Paraphlomis jinggangshanensis (Lamiaceae), a new species from Jiangxi Province, China, is described and illustrated. The new species is morphologically similar to P. intermedia, but can be easily distinguished from the latter by its cordate leaf base (vs. cuneate, decurrent), stem and calyx tube with glandular hairs (vs. short pubescent), and glabrous anthers (vs. ciliate anthers). A phylogenetic analysis, based on ITS regions, suggests that P. jinggangshanensis represents a separate branch in Paraphlomis and is closely related to Clade II. It is currently known only from Jinggangshan National Natural Reserve. Because of its limited distribution and small population size, the species was assessed as Near Threatened (NT) according to the IUCN Red List Categories and Criteria.

Porcine epidemic diarrhea virus (PEDV) has caused substantial economical loss to the Chinese swine industry. To illustrate the genetic characterization of PEDV circulating in China, 205 clinical samples between 2014 and 2018 were collected from 7 provinces in China. 93.17% (191 of 205) of the intestinal and fecal samples were positive for PEDV. 25 S1 amino acid (aa) together with 27 ORF3 genes from 8 provinces were sequenced and analyzed. The phylogenetic trees based on the S1 and ORF3 genes were constructed by the neighbor-joining method using MEGA 7 software. PEDV prevalence was 86.96% (40 of 46) of the swine farms in the 8 provinces and the PEDV positive rate was 93.17% (191 of 205) in the tested samples. Genetic analysis showed CH-JIANGXI-1-2016 CH-JIAGNXI-2-2016, CH-JIANGXI-3-2016 and CH-JIANGXI-2017 had three notable insertions or deletions occurred at aa 59-62, 160, and 139 (140) when compared to all of the strains in this study; moreover, phylogenetic analysis indicated that the four isolates formed a new branch significantly different from G1a, G1b and Indel subtype based on S1 gene: that is the G1c subtype. More research is needed to determine whether the insertions and deletions had biological influence on the virus. The results acquired in the present study showed the genetic diversity of PEDV circulating in 8 provinces, providing information for the development of new diagnostic methods and new vaccines

A number of disorders that harm pig production are linked to porcine circoviruses, including PCV2. PCV2 infection is a substantial contributor to porcine-circovirus-associated illnesses (PCAS) and the post-weaning multi-systemic wasting syndrome (PMWS), which have a significant negative economic impact on pig production. Additionally, PCV infection has been labeled as a global concern to cattle and wildlife. This study’s objectives were to examine the prevalence of PCV1/2/3 in Jiangxi Province, China, and to clarify the epidemiological significance of wild boar in PCV epidemiology. The 2020 hunting seasons resulted in the collection of 138 wild boar samples for PCV1/2/3 detection, which was followed by the genetic clarification of PCV2 strains. According to our data, 21.7% (30/138) of the population had PCV1 positivity, 22.5% (31/138) had PCV2 positivity, and 5.8% (8/138) had PCV3 positivity. Additionally, 10 out of 138 wild boar samples had PCV1 and PCV2 co-infections, while 5 out of 138 wild boar samples had PCV2 and PC3 co-infections. Nineteen full-length PCV2 genomes measuring 1767 nt were recovered from various animal tissues using conventional PCR. Eighteen out of nineteen PCV2 strains were identified as PCV2b by phylogenetic tree analysis, which was completed by the reference strain HLJ2015 obtained from domestic pigs in 2015. Additionally, one genotype of PCV2d JX11-2020 (MW889021) shared a sub-branch with the referenced strain TJ (AY181946), which was isolated in domestic pigs in 2002. This finding raises the possibility that domestic pigs could contract PCV2 strains from wild boar, posing a serious threat to the Jiangxi province of China’s pig production industry.

Abstract To alleviate the problem that the water level drops more and more seriously in recent years, Jiangxi province proposes to set up regulation gates at each of the estuaries of Ganjiang River to raise the water level in Nanchang city during the dry season. The operation of this project will raise the upstream water level, which means the sediment transport rate will decrease and lead to deposition problems. In order to find out the deposition area and development trend, we carried out the suspended sediment deposition test on an existing physical model. The study shows that the upstream sediment is mainly deposited in the reaches near the two deltas. Some special topographic features are more likely to cause siltation, such as the wide and deep thalweg and the backflow areas at the downstream of convex banks. After the operation of the project, the erosion trend of the upper reaches of the central and southern branches will change to siltation, and the siltation of the upper reaches of the Western branch will be further strengthened. Fifteen years later, with the decrease of the upstream sediment intensity, the sediment siltation phenomenon will gradually move downstream, however, this will be a long process.

