The Melanocyte Keratinocyte Transplantation Procedure (MKTP) is an effective surgical replacement of lost melanocytes in recalcitrant vitiligo and pigmentary skin disorders. However, it is only effective in stable vitiligo lesions because active autoimmunity destroys the newly transplanted melanocytes. Despite careful selection of candidates based on the reported clinical stability, the success of the procedure is still unpredictable. MKTP candidates with non-segmental, segmental, and mixed vitiligo, as well as hypopigmented scars and Piebaldism patients were enrolled to our studies. Our aim was first, to investigate the possible immunological mechanisms responsible for the unpredictable post- transplantation outcomes, including T cell subsets and inflammatory chemokines, by correlating these biomarkers with clinical phenotypes, duration of stability, and surgical outcomes. We used suction blister biopsy, a minimally invasive technique that we developed to sample human skin. Moreover, we quantified transplanted melanocytes in the suspension using flow cytometry. Following MKTP, we corelated these biomarkers to the repigmentation score. We found that CD8+ T cells remain in some clinically stable vitiligo lesions, correlate negatively with the post-surgical score of repigmentation, and inversely impact the durability of the responses. Interestingly, the number of transplanted melanocytes in the suspension and the duration of stability do not have prognostic roles. Based on our findings and in a second group of patients, we suppressed the activity of T cells to enhance the outcomes of MKTP. We used Ruxolitinib, JAK1/2 inhibitor, in a triple blinded randomized controlled within subject study, in comparison with Tacrolimus (a calcineurin inhibitor and the standard of care treatment in vitiligo) as well as placebo control. We found lower T cell infiltrate, lower chemokines, and better skin repigmentation in lesions treated with MKTP plus Ruxolitinib or Tacrolimus than in lesions treated with MKTP plus placebo. Lastly, we compared two different types of laser in preparation of the recipient skin for MKTP - ablative versus fractional Er:YAG laser. We found that the ablative laser is combined with minimal CD8+ T cell epidermal infiltrate and superior repigmentation score in comparison to more infiltrate and lower repigmentation score with the fractional laser. Taken together, these results from our studies provide novel insight to predict the optimal surgical candidates and will improve surgical outcomes. It advances the treatment of vitiligo by uncovering the impact of autoimmunity on the success of repigmentation and discovering new approaches to optimize the surgical treatment options in patients with vitiligo and pigmentary skin disorders.

La principale caracteristique de la phosphorylation des proteines cellulaires est sa reversibilite. Dans la cellule, il existe une balance entre la phosphorylation (assuree par des kinases) et la dephosphorylation (assuree par des phosphatases) des proteines. Une modification de cette balance est a l'origine d'une grande partie des mecanismes permettant la transmission des signaux extracellulaires vers le noyau. Les phosphatases sont generalement divisees en trois grandes familles : les serine/threonine phosphatases, les tyrosine phosphatases et les phosphatases a double specificite. Map kinase phosphatase-1 (mkp-1), mkp-2 et mkp-3 sont tous trois membres de la nouvelle famille des phosphatases a double specificite (dsp) qui arborent la sequence consensus : (i/v)hcxagxxr(s/t)g. Ces mkp ont la propriete de dephosphoryler in vitro comme in vivo un substrat archetype, la p42/p44mapk. Afin de determiner l'impact de ces phosphatases sur la regulation des mapk ainsi que leur role dans la proliferation, nous avons entrepris leur etude dans le systeme bien defini des fibroblastes d'hamster chinois, les ccl39. Dans un premier temps, nous avons demontre (i) qu'une forme ectopique de mkp-1 (1-) se localise dans le noyau de cellules quiescentes ou stimulees. (ii) que cette forme surexprimee est capable de bloquer des activites nucleaires de la p42/p44mapk, la transcription de genes de la transition g1/s et enfin la replication de l'adn des cellules transfectees. Dans un second temps, nous avons entrepris la production d'anticorps polyclonaux diriges contre mkp-1 et mkp-2. This diploma thesis deals with an application of the finite element method on elastohydrodynamic (EHD) lubrication simulations. Commercially available software COMSOL is used for the computation, while two different modules for modeling EHD lubrication are described in a detail. Firstly, a new approach using the module Thin-Film Flow is developed, considering and describing some limitations of this approach. This is the very first published work dealing complex with EHD simulation in Thin-Film Flow module. In the second part of the thesis, there was created a model of line contact using the module for the introduction of partial differential equations (PDE). The model is partially verified with available works for different values of the input parameters. Subsequently, the velocity effect of the contact surfaces on the pressure and the lubricant thickness in contact is analyzed. Finally, the last part is examines the influence of the values of some parameters on the final value of the contact pressure and the lubricant thickness, as well as on numerical stability of the entire model.

This diploma thesis deals with the storage tanks for petroleum products. The introductory part is devoted to the distribution of the storage tanks, associated standards and maintenance of tanks. Another part goes about the analysis of real case of a tank collapse. There was created a 3D model of the collapsed tank according to the measured values in the tank. It was created a tank shell model by drawing documentation and analysis of the results of the accident investigation using the finite element method (FEM) by program ANSYS. Calculation was performed on the buckling resistance of the structure according to ČSN EN 1993-1-6 and its comparison with the results of FEM analysis in ANSYS. The aim of the study was to find the critical condition of the structure and determination of the critical internal underpressure and overpressure. At the end there were determined measures for safe operation and settings of the valve members.

This master thesis is focused on computational modelling of critical external as well as internal pressure acting of the repaired tank. During cleaning of pipelines, the gas mixture entered into this storage tank causing loss of integrity in the connection between the roof and the shell. The first part of the thesis is focused on check calculation of the shell in exceptional conditions. The next part describes the repair of the tank including check of the pillar according to ČSN EN 1993 1 1 and ČSN EN 1993-1-6. The main part of the thesis is focused on determination of the buckling resistances of the tank in original as well as repaired state using ANSYS Mechanical software. The results are comparable with original tank, whose buckling resistance was not decreased due to the repair.

K modelovaniu sme si zvolili 3 fázový synchronný stroj s permanentnými magnetmi na povrchu rotora z firmy VUES. Najprv vypočítame parametre stroja a potom spravíme statický model pomocou programu FEMM a Matlab a budeme hladať možné riešenia na 2D dynamický model. Pomocou programu Ansys Workbench spravíme 3D model tohoto stroja budeme a porovnávať výsledky simulácie.

