Sex myoblast migration in the Caenorhabditis elegans hermaphrodite represents a simple, genetically amenable model system for studying how cell migration is regulated during development. Two separable components of sex myoblast guidance have been described: a gonad-independent mechanism sufficient for the initial anterior migration to the mid-body region, and a gonad-dependent mechanism required for precise final positioning (J. H. Thomas, M. J. Stern and H. R. Horvitz (1990) Cell 62, 1041–1052). Here, we demonstrate a role for a Ras-mediated signal transduction pathway in controlling sex myoblast migration. Loss-of-function mutations in let-60 ras, ksr-1, lin-45 raf, let-537/mek-2 or sur-1/mpk-1 cause defects in sex myoblast final positions that resemble those seen in gonad-ablated animals, while constitutively active let-60 ras(G13E) trans-genes allow fairly precise positioning to occur in the absence of the gonad. A mosaic analysis demonstrated that let-60 ras is required within the sex myoblasts to control proper positioning. Our results suggest that gonadal signals normally stimulate let-60 ras activity in the sex myoblasts, thereby making them competent to sense or respond to positional cues that determine the precise endpoint of migration. let-60 ras may have additional roles in sex myoblast guidance as well. Finally, we have also investigated genetic interactions between let-60 ras and other genes important for sex myoblast migration, including egl-15, which encodes a fibroblast growth factor receptor tyrosine kinase (D. L. DeVore, H. R. Horvitz and M. J. Stern (1995) Cell 83, 611–623). Since mutations reducing Ras pathway activity cause a different phenotype than those reducing egl-15 activity and since constitutive Ras activity only partially suppresses the migration defects of egl-15 mutants, we argue that let-60 ras and egl-15 do not act together in a single linear pathway.

Objectives: Utilization of lateral extra-articular tenodesis (LET) in conjunction with anterior cruciate ligament reconstruction (ACLR) has increased in recent years, however, the biomechanical impact of LET, when performed with contemporary techniques, on both load sharing between the ACL graft and the LET and on knee kinematics is not completely clear. The purpose of this study was to quantify the effect of LET performed with ACLR, in the presence of a compromised anterolateral tissues, on (1) forces carried by the ACL graft and the LET and (2) knee kinematics, during simulated pivot shift. Methods: manipulator equipped with a six-axis force-torque sensor. The robot applied multiplanar torques simulating two types of pivot shift (PS) subluxing the lateral compartment at 15° and 30° of knee flexion. The following loading combinations were applied: (PS1) 8 Nm of valgus and 4 Nm of internal rotation torques; (PS2) 100 N compression force, 8 Nm valgus torque, 2 Nm internal rotation torque, and 30 N anterior force. Anteroposterior (AP) translation in the lateral compartment of the knee was recorded in the following states: ACL intact, sectioned, reconstructed and, finally, after sectioning the anterolateral ligament (ALL) and kaplan fibers and performing a LET. ACLR was performed utilizing a bone-patellar tendon-bone autograft, via medial parapatellar arthrotomy. LET was performed using a modified lemaire technique with a metal staple femoral fixation at 60° of flexion in neutral rotation. Resultant forces carried by the ACL graft and LET at the peak applied load in all tested conditions were determined utilizing the principle of superposition and serial sectioning. Results: Under both simulated pivot shift types and at both flexion angles the ACL force decreased with the addition of a LET, with the least force reduction of 39% for PS2 at 15° (p=0.01) and the most force reduction of 80% for PS1 at 30° (p<0.001). While decreasing ACL force, the LET carried at least 43% of the force carried by the ACL graft when tested without LET for PS2 at 15° and 91% of the force carried by the ACL graft at most, for PS1 at 30° (Table 1). For both combinations of multiplananr torques and at both flexion angles, the anterior tibial translation in the lateral compartment decreased for the ACLR+LET knee compared to the intact knee (5.3mm and 7.6mm decrease, for PS1 15° and 30° respectively, p<0.001; 4.4mm p=0.005 and 7.6mm p<0.001, for PS2 15° and 30°, respectively). (Figure 2). Conclusion: During a simulated pivot shift, LET shields the ACL graft from loading. This effect was greatest at 30° of flexion with an 80% drop in ACL graft force. While some shielding of load from the ACL graft can be beneficial, a more significant reduction in the load of the ACL graft may potentially be detrimental to the graft remodeling, maturation and function. The optimal load sharing pattern for improved clinical outcomes is not well understood and merit further investigation. In addition, LET also decreases anterior tibial translation in the lateral compartment to less than that of the intact knee, which represents overconstraint of the lateral compartment. These findings may support the purported “protective” effect of LET on the ACL graft and its important role in stabilizing the lateral compartment in the setting of combined ACL and anterolateral structures deficiency. The influence of overconstraint of the lateral compartment with LET warrants further biomechanical and clinical evaluation. [Table: see text][Figure: see text][Figure: see text]