The Chinese soft-shelled turtle (Pelodiscus sinensis) is an important aquaculture reptile with rich nutritional and medicinal values. In recent decades, the wild resources of P. sinensis have been depleting due to natural and artificial factors. Herein, we report the complete mitochondrial genome of four P. sinensis strains, including the Japanese (RB) strain, Qingxi Huabie (HB) strain, Jiangxi (JB) strain, and Qingxi Wubie (WB) strain. The nucleotide composition within the complete mitogenomes was biased towards A + T with a variable frequency ranging from 59.28% (cox3) to 70.31% (atp8). The mitogenomes of all four strains contained 13 protein-coding genes (PCGs), 22 tRNAs, 2 rRNAs, 1 control region, and a replication origin region of the L-strand replication (OL), which was consistent with most vertebrates. Additionally, the atp8, nad4l, nad6, and nad3 genes possessed high genetic variation and can be used as potential markers for the identification of these P. sinensis strains. Additionally, all PCGs genes were evolving primarily under purifying selection. Through comparative analysis, it was revealed that most of the tRNAs were structurally different in the TψC stem, DHU stem, and acceptor stem. The length of the tandem repeats in the control region was variable in the four P. sinensis strains, ranging from 2 bp to 50 bp. Phylogenetic analysis indicated that all P. sinensis strains clustered into one branch and were closely related to other Trionychinae species. Overall, this study provides mitochondrial genome information for different P. sinensis strains to support further species identification and germplasm resource conservation.

In the context of economic boom and climate change, monitoring the spatio-temporal dynamics of forest fragmentation induced by disturbances and understanding its corresponding associated factors are critical for developing informed forest management strategies. In this study, based on multi-temporal Landsat images acquired from 1999 to 2020, a SVM classifier was first applied to produce high-accuracy land cover maps in Xinyu City. Next, morphological spatial pattern analysis (MSPA) was implemented to characterize the spatio-temporal patterns of forest fragmentation by producing maps of seven fragmentation components, including the core, islet, perforation, edge, bridge, loop, and branch. Then, both natural and human factors responsible for the observed forest fragmentation dynamics were analyzed using the geo-detector model (GDM). The results showed that over the past two decades, Xinyu City experienced a process of significant forest area loss and exacerbating forest fragmentation. The forest area decreased from 1597.35 km2 in 1999 to 1372.05 km2 in 2020. The areal ratio of core patches decreased by 8.49%, and the areal ratio of edge patches increased by 5.98%. Spatially, the trend of forest fragmentation exhibited a progressive increase from the southern and northern regions towards the central and eastern areas. Large-scale forest core patches were primarily concentrated in the northwestern and southwestern regions, while smaller core patches were found in the eastern and central areas. Notably, human activities, such as distance from the roads and land use diversity, were identified as significantly associated with forest fragmentation. The interaction effect of these factors had a greater impact on forest fragmentation than their individual contributions. In conclusion, Xinyu City possesses the potential to further alleviate forest fragmentation by employing the regional differentiation development strategies: (1) intensive development in the northwest and southern regions; (2) high-density development in the western, northwestern, and southern regions, and (3) conservation development in the southwest, northeast, and east-central regions, thus aligning with the path of local social advancement.

The native language will have a negative transfer on the acquisition of second language. Dialects, as a branch of Chinese, also have an influence on English acquisition. The perception of consonants is one of the important foundations of English learning. This study will adopt discrimination as an experimental method to investigate whether there is a significant difference in the influence of different dialects on the perception of English consonants. Three major dialects in Jiangsu province will be taken as a study case to verify the hypothesis that there is a significant difference on the perception of English consonants in different dialects. This study will fill the gap in the overall research on the impact of three major dialects in Jiangsu province on the perception of English consonants, help English language learners with dialect background to reduce the influence of dialects on them and provide some teaching key points to teachers, especially for English language learners in Jiangsu province.