This thesis explores the utilization of FEM software ANSYS in the analysis of the real blade wheel of a radial compressor used in small turbine aviation motors. This apparatus is made by První brněnská strojírna a.s., and its detached site in Velká Bíteš where it has the aviation engineering division. The aim of this thesis is a comprehensive blade wheel analysis composed of several steps. These steps include static and elasto-plastic strength analysis, which describes stress distribution and its assessment from the perspective of boundary conditions of elasticity during the use of the Hooke and the non-linear model of the material. Another step is dynamic analysis; in this case modal analysis, which describes the frequencies and its waveforms. The last part of the thesis contains thermal analysis while considering stationary and non-stationary thermal field. The analysis is then transformed into thermal-stress coupled analysis, which describes stress distribution resulting from thermal expansion of the material. The thesis follows the process of calculation simulations that are used during the design of a real impeller, thus before the creation of a prototype that undergoes real experiments.

Abnormalities in Mitogen Activated Protein Kinase (MAPK) signalling may affect the cells essential processes and influence the cell in acquiring traits to favour tumorigenic transformation and the progression of cancer. Deregulation of the MAPK pathways and uncontrolled crosstalk occurring in cancers may be caused by deregulation by MAP-kinase phosphatases (MKPs) that negatively regulates MAPKs by dephosphorylation. In this study, we were interested in the role of MKP-2 in MAPK-signalling pathways. MKP-2 is known to specifically dephosphorylate the MAP kinases Erk1/2, p38 and JNK. In this study, I was to elucidate key events leading to MKP-2 expression and the role of MKP-2 in regulating and balancing MAPK signaling. Also, I was to analyse the possible involvement of p53 in the deregulation of the MAPK pathways and its correlation with MKP-2 expression. In this report, I suggest a model where the Erk1/2 pathway in conjugation with p53 promote MKP-2 expression. I have also discovered a crosstalk between two different MAPK pathways, i.e between Erk1/2 and Erk5.

This diploma thesis deals with computation modeling of spur gearing using FEM. The methodology of checking calculation was suggested for gears which are not possible to check only by using standard ČSN 01 4686. The analysis of influence of vicinity of stress concentration on stress distribution in tooth root was performed. It is possibility to use the gears with thinner rim than the standard ČSN 01 4686 recommends. The main part of this work is devoted to fatigue crack propagation study in gears with thin rim. Aiding T-stress the influence of constraint and stress multiaxiality on crack propagation was taking into account. The predicted fatigue crack propagation paths were compared with certain experimental data. It is demonstrated that the considering of influence of constraint improves estimation of consequent crack propagation direction and in some cases the influence of constraint can significantly change the estimated crack propagation path.

Residual state of stress in structural materials affect positively or negatively behaviour of component parts. The goal of this scope is not to deal with possible process of creating residual stresses neither about elimination of residual stress, but is focused how to determine magnitude of residual stress by hole-drilling method. We need to know magnitude and direction (angular orientation) of principal stresses to determine how residual state of stress affects behaviour of specimen. The most widely used modern technique for measuring residual stresses is hole-drilling strain-gage method. Hole-drilling method is in scope of this paper and is restricted only for measuring uniform residual stresses of steel specimens with finite dimensions. Structural, linear, elastic and isotropic material model is used with material properties =0,3 and E=2,1[10]^5 MPa. For correct application of this method we need to determine calibration coefficients “a“ and “b“ first. These coefficients are used to determine magnitude and direction of residual stresses in specific depth and diameter of drilled hole for materials with finite dimensions. Geometry and shape of model is simply represented by block with planar faces. Note that numerical determination of calibration coefficients is useful only for one type of strain gauge rosette RY 61 S. Main goals of this thesis are motivation and request to clearly report effectiveness, accuracy and applicability of calibration coefficients in relation to thickness of specimen, dimensions of drilled hole, condition of “through” or “blind” hole and number of drilled increments. High quality and accuracy of created numerical model is necessary too. Numerical simulation of residual stresses by MKP needs to be done to obtain requested results. All results are presented by 3D, 2D graphs and tables and compared with analytical results or results from other authors. Although is this publication focused on numerical modeling using FEM, hole-drilling method has many significant restrictions. The most substantial of them is influence of eccentricity of drilled hole, creation of stress concentration near drilled area and subsequent plastification, influence of geometrical inaccuracy of hole, etc. All these aspects have significant influence of determining calibration coefficients and can not be included into numerical simulation. These problems are closely discussed in background research. All obtained results should be helpful for practical use of calculated calibration coefficients to determine uniform residual stresses of specimens with various thickness and drilled hole. All these results are also applicable only for one type of strain gauge rosette, which is RY 61 S.