During Caenorhabditis elegans vulval development, an inductive signal from the anchor cell stimulates three of the six vulval precursor cells (VPCs) to adopt vulval rather than nonvulval epidermal fates. Genes necessary for this induction include the lin-3 growth factor, the let-23 receptor tyrosine kinase, and let-60 ras. lin-15 is a negative regulator of this inductive pathway. In lin-15 mutant animals, all six VPCs adopt vulval fates, even in the absence of inductive signal. Previous genetic studies suggested that lin-15 is a complex locus with two independently mutable activities, A and B. We have cloned the lin-15 locus by germline transformation and find that it encodes two nonoverlapping transcripts that are transcribed in the same direction. The downstream transcript encodes the lin-15A function; the upstream transcript encodes the lin-15B function. The predicted lin-15A and lin-15B proteins are novel and hydrophilic. We have identified a molecular null allele of lin-15 and have used it to analyze the role of lin-15 in the signaling pathway. We find that lin-15 acts upstream of let-23 and in parallel to the inductive signal.

Abstract During induction of the Caenorhabditis elegans hermaphrodite vulva by the anchor cell of the gonad, six multipotent vulval precursor cells (VPCs) have two distinct fates: three VPCs generate the vulva and the other three VPCs generate nonspecialized hypodermis. Genes that control the fates of the VPCs in response to the anchor cell signal are defined by mutations that cause all six VPCs to generate vulval tissue (Multivulva or Muv) or that cause all six VPCs to generate hypodermis (Vulvaless or Vul). Seven dominant Vul mutations were isolated as dominant suppressors of a lin-15 Muv mutation. These mutations are dominant alleles of the gene let-60, previously identified only by recessive lethal mutations. Our genetic studies of these dominant Vul recessive lethal mutations, recessive lethal mutations, intragenic revertants of the dominant Vul mutations, and the closely mapping semi-dominant multivulva lin-34 mutations suggest that: (1) loss-of-function mutations of let-60 are recessive lethal at a larval stage, but they also cause a Vul phenotype if the lethality is rescued maternally by a lin-34 gain-of-function mutation. (2) The dominant Vul alleles of let-60 are dominant negative mutations whose gene products compete with wild-type activity. (3) lin-34 semidominant Muv alleles are either gain-of-function mutations of let-60 or gain-of-function mutations of an intimately related gene that elevates let-60 activity. We propose that let-60 activity controls VPC fates. In a wild-type animal, reception by a VPC of inductive signal activates let-60, and it generates into a vulval cell type; in absence of inductive signal, let-60 activity is low and the VPC generates hypodermal cells. Our genetic interaction studies suggest that let-60 acts downstream of let-23 and lin-15 and upstream of lin-1 and lin-12 in the genetic pathway specifying the switch between vulval and nonvulval cell types.

