AbstractExtending the three now well-known modes of immigrant labor market incorporation formulated by sociologist Alejandro Portes in 1981, this article proposes that transnationalism can be viewed as a new mode of immigrant labor market incorporation in the age of globalization. The article conceptualizes this new mode of immigrant incorporation and illustrates it with data from unstructured interviews of Chinese transnational migrants in the US and in China. It is found that the transnational mode of incorporation often offers Chinese transnational migrants business and career opportunities and rewards not available in other modes of incorporation, although it sometimes involves risks, uncertainties, and difficulties. Chinese transmigrants with a higher initial social standing, an advanced training, and a dense network are more likely to experience upward social mobility than those without such advantages. Implications of the arguments and findings are also discussed.

Centriole stability and distribution during the mammalian cell cycle was studied by microinjecting biotinylated tubulin into early G1 cells and analyzing the pattern of incorporation into centrioles. Cells were extracted and cold treated to depolymerize labile microtubules, allowing the fluorescent microscopic visualization of the stable centrioles. The ability to detect single centrioles was confirmed by use of correlative electron microscopy. Indirect hapten and immunofluorescent labeling of biotinylated and total tubulin permitted us to distinguish newly formed from preexisting centrioles. Daughter centrioles incorporated biotinylated tubulin, and at mitosis each cell received a centrosome containing one new and one old centriole. We conclude that in each cell cycle tubulin incorporation into centrioles is conservative, and centriole distribution is semiconservative.

Based on a review of existing qualitative research, this study proposes a gender-based interpretation and critique of the incorporation of self-spirituality into mainstream organizations. The article’s findings show that the enactment of self-spirituality in organizations often evokes experiences of tension or objection. The basic premises of self-spirituality culture are concerned with the authentic self and the emphasis on the individual’s awareness of his or her body, thoughts, and feelings is, in fact, a manifesto of the legitimate sources of ‘knowing’ at work. This premise is radically different from rationality as a fundamental principle of knowing and organizing. Self-spirituality further proposes an alternative to workplace relationships, which is based on a radical equality. I argue that these perceptions are embodied in gendered power relations—the relationship between the feminine self-spirituality and the masculine secular organizations. Two main modes of incorporation of self-spirituality into organizations were identified: the ‘domesticated masculine mode of incorporation’, which is presented as a ‘joining force’ in achieving main organizational values, and the ‘feminine’ modes of the incorporation of self-spirituality, which presents a revolutionary alternative to organizations. As for the viability of these modes of incorporation for creating change, self-spirituality in its domesticated masculine mode of incorporation appears to align itself with organizations public domain. In contrast to this, the two ‘feminine’ modes of incorporation create the possibility for self-spirituality to be lived by individuals as an ‘individual wisdom’. The study contributes to organization research by identifying ways in which radical alternatives may be incorporated into organizations.

The incorporation of collaborative filtering recommendation algorithms into blended teaching approaches for college English marks a big step forward in language education. This study investigates the theoretical underpinnings, practical ramifications, and pedagogical issues for adding collaborative filtering algorithms into blended learning environments. Collaborative filtering algorithms produce individualized learning resource recommendations based on user interactions and preferences, increasing student engagement and motivation. However, issues like as data privacy, algorithmic biases, and user approval must be addressed to ensure that collaborative filtering is implemented effectively in the classroom. This study explains the potential of collaborative filtering recommendation algorithms to improve language learning results in college English courses by doing a thorough literature review. It also provides ideas for educators and policymakers on how to handle the complexity of incorporating collaborative filtering recommendation algorithms into blended learning models. Finally, this paper adds to the promotion of novel pedagogical approaches and the improvement of language learning experiences among college students. As a result, the enhanced collaborative filtering recommendation system had a slightly greater recall rate and precision than the pre-optimized method.