Rôle des kinésines mitotiques Eg5 et MKLP-2 dans l’angiogenèse physiologique etpathologique.L’angiogenèse est un phénomène biologique complexe qui correspond à la formation de nouveauxvaisseaux à partir de vaisseaux préexistants. Ce processus essentiel est régulé par des nombreuxfacteurs, dont le plus puissant est le facteur de croissance de l’endothélium vasculaire (VEGF).Des inhibiteurs du VEGF sont actuellement utilisés dans le traitement de nombreux cancerssolides. Leur efficacité est constatée dans plusieurs études mais des résistances contre cesmolécules sont fréquemment observées. Afin d’identifier de nouvelles cibles thérapeutiques dansla voie de signalisation de VEGF, nous avons utilisé le modèle de la membrane chorioallantoïdienne(CAM) de l’embryon de poulet. Les CAM traitées au VEGF pendant 24hdéveloppent de nombreux vaisseaux. Ces tissus ont été isolés pour effectuer une analysetranscriptomique. En dehors des gènes endothéliaux déjà connus pour être régulés par le VEGF,de nouveaux gènes ont été identifiés. Nous avons focalisé notre recherche sur des gènes codantpour les kinésines mitotiques KIF11/Eg5 et KIF20A/MKLP-2 qui ont été fortement induites.Nous avons démontré qu’Eg5 et MKLP-2 sont fortement exprimées au niveau de l’endothéliumdans des tissus sains et dans des cancers solides. Des inhibiteurs chimiques spécifiques d’Eg5(dimethylenastron et ispinesib mesylate) et MKLP-2 (paprotrain) bloquent les étapes clés de laformation des vaisseaux sanguins (prolifération, adhérence et migration des cellules endothéliales),la prolifération des cellules tumorales ainsi que la formation de néo-vaisseaux dans des culturesd’anneaux aortiques. De plus, sur la CAM et chez la souris, l’inhibition de cette même kinésinediminue significativement la croissance et la vascularisation des modèles tumoraux utilisés lors dece projet (le glioblastome et le carcinome rénal). Role of the mitotic kinesins Eg5 and MKLP-2 in physiologic and pathologic angiogenesis.Angiogenesis is a complex biological phenomenon which corresponds to the formation of newblood vessels from pre-existing vessels. This process is regulated by a plethora of differentmolecules with vascular endothelial growth factor (VEGF) being one of the most important ones.VEGF inhibitors are currently used in the treatment of numerous solid cancers. Even though theefficacy of such treatment is prouven by numerous studies, resistance to anti-angiogenic therapy isa common feature. To identify new therapeutic targets downstream of VEGF, we modelized itsaction on the chick chorioallantoic membrane (CAM). VEGF-treated CAMs develop a densevascular network 24h after application. We used chick microarrays to monitor global geneexpression changes in VEGF-induced CAMs. Beside a consistent number of genes alreadydescribed to be regulated by VEGF, numerous unknown genes have been identified. We havefocused our work on the characterization of Eg5/KIF11 and MKLP-2/KIF20A, members of thekinesin family, both strongly upregulated by VEGF.We demonstrated that Eg5 and MKLP-2 are strongly expressed by blood vessels in normal andcancer tissue sections. KIF20A is involved in the proliferation and migration of endothelial cellsin vitro. We showed that chemical inhibitors specific for KIF11/Eg5 (dimethylenastron andispinesib mesylate) affect key steps in the formation of blood vessels (proliferation, adhesion andmigration of endothelial cells) and proliferation of tumor cells (glioma and renal cancer).Furthermore, in experimental glioblastoma and renal cell carcinoma models (CAM and orthotopicimplantation in mice), anti-Eg5 treatment strongly reduces tumor angiogenesis and growth. The diploma thesis deals with comparative analysis of stress and strain of notched machine parts using photoelasticity and finite element method (FEM). Experimental and numerical comparative analyses were performed on three models: hook, J shaped bracket and 3-point bending beam with notch. Equivalent stresses, principal stresses and its directions were analyzed. The emphasis is given on photoelasticity, the theory and principles of photoelasticity are described in more detail. FEM analysis was performed using software ANSYS 11. The work presented includes the preparation of polariscope in the laboratory of Institute of machine and industrial design in order to use it in tutorials focused on experimental stress analysis using photoelasticity.

The presented work is concerning with ductile fracture problematic under monotonic loading which is result of gradual material degradation at large plastic deformation. At present, a large number of models, which calibration is not trivial, are used for its prediction. Ductile fracture mechanisms and cut-off region are described in the literature search part. Next, there is given a summary of criteria which are based on different approaches to ductile fracture. There were proposed and used KHPS and KHPS2 criteria within this PhD thesis. Procedure of assessment of the stress-strain curve and ductile fracture criteria calibration are described in last part of the literature search. A summary of tests, which were done in order to calibrate chosen criteria, is given in experimental part. Various stress states within broad range of stress triaxiality and Lode parameter were reached using these tests. Special type of specimen, by which very low stress triaxiality value is reached, was designed within this work. All specimens were made from the steel 12 050. Wrought rods of one melt with circular cross-section of 27 mm in diameter were used as semi-product. Calibration of chosen ductile fracture criteria was performed using foregoing tests. These criteria were implemented by author into explicit finite element software ABAQUS/Explicit using user subroutine VUMAT. Chosen criteria were used for simulation of multistep extrusion at which formation of internal central cracks occurs. These criteria were also used for simulation of cutting of circular cross-section rods. Results from simulation were compared to experiments which were done by industrial partner J-VST.

The content of this thesis is a model and mesh creation and electromagnetic simulation of synchronous machine. The stator and rotor pack termination effect on the synchronous machine.is analyzed by finite element method in ANSYS program. The type of the stator termination pack is described by four segments. The magnetic field distribution is deformed due to the material properties of used segments. In the next part a methodics of mesh creation is shown. The mesh quality (in term of elements number and elements shape) is important due to the solution accuracy and speed.