During Caenorhabditis elegans male spicule development, four pairs of precursor cells respond to multiple positional cues and establish a pattern of fates that correlates with relative anterior-posterior cell position. One of the extracellular cues is provided by the F and U cells, which promote anterior fates. We show that the genes in the lin-3/let-23 signalling pathway required for hermaphrodite vulval induction also mediate this F/U signal. Reduction-of-function mutations in lin-3, let-23, sem-5, let-60 or lin-45 disrupt the fate of anterior cells. Likewise, activation of the pathway with ubiquitously produced signal results in posterior cells inappropriately adopting the anterior fates even in the absence of F and U. We have further used this genetic pathway to begin to understand how multiple positional cues are integrated to specify cell fate. We demonstrate that lin-15 acts in spicule development as it does in vulval induction, as a negative regulator of let-23 receptor activity. A second extracellular cue, from Y.p, also acts antagonistically to the lin-3/let-23 pathway. However, this signal is apparently integrated into the lin-3/let-23 pathway at some step after lin-45 raf and is thus functionally distinct from lin-15. We have also investigated the role of lin-12 in forming the anterior/posterior pattern of fates. A lin-12 gain-of-function defect is masked by redundant positional information from F and U.

Background: There is concern that utilization of lateral extra-articular tenodesis (LET) in conjunction with anterior cruciate ligament (ACL) reconstruction (ACLR) may disturb lateral compartment contact mechanics and contribute to joint degeneration. Hypothesis: ACLR augmented with LET will alter lateral compartment contact mechanics in response to simulated pivoting maneuvers. Study Design: Controlled laboratory study. Methods: Loads simulating a pivot shift were applied to 7 cadaveric knees (4 male; mean age, 39 ± 12 years; range, 28-54 years) using a robotic manipulator. Each knee was tested with the ACL intact, sectioned, reconstructed (via patellar tendon autograft), and, finally, after augmenting ACLR with LET (using a modified Lemaire technique) in the presence of a sectioned anterolateral ligament and Kaplan fibers. Lateral compartment contact mechanics were measured using a contact stress transducer. Outcome measures were anteroposterior location of the center of contact stress (CCS), contact force from anterior to posterior, and peak and mean contact stress. Results: On average, augmenting ACLR with LET shifted the lateral compartment CCS anteriorly compared with the intact knee and compared with ACLR in isolation by a maximum of 5.4 ± 2.3 mm ( P < .001) and 6.0 ± 2.6 mm ( P < .001), respectively. ACLR augmented with LET also increased contact force anteriorly on the lateral tibial plateau compared with the intact knee and compared with isolated ACLR by a maximum of 12 ± 6 N ( P = .001) and 17 ± 10 N ( P = .002), respectively. Compared with ACLR in isolation, ACLR augmented with LET increased peak and mean lateral compartment contact stress by 0.7 ± 0.5 MPa ( P = .005) and by 0.17 ± 0.12 ( P = .006), respectively, at 15° of flexion. Conclusion: Under simulated pivoting loads, adding LET to ACLR anteriorized the CCS on the lateral tibial plateau, thereby increasing contact force anteriorly. Compared with ACLR in isolation, ACLR augmented with LET increased peak and mean lateral compartment contact stress at 15° of flexion. Clinical Relevance: The clinical and biological effect of increased anterior loading of the lateral compartment after LET merits further investigation. The ability of LET to anteriorize contact stress on the lateral compartment may be useful in knees with passive anterior subluxation of the lateral tibia.

The Caenorhabditis elegans let-60 gene encodes a Ras protein that mediates induction of the hermaphrodite vulva. To better understand how mutations constitutively activate Ras and cause unregulated cell division, we have characterized ga89, a temperature-sensitive, gain-of-function mutation in let-60 ras. At 25°, ga89 increases let-60 activity resulting in a multivulva phenotype. At 15°, ga89 decreases let-60 activity resulting in a vulvaless phenotype in let-60(ga89)/Df animals. The ga89 mutation causes a leucine (L) to phenylalanine (F) substitution at amino acid 19, a residue conserved in all Ras proteins. We introduced the L19F change into human H-Ras protein and found that the in vitro GTPase activity of H-Ras became temperature-dependent. Genetic experiments suggest that LET-60(L19F) interacts with GAP and GNEF, since mutations that decrease GAP and GNEF activity affect the multivulva phenotype of let-60(ga89) animals. These results suggest that the L19F mutation primarily affects the intrinsic rate of GTP hydrolysis by Ras, and that this effect may be sufficient to account for the activated-Ras phenotype caused by let-60(ga89). Our results suggest that a mutation in a human ras gene analogous to ga89 might contribute to oncogenic transformation.

