BCMA CAR T. Methods: This was a single-center retrospective analysis including pts who had been treated with a BCMA-targeted CAR T therapy prior to 12/31/22 and then had disease progression. Data for all subsequent lines of therapy were collected, and best response to therapy was graded as per IMWG criteria. Statistics were descriptive in nature. Results: Thirty pts treated with BCMA CAR T (14 trial, 16 commercial) relapsed. The cohort of median age 61 had notable disease characteristics of 40% with high-risk FISH and 30% with extramedullary disease (EMD). The median number of prior lines of therapy (LOT) was 5, with 80% triple-class refractory and 47% penta-refractory. Patients treated with late BCMA CAR T (≥ 4 prior LOT; n=20) had higher rates of EMD (40% v 10%), triple-class refractory (90% v 60%), and penta-refractory disease (55% v 30%) compared to pts who received early CAR T (< 4 prior LOT; n=10). At data cutoff, 4 pts died without receiving subsequent therapy and 1 pt was pending start of their first regimen. The remaining 25 pts received 58 salvage therapies, of which 54 regimens were evaluable for response. The overall response rate (ORR) for all regimens was 46%. Median overall survival (OS) was not reached, and the 6 and 12-month OS rates were 83% and 77% respectively. ORR and median progression free survival (PFS) stratified by the specific type of salvage therapy are shown in the table. Patients who received a regimen consisting only of agents they had received in a prior LOT had an ORR of 57% and median PFS of 3.6 months. Patients who received early CAR T had a higher ORR to all subsequent salvage therapies (59% v 33%), longer median PFS (4.6 months v 1.6 months), and higher 12-month OS (90% v 70%). Conclusions: For pts with progression after BCMA CAR T, treatment with another BCMA CAR T or transplant had more favorable outcomes. Retreatment with prior therapies resulted in disease control for a fair number of patients, which can serve as a bridge to another novel therapy. The pts treated with early CAR T had more favorable post-progression outcomes than late CAR T, yet 70% of pts who received late CAR T were alive at 1 year post-progression. [Table: see text]

Field investigations onPerna pernawere carried out in 1999 in an unpolluted site (Cap Ghir) and a wastewater-polluted site (Anza) to determine the contents of glycogen, lipids and proteins in mussel samples collected every month. Sample analyses were made using the entire soft masses of male and female mussels or their gonads only. Compared to unpollutedP. perna, the glycogen and lipid concentrations found in polluted mussels showed significantly quantitative variations throughout the year; in contrast, insignificant differences were only found for protein concentrations. Female gonads contained higher rates of lipids than male ones, whereas protein contents were similar. To determine the response of container-grown shrubs to controlled-release fertilizer (CRF) rate when grown in a temperate climate, Polyon® 19–04–10 + Minors, an 8–9 month CRF, was incorporated into growing substrates for ‘Gro-Low’ fragrant sumac (Rhus aromatica Aiton), ‘Goldmound’ spirea (Spiraea × bumalda Burv.) and ‘Bloomerang’® purple lilac (Syringa × ‘Penda’) transplants. Also, a 15–06–11 + Micros, a 10–12 month CRF, was incorporated into growing substrates for ‘Green Mound’ boxwood (Buxus × ‘Green Mound’), ‘Runyan’ yew (Taxus × media) and ‘Emerald’ white-cedar (arborvitae) (Thuja occidentalis L.) transplants, at six rates (0.15, 0.45, 0.75, 1.05, 1.35 and 1.65 kg·m−3 N; 0.25, 0.76, 1.26, 1.77, 2.28 and 2.78 lb·yd−3 N). We observed greater growth index, leaf area, and shoot dry weight at high vs. low CRF rates for the majority of species. Nutrient deficiency symptoms such as light green leaves were observed at low CRF rates for some species, including fragrant sumac, lilac and white-cedar. Optimal species-specific CRF application rates were 1.05 kg·m−3 N (1.77 lb·yd−3 N) for lilac and yew and 0.45 kg·m−3 N (0.76 lb·yd−3 N) for boxwood and white-cedar, while the optimal CRF ranges were 0.75 to 1.35 kg·m−3 N (1.26 to 2.28 lb·yd−3 N) for fragrant sumac and 0.75 to 1.05 kg·m−3 N (1.26 to 1.77 lb·yd−3 N) for spirea. Adjusting CRF application rates based on plant response may provide nursery growers with an efficient tool for managing nursery crop growth and production timing in the temperate climate.

