Der volle Inhalt der QuelleRole of the mitotic kinesins Eg5 and MKLP-2 in physiologic and pathologic angiogenesis.Angiogenesis is a complex biological phenomenon which corresponds to the formation of newblood vessels from pre-existing vessels. This process is regulated by a plethora of differentmolecules with vascular endothelial growth factor (VEGF) being one of the most important ones.VEGF inhibitors are currently used in the treatment of numerous solid cancers. Even though theefficacy of such treatment is prouven by numerous studies, resistance to anti-angiogenic therapy isa common feature. To identify new therapeutic targets downstream of VEGF, we modelized itsaction on the chick chorioallantoic membrane (CAM). VEGF-treated CAMs develop a densevascular network 24h after application. We used chick microarrays to monitor global geneexpression changes in VEGF-induced CAMs. Beside a consistent number of genes alreadydescribed to be regulated by VEGF, numerous unknown genes have been identified. We havefocused our work on the characterization of Eg5/KIF11 and MKLP-2/KIF20A, members of thekinesin family, both strongly upregulated by VEGF.We demonstrated that Eg5 and MKLP-2 are strongly expressed by blood vessels in normal andcancer tissue sections. KIF20A is involved in the proliferation and migration of endothelial cellsin vitro. We showed that chemical inhibitors specific for KIF11/Eg5 (dimethylenastron andispinesib mesylate) affect key steps in the formation of blood vessels (proliferation, adhesion andmigration of endothelial cells) and proliferation of tumor cells (glioma and renal cancer).Furthermore, in experimental glioblastoma and renal cell carcinoma models (CAM and orthotopicimplantation in mice), anti-Eg5 treatment strongly reduces tumor angiogenesis and growth. Der volle Inhalt der QuellePurpose: Mitogen-activated protein kinase (MAPK) phosphatase-1 (MKP-1) regulates by dephosphorylation the activity of the major intracellular signaling pathways mediated by mitogen-activated protein kinase [extracellular signalregulated kinase (ERK), c-Jun NH2-terminal kinase (JNK), and p38]. MKP-1 mediates breast cancer chemoresistance and is repressible by doxorubicin in breast cancer cells. This work is aimed to characterize doxorubicin effects on MKP- 1 and phospho-MAPKs in human breast cancers and to study the clinical relevance of MKP-1 expression in this tumor. Experimental Design: Doxorubicin effects on MKP-1, phospho-ERK1/2 (p-ERK1/2), phospho-JNK (p-JNK), and phospho-p38 were assayed in a broad panel of human breast cancer cells by Western blotting and in human breast cancer were assayed ex vivo by immunohistochemistry. MKP-1 expression was also assayed in a spectrum of normal to malignant breast lesions and in a large series of breast cancer patients with clinical follow-up. Results: MKP-1 was expressed at low levels in normal breast and in usual ductal hyperplasia and at high levels in in situ carcinoma. MKP-1 was overexpressed in approximately 50% of infiltrating breast carcinomas. Similar to what was observed in breast cancer cell lines, ex vivo exposure of breast tumors to doxorubicin downregulated MKP-1, and upregulated activation of ERK1/2 and JNK, in most of studied cases. However, in a proportion of tumors overexpressing MKP-1, doxorubicin did not significantly affect MKP-1 or activation levels of MAPKs. With regard to patient outcome, MKP-1 overexpression was a significant adverse prognostic factor for relapse both by univariate and multivariate analysis. Conclusions: MKP-1 is overexpressed during the malignant transformation of the breast and independently predicts poor prognosis. Furthermore, MKP-1 is repressed by doxorubicin in human breast cancer.