Timing of puberty was assessed by vaginal opening (VO) and preputial separation (PS). MaleLin28bLOF and malelet-7GOF, but not female, mice displayed alteration of pubertal timing, with later PS than controls. In contrast, both male and femaleLin28aGOF mice displayed late onset of puberty. Together, these data point toward a complex system of regulation byLin28a,Lin28b, andlet-7, in whichLin28bandlet-7can impact both puberty and growth in a sex-specific manner, raising the possibility that this pathway may contribute to differential regulation of male and female growth and puberty in humans.

Monte Carlo simulations are used to investigate the effects of acidity (pH) on the primary yields of various chemical species produced in the radiolysis of de-aerated aqueous sulfuric acid solutions over the range from neutral solution to 0.4 mol/L H2SO4. The effects of the quality of radiation, measured in terms of linear energy transfer (LET), have also been studied for LET varying from ~0.3 to 15 keV/µm at ambient temperature. Our results show that an increase in acidity (1 < pH < 4) leads to an increase in the yield [Formula: see text] of the "reducing" free radicals (hydrated electron and H• atom) and a slight increase in G·OH and [Formula: see text], while there is a slight decrease in [Formula: see text] At pH < 1, •OH radicals react with HSO4- anions to form SO4·– radicals, resulting in a steep decrease in G.OH. By contrast, in the range of pH from ~4 to 7, the calculated yield values are independent of sulfuric acid concentration. In both neutral water and 0.4 mol/L H2SO4 (pH 0.46) solutions, the primary molecular yields increase upon increasing LET to ~15 keV/µm with a concomitant decrease in those of free radicals. As an exception, GH. at first increases with LET, reaching a maximum near 6.5 keV/µm before decreasing steeply at higher LET. The results obtained are generally in good agreement with available experimental data over the whole acidity and LET ranges studied. Finally, as an application, we have simulated the radiation-induced oxidation of ferrous sulfate solutions in aerated aq. 0.4 mol/L H2SO4 (Fricke dosimeter) as a function of time up to ~50 s and addressed the effects of LET on the resulting ferric ion yield at 25 °C. The production of Fe3+ ions is highly sensitive to free-radical yields, especially H• atoms (via formation of HO2•), resulting in a marked decline of G(Fe3+) with increasing LET. The general trend of the observed variation of G(Fe3+) with radiation quality is well reproduced by our computed Fe3+ ion yield values.Key words: liquid water, acidic (H2SO4) aqueous solutions, radiolysis, free-radical and molecular yields, linear energy transfer (LET), Fricke dosimeter, Monte Carlo simulations.

Při záchranném archeologickém výzkumu slavkovské šibenice byly objeveny lidské kosterní pozůstatky, kumulované v několika shlucích v neanatomickém uspořádání. Při laboratorním zpracování byly určeny základní demografické charakteristiky souboru. Vzhledem k charakteru nálezu nebylo možno identifikovat kompletní kostry jedinců. Minimální počet jedinců (s využitím pravých stehenních kostí) byl stanoven na 15. Tělesná výška byla odhadnuta s využitím metody organické korelace dle Sjøvolda. Nejvyšší jedinec měřil 173,0 cm (±4,52 cm), nejnižší jedinec měřil 157,5 cm (±4,52 cm). Kromě dospělých jedinců byly objeveny i pozůstatky dvou nedospělých jedinců (dožitý věk byl odhadnut na 16–17 let). Všichni dospělí jedinci příslušeli do věkových kategorií adultus a maturus, nejstarší z nich se dožil kolem 50 let. Mezikosterními pozůstatky se nacházely ostatky jedné ženy. V souboru byly také nalezeny některé velmi zřídka se vyskytující anatomické variety (spina bifida anterior et posterior atlantis, os bregmaticum), doklady komplikovaného hojení traumatu i doklady nezhojených fraktur kostí dlouhého typu vzniklých perimortem. U dvou kostí byly též zaznamenány doklady zánětlivého onemocnění (pravděpodobně syfilis).