Short interspersed nuclear elements (SINEs), one type of retrotransposon, are considered to be ideal molecular markers due to their wide distribution in the genome, high copy number, and high polymorphism. Preliminary studies have identified more than 35,000 SINE-retrotransposon insertion polymorphisms (RIPs) in the pig genome. In this study, 18 SINE-RIPs were used to evaluate the genetic variation and population structure of seven native pig populations and two crossbreeds in the Jiangsu Province of China. Two commercial pig breeds (Duroc and Large White) and one Italian native breed (Sicilian Black pig) were selected as the control. The results showed that all 18 SINE-RIPs were polymorphic among these pigs. The Jiangsu native pig populations (Erhualian, Fengjing, Middle Meishan, Mi, Shawutou, Small Meishan, and Huai) were shown to be more polymorphic than the crossbreeds (Sushan and Sujiang) and external breeds (Sicilian Black pig, Large White, and Duroc) based on the expected heterozygosity and polymorphic information content values. Some native pigs, including Small Meishan, Mi, Middle Meishan, and Erhualian, had a higher degree of inbreeding according to the FIS values. Based on the neighbor-joining tree, all of the Jiangsu native pig populations formed one branch, while the three external pig breeds formed the other branches, with the two crossbreeds containing more than 50% external pig ancestry. The Huai pigs were independent of the other Jiangsu native pigs but shared a common ancestor with Sujiang and Mi. The results provide a new perspective on the population structure of these native pig breeds and will assist with the conservation and utilization of Chinese native pigs.

AbstractJiangxi is one of the provinces in China most seriously affected by the haemorrhagic fever with renal syndrome (HFRS) epidemic. The aim of this paper was to systematically explore the HFRS epidemic in Jiangxi from the perspective of Hantavirus (HV) prevalence in rodents and humans and virus molecular characteristics. Individual information on all HFRS cases in Jiangxi from 2005 to 2021 was extracted from the China Information System for Disease Control and Prevention. All S and M fragment sequences of the Seoul virus and Hantan virus strains uploaded by Jiangxi and its neighbouring provinces and some representative sequences from provinces in China or some countries of Southeast Asia with the highest HV prevalence were retrieved and downloaded from NCBI GenBank. Periodogram and spatial autocorrelation were adopted for temporal periodicity and spatial clustering analysis of the HFRS epidemic. Joinpoint regression was utilized to explore the changing morbidity trend patterns of HFRS. Multiple sequence alignment and amino acid variation analysis were used to explore the homology and variation of strain prevalence in Jiangxi. Based on monthly morbidity time series, the periodogram analysis showed that the prevalence of HFRS had periodicities of 6 months and 12 months. Spatial autocorrelation analysis showed that HFRS distributed in Jiangxi was not random, with a “High-High” clustering area around Gaoan County. HFRS morbidity among the 0 ~ 15-year-old and ~ 61-year-old or older populations in Jiangxi increased significantly during the period of 2008–2015. Generally, HFRS morbidity was significantly positively correlated with the index of rat with virus (IRV) (r = 0.742) in the counties surrounding Gaoan from 2005 to 2019. HTNV strains in Jiangxi were in one independent branch, while the SEOV strains in Jiangxi were relatively more diverse. Both the YW89-15 and GAW30/2021 strains shared approximately 85% nucleotide homology and approximately 97% amino acid homology with their corresponding standard strains and vaccine strains. GAW30/2021 and YW89-15 had some amino acid site variations in nucleoprotein, glycoprotein precursor and RNA-dependent polymerase with their corresponding vaccine strains Z10 (HTNV) and Z37 (SEOV). The HFRS epidemic in Jiangxi has obvious temporal periodicity and spatial clustering, and the significant increase in the non-Immunization Expanded Program (EPI) targeted population (children and elderly) suggests that HFRS vaccination in this population needs to be considered. Although applying the EPI played a certain role in curbing the incidence of HFRS in Jiangxi from the perspective of ecological epidemiology, HTNV and SEOV strains prevalent in Jiangxi have some amino acid site variations compared to their corresponding vaccine strains, suggesting that HV variation needs to be continuously monitored in the future to observe vaccine protective efficiency.

