Longos trechos contínuos em homozigose (LCSH) são regiões ininterruptas de alelos homozigotos originados por consanguinidade, dissomia uniparental (UPD), mecanismos de reparo de DNA e haplótipos ancestrais. Identificamos regiões genômicas de LCSHs através do CMA, avaliando suas frequências e mecanismo de origem em pacientes com deficiência intelectual e/ou transtorno do espectro autista do serviço público de saúde do Brasil Central. Em 90% dos pacientes, pelo menos um LCSH foi identificado com um total de 265 LCSHs observados. De 265 LCSHs, 59% eram recorrentes, observados nos cromossomos 11, 16 e X, e 41% eram não recorrentes e observados nos cromossomos X e autossomos, exceto no cromossomo 19. LCSHs não recorrentes foram originados por consanguinidade em 9,7% dos pacientes, por possível reparo de DNA ou mecanismos de haplótipos ancestrais em 85,4% dos pacientes e por possível UPD segmentar em 4,9% dos pacientes. Nossos achados mostraram a utilidade do CMA de alta densidade de SNPs na identificação de LCSHs, destacando a importância de caracterizar o perfil de LCSHs na população brasileira, especialmente na população do Brasil Central.

Longos trechos contínuos de homozigose (LCSH) são segmentos cromossômicos neutros, resultantes de homozigose cromossômica ou segmentar (HMZ). Os LCSH podem ser detectadas por plataformas de hibridização genômica comparativa por arrays (CGH array) através de sondas específicas para polimorfismo de um único nucleotídeo (SNP). Dificilmente serão realizados exames de CGH array de base populacional no Brasil, para inferir polimorfismos comuns. Portanto, neste trabalho pesquisamos as LCSH em exames de CGH array de pacientes com distúrbios do desenvolvimento (DD) para identificar algumas das variações comuns. Metodologia: Foram analisadas as citobandas que mais frequentemente apresentaram regiões com LCSH em 374 exames de CGH array (Affymetrix CytoScan® HD ou 750k) de pacientes com DD, cujos responsáveis assinaram o termo de consentimento. Os exames foram solicitados por médicos geneticistas e neurologistas dos Hospitais Infantil e Universitário e de clínicas particulares de Florianópolis, e realizados pelo laboratório Neurogene no período de 2013 a 2016. Foram considerados LCSH apenas quando eram superiores a 3Mbp e nos cromossomos autossômicos e frequentes apenas as que apareceram em mais de 5% dos exames, já que estamos lidando com uma população onde o índice de 1% não seria aplicável. Resultados: Dos 374 exames analisados, 4% não apresentaram nenhum LCSH >3Mbp nos cromossomos autossômicos. A região mais envolvida com a homozigose foi a 16p11.2 (59%)[ (~31,451,698-35,220,544) tamanho médio de 3,5Mbp], essa região é conhecida por estar envolvida em rearranjos cromossômicos devido à presença de flanqueamento de duplicações segmentares. Também foi encontrada homozigose frequente em 11p11.2 (18%), 3p21.31(14%), 15q15.1(10%), 1q21.1(10%), 1q21.2(7%), 2q11.1(6%), 10q22.1(6%) e 1p33(5%). Não há genes envolvidos com imprinting nessas regiões [http://www.geneimprint.com], sugerindo que as mesmas podem ser consideradas variações comuns. Outras 300 regiões genômicas com LCSH apresentaram menores frequências e características que podem ser exploradas para interpretações etiológicas, podendo evidenciar genes candidatos a mutações autossômicas recessivas; um grau de consanguinidade; dissomia uniparental; recombinações cromossômicas ou rearranjos. Conclusão: Relatamos uma série de LCSHs que estão presentes em frequência acima de 5% em indivíduos com o neurodesenvolvimento afetado, sugerindo que provavelmente sejam variações normais na população de SC. Esta informação é importante para a interpretação de achados nos exames de CGH array.