Abstract Objectives High adherence to the Mediterranean diet (MedDiet) is associated with a risk reduction on cardiovascular and neurodegenerative diseases, cancer and osteoporosis among others. The aim of the present study is to use ecological momentary assessments (EMAs) as a tool to monitor and promote adherence to the MedDiet. Methods A set of dietary EMAs has been developed aimed at evaluating continuous adherence to the MedDiet. The EMAs are sent daily and enquire about the consumption of 11 key food groups of the Mediterranean diet in the last 24–48 hours in a semi-quantitative way. They have been used to monitor and promote the MedDiet in the frame of a multicomponent life style intervention study which is focused on the prevention of cognitive decline (PENSA study). Participants undergo through a nutritional intervention including individual nutritional sessions and daily EMAs. A total MedDiet adherence score is calculated based on EMAs. Additionally, each food group is monitored individually. On a monthly basis, volunteers receive a personalized feedback including a MedDiet score and its individual components together with recommendations of improvement. Additionally, MedDiet adherence was evaluated at baseline and after 6 months of the intervention using the standardized MEDAS screener. Results Preliminary results (n = 11) show that, following 6 months of the intensive nutritional intervention and dietary EMAs, participants increased significantly their adherence to MedDiet (baseline score: 8 (SD = 1.4) vs 6-month score: 10.6 (SD = 0.9); P &lt; 0.001). At baseline, only 2 volunteers met “high adherence” criteria to the MedDiet whereas at the end of the 6 months, 9 volunteers over 11 were categorized into the “high adherence” group. The most important changes observed in MEDAS 6-month score where in the nuts, fruits and vegetable categories; all of them included in the food groups monitored by EMAs. Conclusions EMAs are a tool to monitor and promote the adherence to the MedDiet. The continuous monitoring of the diet together with the periodic personalized feedbacks send to the volunteers promoted a change in individual's behaviors in favor of the choice of products in line with MedDiet recommendations. Background: Teclistamab is a B-cell maturation antigen (BCMA) targeting bispecific T-cell engager approved in October of 2022 for patients with relapsed-refractory multiple myeloma (RRMM) who have received at least 4 prior lines of therapy (LOT) based on the results of the pivotal MajesTEC-1 trial. In this multicenter retrospective study, we evaluated the real-world safety and efficacy of teclistamab, highlighting patterns of utilization and outcomes, including patients who would have been considered ineligible for MajesTEC-1 trial. Methods: Five U.S. academic centers, part of the US Myeloma Innovations Research Collaborative (USMIRC), contributed data to this retrospective analysis, which included 102 patients with RRMM who received teclistamab as of 7/1/2023. Baseline characteristics were outlined by descriptive analysis. Teclistamab was administered in a step-up dosing manner as per the package insert. All patients had received at least the two step up doses and the first full dose of teclistamab. Patients received antimicrobial prophylaxis, supportive care, and toxicity management per institutional protocols. Responses to therapy including overall response rate (ORR), very good partial response (VGPR), and complete response or better (≥CR) were evaluated using the International Myeloma Working group (IMWG) criteria. Adverse events were graded based on the CTCAE v5.0 criteria. The Kaplan-Meier method was used for progression free survival (PFS), duration of response (DOR), and overall survival (OS) calculation. Results: Of the 102 patients included in this analysis, 25% were non-Hispanic Black and 44% had extramedullary disease (EMD); the median number of prior LOT was 6 (range 4-17). More than 80% (83/102) of the patients would not have met the MajesTEC-1 eligibility criteria; main reasons for ineligibility included: prior BCMA-directed therapy (BDT) (55%), ECOG performance status ≥2 (28%), and baseline cytopenias: grade 3-4 anemia (26%), and grade 3-4 thrombocytopenia (20%). All patients were triple class exposed and 92% were triple class refractory, 78% were penta-class exposed and 67% were penta-class refractory. More than half of the patients (55%) were refractory to prior BDT. Baseline characteristics are summarized in Table 1. Cytokine release syndrome (CRS) was observed in 65% of patients, all of which, except one, were grade 1-2 events, while immune effector cell-associated neurotoxicity syndrome (ICANS) was reported in 15% of the patients, with three patients having grade 3-4 events. The most common grade 3-4 hematological toxicities were neutropenia (23%), leukopenia (18%), thrombocytopenia (17%), and anemia (17%). Infections occurred in 28% of the patients, half of which were characterized as severe. The vast majority (80%) of infections involved the respiratory tract and 7% of patients discontinued teclistamab permanently due to recurrent or severe infections. After a median