In September 2019, approximately 75 to 90% of camphor trees (Cinnamomum camphora) were observed with cankers and branch dieback symptoms in Anyi (N28°32’54’’, E115°37’52’’) and Xinyu (N27°37’38’’, E114°50’25’’) county (Jiangxi Province, China). The symptoms included dark brown to dark, oval-shaped canker lesions, sunken and cracked longitudinally, cracked and evenly swelling, or reddish brown (Figure 1 A-D). Samples were collected from symptomatic branches and were cut into small pieces (ca. 0.5 cm × 0.5 cm × 0.5 cm). Sections were surface sterilized as described by Zhang et al. (2020), then placed on potato dextrose agar amended with 0.01% penicillin and 0.015% streptomycin sulfate and incubated in the laboratory at 25℃ with darkness. After 3 to 5 days, mycelium growing out from tissues were transferred onto PDA medium. In total, 68 fungal isolates including 22 isolates of Diaporthe sp. were obtained from cankers and then were classified into five categories based on morphological characteristics and sequencing of the ITS for morphological representative strains. Pathogenicity tests were conducted in the greenhouse (Figure 1 E-M) and field (Figure 1 N-Q). Branches were surface sterilized and inoculated as described by Prencipe et al. (2017). In the greenhouse, a total of 13 representative isolates (including 6 isolates of Diaporthe sp., 2 isolates of Neofusicoccum sp., 2 isolates of Botryosphaeria sp. and 3 isolates of Colletotrichum sp.) were selected and evaluated using 2-year-old seedlings of camphor tree in pots with 5 replicates per isolate, in which 3 isolates of Collectotrichum sp. had no pathogenicity. Then, two isolates of Diaporthe sp. (Z4 and Z7) were selected for field experiment. In field tests, the same method was used as in the greenhouse. The inoculated and control branches were collected 40 days after inoculation and the fungi were isolated and placed on PDA plates to recover the inoculated fungi and complete Koch’s postulates. Both isolates of Diaporthe sp. produced canker symptoms on the branches. Isolate Z4 caused discoloration also on the branch without wounding. Both isolates produced pycnidia scattered in PDA plates supplemented with stems of alfalfa, were dark brown to black, globose to subglobose (Figure 1 T). Alpha conidia were cylindrical, 5.72-9.98 µm (mean 7.64 µm) × 2.15-3.13 µm (mean 2.69 µm) (n = 30) (Figure 1 S, red arrow), while beta conidia were biguttulate, one-celled, hyaline, non-septate, and 16.21-25.52 µm (mean 21.60 µm) × 0.76~1.65 µm (mean 1.14 µm) (n = 30, green arrow) (Figure 1 S). Five isolates (Z4, S-Z4, P-Z4, Z7 and S-Z7) including those used for pathogenicity test were selected for multi-locus phylogenetic analyses of ITS (White et al., 1990), TEF1-α and TUB2 (Glass et al. 1995) gene sequences, which the accession number was MW036358- MW036362 for ITS, MW052267- MW052271 for TEF1- α, MW052276-MW052280 for TUB2. Based on the phylogenetic tree analysis using IQ-TREE 2, all five isolates were identified as D. eres (Figure 2). D. eres has been reported to cause canker on many different woody plants, such as almond (Holland et al. 2020), peach (Prencipe et al. 2017), hazelnut (Wiman et al. 2019), and so on. However, this is the first report worldwide of D. eres causing disease on Cinnamomum camphora in China.

AbstractAvian leukosis caused by avian leukosis virus (ALV) is one of the most severe diseases endangering the poultry industry. When the eradication measures performed in commercial broilers and layers have achieved excellent results, ALV in some local chickens has gradually attracted attention. Since late 2018, following the re-outbreak of ALV-J in white feather broilers in China, AL-like symptoms also suddenly broke out in some local flocks, leading to great economic losses. In this study, a systematic epidemiological survey was carried out in eight local chicken flocks in Jiangxi Province, China, and 71 strains were finally isolated from 560 samples, with the env sequences of them being successfully sequenced. All of those new isolates belong to subgroup J but they have different molecular features and were very different from the strains that emerged in white feature broilers recently, with some strains being highly consistent with those previously isolated from commercial broilers, layers and other flocks or even isolated from USA and Russian, suggesting these local chickens have been acted as reservoirs to accumulate various ALV-J strains for a long time. More seriously, phylogenetic analysis shows that there were also many novel strains emerging and in a separate evolutionary branch, indicating several new mutated ALVs are being bred in local chickens. Besides, ALV-J strains isolated in this study can be further divided into ten groups, while there were more or fewer groups in different chickens, revealing that ALV may cross propagate in those flocks. The above analyses explain the complex background and future evolution trend of ALV-J in Chinese local chickens, providing theoretical support for the establishment of corresponding prevention and control measures.