Der volle Inhalt der QuelleENGLISH ABSTRACT: Introduction: Cancer is an emerging health problem in South Africa, with breast cancer being one of the leading cancers affecting women globally. Therefore, there is a need to find novel targets to improve the therapeutic options for these patients. A recently proposed target is the mitogen-activated protein kinase phosphatase-1 (MKP-1). Studies have suggested that mitogen-activated protein kinase phosphatases are involved in the development of cancer and play an important role in the response of cancer cells to chemotherapy. Additionally, numerous studies have indicated that there is increased expression of MKP-1 in breast cancers where its over-expression is proposed to be a significant mediator in chemo-resistance. We propose that inhibition of MKP-1 will increase the cytotoxic effect of doxorubicin in breast cancer cells, thus making the cells more responsive to treatment leading to increased cell death through autophagy and apoptosis. Methods: In MDA-MB231 cells, MKP-1 was inhibited using sanguinarine or MKP-1 siRNA and this was compared to a known inducer of MKP-1, dexamethasone. MDA-MB231 cells were treated with doxorubicin alone or in combination with MKP-1 inhibitors or an inducer. Following treatment, cell death was determined by trypan blue and a caspase glo assay as well as with western blotting. Autophagy was determined by western blotting and flow cytometry. LC3 and p62 were used as markers of autophagy and caspase 3 and PARP as apoptosis markers. Likewise, the level of MKP-1 expression under conditions of MKP-1 induction, inhibition or silencing was evaluated by means of western blotting. C57BL6 tumour bearing mice was used to analyse apoptosis and autophagy in vivo under conditions of MKP-1 inhibition, using sanguinarine, together with doxorubicin treatment. Western blotting was used to determine levels of caspase 3, LC3, p62 and MKP-1 expression. Results and discussion: A concentration and time curve indicated that 5 μM doxorubicin reduced cell viability in the MDA-MB231 cells significantly after 24 hours of treatment. MKP-1 expression was significantly reduced with sanguinarine and MKP-1 siRNA. Furthermore, our results indicate a significant increase in apoptosis in MDA-MB231 cells when treated with doxorubicin, under conditions of MKP-1 inhibition or MKP-1 silencing. Also, an increase in autophagic activity was observed following treatment with doxorubicin in combination with sanguinarine. Whole excised tumours of C57BL6 mice also showed an increase in apoptosis and autophagy following treatment with sanguinarine in combination with doxorubicin. This indicates that the inhibition of MKP-1 with sanguinarine sensitized the MDA-MB231 cells and E0771 cell tumours to doxorubicin-induced-apoptosis through a mechanism involving autophagy. Conclusion: This is an encouraging finding that could hopefully be used in future studies to overcome doxorubicin-resistance in breast cancer cells overexpressing MKP-1. Targeting MKP-1 can have potential therapeutic benefits for breast cancer patients by making chemotherapy more effective. Sanguinarine thus has potential to be developed as a clinically relevant inhibitor of MKP-1 which could provide a novel avenue for therapeutic intervention in combination with chemotherapy in breast cancer patients.
AFRIKAANSE OPSOMMING: Inleiding: Kanker is 'n vinnig groeiende gesondheidsprobleem in Suid-Afrika, met borskanker as een van die vernaamste kankers wat vroue wêreldwyd raak. Daar is dus 'n behoefte aan nuwe terapeutiese opsies vir hierdie pasiënte en mitogeen-geaktiveerde proteïenkinase fosfatase-1 (MKP-1) is onlangs voorgestel as ‘n moontlike teiken. Verskeie studies toon dat mitogeen-geaktiveerde proteïenkinase fosfatases betrokke is by die ontwikkeling van kanker en ook belangrike rolspelers is in die reaksie van kanker op chemoterapie. Daarbenewens toon talle studies dat daar verhoogde MKP-1 uitdrukking in borskanker is, asook dat dit ‘n belangrike bemiddelaar is vir die weerstand wat borskanker teen chemoterapie bied. Ons het dus voorgestel dat die inhibisie van MKP-1 die sitotoksiese effek van doxorubicin op borskanker selle sal verhoog; sodoende sal die kanker selle beter reageer op behandeling en dit sal dus lei tot verhoogde seldood deur autofagie en apoptose. Metodes: MKP-1 is geïnhibeer met behulp van sanguinarine of MKP-1 siRNA in MDA-MB231 selle en dit is vergelyk met 'n bekende MKP-1 induseerder, dexamethasone. MDA-MB231 selle is met doxorubicin alleen behandel of in kombinasie met MKP-1 inhibeerders of ‘n induseerder. Seldood is bepaal deur