Der volle Inhalt der QuelleCaenorhabditis elegans is a model organism commonly used to study developmental processes such as vulva formation. This process relies on the precise fate induction of the cells that give rise to the vulva. The resultant cellular fates are arranged into a characteristic pattern, which is conserved across the vulva development of Caenorhabditis and rhabditid nematodes. In contrast to this extensive conservation of the vulval cell fate pattern, my thesis presents two lines of study that revealed signatures of evolution in developmental pathways controlling vulval induction. In the first one, I present a turnover of cis-regulatory motifs required for the cell-specific expression of lin- 3 (EGF), a gene coding for the signalling molecule which induces vulva cell fates. The three transcription factor binding sites required for lin-3 expression in C. elegans are observed only in species of the Elegans group. Using single-molecule FISH and genome editing techniques, I show that the replacement of the endogenous C. elegans binding sites by a 58 bp fragment containing a single site (coming from C. angaria), produces wild-type mRNA levels of lin-3. I conclude that compensatory evolution for the requirement of C. elegans binding sites occurred in cis. In the second line of study, I used a mapping-bysequencing approach and a collection of vulva mutants in Oscheius tipulae, a neighbour of C. elegans in the rhabditid clade, to identify mutations in genes acting on vulva cell fate pattern specification. The only collected mutation to render animals vulvaless corresponds to a deletion of the cis-regulatory region controlling the expression of O. tipulae lin-3, which I confirmed by CRISPR/Cas9 genome editing. Mutants with an excess of vulva induction carry molecular lesions in the plexin/semaphorin pathway, a cell signalling system which I show to act in the antero-posterior positioning of the cells giving rise to the vulva by microscopic analysis. Finally, other O. tipulae animals with disrupted vulval cell fate pattern hold mutations in components of the Wnt pathway; for this reason, I investigated the expression profiles of Wnt ligands of O. tipulae and revealed their conservation with C. elegans. Der volle Inhalt der QuelleFlaxseed is known for their high oil content, omega 3, antioxidants and fiber. To meet growing demand, this requires controlling their production. The aim of this work is to study the effect of environmental factors and cultivar on linseed composition and subsequent pressing process. Ten linseed cultivars were grown in two different locations (north and south of France) during years 2011, 2012 and 2013. Every year have been characterized in terms of pluviometry, sunshine hours and degree days, each climatic factor has been cumulated from flowering to harvest. A strong impact of cultivar on oil, C18:3 and starch content has been noticed whereas the growing location has only affected the starch and lipid contents. Year effect has been recorded on thousand kernel weight, proteins, oil, C18:3 and starch content. Oil expression using the seeds previously characterized has exhibited a strong correlation between protein content and oil yield. To improve the pressing process, dehulling flax seeds before pressing was considered. The study of the pressing flax seeds on hydraulic and continuous press shows the possibility of pressing the dehulling flaxseeds. The concordance between the pressing performance at the laboratory scale and pilot scale was highlighted for the whole and dehulled seeds. This could be suggests a possible transposition to the industrial scale

The class A and B synthetic multivulva (synMuv) genes of C. elegans redundantly antagonize an EGF/Ras pathway to prevent ectopic vulval induction. The class B synMuv genes encode many proteins known to remodel chromatin and repress transcription. The class A synMuv genes likely also function in transcription, although their specific molecular functions are unknown. We have identified a class A synMuv mutation in the promoter of lin-3 EGF, revealing that lin-3 is the key biological target of the class A synMuv genes in vulval development. Using FISH with single mRNA molecule resolution, we found that class AB synMuv double mutants exhibit widespread ectopic lin-3 expression. Our results show that lin-3 EGF is normally expressed in the germline, and many class B synMuv genes have previously been implicated in inhibiting germline fates in somatic cells. We propose that the class A synMuv genes specifically repress ectopic lin-3 EGF expression through a site in the lin-3 promoter and the class B synMuv genes either directly or indirectly repress lin-3 as a consequence of their role in regulating the germline/soma distinction. The class A and B synMuv genes had previously been thought of as two parallel pathways, but we have found that each of those pathways is actually composed of multiple parallel pathways. While class AB synMuv double mutants have a strong Muv phenotype, most class AA synMuv double mutants exhibit a weak Muv phenotype, and most pairs of class B synMuv mutants can enhance each other in sensitized backgrounds, indicating that most genes within a class can function in parallel. We also found that some pairs of synMuv genes cannot act in parallel, indicating that they function together to repress ectopic lin-3 expression. We also report the molecular characterization of the class A synMuv gene lin-38 and the identification of mcd-1 as a class A synMuv gene. lin-38 and mcd-1 encode paralogous zinc-finger proteins. Unlike previously studied class A synMuv genes that function specifically in vulval development by repressing lin-3, both lin-38 and mcd-1 control multiple aspects of development by regulating target genes other than lin-3.

