Objective: Currently, mononuclear cell (MNC) isolation is performed using density gradient methods, which are laborious and time-consuming, requiring a minimum of 10 ml of buffy coat or blood. This method is inadequate for isolating MNCs from small volumes, such as 100 µl. Therefore, we aimed to develop a straightforward, rapid, and cost-effective method for isolating MNCs from minute volumes of blood and buffy coat. Material and Methods: We utilized the MNC isolator to isolate MNCs from microvolumes, such as 100 µl and 500 µl of blood and buffy coat, by simple mixing and centrifugation. The isolated cells were cultured, and ligand-specific magnetic beads were employed to isolate CD4 and CD45 specific cells from the cultured cells. Results: In 15 experiments, the MNC isolator successfully isolated MNCs from small volumes of blood and buffy coat. The cell cultures were established and maintained for up to 3 years without contamination, with the cells remaining healthy. Successful isolations of CD4 and CD45 cells using magnetic beads were achieved, and the cells were successfully frozen and thawed. Conclusion: In this study, we have introduced a simple, cost-effective, and robust method for isolating MNCs from small volumes of buffy coat and blood, suitable for immunological applications and magnetic bead cell isolation. This method is user-friendly and can be adopted in cell culture laboratories worldwide, potentially opening new avenues for the development of novel antibodies, isolation of various cell populations using magnetic beads, and obtaining a large number of T-cells for immunotherapy development, such as CAR T-cell therapies. Aim: The Isolation of nucleic acid is an important step for conducting different molecular assays in many laboratories around the world. It is also a common practice that user is isolating the ribonucleic acid (RNA) from the samples with mini column once and throwing away the supernatant. This makes isolated RNA as limiting factor in many studies as this issue has not been addressed in literature. Therefore, we decided to conduct whether it is a loss of ribonucleic acid during the mini column isolation method. Method: In this research, the mini column isolations were done with different samples of human tissues from placenta and umbilical cords and subsequent isolations of supernatants. Yields and successful isolations of RNA were assessed with spectrometric instrument and real time PCR machine. Results: It was found that there is loss of abundant quantity of RNA during the subsequent isolations. The amount measured with UV spectrometer indicates that some times 2nd and 3rd isolation have more RNA than the first isolation. Realtime PCR for house keeping gene beta actin shows that presence of RNA can be seen up to 6 isolation cycles from supernatants. Conclusion: There is loss of RNA in subsequent isolations with mini column method, therefore it is possible to isolate more RNA from subsequent supernatant isolations. User should do the multiple isolations to get higher yield of RNA.

For the purpose of isolating the vibration transmission from machines to soil and vice versa, this paper presents a new active vibration isolation method, that is, a hybrid vibration isolation method capable of isolating the vibration in the whole frequency range. The hybrid isolation method contains the following three characteristics: (1) a low-pass filter of a conventional elastic support method; (2) a high-pass filter of an active vibration isolation method; (3) suppression of a resonant peak due to an elastic support by a servo damper method; First, this paper presents the principle of the hybrid vibration isolation method and shows a fundamental structure of the system. Then by making the active isolator work as a damper, the suppression of a resonant peak due to an elastic support is achieved. Next, taking into consideration both the control effect and the stability of the system, the design procedure of the hybrid vibration isolation system is shown. Moreover, a control chart to estimate the control effect for suppressing the force transmissibility is presented. Finally, an experiment is carried out, demonstrating that the hybrid isolation method is capable of suppressing the exciting force in almost all the frequency range.

For the purpose of preventing vibration-sensitive optical switches from malfunction caused by broadband vertical vibration, a novel two-stage vibration isolation platform is proposed. The primary stage is a bellows-type isolator of large stroke and low isolation frequency, and the secondary stage is a small-stroke hybrid isolator composed of bellows and voice-coil actuators. In the primary stage, two pre-compressed horizontal bellows and one vertical bellows are used to counter the weight of the switch and to reduce the total height of the isolator. The static properties of the primary stage are analyzed, and the vibration isolation of the platform is investigated. Numerical results indicate that the two-stage platform is effective in isolating vertical vibration. Experiments are also conducted to verify the performance of the platform. It is exhibited that the transmissibility is less than 0 dB over 2 Hz, and the attenuation rate reaches −35 dB/dec at high frequencies. The frequency range of test is 2–200 Hz, and the maximum displacement is 10 mm at 2 Hz. In the secondary stage, the actuators can substantially suppress the resonance peak, and promote isolation performance at low frequencies.

