We examined the antiosteoarthritis effect of a mixture of powdered pomegranate concentrate, eucommiae cortex, and achyranthis radix (5:4:1 w/w) (PEA-Mix). After the injection of monosodium iodoacetate (MIA), PEA-Mixs were orally administered. To assess pain-related behaviors, a von Frey filament test and open field test were performed. We also examined the knee thickness, maximum knee extension angle, bone mineral density (BMD), and compressive strength of the knee joint and performed a histopathologic analysis. The number of COX-2, tumor necrosis factor (TNF)-α, poly (ADP-ribose) polymerase (PARP), and 5-bromo-2′-deoxyuridine immunoreactive cells, the prostaglandin E2 (PGE2) and 5-lipoxygenase (LPO) levels, matrix metalloproteinase (MMP) activity, and mRNA levels of chondrogenesis-related genes were analyzed. PEA-Mix significantly inhibited the MIA-induced decrease in the paw-withdrawal threshold and total distance moved, and the MIA-induced increases in the maximum knee extension angle and knee thickness. Also, the MIA-induced loss of the knee joint articular surface region and decrease in the BMD were significantly suppressed by PEA-Mix. The MIA-induced increases in the 5-LPO, PGE2, MMP-2, MMP-9, COX-2, and TNF-α mRNA levels were reduced by PEA-Mix. PEA-Mix increased the MIA-mediated reduction in the SOX-9 and aggrecan mRNA levels. The number of PARP-positive cells was smaller in PEA-Mix-administered rats than in MIA-administered rats, while the number of 5-bromo-2′-deoxyuridine-positive cells was larger. Therefore, PEA-Mix relieved the MIA-induced pain-related behaviors, chondrocyte proliferation, and anti-inflammatory activity.

The issues of retail and office center structure formation are developed and presented in the existent literature. This article argues that in each case, these issues need to be refined depending on the location, the number of neighborhood and the surrounding streets’ residents, the presence of other shops and services within walking distance, etc. The purpose of this article is to confirm the abovementioned specificity during the marketing research, in order to evaluate the significance of the factors forming the structure of retail and office center for the population and future tenants. A survey of 200 residents and 100 business owners and managers was conducted. The study’s results allow the formulation of a framework of retail and office center, determining the number of tenants, taking into account their preferences, and getting a list of interested tenants. It also shows the way to facilitate optimization of the layout of the center with regards to the requirements of the anchor tenants, high-footfall shops and customers; to develop the best routes to distribute customers across the floors and center’s zones

Abstract Molecular events involved in specification of early hematopoietic system are not well known. In Xenopus, a paired-box homeodomain family (Mix.1-4) has been implicated in this process. Although Mix-like homeobox genes have been isolated from chicken (CMIX) and mice (Mml/MIXL1), isolation of a human Mix-like gene has remained elusive. We have recently isolated and characterized a novel human Mix-like homeobox gene with a predicted open reading frame of 232 amino acids designated the Mix.1 homeobox (Xenopus laevis)–like gene (MIXL). The overall identity of this novel protein to CMIX and Mml/MIXL1 is 41% and 69%, respectively. However, the identity in the homeodomain is 66% to that of Xenopus Mix.1, 79% to that of CMIX, and 94% to that of Mml/MIXL1. In normal hematopoiesis, MIXL expression appears to be restricted to immature B- and T-lymphoid cells. Several acute leukemic cell lines of B, T, and myeloid lineage express MIXL suggesting a survival/block in differentiation advantage. Furthermore, Xenopus animal cap assay revealed that MIXL could induce expression of the α-globin gene, suggesting a functional conservation of the homeodomain. Isolation of theMIXL gene is the first step toward understanding novel regulatory circuits in early hematopoietic differentiation and malignant transformation.