Der volle Inhalt der QuelleFlaxseed oil contain high amount of oméga-3 and present different industrials applications and human health benefits. ln this work, we aimed to identify enzymes allowing the accumulation of high level of omega-3 in linseed plants. ln this context, we evaluated the role of Lysophophatidic acid acyltransferases (LPAAT) and Phospholipases A2 (PLA2) enzymes in the accumulation of omega-3 by in vitro and in vivo approaches. Results showed the presence of a LPAAT2A isoform into flax genome having a high specificity and selectivity toward omega-3. Expression of Lpaat2A and sPia2a gene in Arabidopsis seeds increase seed weight, oil production and omega-3 content (up to 10% and 11%) in transformant seeds respectively. These results showed the presence in linseed plant of an efficient enzymatic system toward omega-3 accumulation. Furthermore, we have evaluated in vitro and in vivo the potential of Litchi and flax LPAAT enzymes toward the production of cyclopropane fatty acids (CFA). These fatty acids are naturally produced by a specific enzyme «CFA synthase» with physico-chemical properties interesting to lubricant and cosmetics industries. ln this context, we have generated Arabidopsis lines expressing E. coli Cfa synthase with or not co-expression of litchi Lpaat gene. Result showed an increase of 25% in the content of CFA in transgenic line co-expressing Cfa synthase and litchi Lpaat comparing to the transgenic lines expressing the CFA synthase. This increase in the level of CFA in transgenic seeds reflects the specificity of Litchi LPAAT toward CFA

Der volle Inhalt der QuelleLinoleic (LA) and α-linolenic (ALA) acids are two polyunsaturated fatty acids. They are essential for humans because we are unable to synthesize them in situ. These two fatty acids are the representatives of omega-6 (LA) and omega-3 (ALA). Although they are essential and therefore necessary, the issue of the omega-6 / omega-3 ratio between these two families is also important to take into account in the diet. The consumption habits of the French over the past thirty years have shifted the balance against omega-3s compared to omega-6s. Or a rebalancing of the omega-6 / omega-3 balance tends to reduce cardiovascular risks. Linseed and camelina oils are endemic crops in France extremely rich in ALA. They are the most suitable for supplementation with omega-3 of plant origin. However, the high contents of polyunsaturated fatty acids in these oils are responsible for their low oxidative stabilities. The consumption of oxidized oils or fats is not recommended because they can contribute toxic molecules. The aim of this project was to improve the shelf life of oils rich in polyunsaturated fatty acids by enhancing the antioxidant extracts of the cakes obtained after crushing the seeds. Among the molecules studied, gallic acid, caffeic acid, propyl gallate as well as pinoresinol and lariciresinol purified from flax showed a real interest in slowing down oxidation, while secoisolariciresinol was ineffective

We surmise that the only simple Lie 2-algebra of this type is W(1, 3). So, our main objective is to study the truthful of this conjecture for these algebras of small dimensions. As a result, we prove that this conjecture is true for all these algebras less than or equal to 16 dimension, and also in some special cases when the dimension is 17.