MKP-1 también media en los mecanismos de quimioresistencia en cáncer de mama y se inhibe por la actividad de la doxorrubucina en modelos celulares de cáncer de mama. Este trabajo aquí presentado tiene como objetivo caracterizar los efectos de la doxorrubicina sobre MKP-1 y sobre las MAPK en cáncer de mama e investigar su relevancia como factor pronóstico en esta enfermedad. Experimental Design: Los efectos de la doxorubicin en MKP-1, la forma fosforilada (p) de ERK1/2 (p-ERK1/2), de p-JNK y de p-p38 se estudiaron en un panel de líneas celulares de cáncer de mama humano por técnicas de Western blot y en tumores humanos cultivados ex vivo por inmunohistoquímica. De forma adicional, MKP-1 se estudió en una serie de tejidos mamarios desde parénquima normal hasta carcinoma infiltrante de mama, y en una cohorte amplia de pacientes de cáncer de mama con seguimiento clínico. Results: MKP-1 se expresaba en niveles bajos en el tejido mamario normal y en la hiperplasia ductal, detectándose sobreexpresión en el carcinoma ductal in situ. MKP-1 también se sobreexpresaba en casi el 50% de los tumores mamarios. De forma similar en las líneas de cáncer de mama y en los tumores cultivados ex vivo, de forma general la doxorrubicina redujo la expresión de MKP-1 e incrementó los niveles de p-ERK1/2 y JNK. Aquellos casos, sin embargo, que sobreexpresaban MKP-1, la doxorrubicina no afectó la expresión de MKP-1 ni de las formas fosforiladas de las MAPK. Por último, la sobreexpresión de MKP-1 se reveló como un factor de mal pronóstico en los análisis uni- y multivariados para la supervivencia libre de enfermedad en las pacientes con cáncer de mama estudiadas. Conclusions: MKP-1 se sobreexpresa en la transformación maligna de la mama y se asocia a una peor supervivencia en las pacientes con cáncer de mama. Además, MKP-1 es inhibida por la doxorrubicina en esta enfermedad.
Purpose: Mitogen-activated protein kinase (MAPK) phosphatase-1 (MKP-1) regulates by dephosphorylation the activity of the major intracellular signaling pathways mediated by mitogen-activated protein kinase [extracellular signalregulated kinase (ERK), c-Jun NH2-terminal kinase (JNK), and p38]. MKP-1 mediates breast cancer chemoresistance and is repressible by doxorubicin in breast cancer cells. This work is aimed to characterize doxorubicin effects on MKP- 1 and phospho-MAPKs in human breast cancers and to study the clinical relevance of MKP-1 expression in this tumor. Experimental Design: Doxorubicin effects on MKP-1, phospho-ERK1/2 (p-ERK1/2), phospho-JNK (p-JNK), and phospho-p38 were assayed in a broad panel of human breast cancer cells by Western blotting and in human breast cancer were assayed ex vivo by immunohistochemistry. MKP-1 expression was also assayed in a spectrum of normal to malignant breast lesions and in a large series of breast cancer patients with clinical follow-up. Results: MKP-1 was expressed at low levels in normal breast and in usual ductal hyperplasia and at high levels in in situ carcinoma. MKP-1 was overexpressed in approximately 50% of infiltrating breast carcinomas. Similar to what was observed in breast cancer cell lines, ex vivo exposure of breast tumors to doxorubicin downregulated MKP-1, and upregulated activation of ERK1/2 and JNK, in most of studied cases. However, in a proportion of tumors overexpressing MKP-1, doxorubicin did not significantly affect MKP-1 or activation levels of MAPKs. With regard to patient outcome, MKP-1 overexpression was a significant adverse prognostic factor for relapse both by univariate and multivariate analysis. Conclusions: MKP-1 is overexpressed during the malignant transformation of the breast and independently predicts poor prognosis. Furthermore, MKP-1 is repressed by doxorubicin in human breast cancer.

Although the use of curriculum materials and MKT both influence the quality of mathematics instruction, no relationship between teachers' curricular reasoning and MKT was found. Further, teachers' curricular reasoning across algebra and statistics was very similar despite differences in these fields and differences in MKT. A model for thinking about the knowledge needed to provide high quality instruction is proposed by considering the role of curricular reasoning and MKT in instruction.

Hyper-heuristics are a class of high-level search methodologies which operate over a search space of heuristics rather than a search space of solutions. Hyper-heuristic research has set out to develop methods which are more general than traditional search and optimisation techniques. In recent years, focus has shifted considerably towards cross-domain heuristic search. The intention is to develop methods which are able to deliver an acceptable level of performance over a variety of different problem domains, given a set of low-level heuristics to work with. This thesis presents a body of work investigating the use of selection hyper-heuristics in a number of different problem domains. Specifically the use of crossover operators, prevalent in many evolutionary algorithms, is explored within the context of single-point search hyper-heuristics. A number of traditional selection hyper-heuristics are applied to instances of a well-known NP-hard combinatorial optimisation problem, the multidimensional knapsack problem. This domain is chosen as a benchmark for the variety of existing problem instances and solution methods available. The results suggest that selection hyper-heuristics are a viable method to solve some instances of this problem domain. Following this, a framework is defined to describe the conceptual level at which crossover low-level heuristics are managed in single-point selection hyper-heuristics. HyFlex is an existing software framework which supports the design of heuristic search methods over multiple problem domains, i.e. cross-domain optimisation. A traditional heuristic selection mechanism is modified in order to improve results in the context of cross-domain optimisation. Finally the effect of crossover use in cross-domain optimisation is explored.

Disertační práce se věnuje problematice obecných koncentrátorů napětí v anisotropních prostředích. Zejména se jedná o problém trhlin končících na rozhraní dvou různých materiálů, či problém obecného více-materiálového klínu. Cílem práce je vytvořit komplexní nástroj pro posuzování obecných koncetrátorů napětí tj, popis pole napětí v jeho okolí, zahrnutí případného vlivu přemostění trhliny do výsledného pole napětí a definici lomových kritérií pro obecný koncentrátor v anisotropním prostředí. U popisu pole napětí je využit tzv. Lechnického-Strohův formalismus a technika spojitě rozložených dislokací využívající teorii komplexních potenciálů. V práci je rovněž široce uplatněn tzv. dvoustavový "psí"-integrál (pro výpočet různých součinitelů asymptotického rozvoje pro napětí), založený na Bettiho recipročním teorému v kombinaci s metodou konečných prvků. Pro formulaci lomových kritérií je použita teorie tzv. „konečné lomové mechaniky“ a teorie sdružených asymptotických rozvojů. Studován je především vztah mezi ohybem trhliny podél rozhraní a její případnou pentrací do základního materiálu. Veškeré potřebné výpočty jsou prováděny v matematických softwarech MAPLE 10.0, MATLAB 7.1 a konečnoprvkovém systému ANSYS 10.0.

The following study aims to examine the special mathematical knowledge needed in order to teach mathematics. Furthermore, the study attempts to explore how teachers’ views on the knowledge needed in order to teach mathematics affects their student’s opportunities to develop their conceptual understanding. Qualitative and quantitative empirical data was attained by observations and complementary interviews. A total of three teachers, all working at the same school, was observed and interviewed. The study used Ball, Thames & Phelps (2008) practice-based theory of mathematical knowledge for teaching, MKT, as its theoretical framework when analyzing the empirical data. The result of the observations displays that math teachers tend to use common content knowledge far more than specialized content knowledge during their lessons. The outcome of this also study reveals that there is a tendency among teachers to interfuse mathematical concepts with terminology. Conceptual understanding is equated with the use of correct terminology. The students are not exposed to the underlying ideas of the mathematical concepts. The study also concludes that there seems to be a sectioning between the mathematical content taught in grade 4-6 from the rest of the content being taught in elementary school, with a low number of connections being made between mathematical topics and concepts included in the curriculum.