Background: Patients who undergo anterior cruciate ligament (ACL) reconstruction (ACLR) can have a persistent postoperative pivot shift. Performing lateral extra-articular tenodesis (LET) concurrently has been proposed to address this; however, LET femoral fixation may interfere with the ACLR femoral tunnel, which could damage the ACL graft or its fixation. Purpose: To evaluate the safe maximum implant or tunnel depth for a modified Lemaire LET when combined with ACLR anteromedial portal femoral tunnel drilling and to validate the safe LET drilling angles to avoid conflict with the ACLR femoral tunnel. Study Design: Descriptive laboratory study. Methods: Twelve fresh-frozen cadaveric knees were used. With each knee at 120° of flexion, an ACLR femoral tunnel in the anteromedial bundle position was created arthroscopically via the anteromedial portal using a 5-mm offset guide, a guide wire, and an 8-mm reamer, which was left in situ. A modified Lemaire LET was performed using a 1 cm-wide iliotibial band strip harvested with the distal attachment intact, to be fixed in the femur. The desired LET fixation point was identified with an external aperture 10 mm proximal and 5 mm posterior to the fibular collateral ligament’s femoral attachment, and a 2.4-mm guide wire was drilled, aiming at 0°, 10°, 20°, or 30° anteriorly in the axial plane and at 0°, 10°, or 20° proximally in the coronal plane (12 different drilling angle combinations). The relationship between the LET drilling guide wire and the ACLR femoral tunnel reamer was recorded for each combination. When a collision with the femoral tunnel was recorded, the LET wire depth was measured. Results: Collision with the ACLR femoral tunnel occurred at a mean LET wire depth of 23.6 mm (range, 15-33 mm). No correlation existed between LET wire depth and LET drilling orientation ( r = 0.066; P = .67). Drilling angle in the axial plane was significantly associated with the occurrence of tunnel conflict ( P < .001). However, no such association was detected when comparing the drilling angle in the coronal plane ( P = .267). Conclusion: Conflict of LET femoral fixation with the ACLR femoral tunnel using anteromedial portal drilling occurred at a mean depth of 23.6 mm but also at a depth as little as 15 mm, which is shorter than most implants. When longer implants or tunnels are used, the orientation should be directed at least 30° anteriorly in the axial plane to minimize the risk of tunnel conflict, bearing in mind the risk of joint violation. Clinical Relevance: This study provides important information for surgeons performing LET in combination with ACLR anteromedial portal femoral tunnel drilling regarding safe femoral implant or tunnel length and orientation.

O trabalho teve como objetivo avaliar o uso do lodo de esgoto tratado (LET) associado a substrato à base de casca de pinus (SC1) ou à base de fibra de coco (SC2), em diferentes proporções, como substrato para produção de mudas de vinhático. O experimento foi conduzido em casa de vegetação, localizada na unidade de apoio a pesquisa da Universidade Estadual do Norte Fluminense, de junho a novembro de 2016. Utilizaram-se 20, 40, 60, 80% de LET associados ao SC1 ou ao SC2, 100% de LET e o tratamento com SC1 com adubação convencional (controle). O lodo de esgoto foi higienizado com adição de cal a 15% do peso seco. Os resultados revelam que as menores proporções de LET no substrato resultaram em mudas com melhor crescimento em altura, diâmetro do coleto, área foliar, massa seca da parte aérea e do sistema radicular, comprimento e volume radicular. A nodulação foi negativamente influenciada nas mudas produzidas em proporções acima de 20% de LET. A agregação do torrão ao substrato não foi influenciada pela adição de LET, entretanto, houve aumento da curvatura do torrão nas mudas produzidas em maiores quantidades de LET. O conteúdo da parte aérea de N, P e K foi menor nas mudas produzidas em substrato com LET. Para produção de mudas de vinhático, sugere-se a proporção de 20% de lodo de esgoto associado ao SC1 ou SC2, não havendo prejuízos em relação às mudas produzidas em substrato comercial adubado, utilizado comercialmente.