Der volle Inhalt der QuelleAn even cycle decomposition (ECD) of an Eulerian graph is a partition of the edge-set into even cycles. We color the even cycles so as two cycles sharing at least one vertex receive distinct colors. If m is the minimum number of required colors, then we say that the even cycle decomposition has index m. The notion of an ECD of index m is connected to the palette index of a graph, a chromatic parameter describing a graph by the minimum number of palettes of its vertices. In particular, the possible values for the palette index of a 4-regular graph are 1, 3, 4 and 5. It is 3 if and only if the graph has an even 2-factor or an ECD of index 3. There exist infinite families of 4-regular graphs with an ECD of index 3 . As far as we know, no example of 4-regular graph whose edge set can be partitioned into even cycles and every ECDs has index larger than 3 is known. Motivated by the problem on the existence of such a 4-regular graph, we study ECDs in 4-regular line graphs of class 2 cubic graphs. For some of the infinite families of class 2 cubic graphs that are characterized by an arbitrary large oddness, we can find an ECD of index 3 in the corresponding line graph.

Syftet med arbetet var att ta fram en flödesanalys över vilka montage som krockar flödesmässigt vid en förflyttning av lina 3. En undersökning av olika möjligheter att tilta/vrida hytten gjordes för att hitta en bra arbetsergonomi åt montören. En flödesanalys över vilka montage som krockar vid en förflyttning av lina 3 togs fram med hjälp av processflödesanalys. I slutet av arbetet gjordes ett förslag över vilka montage som bör flyttas till de olika monteringslinorna. Med de förslag som gjorts angående hur man kan vrida hytten, samt vilka montage som ska flyttas till diverse monteringslinor kan man genomföra en omorganisation av lina 3.

Transit agencies face a variety of challenges, from increasing ridership to changes in infrastructure, to system expansions, all of which require significant preparation to accommodate the changes without affecting passengers or the agency negatively. The MBTA Green Line, a Boston light rail system serving nearly 230,000 average weekday passengers, will be undergoing major changes in the next two decades, including nearly doubling of ridership and system expansion. In order to prepare for these changes, measures need to be taken to increase capacity and plan for operations on the new segment. Starting in Fall 2010, the MBTA added to a 2-car train operation, and subsequently increased, a number of 3-car trains on three of the four Green Line branches, in order to begin to address the capacity issue. This thesis analyzes service performance before and after implementation of 3-car trains to find that although scheduled capacity increased slightly, the actual capacity of the system remained constant during the morning peak period and decreased during the evening peak period. Furthermore, there were some negative impacts with respect to passenger waiting time and running times, thus worsening the overall passenger experience. However, since 3-car trains will be required for increasing capacity on the Green Line, it is recommended that trials of 3-car trains continue, with the restriction that only 2- or 3-car trains operate on a branch. Furthermore, field observations at terminal stations on two of the branches show differences in operations management practices, which help explain some variability in service along the route, and point to strategies to improve service reliability.