ENGLISH ABSTRACT: Introduction: Cancer is an emerging health problem in South Africa, with breast cancer being one of the leading cancers affecting women globally. Therefore, there is a need to find novel targets to improve the therapeutic options for these patients. A recently proposed target is the mitogen-activated protein kinase phosphatase-1 (MKP-1). Studies have suggested that mitogen-activated protein kinase phosphatases are involved in the development of cancer and play an important role in the response of cancer cells to chemotherapy. Additionally, numerous studies have indicated that there is increased expression of MKP-1 in breast cancers where its over-expression is proposed to be a significant mediator in chemo-resistance. We propose that inhibition of MKP-1 will increase the cytotoxic effect of doxorubicin in breast cancer cells, thus making the cells more responsive to treatment leading to increased cell death through autophagy and apoptosis. Methods: In MDA-MB231 cells, MKP-1 was inhibited using sanguinarine or MKP-1 siRNA and this was compared to a known inducer of MKP-1, dexamethasone. MDA-MB231 cells were treated with doxorubicin alone or in combination with MKP-1 inhibitors or an inducer. Following treatment, cell death was determined by trypan blue and a caspase glo assay as well as with western blotting. Autophagy was determined by western blotting and flow cytometry. LC3 and p62 were used as markers of autophagy and caspase 3 and PARP as apoptosis markers. Likewise, the level of MKP-1 expression under conditions of MKP-1 induction, inhibition or silencing was evaluated by means of western blotting. C57BL6 tumour bearing mice was used to analyse apoptosis and autophagy in vivo under conditions of MKP-1 inhibition, using sanguinarine, together with doxorubicin treatment. Western blotting was used to determine levels of caspase 3, LC3, p62 and MKP-1 expression. Results and discussion: A concentration and time curve indicated that 5 μM doxorubicin reduced cell viability in the MDA-MB231 cells significantly after 24 hours of treatment. MKP-1 expression was significantly reduced with sanguinarine and MKP-1 siRNA. Furthermore, our results indicate a significant increase in apoptosis in MDA-MB231 cells when treated with doxorubicin, under conditions of MKP-1 inhibition or MKP-1 silencing. Also, an increase in autophagic activity was observed following treatment with doxorubicin in combination with sanguinarine. Whole excised tumours of C57BL6 mice also showed an increase in apoptosis and autophagy following treatment with sanguinarine in combination with doxorubicin. This indicates that the inhibition of MKP-1 with sanguinarine sensitized the MDA-MB231 cells and E0771 cell tumours to doxorubicin-induced-apoptosis through a mechanism involving autophagy. Conclusion: This is an encouraging finding that could hopefully be used in future studies to overcome doxorubicin-resistance in breast cancer cells overexpressing MKP-1. Targeting MKP-1 can have potential therapeutic benefits for breast cancer patients by making chemotherapy more effective. Sanguinarine thus has potential to be developed as a clinically relevant inhibitor of MKP-1 which could provide a novel avenue for therapeutic intervention in combination with chemotherapy in breast cancer patients.
AFRIKAANSE OPSOMMING: Inleiding: Kanker is 'n vinnig groeiende gesondheidsprobleem in Suid-Afrika, met borskanker as een van die vernaamste kankers wat vroue wêreldwyd raak. Daar is dus 'n behoefte aan nuwe terapeutiese opsies vir hierdie pasiënte en mitogeen-geaktiveerde proteïenkinase fosfatase-1 (MKP-1) is onlangs voorgestel as ‘n moontlike teiken. Verskeie studies toon dat mitogeen-geaktiveerde proteïenkinase fosfatases betrokke is by die ontwikkeling van kanker en ook belangrike rolspelers is in die reaksie van kanker op chemoterapie. Daarbenewens toon talle studies dat daar verhoogde MKP-1 uitdrukking in borskanker is, asook dat dit ‘n belangrike bemiddelaar is vir die weerstand wat borskanker teen chemoterapie bied. Ons het dus voorgestel dat die inhibisie van MKP-1 die sitotoksiese effek van doxorubicin op borskanker selle sal verhoog; sodoende sal die kanker selle beter reageer op behandeling en dit sal dus lei tot verhoogde seldood deur autofagie en apoptose. Metodes: MKP-1 is geïnhibeer met behulp van sanguinarine of MKP-1 siRNA in MDA-MB231 selle en dit is vergelyk met 'n bekende MKP-1 induseerder, dexamethasone. Seldood is bepaal deur middel van ‘n trypan blou en kaspase toetsingsmetode, asook met die westelike kladtegniek. Autofagie is bepaal deur westelike kladtegniek en vloeisitometrie. LC3 en p62 is gebruik as merkers van autofagie en kaspase 3 en PARP is as apoptose merkers gebruik. MKP-1 uitdrukking is geëvalueer deur middel van westelike kladtegniek. C57BL6 muise met kankeragtige gewasse is gebruik om apoptose en autofagie in vivo te ondersoek. MKP-1 is geïnhibeer met sanguinarine en die muise is behandel met ‘n kombinasie van sanguinarine en doxorubicin. Kaspase 3, LC3, p62 en MKP-1 uitdrukking is bepaal deur middel van die westelike kladtegniek. Resultate en bespreking: ‘n Konsentrasie en tyd kurwe het aangedui dat 5 μM doxorubicin die MDA-MB231 selle se lewensvatbaarheid aansienlik verminder het na 24 uur. MKP-1 uitdrukking is ook aansienlik verminder met sanguinarine en MKP-1 siRNA. Verder dui die resultate op 'n beduidende toename in apoptose in MDA-MB231 selle na behandeling met doxorubicin onder toestande van MKP-1 inhibisie. 'n Toename in autofagiese aktiwiteit is waargeneem na behandeling met doxorubicin en sanguinarine. Die kankeragtige gewasse van die C57BL6 muise toon ook 'n toename in apoptose en autofagie na behandeling met sanguinarine en doxorubicin. Hierdie resultate dui daarop dat die inhibisie van MKP-1 met sanguinarine die MDA-MB231 selle en E0771 sel gewasse gesensitiseer het tot doxorubicin-geïnduseerde apoptose deur middel van ‘n meganisme wat autofagie insluit. Gevolgtrekking: Hierdie bevinding kan hopelik in toekomstige studies gebruik word om doxorubicin weerstand te oorkom in borskanker selle waar MKP-1 verhoog is. Deur MKP-1 te teiken, kan dit lei tot potensiële terapeutiese voordele vir borskanker pasiënte en sodoende kan dit chemoterapie meer effektief maak. Sanguinarine het dus die potensiaal om ontwikkel te word as ‘n klinies relevante inhibeerder van MKP-1 wat sodoende kan dien as terapeutiese intervensie in kombinasie met chemoterapie vir borskanker pasiënte.