follow up of 3.2 months, the ORR was 64% for the entire cohort; ORR was 57%, 66% and 45% for patients who were BDT-refractory, penta-refractory, or had EMD, respectively. Of those who did respond, responses were found to be deep (72% ≥VGPR; 45% ≥CR). Patients that would have been ineligible for the MajesTEC-1 trial had an ORR of 59%. Detailed responses for the groups of interest are reported separately in Table 1. The estimated 6-month PFS, DOR, and OS of the entire cohort were 38% (95% CI, 26-56), 54% (95% CI, 34-85) and 68% (95% CI, 58-80), respectively (Figure 1). No treatment-related mortality was observed, although 26% of the patients had died at the time of data cut-off, with 89% of the deaths attributed to disease progression. Conclusion: This is the largest real-world experience of the safety and efficacy of teclistamab for RRMM. Overall, teclistamab was well tolerated with no major safety concerns, despite worse performance status and cytopenias than the MajestTEC-1 trial population. Moreover, despite the increased incidence of high-risk features, including penta-refractoriness, BDT refractoriness, and presence of EMD in this heavily pre-treated RRMM patient population, early efficacy assessment of teclistamab in a real-world setting was encouraging.

The residual value of manganese fertiliser for seed (grain) production of lupin (Lupinus angustifolius L.) was measured in 2 long-term (16 years) field experiments on 2 different soils (sand, lateritic ironstone gravel–sand) when manganese sulfate was placed (drilled) with the seed at about 5 cm while sowing. Relative to the nil-manganese treatment, additions of manganese fertiliser increased yields by about 1.0 t seed/ha and decreased the amount of split seed from about 80% to less than 5%. Additions of manganese fertiliser did not affect plant density (measured 1 month after sowing) or the yield of dried shoots. The residual value of manganese fertiliser was measured in 1994 for grain production of lupin, for manganese applied once only to plots not treated with manganese before, either in 1994 (current manganese) or in a previous year (previous manganese), applied in one of the following years: 1978, 1982, 1988, 1991. The residual value of previous manganese for grain production decreased relative to the effectiveness of current manganese, the decrease increased with increasing time since application. The decrease in the relative value varied with soil type. For example, the sandy soil showed a larger decrease in the effectiveness of manganese compared with the gravelly sand. For the sandy soil, 16 years after application the manganese application was about one fifth as effective as current manganese, for lupin grain production. As measured using soil test manganese, critical soil manganese values for diethtlenetriamine penta-acetic acid, ammonium acetate, and ammonium acetate plus hydoxy quinone extraction varied with soil type. For example, the diethtlenetriamine penta-acetic acid critical concentration of manganese for grain yield for the sand was about 2 mg/kg compared with about 4 mg/kg for the gravelly sand. The critical concentration of manganese in the youngest vegetative bud for maximum seed yield was about 50 mg/kg while the critical concentration of manganese in the stem at mid-flowering of the main stem was about 20 mg/kg. Having a manganese concentration above the critical concentration of manganese in either plant tissues resulted in split seed levels of less than 5%.

European Radiation Dosimetry Group (EURADOS) Working Group 7 is a network on internal dosimetry that brings together researchers from more than 60 institutions in 21 countries. The work of the group is organised into task groups that focus on different aspects, such as development and implementation of biokinetic models (e.g. for diethylenetriamine penta-acetic acid decorporation therapy), individual monitoring and the dose assessment process, Monte Carlo simulations for internal dosimetry, uncertainties in internal dosimetry, and internal microdosimetry. Several intercomparison exercises and training courses have been organised. The IDEAS guidelines, which describe – based on the International Commission on Radiological Protection’s (ICRP) biokinetic models and dose coefficients – a structured approach to the assessment of internal doses from monitoring data, are maintained and updated by the group. In addition, Technical Recommendations for Monitoring Individuals for Occupational Intakes of Radionuclides have been elaborated on behalf of the European Commission, DG-ENER (TECHREC Project, 2014–2016, coordinated by EURADOS). Quality assurance of the ICRP biokinetic models by calculation of retention and excretion functions for different scenarios has been performed and feedback was provided to ICRP. An uncertainty study of the recent caesium biokinetic model quantified the overall uncertainties, and identified the sensitive parameters of the model. A report with guidance on the application of ICRP biokinetic models and dose coefficients is being drafted at present. These and other examples of the group’s activities, which complement the work of ICRP, are presented.