Resumo: A invasão do endométrio pelo trofoblasto extraviloso é fundamental no desenvolvimento do feto e da placenta, processo este controlado por fatores ligados à atividade imunológica e hormonal que, quando alterada, pode resultar em interrupção da gestação e/ou geração das chamadas doenças trofoblásticas gestacionais. Em algumas situações, pode haver evolução para o coriocarcinoma, neoplasia maligna do trofoblasto, em que há evidências da atuação das moléculas ligadas ao processo de fusão celular e inflamação. Porém, os estudos neste tema são incipientes e inconclusivos. Considerando essas informações, o objetivo deste trabalho é estudar de forma comparativa a expressão das citocinas LIF, IL-6 e do ativador de transcrição STAT-3, além da telomerase, em material de aborto, de placenta normal a termo e de coriocarcinoma. Métodos: a expressão destas moléculas foi avaliada pelos métodos: imunoistoquímica (IHQ), imunofluorescência (IF), Western Blotting (WB) e Real-Time PCR (RT-PCR), em amostras de material de aborto, placenta normal a termo e coriocarcinoma (N=12 cada um). Os ensaios de WB e Real-Time PCR empregaram material a fresco de placenta normal a termo e seu cultivo celular e cultura da linhagem BeWo. Resultados: no material de aborto, as reações de IHQs evidenciaram expressão moderada de IL-6 em 58,4% dos casos e intensa de STAT-3 em 33,3%. Na placenta normal, observou-se intensa marcação de IL-6 em 50% e de STAT- 3 em 16,7% dos casos, enquanto que, no coriocarcinoma, houve expressão intensa de IL-6 em 50% e de STAT-3 em 75% dos casos. Por outro lado, as reações para LIF tiveram expressão nula em todos os três grupos. Pelo WB houve expressão proteica de IL-6 apenas no material fresco de placenta normal e ausência de expressão na sua cultura primária e na linhagem BeWo; LIF não foi expresso em todos os grupos estudados. STAT-3 foi detectado no citoplasma em todos os grupos, entretanto, a expressão nuclear da STAT-3 fosforilada (pSTAT-3) não foi observada na IF e nem pelo WB. Na análise gênica pelo RTPCR houve forte expressão de IL-6 e STAT-3 no material fresco de placenta normal e expressão muito fraca na cultura primária de placenta normal e na linhagem BeWo; a expressão de LIF foi muito fraca em todos os grupos. Apenas a linhagem BeWo demonstrou forte expressão gênica da telomerase, contrastando com a completa falta de expressão no material fresco de placenta normal e em sua cultura primária. Conclusão: A intensa expressão IHQ de IL-6 e STAT-3 no coriocarcinoma indica a atuação de ambas na carcinogênese. A expressão proteica de IL-6 no material fresco de placenta normal e sua ausência no material de cultura primária e na linhagem BeWo pode ser ocasionado pelo contato célula-a-célula nas culturas aderentes, inibindo o crescimento celular e, consequentemente, as vias de sinalização. A falta de expressão da pSTAT-3 tanto na IF como por WB demonstra que a via JAK-STAT está sendo desativada. A ausência de expressão de LIF, em todos os métodos estudados, sugere que esta citocina poderia estar sendo inibida por meio de proteínas SOCS3 ou, atuando, de modo indireto, na proliferação celular do coriocarcinoma. O aumento da atividade da telomerase nas células BeWo reforça sua relação com o fenótipo maligno e a aponta como um bom marcador para progressão da doença
Abstract: The invasion of the endometrium by extravillous trophoblast is a fundamental process in the growth of the fetus and placenta. The process is controlled by factors related to the immune and hormonal activity that, when changed, may result in termination of pregnancy and development of so-called gestational trophoblastic diseases. In some cases, changes can result in malignancy, in which some molecules play a role in cell fusion process and inflammation, although studies in this area are inconclusive. Considering this information, the study had the aim of investigating the expression of cytokines LIF, IL-6, STAT- 3 and the function of telomerase to understand their participation in abortion, in normal at term placenta and choriocarcinoma. Methods: The expression of the molecules was assessed by immunohistochemical assay (IHC), immunofluorescence (IF), Western Blotting (WB) and Real-Time PCR (RT - PCR) using fixed material from biopsies of abortions, normal at term placentas and choriocarcinoma along with fresh tissue of normal at term placenta and their primary culture and BeWo cell line. Paraffin embedded material used in IHC and IF assays were obtained from the Department of Pathology files. Tests of WB and Real-Time PCR employed fresh material, obtained from cell cultures of normal at term placenta and the BeWo line. Results: IHC reactions to abortion biopsies showed moderate staining for IL-6 in 58.4% of cases and intense for STAT-3 in 33.3 % of cases. In biopsies of normal placenta, there was intense reaction for IL-6 in 50% of cases, intense for STAT-3 in 16.7%; choriocarcinoma showed intense staining for IL- 6 in 50% of cases and also for STAT-3 in 75% of cases. On the other hand, LIF expression was missing in all three groups. WB analyses showed IL-6 protein in fresh material from normal placentas, but no expression in placenta primary cultures and BeWo line. LIF was absent in all groups. Cytoplasmic STAT-3 was observed in all groups, while the nuclear expression of phosphorylated STAT-3 was absent. On gene analyses a strong expression of IL-6 and STAT- 3 was observed from fresh normal placenta, but very weak expression in primary cultures of normal placenta and BeWo cell line. LIF expression was very weak in all groups. In regard to the gene expression of telomerase, it was strong in the BeWo line which contrasted with its complete lack of expression in fresh normal placenta and its primary culture. Conclusion: The high expression of IL-6 and STAT-3 in biopsies of choriocarcinoma indicates the role of both in tumor progression. Regarding protein expression, the presence of IL-6 in the material from fresh normal placenta, and its absence in primary culture and BeWo line may be caused by the cell-to-cell contact cultures by inhibiting cell growth and thus signaling pathways. However, the lack of expression of phosphorylated STAT-3 whether through IF or WB shows that its JAK-STAT pathway is inhibited. Lack of expression of the LIF suggests that it might be involved indirectly in choriocarcinoma cell proliferation or be inhibited by SOCS3 protein. Moreover, the increased telomerase activity of BeWo cells enhances their relation to the malignant phenotype and indicates a good marker for disease progression