In July 2018, a virus (JXLC1806-2) was isolated from Culicoides biting midges collected in Lichuan County, Jiangxi Province, China. The virus isolate showed significant cytopathic effects within 48 hours after inoculation with mammalian cells (BHK-21). JXLC1806-2 virus could form plaques in BHK-21 cells, and the virus titer was 1×105.6 pfu/mL. After inoculation with the virus, suckling mice developed disease and died. The nucleotide and amino sequence analysis showed that the JXLC1806-2 virus genome was composed of S, M and L segments. Phylogenetic analysis showed that the S, M and L genes of JXLC1806-2 virus belonged to the Tete serogroup, Orthobunyavirus, but formed an independent evolutionary branch from the other members of the Tete serogroup. The results showed that the JXLC1806-2 virus, which was named as Lichuan virus, is a new member of Tete serogroup, and this is the first time that a Tete serogroup virus has been isolated in China.

Freshwater fungi refer to the fungi that depend on the freshwater habitats for the whole life cycle or part of their life cycle. In this context, a new aquatic hyphomycete was isolated from decaying wood in a freshwater habitat in Jiangxi Province, China. Dictyochaeta jiangxiensis sp. nov., a new aquatic hyphomycete, is characterised by its unbranched, septate, base-fertile conidiophores with multisepta and single phialide at the apex, brown, sterile seta, monophialidic, subcylindrical conidiogenous cells narrowing below the funnel-shaped collarette, hyaline, unicellular, thin-walled, smooth, guttulate, falcate to subclavate conidia narrowly rounded at both ends with hair-like appendages. Phylogenetically, the new species Dictyochaeta jiangxiensis clustered together with Dictyochaeta brevis MFLU 19-0216 in a well-supported clade, but formed a separate branch. In order to better define the taxonomic status of the new species, a phylogenetic tree of most closely-related taxa in Chaetosphaeriaceae was established, based on multi-locus sequences (ITS and LSU). The novel species is described and illustrated. Newly-generated molecular data of Dictyochaeta jiangxiensis is also provided.

Wilt is one the most serious soil-born fungal diseases of Chinese yam (Dioscorea polystachya Turczaninow cv. Tiegun), affecting plant production in many growing regions in Jiangxi province, China. The average annual incidence of wilt is 45-80%. In 2020, affected plants with wilt symptoms including withered and dried leaves, vascular discoloration, and brown necrotic stem lesions at the soil line or in the crown of the plant (Figure 1A-D) were collected from the Ruichang, Yongfeng and Taihe production areas (four fields per location) in Jiangxi province. A total of fifteen isolates were recovered from the infected stem tissues of Chinese yam and purified by single spore cultures on PDA growth medium. The fifteen isolates were similar in morphology so isolate JXRC11 was selected to be representative of the group. Pure fungal colonies of JXRC11were found to be round, white, with margin entire (Figure 1E). Macroconidia with 3-5 septations were straight to slightly curved, 23.8-40.3 µm in length and 2.6-3.9 µm in width, with predominantly 5-septate macroconidia on carnation leaf agar (CLA) (Figure 1F). However, neither microconidia or chlamydospores were observed on CLA. The morphological characteristics of the isolate were consistent with the description observed previously for Fusarium humuli species complex (Wang et al. 2019). To confirm morphological identification, ITS, CAM, TEF-1α, RPB1 and RPB2 were amplified using the primers ITS5/ITS4 (White et al. 1990), CL1/CL2A (O'Donnell et al. 2000), EF1/EF2 (O'Donnell et al. 1998), Fa/G2R (O'Donnell et al. 2010), and 5f2/11ar (O'Donnell et al. 2010), respectively. BLASTn analysis of the ITS sequence (GenBank accession no. MZ768912), EF-1α (MZ824669), CAM (MZ824670), RPB1 (MZ824672) and RPB2 (MZ824673) alignment showed 99.55%, 99.68%, 99.85%, 97.61% and 99.76% identity to those of F. humuli CQ1039 (MK280845, MK289570, MK289712, MK289840 and MK289724), respectively. Multilocus phylogenetic analyses showed that the sequences of ITS, CAM, EF-1α, RPB1, and RPB2 of the isolate belonged to the incarnatum clade (FIESC-33) of the F. incarnatum-equiseti species complex with an independent branch (Figure 2). Pathogenicity tests were conducted on one-month-old Chinese yam seedlings using a seedling root dip method (Li et al. 2013). The roots and rhizomes of seedlings grown to two meters in height were dipped into spore suspensions (1×106 spores/mL) of isolate JXRC11 for 30 min and then transferred into 20 cm diameter plastic pots containing steam-sterilized soil and placed in a greenhouse under 12 h photoperiod. After 15 d, the inoculated seedlings showed typical wilt symptoms similar to those observed in farm fields, whereas the control remained unaffected (Figure 1G-H). The pathogen was then re-isolated from the infected plants, the re-isolations were identified as F. humuli by sequencing EF-1α, fulfilling the Koch's postulates. It has been reported that the pathogen F. oxysporum Schlecht causes Fusarium wilt in five species of Dioscorea (Nwankiti and Arene, 1978). Moreover, at least 5 species of Fusarium were identified as a causative agent of Chinese yam wilt (Fang et al. 2020). To our knowledge, this is the first report of Fusarium wilt disease on Chinese yam caused by a member of the F. humuli in Jiangxi, China. This report will contribute to developing management strategies to control the disease.