Background Afghanistan has the lowest routine immunization coverage, according to the WHO-UNICEF reports. The coverage rate of Penta3 in Iran, India, Pakistan, and Afghanistan are estimated to be 99%, 89%, 75%, and 66%, respectively. Objective This study aimed to find the real immunization coverage in urban areas and factors related to vaccinated and unvaccinated 12- to 23-month children, in 2018, in Sarepol province. Methods A descriptive cross-sectional study with probability proportional to size (PPS) cluster sampling was conducted and modified for application to surveys of immunization coverage. We selected 30 clusters and randomly selected 7 households from each cluster in the urban setting of Sarepol province. The children’s age was calculated, in months, with respect to the 1st day of the survey. We designed a comprehensive questionnaire, and 210 questionnaires were filled. The data were managed and analyzed in Epi Info v.7. Results This survey shows EPI routine coverage for 12- to 23-month children for BCG, Meales-1, Penta1, and Penta3, which were 97.14%, 77.14%, 93.81%, and 83.81%, respectively. A full immunization coverage by gender—80.18% for girls and 71.15% for boys—was reported. The dropout rate of vaccination among Penta1, Penta3, and BCG was 9.27%, and for Measles-1 was 18.90%. Moreover, 2.86% of 12- to 23-month children did not receive any vaccine in these urban areas. Children’s illness, emigration, distant health facilities, and the gaps between the doses were reported by the respondents as the main reasons for incomplete or no vaccination. The valid doses administered for BCG, measles, and Penta3 were calculated to be 93.80%, 71.43%, and 80%, respectively. Conclusions It is observed that access and use of immunization services in urban areas have improved because full immunization was 75% compared with the AHS-2018 survey’s 61%. However, there are still many children who have not received any vaccine. High immunization dropout rates could be overcome by creating awareness of the program and of the importance of second and third doses of penta, polio, and measles vaccines. Measles coverage is very low, and we are expecting more outbreaks in urban areas. We therefore suggest that the Ministry of Public Health better enhance awareness and implement measles campaigns.

AbstractAsian green mussel is commercially farmed in tidal waters in several enclosed bays in Sabah, Malaysia. In this study, two areas on the west coast of Sabah – Ambong Bay and Marudu Bay – were selected for the monitoring of the growth and mortality rates of green mussels farmed in suspension raft. Both growth and survival rates were then correlated with physicochemical parameters (dissolved oxygen, pH, salinity, temperature, water transparency, chlorophyll-a), nutrients (phosphate (PO43-), ammonia (NH3-N), nitrate (NO3-N) and nitrite (NO2-N) and condition index of mussel from each study site, as well as between the study sites. The twelve-month growth study (September 2017 to August 2018) was started with a total of 180 mussel specimens (90 at each site). The initial size (mean) of the mussel seed used was 47.7 ± 3.5 mm and 51.1 ± 3.9 mm for Marudu Bay and Ambong Bay, respectively. Mussels in Marudu Bay attained mean size of 73.47 ± 11.05 mm (SGR 0.17% ± 0.22) compared to 64.05 ± 7.44 mm (SGR 0.11% ± 0.22) for Ambong Bay at the end of the experiment. The cumulative mortality rates were 9.2% ± 4.9 and 55.5% ± 30.0 for Marudu Bay and Ambong Bay, respectively. The Pearson correlation indicated a significant positive relationship between mortality and water transparency (r = 0.684, p<0.01). There was a significant negative relationship between ammonia in seawater and mussel mortality (r = -0.561, p<0.01), as well as significant negative relationships between nitrate and growth (r = -0.480, p<0.05) and mortality (r = -0.460, p<0.05), as indicated by Spearman’s Rank-order Correlation analysis. Overall, the growth performance of green mussels farmed in Marudu Bay was better than in Ambong Bay, however, the mortality of mussels in Ambong Bay was higher.