Thesis deals with determination of layerwise mechanical properties of composite ceramics by indirect method, namely Youngs modulus. Based on literature review, it was found that a method to determine elastic properties of one or more components of multi layered composites based on experimental modal analysis and finite element modal analysis, or analytical approach exists. Method based on FE modal analysis was applied to ceramic laminate, where it was attempt to determine youngs modulus of one component. Beyond that, it was attempt to determine Youngs moduli of both components using first two bending resonant frequencies. Results were unsatisfying. Sensitivity analysis showed that layers with unknown Youngs modulus were overly sensitive to small changes in input parameters due to their small relative thickness with respect to thickness of laminate and due to location in laminate. Based on this conclusion, recommendations were made with respect to suitable geometry of test specimens.

Mitogen-activated protein kinase phosphatase-2 (MKP-2) is a type 1 nuclear dual specific phosphatase (DUSP4) and an important immune regulator. It specifically dephosphorylates the MAP kinases ERK and JNK to influence pro- and antiinflammatory cytokine production. MKP-2 has recently been shown to play a significant role in controlling Leishmania mexicana infection (Al-Mutairi et al., 2010) primarily by influencing macrophage activity. However, information on the effect of MKP-2 deletion at the molecular level on macrophage development and function is limited. This project utilised a novel DUSP4 gene knockout mouse and investigated the effects of MKP-2 deletion on M-CSF induced MAPK signalling and macrophage development as well as macrophage gene expression. Experiments in bone marrow derived macrophages demonstrated that in response to M-CSF macrophage, proliferation was reduced following to MKP-2 deletion. This was correlated with ERK phosphorylation, the expression of CD115 and CD34 on macrophage progenitors as well as the induction of genes related to macrophage differentiation and proliferation, colony stimulating factor-2 (Csf2) and monocyte to macrophage differentiation associated (Mmd) genes. In addition a comparative microarray gene expression analysis was conducted on MKP-2-/- and MKP-2+/+ macrophages following (LPS) or (IL-4) activation. As demonstrated previously, and associated with a role for MKP-2 in antimicrobial activity, arginase-1 expression was up-regulated in MKP-2-/- compared with MKP- 2+/+ macrophages. Surprisingly, and in contrast, we found that other alternative activation markers Ym1 (Chi3l3) and Fizz1/Retnla (Relm-a) were significantly reduced in MKP-2-/- macrophages when compared with their wild-type counterparts. As both Ym1 and Fizz1 have been implicated to play a major role in extracellular matrix disposition this suggests a significant role for MKP-2 in wound healing. Collectively, the findings in the current study have established that MKP-2 plays an important role in macrophage development and immune function.