Abstract NFS-60 and FDCP-Mix cells are interleukin-3--dependent multipotent hematopoietic cells that can differentiate in vitro into mature myeloid and erythroid cells. Retrovirus-mediated transfer of the human colony- stimulating factor-1 (CSF-1) receptor gene (c-fms) enabled NFS-60 cells but not FDCP-Mix cells to proliferate in response to CSF-1. The phenotype of NFS-60 cells expressing the human CSF-1 receptor (CSF-1R) grown in CSF-1 did not grossly differ from that of original NFS-60 as assessed by cytochemical and surface markers. Importantly, these cells retained their erythroid potentiality. In contrast, a CSF-1-dependent variant of NFS-60, strongly expressing murine CSF-1R, differentiated into monocyte/macrophages upon CSF-1 stimulation and almost totally lost its erythroid potentiality. We also observed that NFS-60 but not FDCP-Mix cells could grow in response to stem cell factor, (SCF), although both cell lines express relatively high amounts of SCF receptors. This suggests that SCF-R and CSF-1R signalling pathways share at least one component that may be missing or insufficiently expressed in FDCP-Mix cells. Taken together, these results suggest that human CSF-1R can use the SCF-R signalling pathway in murine multipotent cells and thereby favor self-renewal versus differentiation.

Abstract A study was conducted to characterize the response of Gaillardia aristata Pursh to salinity (0.8, 2.0 or 4.0 dS/m) and growing media: 100% perlite (Perlite), 100% Sunshine Mix No. 4 (Mix), 1 to 1 (by vol) perlite and Sunshine Mix No. 4 (Perlite Mix), or 1 to 1 Sunshine Mix No. 4 and composted mulch (Mix Mulch). Type of medium did not influence shoot dry weight (DW). However, root to shoot DW ratio was highest for plants grown in Perlite. Shoot DW of plants irrigated with tap water (0.8 dS/m) was higher compared to those irrigated with saline solution at 2.0 or 4.0 dS/m, except for those grown in Mix. Salinity did not alter the root to shoot DW ratio. In general, elevated salinity led to relatively short plants. Plants were taller when grown in Perlite or Mix Mulch with fewer lateral shoots compared to plants grown in Mix and Perlite Mix. Flower bud abortion occurred in plants grown in Mix or Perlite Mix, while this phenomenon was not observed in plants grown in Perlite or Mix Mulch. Overall, plants performed better in Perlite and Mix Mulch than Mix and Perlite Mix.

Objective. To observe whether there are sexual-related differences in response to mid- or low-premixed insulin in type 2 diabetic patients. Methods. This was an analysis of CGM data of a previous study. After screening, patients with longstanding T2D receive a 7-day continuous subcutaneous insulin infusion (CSII) therapy, and then subjects were randomly assigned 1 : 1 into two groups receiving Novo Mix 30 or Humalog Mix 50 regimen for a 2-day phage, followed by a 4-day cross-over period. A 4-day continuous glucose monitoring (CGM) was performed during the cross-over period. The primary endpoint was the differences in glycemic control between male and female patients receiving mid- or low-premixed insulin therapy. Results. A total of 102 patients (52 men and 50 women) completed the study. Our data showed that male patients had significant decrease in mean glucose levels monitored by CGM after three meals during Humalog Mix 50 treatment period compared to those received Novo Mix 30 regimen (0900: 11.0±2.5 vs. 12.2±2.8, 1000: 9.9±2.9 vs. 11.3±3.1, 1200: 8.0±1.9 vs. 9.1±2.5, 1400: 9.2±2.3 vs. 10.3±2.5, and 2000: 7.3±2.1 vs. 8.2±2.4 mmol/L, p<0.05, respectively). In addition, male patients receiving Novo Mix 30 experienced a significantly increased hypoglycemic duration compared to those of receiving Humalog Mix 50 (0 (0, 4.8) vs. 0 (0, 0), p<0.05). Conclusion. Our data indicate that male patients with T2D receiving mid-premixed insulin analogue regimen may have a potential benefit of improvement in glycemic control compared to female patients. ABSTRACT The release of crude oil into a southern California river from an earthquake-related pipeline rupture resulted in an initial rise in TPH and total BTEX concentrations in the water column. No toxicity was observed in aquatic toxicity tests conducted with rainbow trout and threespine stickleback. Statistically significant differences noted in several of the indices of macroinvertebrate community structure at sites downstream from the oil entry location appeared to be due to confounding factors including the spill. The assessment of incident-related impacts was complicated by several earthquake-related phenomena.