Der volle Inhalt der QuelleIn the first part we present the Weyl algebra and our results concerning its finite-dimensional Lie subalgebras. The second part is devoted to a more exotic algebraic structure, the Lie algebra of order 3. We set the basis of a theory of deformations and contractions of these algebraic structures. We then concentrate on a particular such Lie algebra of order 3 which extends in a non-trivial way the Poincaré algebra, this extension being different of the supersymmetric extension. We then focus on the construction of a field theoretical model based on this algebra, the cubic supersymetry (3SUSY). For this purpose we obtain bosonic multiplets with whom we construct invariant Lagrangians. We then study the compatibility between this new symmetry and the abelian gauge symmetry. Furthermore, the analyse of possible interactions shows that interactions terms are not allowed by the 3SUSY invariance. Finally we establish results regarding the extension in arbitrary dimensions of our model

Technological advances in portable devices imply not only the development of devices but also the need to enhance their power sources. For instance, cellular phones with high technology incorporating Bluetooth, Infrared, Digital Camera, Games, GPS, Internet Browsing, etc., need high performance rechargeable batteries. Lithium ion-conducting glass-ceramics with NASICONtype structure, based on the Li1+xMxM'2-x(PO4)3 system, where M = Al, Cr, Fe, Ga and M' = Ge, Ti, Hf, are considered good candidates as solid electrolytes in lithium ion batteries due to their chemical stability, easy fabrication and high ionic conductivity. In this work, the Li1+xAlxTi2-x(PO4)3 glass composition was synthesized by the traditional method known as Splat Cooling. Glass-ceramics with the same composition as the precursor glass but with different microstructures were obtained by controlled crystallization, using single and double heat treatments. The parent glass was characterized by chemical and thermal analysis and the latter enabled tailoring of the crystallization heat treatments. X-ray diffraction (XRD) results indicated that the resulting glassceramics exhibit the desired NASICON-type structure as primary phase, as well as the segregation of insulating phases, i.e., AlPO4 and/or TiO2, which do not affect the electrical properties. An examination of the microstructure by scanning electron microscopy (SEM) and measurements of electrical conductivity by impedance spectroscopy (IS), combined with the XRD analysis, indicated that the electrical conductivity of single heat-treated samples increases with the heat treatment temperature and hence with the increase in crystallinity. Samples synthesized by double heat treatment showed increasing electrical conductivity with decreasing nucleation treatment time and consequently with increasing average grain size.
Os avanços tecnológicos em dispositivos portáteis trazem outro aspecto que chama a atenção, tanto ou mais importante do que os próprios dispositivos desenvolvidos, i.e., a necessidade de fontes de alimentação mais poderosas. Por exemplo, telefones celulares com tecnologia de ponta que incorporam Bluetooth, Infravermelho, Câmara fotográfica, Jogos, GPS, Navegação na Internet, etc., requerem baterias recarregáveis de alto desempenho. Vitrocerâmicas condutoras por íon lítio com estrutura tipo NASICON, concretamente baseadas no sistema Li1+xMxM 