Resumo: A proteína MKP-3 (MAPK phosphatase-3) possui capacidade de associar-se fisicamente e desfosforilar a FoxO1 no fígado, resultando em aumento da gliconeogênese e hiperglicemia na condição de obesidade e diabetes. Por outro lado, o exercício físico é uma das estratégias não farmacológicas mais utilizadas para a melhora do perfil glicêmico em pacientes diabéticos, no entanto, suas ações no tecido hepático são pouco conhecidas. Assim, o objetivo deste trabalho foi investigar os efeitos do treinamento físico sobre a expressão proteica da MKP-3, bem como sua interação com a FoxO1 em fígado de animais obesos. Ademais, tem como objetivo avaliar o mecanismo pelo qual o exercício físico é capaz de inibir a proteína MKP-3 em tecido hepático. Foram utilizados camundongos Swiss machos que receberam por 16 semanas uma dieta padrão ou rica em gordura. Os animais foram distribuídos aleatoriamente em seis grupos: controle (C), obeso sedentário (OB), obeso exercitado (EXE), obeso tratado com oligonucleotídeo antisense MKP-3 (OB-ASO), obeso tratado com sense MKP-3 (OB-sense) e obeso submetido ao treinamento físico e ao tratamento com antisense MKP-3 concomitantemente (OB-EXE-ASO). O protocolo de treinamento físico consistiu de natação com sessões de 60 minutos, 5 vezes por semana, com carga equivalente a 5% da massa corporal total do animal. O tratamento com antisense foi realizado através de injeções duas vezes ao dia, com um volume total de 2.0 ?l por dose (4.0 nmol/?l) por 5 dias. Ao final do experimento foram realizados os testes de tolerância ao piruvato (TTP) e teste de tolerância à insulina (TTI). Além disso, foram avaliados parâmetros fisiológicos (ingestão calórica, massa corporal, glicemia e insulinemia de jejum) e a análise molecular das proteínas (MKP-3, FoxO1, PGC1?, HNF-4?, PEPCK, G6Pase, ERK e CK2?) através das técnicas de imunoblot e imunoprecipitação. Utilizou-se a técnica de imunohistoquímica para análise da co-localização da MKP-3 no fígado dos animais. Os resultados obtidos demonstraram que o treinamento físico diminuiu expressão proteica de MKP-3 e a associação FoxO1/MKP-3 no fígado dos animais obesos. Além disso, camundongos obesos treinados apresentaram maiores níveis de FoxO1 fosforilada e diminuição da expressão das proteínas PGC-1? e HNF-4???no fígado se comparado aos animais obesos não submetidos ao treinamento físico. Verificam-se também menores níveis das enzimas gliconeogênicas PEPCK e G6Pase nos animais obesos treinados se comparado aos seus pares sedentários. Estes resultados a nível molecular foram acompanhados por alterações fisiológicas incluindo aumento da sensibilidade à insulina no fígado, menor produção hepática de glicose e redução da hiperglicemia nos camundongos obesos, independentemente da redução da massa corporal total. Ademais, não foram encontrados efeitos aditivos sobre os parâmetros fisiológicos e moleculares com o tratamento de oligonucleotídeo antisense MKP-3 e treinamento físico realizado concomitantemente nos animais obesos. Por fim, os efeitos supressivos do treinamento físico sobre a proteína MKP-3 parecem estar relacionados, no mínimo em parte, a diminuição na fosforilação das ERKs no fígado de camundongos obesos
Abstract: The protein MKP-3 (MAPK phosphatase-3) has the ability to physically associate with and dephosphorylate the FoxO1 in the liver, resulting in increased gluconeogenesis and hyperglycemia in the condition of obesity and diabetes. On the other hand, exercise is one of the most used non-pharmacological strategies for improving glycemic control in diabetic patients; however, their actions in liver tissue are poorly explored. The objective of this study was to investigate the effects of exercise on the expression of MKP-3 and its interaction with FoxO1 in the liver of obese animals. Furthermore, aimed at evaluating the mechanism by which physical exercise is able to inhibit protein MKP-3 in the liver. We used male Swiss mice that received a standard diet or high fat for 16 weeks. The animals were randomly divided into six groups: control (C), sedentary obese (OB), obese exercised (EXE), obese treated with MKP-3 antisense oligonucleotide (ASO-OB), obese treated with sense MKP-3 (OB- sense) and obese subjected to physical training and treatment with antisense MKP-3 concomitantly (OB-EXE-ASO). The protocol consisted of swimming exercise training with sessions of 60 minutes, five times per week with a load equivalent to 5% of the total body mass of the animal. Treatment with antisense was performed by injection twice daily, with a total volume of 2.0 ul per dose (4.0 nmol / microl) for 5 days. At the end of the experiment were performed to pyruvate tolerance tests (PTT) and insulin tolerance test (ITT). Furthermore, physiological parameters were evaluated (caloric intake, body weight, blood glucose and fasting insulin) and molecular analysis of proteins (MKP-3, FoxO1, PGC1a, HNF-4a PEPECK, G6Pase, ERK and CK2?) by immunoblotting and immunoprecipitation techniques. The immunohistochemistry technique was utilized to evaluate the co-localization of MKP-3 in the liver of the animals. The results showed that physical training decreased the protein expression of MKP-3 and association FoxO1/MKP-3. Further, trained obese mice had higher levels of phosphorylated FoxO1 and decreased PGC-1? and HNF-4??? There was also a decrease in the protein levels of PEPCK and G6Pase in the liver of mice exercised compared to sedentary obese mice. These results at molecular level were accompanied by physiological changes including increased insulin sensitivity in the liver, reduced hepatic glucose production and reduce hyperglycemia in obese mice, regardless of reducing total body mass. In addition, there were no additive effects of treatment with antisense oligonucleotide MKP-3 and physical training performed concurrently in obese animals. Finally, the suppressive effects of physical training on MKP-3 protein appear to be related at least in part, the decrease in phosphorylation of ERKs in the livers of obese mice

Opettajan ammatillinen itseymmärrys tarkoittaa käsitystä itsestä opettajana. Se kuvaa sitä, miten opettaja ymmärtää itsensä suhteessa työhön ja ammatillisuuteen. Ammatillinen itseymmärrys muuttuu uusien kokemusten ja oppimisen myötä. Opettajan vahva ammatillinen itseymmärrys auttaa työssä jaksamisessa sekä oman pedagogisen toiminnan ja valintojen perustelussa. Ammatillinen itseymmärrys kehittyy huomattavasti jo opintojen aikana. Erityisesti opetusharjoitteluissa saadut myönteiset kokemukset tukevat ammatillisen itseymmärryksen kehittymistä opintojen aikana. Jotta opiskelijan ammatillista kehittymistä voidaan tukea mahdollisimman hyvin, on tärkeä tietää, mitkä tekijät opetusharjoittelussa voivat tukea tai haitata ammatillisen itseymmärryksen kehittymistä. Maisterivaiheen koulutyöskentely -opintojakso (MKT-harjoittelu) on yksi Oulun yliopiston luokanopettajakoulutukseen kuuluvista harjoitteluista. Se suoritetaan Oulun normaalikoululla yleensä neljännen tai viidennen opintovuoden aikana. Tutkielmani tavoitteena on selvittää, mitä luokanopettajaopiskelijat kertovat MKT-harjoittelun merkityksestä ammatillisen itseymmärryksen kehittymiselle. Keskityn erityisesti opiskelijoiden nimeämiin ammatillisen itseymmärryksen kehittymistä tukeviin ja haittaaviin tekijöihin. Tutkimuskysymykseni on: ”Mitkä tekijät MKT-harjoittelussa ovat opiskelijoiden mielestä merkittäviä ammatillisen itseymmärryksen kehittymisen kannalta?” Tarkentavia alakysymyksiä ovat: ”Mitkä tekijät MKT-harjoittelussa tukevat ammatillisen itseymmärryksen kehittymistä?” sekä ”Mitkä tekijät MKT-harjoittelussa haittaavat ammatillisen itseymmärryksen kehittymistä?” Tutkimusaineisto koostuu neljän maisterivaiheen luokanopettajaopiskelijan teemahaastatteluista ennen MKT-harjoittelua ja sen päättymisen jälkeen. Haastatteluaineisto on analysoitu aineistolähtöisesti ja analyysin loppuvaiheessa aineistoon on yhdistetty teoria itseymmärryksen osa-alueista. Tutkimukseni osoittaa, että MKT-harjoittelun ohjaava opettaja voi sekä tukea että haitata opiskelijan ammatillisen itseymmärryksen kehittymistä. Harjoittelun aikaiset onnistumiset sekä toimiva vuorovaikutus oppilaiden kanssa tukevat ammatillisen itseymmärryksen kehittymistä. Lisäksi ammatillisen itseymmärryksen kehittyminen MKT-harjoittelun aikana on yhteydessä opiskelijan käsitykseen tulevaisuudesta. Erityisesti ohjaavan opettajan palautteella oli tässä tutkimuksessa merkitystä opiskelijan ammatillisen itseymmärryksen kehittymiselle. Palaute oli yhteydessä myös opiskelijan tulevaisuuskuvaan. Koska MKT-harjoittelu on viimeisiä luokanopettajakoulutuksessa suoritettavia opintojaksoja, harjoittelun aikaisella ammatillisen itseymmärryksen kehittymisellä voi olla suuri merkitys siihen, millaisia opettajia koulutuksesta valmistuu. Ohjaavan opettajan palautteen merkitys tulisi huomioida opetusharjoitteluiden toteutuksessa, erityisesti ammatillisen itseymmärryksen kehittymistä haittaavaan palautteen osalta.