This article analyzes sociocultural transnational linkages among Colombian, Dominican, and Salvadoran immigrants in the United States. It emphasizes the importance of comparative analysis and yields three main findings. First, participation in any particular transnational activity is low, but participation over all the different forms of transnational practices is extended. Second, the process of incorporation does not weaken transnational participation. Third, there is more than one causal path that can account for the rise of transnational sociocultural practices. The different paths can be explained by reference to the context of reception and the mode of incorporation of each group.

Mode choice models have been used widely to forecast the relative probabilities of using available travel modes. These depend on mode-related and traveler-related characteristics. On the other hand, smartphones are increasingly being used to collect sensors’ data relating to trips made after selection of a suitable mode. Such sensors’ data may be correlated with decision-making process of travelers regarding travel mode selection. Discrete Choice Modelling is used to simulate this decision-making process by computing utilities of various travel alternatives, and then calculating their respective probabilities of being selected. In this paper, multinomial logit (MNL) mode choice model is utilized to enhance the prediction capacity of supervised learning algorithm i.e. Weighted Random Forest. To make the procedure less energy-intensive, GPS data was used only to locate the origin and destination of any trip, to be incorporated in mode choice model. Afterwards only accelerometer data was utilized in feature selection for the learning algorithm. One tenth of the classified data was used to train the algorithm whereas rest was used to test it. Results suggested that with incorporation of MNL, the overall prediction accuracy of learning algorithm was increased from 93.75% to 99.08%.

La thèse vise à analyser ce qui détermine les conditions d’accès au marché du travail (la mise au travail) des personnes venues en France par le système d’asile (les gouverné·es de l’asile) et à penser les trajectoires économiques, prises dans des modalités d’assignation et d’exploitation spécifiques, d’une immigration cadrée comme politique. En s’appuyant sur un dialogue critique avec le concept de mode d’incorporation, les premiers chapitres historiques reconstituent, à partir d’archives croisées d’administrations et d’associations, l’incorporation économique différentielle (c’est-à- dire structurant et structurée par différents rapports sociaux de domination) des réfugié·es en France, des années 1920 aux années 2010. Je retrace ainsi l’émergence d’un régime spécifique de mise au travail des réfugié·es, puis sa scission à partir des années 1980, en analysant dans chaque configuration historique le jeu croisé des individus et organisations formant le sous-champ bureaucratique de gestion des réfugié·es : administrations, organisations et membres de l’espace de la cause des réfugié·es ou acteurs économiques. La spécificité de la mise au travail des réfugié·es est alors à chercher dans les droits acquis, dans les dispositifs concrets ou encore dans des modes spécifiques de contrôle des mobilités (camps et centres). À partir d’un terrain au ministère de l’Intérieur et d’entretiens réalisés en 2020 et 2021 avec des acteurs associatifs, privés lucratifs et publics, je m’intéresse ensuite à la politique publique dite d’« intégration professionnelle des réfugiés » déployée à partir de 2015. J’étudie la genèse sous tension de cette politique à la lumière des clivages du champ administratif. Je montre l’effet de l’investissement d’une multiplicité d’acteur·ices du privé lucratif sur le cadrage discursif de l’emploi des réfugié·es, l’organisation de l’action publique et la structuration du champ associatif. J’analyse également la production d’un français au rabais et d’une garantie de compétences dégradées par des associations d’intermédiation de l’emploi des réfugié·es prises en étau entre l’État et le marché. Enfin, à partir de l’exploitation statistique de l’enquête ELIPA 2 et d’entretiens réalisés auprès de personnes exilées, je tente de comprendre les statuts et trajectoires d’activité des réfugié·es à la fin des années 2010 en articulant méthode qualitative et méthodes quantitatives dans une perspective intersectionnelle. À travers différentes périodes et différentes méthodes, je propose donc d’analyser la mise au travail des gouverné·es de l’asile en France comme un produit des modes d’incorporation économique différentiels. Si les conditions matérielles et juridiques des réfugié·es favorisent dans l’ensemble la disponibilité de leur force de travail dans les segments subalternes de l’appareil productif, venant renforcer une division classiste, genrée et ethnoraciale du travail, certains groupes et certaines personnes sont à même d’opposer aux contraintes institutionnelles des ressources collectives ou individuelles, c’est-à-dire d’être un peu moins gouverné·es
The aim of this thesis is to analyze the factors that determine the conditions of access to the labor market for individuals who arrive in France through the asylum system – whom I refer to as “governed by asylum”. I explore the economic trajectories of these immigrants often framed as political. Drawing on a critical dialogue with the concept of “modes of incorporation”, the first chapters take a historical perspective in order to trace the differential modes of economic incorporation of refugees in France from the 1920s to the 2010s, which both structure and are structured by various social relations of domination. Using a combination of archival materials from government agencies and charities, I retrace the development of a specific system for employing refugees and its fragmentation from the 1980s onwards. In each historical configuration, I analyze the interactions between individuals and organizations within the bureaucratic subfield of refugee management : administrations, nonprofit organizations and members of the field of refugee advocacy, as well as economic actors. The specific nature of the process of refugee labor market integration is to be found in the various rights acquired, in concrete arrangements or in specific modes of mobility control (camps and accommodation centers). Based on fieldwork conducted at the French Interior Ministry and interviews carried out in 2020 and 2021 with actors from the nonprofit, public and private sectors, I then focus on the public policy concerning the “professional integration of refugees” introduced from 2015 onwards. I examine the policy’s contentious origin in light of divisions within the administrative field and demonstrate how the involvement of various private actors influences the discursive framing of refugee employment, the organization of public action and the structure of the nonprofit organizations field. I also analyze how nonprofits, acting as intermediaries in refugee employment and caught between state and market pressures, produce a simplified version of the French language proficiency among refugees and downgraded professional qualifications. Finally, based on a statistical analysis of the ELIPA 2 survey as well as interviews with exiles, I aim to scrutinize the employment status and trajectories of refugees at the end of the 2010s by combining qualitative and quantitative methods from an intersectional point of view. Across different periods and different methods, I propose to consider the employment of refugees and asylum seekers in France as a result of differential modes of economic incorporation. While the material and legal conditions of refugees generally restrict their labor power to the subordinate jobs of the productive system – reinforcing a classist, gendered and racial division of labor – certain groups and individuals manage to be somewhat less governed than others, countering institutional constraints through collective or individual resources