In autumn 2020, leaf blight was observed on rice (Oryza sativa L., variety Zhongzao39, Yongyou9, Yongyou12, Yongyou15, Yongyou18, Yongyou1540, Zhongzheyou8, Jiafengyou2, Xiangliangyou900 and Jiyou351) in the fields of 17 towns in Zhejiang and Jiangxi Provinces, China. The disease incidence was 45%-60%. Initially, water-soaked, linear, light brown lesions emerged in the upper blades of the leaves, and then spread down to leaf margins, which ultimately caused leaf curling and blight during the booting-harvest stage (Fig. S1). The disease symptoms were assumed to be caused by Xanthomonas oryzae pv. oryzae (Xoo), the pathogen of rice bacterial blight. 63 isolates were obtained from the collected diseased leaves as previously described (Hou et al. 2020). All isolates showed circular, smooth-margined, yellow colonies when cultured on peptone sugar agar (PSA) medium for 24h at 28℃. The cells were all gram-negative and rod-shaped with three to six peritrichous flagella; positive for catalase, indole, glucose fermentation and citrate utilization, while negative for oxidase, alkaline, phenylalanine deaminase, urease, and nitrate reductase reactions. 16S rRNA gene sequence analysis from the 6 isolates (FY43, JH31, JH99, TZ20, TZ39 and TZ68) revealed that the amplified fragments shared 98% similarity with Pantoea ananatis type strain LMG 2665T (GenBank JFZU01) (Table S3). To further verify P. ananatis identity of these isolates, fragments of three housekeeping genes including gyrB, leuS and rpoB from the 6 isolates were amplified and sequenced, which showed highest homology to LMG 2665T with a sequence similarity of 95%-100% (Table S3). Primers (Brady et al. 2008) and GenBank accession numbers of gene sequences from the 6 isolates are listed in Table S1 and Table S2. Phylogenetic analysis of gyrB, leuS and rpoB concatenated sequences indicated that the 6 isolates were clustered in a stable branch with P. ananatis (Fig. S2). Based on the above morphological, physiological, biochemical and molecular data, the isolates are identified as P. ananatis. For pathogenicity tests, bacterial suspension at 108 CFU/mL was inoculated into flag leaves of rice (cv. Zhongzao39) at the late booting stage using clipping method. Water was used as a negative control. The clipped leaves displayed water-soaked lesions at 3 to 5 days after inoculation (DAI); then the lesion spread downward and turned light brown. At about 14 DAI, blight was shown with similar symptoms to those samples collected from the rice field of Zhejiang and Jiangxi provinces (Fig. S1). In contrast, the control plants remained healthy and symptomless. The same P. ananatis was re-isolated in the inoculated rice plants, fulfilling Koch’s postulates. In the past decade, P. ananatis has been reported to cause grain discoloration in Hangzhou, China (Yan et al. 2010) and induce leaf blight as a companion of Enterobacter asburiae in Sichuan province, China (Xue et al. 2020). Nevertheless, to the best of our knowledge, this is the first report of P. ananatis as the causative agent of rice leaf blight in southeast China. This study raises the alarm that the emerging rice bacterial leaf blight in southeast China might be caused by a new pathogen P. ananatis, instead of Xoo as traditionally assumed. Further, the differences of occurrence, spread and control between two rice bacterial leaf blight diseases caused by P. ananatis and Xoo, respectively need to be determined in the future.

