Vertebrate kinetochores contain over 50 different proteins organized into three distinct regions: the inner plate, outer plate, and fibrous corona. The present study characterizes numerous precursors of kinetochore assembly in a system free of centromeric chromatin, Xenopus extracts. Hydrodynamic analysis suggests there are a minimum of two monomeric proteins and six preassembled complexes that accumulate on centromeres to form the kinetochore. The inner and outer kinetochore assemble from at least two distinct kinetochore complexes containing the proteins Mis12, Zwint, and Ndc80, all of which interact by immunoprecipitation. There is also a network of interactions between the fibrous corona proteins that is dissociated by microtubules. We quantify the number of molecules of specific proteins assembled into a single kinetochore. There are between 800 and 1200 molecules of the measured inner and outer kinetochore proteins, demonstrating that the components in these regions are in similar stoichiometry. In contrast, the measured fibrous corona proteins are present at 250-300 molecules per kinetochore. Zwint, but not Mis12, requires the Ndc80 complex for assembly into the kinetochore. Further, Ndc80 requires Zwint for assembly, indicating a codependency for these two proteins. Our data provide a model for the structural architecture and assembly pathway of the vertebrate kinetochore.

Thereby, a new defect rate prediction model is proposed and validated with three copier assembly cases.FindingsThe new proposed model is much more accurate and stable in the human‐induced assembly defect prediction in copier production.Practical implicationsThe proposed model can be used to ensure the assembly quality by removing potential defects at the structure and process design stages. Meanwhile, with this model, the interactions between the engineers and designers can be more effective.Originality/valueThis paper presents a novel assembly defect rate prediction model for copier assembly quality management.

According to the structure and characteristics of the HSK-A63 tool holder, we design and retrofit the non-standard tool holder with special function module. Establish and assemble three-dimensional entity models of embedded temperature measuring tool holder, ER collet/nut and flat end mill with Pro/E 5.0. Based on the establishment of the reasonable boundary conditions and meshing of the tool holder assembly, natural frequency and vibration modes of the tool holder assembly are calculated by modal analysis with the Ansys Workbench. These analysis results are the important research basis for the dynamic characteristics designing and dynamic balance researching to the tool system.

Electromagnetic actors (solenoids) are used in automotive and automation technology as valve actors for pneumatics and hydraulics or as actuating and pressure solenoids. During assembly there are often press-in and caulking processes used. A central quality aspect of the final control elements is the adherence to defined actuating force ranges in order to ensure the switching process and at the same time to keep wear within limits. These defined force ranges can currently only be maintained by additional heat treatment of the parts. The hysteresis of the assembled unit can be influenced by the caulking. The objective of this paper is to propose the potential reduction of annealing processes by revealing the interdependencies between the magnetic interactions of the individual components and the assembly process parameters. In addition, errors in the caulking process are detected and prevented at an early stage. A magnetic characterization of the soft magnetic components and assemblies before and during their assembly is proposed. Furthermore, the integration of the measurement data into a process control system is intended. The precondition is the development and construction of measuring equipment and systems that can be used to assess the magnetic properties at an early stage of assembly. The assembly can then be adjusted in order to compensate batch fluctuations. As a consequence new materials can be used to eliminate complex annealing processes prior to assembly.

Microtubules are key players in cellular self-organization, acting as structural scaffolds, cellular highways, force generators and signalling platforms. Microtubules are polar filaments that undergo dynamic instability, i.e. transition between phases of growth and shrinkage. This allows microtubules to explore the inner space of the cell, generate pushing and pulling forces and remodel themselves into arrays with different geometry and function such as the mitotic spindle. To do this, eukaryotic cells employ an arsenal of regulatory proteins to control microtubule dynamics spatially and temporally. Plants and microorganisms have developed secondary metabolites that perturb microtubule dynamics, many of which are in active use as cancer chemotherapeutics and anti-inflammatory drugs. Here, we summarize the methods used to visualize microtubules and to measure the parameters of dynamic instability to study both microtubule regulatory proteins and the action of small molecules interfering with microtubule assembly and/or disassembly.

Biocompatible quaternary chitosan/sodium alginate multilayer microcapsules were prepared by layer-by-layer (LBL) self-assembly on the template of monodispersed melamine formaldehyde resin microspheres (MF). The process of self-assembly was monitored by measuring the surface zeta-potential of colloidal particles. The particle size was determined by digital light scattering (DLS) after each deposition, and the average thickness of monolayer film was revealed to be 3.9 nm. Using rhodamine B-labeled quaternary chitosan as the positive polyelectrolyte and sodium alginate as the negative polyelectrolyte, self-assembled multilayer microcapsules with strong red-light emitting were obtained and observed with fluorescence microscope. The fluorescent microcapsules self-assembled from the biocompatible natural polysaccharides may be potentially applied in drug delivery and fluorescence diagnosis.