In this thesis, we propose a methodology for incorporating attitudinal data in a choice model to capture unobservable heterogeneity across the population. The key features of this approach are, 1) the concept of latent attitudes, and the assumption that 2) the respondent's answers to psychometric attitudinal questions relating to the importance of attributes are manifestations of these attitudes and that 3) those attitudinal data bring sufficient information to capture unobservable heterogeneity across the population in the context of choice behavior. Each individual is probabilistically assigned to a finite number of segments according to his/her own value of latent attitudinal variable(s) as well as to threshold parameter(s) common to the population. Segment-specific parameters are estimated simultaneously. An empirical case study on shopping trip mode choice demonstrates the effectiveness of the methodology.

This thesis makes a contribution to three areas of sociological thought. First, it is concerned with the elaboration and extension of the political economy approach to migration as it is represented in the work of Stephen Castles and his various co-authors. It suggests that the work of Castles, et al., is relatively silent on the role of the state, and ideological relations in the structuration of migration. In seeking to further refine the political economy framework as it is applied to migration, this thesis draws upon two other sets of literature which, in part, have emerged as counters to some of the more economistic of their formulations. In this light, the second area of sociological literature I draw upon is the recent work on the concepts of free and unfree labour. Finally, this thesis is informed by an analysis of recent debates on the concept of racialization. In synthesizing these three strands of sociology, this thesis advances the theoretical claim that political economy oriented theorists should focus on modes of incorporation, or the manner in which foreign-born labour articulates with capital and the state. Within this context, four distinct modes of incorporation under capitalism are identified. These modes of incorporation are designated as: free immigrant labour, unfree immigrant labour, free migrant labour and unfree migrant labour. This thesis suggests that agents are subject to particular modes of incorporation, in part, on the basis on the process of racialization. This thesis uses the cases of late nineteenth and early twentieth century Chinese migration to Canada, and the post-1945 migration of farm labourers, from a number of source countries, including, specifically, the Netherlands, Poland, Germany, and the Caribbean, to the south western Ontario fruit and vegetable industry to highlight the centrality of the state in the process of migration, and the differential modes of incorporation of foreign-born persons into sites in production relations. Furthermore, the process of racialization is seen to have an impact on whether particular groups are allowed entry to a social formation, and upon how they are incorporated into sites in production relations.