The genus Taxus is the natural material of the anticancer drug paclitaxel (Xiong et al. 2021). Harvesting sources of paclitaxel from the wild has greatly decreased the population of these trees. One of the taxus species, Taxus × media Rehder, a natural hybrid of taxus trees, has a higher paclitaxel content (Zhou et al. 2019). It has been introduced and cultivated in Sichuan, Chongqing, Yunnan, Zhejiang, Jiangxi, and other places in China. In 2021, approximately 20% of T. media (an average 30% of the affected area per tree) showed obvious shoot and leaf blight symptoms in a plantation of taxus trees (about 40 ha of the planting area), located in Sandaoyan county, Sichuan province, China (GPS, 103°94′60″N, 30°84′97″E). Initially, brown necrotic spots appeared on shoots. Gradually, the spots increased in number, expanded to the leaf attached to the branch, and caused wilting of the shoots and leaves. To identify the pathogen, symptomatic samples were randomly collected. Lesion margins of the diseased leaves and barks were surface sterilized for 1 min in 75% ethanol, rinsed with sterile distilled water three times, dried with sterile filter paper, placed on potato dextrose agar (PDA) amended with streptomycin sulfate (50 mg/liter), and incubated at 28°C in the dark. Six purified fungal isolates were obtained. Collected isolates with similar morphology were described as Botryosphaeria spp. (Zhang et al. 2021). The colonies were initially white, gradually became dark gray with dense erial mycelium after 5 days, and formed black pycnidia (Dimensions, 121.3 to 134.6 μm, n = 5) after 16 days. Conidia were fusiform, aseptate, transparent, and thin-walled (23.6 ± 1.2 × 7.27 ± 1.3 μm, n = 50), similar to B. dothidea (Hattori et al. 2021). For pathogenicity testing, ten 2-year-old seedlings of T. media were selected. Fungal cakes of the isolate Tmsdy-2 were applied to the punctured stems of seedlings and covered with Parafilm. Pieces of sterile medium were used as controls. All the seedlings were incubated at 25 ± 2°C, 50% relative humidity, and 16 h of light in a greenhouse. Four days later, the inoculated seedlings developed brown spots and were blighted in 14 days, with symptoms similar to the original diseased plants. The controls remained healthy. The same fungus was reisolated from the infected tissues and subsequently identified by morphological characteristics and DNA sequence analysis. The pathogenicity test was repeated three times with similar results, confirming Koch’s postulates. For molecular identification, the DNA of the isolates was extracted using a Quick-DNA Extraction Kit (Tiangen Biotech, Beijing). The ITS, LSU, SSU, TUB2, and TEF 1-α genes were amplified with the primer pairs ITS1/ITS4, LR0R/LR05, NS1/NS4 (Li et al. 2018), Bt2a/Bt2b, and EF1-728F/EF1-986R (Hattori et al. 2021), respectively. The generated sequences were deposited in GenBank with accession numbers OQ179939 (ITS), OQ179940 (LSU), OQ179942 (SSU), OQ268596 (TUB2), and OQ268597 (TEF 1-α). BLAST analyses showed >99.65% identity with previously deposited sequences of B. dothidea in GenBank. Based on the maximum likelihood method, phylogenetic analysis revealed 100% bootstrap support values with B. dothidea. The fungus was identified as B. dothidea based on morphological and multilocus phylogenetic analyses. To our knowledge, this is the first report of B. dothidea causing shoot and leaf blight of T. media in China. These results will contribute to developing control strategies for this disease.

Abstract The Tancheng–Lujiang (Tanlu) fault zone is the most active fault zone in eastern China. In this zone, the Anqiu–Juxian fault represents the most recently active fault and has the clearest surface traces and the highest seismic risk. This study comprehensively analyzes the kinematic characteristics of the Jiangsu segment of the Anqiu–Juxian fault using field geological surveys, trenches, shallow seismic reflection surveys, combined borehole section exploration, and middepth seismic reflection surveys. The results show that the Jiangsu segment of the Anqiu–Juxian fault features a single branch in the bedrock outcrop area, with reverse strike-slip motion near North Maling Mountain and Chonggang Mountain and normal strike-slip motion near South Maling Mountain. The sedimentary zone features two normal strike-slip faults (east and western branches), which represent the synsedimentary boundaries of a half-graben rift basin. The kinematic process is represented by rotational movement along the strike-slip fault with a curved path. The resulting tensile and compressive stresses are accommodated by dip-slip movement at both ends of the strike-slip fault. The activity of the Jiangsu segment of the Anqiu-Juxian fault can be divided into two periods. The first period of activity occurred before the later part of the Late Pleistocene, when movement along this curved segment occurred, forming the western branch of the Xinyi segment and the eastern branch of the Suqian segment. The second period of activity started in the later part of the Late Pleistocene and continues today. It is characterized by activity on the western branch of the Xinyi segment and the western branch of the Suqian segment of the Jiangsu segment, while the eastern branch of the Xinyi segment and the eastern branch of the Suqian segment became inactive and can be considered Late Pleistocene faults. The maximum vertical slip rate of the Jiangsu segment of the Anqiu–Juxian fault since the Pleistocene has been 0.28 mm/a. The Jiangsu segment of the Anqiu–Juxian fault formed via dextral strike-slip faulting, mainly due to the southward movement of the region to the east of the fault.