Purpose – This paper aims to provide an assembly method to improve cylindrical components assembly quality. The proposed method not only could be applied to tolerance allocation, but also could guide the assembly of cylindrical components. Design/methodology/approach – The paper claims to provide a stack-build assembly method using a connective assembly model to take the location and orientation tolerances of a rotor stage into account. Through the separate analysis of the location and orientation tolerances propagation process in the assembly, the quality of the final assembly of the rotationally symmetric cylindrical components assembly could be improved by properly selecting component orientations to minimize the eccentric deviation in the assembly. Findings – The effectiveness of the proposed stack-build assembly technique in improving the tolerance propagation in the assembly of cylindrical components was verified through experiments run with a measuring machine. A real aero-engine rotor was assembly using the proposed method; compared to the direct-build assembly technique, which had the component orientations without consideration, the stack-build assembly technique could be used to reduce the eccentric deviation in cylindrical components assembly by nearly 50 per cent. Originality/value – Different with the old methods, the new method defined the tolerances in detail, such as perpendicularity and angle of the lowest point, and could guide the assembly by the features of surfaces on different components. Through measuring the special tolerances of surfaces on the components, the best assembly angle for each component could be obtained.

The standard tapping machine used for the ASTM and ISO tests does not require the test engineer to measure the input force in the system, instead, just relies on measuring the sound pressure level (SPL) output. However, the input force depends on the assembly itself being tested. The input force levels are lower for lightweight assemblies like hardwood floors as compared to heavyweight assemblies like concrete. Without knowledge of this input force, the output SPL levels cannot and should not be compared using the IIC (Impact Insulation Class) rating. In this work, we measured the input force levels for the same tapping machine on different floors. We also measured the floor impedance for different assemblies and their comparison is also shown. This work shows the importance of measuring input forces for the standard floor-ceiling assembly impact tests

In vitro self-assembled collagen fibrils form a variety of different structures during dialysis. The self-assembly is dependent on several parameters, such as concentrations of collagen and α1-acid glycoprotein, temperature, dialysis time, and the acid concentration. For a detailed understanding of the assembly pathway and structural features like banding pattern or mechanical properties it is necessary to study single collagen fibrils. In this work we present a fully automated system to control the permeation of molecules through a membrane like a dialysis tubing. This allows us to ramp arbitrary diffusion rate profiles during the self-assembly process of macromolecules, such as collagen. The system combines a molecular sieving method with a computer assisted control system for measuring process variables. With the regulation of the diffusion rate it is possible to control and manipulate the collagen self-assembly process during the whole process time. Its performance is demonstrated by the preparation of various collagen type I fibrils and native collagen type II fibrils. The combination with the atomic force microscope (AFM) allows a high resolution characterization of the self-assembled fibrils. In principle, the represented system can be also applied for the production of other biomolecules, where a dialysis enhanced self-assembly process is used.

Assembly performance test bed is an indispensable equipment in the development and production of air compressor assembly. The test bed studied is a mechanical, electronic and hydraulic integrated equipment and its measuring and controlling system mainly consist of computer system, process channel, control object and measuring and controlling software. Guided by the “soft hardware” design conception, two “soft” subsystems was adopted in a one-level computer system to replace the generally adopted two-level “hard” system consisting of measuring and controlling subsystem and management subsystem. This improvement allows the more applications of integrated sensor and intelligent controller, resulting in a simpler hardware and greater openness. The practice indicated that, with rapid parameter adjustment, accurate measuring, large testing scope and high automation, the system meets all the technical indexes of test requirement.

Assembly is one of the most important procedure in manufacturing complex miniature products. Batch assembly is limited by the manual manipulation, for which specially trained technicians is required. In order to automatically assembly certain complex miniature products consisting of multiple parts with different shape and various dimension, and also to reduce the production cost and improve efficiency, an assembly system was developed. Hardware architecture of the system was discussed in detail. Measurement and control strategies in assembling procedure were presented. Local feature extraction and fitting algorithms, reference datum setting were applied to overcome the occlusion. Linear and circular feature fitting from multiple local images of parts, coordinate transformation and the compensation of stages assembly error angle among three local coordinates were adopted to improve the measuring accuracy of vision system. Microscale force sensor was introduced to control the contact force, and the accuracy obtained is less than 1mN. Torque sensor was adopted for sensing the torque during the screw thread assembling, stepping motor and flexible coupler were applied to make the torque smoothly transferred, all of which help precisely measuring and controlling the torque during the M1.4 screw thread assembly.

Multi-station turntable, widely used in industrial automation field, has become an indispensable device in the automation assembly line. The positioning accuracy of traditional mechanical turntable is largely affected by the machining accuracy of Geneva wheel, Ratchet and Cam. The positioning accuracy level has a crucial influence on processing and assembly accuracy, and production efficiency. This paper presents a measuring method to measure positioning accuracy by eddy current sensor. To achieve the positioning accuracy the writer measures the distance between the probe and the measuring block in the circumferential tangent direction. When the turntable turns to a station, the sensor probe is manually adjusted to align it with the measuring block. In this way, the positioning accuracy can be measured conveniently and directly.