A topographic steering river meandering (TSRM) model based on continuity of a simplified flow field through bends is developed. The equilibrium coupling between helical flow and sediment transport, as in Ikeda [1989] determines downstream variation of transverse bed slope. The model tests the hypothesis that meander development patterns can be captured under the assumptions: (a) bank shear stress arises from forces associated with topographically induced convective accelerations; (b) turbulent boundary layer dissipation of these forces at the banks is sufficiently represented by gaussian smoothing at a parameterized scale; and (c) lateral migration of the channel is proportional to bank shear stress. The resulting TSRM model produces realistic complex meander patterns and scroll bar-like topography. Model compound bend formation is compared to a field case and found to arise from the nonlinear interaction of bank roughness and channel hydraulics scales. When the latter is short relative to the former, maximum bank shear stress occurs early in the bend and leads to compound bend formation. New statistical stream sinuosity and spatial coordinate variation measures are applied to both natural and model streams and reveal secondary sinuosities arising from compound bend formation in both cases. Scroll bar topography and channel bank roughness are studied in the field to compare natural and model mechanisms. A channel-hillslope integrated landscape development (CHILD) model incorporates the TSRM model. The CHILD model represents the landscape as an irregular, Delaunay triangulated mesh of landscape nodes that may be moved, deleted, or added to accommodate meandering channels that are in general discretized at different spatial resolution than the surrounding landscape. The interactions among meandering, bank erodibility's bank height dependence, and uplift rate in a detachment-limited river valley are examined. An equilibrium landscape adjusts to the onset of meandering and approaches a new dynamic equilibrium. For the detachment-limited case, the hypothesis that meandering is more active when uplift is quiescent is rejected. When bank erodibility's bank height dependence is greater, bend scale sinuosity is smaller, but the tendency toward multi-bend loop formation is reinforced.

Dissertation to obtain a Master Degree in Biotechnology
This work is divided in two parts comprising two distinct areas of science that are bonded with a functional purpose: study of the polyphenolic composition in leaves of three coffee genotypes and their responses to cold and drought stresses; and analysis of the effect of incorporation of some polyphenolic compounds in a microemulsion model system. Three genotypes of coffee, Icatu Obatã and Apoatã, were subjected to temperature and water content stress conditions and their sensitivity or resistance to the treatments imposed was investigated. Responses were analyzed from phenolic fraction point of view in the leaves of coffee plants. It was found that polyphenols are actually involved in the response to these abiotic stresses and that the 4.5-diCQA seems to have an essential role in the recovery from cold in Obatã. Various phenolic compounds were identified and quantified, being the majority: 5-CQA, epicatechin, procyanidin, 3,4-diCQA, 3.5-diCQA, 4.5-diCQA, 5-FQA and mangiferin that was discovery as a new polyphenol in C. arabica and also works as a biomarker to differentiate between C. arabica and C. canephora. It was confirmed that Apoatã (Coffea canephora) possesses a greater amount of polyphenols than Obatã and Icatu (Coffea arabica). It was ascertained that Icatu and Obatã seem to be more resistant to cold stress than Apoatã, but more sensitive to drought than Apoatã. The model microemulsions is build up from a nonionic surfactant, C12E5, decane and water, a system already well characterized. Two antioxidants, 5-CQA and α-tocopherol, and a drug, lidocaine, were incorporated in microemulsions, being the effect of its addition studied through phase diagrams and DLS. A general model was proposed, arguing that the addition of a hydrophilic molecule increases the temperature of the microemulsion phase (micelles), while a hydrophobic decreases, which is of most importance for pharmaceutical applications. The antioxidant activity is not significantly affected within and outside the microemulsion and was determined the existence of synergistic effect when mixing 5-CQA and α-tocopherol in this system.