The aim of this study was to investigate the expression patterns of miRNA-let 7a, 7b, and 7c in bronchoalveolar lavage fluid in infants with asthma and airway foreign bodies. Between January 2016 and February 2017, 27 infants were included and divided into observation group (infants with asthma, n = 15) and control group (infants with airway foreign bodies, n = 12). The differential expression profiles of miRNA-let 7a, 7b, and 7c were determined by reverse transcription–polymerase chain reaction in bronchoalveolar lavage fluid (BALF) from infants of the 2 groups. The BALF was collected from infants undergoing flexible bronchoscopy. MiRNA-let 7a, 7b, and 7c increased significantly in infants from observation group as compared with control group (2.72 ± 0.48 vs 1, 8.23 ± 1.64 vs 1, 3.16 ± 0.62 vs 1, respectively). The increased expression of miRNA-let 7a, 7b, and 7c were associated with the asthma of infants.

Written records on little people (Homo floresiensis) or ‘Hobbits’ are legions in either occidental or oriental history, let alone the excavation finding of a 1.06 meter (3.6 feet) 30-year-old adult female at Liang Bua cave on the remote Indonesian island of Flores in 2003. In English and Chinese literature, there are indeed no meagre narratives of little people, let alone the records found in the Chinese historical documentation and Buddhist scriptures as early as 770 BC. The main thrust of this qualitative research is to examine the little people in literature believed to be a different species or new human by comparing English and Chinese mythologies, literary creations with historical documentations and current archeological findings in light of historical research—an approach which identifies social and cultural history drawing from three main sources, namely, primary, secondary and oral tradition where accessible.

Historický výzkum umírání a smrti je již dlouhou dobu ovlivňován sociologickými a antropologickými přístupy. Přesto se teprve zhruba od 80. let minulého století historikové zabývají též otázkou sociálního užití smrti, tématem, jež hraje významnou roli v historickém bádání posledních let. To se totiž mj. soustředilo na problematiku rituálů smrti jako prezentace a rekonstrukce sociálního řádu. Epocha raného novověku, která je předmětem této studie, se ovšem vyznačovala řadou specifik, v daném kontextu především významem konfesijního faktoru jako výsledku evropské reformace. Pohřby a rituály smrti se následně často stávaly jednou z oblastí, v nichž byla vyjadřována konfesijní identita. Především v konfesijně smíšených regionech, zejména ve městech, se proto v případech vyhrocení situace jednotlivé (konfesijně definované) komunity snažily napadat rituály jiných konfesí za účelem potvrzení vlastní čistoty a správnosti vyznání. Právě v těchto momentech lze i na příkladu raněnovověké společnosti pozorovat některé ze základních antropologických principů. Členství v komunitě hrálo nanejvýš důležitou úlohu a začlenění do ní prostřednictvím tradičního rituálu a ctnostného pohřbu garantovalo též stálé místo ve společenství křesťanů (věřících). Naopak separace a vyloučení měly za následek zamezení symbolickému kontaktu mezi zemřelými a žijícími, bez něhož nebylo možno dosáhnout spásy.