middel van ‘n trypan blou en kaspase toetsingsmetode, asook met die westelike kladtegniek. Autofagie is bepaal deur westelike kladtegniek en vloeisitometrie. LC3 en p62 is gebruik as merkers van autofagie en kaspase 3 en PARP is as apoptose merkers gebruik. MKP-1 uitdrukking is geëvalueer deur middel van westelike kladtegniek. C57BL6 muise met kankeragtige gewasse is gebruik om apoptose en autofagie in vivo te ondersoek. MKP-1 is geïnhibeer met sanguinarine en die muise is behandel met ‘n kombinasie van sanguinarine en doxorubicin. Kaspase 3, LC3, p62 en MKP-1 uitdrukking is bepaal deur middel van die westelike kladtegniek. Resultate en bespreking: ‘n Konsentrasie en tyd kurwe het aangedui dat 5 μM doxorubicin die MDA-MB231 selle se lewensvatbaarheid aansienlik verminder het na 24 uur. MKP-1 uitdrukking is ook aansienlik verminder met sanguinarine en MKP-1 siRNA. Verder dui die resultate op 'n beduidende toename in apoptose in MDA-MB231 selle na behandeling met doxorubicin onder toestande van MKP-1 inhibisie. 'n Toename in autofagiese aktiwiteit is waargeneem na behandeling met doxorubicin en sanguinarine. Die kankeragtige gewasse van die C57BL6 muise toon ook 'n toename in apoptose en autofagie na behandeling met sanguinarine en doxorubicin. Hierdie resultate dui daarop dat die inhibisie van MKP-1 met sanguinarine die MDA-MB231 selle en E0771 sel gewasse gesensitiseer het tot doxorubicin-geïnduseerde apoptose deur middel van ‘n meganisme wat autofagie insluit. Gevolgtrekking: Hierdie bevinding kan hopelik in toekomstige studies gebruik word om doxorubicin weerstand te oorkom in borskanker selle waar MKP-1 verhoog is. Deur MKP-1 te teiken, kan dit lei tot potensiële terapeutiese voordele vir borskanker pasiënte en sodoende kan dit chemoterapie meer effektief maak. Sanguinarine het dus die potensiaal om ontwikkel te word as ‘n klinies relevante inhibeerder van MKP-1 wat sodoende kan dien as terapeutiese intervensie in kombinasie met chemoterapie vir borskanker pasiënte.

Resumo: A proteína MKP-3 (MAPK phosphatase-3) possui capacidade de associar-se fisicamente e desfosforilar a FoxO1 no fígado, resultando em aumento da gliconeogênese e hiperglicemia na condição de obesidade e diabetes. Por outro lado, o exercício físico é uma das estratégias não farmacológicas mais utilizadas para a melhora do perfil glicêmico em pacientes diabéticos, no entanto, suas ações no tecido hepático são pouco conhecidas. Assim, o objetivo deste trabalho foi investigar os efeitos do treinamento físico sobre a expressão proteica da MKP-3, bem como sua interação com a FoxO1 em fígado de animais obesos. Ademais, tem como objetivo avaliar o mecanismo pelo qual o exercício físico é capaz de inibir a proteína MKP-3 em tecido hepático. Foram utilizados camundongos Swiss machos que receberam por 16 semanas uma dieta padrão ou rica em gordura. Os animais foram distribuídos aleatoriamente em seis grupos: controle (C), obeso sedentário (OB), obeso exercitado (EXE), obeso tratado com oligonucleotídeo antisense MKP-3 (OB-ASO), obeso tratado com sense MKP-3 (OB-sense) e obeso submetido ao treinamento físico e ao tratamento com antisense MKP-3 concomitantemente (OB-EXE-ASO). O protocolo de treinamento físico consistiu de natação com sessões de 60 minutos, 5 vezes por semana, com carga equivalente a 5% da massa corporal total do animal. O tratamento com antisense foi realizado através de injeções duas vezes ao dia, com um volume total de 2.0 ?l por dose (4.0 nmol/?l) por 5 dias. Ao final do experimento foram realizados os testes de tolerância ao piruvato (TTP) e teste de tolerância à insulina (TTI). Além disso, foram avaliados parâmetros fisiológicos (ingestão calórica, massa corporal, glicemia e insulinemia de jejum) e a análise molecular das proteínas (MKP-3, FoxO1, PGC1?, HNF-4?, PEPCK, G6Pase, ERK e CK2?) através das técnicas de imunoblot e imunoprecipitação. Utilizou-se a técnica de imunohistoquímica para análise da co-localização da MKP-3 no fígado dos animais. Os resultados obtidos demonstraram que o treinamento físico diminuiu expressão proteica de MKP-3 e a associação FoxO1/MKP-3 no fígado dos animais obesos. Além disso, camundongos obesos treinados apresentaram maiores níveis de FoxO1 fosforilada e diminuição da expressão das proteínas PGC-1? e HNF-4???no fígado se comparado aos animais obesos não submetidos ao treinamento físico. Verificam-se também menores níveis das enzimas gliconeogênicas PEPCK e G6Pase nos animais obesos treinados se comparado aos seus pares sedentários. Estes resultados a nível molecular foram acompanhados por alterações fisiológicas incluindo aumento da sensibilidade à insulina no fígado, menor produção hepática de glicose e redução da hiperglicemia nos camundongos obesos, independentemente da redução da massa corporal total. Ademais, não foram encontrados efeitos aditivos sobre os parâmetros fisiológicos e moleculares com o tratamento de oligonucleotídeo antisense MKP-3 e treinamento físico realizado concomitantemente nos animais obesos. Por fim, os efeitos supressivos do treinamento físico sobre a proteína MKP-3 parecem estar relacionados, no mínimo em parte, a diminuição na fosforilação das ERKs no fígado de camundongos obesos