This study was carried out for isolating some pathogenic types of positive and negative bacteria for Gram stain, which cause the infection of the vaginitis in infected women and its diagnosis by routine laboratory methods and confirm the diagnosis using Vitek2compact system by collecting fifty samples of the swab of the cervix in the period between October 2015 till April 2016 from the in patients and out patients women in Salahuddin Hospital for the age groups between (14-50) years.The results of isolation show the percentage of bacterial growth was 78%, it reached samples in 78%.The results of the diagnosis of routine diagnostic tests, which were confirmed by using VITEK comparte2 system that there was a diversity of bacterial species including bacterial species from the source of isolation of vaginal infection, where the Escherichia coli bacteria had the highest isolation rate of isolation in 22%, followed by other bacterial species with different isolations. It was observed that there was a variation in the isolation rates and bacterial infections species of the vagina where bacterial isolates were isolated Proteus mirabilis isolates in 20.5% and as Klebsiella pneumonia showed isolations in 10.3%. For Salmonicida Aeromonas and Staphylococcus hominis Bacteria, the isolates was in 7.7%, While Pseudomonas luteola, Pantoes spp, Pseudomonas oryzihabitans, Ochrobactrum anthropic, Sphingomonas paucimbilis bacteria, isolations were isolated in 5.1 %.All isolates were characterized by their high resistance to ten different types of antibiotics, which represent: Gentamicin, Oxacillin, Tetracyclin, Cefotaxme, Amplicillin Novabiocin, Chloramphenicol, Rifampin, Trimethoprim and Azithromycin. The bacterial isolates gave absolute resistance to 100% of the majority of antibiotics used in our current study, while most isolates were sensitive to antagonists Gentamicin

The isolating system absorbs part of the earthquake energy before transferring it to the structure, by shifting the natural period of the isolated structure. This period shift results in a reduction in the inertial forces. It is clear that the effects of near-fault (NF) ground motions with large velocity pulses can bring the seismic isolation devices to critical working conditions. In this study, two three-dimensional RC buildings with the heights of 9.0m and 21.0m which are supported by Triple Friction Pendulum Bearing (TFPB) isolators are idealized. Various TFPB configurations are selected for isolation systems. There are also viscous dampers to limit the excess deformation of isolators. Nonlinear time history analyses were performed by using OpenSees to study the influence of supplemental dampers on structural responses such as isolator displacements and maximum drifts under ten near-fault ground motion records. The results show noticeable reduction in isolator displacement when using dampers. However, maximum drift rises considerablely. Moreover by increasing the period range or reducing the damping ratio of isolation system, maximum driftreduces but the displacement of isolator increases.

300 samples from different infection sourcel for a patients and revisers in Azadi hospital, Kirkuk generic hospital, and children hospital, from various ages for both genders, to isolate Escherichia coli bacteria and diagnosis it, and study the genetic variety which distribution to clinical samples by (140) urine samples, (45) diarrhea samples, (40) wound swab, (40) burn swab, and (35) from otitis media infection. The growth isolation on the different cultural media was diagnosed from the appearance, microscopical, and cultural characteristics. The results the diagnosis showed that (75) clinical isolation about (38.65%) refers to E. coli bacteria distributed by (9) isolation at ratio (31%) from the burn swab, (6) isolation at ratio (26.1) from wound swab, (4) isolation from otitis media infection (19%), (13) isolation from the stool about (40.6%), and (43) isolation (84.3%) from the urinary tract infection (UTI). The sensitivity of the clinical and ecological isolations was studied for nine antibiotics from different antibiotics, broadly. Where the highest resistance for the antibiotics in the clinical isolations to both (ampicillin and cephalexin) reaching in the rate of (97.3%) and (92%), respectively. While the lowest resistance rate for the clinical isolations was to the (Amikacin) in the rate of (17.3%). It has been selected 24 clinical isolations, and 24 ecological isolations from E. coli isolations depending on here resistance to many of antibiotics. To study the genetic variation for these isolations and the genetic markers it has been used a type of molecular markers which is depending on PCR technical it is: randomly Amplified polymorphic DNA (RAPD).