We have studied the role of the activin immediate-early response gene Mix.1 in mesoderm and endoderm formation. In early gastrulae, Mix.1 is expressed throughout the vegetal hemisphere, including marginal-zone cells expressing the trunk mesodermal marker Xbra. During gastrulation, the expression domains of Xbra and Mix.1 become progressively exclusive as a result of the establishment of a negative regulatory loop between these two genes. This mutual repression is important for the specification of the embryonic body plan as ectopic expression of Mix.1 in the Xbra domain suppresses mesoderm differentiation. The same effect was obtained by overexpressing VP16Mix.1, a fusion protein comprising the strong activator domain of viral VP16 and the homeodomain of Mix.1, suggesting that Mix.1 acts as a transcriptional activator. Mix.1 also has a role in endoderm formation. It cooperates with the dorsal vegetal homeobox gene Siamois to activate the endodermal markers edd, Xlhbox8 and cerberus in animal caps. Conversely, vegetal overexpression of enRMix.1, an antimorphic Mix.1 mutant, leads to a loss of endoderm differentiation. Finally, by targeting enRMix.1 expression to the anterior endoderm, we could test the role of this tissue during embryogenesis and show that it is required for head formation.

The paper argues that the scope of a Member State’s right to electricity mix guaranteed by Article 194(2) TFEUis limited considerably. The right of a Member State to decide on the choice of its energy sources and the general structure of its energy supply in the electricity sector is bordered by the same rights of the other Member States.

Introduction. The research featured the physicochemical and functional properties of tiger nut (Cyperus esculentus), quinoa (Chenopodium quinoa), and their mixes. The research objective was to study the effects of their interfacial interaction and the qualitative characteristics of meat model systems, as well as general opportunities for their use in meat products. Study objects and methods. The tiger nut is a traditional plant that grows in Egypt. It is often used in beverages and bakery. The quinoa is a cereal product that grows mainly in Peru and Colombia. A set of experiments was conducted to determine such colloidalchemical parameters as optical density, electrical conductivity, solubility, pH, and effective viscosity. The studied additives proved to be synergists. The research also included the organoleptic characteristics of meat model samples prepared with a mix of tiger nut and quinoa at different ratios as a partial replacement of raw meat. The mix substituted 5% of the meat system. Tiger nut and quinoa used as a substitute for raw meat positively affected such indicators of the finished product as taste, smell, color, and aroma. The obtained data were analyzed with the help of a specialized software package “Multimeat Expert”. The optimal ratio of tiger nut and quinoa in the mix was determined as 1:1. Results and discussion. The mix of tiger nut and quinoa “TiQi” contained 11.38% of dietary fiber, 14.35% of lipids, and 9.37% of protein. In addition, the mix contained a large amount of minerals: potassium – 636.61 mg/100 g, calcium – 68.54 mg/100 g, and vitamins C, E, and B. Its antioxidant activity reached 20.41 mg/g. Conclusion. The obtained mix of tiger nut and quinoa could be used as a functional ingredient in meat products to increase the content of dietary fibers, as well as vitamins and minerals. In addition, “TiQi” demonstrated good antioxidant activity and increased the shelf life of meat products.