Abstract: The protein MKP-3 (MAPK phosphatase-3) has the ability to physically associate with and dephosphorylate the FoxO1 in the liver, resulting in increased gluconeogenesis and hyperglycemia in the condition of obesity and diabetes. On the other hand, exercise is one of the most used non-pharmacological strategies for improving glycemic control in diabetic patients; however, their actions in liver tissue are poorly explored. The objective of this study was to investigate the effects of exercise on the expression of MKP-3 and its interaction with FoxO1 in the liver of obese animals. Furthermore, aimed at evaluating the mechanism by which physical exercise is able to inhibit protein MKP-3 in the liver. We used male Swiss mice that received a standard diet or high fat for 16 weeks. The animals were randomly divided into six groups: control (C), sedentary obese (OB), obese exercised (EXE), obese treated with MKP-3 antisense oligonucleotide (ASO-OB), obese treated with sense MKP-3 (OB- sense) and obese subjected to physical training and treatment with antisense MKP-3 concomitantly (OB-EXE-ASO). The protocol consisted of swimming exercise training with sessions of 60 minutes, five times per week with a load equivalent to 5% of the total body mass of the animal. Treatment with antisense was performed by injection twice daily, with a total volume of 2.0 ul per dose (4.0 nmol / microl) for 5 days. At the end of the experiment were performed to pyruvate tolerance tests (PTT) and insulin tolerance test (ITT). Furthermore, physiological parameters were evaluated (caloric intake, body weight, blood glucose and fasting insulin) and molecular analysis of proteins (MKP-3, FoxO1, PGC1a, HNF-4a PEPECK, G6Pase, ERK and CK2?) by immunoblotting and immunoprecipitation techniques. The immunohistochemistry technique was utilized to evaluate the co-localization of MKP-3 in the liver of the animals. The results showed that physical training decreased the protein expression of MKP-3 and association FoxO1/MKP-3. Further, trained obese mice had higher levels of phosphorylated FoxO1 and decreased PGC-1? and HNF-4??? There was also a decrease in the protein levels of PEPCK and G6Pase in the liver of mice exercised compared to sedentary obese mice. These results at molecular level were accompanied by physiological changes including increased insulin sensitivity in the liver, reduced hepatic glucose production and reduce hyperglycemia in obese mice, regardless of reducing total body mass. In addition, there were no additive effects of treatment with antisense oligonucleotide MKP-3 and physical training performed concurrently in obese animals. Finally, the suppressive effects of physical training on MKP-3 protein appear to be related at least in part, the decrease in phosphorylation of ERKs in the livers of obese mice

Der volle Inhalt der QuelleThe Swedish national agency of education has written reports about what the first-teacher do in general. The purpose with this study is to exemplify what the first-teacher in mathematics do and what it possibly can result in, both for colleagues and pupils. By making qualitative interviews with seven first-teachers in mathematics the study illustrates how they work and what outcome they experience. The results show that the first-teacher assignments differs, for example, one teacher does nothing above her regular teaching in her assignment more than having a title. Another first-teacher actively work with screening to assess pupils’ knowledge in mathematics, enables teaching of practical mathematics and is responsible for different projects in developing competence for colleagues. The first-teachers work is done under different conditions, such as time assigned to the mission, number of pupils at the school they work at and other on-going projects. Something that is crucial for the first-teacher efforts is, in part, how committed principal and school leadersare, and what demands they make, partly how passionate and skilful the first-teacher is herself.