This paper analyses and compares the treatment of sexual health in Library of Congress Subject Headings (LCSH) and Répertoire de vedettes-matière de l’Université Laval (RVM) using three of Bowker and Star’s (1999) infrastructural inversion techniques: practical politics, convergence, and resistance. Our findings reveal that neither LCSH nor RVM offer a holistic representation of sexual health (practical politics), that LCSH’s topical representation of sexual health limits access to relevant material (convergence), and that the enhancement of LCSH through user-added content could improve but not replace these systems (resistance).

One of the most important tasks of a librarian is the assignment of appropriate subject(s) to a resource within a library’s collection. The subjects usually belong to a controlled vocabulary that is specifically designed for such a task. The most widely adopted controlled vocabulary across libraries around the world is the Library of Congress Subject Headings (LCSH). However, there seems to be a shifting from traditional LCSH to modern thesauri. In this paper, a methodology is proposed, capable of incorporating thesauri into existing LCSH-based Information Retrieval–IR systems. In order to achieve this, a mapping methodology is proposed capable of providing a common structure consisting of terms belonging to LCSH and/or a thesaurus. The structure is modeled as a Simple Knowledge Organization System (SKOS) ontology, which can be employed by appropriate subject-based IR systems. As a proof of concept, the proposed methodology is applied to the DSpace-based University of Piraeus digital library.

Os transtornos do desenvolvimento neurológico podem ocorrer por variações no número de cópias, rearranjos cromossômicos, mutações pontuais e alterações epigenéticas. Além disso, longos trechos contínuos em homozigose (LCSH) podem influenciar no desenvolvimento de desordens neurológicas. Mecanismos responsáveis pelo surgimento de longos trechos contínuos de homozigose compreendem dissomia uniparental (UPD), homozigose ancestral e consanguinidade dos pais. No presente estudo relatamos o caso de três garotas encaminhadas ao Laboratório de Citogenética Humana e Genética Molecular – LaGene/Lacen/SES, com indicação clínica de atraso no desenvolvimento neuropsicomotor, que foi observada a presença de LCSH no cromossomo 11 através da técnica de Análise Cromossômica por Microarranjo. A citogenética convencional com bandeamento GTG usando o software IKAROS® (Metasystems Corporation, Alemanha) mostrou cariótipo do sexo feminino (46,XX) para as três meninas. O DNA genômico das três pacientes foi isolado usando o kit GE HealthCareIllustraTMbloodGenomicPrep Mini Spin. Em seguida, a Análise Cromossômica por Microarranjo usando o GeneChip®CytoScanHDTM da Affymetrixe filtro de análise para LCSH > 5000 pb, revelou para as três pacientes uma LCSH com tamanho em torno de 5,27 Mb na região cromossômica 11p11.2 abrangendo 31 genes mórbidos. A LCSH foi observada em uma região do cromossomo 11 que é classificada como de retardo mental autossômico recessivo (OMIM614344), que já foi relatada para uma família de indivíduos consanguíneos que apresentavam severa deficiência intelectual. Ademais, a região 11p11.2 está relacionada com a Síndrome de Potocki-Shaffer (OMIM601224), classificada como uma síndrome rara de deleção de genes contíguos devido à haploinsuficiência da região 11p12-p11.2, sendo caracterizada fenotipicamente por atraso no desenvolvimento, deficiência intelectual e anormalidades craniofaciais. É importante que seja feito estudo de expressão dos genes, presentes na região em homozigose, para observar o comportamento gênico e a correlação com o fenótipo clínico da deficiência intelectual observado nas pacientes. Além disso, é recomendado que estas famílias façam o aconselhamento genético com o objetivo de tentar entender o mecanismo que gerou a formação de longos trechos em homozigose, fazer o cálculo de risco de recorrência e proporcionar uma melhor qualidade de vida para estas famílias.