ABSTRACT EGL-15 is a fibroblast growth factor receptor in the nematodeCaenorhabditis elegans. Components that mediate EGL-15 signaling have been identified via mutations that confer a Clear (Clr) phenotype, indicative of hyperactivity of this pathway, or a suppressor-of-Clr (Soc) phenotype, indicative of reduced pathway activity. We have isolated a gain-of-function allele of let-60 ras that confers a Clr phenotype and implicated bothlet-60 ras and components of a mitogen-activated protein kinase cascade in EGL-15 signaling by their Soc phenotype. Epistasis analysis indicates that the gene soc-1 functions in EGL-15 signaling by acting either upstream of or independently of LET-60 RAS. soc-1 encodes a multisubstrate adaptor protein with an amino-terminal pleckstrin homology domain that is structurally similar to the DOS protein in Drosophilaand mammalian GAB1. DOS is known to act with the cytoplasmic tyrosine phosphatase Corkscrew (CSW) in signaling pathways inDrosophila. Similarly, the C. elegans CSW ortholog PTP-2 was found to be involved in EGL-15 signaling. Structure-function analysis of SOC-1 and phenotypic analysis of single and double mutants are consistent with a model in which SOC-1 and PTP-2 act together in a pathway downstream of EGL-15 and the Src homology domain 2 (SH2)/SH3-adaptor protein SEM-5/GRB2 contributes to SOC-1-independent activities of EGL-15.

Nevertheless, most commentators insist upon interpreting καθευδώ in 5.10, not with the meaning it has in 5.6–7, but with the same meaning as καιμαομαι in 4.13–15 – those who have physically died. This also requires that ‘awake’ (γρηγορωμεν) in 5.10 be interpreted as physically ‘alive’ rather than ethically or spiritually ‘vigilant’, the sense it has in 5.6.

In the article we present the research results carried out in 2015. The research was focused on recognition of adolescents’ opinions and attitudes (15–18 years of age) towards physical activities and sport from different points of view. A subtask dealt with comparing the research of compatible items resulting of the questionnaire PASA conducted in 2000 (Slepička et al., 2001). The work hypothesis assumed certain differences in the real estimates of own sporting and physical activities including the selection of practised sports. Data were analysed by the questionnaire PASA (physical activities and sport in adolescents), which was presented among adolescents at secondary schools (Grammar schools). In general, 1067 respondents were inquired, 468 of them boys and 599 girls. Regarding the results it is clear that in 2000 more than 50 % of boys aged 15 to 18 years of age did physically nothing at all or were just hardly keeping current physical condition. In 2015 only 40 %, which means improvement of about 10 % in favor of physical activities and sports among boys. As for the girls it was over 61 % (2000) and 45 % (2015), and that was a surprising improvement in favor of physical activities and sports of 16,1 %. Within the fifteen-year period, the selection of sports among boys, was almost identical. They preferred football and cycling. in the third place it was sightly different – in 2000, they preferred martial arts while in the year 2015 jogging. The girls differed more in the year 2000, the ranking was – aerobics, volleyball and cycling, but in 2015 – beach volleyball, work out (home exercise) and orienteering.

Analizirali smo letne in sezonske količine ter trende za 12 meteoroloških postaj v panonskem in obpanonskem delu Srbije za obdobje 1949–2010. Rezultati kažejo trend zmanjševanja padavin pozimi in spomladi ter trend porasta poleti in jeseni. Časovne serije letnih količin padavin kažejo porast padavin na večini preučevanih postaj in so posledica bolj namočenih let ter ekstremnih padavinskih dogodkov v zadnjih 15 letih. V preučevanem obdobju je bilo tudi 10 let z močno in ekstremno sušo s tendenco pojavljanja v več zaporednih letih. Dobro sliko o razporeditvi padavin je dala tudi metoda razvrščanja v skupine (cluster analiza).