In order to study isolation of bearings, structure seismic isolating model is established based on new-style isolation bearings. Through the input of seismic excitation, acceleration peak and storey drift are regarded as basic discrimination factor for studying seismic response, and structure seismic isolating model is contrasted with structure seismic non-isolating model. The example shows that the new-style of isolation bearings not only can isolate in the horizontal direction, but also isolation ability is better in the vertical direction.

Two models have previously been commonly used to predict the isolation performance of a hydraulic leveraged dynamic anti-resonance vibration isolator. The models have deficiencies, however, in considering the volumetric stiffness of the isolator. In this paper, a new model is proposed to improve the accuracy by reasonably taking the volumetric stiffness into consideration. The model is validated by a carefully designed experiment. The influence of the volumetric stiffness on the isolation performance of the isolator is investigated by the validated model. The results indicate that the volumetric stiffness of the isolator and its location in a mathematical model have a significant influence on the isolation frequency and the second natural frequency of the isolator. The dependence of the volumetric stiffness on the isolation frequency, the attenuation capacity at the isolation frequency, and the bandgap of the isolator is presented. The investigation will enhance the understanding of the hydraulic leveraged dynamic anti-resonance vibration isolator, and facilitate the design and exploitation of this type of isolator in the field of vibration isolation.

Conventional vibration isolators are usually assumed to be massless for the purpose of modelling. This simplification tends to overestimate the isolator performance because of neglecting the internal resonances (IRs) due to the distributed mass effects in the isolator, which is especially important for lightly damped metallic isolators. Previous research on the problem of IRs is not particularly comprehensive, because it does not clarify the characteristics of the distributed parameter isolator. Furthermore, with the development of active vibration isolation, there is a need to investigate the effects of isolator IRs on the control performance and stability for commonly used control strategies. Effective ways to attenuate these effects are also required. This thesis concerns the active vibration isolation of a piece of delicate equipment mounted on a distributed parameter isolator, which is modelled as different idealised configurations under various types of deformation. The model is first developed to determine the effects of IRs on a single-degree-of-freedom system with a distributed parameter isolator. This analysis is then extended to include the resonance behaviour of the supporting structure. Simple expressions are derived which describe the behaviour of various types of distributed parameter isolator. The parameters which control the isolator performance at various frequencies are clarified theoretically and experimentally. The effects of IRs on control performance and stability of several control strategies are determined and compared. Absolute Velocity Feedback (AVF) control is shown to be the optimal solution to minimise the mean square velocity of the equipment mass supported by a distributed parameter isolator. A stability condition for an AVF control system containing a distributed parameter isolator is proposed. Based on this condition, different approaches to stabilize such a control system are presented. Experimental work is carried out to validate the theoretical results. Based on the improved knowledge of the characteristics of IRs in the distributed parameter isolator, different approaches which can suppress the IRs are proposed. AVF control with more damping in the isolator is demonstrated to be effective in attenuating the IRs theoretically and experimentally. Absolute velocity plus acceleration feedback control and AVF control on a fraction of the isolator length are also shown theoretically to be effective ways to attenuate the IRs and improve the isolation performance over a broad range of frequencies.

My work investigates a lifelong passion for buildings, homes, and architecture. Buildings are more then shelters made of wood and screws; I personify them as if they were living breathing creatures. I’m particularly drawn to rundown and abandoned structures because I empathize with these sad looking buildings that were once majestic. This personal connection exists because I view them as versions of my former self, rough around the edges with a promising interior. Expanding beyond vacant and decaying buildings, I portray my experiences within fabricated architectural systems. I communicate with building materials and architecture through their many layers of physical and socially constructed histories. Drawing from my memories and feelings surrounding my brother’s death and subsequent drug addiction, I’ve focused on the emotions of abandonment, isolation, absence, and loneliness.