This paper provides an analysis of data collected on the continued prevalence of outdated, marginalizing terms in contemporary cataloguing practices, stemming from the Library of Congress Subject Heading term “Indians” and all its related terms. Using Manitoba Archival Information Network’s (MAIN) list of current LCSH and recommended alternatives as a foundation, we built a dataset from titles published in the last five years. We show a wide distribution of LCSH used to catalogue fiction and non-fiction, with outdated but recognized terms like “Indians of North America-History” appearing the most frequently and ambiguous and offensive terms like “Indian gays” appearing throughout the dataset. We discuss two primary problems with the continued use of current LCSH terms: their ambiguity limits the effectiveness of an institution’s catalog, and they do not reflect the way Indigenous Peoples, Nations, and communities in North America prefer to represent themselves as individuals and collectives. These findings support those of parallel scholarship on knowl­edge organization practices for works on Indigenous topics and provide a foundation for further work.

Abstract Abstract 4692 Allogeneic hematopoietic cell transplantation (HCT) is the most effective curative therapy in patients acute myeloid leukemia (AML) and myelodisplastic syndromes (MDS). Despite improvements in the clinical management, relapse from the original disease is still one of the main causes of treatment failure. There is scarce available data on the influence of genomic changes in relapsed patients after HCT, in particular in HLA identical transplantation. Selective pressure mediated by T cells can result in genomic changes in the HLA region on chromosome 6, as shown by several animal models and in human haploidentical transplantation. In this study we examined genome wide DNA copy number changes and long contiguous stretches of homozygosity (LCSH) in 21 patients with a diagnosis of AML or MDS/AML undergoing HCT. SNP-chip analysis revealed a total of 162 genomic aberrations (GA) in 21 pairs of samples before allogeneic transplantation and at relapse in patients diagnosed with AML or AML/MDS. Total GA were significantly higher after transplant compared with the pre transplant samples (p<0.05) only in the group of patients with normal cytogenetics. Within this last group of patients, only a small deletion was detected in the pre transplant sample not detected by conventional cytogenetics. In contrast, in the group of patients with abnormal/complex karyotype, 5 additional deletions and 2 duplications were detected in 5 patients before transplantation. The median number of GA per patient pre transplant was 3 (range 0–19), while the median number of GA at relapse post transplantation was 5 (range 1–20). The median copy number loss per patient was 1.6 before transplant and 2.8 at relapse, while the median number of LCSH at the same time points was 2 and 2.8 respectively. Copy number gains were significantly less common (p<0.05) with a median of 0.5 per patient before transplant and 1 at relapse. Of 21 patients, eleven (52%) shared the same GA before transplant and at relapse indicating common clonality. Conversely 26 novel deletions, 9 LCSH and 6 duplications arose at relapse detected in 17 patients (76%). Of interest 3 out of 9 of these new LCSH involving chromosomes 6p, 20q and 22q, were recurrent GA at relapse detected in 2, 5 and 4 patients respectively. HLA typing of the blasts from patients with LCSH in chromosome 6p revealed a loss of the patient specific band at the mismatched locus leading to homozygosity for the HLA haplotype shared by the patient and the donor. LCSH was the most frequent lesion present in 81% of the patients followed by deletions (57%), trisomies (48%) and duplications (24%). All chromosomes had one or more GA, with significant differences among the 22 autosomes. Chromosome 2 had only 2 GA in the post transplant sample while chromosome 17 had 9 in the same time point. The most frequent lesion was LCSH in chromosome 20 q11.21 in the relapse sample, occurring in 5 of 21 patients (24%). Another recurrent GA was a deletion in chromosome 12 p13 detected in 4 patients (19%) at relapse. To confirm the SNP-chip results detected in allogeneic transplanted patients, extensive validation analysis was performed. FISH signal for probes TP53 (17q13.1), ETV6 (12p13) and ATM (11q 22.3) revealed one signal. These regions also showed hemizygous deletion by SNP-chips analysis. D8Z2 probe revealed three signals in patients with trisomy 8 detected with the array platform. Two patients with LCSH in the short arm of chromosome 9 p24 were homozygous for JAK2 mutation as detected by pyrosequencing. STR analysis using the polymorphic marker YNZ 22 confirm the allelic loss of at least one allele in the 17 p13.3 region. Results of SNP, FISH, pyrosequencing and STR analysis were completely congruent. These validation results suggest that SNP analysis reflected genomic changes. Patients with late onset GvHD (> day 100 post transplantation) had a significant higher number of GA after transplantation (p<0.05). No significant association was found between acquired GA and the rest of the clinical variables analyzed including outcome. Our results indicate that GA in patients undergoing HCT are likely to play a role. LCSH in chromosome 6p can result in immune escape by leukemic cells from graft-versus-leukemia effect that can lead to relapse. In addition homozygosity in mutated genes such as FLT3 (13 q12) and JAK2 (9 p24) due to deletion or LCSH has important implication for selecting a treatment in patients that relapse after HCT. ABSTRACT Background Observational investigations into the health impacts of low-calorie sweeteners (LCSs) in humans fail to adequately identify or fully characterize LCS consumption. Objectives We aimed to utilize a novel biomarker approach to investigate exposure to 5 LCSs and to test whether reported low-calorie sweetened beverage (LCSB) consumption effectively identifies exposure to LCSs in adults. Methods In this cross-sectional analysis, 2 population studies were conducted in adults. Urinary excretions of 5 LCSs, namely acesulfame-K, saccharin, cyclamate, sucralose, and steviol glycosides, were simultaneously determined using LC tandem-MS. In Study 1, previously collected 24-h urine samples (n = 357) were analyzed. In Study 2, previously collected 24-h urine samples (n = 79) were analyzed to compare urinary excretions of LCSs with self-reported LCSB consumption for identifying LCS exposure. Exposure to LCSs was characterized using descriptive statistics and chi-square tests were performed to assess associations between age-groups and LCS excretion, and to assess the proportion of individuals identified as LCS consumers using biomarker data or reported LCSB consumption. Results A total of 341 adults (45% men) and 79 adults (39% men) were included in the final analysis of Studies 1 and 2, respectively. In Study 1, &gt;96% of samples contained ≥1 LCS and almost 60% contained ≥3 LCSs. A greater proportion of younger adults (&lt;40 y old) excreted ≥3 LCSs than older adults (&gt;40 y old) (P &lt; 0.001). In Study 2, a much higher prevalence of LCS consumption was observed using biomarker data (92%) than reported LCSB consumption (6%) (P &lt; 0.001). Conclusions This work indicates widespread exposure to LCSs, suggesting that population-based research to date into LCS exposure and health may be flawed. Therefore, a urinary biomarker approach offers considerable potential for more robust investigations in this area.