ABSTRACTCultivation of green mussels (Perna viridis) using multilevel plastic baskets has been carried out in the waters of Salemo Island, Pangkep Regency, Indonesia. The purpose of this study was to determine the oceanographic parameters that affect mussel cultivation at different water depths. The research treatment was placement on the surface and bottom of the waters (five meters deep) with different stocking densities. Green mussel cultivation is carried out for 4 months, namely May, June, July and August. Measurement of oceanographic parameters was carried out every month during green mussel cultivation, in situ, namely temperature, turbidity, salinity, pH, DO and those analyzed in the laboratory, namely TSS, TOM, Nitrate, Phosphate and the composition and abundance of plankton. Growth measurements of green mussels were also carried out, namely absolute weight and survival (SR). The results showed that the condition of oceanographic parameters differed between the surface and bottom waters, especially the parameters of TSS, turbidity, TOM, nutrients and plankton abundance. Background: The global COVID-19 pandemic is overstretching existing health systems. The COVID-19 pandemic has prompted the world to implement drastic prevention methods based on limiting population movements that have an impact on public health policies such as vaccination. This study aimed to assess the impact of COVID-19 on routine immunization at an immunoprophylaxis clinic of the tertiary care centre.Methods: a record-based cross-sectional study was carried out during the 4 months, (February to May) 2020 in the Immunization Clinic of Tertiary Care Center of central India. Vaccines included in the study were OPV, IPV, ROTA, PENTA, MR, and DPT. Data analysis was done by using software open EPI, p<0.05 was considered to be statistically significant.Results: The study showed a prominent decline in the percentage of children who came for immunization during the period of lockdown due to the COVID-19 Pandemic. This downward trend continued from the month of February, as the months progressed and till May arrived, the fraction of children vaccinated in 2020 as compared to 2019 saw a very significant reduction, (p<0.000). individual vaccine-wise immunization status of under 5 children also shows a downward trend in the year 2020 as compared to the year 2019.Conclusions: There was a sharp decline in the utilization of immunization services in a tertiary care center in central India.

Perna perna(Linnaeus, 1758) is the main marine bivalve mussel yielded commercially in Brazil. In spite of this, scientific data is very scarce regarding its productivity in tropical shallow waters. The Condition Index (CI) is used worldwide in mariculture to assess animal health, harvest time, and yield. In this study, the authors used CI results from nine different methods to assess the season effect on the mussel CI and also to evaluate the potential yield of three southern Brazilian bays. The results from nine CI methods were used for the comparison of the seasonality and yield of mussels reared in three marine bays. Sampling was carried out monthly within two 4-month periods, from December 2008 to August 2009. The results show a trend for seasonal effects on the CI results. The winter months showed the highest and the lowest values. Between bays, higher CI values were detected in animals reared at Sepetiba Bay, followed by Guanabara Bay and Ilha Grande Bay. We suggest that the CI (that considers the ratio between bivalve soft tissue wet weight and total length) should be used by fishermen, since this formula was able to detect differences between sites and is more easily applied.

Background: Vaccination is a cost-effective way of preventing disease and promoting health. Most vaccines have little or no side effects. Although most vaccine adverse events are self-limiting and non-life threatening, some may be severe with long term health implications. Massive proteinuria with Nephrotic syndrome (NS) and severe Acute kidney injury (AKI) secondary to acute interstitial nephritis and membranous nephropathy have been reported rarely following the 2009 H1N1 infuenza vaccine. There has been no reported case of massive proteinuria with severe AKI following Pentavalent vaccination in Nigerian children. Case summary: We report a previously healthy two months old female who developed sudden onset generalized urticarial rash, acute pulmonary oedema, anuria, proteinuria (++), haematuria(+) and severe AKI (serum creatinine- 140umol/L, urea- 7.3umol/L, potassium-4.5umol/L; following vaccination with pentavalent vaccine. Symptoms resolved following fluid management and diuretics. A month later, she had another episode of AKI with massive proteinuria within 24 hours of a 2nd dose of Pentavalent vaccine. She also responded well to conservative management and has remained stable post discharge. She is still being followed up. It is possible that some cases of NS with AKI and glomerulonephritis may be secondary to hitherto unrecognized underlying pathology such as Vaccine induced membranous nephropathy (VIMN). Conclusion: Thus, vaccine induced AKI should be suspected in children who develop sudden-onset AKI following immunization particularly in resource-poor centers where biopsy and electron microscopy are not readily available, Key words: Penta vaccine, Membranous nephropathy, Vaccination