Experimental Autoimmune Encephalomyelitis (EAE) is an animal model of multiple sclerosis (MS), causing a demyelinating central nervous system (CNS) inflammation which resembles the main pathological features of MS. Mitogen-activated protein kinases (MAPKs), which are key components in the molecular response leading to MS/EAE pathogenesis, are regulated by MAPK phosphatases (MKPs), enzymes which dephosphorylate phosphotyrosine and phosphothreonine residues. It has previously been shown that MKP-2 modulates the inflammatory response during both acute lung injury and sepsis. Therefore in the present study we investigated the role of MKP-2 in the development of the neurological autoimmune disease, MS, using a murine EAE model. We first observed significantly increased expression of MKP-2 mRNA in the spinal cord of EAE mice compared with PBS controls. Subsequently, to understand the function of MKP-2 in vivo, we utilised MKP-2 deficient mice, inducing EAE in MKP-2 KO and WT littermates. Our data show that MKP-2 KO mice displayed significantly reduced EAE susceptibility, associated with diminished CNS inflammation and cellular infiltration, decreased expression of key cytokines and chemokines (IL-17, IFNγ, IL-6, IL-2 and CCL2), reduced frequency of CD4⁺ T cells, CD8⁺ T cells and B cells in spleen and dLN tissue as well as downregulated nitric oxide (NO) production in MKP-2 KO EAE mice. We further analysed the role of MKP-2 in two key immune cells involved in EAE pathogenesis. Upon LPS stimulation, MKP-2 deficient bone marrow-derived dendritic cells expressed less MHC-II while producing more IL-6, TNF-α and IL-10, whereas MKP-2 KO bone marrow-derived macrophages displayed a unique M1 and M2 mixed phenotype, with reduced NO production (M1) and increased CD206 expression (M2) but increased IL-6 and TNF-α, which are more associated with M1 responses. Therefore this report suggests that MKP-2 is essential to the pathogenic response of EAE, and that inhibition of MKP-2 expression or function may be a viable strategy in the treatment of autoimmune inflammatory diseases such as MS.

Skolverket har skrivit rapporter om förstelärare generellt. Syftet med denna studie var att exemplifiera vad försteläraren i matematik gör och vad det kan medföra, både för kollegor och elever. I studien genomfördes kvalitativa intervjuer med sju förstelärare i matematik vilket synliggjorde exempel på hur förstelärare i matematik arbetar och vad de upplever att det har för effekter. Studiens resultat visade bland annat att det förstelärarna i matematik gör i sitt uppdrag skiljer sig markant. En förstelärare gör i princip ingenting i sitt uppdrag mer än att ha en titel. En annan förstelärare arbetar aktivt med screening för att bedöma elevers kunskaper i matematik, möjliggör för undervisning av praktisk matematik och driver projekt för att kompetensutveckla lärarkollegor i kommunen. Förstelärarnas arbete sker på olika villkor. Villkoren handlar bland annat om vilken tid som finns avsatt för uppdraget, hur stort antal elever som går på skolan försteläraren arbetar på och vilka andra projekt som drivs parallellt på skolan. Något som är avgörande för hur förstelärarens insatser ser ut är dels hur engagerade huvudmännen i form av rektorer och skolchefer är och vilka krav de ställer, dels hur passionerad och drivande försteläraren själv är.
The Swedish national agency of education has written reports about what the first-teacher do in general. The purpose with this study is to exemplify what the first-teacher in mathematics do and what it possibly can result in, both for colleagues and pupils. By making qualitative interviews with seven first-teachers in mathematics the study illustrates how they work and what outcome they experience. The results show that the first-teacher assignments differs, for example, one teacher does nothing above her regular teaching in her assignment more than having a title. Another first-teacher actively work with screening to assess pupils’ knowledge in mathematics, enables teaching of practical mathematics and is responsible for different projects in developing competence for colleagues. The first-teachers work is done under different conditions, such as time assigned to the mission, number of pupils at the school they work at and other on-going projects. Something that is crucial for the first-teacher efforts is, in part, how committed principal and school leadersare, and what demands they make, partly how passionate and skilful the first-teacher is herself.

Den här kvalitativa forskningsstudien fokuserar på vilka kunskaper lärare anser behövs vid utomhuspedagogik i matematik. Syftet med forskningsstudien är att undersöka vad som krävs av läraren för att planera samt genomföra en geometrilektion vid utomhuspedagogik. Undersökningen bygger på en kvalitativ ansats där textanalys och semistrukturerade intervjuer, där lärarna får ge sin bild av lektionsplaneringen, som utgör en del av studiens datainsamling. Studien utgår ifrån en hermeneutisk ansats, med utgångspunkt i Ball m.fl. teoretiska ramverk MKT samt van Hieles teori. Den här studien fokuserar på hur tre verksamma lärare planerat en geometrilektion och vilka kunskaper lärarna anser sig behöva för att bedriva matematiklektionen utomhus. Sammanfattningsvis visar studiens resultat att lärarna uppskattar utomhuspedagogik, men att det däremot sker i mindre utsträckning på grund av väder, tid och resurser. Resultatet visar även att lärarna har kunskaper för att undervisa i matematik men de visar däremot på olika kunskaper utifrån teorin MKT. Lärarna visar på olika kunskaper vid utformandet av sin lektionsplanering i geometri samt att de använder sig av läroplanen i olika stor utsträckning. Avslutningsvis visar resultatet att lärarna anser att de vid undervisning av geometri utomhus behöver vara organiserade, ha struktur och en genomtänkt planering. De menar även att det behövs en varierarnade undervisning inom geometri, där eleverna får arbeta konkret och samtala med varandra.