The objective of this study was to design and evaluate new primers for species-specific detection of L. infantum chagasi using PCR. Two combinations of primer pairs were established with the aim of obtaining specific amplification products from the L. infantum chagasi 18S rRNA gene. The combinations of the primer pairs and the respective sizes of the PCR products, based on the U422465 GenBank reference sequence of L. infantum chagasi, were: LCS1/LCS3 (259 bp) and LCS2/LCS3 (820 bp). It was concluded that the new PCR assays optimized using the primer pairs LCS1/LCS3 and LCS2/LCS3 were effective for specific detection of L. infantum chagasi, with analytical sensitivity to detect 1 pg/µL of DNA.

Purpose – The purpose of this paper is to explore relationships between social tagging and key poststructuralist principles; to devise and construct an analytical framework through which key poststructuralist principles are converted into workable research questions and applied to analyse Librarything tags, and to assess the validity of performing such an analysis. The research hypothesis is that tagging represents an imperfect analogy for the poststructuralist project. Design/methodology/approach – Tags from LibraryThing and from a library OPAC were compared and constrasted with Library of Congress Subject Headings (LCSH) and publishers’ descriptions. Research questions derived from poststructuralism, asked whether tags destabilise meaning, whether and how far the death of the author is expressed in tags, and whether tags deconstruct LCSH. Findings – Tags can temporarily destabilise meaning by obfuscating the structure of a word. Meaning is destabilised, perhaps only momentarily, and then it is recreated; it might resemble the original meaning, or it may not, however any attempt to make tags useful or functional necessarily imposes some form of structure. The analysis indicates that in tagging, the author, if not dead, is ignored. Authoritative interpretations are not pervasively mimicked in the tags. In relation to LCSH, tagging decentres the dominant view, but neither exposes nor judges it. Nor does tagging achieve the final stage of the deconstructive process, showing the dominant view to be a constructed reality. Originality/value – This is one of very few studies to have attempted a critical theoretical approach to social tagging. It offers a novel methodological approach to undertaking analysis based on poststructuralist theory.