Large scale software development efforts in the past decade have posed a problem in terms of the reliability of the software. The size and complexity of software that is being developed is growing rapidly and integrating diverse pieces of software in the operational environment also poses severe reliability issues, resulting in increased development and operational costs. A number of reliability models have been defined in the literature to deal with problems of this kind. However, most of these models treat the system as a "black box" and do not consider the complexity of the software in its reliability predictions. Also, reliability is predicted after the system had been completely developed leaving little scope for any major design changes to improve system reliability. This thesis reports on an effort to develop a reliability model based on complexity metrics which characterize a software system and runtime metrics which reflect the degree of testing of the system. A complete development of the reliability model is presented here. The model is simple and reflects on our intuition of the software development process and our understanding of the significance of the complexity metrics. Credibility analysis is done on the model by simulating a number of systems and applying the model. Data collected from three FORTRAN coded systems developed for NASA Goddard was used as representative of the actual software systems. An analysis of the results is finally presented.

Abstract The restoring forces on membranes are due to the applied tension, while the restoring forces for plates are due to the flexural rigidity of the plate’s material. The transition to two dimensions introduces some features that did not show up in our analysis of one-dimensional vibrating systems. Instead of applying boundary conditions at one or two points, those constraints will have to be applied along a line or a curve. In this way, incorporation of the boundary condition is linked inexorably to the choice of coordinate systems used to describe the resultant normal mode shape functions. For two-dimensional vibrators, two indices are required to specify the frequency of a normal mode, fm,n, with the number of modes in a given frequency interval increasing in proportion to the center frequency of the interval, even though that interval remains a fixed frequency span. It is also possible that modes with different mode numbers might correspond to the same frequency of vibration, a situation that is designated as “modal degeneracy.” A membrane’s response to sound pressures provides the basis for broadband condenser microphone technology that produces signals related to the electrical properties of that capacitor and the charge stored on its plates.

AbstractDependent type theory gives an expressive type system facilitating succinct formalizations of mathematical concepts. In practice, it is mainly used for interactive theorem proving with intensional type theories, with PVS being a notable exception. In this paper, we present native rules for automated reasoning in a dependently-typed version (DHOL) of classical higher-order logic (HOL). DHOL has an extensional type theory with an undecidable type checking problem which contains theorem proving. We implemented the inference rules as well as an automatic type checking mode in Lash, a fork of Satallax, the leading tableaux-based prover for HOL. Our method is sound and complete with respect to provability in DHOL. Completeness is guaranteed by the incorporation of a sound and complete translation from DHOL to HOL recently proposed by Rothgang et al. While this translation can already be used as a preprocessing step to any HOL prover, to achieve better performance, our system directly works in DHOL. Moreover, experimental results show that the DHOL version of Lash can outperform all major HOL provers executed on the translation.

Aerodynamic interactions impact multirotor vehicle performance throughout its entire flight envelope and change with vehicle orientation, attitude, and forward flight speed. This paper presents efforts in incorporating these interaction effects into a reduced-order numerical quadrotor model informed by experimental flight test data. The interaction model employed system identification tools to compensate for discrepancies between actual rotor performance data and a Blade Element Theory (BET) based baseline model. Incorporation of the interaction model derived from system identification techniques improved the accuracy of model predicted rotor performance. The interaction model also provided insight into interaction effects predominantly influencing rotor performance for multiple flight conditions. The results demonstrate the utility of system identification techniques for accurate multirotor modeling capabilities.