143 Background: Activation of the PI3K/AKT/mTOR and cyclin D–CDK4/6–INK4–Rb pathways has been implicated in endocrine therapy resistance in pts with advanced HR+ breast cancer (BC). BYL719 (BYL), an α-isoform selective PI3K inhibitor and LEE011 (LEE), a CDK4/6 inhibitor have demonstrated clinical activity in pts with advanced HR+ BC and advanced solid tumors, respectively. In preclinical HR+ BC models, the combination of LEE, BYL, and letrozole (LET) had enhanced antitumor activity vs either agent alone. Here we report results from Arm (A)1 (LEE + LET) and A2 (BYL + LET) of the Ph Ib part of a Ph Ib/II, 3-arm study of LEE, BYL, and LET in pts with advanced ER+ BC (CLEE011X2107/NCT01872260). Methods: Postmenopausal women with advanced ER+, HER2− BC receive daily oral doses of LEE (3-wks on/1-wk off; A1) or BYL (continuous; A2), plus fixed, daily LET (2.5 mg, continuous), as part of a 28-day cycle. Primary objective: determine the MTD and/or RP2D of A1 and A2. Dose escalation is guided by a Bayesian Logistic Regression Model using the escalation with overdose control principle and real-time PK. Safety and preliminary efficacy are also assessed. Results: As of March 28, 2014, 10 pts have been treated with 600 mg LEE + LET (A1), and 7 pts with 300 mg BYL + LET (A2). One DLT was observed (A1: Grade [G]4 neutropenia; data cut-off: May 15). Common (all grade >30%) study drug-related AEs (all grade/G3–4) were: A1: neutropenia (90%/50%) and nausea (40%/0%); A2: hyperglycemia (57%/14%), decreased appetite, diarrhea, and nausea (43%/0% each). PK for LEE and BYL on Days 1 and 21, and LET on Day 1, are comparable to historic single-agent data. PK of LET at steady state is being evaluated. In A1, 6 pts had known responses: 1 PR, 2 SD, 1 NCRNPD, and 2 PD. In A2 5 pts had known responses: 2 SD, and 3 NCRNPD. Conclusions: LET plus LEE or BYL had an acceptable safety profile and preliminary signs of clinical activity have been observed. Upon determination of the MTD/RP2D in A1 and A2, enrollment into A3 (LEE + BYL + LET) will commence. The randomized Ph II part of the trial will compare LET + LEE or BYL with LET + LEE + BYL. MONALEESA-2 (CLEE011A2301/NCT01958021), a Ph III, randomized, double-blind, placebo-controlled study of LEE + LET in untreated advanced HR+ BC is currently recruiting pts. AbstractExperiments have been performed on the disturbance of a high-Reynolds-number turbulent boundary layer by three forward steps with sizes close to 3.8, 15 and 60 % of the boundary layer thickness. Particular attention is focused on the impact of the steps on the fluctuating surface pressure field. Measurements were made from 5 boundary layer thicknesses upstream to 22 boundary layer thicknesses downstream of the step, a distance equivalent to over 600 step heights for the smallest step size. Flow speeds of 30 and $\def \xmlpi #1{}\def \mathsfbi #1{\boldsymbol {\mathsf {#1}}}\let \le =\leqslant \let \leq =\leqslant \let \ge =\geqslant \let \geq =\geqslant \def \Pr {\mathit {Pr}}\def \Fr {\mathit {Fr}}\def \Rey {\mathit {Re}}60\ \mathrm{m}\ {\mathrm{s}}^{-1}$ were studied, corresponding to boundary layer momentum thickness Reynolds numbers of 15 500 and 26 600 and step size Reynolds numbers from 6640 to 213 000. The steps produce a disturbance to the boundary layer pressure spectrum that scales on step size and decays remarkably slowly with distance downstream. When normalized on step height and free-stream velocity, the disturbance is self-similar and appears to develop almost independently of the enveloping boundary layer. The disturbance is still clearly visible at 150 step heights downstream of the mid-size step. Pressure correlations show the disturbance to be characterized by organized quasiperiodic motions that become visible well downstream of reattachment. The coherence and scale of these motions, as seen in the wall pressure correlations, scales on the step height and thus their visibility relative to the boundary layer grows rapidly as the step size is increased.