This study aimed to present the advancements in different breeding stages of super sweet corn (SS), from heterotic field corn populations to the single-cross of SS. Two parental field corn populations, four backcrossed SS populations, four SS intervarietal hybrids (SS-IH), eighteen SS single hybrids (SS-SCH) and two controls were evaluated in two environments over two crop years. The SS-IH and SS-SCH categories presented the highest ear yield, and SS-SCH presented the highest ear yield without husk (11.3 t ha-1). Although SS-SCH was the most productive category, it presented the lowest husk coverage quality. The ear yield and husk cover traits expressed the highest heterosis based on the midparents. The hybrid genotype categories (SS-SCH, SS-IH and controls) had the greatest ear length and diameter, as well as plant and ear height. It was possible to conclude that the techniques used to exploit heterosis are efficient in increasing SS yield but not husk coverage, since this trait requires productive parents. Single-crosses, such as LCSH-116 x LP8HS-129 and LCSH-116 x LP8HS-130, stood out as promising genotypes for registration since they recorded high husk coverage yield and quality.

There is growing interest in food and cooking in the United States, and cookbooks are published on every topic. Library standards for subject analysis must accurately represent and organize cookbooks and materials on cooking. This paper describes a research project that examined the subject of cooking in the Library of Congress Subject Headings and the Library of Congress Classification using the work of Hope Olson as a framework. It examined how the subject headings and classification numbers are constructed, how they changed over time, and how national and ethnic cuisines are treated in each standard.

The concept of ‘social tagging’ has gained popularity nowadays due to the emergence of web 2.0 technologies. Those technologies led to the practice of associating metadata with digital resources among users through collaboratively or socially for self-information retrieval. Many researchers have opined that social tags can enhance the use of library collections. The present study was predominantly carried out to compare social tags collected from the LibraryThing website with Library of Congress Subject Heading (LCSH) descriptors collected from the Library of Congress Online Catalogue applied for thousand book titles in the field of Economics. The study also aimed to know whether social tags can be applied in the library database or not. The findings elucidate that users mostly use descriptors (47.39 %) as tags than expert’s usage of tags (12.77 %) as descriptors. Spearman’s correlation suggests that 75 per cent chance where tags and descriptors can be used simultaneously in overlapping terms. The Jaccard similarity coefficient identifies that users and experts use different terminologies to annotate the books. Users and experts use at least one common keyword for major book titles (908). Users mostly sought title based keywords but experts use mostly subject-based terminologies. The study further clarifies that social tags may be incorporated into the library databases but cannot replace LCSHs. The accessibility and usage of documents especially in the field of economics may be enhanced once the notion of social tags is incorporated with the library OPAC.

Congenital adrenal hyperplasia (CAH) is a group of autosomal recessive disorders caused by enzyme deficiencies required for cortisol biosynthesis in the adrenal cortex. The majority of CAH are due to the deficiency of the 21-hydroxylase enzyme, while 3β-hydroxysteroid dehydrogenase type 2 deficiency accounts for less than five percent of all CAH cases. We report two Moroccan twins from a spontaneous triplet pregnancy. The 46,XY newborn exhibited a disorder of sexual differentiation (DSD) with hypo virilization, while the 46,XX newborn had normal female external genitalia. In the first week of life, they showed hyponatremia and primary adrenal insufficiency with a slight 17OHP elevation and increased DHEAS and renin levels. The aCGH-SNP analysis disclosed a 8.36 Mb long contiguous stretch of homozygosity (LCSH) on chromosome 1p13.2-p11.2 including the candidate HSD3B2 gene, a LCSH of 7.3 Mb on 14q31.1-q32.11, and a 7 Mb duplication on 10q22.3-q23.2. Clinical exome sequencing revealed the biallelic c.969T > G (p.Asn323Lys) HSD3B2, likely pathogenic, variant in both of the affected twins. This case emphasizes the importance of a prompt molecular diagnosis performed through the combination of aCGH and clinical exome, both for establishment of correct therapy and for follow-up, as the newborns also carry a genomic rearrangement with possible clinical implications.