Abstract Flexible robots with both link flexibility and joint flexibility have received increasing attention recently. In modeling the flexible robots with the assumed mode method, the model accuracy is highly dependent on the mode shapes of the link deflection. For flexible-link, flexible-joint robots, conventionally used clamped-free or pinned-free modes may cause large errors. To address this problem, this paper presents a systematic approach to dynamic modeling and mode analysis of a single-link flexible robot, which has a flexible joint and a hub at the base end and a payload at the free end. Accurate modes of the system are obtained. The following important conclusions are obtained: (1) Even a small joint flexibility can significantly affect the system frequencies; (2) The fundamental frequency is sensitive to the change in the payload and is not sensitive to the change in the hub inertia.

Phonon scattering from media with embedded spherical nanoparticles has been studied extensively over the last decade due to its application to reducing the thermal conductivity of thermoelectric materials. However, similar studies of thermal transport in fiber-embedded media have received little attention. Calculating the thermal conductivity tensor from microscopic principles requires knowledge of the scattering cross section spanning all possible incident elastic wave orientations, polarizations and wavelengths including the transition from Rayleigh to geometric scattering regimes. In this paper, we use continuum mechanics to develop an analytic treatment of elastic wave scattering for an embedded cylinder and show that a classic treatise on the subject contains important errors for oblique angles of incidence, which we correct. We also develop missing equations for the scattering cross section at oblique angles and study the sensitivity of the scattering cross section as a function of elas-todynamic contrast mechanisms. In particular, we find that for oblique angles of incidence, both elastic and density contrast are important mechanisms by which scattering can be controlled, but that their effects can offset one another, similar to the theory of reflection at flat interfaces. The solution developed captures the scattering physics for all possible incident elastic wave orientations, polarizations and wavelengths including the transition from Rayleigh to geometric scattering regimes, so long as the continuum approximation holds. The method thus enables incorporation of coherent scattering models into calculations of the thermal conductivity tensor for media with nanofibers.

In the context of a continuously developing electrical world where the systems are becoming more complex, the demands on fuel consumption and greenhouse gases reduction are also becoming stricter. Additionally, more vessels are looking into using various energy sources – batteries, variable speed generators, etc. – to improve their operation. For such systems, an energy management system (EMS) becomes a vital component. The task of an EMS is to optimize the operation based on a specific goal or goals. Mostly it is the optimization of fuel consumption, and thus also the exhaust pollution reduction, that the EMS is striving to achieve. After implementing the EMS on seagoing ferries (predictable load cycle) and a super yacht (non-predictable load cycle), RH Marine has analysed the measured data and has seen a considerable reduction of fuel consumption, of even up to 38%. With the initial goals achieved, the EMS operation can now be expanded to include more tasks. A further development of the EMS is to incorporate lifetime, wear and tear, and maintenance requirements of equipment into it. Doing so makes it possible to optimise on total cost of ownership (TCO). As a proof of concept the battery lifetime has been incorporated. Extensive simulations demonstrate that the battery lifetime can be extended, or better: a required lifetime can be reached. This can be realised by carefully observing lifetime determining quantities, like state of charge (SOC), magnitude of charging and discharging currents. This is achieved with a minimal impact on the previously obtained reduction in fuel consumption. The same is valid for other energy sources, such as diesel-generator sets (DGs). By incorporating effects of start-stop cycles on wear and tear, the use of these can be optimised regarding TCO as well. The better the load can be predicted, the better the EMS will perform. The self-learning function of the EMS will make use of all available data, both from past and future, like a planned combat mode. The paper describes how the above has been integrated into the EMS. The simulations that were performed to prove the concept as described are presented and further developments in the near future will be announced.

The META 21 Integrated Assessment Model (Dietz et al., 2021) represents a fairly comprehensive climate change simulation model incorporating a number of features: (a) a recent simple climate model; (b) down-scaling of temperature change to the country level; (c) integration of a number of bio-physical tipping points linked to rising temperatures; and (d) economic damages linked to rising sea levels and temperature using recent country-level estimates from the literature. The original implementation of META 21 was in Excel and linked to the @Risk Excel add-in for performing Monte Carlo-type analysis. This paper reflects a translation of META 21 into GAMS. One of the key purposes is to allow for ready incorporation of many of the features of META 21 into other models—notably CGE-based IAMs. To replace the features provided by the @Risk Excel add-in, this paper also introduces a software package that produces random deviates—and, similar to @Risk, uses the Latin Hypercube Sampling (LHS) approach, which is a stratified sampling technique intended to cover the entire sampling space efficiently and, in addition, orders the sample to reflect a desired correlation matrix for the sampled random variables.