Abstract Introduction: Selinexor is a novel, oral Selective Inhibitor of Nuclear Export (SINE) compound that blocks exportin 1 (XPO1). Selinexor treatment results in nuclear accumulation and activation of tumor suppressor proteins, inhibition of NF-kB, and translational suppression of several oncoprotein mRNAs (e.g., c-myc, cyclin D).Multiple myeloma (MM) remains incurable, and most patients (pts) eventually progress through standard drug classes of proteasome inhibitors (PIs), immunomodulatory drugs (IMiDs), anti-CD38 mAbs and others. The increased use of combinations in MM treatment, (PIs/IMiDs/mAbs), has led to a growing number of pts with penta-refractory MM (pts that have been treated with bortezomib (bort), carfilzomib (carfil), lenalidomide (len), pomalidomide (pom) and daratumumab (dara)). Active novel therapies with different mechanisms of actions are needed to address this unmet medical need. Part 1 of STORM enrolled pts with both quad- (bort, carfil, len, pom, treated MM) or penta-refractory MM and demonstrated an overall response rate (ORR) of 21% (Vogl et al, JCO 2018). Based on these findings, the Pivotal Part 2 of STORM was initiated, enrolling an additional cohort of 122 patients with penta-refractory MM. Methods: Pts with penta-refractory MM were treated with 80 mg selinexor plus 20 mg dexamethasone (Sd) twice weekly. Pts must have received an alkylator, bort, carfil, len, pom and dara, and had MM refractory to ≥1 PI, ≥1 IMiD, dara, a glucocorticoid, and their last therapy. Pts must have a total ANC ≥1000 mm3,platelets ≥50k/mm3 (or ≥75k if marrow plasma cells <50%), and creatinine clearance ≥20 mL/min. The primary endpoint was ORR. Secondary endpoints: duration of response (DOR), clinical benefit rate (CBR), progression free survival (PFS), and overall survival (OS). Efficacy was assessed by an Independent Review Committee (IRC) based on IMWG criteria. OS was also compared to a cohort of pts with PI, IMiD, dara refractory MM from the Flatiron Health Analytic Database (FHAD), (ref: ASH 2018 abs ID: 116493),who met all the inclusion criteria for STORM. Results: As of 1-Jun-2018, 122 pts (71 M/ 51 F) were enrolled in 38 sites (US and EU). Pt characteristics were [medians (range)]: age 65 yrs (40-85); 7 (3 - 18) prior treatment regimens, 6.6 yrs (<1 - 23.4) from initial MM diagnosis.65 pts (53%) had high risk cytogenetics, 86 pts (70%) had prior dara in combination, 102 pts (84%) had prior stem cell transplantation, 2 pts had prior CAR-T therapy. All pts enrolled with progressive disease (PD), 72% of pts had increases (3% - 792%) in MM markers from screening to C1D1 (median 11 days). Frequently reported Sd treatment related adverse events (AEs) included (all grades, grades 3/4): thrombocytopenia (67%, 53%), nausea (67%, 10%), fatigue (68%, 21%), anorexia (50%, 2%), anemia (46%, 28%), and weight loss (46%, 0%). Eight pts remain on study and 114 pts discontinued treatment (most commonly for PD). There were 4 deaths on treatment: sepsis, respiratory failure, pulmonary embolism, and an unrelated, unspecified cardiac event. IRC determined ORR (≥PR) was 26.2%, with 6.5% ≥ very good partial response, including 2 stringent complete responses (sCRs; MRD negative at 1:10-6 and at 1:10-4sensitivity). Both pts who relapsed after CAR-T achieved PRs. The CBR (≥minimal response, [MR]) was 39.3%, and 79% of pts achieved ≥stable disease (SD). Responses typically occurred within the first month. Medians: DOR 4.4 months (mo) (range <1 - 10 mo), PFS 3.7 mo, and OS 8.0 mo. Pts with ≥MR had significantly longer OS than pts with PD/NE (median not reached vs 1.9 mo, p=<0.0001). Compared to the FHAD cohort, STORM cohort had longer OS (Figure 1, HR 0.41, p=0.0001). Conclusions: Results of the pivotal STORM Part 2 in penta (PI, IMiD, dara)-refractory MM demonstrated that oral selinexor plus low-dose dexamethasone (Sd) was highly active with an ORR of 26.2%. Importantly, responses were rapid and deep with 2 patients achieving sCRs (both MRD negative) in these heavily pre-treated penta-refractory MM pts (median 7 prior regimens, 53% high risk). AEs are a function of dose/schedule/disease severity and can be managed with dose modifications and supportive care. No major organ toxicity was observed and AEs were typically transient and reversible. Sd is an all-oral, first in class mechanism with novel MOA and represents a potential therapeutic option to the growing number of pts with penta-refractory MM who have exhausted approved therapies.