2-x(PO4)3, onde M = Al, Cr, Fe, Ga e M = Ge, Ti, Hf, são consideradas boas candidatas como eletrólitos sólidos em baterias de íon lítio devido à sua estabilidade química, facilidade de fabricação e alta condutividade iônica. Neste trabalho foi obtido o vidro de composição Li1.2Al0.6Ti1.6(PO4)2.9 usando o método tradicional de resfriamento rápido conhecido como Splat Cooling. Vitrocerâmicas de composição igual à do vidro matriz, com diferentes microestruturas, foram obtidas por cristalização controlada mediante tratamentos térmicos simples e duplos. O vidro precursor foi caracterizado por análise química e térmica. Esta última permitiu projetar os tratamentos térmicos de cristalização. Os resultados de difração de raios X (DRX) mostraram a obtenção da estrutura tipo NASICON como fase principal, com presença das fases isolantes AlPO4 e/ou TiO2, que não afetam as propriedades elétricas. O estudo da microestrutura e da condutividade elétrica, usando microscopia eletrônica de varredura (MEV) e espectroscopia de impedância (EI), respectivamente, evidenciaram o aumento da condutividade com o crescimento do tamanho médio do grão, atribuído ao maior contato entre grãos e, provavelmente, a um melhor alinhamento dos canais na estrutura do LiTi2(PO4)3. A máxima condutividade de 1,3 x 10-3 S/cm foi atingida na amostra obtida mediante tratamento térmico simples em 1000 °C durante 20 minutos a qual apresentou um tamanho médio de grão de 350 nm.

This work reports the synthesis of zeolites with different compositions (pure silica, Si/Ti and Si/Al), via hydroxide and fluoride medium using the cation 1-butyl-3- methylimidazolium as structure directing agent. Initially, the cation was synthesized in chloride form and used for the synthesis in hydroxide medium. An anion-exchange (Cl- for OH-) was required for the synthesis in fluoride medium. Different reactants were used for the formation of gels synthesis, resulting in the crystallization of MFI and TON phases, the latter predominant in many compositions. The cation and synthesized zeolites obtained were characterized by different techniques such as NMR, TG/DTG, XRD, SEM, N2 adsorption and desorption, DRS and EPMA. Besides characterizing the cation and zeolites, the mother liquor of hydroxide synthesis was characterized and it was possible to observe a modification of the cation in the synthesis conditions employed. The materials synthesized in this work can be applied in catalytic reactions and adsorption
Este trabalho relata a s?ntese de ze?litas de diferentes composi??es (puramente sil?cicas, Si/Ti e Si/Al), via meio b?sico e fluor?drico, utilizando o c?tion 1-butil-3- metilimidaz?lio como agente direcionador de estrutura. Inicialmente o c?tion foi sintetizado e utilizado na forma de cloreto para as s?nteses em meio b?sico e uma troca ani?nica (Cl- para OH-) foi necess?ria para as s?nteses em meio fluor?drico. Diferentes reagentes foram empregados para a forma??o dos g?is de s?ntese, resultando na cristaliza??o das fases MFI e TON, esta ?ltima predominante em muitas composi??es de gel testadas. O c?tion sintetizado e as ze?litas obtidas foram caracterizados por diferentes t?cnicas, como RMN, TG/DTG, DRX, MEV, adsor??o e dessor??o de N2, DRS e EPMA. Al?m de caracterizar o c?tion e as ze?litas, a ?gua-m?e das s?nteses em meio b?sico foi caracterizada e foi poss?vel verificar uma modifica??o do c?tion nas condi??es de s?ntese empregadas.