Fruit deterioration is a consequence of a genetically-determined fruit ripening and senescence programs, in which developmental factors lead to a climacteric rise of ethylene production in ethylene-sensitive fruits such as tomato and banana. Breeding of tomato with extended fruit shelf life involves the incorporation of a mutation in RIN, a MADS-box transcription factor participating in developmental control signalling of ripening. The RIN mode of action is not fully understood, and it may be predicted to interact with other MADS-box genes to execute its effects. The overall goal of this study was to demonstrate conservation of ripening control functions between banana and tomato and thus, the potential to genetically extend shelf-life in banana based on tools developed in tomato. The specific objectives were: 1. To increase the collection of potential RIN-like genes from banana; 2. To verify their action as developmental regulators; 3. To elucidate MADS-box gene mode of action in ripening control; 4. To create transgenic banana plants that express low levels of endogenous Le-RIN- like, MaMADS- gene(s). We have conducted experiments in banana as well as in tomato. In tomato we have carried out the transformation of the tomato rin mutant with the MaMADS1 and MaMADS2 banana genes. We have also developed a number of domain swap constructs to functionally examine the ripening-specific aspects of the RIN gene. Our results show the RIN-C terminal region is essential for the gene to function in the ripening signalling pathway. We have further explored the tomato genome databases and recovered an additional MADS-box gene necessary for fruit ripening. This gene has been previously termed TAGL1 but has not been functionally characterized in transgenic plants. TAGL1 is induced during ripening and we have shown via RNAi repression that it is necessary for both fleshy fruit expansion and subsequent ripening. In banana we have cloned the full length of six MaMADS box genes from banana and determined their spatial and temporal expression patterns. We have created antibodies to MaMADS2 and initiated ChI assay. We have created four types of transgenic banana plants designed to reduce the levels of two of the MaMADS box genes. Our results show that the MaMADS-box genes expression in banana is dynamically changing after harvest and most of them are induced at the onset of the climacteric peak. Most likely, different MaMADS box genes are active in the pulp and peel and they are differently affected by ethylene. Only the MaMADS2 box gene expression is not affected by ethylene indicating that this gene might act upstream to the ethylene response pathway. The complementation analysis in tomato revealed that neither MaMADS1 nor MaMADS2 complement the rin mutation suggesting that they have functionally diverged sufficiently to not be able to interact in the context of the tomato ripening regulatory machinery. The developmental signalling pathways controlling ripening in banana and tomato are not identical and/or have diverged through evolution. Nevertheless, at least the genes MaMADS1 and MaMADS2 constitute part of the developmental control of ripening in banana, since transgenic banana plants with reduced levels of these genes are delayed in ripening. The detailed effect on peel and pulp, of these transgenic plants is underway. So far, these transgenic bananas can respond to exogenous ethylene, and they seem to ripen normally. The response to ethylene suggest that in banana the developmental pathway of ripening is different than that in tomato, because rin tomatoes do not ripen in response to exogenous ethylene, although they harbor the ethylene response capability This study has a major contribution both in scientific and agricultural aspects. Scientifically, it establishes the role of MaMADS box genes in a different crop-the banana. The developmental ripening pathway in banana is similar, but yet different from that of the model plant tomato and one of the major differences is related to ethylene effect on this pathway in banana. In addition, we have shown that different components of the MaMADS-box genes are employed in peel and pulp. The transgenic banana plants created can help to further study the ripening control in banana. An important and practical outcome of this project is that we have created several banana transgenic plants with fruit of extended shelf life. These bananas clearly demonstrate the potential of MaMADS gene control for extending shelf-life, enhancing fruit quality, increasing yield in export systems and for improving food security in areas where Musaspecies are staple food crops.