Abstract Background: Hematopoietic stem cell transplantation (HSCT) is cure for thalassemia major (TM). However, a suitable donor (HLA matched sibling and unrelated donor) for HSCT is less than 50%. Alternative donors were recently used in TM HSCT. Some study have found that thalassemia-free survival (TFS) was approximately 70% in haploidentical HSCT (h-HSCT) or unrelated cord blood (UCB) transplant for TM patients. So, it is necessary to find out a better h-HSCT for TM patients. In our early practice in leukemic HSCT we found that outcomes were improved by adding UCB to post-transplant cyclophosphamide (PT/Cy) h-HSCT. The latter associated with high mortality related transplant (32%). Henceforth, we used this termed haplocord transplant in TM. Aim: To develop a high TFS h-HSCT protocol for TM patients. Patients and methods First 10 patients with median age 8 (5-17) old years received NF-13-PT/Cy-TM protocol (fig. 1), in which, UCB was added on day 6 after PT/CY h-HSCT. Following 9 patients with age 9 (4-15) old years received NF-14-PT/Cy-TM protocol (fig. 2), in which three doses Thymoglobuline were added to NF-13-PT/Cy-TM protocol. Cyclophosphamide on day 3 and day 4 after transplant were both GVHD prophylaxis for h-HSCT and conditioning for UCB transplant. The HLA (at HLA-A, -B, -C and ¨CDRB1) for the pair of recipient and donor was 2-loci and more mismatched in h-HSCT and 2-loci and less mismatched in UCB. Results The results of haplocord transplants for all patients were showed in table 1. For first 10 patients, final cord blood engrafted in 4 patients; final haploidentical donor engrafted in 3 patients, 2 patients had a primary rejection. One had a secondary rejection and gave up therapy and died of infection. One patient died of grade IV acute GVHD. TFS is 6/10. For second group patients, final cord blood engrafted in 4 patients; final haploidentical donor engrafted in 3 patients, mixed donor engrafted in 2. No patient rejected his graft; All 9 patients live with transfusion independence. Summary Our data showed that UCB followed PT/Cy h-HSCT using NF-14-PT/Cy-TM protocol improved the results of alternative donor transplant in thalassemia major. Maryland Chesapeake Bay crab industry generates ≈20 tons of crab waste annually. The crab waste compost (CWC) was a mixture of crab chum and saw dust that had an initial EC of ≈26 dS·m–1. In Fall 1994, soft-pinched single stem `Annette Hegg Red' poinsettias (Euphorbia pulcherrima) in 15-cm azalea pots were grown in media containing Sunshine mix, 1 CWC: S base mix (BM), 1 CWC: 2 BM,. 1 CWC: 1 BM, or 2 CWC: 1 BM. Base mix is a 1 peat: 1 perlite (v/v). Fertigation treatment with 266 mg·liter–1 N from 30N–4.4P–8.8K was began on the 1st, 2nd, or 3rd week after potting. The total fertigations ranged from 8 to 10 for the 13-week study. With Sunshine mix, shoot height and canopy diameter were ≈15% greater than with 16% CWC mix and were ≈27% greater than with 67% CWC mix. There was a ≈10% decreased in the shoot height and canopy diameter with increasing %CWC in mix from 17% to 67%, but there was no difference in number of branches among the CWC mixes.

Bacteria in biofilms are encased in an extracellular polymeric matrix that limits exposure of microbial cells to lethal doses of antimicrobial agents, leading to resistance. In Pseudomonas aeruginosa, biofilm formation is regulated by cell-to-cell communication, called quorum sensing. Quorum sensing facilitates a variety of bacterial physiological functions such as swarming motility and protease, pyoverdine, and pyocyanin productions. Peptide mix from the marine mollusc, Olivancillaria hiatula, has been studied for its antibiofilm activity against Pseudomonas aeruginosa. Microscopy and microtiter plate-based assays were used to evaluate biofilm inhibitory activities. Effect of the peptide mix on quorum sensing-mediated processes was also evaluated. Peptide mix proved to be a good antibiofilm agent, requiring less than 39 μg/mL to inhibit 50% biofilm formation. Micrographs obtained confirmed biofilm inhibition at 1/2 MIC whereas 2.5 mg/mL was required to degrade preformed biofilm. There was a marked attenuation in quorum sensing-mediated phenotypes as well. At 1/2 MIC of peptide, the expression of pyocyanin, pyoverdine, and protease was inhibited by 60%, 72%, and 54%, respectively. Additionally, swarming motility was repressed by peptide in a dose-dependent manner. These results suggest that the peptide mix from Olivancillaria hiatula probably inhibits biofilm formation by interfering with cell-to-cell communication in Pseudomonas aeruginosa.