Constructed Isolation investigates the development of a collection of architectural devices and operations that respond to and have evolved from the specific nature of residential suburban Perth, Western Australia. These devices explore the potential for ambiguity to create space that transforms the occupant from being a passive observer into a dynamic participant. It collates a collection of projects by iredale pedersen hook architects and examines the devices under the thematic concerns of geometry, place, time and craft. The Masters Thesis identifies the potential for Perth's geographic isolation to manifest as a positive construct, developing a unique architecture that opportunistically reinterprets past traditions to create individual environments. 'Constructed Isolation' is an ambitious idea where identity may begin with the individual dwelling and collectively evolve to create a constellation of considerable influence. It demonstrates how these devices may result in new propositions for suburbia, how big ideas with small-scale projects and a little irrational joyousness may contribute to or at least aspire to create change. These small projects will be viewed as a continuation of Perth's 'Heroic Period of Modernism', the fertile and ambitious but incomplete project from the 1950s and '60s. This will also focus on the development and change that occurred as a result of the reflective practice process, while identifying potential opportunities for the future.

In this study, seismic roof isolation solutions were investigated with a case study of Halkapinar Gymnasium, izmir. In the first part, seismic isolation was explanied and philosophy behind it was given. In the second part, existing seismic roof isolators, elastomeric bearing and viscous damper, were investigated with different support isolation combinations. In the third part, two more types of seismic isolators, lead rubber bearing and friction pendulum isolators, were also analyzed by using the same model and all results were compared with each other and that of non-isolated roof system. Furthermore, bilinear effect of the non-linear isolators were also studied. Finally, all results were compared with each other considering structure responses and effectiveness of the seismic isolators. It has been tried to find the most efficient seismic isolation solution for Halkapinar Gymnasium.

The single-degree-of-freedom (SDOF) system is the most widely used model for vibration isolation systems. The SDOF system is a simple but worthy model because it quantifies many results of an isolation system. For instance, a SDOF model predicts that the high frequency transmissibility increases when the isolator has passive damping although this does not occur for an isolator implementing active damping. A severe limitation of this system is that it cannot be used when the base and/or equipment are flexible. System flexibility has been considered in previous literature but the flexibility has always been approximated which leads to truncation errors. The analysis used in this work is more sophisticated in that it can model the system flexibility without the use of any approximations. Therefore, the true effects of system flexibility can be analyzed analytically. Current literature has not fully explored the choice of mount frequency or actuator placement for flexible systems either. It is commonly suggested that isolators should be designed with a low-frequency mount. That is, the isolator frequency should be much lower than any of the system frequencies. It is shown that these isolators tend to perform best in an overall sense; however, mount frequencies designed between system modes tend to have a coupling effect. That is, the lower frequencies have such a strong interaction between each other that when isolator damping is present, multiple system modes are attenuated. Also, when the base and equipment are flexible, isolator placement becomes a critical issue. For low-frequency mount designs, the first natural frequency can shift as much as 15.6% for various isolator placements. For a mid-frequency mount design, the shift of the first three modes can be as high as 34.9%, 26.6% and 11.3%, respectively, for varying isolator placements. This chapter explores the distinctive features of solitary confinement and the ways in which isolation is endemic in the structure of U.S. prisons. It analyze the expansion of solitary confinement, the debate about the lawfulness of the practice in the federal courts, and recent data on the widespread use of solitary confinement in state and federal prison systems. The U.S. Supreme Court has not prohibited the practice but has licensed courts to inquire about whether a particular version imposes an “atypical and significant hardship” on an individual. If such a showing is made, then prisons must provide some procedural buffers against arbitrary placements. Nation-wide data demonstrate that more than 60,000 individuals were, as of 2018, placed in cells for 15 days or more for 22 hours or more. Almost 4,000 people have been so confined for three years or more. Solitary confinement is thus all too “typical” a facet of prison life. Its commonplace occurrence ought not insulate solitary confinement from the conclusion that it is an illicitly cruel practice that debilitates individuals. The complexity of ending solitary confinement terms not only from its widespread use but also from the ways in which U.S. prisons are committed to many practices that are isolating and disabling of individuals.