The information retrieval system contains either a list of subject terms (taxonomy) or a list of collaborative tags (folksonomy) or both. The taxonomy and folksonomy come together as called hybrid subject devices. The main purpose of this paper is to apply machine learning techniques in the dataset from the library domain like others and analyse a large quantity of data for critical problems with accuracy. This research reveals to perform EDA (Exploratory data analysis), prediction analysis, and similarity measurement between folksonomy and taxonomy terms with new emerging technologies. Data science deals with big data that means unstructured data, messy data, a large volume of data. The size is of a large amount of data in terms of GB, TB. Machine learning tools manage this type of data. Usually, the Excel, or other spreadsheets package could not manage the file size in GB or TB, and that’s why ML tools, and techniques are applied. At present, the library science domain also contains a large amount of data like 20/30 years of circulation data or subject descriptors, collaborative tags etc. Library professionals can apply machine learning tools for analysing this kind of data in the library domain. In this paper, the authors have introduced the applications of tools and techniques in the library domain and they have tested with 2642 taxonomy and folksonomy terms. This research work includes – EDA, prediction analysis, and similarity measurement of a folksonomy and taxonomy dataset. In the EDA part, the research work has performed a lot of analysis that includes frequency of LCSH (Library of Congress Subject Heading - taxonomy) terms, pair plots, joint plots, and heat map of LCSH and folksonomy terms. The logistic regression (LR) model for prediction analysis has been used in the folksonomy and taxonomy dataset. These 2642 terms of folksonomy and taxonomy both terms are taken as data for this research work. The EDA has been performed with the attributes in the dataset. The accuracy value of logistic regression (f1- score) is 0.37 at the training percentage of 69. The percentage of similarity between LCSH terms and folksonomy terms is 30 per cent (0.30151134), and the angle between these two vectors is 27 degrees. The novelty of this research work is that library data has been analysed using machine learning techniques the ever used before.

Since the 1950s, the Library of the Université Laval has devoted a great deal of effort and time to its subject headings list. This list, the Répertoire de vedettes-matière (RVM), is an adaptation of LCSH. Sources from specialised thesauri such as the MeSH and AAT supplement the headings. Following a brief historical overview of the RVM, the methodology for integrating AAT descriptors into RVM is discussed.This article is a revised version of a paper delivered to the 23rd Annual Conference of ARLIS/NA, Montréal, March 10th-15th, 1995.

Archival representations strive to convey the original context around historical collections, but perspectives on certain topics and materials may change over time. Such shifting perspectives pose problems for providing appropriate subject access without obscuring contexts or misrepresenting resources in their broader cultural settings. This study seeks to examine these problems through an investigation of historical materials associated with American sideshows, public opinions on which changed drastically from the 19th to the 20th centuries. Using as a case the Ronald G. Becker Collection of Charles Eisenmann Photographs, an archival collection of digitized images of sideshow performers, we explored a variety of systems of subject representation for major concepts and terms relevant to sideshow performers and activities. Combining keyword extraction and descriptive analysis of current collection metadata (e.g., LCSH) with comparisons to other popular systems of subject representation, we found complex and conflicting perspectives on concepts such as dwarfism, hypertrichosis, and obesity. Results revealed that current LCSH-based subject metadata for these materials tends to reduce historical and demographic context around performers and highlight contemporary medical perspectives. At the same time, original language used in these images is now often seen as demeaning, exploitative, and offensive. Furthermore, neither of these perspectives may match with self-identifying language used in contemporary society. Taken together, however, these sets of terminologies and their relationships may provide a more robust representation of changing perceptions and terminologies over time. Findings from this study reveal the potentials of utilizing a faceted approach and multiple subject languages together to further clarify and contextualize archival collections.