In the S-LBBP stage, it is rsR. The predominant morphology is with r/R waves in S-LBBP, which includes variations like rSR, rsR, rSr, rsr, rR, rs, rS, and R type, making up 96.7% of the total. The r waves in lead V1 are associated with agitated myocardium conducted from the left bundle branch.ConclusionThe initial r-wave in lead V1 may be a marker during the follow-up of patients with selective LBB capture.

Adenia globosa, as an excellent indoor ornamental plant, is planted in Tropical Botanical Museum, Nanjing Zhongshan Botanical Garden, Jiangsu Province, China. In September 2022, a new stem basal rot disease was observed on A. globosa seedlings, being planted here. Stem basal rot were observed on approximately 80% of A. globosa seedlings. The basal stem of cutting seedlings appeared decayed, and stem tip eventually turned dry due to water loss (Figure S1A). To isolate the pathogen, three diseased stems were collected from three cuttings planted in different pots of the Tropical Botanical Museum. The stem sections (3 to 4 mm) were excised from the margins between healthy and diseased tissues, surface sterilized in 75% ethanol for 30 s and 1.5% NaClO for 90 s, rinsed three times in sterilized distilled water, plated on potato dextrose agar (PDA) and incubated at 25℃ in the dark. Pure cultures were obtained by monosporic isolation. Eight isolates were obtained, and all identified as Lasiodiplodia sp.. The colonies morphology of cultures, growing on PDA were cotton-like, the primary mycelia were black gray after 7 days, and the reverse sides of PDA plates were similar to front sides in color (Figure S1B). A representative isolate, QXM1-2 was selected for the further study. Conidia of QXM1-2 were oval or elliptic, with a mean size of 11.6 µm×6.6 µm (n=35). The conidia are colorless and transparent in the early stage, and become dark brown with one-septum in the later stage (Figure S1C). The conidiophores produced conidia after nearly four weeks of cultivation on PDA plate (Figure S1D). The conidiophore was a transparent cylindrical structure, with a size of (6.4-18.2) µm × (2.3-4.5) µm ( n = 35). These characteristics were consistent with the description of Lasiodiplodia sp. (Alves et al. 2008). The internal transcribed spacer regions (ITS), translation elongation factor 1-alpha (TEF1α) and β-tubulin (TUB) genes (GenBank Accession No.OP905639, No.OP921005, and No.OP921006, respectively) were amplified and sequenced with the primer pairs ITS1/ITS4 (White et al. 1990), EF1-728F/EF1-986R (Alves et al. 2008) and Bt2a/Bt2b (Glass and Donaldson 1995), respectively. They had 99.8-100% homology to the ITS (504/505 bp) of Lasiodiplodia theobromae strain NH-1 (MK696029), TEF1α (316/316 bp) of strain PaP-3 (MN840491), and TUB (459/459 bp) of isolate J4-1 (MN172230). A neighbor-joining phylogenetic tree was generated by combining all sequenced loci in MEGA7. The isolate QXM1-2 clustered in the L. theobromae clade with 100% bootstrap support (Figure S2). To test pathogenicity, three A. globosa cutting seedlings that previously had been wounded with a sterile needle were inoculated with 20 μL conidia suspension (1×106 conidia/mL) on the stem base. The seedlings inoculated with 20 µL sterile water was used as the control. All plants were covered with clear polyethylene bags to keep moisture in a greenhouse (25℃, 80% relative humidity). The experiment was repeated three times. After 7 days post-inoculation, typical stem rot were found on the treated cutting seedlings and the control seedlings did not have any symptoms (Figure S1E-F). The same fungus, identified by morphological characteristics and sequencing using ITS, TEF1α and TUB genes, was isolated from the diseased tissues of the inoculated stems to complete Koch's postulates. This pathogen has been reported infecting the branch of castor bean (Tang et al. 2021) and root of Citrus (Al-Sadi et al. 2014). For our knowledge, this is the first report of L. theobromae infecting A. globosa in China. This study provides an important reference for the biology, epidemiology of L. theobromae.