Annotation:
Gaillardia aristata Foug. is a hardy, drought-tolerant perennial found throughout much of the United States. Little information exists on the salt tolerance of this plant when grown in various growing media. A study was conducted to characterize the response of G. aristata to three salinity levels (0.8, 2.0, or 4.0 dS/m) and four growing media: 1) 100% perlite; 2) 1 perlite: 1 Sunshine mix No. 4 (v/v); 3) 100% Sunshine mix No. 4; or 4) 1 Sunshine mix No. 4: 1 composted mulch (v/v). The type of medium influenced the dry weight of roots but not shoots, while salinity significantly influenced the dry weight of both shoots and roots. The dry weight of shoots was higher in plants irrigated with tap water (0.8 dS/m) compared to those irrigated with saline solution at 2.0 or 4.0 dS/m except for those grown in 100% Sunshine mix. The ratio of root to shoot dry weight was not influenced by salinity, but was highest in the plants grown in 100% perlite. Both medium and salinity affected plant height. Elevated salinity reduced plant height. Plants were taller when grown in 100% perlite and in 1 Sunshine mix: 1 composted mulch. However, plants had fewer lateral shoots when grown in 100% perlite or 1 Sunshine mix: 1 composted mulch. Some of the flower buds aborted when grown in 100% Sunshine mix or 1 perlite: 1 Sunshine mix compared to none in plants grown in 100% perlite or 1 Sunshine mix: 1 composted mulch. These results indicate that growth and morphology of G. aristata were affected by not only salinity, but also the type of medium.

Twenty growing buffalo calves were fed on a basal diet consisting of wheat straw and concentrate mixture in a randomised block design, to study the effect of feeding phytogenic feed additives on growth performance, nutrient utilisation and methanogenesis. The four groups were viz. control (no additive), Mix-1 (ajwain oil and lemon grass oil in 1 : 1 ratio @ 0.05% of dry matter intake), Mix-2 (garlic and soapnut in 2 : 1 ratio @ 2% of DMI) and Mix-3 (garlic, soapnut, harad and ajwain in 2 : 1 : 1 : 1 ratio @ 1% of DMI). The experimental feeding was continued for a period of 8 months. A metabolism trial was conducted after 130 days of feeding. Methane emission from animals was measured by open-circuit indirect respiration calorimeter. The feed conversion efficiency was higher by 9.5% in Mix-1, 7% in Mix-2 and 10.2% in Mix-3 group than in control. The digestibility of nutrients was similar except crude protein, which was improved (P < 0.05) in treatment groups. All buffalo calves were in positive nitrogen balance. Comparative faecal nitrogen decreased and urinary nitrogen increased in all the supplemented groups compared with in the control group. Methane emission (in terms of L/kg dry matter intake and L/kg digestible dry matter intake) was reduced by 13.3% and 17.8% in Mix-1, 10.9% and 13.5% in Mix-2 and 5.1% and 9.8% in Mix-3 groups as compared with control. When expressed in L/kg organic matter intake and L/kg digestible organic matter intake, methane production was reduced by 13.3% and 16.7% in Mix-1, 10.9% and 12.9% in Mix-2 and 5.1% and 8.4% in Mix-3 groups compared with the control group. These feed additives inhibited methane emission without adversely affecting feed utilisation by the animals. The faecal energy, urinary energy and methane energy losses were not affected (P > 0.05) due to feeding of these additives. Further, long-term feeding experiments should be conducted on a large number of animals to validate these effects before they can be recommended for use at a field level.