A three-dimensional (3D) seismic isolation system for FBR building is under development. The proposed vertical isolation system consists form hydraulic cylinders with water-based liquid and accumulators to support large vertical static load and to realize low natural frequency in the vertical direction. For horizontal isolation, laminated rubber isolator or sliding type isolator will be combined. Because the major part of the feasibility of this isolation system depends on the sealing function and durability of the hydraulic cylinder, a series of feasibility tests of the hydraulic cylinder have been conducted to verify the reliability against seismic load and seismic motion. This paper describes the specification of the seismic isolations system, seismic response characteristics and the results of the feasibility tests of the seal. This study was performed as part of a government sponsored R&D project on 3D seismic isolation.

Issues addressed in this paper are related to the isolation and vibration transmission of vibration-sensitive systems mounted on a flexible support. The normal operation of certain electronic, optical and mechanical systems requires a vibration-free environment. To obtain such an environment, these systems are usually isolated from their supports with soft springs. When an array of such systems is needed, due to space constraints or other reasons, they are typically mounted on a common support, which in practice is flexible. Although such a design generally is effective in isolating vibration from ground support, vibration from one system, due to excitations other than from its support, can easily transmit to nearby systems. The level of the transmitted vibration (also known as vibration interaction) can be very significant, especially when all the systems are designed identically for simplicity and with less damping for effective ground vibration isolation. Isolator frequency separation (decoupling), viscous and viscoelastic damping are studied for the reduction of vibration transmission among the systems and their effects on system isolation are discussed. It is found that although the isolator frequency separation and viscous damping could be used for vibration transmission reduction among the isolated systems on the flexible support, the addition of viscoelastic damping reduces the vibration transmission without sacrificing their isolation performance. The difference between viscous and viscoelastic damping on ground vibration isolation is explained theoretically in the final part of the paper using a one degree of freedom model.

Maintenance procedures conducted on pipelines often require isolation of a section of the line. Valve replacement, tie ins, pipe replacement are a few examples of procedures which normally require the pipeline to be taken out of service and drained down, or solid material stops intrusively applied to the pipeline. Pipeline isolation tools have been available and applied for many years; these tools required pressure differential or tethered control to activate and deactivate the isolating mechanism. While these tools were applied with success, there was no way to accurately monitor this early generation of tool for performance and determine with reliability whether the tool was properly set. These tools had the capability of performing one task, once the tool had completed its task, it was removed from the pipeline and readied for its next application. A new isolation tool has been developed and proven for pipeline application and this paper will discuss the Multi-Set Isolation Tool (Iso-Tool), its unique features, development and application.

By setting the isolation system between the foundation of the house and the foundation ground, it makes it possible to scale back the influence of the earthquake. A sliding isolator, which is compact and has high isolation characteristics, had been developed. This isolation system of the house have found effective and had high performance. This system was applied to the vending machine. Although the sliding isolator has been shown to be effective, it required the wide space around the vending machine. By using the base isolation combined with the air damper, it allowed to use in the narrow space area. It is useful when the vending machine is set in front of the wall. A sliding isolator combined with air damper has high isolation characteristics.

There is no specific guidance available on the use of electrical isolation devices and how to protect against faults. Because of this, the practice of isolation is varied, which at times may lead to a possible loss of pipeline integrity or electrical safety hazards. Electrically isolating devices are common in cathodically protected piping systems. They can be used to electrically isolate the pipeline from terminal facilities and pumping systems, to divide the pipeline into sections for individual CP, or to manage stray current interference. NACE International defines these devices as an Electrical Isolating Joint. This document addresses the conditions when the use of isolating devices is indicated and outlines the conditions when installing an isolating device may either create an adverse condition or require additional measures (such as protective devices) to ensure the proper operation. Installation of electrical isolation (EI) devices may be a requirement for cathodically protected underground pipe. The primary purpose of the isolation is to open an electrical circuit formed by the protected structure and other metallic objects such as terminal facilities, pumping and compressor stations, metering stations, river crossings, etc.

Pipeline isolation tools from three different manufacturers were evaluated using full-scale testing and numerical modeling to evaluate stresses generated in 24-inch diameter pipe material considering tool-induced loads and internal pressure. Finite Element Analysis (FEA) was conducted to calculate stresses and strains considering different pipe sizes, material grades, and internal pressures. The FEA results were used to generate a user-friendly parametric tool that was validated with measurements made using strain gauges installed on the test spools. The program demonstrated that the isolation tools are an effective means for isolating pressures without inducing excessive levels of stress or damage to the internal pipe surfaces.