A comprehensive performance testing table, on which 9 testing items can be done, of the constant velocity joint drive shaft assembly is developed. The principle of the system is introduced and the mechanical structure and the computer measuring and control system of it are discussed particularly. Some intelligent measuring methods, such as fuzzy method, are used in the system and an example of testing the slip curve characteristic of the joint assembly by fuzzy measuring method is presented in detail, including its membership function and fuzzy rules of fuzzy reasoning. The slip curve measured by traditional method compared with that by fuzzy method. The experimental results show that the measurement precision by fuzzy method is more accurate than that by the traditional one.

We have developed methods to study these events using tethered lipid bilayers, which can be probed by impedance spectroscopy and surface plasmon resonance. The use of wireless devices has greatly increased in the last decade, and it has been one of the most widely used medium of information transmission. Within the wireless devices the wireless sensor networks are the most contemporary and most commonly researched field. The work deals with the industrial use of wireless sensor networks and more precisely with monitoring and controlling industrial assembly lines. The focus of this study is localization by the use of wireless technology in the above mentioned environment. In the experiment wireless sensors are placed on the base elements of currently being assembled products. The developed system is able to specify the precise place of the product in the assembly line and record the time of localization. By the use these information the time of assembling the product can be monitored. For determining the place of the product the Received signal strength indication – RSSI has been used. The current position of the product is calculated by a neural network. The use of these sensors makes possible the measuring and recording of the influences on the product during the assembly, such as the effects of temperature, humidity, or if the product has been hit or damaged. By the use of these wireless sensor networks the quality of the assembled products can be improved and the process of assembly can be optimized.

The article discusses the method for interchangeability method. To reduce the volume of work in progress, the design of a fixed combined compensator is developed, which provides stepwise regulation. The article reducing the work in progress by stepwise regulation in multilink dimension chains when assembled using the group proposes a rational technology for the use of combined compensator, taking into account the peculiarities of the group interchangeability method. In order to reduce the work in progress during assembly using the group interchangeability method, two ways of using the combined compensator have been developed: 1 - the inclusion of a compensator in each dimensional chain collected by the group interchangeability method; 2-application of the compensator only at the stage of formation of work in progress during group assembly by replacing one of the parts with it. The peculiarity of the methods is the use of the informational advantage of group assembly — information about the actual dimensions necessary for sorting parts into groups. This eliminates the step of measuring the closing link during assembly and simplifies the design of the compensator, reducing the number of control steps.

AbstractDeveloping an implantable, telemetric pressure measuring system for venous applications makes a high degree of miniaturization necessary. Thus the influence on the measurement environment is minimized and the risk of thrombosis at small flow blood velocities is decreased. But these systems are limited in terms of accuracy and resolution. The asked system requirements could only be reached by optimising the assembly and encapsulation techniques. To achieve the high degree of miniaturization numerical simulations were performed on the shape and size of the implant and led to the development of a specific metal housing consisting of two main components. A small measuring chamber will be placed into the portal vein and is rigidly fixed to a flat circular part that contains the pressure sensor chip and a transponder board and will be located outside on top of the vein. The main focus of the assembly process was based on a stress-free design and mounting of the components.

To address the problems of low efficiency, high cost and low automatic level existing in traditional laser tracking measurement systems, a laser tracking-vision guiding measurement system for large-scale parts assembly is introduced in this paper. The system is composed of mono-camera and a laser tracker, can fulfill real-time tracking and automatic measurement throughout the whole assembly process. A global calibration method based on public planes and a method for finding the 3D positions of the target reflectors based on monocular vision measuring are expounded. Mathematical model is established, measurement system is built, and the experiment is accomplished. The experiment result indicates that the proposed system features relative high degree of automation and high measuring speed, has future in practical application.

The effect of the tooth contact and alignment error of the hypoid gear assembly on transmission error was investigated with a new measuring apparatus which can measure the transmission errors of loaded hypoid gears assembled into a final drive unit. Measurements indicate that transmission error predictions made with the TCA and LTCA — analytical tools developed by Gleason Works for a no-load and loaded state, respectively — have sufficient accuracy when actual data on the gear tooth surface and alignment error of the gear assembly are used in the calculations. A systematic examination has also been made of the effects of tooth contact and gear assembly alignment error on transmission error on the basis of the LTCA calculations. It was found that the transmission errors relative to the applied load varied not only according to the tooth contact but also the no-load transmission error of the gears. This relationship was also examined by taking into account the effects of the gear dimensions. It was confirmed through calculation and experiment that a small module design was effective in reducing transmission error.
